Abstract Influenza remains a serious issue for public health and it’s urgent to discover more effected drugs against influenza virus. Rhamnocitrin, as a flavonoid, its effect on influenza virus infection remains poorly explored. In this study, rhamnocitrin showed antiviral effect and anti-apoptosis on influenza virus A/Aichi/2/1968 (H3N2) in MDCK cells and A549 cells. In addition, molecular docking revealed that rhamnocitrin have good binding activity with the target proteins cGAS and STING, molecular dynamic simulation and surface plasmon resonance showed that rhamnocitrin could form a stable complex with the above proteins. Moreover, the qPCR and western blot assays further verified that rhamnocitrin could reduce type I IFN and proinflammatory cytokines production by inhibiting the cGAS/STING pathway. Taken together, the results suggest that rhamnocitrin could be a potential anti-viral agent against influenza.

Abstract. Background:. While some studies have reported the impact of COVID-19 vaccines on immune recovery and human immunodeficiency virus (HIV) reservoirs in persons living with HIV (PLWH), their findings remain inconsistent. Furthermore, limited data exist on the safety of multiple COVID-19 vaccines among PLWH. This study aimed to investigate the safety of heterological COVID-19 vaccine administration in PLWH receiving antiretroviral treatment (ART) and its effects on HIV reservoirs and immune recovery. Such an investigation can help provide a basis for vaccinations against COVID-19 in PLWH. Methods:. In this cross-sectional study, PLWH receiving ART were recruited from the outpatient clinic of the Department of Infectious Diseases at the Fifth Medical Center of the Chinese PLA General Hospital from September 1, 2021 to December 31, 2021. Fifty-three PLWH receiving ART were vaccinated against COVID-19, with 35 receiving an inactivated vaccine (IN group) and 18 receiving a recombinant protein (CHO cells) vaccine (CHO group) as the third dose. We assessed the changes in liver and kidney functions, blood lipids, and other laboratory parameters at 0, 1, 2, 4, 7, 8, 10, and 13 months after the first vaccination dose. We also monitored CD4+ and CD8+ T cell counts using flow cytometry at 0, 1, 2, 4, 7, 8, 10, and 13 months after the first vaccination dose, as well as levels of cell-associated HIV DNA (CA HIV DNA) and cell-associated HIV RNA (CA HIV RNA) in peripheral blood using the corresponding quantitative detection kits at 0, 2, and 8 months after the first vaccination dose. Results:. No severe adverse events related to either type of COVID-19 vaccine were observed in HIV patients, irrespective of their CD4+ T cell levels. Both the IN and CHO groups showed significant increases in CD4+ T cell counts during follow-up, CD4+ T cells increased from 438 ± 45 cells/μL to 557 ± 48 cells/μL in the CHO group, while in the IN group, it increased from 448 ± 34 cells/μL to 572 ± 39 cells/μL (both P < 0.001). CA HIV DNA initially decreased and then increased in both groups. CA HIV RNA levels showed significantly decreased in both group after three doses of vaccination (P < 0.01 in CHO group and P < 0.001 in IN group). CD8+ T cells increased from 608 ± 77 cells/μL to 812 ± 116 cells/μL in the CHO group (P < 0.01), while in the IN group, it increased from 702 ± 71 cells/μL to 852 ± 68 cells/μL(P < 0.001) from baseline to the 13 month of following up. Conclusion:. Our data suggest that the SARS-CoV-2 booster vaccine is safe for PLWH on ART, although it may affect HIV reservoirs and CD8+ T cell counts.

Abstract This study examines the impact of geopolitical risk on energy transition, focusing on the moderating roles of environmental regulations and green innovation within OECD countries. By employing a multivariate linear and nonlinear regression model, we identify a substantial positive effect of geopolitical risk on energy transition. Our analysis indicates that stronger environmental regulations and advancements in green innovation significantly amplify this effect. Through threshold effect bootstrap sampling tests, we detect a nonlinear relationship between geopolitical risk and energy transition at varying levels of environmental regulation and green innovation. We also explore lag effects, revealing that the influence of geopolitical risk on energy transition grows stronger over time. The inclusion of interaction terms in our analysis further clarifies the moderating influences of environmental regulation and green innovation. Utilizing a range of geopolitical risk indicators and regression methods, our findings are robust, consistently highlighting the proactive role of geopolitical risk in fostering energy transition. These insights highlight the importance of integrated strategies that harness environmental regulations and technological innovation to facilitate a resilient and efficient energy transition in the face of challenges posed by geopolitical uncertainties.

Rorippa palustris Besser 1821, a species of Brassicaceae, is widely distributed around the world and used for both food and traditional Chinese medicinal purposes. Despite the plant’s significance, its genetic diversity must be better understood. In this study, we have successfully assembled and characterized a complete plastome of R. palustris, marking a significant advancement toward comprehending its genetic composition. The plastome is 154,674 bp long and harbors 128 genes, including 83 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Our phylogenomic analysis indicated that R. palustris is closely related to R. curvipes. These findings are crucial for conserving and utilizing this important plant species. They also highlight the potential for future research into the evolution and preservation of R. palustris, which could be advantageous in pharmaceutical applications.

Objective To understand the prevalence and influencing factors of different types of obesity among middle-aged and elderly Han and Tibetan residents in Gansu province, and to provide evidence for the prevention and control of obesity. Methods Data of 18 114 middle-aged and elderly adults aged 35 to 75 years old with complete body measurement information were analyzed. The data were collected in the baseline survey of "Gansu Province′s Urban and Rural Natural Population Cohort Establishment and Tumor Follow-up Study" of the "Regional Ethnic Cohort Study in Northwest China" conducted from November 2018 to May 2019. Body mass index (BMI), waist circumference (WC), body fat percentage (BFP) and visceral fat index (VFI) were used to assess the prevalence of general obesity, central obesity, body fat obesity, and visceral obesity, respectively. Multivariable unconditional logistic regression analyses were used to explore the influencing factors of different types of obesity. Results Among the 18 114 middle-aged and elderly adults enrolled in Gansu Province, there were 16 999 Han (93.8%) and 1 115 Tibetan (6.2%) ones. The prevalence rates of general obesity, central obesity, body fat obesity, and visceral obesity among middle-aged and elderly Han and Tibetan residents were 12.5% and 9.0%, 31.4% and 33.9%, 50.7% and 41.9%, 40.0% and 27.7%, respectively. The prevalence rates of general obesity, body fat obesity, and visceral obesity among Han adults were higher than those among Tibetan residents (all P < 0.05). The results of multivariable unconditional logistic regression analysis showed that being female was a risk factor for body fat obesity among middle-aged and elderly Han residents and for general obesity, central obesity, and body fat obesity among middle-aged and elderly Tibetan residents in Gansu province, but was a protective factor for general obesity, central obesity, visceral obesity among middle-aged and elderly Han residents and for visceral obesity among middle-aged and elderly Tibetan residents in Gansu province. Age ≥ 45 years old was a risk factor for general obesity, central obesity, body fat obesity, and visceral obesity among middle-aged and elderly Han residents and for body fat obesity among middle-aged and elderly Tibetan residents in Gansu province. Primary school education and above was a protective factor for central obesity among middle-aged and elderly Han residents in Gansu province. Being married was a risk factor for general obesity and visceral obesity among middle-aged and elderly Han residents in Gansu province. Being a non-farmer was a risk factor for general obesity and central obesity among middle-aged and elderly Han residents in Gansu province. Per capita annual household income ≥ 4 000 Yuan was a risk factor for visceral obesity among middle-aged and elderly Han residents in Gansu province. Smoking was a protective factor for general obesity, body fat obesity, and visceral obesity among middle-aged and elderly Han residents in Gansu province. Drinking was a risk factor for central obesity among middle-aged and elderly Tibetan residents in Gansu province. ConclusionThe prevalence rates of general obesity, body fat obesity, and visceral obesity were higher among middle-aged and elderly Han residents than those among middle-aged and elderly Tibetan residents in Gansu province, and gender was a common influencing factor for the four types of obesity among middle-aged and elderly Han and Tibetan residents in Gansu province.

Lindernia crustacea (L.) F. Muell 1882, a species in the Linderniaceae family, holds traditional medicinal value in China. To investigate its genetic diversity, we assembled, annotated, and characterized the first complete chloroplast genome of L. crustacea using Illumina sequencing data and various bioinformatics tools. The genome is 153,647 bp in length, with a GC content of 37.6%. It exhibits a typical quadripartite structure, consisting of a large single-copy region (LSC) of 85,411 bp, a small single-copy region (SSC) of 18,724 bp, and two inverted repeat sequences (IRa and IRb) of 25,816 bp each. The genome was predicted to contain 131 genes, including 87 protein-coding genes, 36 tRNA genes, and eight rRNA genes. Phylogenomic analysis indicated that L. crustacea is closely related to L. stricta. These findings provide a foundation for further research on the evolution and potential medicinal applications of the Linderniaceae family.

ObjectiveExperimental animal models have been used for decades to study the development and progression of chronic obstructive pulmonary disease (COPD). However, there is a lack of methods for constructing animal models of COPD for optimal modelling. This systematic literature review (SLR) aimed to assess the various methods used to establish COPD animal models, highlight their advantages and limitations, and explore more optimized approaches for establishing such models.MethodsA systematic search was performed in four English databases (PubMed, Embase, Web of Science, and the Cochrane Library) and four Chinese databases (Chinese Biomedical Literature Database, China National Knowledge Infrastructure, China Science and Technology Journal Database, and Wanfang Database). Of the 8,015 retrieved full-text manuscripts, 453 were selected.ResultsSmoking (n = 140), smoking combined with lipopolysaccharide (LPS) (n = 275), smoking combined with protease drip (PPE) (n = 10), smoking combined with bacteria (n = 23), and smoking combined with particulate matter (PM2.5) (n = 5) were the most used methods for establishing animal models of COPD. Rats and mice were the most frequently selected experimental animals, with male animals accounting for 79.47% of the total. A total of 92.49 and 29.14% of the articles reviewed considered lung pathology of experimental animals only and lung pathology and lung function tests, respectively.ConclusionOur review suggests that the best way to establish an animal model of COPD is to combine smoking with LPS. Sulfate-reducing Bacteria (SRB) corrosion is a serious threat to the safety of marine pipelines.To reveal the influence of the corrosion behavior of X60 pipeline steel in the presence of SRB cultured in seawater and mud from the East China Sea The corrosion behavior of X60 pipeline steel was studied with weight loss measurements, microstructure and membrane composition analysis, electrochemical measurements The corrosion rate in sterile seawater is 0.11 mm/y, whereas, in SRB-infested seawater, it increases by 245 % to 0.38 mm/y. In sterile sea mud, the corrosion rate is 0.15 mm/y, but in SRB-infested sea mud, it increases by 87 % to 0.28 mm/y. In the corrosive environment of seawater and mud in the East China Sea, SRB significantly accelerates the microbial corrosion of X60 pipeline steel. These findings provide theoretical guidance for further research on SRB corrosion mechanisms and corrosion control of marine pipelines.

This study utilized a rapid detection technique, Isothermal Capture Transient Spectroscopy (ICTS), to identify and evaluate defects related to GaN device surface and buffer traps, and investigated the mechanisms by which they affect the dynamic performance of GaN based devices. Interface states and dielectric bulk traps were analyzed, unveiling distinctive device behavior under positive gate pulse-biased ICTS. The observed S-shaped threshold voltage (VTH) instability and dynamic gate-drain capacitance (CGD) issues were directly associated with these interface states and bulk traps. Negative gate pulse-biased ICTS revealed the presence of logarithmic electron trapping originating from extended electron traps, and spatial wide distributed hole traps within the GaN buffer layer, significantly impacting the dynamic on-resistance (RON) during off-state stress of the devices. The rapid assessment capabilities of this method provide a valuable tool for optimizing GaN epitaxy and manufacturing processes.

Abstract Chironomids are one of the most abundant aquatic insects and are widely distributed in various biological communities. However, the lack of high-quality genomes has hindered our ability to study the evolution and ecology of this group. Here, we used Nanopore long reads and Hi-C data to produce two chromosome-level genomes from mixed genomic data. The genomes of Smittia aterrima (SateA) and Smittia pratorum (SateB) were assembled into three chromosomes, with sizes of 78.45 Mb and 71.56 Mb, scaffold N50 lengths of 25.73 and 23.53 Mb, and BUSCO completeness of 98.5% and 97.8% (n = 1,367), 5.68 Mb (7.24%) and 1.94 Mb (2.72%) of repetitive elements, and predicted 12,330 (97.70% BUSCO completeness) and 11,250 (97.40%) protein-coding genes, respectively. These high-quality genomes will serve as valuable resources for comprehending the evolution and environmental adaptation of chironomids.

Maize leaf spot, which is caused by Curvularia lunata (Wakkre) Boedijn, was epidemic in the maize-growing regions of northeastern and northern China in the mid-1990s, where it led to large yield losses. Since then, the epidemic has evolved into a kind of common disease. In recent years, however, a tendency of becoming an epidemic disease again has been observed in some areas in China due to significant changes in climate, farming, systems and crop varieties. The significance of methyl-(5hydroxymethyl) furan-2-carboxylate (M5HF2C) as a nonspecific host toxin in causing maize leaf spot disease has been demonstrated in previous research. However, the key enzymes involved in M5HF2C toxin synthesis remain unclear. In our study, we demonstrate that the synthesis of M5HF2C toxin starts from a precursor substrate in the pathogen, furfural, which is then catalytically dehydrogenated into furoic acid via an alcohol dehydrogenase (CLADH6). The furoic acid was further confirmed as one of the raw materials for the biosynthesis of M5HF2C toxin based on deletion mutants of the alcohol dehydrogenase gene (Cladh6) in C. lunata, which had reduced M5HF2C toxin-producing ability; however, this ability could be restored in all deletion mutants through complementation with furoic acid, thereby confirming that furoic acid is an intermediate in the biosynthesis of M5HF2C toxin. In summary, the biosynthesis process of M5HF2C toxin in C. lunata involves three transformation steps: (1) from xylose to furfural; (2) then from furfural to furoic acid; and (3) eventually from furoic acid to M5HF2C toxin. Our research findings provide new clues in elucidating the major steps in the process of M5HF2C toxin biosynthesis in C. lunata.

Objective Sulfate-reducing bacteria (SRB) pose a threat to the safe operation of shale-gas-gathering pipelines. Therefore, it is essential to explore the role played by SRB in dedicated pipelines. Methods In this work, the corrosion behavior of SRB was investigated by organic carbon starvation immersion experiments combined with cell number monitoring, corrosion weight loss recordings, morphology and profile observations and electrochemical measurements. Results In experiments with sodium lactate content ranging from 0–3500 ppm, the corrosion rate and pitting depth were the highest at 350 ppm. Conclusions The results indicated that the reduction in carbon sources leads to bacterial starvation, which directly obtains electrons from metals and exacerbates corrosion. It is not appropriate to use the content of bacteria to determine the strength of bacterial corrosion.

For centuries, Laggera pterodonta (LP), a Chinese herbal medicine, has been widely employed for treating respiratory infectious diseases; however, the mechanism underlying LP's effectiveness against the influenza A/Aichi/2/1968 virus (H3N2) remains elusive. This study aims to shed light on the mechanism by which LP combats influenza in H3N2-infected mice. First, we conducted quasi-targeted metabolomics analysis using liquid chromatography-mass spectrometry to identify LP components. Subsequently, network pharmacology, molecular docking, and simulation were conducted to screen candidate targets associated with AKT and NF-κB. In addition, we conducted a series of experiments including qPCR, hematoxylin-eosin staining, flow cytometry, immunohistochemistry, and enzyme-linked immunosorbent assay to provide evidence that LP treatment in H3N2-infected mice can reduce pro-inflammatory cytokine levels (TNF-α, IL-6, IL-1β, and MCP-1) while increasing T cells (CD3+, CD4+, and CD8+) and syndecan-1 and secretory IgA expression. This, in turn, aids in the prevention of excessive inflammation and the fortification of immunity, both of which are compromised by H3N2. Finally, we utilized a Western blot assay to confirm that LP indeed inhibits the AKT/NF-κB signaling cascade. Thus, the efficacy of LP serves as a cornerstone in establishing a theoretical foundation for influenza treatment.

Background Chronic obstructive pulmonary disease (COPD) and hyperuricaemia are both characterised by systemic inflammation. Preventing chronic diseases among the population with common metabolic abnormality is an effective strategy. However, the association of hyperuricaemia with the higher incidence and risk of COPD remains controversial. Therefore, replicated researches in populations with distinct characteristics or demographics are compellingly warranted.Methods This cohort study adopted a design of ambispective hospital-based cohort. We used propensity score matching (PSM) and inverse probability of treatment weighting (IPTW) to minimise the effects of potential confounding factors. A Cox regression model and restricted cubic spline (RCS) model were applied further to assess the effect of serum urate on the risk of developing COPD. Finally, we conducted a two-sample Mendelian randomisation (MR) analysis to explore evidence of causal association.Results There is a higher incidence in the population with hyperuricaemia compared with the population with normal serum urate (22.29/1000 person-years vs 8.89/1000 person-years, p=0.009). Background: Denglao Qingguan decoction (DLQGD) has been extensively utilized for the treatment of colds, demonstrating significant therapeutic efficacy. Human Coronavirus 229E (HCoV-229E) is considered a crucial etiological agent of influenza. However, the specific impact and underlying mechanisms of DLQGD on HCoV-229E remain poorly understood. Methods: Active ingredients and targets information of DLQGD were collected from Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP), literature search, and Swiss ADEM database. The Genecard database was used to collect HCoV-229E related targets. We built an “ingredient-target network” through Cytoscape. Protein - Protein interaction (PPI) networks were mapped using the String database. The Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) were enriched using the DAVID database. Then, we used molecular docking techniques to verify the binding activity between the core compounds and the core gene targets. Finally, in vitro experiments were conducted to validate DLQGD's antiviral activity against HCoV-229E and assess its anti-inflammatory effects. Results: In total, we identified 227 active components in DLQGD. 18 key targets involved in its activity against HCoV-229E. Notably, the core active ingredients including quercetin, luteolin, kaempferol, β-sitosterol, and apigenin, and the core therapeutic targets were CXCL8, RELA, MAPK14, NFKB1, and CXCL10, all associated with HCoV-229E. KEGG enrichment results included IL-17 signaling pathway, Toll-like receptor signaling pathway, RIG-I-like receptor signaling pathway and so on. The core active ingredients and the core therapeutic targets and Human Aminopeptidase N (ANPEP) all showed good binding activity by molecular docking verification. In vitro, DLQGD exhibited anti-HCoV-229E activity and anti-inflammatory effects. Conclusion: Our study suggests that DLQGD has both effects of anti-HCoV-229E and anti-inflammatory. The core active ingredients (quercetin, luteolin, kaempferol, β-sitosterol, apigenin) and the core therapeutic targets (CXCL8, RELA, MAPK14, NFKB1, CXCL10) may play key roles in the pharmacological action of DLQGD against HCoV-229E.

Abstract Background Zostera marina L., or eelgrass, is the most widespread seagrass species throughout the temperate northern hemisphere. Unlike the dry seeds of terrestrial plants, eelgrass seeds must survive in water, and salinity is the key factor influencing eelgrass seed germination. In the present study, transcriptome and proteome analysis were combined to investigate the mechanisms via which eelgrass seed germination was stimulated by low salinity, in addition to the dynamics of key metabolic pathways under germination. Results According to the results, low salinity stimulated the activation of Ca2+ signaling and phosphatidylinositol signaling, and further initiated various germination-related physiological processes through the MAPK transduction cascade. Starch, lipids, and storage proteins were mobilized actively to provide the energy and material basis for germination; abscisic acid synthesis and signal transduction were inhibited whereas gibberellin synthesis and signal transduction were activated, weakening seed dormancy and preparing for germination; cell wall weakening and remodeling processes were activated to provide protection for cotyledon protrusion; in addition, multiple antioxidant systems were activated to alleviate oxidative stress generated during the germination process; ERF transcription factor has the highest number in both stages suggested an active role in eelgrass seed germination. Conclusion In summary, for the first time, the present study investigated the mechanisms by which eelgrass seed germination was stimulated by low salinity and analyzed the transcriptomic and proteomic features during eelgrass seed germination comprehensively. The results of the present study enhanced our understanding of seagrass seed germination, especially the molecular ecology of seagrass seeds.

Abstract The extensive use of neonicotinoids on food crops for pest management has resulted in substantial environmental contamination. It is imperative to develop an effective remediation material and technique as well as to determine the evolution pathways of products. Here, novel ball-milled nitrogen-doped biochar (NBC)-modified zero-valent iron (ZVI) composites (named MNBC-ZVI) were fabricated and applied to degrading neonicotinoids. Based on the characterization results, NBC incorporation introduced N-doped sites and new allying heterojunctions and achieved surface charge redistribution, rapid electron transfer, and higher hydrophobicity of ZVI particles. As a result, the interaction between ZVI particles and thiamethoxam (a typical neonicotinoid) was improved, and the adsorption–desorption and reductive degradation of thiamethoxam and ·H generation steps were optimized. MNBC-ZVI could rapidly degrade 100% of 10 mg·L−1 thiamethoxam within 360 min, its reduction rate constant was 12.1-fold greater than that of pristine ZVI, and the electron efficiency increased from 29.7% to 57.8%. This improved reactivity and selectivity resulted from increased electron transfer, enhanced hydrophobicity, and reduced accumulation of iron mud. Moreover, the degradation of neonicotinoids occurred mainly via nitrate reduction and dichlorination, and toxicity tests with degradation intermediates revealed that neonicotinoids undergo rapid detoxification. Remarkably, MNBC-ZVI also presented favorable tolerance to various anions, humic acid, wastewater and contaminated soil, as well as high reusability. This work offers an efficient and economic biochar-ZVI remediation technology for the rapid degradation and detoxification of neonicotinoids, significantly contributes to knowledge on the relevant removal mechanism and further advances the synthesis of highly reactive and environmentally friendly materials. Falkner-Skan aspects are revealed numerically for a non-homogeneous hybrid mixture of 50% ethylene glycol-50% water, silver nanomaterials Ag, and molybdenum disulfide nanoparticles MoS2 during its motion over a static wedge surface in a Darcy-Forchheimer porous medium by employing the modified Buongiorno model. The Brownian and thermophoresis mechanisms are included implicitly along with the thermophysical properties of each phase via the mixture theory and some efficient phenomenological laws. The present simulation also accounts for the impacts of nonlinear radiative heat flux, magnetic forces, and Joule heating. Technically, the generalized differential quadrature method and Newton-Raphson technique are applied successfully for solving the resulting nonlinear boundary layer equations. In a limiting case, the obtained findings are validated accurately with the existing literature outcomes. The behaviors of velocity, temperature, and nanoparticles volume fraction are discussed comprehensively against various governing parameters. As crucial results, it is revealed that the temperature is enhanced due to magnetic field, linear porosity, radiative heat flux, Brownian motion, thermophoresis, and Joule heating effects. Also, it is depicted that the hybrid nanoliquids present a higher heat flux rate than the monotype nanoliquids and liquids cases. Moreover, the surface frictional impact is minimized via the linear porosity factor. Furthermore, the surface heat transfer rate receives a prominent improvement due to the radiative heat flux inclusion.

Abstract Sm1 and Chit42 of Trichoderma have been universally confirmed as crucial biocontrol factors against pathogen infection through induced resistance and mycoparasitism, respectively. However, not enough work has been conducted to understand the novel function of fused expression of these two proteins in Trichoderma. The results of this study demonstrated that Sm1-Chit42 protein (SCf) engineered T. afroharzianum strain OE:SCf exerted synergistic inhibition to Botrytis cinerea growth at multiple stages of mycoparasitic interaction of T. afroharzianum and B. cinerea including chemotropism sensing, hyphal coiling, hydrophobicity modulation, cell wall adhesion, virulence reduction and pathogen killing by ROS. These results highlight a novel mycoparasitic system in Trichoderma strains engineered with Sm1-Chit42 chimeric protein to combat B. cinerea growth and reproduction, which would lay a strong foundation for exploring a new engineered Trichoderma biofungicide created with chimeric proteins in the future.

Abstract The family Chironomidae is represented by seven subfamilies in China, among which Chironominae and Orthocladiinae are the most diverse. To gain a better understanding of the architecture and evolution of the mitogenomes of Chironomidae, we sequenced mitogenomes of twelve species (including two published species) of the two subfamilies Chironominae and Orthocladiinae, and comparative mitogenomic analyses were performed. Thus, we identified highly conserved genome organization of twelve species with regard to genome content, nucleotide and amino acid composition, codon usage, and gene characteristics. The K a /K s values of most protein-coding genes were far smaller than 1, indicating that these genes were evolving under purifying selection. Phylogenetic relationships between the family Chironomidae were reconstructed using 23 species representing six subfamilies, based on protein-coding genes and rRNAs using Bayesian Inference and Maximum Likelihood. Our results suggested the following relationship within the Chironomidae: (Podonominae + Tanypodinae) + (Diamesinae + (Prodiamesinae + (Orthocladiinae + Chironominae))). This study contributes to the mitogenomic database of Chironomidae, which will be significant for studing the mitogenome evolution of Chironomidae.

BackgroundAccording to GLOBOCAN 2020, lymphoma ranked as the 9th most common cancer and the 12th leading cause of cancer-related deaths worldwide. Traditional diagnostic methods rely on the invasive excisional lymph node biopsy, which is an invasive approach with some limitations. Most lymphoma patients are diagnosed at an advanced stage since they are asymptomatic at the beginning, which has significantly impacted treatment efficacy and prognosis of the disease.MethodThis study assessed the performance and utility of a newly developed blood-based assay (SeekInCare) for lymphoma early detection. SeekInCare utilized protein tumor markers and a comprehensive set of cancer-associated genomic features, including copy number aberration (CNA), fragment size (FS), end motif, and lymphoma-related virus, which were profiled by shallow WGS of cfDNA.ResultsProtein marker CA125 could be used for lymphoma detection independent of gender, and the sensitivity was 27.8% at specificity of 98.0%. After integrating these multi-dimensional features, 77.8% sensitivity was achieved at specificity of 98.0%, while its NPV and PPV were both more than 92% for lymphoma detection. The sensitivity of early-stage (I-II) lymphoma was up to 51.3% (47.4% and 55.0% for stage I and II respectively). After 2 cycles of treatment, the molecular response of SeekInCare was correlated with the clinical outcome.ConclusionIn summary, a blood-based assay can be an alternative to detect lymphoma with adequate performance. This approach becomes particularly valuable in cases where obtaining tissue biopsy is difficult to obtain or inconclusive.

In this paper, numerical simulation for the two-dimensional flow of tangent hyperbolic fluid in the manifestation of nanoparticles, heat source and joule heating is considered. For motivation of problem, the role of Riga plate is investigated. Chemical reaction and Brownian motion are the parts of the analysis. The set of PDEs of Tangent hyperbolic nanofluids with swimming germs is converted in to couple of ODEs by employing well-known approach namely shooting scheme. The resulting highly nonlinear and coupled ODEs are solved by using ND solve tool of Wolfram Mathematica 11. Numerical outcome of involving parameters of interest like Lewis number Le, Prandtl number Pr, Chemical diffusion k, Peclet number Pe, Weissenberg number We, Bioconvection Lewis number Lb, nonlinear radiation, Biot number Bi, modified Hartmann number Q and magnetic parameter M on tangent hyperbolic nanofluids velocity f″, temperature θ, concentration φ and density of germs distribution are discussed in the form of graphical trend and tables. We professed that the speed of fluid degenerated for the increasing value of power-law index n and magnetic parameter M. The Nusselt number growths for growing values of the thermophoresis parameter Nt, Biot number Bi and radiation parameter Nr.

Abstract Environmental exposure to ambient polycyclic aromatic hydrocarbons (PAHs) can disturb the immune response. However, the evidence on adverse health effects caused by exposure to PAHs emitted from specific sources among different vulnerable subpopulations is limited. In this cross‐sectional study, we aimed to evaluate whether exposure to source‐specific PAHs could increase systemic inflammation in older adults. The present study included community‐dwelling older adults and collected filter samples of personal exposure to PM2.5 during the winter of 2011. Blood samples were collected after the PM2.5 sample collection. We analyzed PM2.5 bound PAHs and serum inflammatory cytokines (interleukin (IL)1β, IL6, and tumor necrosis factor alpha levels. The Positive Matrix Factorization model was used to identify PAH sources. We used a linear regression model to assess the relative effects of source‐specific PM2.5 bound PAHs on the levels of measured inflammatory cytokines. After controlling for confounders, exposure to PAHs emitted from biomass burning or diesel vehicle emission was significantly associated with increased serum inflammatory cytokines and systemic inflammation. These findings highlight the importance of considering exposure sources in epidemiological studies and controlling exposures to organic materials from specific sources.

Abstract A steady, incompressible, two-dimensional Sisko-nanofluid flow towards the horizontal direction with no movement in the vertical direction is considered on a stretching/shrinking surface. The power law component (Sisko model) is incorporated under the regime of the porous medium. A magnetic impact is included coming from the MHD in the surface normal direction. In addition, thermal radiation, Brownian diffusion, and thermophoresis are involved in the governing system of equations obtained from the Navier–Stokes model in two-dimensional flow systems. The PDEs are converted into the one-dimensional system using suitable transformations and solved by Galerkin weighted residual method validated with the spectral collocation method. The optimization analysis is performed on heat transfer and skin-friction factors using response surface methodology. The impact of the parameters involved in the model has been testified and is provided in graphical forms. The outcomes indicate that for the values of the porosity factor fluctuating between [0, 2.5], the velocity profile and corresponding boundary layer thickness are lesser towards the maximum value of the parameter, and the results are opposite as the parameter approaches zero. The optimization and sensitivity analysis shows that the transport of heat sensitivity towards thermal radiation, Brownian diffusion, and thermophoresis declined whenever the Nt and Nb increased from low to high and at the medium level of thermal radiation. An increment in the Forchheimer parameter increases the sensitivity of the rate of friction factor, whereas increasing the Sisk-fluid parameter has the reverse effect. Elongation processes like those of pseudopods and bubbles make use of such models. The idea is also widely used in other sectors, such as the textile industry, glass fiber production, cooling baths, paper manufacture, and many more.

Abstract Objective To study the survival prediction value of lymph node ratio (LNR) and preoperative thyroglobulin (Tg) in the prognosis of thyroid papillary carcinoma (PTC). Methods A total of 495 patients with PTC and lymph node metastasis treated at the Cancer Hospital of Xinjiang Medical University were selected for a retrospective study. The disease‐free survival (DFS) of patients was the follow‐up endpoint. DFS was calculated for all patients. The Cox proportional risk regression model and nomogram were used to predict the survival prognosis of PTC with lymph node metastasis by index. LNR and preoperative Tg level cutoff values were obtained using ROC curves. To express DFS, Kaplan–Meier survival curves were created. Using 3‐ and 5‐year calibration curves and AUC values, the prognostic models' precision and discrimination were assessed. Clinical decision curve analysis was used to forecast clinical benefitability. Finally, the results were validated using internal cross‐validation. Results The cutoff values of LNR and preoperative Tg level were 0.295 and 50.24, respectively, and they were divided into two groups according to the cutoff values. Abstract Background There is an urgent need for effective treatment of patients with relapsed/refractory diffuse large B‐cell lymphoma (R/R‐DLBCL). This trial investigated the efficacy of decitabine in combination with rituximab, cisplatin, cytarabine, dexamethasone (RDHAP) in R/R‐DLBCL. Methods 56 patients were divided into two groups (decitabine‐RDHAP group. n = 35; RDHAP group, n = 21). The primary endpoints were the overall response rate (ORR) and duration of remission (DOR). Secondary objectives were toxicity, progression‐free survival (PFS), and overall survival (OS). Results The ORR was 40% and 33% for decitabine‐RDHAP and RDHAP groups, respectively, with no difference between the groups. The DOR for the decitabine‐RDHAP regimen was higher than that for the RDHAP regimen (p = 0.044). After a median follow‐up of 12.0 months, the median PFS and OS were 7.0 and 17.0 months for in the decitabine‐RDHAP group and 5.0 and 9.0 months in the RDHAP group with no significant differences between the two groups (p = 0.47, 0.17). The incidence of adverse events was not significantly different between groups. Conclusion The decitabine‐RDHAP regimen is effective and well tolerated, and is a promising salvage regimen for R/R‐DLBCL.

Motivated by the thermal importance of feeble electrically conducting nanofluids and their flow controls in many industrial and engineering applications, the present scrutinization intended to evidence comprehensively the main electro-magneto-hydrothermal and mass aspects of convective non-homogeneous flows of alumina-based pure water nanofluids Al2O3-H2O over a horizontal flat surface of an electromagnetic actuator (i.e., Riga pattern, which is embedded geometrically in a Darcy-Forchheimer porous medium. Further, the present nanofluid flow model is formulated realistically under the umbrella of the renovated two-phase Buongiorno’s approach with the inclusion of Brownian motion and thermophoresis diffusive phenomena, in which the vertical component of the nanoparticles’ mass flux tend to vanish at the limiting contact surface due to its impermeability trend. For streamlining the technical handling of the present nanofluid flow problem, the governing partial differential equations (PDEs) are simplified mathematically by adopting the physical approximations of the boundary layer theory and then transformed into a differential structure of ordinary differential equations (ODEs) based on several similarity changes. Methodologically, the resulting nonlinear coupled ODEs are solved numerically via a validated differential quadrature procedure. Besides, the generated graphical demonstrations show that the nanofluid temperature is enhanced significantly with the porosity factors, the nanoparticles’ loading, the convective heating strength, and the thermophoresis process. However, the porosity factors and the nanoparticles’ loading exhibit a slowing-down impact on the nanofluid motion. Usefully, it is revealed from the obtained GDQM - NRT datasets that the nanoparticles’ loading and the porosity factors express an important improvement in the strength of the surface viscous drag forces, whereas the induced electromagnetic field shows a reverse viscous frictional impact.

Abstract Background Stratification of DNA mismatch repair (MMR) status in patients with colorectal cancer (CRC) enables individual clinical treatment decision making. The present study aimed to develop and validate a deep learning (DL) model based on the pre-treatment CT images for predicting MMR status in CRC. Methods 1812 eligible participants (training cohort: n = 1124; internal validation cohort: n = 482; external validation cohort: n = 206) with CRC were enrolled from two institutions. All pretherapeutic CT images from three dimensions were trained by the ResNet101, then integrated by Gaussian process regression (GPR) to develop a full-automatic DL model for MMR status prediction. The predictive performance of the DL model was evaluated using the area under the receiver operating characteristic curve (AUC) and then tested in the internal and external validation cohorts. Additionally, the participants from institution 1 were sub-grouped by various clinical factors for subgroup analysis, then the predictive performance of the DL model for identifying MMR status between participants in different groups were compared. Results The full-automatic DL model was established in the training cohort to stratify the MMR status, which presented promising discriminative ability with the AUCs of 0.986 (95% CI 0.971–1.000) in the internal validation cohort and 0.915 (95% CI 0.870–0.960) in the external validation cohort. In addition, the subgroup analysis based on the thickness of CT images, clinical T and N stages, gender, the longest diameter, and the location of tumors revealed that the DL model showed similar satisfying prediction performance. Conclusions The DL model may potentially serve as a noninvasive tool to facilitate the pre-treatment individualized prediction of MMR status in patients with CRC, which could promote the personalized clinical-making decision.

Objective: COPD is the most common chronic respiratory disease with complex environmental and genetic etiologies. It was reported that EPAS1 might participate in the occurrence and development of respiratory diseases. However, the association between EPAS1 and COPD was unclear. Methods: First, a case-control study enrolling 1130 COPD patients and 1115 healthy controls in Guangzhou was conducted to clarify the association between EPAS1 polymorphisms and COPD susceptibility. Secondly, a prevalence study recruited 882 participants in Gansu to verify the effect of positive polymorphisms on lung function. Finally, the 10-year absolute risk considering environmental factors and genetic variations was calculated by the method of Gail and Bruzzi. Results: EPAS1 rs13419896 AA genotype reduced COPD risk in southern Chinese (AA vs. GG: adjusted OR = 0.689, 95% CI = 0.498–0.955; AA vs. GG/GA: adjusted OR = 0.701, 95% CI = 0.511–0.962). Further, the rs13419896 A allele was significantly associated with higher pre-FEV1/pre-FVC in both the Guangzhou and Gansu populations (P A were finally retained to develop a relative risk model for males. Smoking status, biomass as fuels, and rs13419896 G > A were retained in the female model. The population-attributable risk of the male or female model was 0.457 (0.283–0.632) and 0.421 (0.227–0.616), respectively. Conclusions: This study first revealed that EPAS1 rs13419896 G > A decreased COPD susceptibility and could be a genetic marker to predict the 10-year absolute risk for COPD.

Abstract Background DNA mutations of diverse types provide the raw material required for phenotypic variation and evolution. In the case of crop species, previous research aimed to elucidate the changing patterns of repetitive sequences, single-nucleotide polymorphisms (SNPs), and small InDels during domestication to explain morphological evolution and adaptation to different environments. Additionally, structural variations (SVs) encompassing larger stretches of DNA are more likely to alter gene expression levels leading to phenotypic variation affecting plant phenotypes and stress resistance. Previous studies on SVs in rice were hampered by reliance on short-read sequencing limiting the quantity and quality of SV identification, while SV data are currently only available for cultivated rice, with wild rice largely uncharacterized. Here, we generated two genome assemblies for O. rufipogon using long-read sequencing and provide insights on the evolutionary pattern and effect of SVs on morphological traits during rice domestication. Results In this study, we identified 318,589 SVs in cultivated and wild rice populations through a comprehensive analysis of 13 high-quality rice genomes and found that wild rice genomes contain 49% of unique SVs and an average of 1.76% of genes were lost during rice domestication. These SVs were further genotyped for 649 rice accessions, their evolutionary pattern during rice domestication and potential association with the diversity of important agronomic traits were examined. Genome-wide association studies between these SVs and nine agronomic traits identified 413 candidate causal variants, which together affect 361 genes. An 824-bp deletion in japonica rice, which encodes a serine carboxypeptidase family protein, is shown to be associated with grain length. Conclusions We provide relatively accurate and complete SV datasets for cultivated and wild rice accessions, especially in TE-rich regions, by comparing long-read sequencing data for 13 representative varieties. The integrated rice SV map and the identified candidate genes and variants represent valuable resources for future genomic research and breeding in rice.

Abstract Background Trichoderma spp. are important agricultural biocontrol microorganisms that are often used as effective components of microbial fungicides and microbial biofertilizers. However, most of these products are prepared by a single strain in monoculture, which significantly limits the biocontrol efficiency and stability of Trichoderma products. Therefore, the establishment of a design and screening approach for consortia with multi-Trichoderma strains for co-culture is of great importance to overcome the shortage of traditional Trichoderma biocontrol products. Results First, 15 Trichoderma strains were screened in terms of mycelium growth rate, antagonistic activity to a variety of pathogens, stress tolerance to high temperature and salt stress, and cucumber seedling growth promotion level. Then, the combinations of Trichoderma asperellum GDSF1009 (CGMCC NO. 9512), Trichoderma asperelloides Z4-1 (CGMCC NO. 40245), Trichoderma harzianum 10569 (CGMCC NO. 40246), and T. asperellum 10264 (CGMCC NO. 22404) were finally screened as an optimal consortium for co-culture underlying the levels of plant growth-promoting and antagonistic activity to Fusarium oxysporum and seed germination promotion relative to the monoculture of a single strain. Consortia with multiple co-cultured strains were found to generate larger amounts of free amino acids than those from the monoculture of a single strain, and a pot assay also indicated that metabolites of co-cultures were able to promote cucumber seedling growth superior to that with monoculture of a single strain, even though the promotion was better than from simply mixed cultures from each of the four Trichoderma strains. Taken together, the co-culture consortia composed of the four compatible interactive Trichoderma strains was a potential novel multiple strain biocontrol agent based on the combination of synthetic consortia design and co-culture. In the field experiment, we found that the growth-promoting effect of the co-culture fermentation filtrate was better than that of the single culture fermentation filtrate. Compared with T-Z4-1, T-1009, T-10264 and T-10569, the plant height of cucumber was increased by 22.99%, 42.06%, 24.18% and 30.09%, respectively, and the stem diameter was increased by 16.59%, 18.83%, 13.65% and 14.70%, respectively. Conclusion An approach to designing and screening Trichoderma consortia for co-culture was established. The consortia co-culture presented a better performance in antagonistic activity and cucumber growth compared with a monoculture of a single strain. Thus, it is of great significance to lay the foundation for the creation of a novel Trichoderma biofungicide or biomanure to resist cucumber Fusarium wilt and promote cucumber growth.

BackgroundSelecting features related to postoperative infection following cardiac surgery was highly valuable for effective intervention. We used machine learning methods to identify critical perioperative infection-related variables after mitral valve surgery and construct a prediction model.MethodsParticipants comprised 1223 patients who underwent cardiac valvular surgery at eight large centers in China. The ninety-one demographic and perioperative parameters were collected. Random forest (RF) and least absolute shrinkage and selection operator (LASSO) techniques were used to identify postoperative infection-related variables; the Venn diagram determined overlapping variables. The following ML methods: random forest (RF), extreme gradient boosting (XGBoost), Support Vector Machine (SVM), Gradient Boosting Decision Tree (GBDT), AdaBoost, Naive Bayesian (NB), Logistic Regression (LogicR), Neural Networks (nnet) and artificial neural network (ANN) were developed to construct the models. We constructed receiver operating characteristic (ROC) curves and the area under the ROC curve (AUC) was calculated to evaluate model performance.ResultsWe identified 47 and 35 variables with RF and LASSO, respectively. Twenty-one overlapping variables were finally selected for model construction: age, weight, hospital stay, total red blood cell (RBC) and total fresh frozen plasma (FFP) transfusions, New York Heart Association (NYHA) class, preoperative creatinine, left ventricular ejection fraction (LVEF), RBC count, platelet (PLT) count, prothrombin time, intraoperative autologous blood, total output, total input, aortic cross-clamp (ACC) time, postoperative white blood cell (WBC) count, aspartate aminotransferase (AST), alanine aminotransferase (ALT), PLT count, hemoglobin (Hb), and LVEF. The prediction models for infection after mitral valve surgery were established based on these variables, and they all showed excellent discrimination performance in the test set (AUC > 0.79).ConclusionsKey features selected by machine learning methods can accurately predict infection after mitral valve surgery, guiding physicians in taking appropriate preventive measures and diminishing the infection risk.

Chronic obstructive pulmonary disease (COPD) is a complex respiratory disorder influenced by various factors and involving multiple genes. Respiratory dysfunction in COPD patients leads to hypoxia, resulting in limited oxygen uptake. Peroxisome proliferator-activated receptor alpha (PPARA) is a plateau-adapted gene that regulates respiratory function in populations adapted to high-altitude areas through multiple pathways. Interestingly, PPARA expression is higher in long-term inhabiting Tibetan populations that have adapted to the plateau environment. However, in patients with COPD, the expression of PPARA is downregulated, leading to dysregulation of the hypoxia-inducible factor pathway. Moreover, abnormal PPARA expression in lung epithelial cells triggers inflammatory responses, oxidative stress, and disrupted lipid metabolism, thereby exacerbating disease progression. Thus, this paper explored the mechanism underlying the role of plateau-adapted PPARA in COPD, providing essential theoretical insights into the treatment and prevention of COPD in high-altitude regions.

Background: Environmental and genetic factors are jointly involved in the development of chronic obstructive pulmonary disease (COPD). The EGLN1 gene is a major factor in upstream regulation of the hypoxia-inducible pathway. EGLN1 negatively regulates the hypoxia-inducible factors HIF-lα and HIF-2α by regulating the concentration of oxygen, mainly in a hypoxic environment. Hypoxia is a common physiologic condition during the progression of COPD, and several studies have identified genetic variants in EGLN1 as a key factor in the adaptation to hypoxic environments. However, it is still unclear whether there is an association between EGLN1 variants and the risk of developing COPD. Methods: A case-control study was conducted in the Gannan Tibetan Autonomous Prefecture, Gansu Province. A total of 292 COPD patients and 297 healthy controls were enrolled to assess the association of EGLN1 single nucleotide polymorphisms (SNPs) (rs41303095 A>G, rs480902 C>T, rs12097901 C>G, rs2153364 G>A) with COPD susceptibility. Results: The EGLN1 rs41303095 A>G, rs480902 C>T, rs12097901 C>G, and rs2153364 G>A polymorphisms were not associated with COPD susceptibility (p > 0.05). Conclusions: The EGLN1 rs41303095 A>G, rs480902 C>T, rs12097901 C>G and rs2153364 G>A polymorphisms were found in this study not to be associated with susceptibility to COPD in Gannan Tibetans.

This paper proposes an event-triggered fault-tolerant time-varying formation control method dedicated to multiple unmanned aerial vehicles (UAVs). We meticulously design a formation-tracking controller with a predefined tracking performance to accommodate the presence of actuator faults and external disturbances. Firstly, the formation-tracking controller acquires the desired heading using the line-of-sight algorithm. Secondly, in the presence of actuator faults and external disturbances, we introduce the radial basis function neural network (RBFNN) and adaptive law tracking control to effectively compensate for their effects. Additionally, we design adaptive tracking controllers and event-triggering conditions to increase the computational frequency. The predefined tracking performance, implemented via a Lyapunov function, ensures the convergence of the tracking error over time. Finally, we conduct a thorough analysis of the system’s stability, successfully eliminating the possibility of Zeno behavior. The simulation results thoroughly validate the effectiveness of the theoretical analysis.

Abstract Fluid–structure interaction (FSI) gained a huge attention of scientists and researchers due to its applications in biomedical and mechanical engineering. One of the most important applications of FSI is to study the elastic wall behavior of stenotic arteries. Blood is the suspension of various cells characterized by shear thinning, yield stress, and viscoelastic qualities that can be assessed by using non-Newtonian models. In this study we explored non-Newtonian, incompressible Casson fluid flow in a bifurcated artery with a stenosis. The two-dimensional Casson model is used to study the hemodynamics of the flow. The walls of the artery are supposed to be elastic and the stenosis region is constructed in both walls. Suitable scales are used to transform the nonlinear differential equations into a dimensionless form. The problem is formulated and discretized using Arbitrary Lagrangian–Eulerian (ALE) approach. The finite element method (FEM) technique is used to solve the system of equations, together with appropriate boundary conditions. The analysis is carried out for the Bingham number, Hartmann number, and Reynolds number. The graphical results of pressure field, velocity profile, and load on the walls are assessed and used to study the influence of hemodynamic effects on stenotic arteries, bifurcation region, and elastic walls. This study shows that there is an increase in wall shear stresses (WSS) with increasing values of Bingham number and Hartmann number. Also, for different values of the Bingham number, the load on the upper wall is computed against the Hartmann number. The result indicate that load at the walls increases as the values of Bingham number and Hartmann number increase.

Objective: This study aimed to evaluate the safety, efficacy, and feasibility of the rituximab, fotemustine, pemetrexed, and dexamethasone (R-FPD) regimen followed by whole-brain radiotherapy (WBRT) for patients with primary central nervous system lymphoma (PCNSL). Methods: A prospective, single-center phase II clinical trial was conducted. Patients with PCNSL newly diagnosed at the First Affiliated Hospital of Zhengzhou University between July 2018 and July 2020 were studied. The R-FPD regimen consisted of rituximab (375 mg/m2 i.v. on D0), fotemustine (100 mg/m2 i.v. on D1), pemetrexed (600 mg/m2 i.v. on D1), and dexamethasone (40 mg i.v. on D1-5). Patients 60 years or younger who showed a complete response (CR) were treated with 23.4 Gy of WBRT after the end of chemotherapy; those older than 60 years with CR were treated with a wait-and-see approach; and those who did not show CR after the 4th cycle of chemotherapy were given salvage WBRT 30 Gy + local tumor field irradiation up to 45 Gy, regardless of age. Results: A total of 30 patients were included. After 2 cycles, the objective response rate (ORR) was 96.5% (28/29, 1 CR, 27 PR, 0 SD, and 1 PD). After 4 cycles, the ORR was 73.1% (19/26, 11 CR, 8 PR, 4 SD, and 3 PD). After WBRT, the ORR was 90.9% (10/11, 7 CR, 3 PR, and 1 SD). The grade III and IV toxicity responses were mainly leukopenia (20.0%), thrombocytopenia (23.3%), and anemia (10.0%). Conclusions: Fotemustine-based therapy in combination with rituximab chemotherapy followed by WBRT can improve outcomes, providing ORR benefits and favorable tolerability in patients newly diagnosed with PCNSL.

To grasp the geometry characteristics and precision manufacturing method of it, the analytical method for toroidal helix is presented. The mathematical model of toroidal helix is developed, and the geometry characteristics of helix angle, axial tooth thickness, and tooth surface undercutting are analyzed. The helix angle of toroidal helix presents the convex shape, and the undercutting characteristic is affected by the pressure angle, helix angle, transmission ratio, and normal modulus. Based on the toroidal helix of tooth surface, the precision manufacturing method is proposed, and the measuring method is developed. The hourglass worm sample was manufactured and measured, and the transmission accuracy of hourglass worm drive is tested. The results show that the manufacturing and measuring method of toroidal helix is effective and feasible.

BackgroundChronic Obstructive Pulmonary Disease (COPD) is a common and harmful disease that requires an effective tool to early screen high-risk individuals. Gansu has unique environments and customs, leading to the different prevalence and etiology of COPD from other regions. The association between altitude and COPD once attracted epidemiologists' attention. However, the prevalence in Gansu and the role of altitude are still unclarified.MethodsIn Gansu, a multistage stratified cluster sampling procedure was utilized to select a representative sample aged 40 years or older. The questionnaire and spirometry examination were implemented to collect participants' information. The diagnosis and assessment of COPD were identified by the Global Initiative for Chronic Obstructive Lung Disease (GOLD) criterion, while post-bronchodilator FEV1/FVC < LLN was for sensitivity analysis. Furthermore, the effect of high altitude on COPD was evaluated by the logistic regression model after propensity score matching (PSM). Finally, the participants were randomly divided into training and validation sets. The training set was used to screen the relative factors and construct a nomogram which was further assessed by the receiver operating characteristic (ROC) curve, calibration curve, and decision curve analysis (DCA) in the two sets.ResultsThere were 2,486 eligible participants in the final analysis, of which 1,584 lived in low altitudes and 902 lived in high altitudes. Based on the GOLD criterion, the crude and standardized prevalences in Gansu were 20.4% (18.7–22.0) and 19.7% (17.9–21.6). After PSM, the logistic regression model indicated that high altitude increased COPD risk [PSM OR: 1.516 (1.162–1.978)]. Altitude, age, sex, history of tuberculosis, coal as fuel, and smoking status were reserved for developing a nomogram that demonstrated excellent discrimination, calibration, and clinical benefit in the two sets.ConclusionsCOPD has become a serious public health problem in Gansu. High altitude is a risk factor for COPD. The nomogram has satisfactory efficiency in screening high-risk individuals.

Abstract Background Long noncoding RNAs (lncRNAs) are emerging as master regulators for gene expression and thus play a vital role in human tumorigenesis and progression. But the involvement of novel lncRNAs in non-small cell lung cancer (NSCLC) remains largely unelucidated. Methods A total of 170 NSCLC and their adjacent non-tumor tissues were enrolled to detect the expression of Lnc-LSAMP-1 by RT-qPCR. The effects of Lnc-LSAMP-1 on cell proliferation, migration, invasion and drug-sensitivity were determined by in vitro and in vivo experiments. The proteins that interact with Lnc-LSAMP-1were confirmed by RNA pull-down assay. RNA-sequencing were used to identify the potential targets of Lnc-LSAMP-1 in NSCLC. Results We found that Lnc-LSAMP-1 was significantly down-regulated in 170 cases of NSCLC tissues when compared to their adjacent non-cancerous tissues. Loss expression of Lnc-LSAMP-1 was notably correlated with unfavorable prognosis of NSCLC patients. The ectopic expression of Lnc-LSAMP-1 drastically inhibited lung cancer cell proliferation, viability, invasion and migration ability, arrested cell cycle and facilitated apoptosis. Chemotherapy sensitization experiments showed that over-expressed Lnc-LSAMP-1 enhanced the inhibition of cell proliferation induced by TKI. Mechanistically, Lnc-LSAMP-1-LSAMP formed a complex which could protect the degradation of LSAMP gene, and thus exerted crucial roles in NSCLC progression and TKI targeted treatment. Conclusions Consequently, our findings highlight the function and prognostic value of Lnc-LSAMP-1 in NSCLC and provide potential novel therapeutic targets and prognostic biomarkers for patients with NSCLC.

Abstract Objectives To retrospectively analyse the potential influencing factors of CT-guided hook wire localization failure prior to thoracoscopic resection surgery of ground glass nodules (GGNs), and determine the main risk elements for localization failure. Methods In all, 372 patients were included in this study, with 21 patients showing localization failure. The related parameters of patients, GGNs, and localization were analysed through univariate and multiple logistic regression analysis to determine the risk factors of localization failure. Results Univariate logistic regression analysis indicated that trans-fissure (odds ratio [OR] 4.896, 95% confidence interval [CI] 1.489–13.939); trans-emphysema (OR 3.538, 95% CI 1.343–8.827); localization time (OR 0.956, 95% CI 0.898–1.019); multi-nodule localization (OR 2.597, 95% CI 1.050–6.361); and pneumothorax (OR 10.326, 95% CI 3.414–44.684) were risk factors for localization failure, and the p-values of these factors were

Magnesium hydride (MgH2) is a potential material for solid-state hydrogen storage. However, the thermodynamic and kinetic properties are far from practical application in the current stage. In this work, two-dimensional vanadium carbide (V2C) MXene with layer thickness of 50−100 nm was fist synthesized by selectively HF-etching the Al layers from V2AlC MAX phase and then introduced into MgH2 to improve the hydrogen sorption performances of MgH2. The onset hydrogen desorption temperature of MgH2 with V2C addition is significantly reduced from 318 °C for pure MgH2 to 190 °C, with a 128 °C reduction of the onset temperature. The MgH2 + 10 wt% V2C composite can release 6.4 wt% of H2 within 10 min at 300 °C and does not loss any capacity for up to 10 cycles. The activation energy for the hydrogen desorption reaction of MgH2 with V2C addition was calculated to be 112 kJ mol−1 H2 by Arrhenius's equation and 87.6 kJ mol−1 H2 by Kissinger's equation. The hydrogen desorption reaction enthalpy of MgH2 + 10 wt% V2C was estimated by van't Hoff equation to be 73.6 kJ mol−1 H2, which is slightly lower than that of the pure MgH2 (77.9 kJ mol−1 H2). Microstructure studies by XPS, TEM, and SEM showed that V2C acts as an efficient catalyst for the hydrogen desorption reaction of MgH2. The first-principles density functional theory (DFT) calculations demonstrated that the bond length of Mg−H can be reduced from 1.71 Å for pure MgH2 to 2.14 Å for MgH2 with V2C addition, which contributes to the destabilization of MgH2. This work provides a method to significantly and simultaneously tailor the hydrogen sorption thermodynamics and kinetics of MgH2 by two-dimensional MXene materials.

Trichoderma, a well-known and extensively studied fungal genus, has gained significant attention for its remarkable antagonistic abilities against a wide range of plant pathogens. In this study, a total of 108 Trichoderma isolates were screened through in vitro dual antagonistic assays and culture filtrate inhibition against Fusarium graminearum. Of these, the YNQJ1002 displayed noteworthy inhibitory activities along with thermal stability. To validate the metabolic differences between YNQJ1002 and GZLX3001 (with strong and weak antagonism, respectively), UPLC-TOF-MS/MS mass spectrometry was employed to analyze and compare the metabolite profiles. We identified 12 significantly up-regulated metabolites in YNQJ1002, which include compounds like Trigoneoside, Torvoside, trans,trans-hepta-2,4,6-trienoic acid, and Chamazulene. These metabolites are known for their antimicrobial properties or signaling roles as components of cell membranes. Enriched KEGG analysis revealed a significant enrichment in sphingolipid metabolism and linoleic acid metabolism, as well as autophagy. The results demonstrated that YNQJ1002’s abundance of antimicrobial substances, resulting from specific metabolic pathways, enhanced its superior antagonistic activity against F. graminearum. Finally, YNQJ1002 was identified using the ITS, tef1-1α, and rpb2 regions, with MIST system sequence matching confirming its classification within the species. Overall, we have obtained a novel strain, T. asperellum YNQJ1002, which is rich in metabolites and shows potential antagonistic activity against F. graminearum. This study has opened promising prospects for the development of innovative Trichoderma-derived antifungal compounds, featuring a unique mechanism against pathogens.

Background: The occurrence and development of chronic obstructive pulmonary disease (COPD) are regulated by environmental and genetic factors. In hypoxia, Erythropoietin (EPO) satisfies the body’s need for oxygen by promoting the production of red blood cells. Hypoxia was proven to be a common physiological condition in COPD progression and associated with many complications. Some studies have found that EPO is involved in the development of COPD. But the mechanism has not been fully proven. Methods: We conducted a case-control study enrolled 1095 COPD patients and 1144 healthy controls in Guangdong Province to evaluate the association between EPO polymorphisms (rs1617640 A>C, rs507392 A>G, rs564449 G>T) and COPD susceptibility. 872 participants from southern Gansu Province were recruited to verify the effect of EPO polymorphisms on lung function. Results: EPO rs1617640 C allele reduced COPD susceptibility in southern Chinese significantly (AC vs. AA: adjusted Odds ratio (OR) = 0.805, 95% CI = 0.669–0.969; AC+CC vs. AA: adjusted OR = 0.822, 95% CI = 0.689–0.980). However, there was no association between rs507392 A>G and rs564449 G>T polymorphisms and COPD susceptibility (p > 0.05). We further observed that the rs1617640 C allele was associated with higher FEV1 and FVC in Guangdong and Gansu populations significantly (both p < 0.05). In brief, the level of FEV1 and FVC increased with the C allele number. We modeled the relative risk for men and women, in which the population-attributable risks chances were 0.449 (0.258–0.641) and 0.262 (0.128–0.396) respectively. In this model, smoking status, coal as fuels, education level, and rs1617640 A>C were finally retained for males, while smoking status, biomass as fuels, and1617640 A>C were retained for females. In the end, using the method developed by Gail and Bruzzi, we fitted a 10-year absolute risk model for southern Chinese with different individual relative risks, which was presented as a table. Conclusions: In conclusion, this study found that EPO rs1617640 A>C polymorphism is associated with COPD susceptibility in southern Chinese, and the C allele was associated with better lung function. In addition, it could also be considered a genetic marker associated with environmental factors to predict the absolute 10-year risk of COPD in southern Chinese.

Abstract Objective The purpose of this study was to compare imaging features between COVID-19 and mycoplasma pneumonia (MP). Materials and methods The data of patients with mild COVID-19 and MP who underwent chest computed tomography (CT) examination from February 1, 2020 to April 17, 2020 were retrospectively analyzed. The Pneumonia-CT-LKM-PP model based on a deep learning algorithm was used to automatically quantify the number, volume, and involved lobes of pulmonary lesions, and longitudinal changes in quantitative parameters were assessed in three CT follow-ups. Results A total of 10 patients with mild COVID-19 and 13 patients with MP were included in this study. Abstract Chlorogenic acids (CGAs) are important secondary metabolites produced in sweet potato. However, the mechanisms of their biosynthesis and regulation remain unclear. To identify potential genes involved in CGA biosynthesis, analysis of the dynamic changes in CGA components and RNA sequencing were performed on young leaves (YL), mature leaves (ML), young stems (YS), mature stems (MS) and storage roots (SR). Accordingly, we found that the accumulation of six CGA components varied among the different tissues and developmental stages, with YS and YL recording the highest levels, while SR exhibited low levels. Moreover, the transcriptome analysis yielded 59,287 unigenes, 3,767 of which were related to secondary-metabolite pathways. The differentially expressed genes (DEGs) were identified based on CGA content levels by comparing the different samples, including ML vs. YL, MS vs. YS, SR vs. YL and SR vs. YS. A total of 501 common DEGs were identified, and these were mainly implicated in the secondary metabolites biosynthesis. Additionally, eight co-expressed gene modules were identified following weighted gene co-expression network analysis, while genes in darkgrey module were highly associated with CGA accumulation. Darkgrey module analysis revealed that 12 unigenes encoding crucial enzymes (PAL, 4CL, C4H, C3H and HCT/HQT) and 42 unigenes encoding transcription factors (MYB, bHLH, WD40, WRKY, ERF, MADS, GARS, bZIP and zinc finger protein) had similar expression patterns with change trends of CGAs, suggesting their potential roles in CGA metabolism. Our findings provide new insights into the biosynthesis and regulatory mechanisms of CGA pathway, and will inform future efforts to build a genetically improve sweet potato through the breeding of high CGA content varieties.
