ObjectiveThe use of pesticides is one of the root causes of food safety problems. Pesticide exposure and pesticide residues can not only lead to environmental pollution issues but also seriously affect human health. In order to meet the rapid and sensitive detection needs of pesticide residues in agricultural products, a method based on lemon juice reduction to prepare silver nanoparticles (AgNPs) is reported in this research.MethodsFirst, fresh lemon juice was filtered through filter paper and diluted to a 2% lemon juice aqueous solution. Then, a certain concentration of AgNO3 solution, 50 mm NaOH solution were prepared and stored at room temperature. Then, 10 mL ddH2O, 2 mL NaOH, 2 mL 2% lemon juice, and 5 mL AgNO3 solution were mixed. When the solution turned to a clear yellow color, the solution was centrifuged to obtain AgNPs. The morphology and structure of AgNPs were observed by transmission electron microscopy (TEM). In order to verify the successful synthesis of the nanoparticles and the distribution characteristics of the nanoparticles, ultraviolet spectroscopy was used for measurement and analysis, and 4-ATP was used as a SERS probe to preliminarily verify the SERS enhancement performance of AgNPs. Furthermore, the content of the main reducing components in lemon juice, namely ascorbic acid, glucose, and fructose was analyzed. The content of ascorbic acid in lemon juice was determined by high-performance liquid chromatography, and the content of glucose and fructose in lemon juice was determined by UV-visible spectrophotometry. To verify the stability and uniformity of the SERS signal of the nanoparticles, 4-ATP was used as an surface enhancement of raman scattering (SERS) probe for detection analysis. The stability of the SERS performance of the colloidal substrate within 41 days and the SERS performance at temperatures ranging from 0-80 °C were analyzed. Using 4-ATP as the SERS probe, the experimental conditions were optimized for the preparation of AgNPs by the lemon juice method, including pH and AgNO3 concentration. To validate the practical usability of the nanoparticles, the solutions of paraquat and carbendazim and the detection limits of pesticide residues on different fruits and vegetables were detected by SERS.Results and discussionsThe method for preparing AgNPs has the advantages of simple operation, green and easy synthesis. The particle morphology and size of the prepared AgNPs were basically uniform, with a size of about 20 nm. The ultraviolet-visible spectrum of AgNPs solution showed that the absorption peak was about 400 nm and the peak shape was narrow, indicating that the colloidal solution had good homogeneity. The detection limit of 4-ATP as the SERS probe was 10-14 M, indicating that the nanoparticle had a good SERS. In addition, the content of ascorbic acid, the main reducing ingredient, in lemon juice measured by high-performance liquid chromatography (HPLC) was 395.76 μg/mL. The contents of glucose and fructose, which were the main reducing components in lemon juice, were 5.95 and 5.90 mg/mL, respectively. Furthermore, the characterization and analysis results of the AgNPs prepared by the mixed reducing solution prepared according to the concentration data of each component showed that the AgNPs obtained were also uniform in morphology and size, with a diameter of about 20 nm, but the SERS enhancement performance was not as good as that of the AgNPs reduced by lemon juice. The SERS signal uniformity of the AgNPs reduced by lemon juice analyzed results showed that the peak intensity of the SERS spectral of 4-ATP at different sites at the same concentration was not significantly changed for 15 times, and its standard deviation RSD=5.03%, which was much lower than the intersubstrate RSD value (

A methodology for the non-targeted detection of milk powder authenticity using Raman imaging was proposed in the present study. Meanwhile, a novel flow-based discrimination neural network was developed to extract the deep feature of the Raman image of milk powder. Using a combination of possibility distribution transformation and non-volume preserving strategies, feature distribution of authentic milk powder was constructed to distinguish between normal and adulterated milk powder samples. As a result, this method could identify various adulterated samples with an accuracy higher than 97.3%, and the limit of detection was 0.3%. The present methodology was characterized by a wide range of applicability, high precision, convenience and rapidity and could meet the demand of milk powder authenticity detection in practice, which may also provide a new approach for non-targeted detection of the authenticity of other non-homogenous food systems.

Ougan (Citrus Suavissima Hort. ex Tanaka) as a long history of edible and medicinal fruit, has a unique taste, and can clear heat, detoxify, eliminate phlegm and relieve cough. However, because of its slight bitterness, it has a strong regional consumption and is not widely accepted. This review focuses on the preparation technology and content analysis of nutritional and functional components in Ou orange, summarizes the research and development status of the processing products of ou orange, such as traditional "Gan er wen" and tangerine peel, as well as new Ougan enzyme, fruit vinegar and fruit wine, and points out the future research direction in the field of deep processing of ou orange.

Corneal neovascularization is a serious corneal pathological change caused by various factors. The drug delivery system is of great significance for the effective treatment of corneal neovascularization. Herein, we developed and characterized a monolith/hydrogel composite as the triamcinolone acetonide (TA) carrier for curing corneal neovascularization. The composite was prepared by photo-initiated free radical polymerization of multi-methacrylate substituted dodecamine organic molecular cage and post-modified by the sequential photo-initiated free radical polymerization of acrylated gelatin. The globular morphology and structural property of as-prepared composites were evaluated by scanning electron microscopy, Fourier-transform infrared spectroscopy and solid-state cross polarization magic angle spinning carbon-13 nuclear magnetic resonance. Then swelling ratio and the TA loading capacity were investigated then. Compared with gelatin hydrogel, the composites exhibited a decreased swelling ratio and an improved loading capacity. With good biocompatibility, the composite can sustainedly release TA for up to 28 days, and effectively inhibit corneal neovascularization with an alkali burn-induced corneal neovascularization model. Additionally, tandem mass tags-labeled quantitative proteomics were performed to identify differentially expressed proteins between vascularized and devascularized corneas. The Kyoto Encyclopedia of Genes and Genomes enrichment analysis revealed that the inhibition process could be primarily linked to the fibrinolytic system. These results demonstrated the potential of monolith/hydrogel composites as delivery systems in the therapy for biomedical diseases.

Abstract Pseudomonas aeruginosa infection is a severe acute suppurative ulcer that engulfs virtually the entire tissue in a short period and leads to devastating destruction. Antibiotic therapy is a common approach for the prophylaxis and treatment of P. aeruginosa infection. However, it is often associated with serious side effects, complications, and multidrug resistance. Therefore, it has been a long-standing challenge to explore safe and effective methods for controlling P. aeruginosa infection. Herein, tannin-coordinated nanozyme composite-based hybrid hydrogels (TCNH) are developed and characterized for the prophylactic treatment of P. aeruginosa and multidrug-resistant P. aeruginosa infections using mouse keratitis as the animal model. The TCNH eye drops are constructed by photoinitiated free radical polymerization of acetylated gelatin solution containing self-synthesized tannin-coordinated Co3O4/Ag nanozyme composite. The as-prepared TCNH displays good dispersibility, peroxidase-like activity and in vitro/in vivo biocompatibility. The nanozyme composite in TCNH seems to penetrate the interior of bacteria and exhibited significant broad-spectrum antibacterial activity owing to its intrinsic and nanozymic catalytic properties. Furthermore, TCNH eye drops can be successfully applied to treat P. aeruginosa and multidrug-resistant P. aeruginosa keratitis. The findings of this study reveal the potential of tannin-coordinated nanozyme composite-based hybrid hydrogel eye drops for treating infectious diseases. Graphical Abstract

Abstract This study aimed to perform qualitative and quantitative examination of DNA degradation during the digestion process in the mouse gut through PCR, qPCR and short tandem repeat (STR) analysis. Human blood leukocytes were gavaged into the digestive tract in mice. GAPDH, TH01, TPOX and D7S820 genes in the contents of the stomach and small intestine were analyzed with PCR and qPCR at various times pre- and post-gavage. Through STR analysis, 21 human genomic DNA loci were analyzed. The half-life of DNA degradation, and the relationship between the average peak area and digestion time were determined. The PCR results showed bands of amplified genes at pre-gavage (0 min) and post-gavage (40, 80 and 120 min) from the mouse stomach contents, whereas no DNA bands from small intestinal chyme were observed after gavage. The qPCR results revealed a significant decrease in DNA concentrations during 40–120 min in the mouse stomach after gavage. At 120 min, 85.62 ± 8.10% of the DNA was degraded, and the half-life of exogenous DNA degradation in the mouse stomach was 70.50 ± 5.46 min. At various digestion times, almost no target genes were detected in the mouse small intestinal chyme. STR analysis showed a decrease in allele numbers with bowel advancement in the small intestine in mice. The degradation of exogenous DNA was higher in the mouse stomach during the first 2 h, and almost complete degradation was observed within 40 min after entering the small intestine in mice.

The corneal endothelium is critical for maintaining corneal clarity by mediating hydration through barrier and pump functions. Progressive loss of corneal endothelial cells during aging has been associated with the development of Fuchs endothelial corneal dystrophy (FECD), one of the main causes of cornea-related vision loss. The mechanisms underlying FECD development remain elusive. Single-cell RNA sequencing of isolated healthy human corneas discovered 4 subpopulations of corneal endothelial cells with distinctive signatures. Unsupervised clustering analysis uncovered nuclear enriched abundant transcript 1 (NEAT1), a long non-coding RNA (lncRNA), as the top expressed gene in the C0-endothelial subpopulation, but markedly downregulated in FECD. Consistent with human corneas, a UVA-induced mouse FECD model validated the loss of NEAT1 expression. Loss of NEAT1 function by an in vivo genetic approach reproduced the exacerbated phenotype of FECD by ablating corneal endothelial cells. Conversely, gain of function by a CRISPR-activated adenoviral delivery system protected corneas from UVA-induced FECD. Our findings provide novel mechanistic insights into the development of FECD, and targeting NEAT1 offers an attractive approach for treating FECD.

AIM: To explore the expression of cGAS/STING signaling components in Mooren's ulcer (MU). METHODS: Samples were obtained from ten MU patients, and eight residual corneal-scleral rings of healthy donor corneas for controls. Human corneal epithelial cells (HCECs) were used to evaluate the effect of cGAS/STING signaling pathway. Immunohistochemistry (IHC) and Western blot were used to examine the expression of cGAS, STING, and phosphorylated interferon regulatory factor 3 (p-IRF3) in MU tissues. The expression of interferon-β (IFN-β) and interferon-stimulated genes (ISGs) was quantified by real-time polymerase chain reaction (PCR) and enzyme-linked immunosorbent assay (ELISA). RESULTS: The protein levels of cGAS and STING in MU samples were significantly elevated when compared with the healthy controls by Western blot and IHC. After stimulation with cGAMP, real-time PCR and ELISA showed a dramatic increase of IFN-β and ISGs (containing CXCL10, IFIT1, and IL-6) in HCECs. Moreover, HCECs treated with cGAMP was characterized by increased phosphorylation and more nuclear translocation of IRF3. Meanwhile, increased p-IRF3 was observed in MU samples via IHC and Western blot. CONCLUSION: The pronounced expression of cGAS/STING signaling components in the patients with MU and probably contribute to the onset and development of MU.

AIM: To explore the secretome efficacy in tumor necrosis factor (TNF)-α stimulated mouse mesenchymal stem cells (MSCs) in a murine model of corneal limbal alkali injury. METHODS: Corneal limbal stem cell deficiency (LSCD) was created in the eyes of male C57 mice. Concentrated conditioned medium from TNF-α stimulated MSCs (MSC-CMT) was applied topically for 4wk, with basal medium and conditioned medium from MSCs as controls. Corneal opacification, corneal inflammatory response, and corneal neovascularization (NV) were evaluated. Corneal epithelial cell apoptosis, corneal conjunctivation, and inflammatory cell infiltration were assessed with TUNEL staining, CK3 and Muc-5AC immunostaining, and CD11b immunofluorescence staining, respectively. The effect of TSG-6 was further evaluated by knockdown with short hairpin RNA (shRNA). RESULTS: Compared to the controls, topical administration of MSC-CMT significantly ameliorated the clinical symptoms of alkali-induced LSCD, with restrained corneal NV, reduced corneal epithelial cell apoptosis, and inhibition of corneal conjunctivation. In addition, MSC-CMT treatment significantly reduced CD11b+ inflammatory cell infiltration, and inhibited the expression of pro-inflammatory cytokines (IL-1β, TNF-α and IL-6). Furthermore, the promotion of corneal epithelial reconstruction by MSC-CMT was largely abolished by TSG-6 knockdown. CONCLUSION: Our study provides evidence that MSC-CMT enhances the alleviation of corneal alkali injuries, partially through TSG-6-mediated anti-inflammatory protective mechanisms. MSC-CMT may serve as a potential strategy for treating corneal disorders.

The relationship between molecular geometries, crystal structures and charge mobilities of hexathiapentacene (HTP) and three of its derivatives (2Se-HTP, 4Se-HTP, 6Se-HTP) were studied using the density functional theory combined with a hopping mechanism at the molecular and crystal level. The effect of Se substitution on the charge mobility was discussed. The calculated results showed that the derivatives exhibit good planarity and the molecular geometries show little variation during the charge transfer process. The electron mobility is 1.20 cm2 V-1 S-1 for HTP and 2.30 cm2 V-1 S-1 for 6Se- -HTP, which are much larger than the corresponding hole ones, indicating that HTP and 6Se-HTP are good candidates for n-type organic semiconductors. However, 2Se-HTP and 4Se-HTP have comparable hole and electron motilities and are suitable for ambipolar semiconductors.

Corneal wound healing is a highly regulated biological process that is of importance for reducing the risk of blinding corneal infections and inflammations. Traditional eye drop was the main approach for promoting corneal wound healing. However, its low bioavailability required a high therapeutic concentration, which can lead to ocular or even systemic side effects. To develop a safe and effective method for treating corneal injury, we fabricated rutin-encapsulated gelatin hydrogel/contact lens composites by dual crosslinking reactions including in situ free radical polymerization and carboxymethyl cellulose/N-hydroxysulfosuccinimide crosslinking. In vitro drug release results evidenced that rutin in the composites could be sustainedly released for up to 14 days. In addition, biocompatibility assay indicated nontoxicity of the composites. Finally, the effect of rutin-encapsulated composites on the healing of the corneal injury in rabbits was investigated. The injury was basically cured in corneas using rutin-encapsulated composites (healing rate, 98.3% ± 0.7%) at 48 h post-operation, while the damage was still present in corneas using the composite (healing rate, 87.0% ± 4.5%). Further proteomics analysis revealed that corneal wound healing may be promoted by the ERK/MAPK and PI3K/AKT signal pathways. These results inform a potential intervention strategy to facilitate corneal wound healing in humans.

Introduction: Furcation involvement (FI) in multi-rooted teeth is challenging for proper oral hygiene, clinical treatment, and leads to poor prognosis. Traditional treatment modalities often result in sacrificing periodontal bone. Multiple regenerative approaches have been attempted to treat furcation defects, but complete regeneration of the periodontal apparatus in grade III furcation has not been reported. Platelet rich fibrin (PRF) shows great potential in enhancing tissue regeneration, angiogenesis, and prevention of infection. This case report introduces a treatment combining allogenic bone grafts with PRF to treat mandibular grade III furcation lesions with a one-year follow-up. Case presentation: Two patients presented with grade III FIs of the mandibular first molars, with intrabony defects requiring guided tooth regeneration (GTR). PRF was collected from each patient to serve as biologics, by mixing with allogenic bone graft, and packed into the furcation and intrabony defects. The PRF membranes were also used for space maintenance. The twelve-month postoperative follow-up demonstrated quicker tissue healing, significant pocket reduction, clinical attachment gain, as well as radiographic bone fill in both cases. Conclusion: Successful periodontal regeneration of grade III furcation defects can be achieved by using PRF in combination with bone allograft.

Abstract Background Major bleeding and allogeneic transfusion leads to negative outcomes in patients receiving cardiac surgery with cardiopulmonary bypass (CPB). Ulinastatin, a urine trypsin inhibitor, relieves systemic inflammation and improves coagulation profiles with however sparse evidence of its effects on blood loss and allogeneic transfusion in this specific population. Methods In this prospective randomized controlled trial, 426 consecutive patients receiving open heart surgery with CPB were randomly assigned into three groups to receive ulinastatin (group U, n = 142), tranexamic acid (group T, n = 143) or normal saline (group C, n = 141). The primary outcome was the total volume of post-operative bleeding and the secondary outcome included the volume and exposure of allogeneic transfusion, the incidence of stroke, post-operative myocardial infarction, renal failure, respiratory failure and all-cause mortality. A ten-year follow-up was carried on to evaluate long-term safety. Results Compared with placebo, ulinastatin significantly reduced the volume of post-operative blood loss within 24 h (688.39 ± 393.55 ml vs 854.33 ± 434.03 ml MD − 165.95 ml, 95%CI − 262.88 ml to − 69.01 ml, p

Patients were found to experience more pain during their second eye cataract surgery compared with their first eye surgery. This study aimed to explore the inflammatory alterations along time in the fellow eye after the first eye surgery and to reveal the underlying mechanism. Eighty patients with bilateral cataracts were recruited and were divided into four groups based on the time of having the second eye surgery. The second eye aqueous humor samples were collected just before surgery and analyzed by mass spectrometry and PCR array. Cytokine activity was enriched in the aqueous humor of the contralateral eye with granulocyte colony-stimulating factor CSF3 significantly upregulated at both gene and protein levels. Rabbits with or without superior cervical ganglionectomy (SCGx) were subjected to lensectomy to mimic human situations. In both human and rabbit models, the fellow eye CSF3 peaked at 1 week post the first eye surgery. Consistently, more neutrophils were recruited to the contralateral eye aqueous humor. Corneal sensitivity and trigeminal electrophysiology were recorded to imply the pain severity in rats receiving capsulorrhexis with or without SCGx. A more intense pulse was detected in the contralateral trigeminal ganglion after the rat received one eye surgery. SCGx could effectively reduce the fellow corneal sensitivity and trigeminal nerve pain. These alterations were under direct regulation of the sympathetic nerves on the surgical eye side. Our results suggest that CSF3 and sympathetic activity could serve as potential analgesic targets during ocular surgeries.

Diabetes mellitus (DM) is a complex metabolic disorder. Long-term hyperglycemia may induce diabetic keratopathy (DK), which is mainly characterized by delayed corneal epithelial regeneration. MicroRNAs (miRNAs) have been reported to play regulatory roles during tissue regeneration. However, the molecular mechanism by which miRNAs influence epithelial regeneration in DK is largely unknown. In this study, we performed miRNA and mRNA sequencing of regenerative corneal epithelium tissue from streptozotocin-induced type 1 diabetic (T1DM) and wild-type mice to screen for differentially expressed miRNAs and mRNAs. Based on regulatory network analysis, miR-223-5p was selected for subsequent experiments and Hpgds was then identified as a direct target gene. MiR-223-5p downregulation significantly promoted diabetic corneal epithelial wound healing and nerve regeneration. However, the beneficial effects of miR-223-5p inhibition were abolished by an Hpgds inhibitor. Furthermore, mechanistic studies demonstrated that miR-223-5p suppression ameliorated inflammation and enhanced cell proliferation signaling in DK. Taken together, our findings revealed that the regulatory role of miR-223-5p in diabetic corneal epithelial and nerve regeneration by mediating inflammatory processes and cell proliferation signaling. And silencing miR-223-5p may contribute to the development of potential therapeutic strategies for DK.

To overcome the disadvantage of clinical manual sleep staging, a convenient, economical, and efficient multi-class automatic sleep staging method is proposed based on long short-term memory network (LSTM) using single-lead electrocardiogram signals. From electrocardiogram signals, heart rate variability and respiratory signals were calculated, and, then, totally 25 features were extracted. Four different classifiers, including the two-class classifier to distinguish between wake and sleep, the three-class classifier to distinguish wake, non-rapid eye movement sleep, and rapid eye movement, the four-class classifier to distinguish wake, light sleep, slow wave sleep, and rapid eye movement, and the five-class classifier to distinguish wake, sleep stage N1, sleep stage N2, sleep stage N3, and rapid eye movement, were constructed using the LSTM. The single-lead electrocardiogram data from 238 patients with full sleep stages during sleep were used for the training set and the data from other 60 patients were regarded as a validation set. The rest of 75 patients have left aside for testing set. The accuracy of two-class, three-class, four-class, and five-class sleep staging was 89.84%, 84.07%, 77.76% and 71.16% and the Cohen's kappa statistic k was 0.52, 0.58, 0.55, and 0.52, respectively, which realized the moderate agreement with clinical analysis. When expanding the dataset to extra 1068 patients with missing sleep stages, the accuracy has no obvious reduction but the Cohen's kappa statistic k dropped to 0.51, 0.52, 0.48 and 0.43, respectively. The proposed method, in this paper, is promising for low-cost, efficient, and convenient sleep staging in home care monitoring.

Abstract The aging of many mammalian tissues is associated with loss of functional adult stem cells, especially bone marrow-derived mesenchymal stem cells (BMSCs). This study was aimed to analyze the biological effect of chick embryo (CE) and nutrient mixture (NM) on the BMSCs of aging rats. The aging rat model was established to be induced by D-galactose (500 mg/kg/d) for 90 days. Meanwhile, aging rats were fed with CE and NM in different dose manner by intragastric administration. At the end of the experimental period, serum was collected from rats and used for BMSCs culture. Flow cytometric analysis was used to investigate the BMSCs surface markers. Alizarin Red and oil red O staining were performed to evaluate the multi-lineage differentiation of BMSCs. The results showed that CE plus NM increased the telomere length of BMSCs and promoted BMSCs proliferation. Moreover, CE plus NM administration promoted BMSCs differentiation into osteoblasts and suppressed differentiation into adipocytes. High-throughput sequencing analysis revealed that there were 326 genes were up-regulated and 59 genes were down-regulated in BMSCs of aging rats treated with CE plus NM. In conclusion, CE plus NM supplement had potential to delay aging through the recovery of BMSCs senescence and could be used as a safe effective approach for nutritional therapy of anti-aging.

Small ‘aircraft’ and other systems, such as timely early warning, all-weather monitoring, strong expulsion and safety capture, combine the two information together to build a model of accurate information, rapid organisation, well-informed decision-making, well-managed and effective safety and prevention, to ensure the safety of the prison.

AIM:To solidify the involvement of Saa-related pathway in corneal neovascularization (CorNV). The pathogenesis of inflammatory CorNV is not fully understood yet, and our previous study implicated that serum amyloid A (Saa) 1 (Saa1) and Saa3 were among the genes up-regulated upon CorNV induction in mice.METHODS:Microarray data obtained during our profiling project on CorNV were analyzed for the genes encoding the four SAA family members (Saa1-4), six reported SAA receptors (formyl peptide receptor 2, Tlr2, Tlr4, Cd36, Scarb1, P2rx7) and seven matrix metallopeptidases (Mmp) 1a, 1b, 2, 3, 9, 10, 13 reportedly to be expressed upon SAA pathway activation. The baseline expression or changes of interested genes were further confirmed in animals with CorNV using molecular or histological methods. CorNV was induced in Balb/c and C57BL/6 mice by placing either three interrupted 10-0 sutures or a 2 mm filter paper soaked with sodium hydroxide in the central area of the cornea. At desired time points, the corneas were harvested for histology examination or for extraction of mRNA and protein. The mRNA levels of Saa1, Saa3, Fpr2, Mmp2 and Mmp3 in corneas were detected using quantitative reverse transcription-PCR, and SAA3 protein in tissues detected using immunohistochemistry or western blotting.RESULTS:Microarray data analysis revealed that Saa1, Saa3, Fpr2, Mmp2, Mmp3 messengers were readily detected in normal corneas and significantly up-regulated upon CorNV induction. The changes of these five genes were confirmed with real-time PCR assay. On the contrary, other SAA members (Saa2, Saa4), other SAA receptors (Tlr2, Tlr4, Cd36, P2rx7, etc), or other Mmps (Mmp1a, Mmp1b, Mmp9, Mmp10, Mmp13) did not show consistent changes. Immunohistochemistry study and western blotting further confirmed the expression of SAA3 products in normal corneas as well as their up-regulation in corneas with CorNV.CONCLUSION:SAA-FPR2 pathway composing genes were expressed in normal murine corneas and, upon inflammatory stimuli challenge to the corneas, their expressions were up-regulated, suggesting their roles in pathogenesis of CorNV. The potential usefulness of SAA-FPR2 targets in future management of CorNV-related diseases deserves investigation.

Load-normalized strain energy increments between consecutive load steps are aggregated through the Kreisselmeier-Steinhauser (KS) function, and the KS function is proposed as a stiffness criterion of geometrically nonlinear structures. A topology optimization problem is defined to minimize the KS function together with the perimeter of structure and a volume constraint. The finite element analysis is done by remeshing, and artificial weak material is not used. The topology optimization problem is solved by using the level set method. Several numerical examples in two dimensions are provided. Other criteria of stiffness, i.e., the end compliance and the complementary work, are compared.

Hyperosmolarity has been recognized as an important pathological factor in dry eye leading to ocular discomfort and damage. As one of the major neuropeptides of corneal innervation, substance P (SP) has been shown to possess anti-apoptotic effects in various cells. The aim of this study was to determine the capacity and mechanism of SP against hyperosmotic stress-induced apoptosis in cultured corneal epithelial cells. The cells were exposed to hyperosmotic stress by the addition of high glucose in the presence or absence of SP. The results showed that SP inhibited hyperosmotic stress-induced apoptosis of mouse corneal epithelial cells. Moreover, SP promoted the recovery of phosphorylated Akt level, mitochondrial membrane potential, Ca2+ contents, intracellular reactive oxygen species (ROS) and glutathione levels that impaired by hyperosmotic stress. However, the antiapoptotic capacity of SP was partially suppressed by Akt inhibitor or glutathione depleting agent, while the neurokinin-1 (NK-1) receptor antagonist impaired Akt activation and ROS scavenging that promoted by SP addition. In conclusion, SP protects corneal epithelial cells from hyperosmotic stress-induced apoptosis through the mechanism of Akt activation and ROS scavenging via the NK-1 receptor.

Artificial intelligence (AI) has the potential to support self-regulated learning (SRL) because of its strong anthropomorphic characteristics. However, most studies of AI in education have focused on cognitive outcomes in higher education, and little research has examined how psychological needs affect SRL with AI in the K-12 setting. SRL is a self-directed process driven by psychological factors that can be explained by the three basic needs of self-determination theory (SDT), i.e., autonomy, competence, and relatedness. This study fills a research gap by examining the moderating effects of need satisfaction and gender in predicting SRL among Grade 9 students. The results indicate that girls perceive more need support than boys. In predicting SRL, satisfaction of the need for autonomy and competence is moderated by both gender and AI knowledge, whereas satisfaction of the need for relatedness is moderated by gender only. Particularly among girls, the effects of autonomy and competence more strongly predict SRL when AI knowledge is low. These findings confirm the gender differences in need satisfaction when predicting SRL with a chatbot. The findings have implications for both teacher instruction and the design and development of intelligent learning environments.

The anthropomorphic characteristics of artificial intelligence (AI) can provide a positive environment for self-regulated learning (SRL). The factors affecting adolescents' SRL through AI technologies remain unclear. Limited AI and disciplinary knowledge may affect the students' motivations, as explained by self-determination theory (SDT). In this study, we examine the mediating effects of needs satisfaction in SDT on the relationship between students' previous technical (AI) and disciplinary (English) knowledge and SRL, using an AI conversational chatbot. Data were collected from 323 9th Grade students through a questionnaire and a test. The students completed an AI basic unit and then learned English with a conversational chatbot for 5 days. Confidence intervals were calculated to investigate the mediating effects. We found that students' previous knowledge of English but not their AI knowledge directly affected their SRL with the chatbot, and that satisfying the need for autonomy and competence mediated the relationships between both knowledge (AI and English) and SRL, but relatedness did not. The self-directed nature of SRL requires heavy cognitive learning and satisfying the need for autonomy and competence may more effectively engage young children in this type of learning. The findings also revealed that current chatbot technologies may not benefit students with relatively lower levels of English proficiency. We suggest that teachers can use conversational chatbots for knowledge consolidation purposes, but not in SRL explorations.

Real-world problems serve as authentic learning contexts for students to integrate science, technology, engineering, and mathematics (STEM) knowledge, enhancing their interest and fostering their identity in STEM. Primary school students' unfamiliarity with real-world problems can be addressed through community engagement; however, research in this area is insufficient. This study used self-determination theory (SDT) to examine and explain the effect of community engagement on student STEM interest and identity development. The mixed-methods sequential explanatory design was adopted, and two experimental groups were set up: community engagement (CE; N = 72) and non-community engagement (NCE; N = 69). The CE group reported significantly higher needs satisfaction levels with autonomy, competence, and relatedness and greater STEM interest and identity. The CE group also expressed that the autonomy encouraged them to learn beyond the classroom and helped them engage with the community; that they were able to build better products through iterations and feedback from users; and that they experienced meaningful learning by creating useful products for others. These findings support the pedagogical value of community engagement projects to foster autonomous motivation. This study explicates how young students' STEM interest and identity could be fostered using SDT contextualized as a community-based STEM engagement project. Through this study, educators worldwide are provided with practical suggestions on using the tested approach.
