In order to understand the role of GA in heterosis of Brassica napus at seeding stage and effect of temperature on GA synthesis. In this study, the hybrid combination YG2009 × YC4 with strong yield advantage of hybrid F1 and its parents YG2009 and YC4 were selected as experimental materials, which grew at 5, 15 and 25°C, respectively. The relationship between agronomic traits and total GA content of hybrid F1 and parents at 20 days of seedling age and the trend of total GA content with temperature were analyzed, and the relative transcription level of key enzyme genes in GA synthesis pathway was analyzed by qPCR. The results showed that temperature significantly affected the GA content in B. napus seedlings, which in turn affected the growth of seedlings. With the increase of temperature, the phenotypic traits such as plant height, leaf number, leaf width, leaf length, fresh weight and dry weight of different materials showed a significant upward trend. The total GA content in plants changed significantly with temperature. Canonical correlation analysis showed that phenotypic traits were positively correlated with the total GA content, that is, the total GA content increased, which was beneficial to the growth of B. napus seedlings. The total GA content in YG2009 × YC4 plants increased with the increase of temperature, and reached the highest at 25°C. The total GA content in YG2009 × YC4 plants was 83.01 % higher than that of female parent YG2009 and 100.72 % higher than that of male parent YC4, reaching a significant difference level. At 25°C, the relative transcription level analysis of key enzyme genes of GA metabolism in different parts showed that there were different transcriptions of key enzyme genes in different parts of B. napus, resulting in a large difference in the total GA content in each part. Grey correlation analysis showed that the relative transcription level of GA2ox6 had the highest grey correlation degree with the total GA content in roots, the relative transcription level of GID1 had the highest grey correlation degree with the total GA content in stems, and the relative transcription level of GA20ox4 had the highest grey correlation degree with the total GA content in leaves. It was speculated that the relative transcription levels of GA2ox6, GID1 and GA20ox4 had a great influence on the total GA content in plants. [ABSTRACT FROM AUTHOR]

Planting density is one of the most important factors to improve repeseed yield, generally 30x104-45x 104 plants/hm² is recommended in national variety trials and farmer planting in China. However, poor weather and soil conditions will affect the emergence of rape seedlings. For a long time, rape farmers have formed the sowing habit of greatly increasing the sowing amount, for example, the sowing amount of farmers in Hubei, Hunan and Jiangxi reaches 7.5kg/hm². High seeding rate can produce more than 180x104 plants/hm². In period of emergence, if current rapeseed varieties can adapt to high seeding rate is unclear. In this study, 54 rapeseed varieties planted with 7.5 kg/hm² seeding rate which simulated farmer's seeding habit. The results showed that a large range genetic variation existed in no-effective plant density, plant pods, effective plant density and yield. Under high seeding rate, plant height, number of seeds per silique, plant weight and effective plant density were significantly correlated with yield. Compared to the low-yield type varieties, high-yield type varieties increased 67.6% in yield. Multivariate correlation analysis showed that effective plant density, seed number per silique and plant dry weight had a significant relationship with yield, effective plant density had the largest path coefficient on yield (0.6849), followed by plant dry weight, plant height, seed number per silique and seed weight. Different varieties could form different effective plant density, 70x104-120x104 plants/ hm² effective plant density ranges would produce higher seed yield. In order to adapt to the high seeding rate, we should select suitable varieties with higher seeding rate field trial. For genetic improving and variety breeding, we should focus on the same traits such as strong forming ability of effective plant density, seed and pod number, which related to early vigor, plant dry weight, seed number per silique, seed weight. Two quasi-periodic oscillations in the tropical atmosphere with similar oscillation period--the stratospheric quasi-biennial and the Southern oscillations--and the relationship between these two oscillations are examined using the Principal Oscillation Pattern (POP) analysis technique. The POP analysis of the equatorial strutospheric dataset provides a compact description of the QBO. The oscillation features identified by the POP analysis, namely, the spatial structure, the characteristic times of the oscillation, and the asymmetry in downward propagation, are almost identical to those found by earlier studies using more conventional analyses. The simultaneous POP analysis of the equatorial zonal surface wind and sea surface temperature indicates a well-defined cyclic behavior of the SO. In contrast to the very regular QBO, the SO appears to be much more noisy with intermittent quiet phases. A spectral analysis of the complex POP coefficient time series and the SO index reveals a negligible correlation between the two processes. A POP analysis of the combined equatorial dataset of stratospheric wind, zonal surface wind, and SST also indicates no relation between the QBO and the SO. Two independent modes are identified, one of them completely describing the QBO and the other representing the entire SO. No linear relationship is found between the two modes either in space or in time. It is concluded that the SO and the QBO are two independent processes in the tropical atmosphere with similar time scales.

Correlation between gene expressions and physiologies was investigated under drought stress using 8 tolerant and susceptive rapeseed materials. The genes included 6 drought - related genes ( btg - 26, BnD22, P5CS, CaM, BADH and ADC). Results showed that the expression profiles were different among the genes between tolerant rape Q2 and susceptive materials Zs6 and S95 - 3. Among the 5 indicators, leaf wilting index (LWI) and relative electrolyte leakage (REL) varied largely among the varieties under drought. Correlation analysis showed that after 3h of drought, P5CS was significantly correlated to LWI and REL, BADH was correlated with REL. After 24h of drought, btg - 2 6 was significantly correlated with relative water content and REL. Results indicated that the above genes could be appraisal indices of drought - tolerance. A description was given to a time-domain technique for real-time estimation of wave-induced vertical bending moment from ship motion measurements. The estimated values can be compared with those obtained from the strain gauge readings to ensure the validity of hull response monitoring systems. Artificial neural networks were used to model the time-domain relationship between wave-induced vertical bending moment and coupled heave and pitch motions. Objective: To study the relationship between the solar term of onset of acute myocardial infarction (AMI) and its syndrome types in traditional Chinese medicine (TCM)., Methods: The clinical data about 430 patients with AMI hospitalized in Foshan Hospital of TCM from February 4th 2003 (Beginning of Spring) to February 3rd 2008 (Beginning of Spring) were collected, and the solar term of onset as angle coordinate was regarded, then the peak phase of the onset solar term in each syndrome type of AMI was calculated by circular statistical analysis., Results: Among 430 patients with AMI, 134 patients were considered to have qi stagnancy and blood stasis syndrome, 188 patients showed the syndrome of turbid sputum obstruction, 29 of them showed deficiency of yin-blood, and 79 showed deficiency of yang qi. The clinical manifestation of AMI was mainly asthenia syndrome (qi stagnancy and blood stasis+turbid sputum obstruction, 74.9%). According to the circular statistical analysis, the peak of the solar terms of AMI onset occurred at the Beginning of Spring in all cases (r=0.127 4, P<0.01), and standard deviation (s)=116.300 6 degree angle, showed it mainly occurred in winter and spring. As the peak of the onset of qi stagnancy and blood stasis occurred at Winter Solstice and Lesser Cold (r=0.200 5, P<0.01), its peak occurred in winter; the turbid sputum obstruction syndrome occurred at Spring Equinox (r=0.147 0, P<0.05), mainly in spring, yet the symptoms of above two peaks were generally mild. Besides, there was no significant difference in onset of the solar term in regard to onset of deficiency of yin-blood and deficiency of yang qi (both P>0.05)., Conclusion: There is a close relationship between periodicity of the solar terms and onset of AMI. The main treatment for AMI is to expel turbid sputum, activate blood to resolve stasis and promote blood circulation to relieve pain; also the method of activating blood to resolve stasis is frequently contemplated in winter, and the method of expelling turbid sputum is the main strategy in spring.

Objective: To construct a subtracted cDNA library of differentially expressed genes in eosinophils from asthma patients., Methods: Suppression subtractive hybridization (SSH) was used to isolate the cDNA fragments of differentially expressed genes in the eosinophils of asthma patients before and after treatment. The cDNA fragments were directly inserted into T/A cloning vector to establish the subtractive library, followed by amplification of the library through E. coli transformation with calcium chloride and screening of blue and white clones of the transformants. One hundred positive bacterial clones were randomly picked and identified by colony PCR., Results: The amplified library contained more than 3,000 positive bacterial clones. Analysis of the randomly selected 100 white clones by PCR showed that 90% of the clones contained 100-500 bp inserts, which might be the cDNA fragments of differentially expressed genes in eosinophils of asthma patients before treatment., Conclusion: A subtracted cDNA library of differentially expressed genes in the eosinophils of asthma patients before and after treatment is constructed successfully by SSH and T/A cloning techniques, which lays a solid foundation for screening and cloning new specific differentially.expressed genes in the eosinophils of asthma patients.

Objective: To study genes that differentially expressed in peripheral blood eosinophils of patient with asthmatic., Methods: Total RNA extracted from eosinophils of patient at the time of exacerbation was taken as the tester and the total RNA from eosinophils of the same patient after improvement as driver. The ds cDNA was synthesized by SMART PCR cDNA Synthesis technology. The differentially expressed genes were obtained by suppression subtractive hybridization (SSH) technology and were further identified by Southern blot., Results: Twelve differentially expressed genes including transformation growth factor beta activated kinase like (TAK1L) protein, cGMP gated channel protein were obtained., Conclusion: Increased expression of these genes were involved in the regulation of proinflammatory response, signal transduction, energy metabolism and cell apoptosis, which may help to clarify the molecular mechanism of eosinophils in asthma and may provide the theoretical base for finding out the new medicines towards eosinophils.

Objective: To explore the molecular mechanism of eosinophils for its role in bronchial asthma., Methods: The total RNA extracted from the eosinophils of patients during the onset of asthma was used as the tester and the total RNA obtained after treatment served as the driver. cDNA suppression subtractive hybridization (SSH) was performed using the protocols described in the Clontech SMART PCR cDNA Syn thesis Kit and PCR-Select cDNA Subtraction Kit. The subtracted cDNA was then inserted into T vector to generate subtracted cDNA library. Clones of the subtracted cDNA library were screened by hybridization and the insert sequence of the positive clones was compared with the sequence in the GenBank., Results: A total of 400 clones selected from the subtracted cDNA library were amplified by PCR and about 85% of these clones contained inserts. Six differential cDNA fragments were acquired after two differential screening. These genes were involved in the regulation of proinflammatory response, signal transduction, energy metabolism and cell apoptosis., Conclusion: Differentially expressed genes of the eosinophils during the onset and the remission stage of bronchial asthma can be effectively cloned by SSH, which provides a solid foundation for clarifying the molecular mechanism of eosinophils in asthma and a theoretical base for clinical treatment and prevention of asthma.
