Data-driven approaches such as neural networks are increasingly used for deep excavations due to the growing amount of available monitoring data in practical projects. However, most neural network models only use the data from a single monitoring point and neglect the spatial relationships between multiple monitoring points. Besides, most models lack flexibility in providing predictions for multiple days after monitoring activity. This study proposes a sequence-to-sequence (seq2seq) two-dimensional (2D) convolutional long short-term memory neural network (S2SCL2D) for predicting the spatiotemporal wall deflections induced by deep excavations. The model utilizes the data from all monitoring points on the entire wall and extracts spatiotemporal features from data by combining the 2D convolutional layers and long short-term memory (LSTM) layers. The S2SCL2D model achieves a long-term prediction of wall deflections through a recursive seq2seq structure. The excavation depth, which has a significant impact on wall deflections, is also considered using a feature fusion method. An excavation project in Hangzhou, China, is used to illustrate the proposed model. The results demonstrate that the S2SCL2D model has superior prediction accuracy and robustness than that of the LSTM and S2SCL1D (one-dimensional) models. The prediction model demonstrates a strong generalizability when applied to an adjacent excavation. Based on the long-term prediction results, practitioners can plan and allocate resources in advance to address the potential engineering issues.

A magnitude 7.3 earthquake occurred at 7:58 a.m. (Beijing time) on April 3, 2024, in the sea area of Hualien County, Taiwan Province, China (23.81°N，121.74°E), which had a large seismic hazard impact and attracted widespread attention at home and abroad. This paper introduces the geological and tectonic background of the earthquake and its disaster influence, as well as the volcanic activity on Kueishantao volcano that partially collapsed as a result of the seismic effect, and focuses on a comprehensive analysis of the possibility of whether the earthquake will trigger the volcanic activity. Through the analysis of seismic and hyperspectral gas data, it can be seen that the Kueishantao volcano has not yet shown obvious anomalous responses. Then the probability of triggering is quantitatively judged by introducing the non-dimensional index of earthquake triggering potential volcanoes，and the TRIGI value is 19.8 (> 10), which indicates that the probability of earthquake-triggered eruption of Kueishantao volcano is relatively low in the short term. At present, the volcano is still in the state of dormancy from the analysis of qualitative and quantitative aspects. In the future, we will continue to collect and improve macro and micro precursor anomaly data before volcanic eruption, track and judge the state of volcanic activity, and draw more accurate and reliable conclusions about the triggering possibility.

IntroductionRoad cracks significantly shorten the service life of roads. Manual detection methods are inefficient and costly. The YOLOv5 model has made some progress in road crack detection. However, issues arise when deployed on edge computing devices. The main problem is that edge computing devices are directly connected to sensors. This results in the collection of noisy, poor-quality data. This problem adds computational burden to the model, potentially impacting its accuracy. To address these issues, this paper proposes a novel road crack detection algorithm named EMG-YOLO.MethodsFirst, an Efficient Decoupled Header is introduced in YOLOv5 to optimize the head structure. This approach separates the classification task from the localization task. Each task can then focus on learning its most relevant features. This significantly reduces the model’s computational resources and time. It also achieves faster convergence rates. Second, the IOU loss function in the model is upgraded to the MPDIOU loss function. This function works by minimizing the top-left and bottom-right point distances between the predicted bounding box and the actual labeled bounding box. The MPDIOU loss function addresses the complex computation and high computational burden of the current YOLOv5 model. Finally, the GCC3 module replaces the traditional convolution. It performs global context modeling with the input feature map to obtain global context information. This enhances the model’s detection capabilities on edge computing devices.ResultsExperimental results show that the improved model has better performance in all parameter indicators compared to current mainstream algorithms. The EMG-YOLO model improves the accuracy of the YOLOv5 model by 2.7%. The mAP (0.5) and mAP (0.9) are improved by 2.9% and 0.9%, respectively. The new algorithm also outperforms the YOLOv5 model in complex environments on edge computing devices.DiscussionThe EMG-YOLO algorithm proposed in this paper effectively addresses the issues of poor data quality and high computational burden on edge computing devices. This is achieved through optimizing the model head structure, upgrading the loss function, and introducing global context modeling. Experimental results demonstrate significant improvements in both accuracy and efficiency, especially in complex environments. Future research can further optimize this algorithm and explore more lightweight and efficient object detection models for edge computing devices.

Abstract Field observations in sedimentation and erosion-prone areas indicate that most natural sand deposits may contain a certain amount of non-plastic fines and are often under anisotropic stress conditions. A series of triaxial compression tests were performed on clean and silty sand with fines content f c ranging from 0 to 20% at an initial mean effective stress of p 0′ = 100 kPa and varying consolidation conditions to understand the impact of initial stress anisotropy on undrained shear behavior. The results indicate that the state parameter ψ is a superior predictor for characterizing the responses of sand-fines mixtures compared to the global void ratio and relative density. A comparison of the behavior of clean and silty sand with a constant ψ (= − 0.03) confirms that the sample with 10% f c exhibits the strongest dilation and greatest shear resistance, irrespective of the consolidation conditions. It is also demonstrated that the initial stress anisotropy with a comparably higher static stress ratio η s typically diminishes the shear strength of mixtures. However, the influence of initial stress anisotropy on soil stiffness is not unilateral. The sample consolidated to a negative η s is stiffer than that under isotropic consolidation, while the presence of a positive η s leads to a decrease in the secant Young's modulus.

Abstract With the booming of cloud-based digital twin systems, monitoring key performance indicators has become crucial for ensuring system security and reliability. Due to the massive amount of monitoring data generated, data compression is necessary to save data transmission bandwidth and storage space. Although the existing research has proposed compression methods for multivariate time series (MTS), it is still a challenge to guarantee the correlation between data when compressing the MTS. This paper proposes an MTS Collaborative Compression (MTSCC) method based on the two-step compression scheme. First, shape-based clustering is implemented to group the MTS. Afterward, the compressed sensing is optimized to achieve collaborative compression of grouped data. Based on a real-world MTS dataset, the experimental results show that the proposed MTSCC can effectively preserve the complex temporal correlation between indicators while achieving efficient data compression, and the root mean squared error of correlation between the reconstructed and original data is only 0.0489 in the case of 30% compression ratio. Besides, it is verified that using the reconstructed data in the production environment has almost the same performance as using the original data.

Background: This pragmatic clinical trial aims to determine the efficacy and safety of add-on Astragalus membranaceus (AM) for cognition and non-cognition in patients with of mild to moderate Alzheimer's disease complicated with orthostatic hypotension in orthostatic hypotension, elucidate the underlying mechanisms, identify related response predictors, and explore effective drug components. Methods: This is an add-on, assessor-blinded, parallel, pragmatic, randomized controlled trial. At least 66 adults with mild to moderate Alzheimer's disease (AD) and OH aged 50–85 years will be recruited. Participants will be randomized in a 1:1:1 ratio to receive 24 weeks of routine care or add-on low dose AM or add-on high dose AM group. The primary efficacy outcome will be measured by the Alzheimer's Disease Assessment Scale-Cognitive Subscale, Chinese version. Secondary efficacy outcome assessment will include neuropsychological tests, blood pressure, plasma biomarkers, multimodal electroencephalograms, and neuroimaging. Safety outcome measures will include physical examinations, vital signs, electrocardiography, laboratory tests (such as hematologic and blood chemical tests), and adverse event records. Ethics and dissemination: This trial was approved and supervised by Fujian Medical University Union Hospital (2021KJCX040). Independent results, findings will be published in peer-reviewed journals and presented at national and international conferences. Backgrounds TBC1D family members (TBC1Ds) are a group of proteins that contain the Tre2-Bub2-Cdc16 (TBC) domain. Recent studies have shown that TBC1Ds are involved in tumor growth, but no analysis has been done of expression patterns and prognostic values of TBC1Ds in hepatocellular carcinoma (HCC). Methods The expression levels of TBC1Ds were evaluated in HCC using the TIMER, UALCN and Protein Atlas databases. The correlation between the mRNA levels of TBC1Ds and the prognosis of patients with HCC in the GEPIA database was then analyzed. An enrichment analysis then revealed genes that potentially interact with TBC1Ds. The correlation between levels of TBC1Ds and tumor-infiltrating immune cells (TIICs) in HCC were studied using the TIMER 2.0 database. Finally, a series of in vitro assays verified the role of TBC1Ds in HCC progression. Results This study revealed the upregulated expression of TBC1Ds in HCC and the strong positive correlation between the mRNA levels of TBC1Ds and poor prognosis of patients with HCC. The functions of TBC1Ds were mainly related to autophagy and the AMPK pathway. There was also a significant correlation between level of TBC1Ds and tumor-infiltrating immune cells (TIICs) in HCC. The promoting role of TBC1Ds in HCC progression was verified in vitro assays. Conclusion The results of this analysis indicate that TBC1Ds may serve as new biomarkers for early diagnosis and treatment of HCC.

Background: Inflammation is critical in the etiology and progression of acute respiratory distress syndrome (ARDS). This study aims to rigorously assess the predictive capacity of systemic immune-inflammation index (SII) in determining the outcomes of patients with ARDS. Methods: Patient data were extracted from version 2.2 of the Medical Information Mart for Intensive Care IV (MIMIC-IV). The Receiver Operating Characteristic (ROC) curve was deployed to determine the optimal cutoff value for the SII, facilitating the stratification of participants into distinct cohorts based on SII levels. The relationship between SII and survival outcomes was rigorously evaluated using Cox proportional hazards models. The association between SII and patient survival was rigorously examined using Cox proportional-hazard models. The impact of varying SII levels on mortality was quantitatively assessed through these models, with the results articulated as hazard ratios (HRs) and 95% confidence intervals (CIs). Three distinct models were formulated for this analysis: Model 1 employed univariate Cox regression to relate SII with mortality; Model 2 introduced adjustments for age and sex; and Model 3 extended these adjustments to include age, sex, race, SAPS II, APSIII, Hemoglobin, Albumin, Pneumonia, SpO2, and SBP. Results: Post-application of the inclusion criteria, a cohort of 976 eligible patients was delineated for detailed examination. Univariate analysis focusing on 30-day mortality within the SII ≥1694, the hazard ratio (HR) was 1.42 (95% confidence interval (CI): 1.11, 1.81). However, after adjusting for confounding factors such as age, sex, race, Simplified Acute Physiology Score II (SAPS II), Acute Physiology Score (APS) III, Hemoglobin, Albumin, Pneumonia, SpO2, and Systolic Blood Pressure (SBP), an SII value of ≥1694 was identified as an independent and significant risk factor for mortality in patients with ARDS, with an HR of 1.38 (95% CI: 1.08–1.77, P = 0.0016). This trend was consistent for 90-day and one-year mortality rates. Conclusions: SII surfaced as an autonomous determinant of mortality in ARDS patients, affirming its status as an accessible and dependable prognostic indicator for individuals newly diagnosed with this critical condition. Additional research is imperative to further elucidate the prognostic implications of SII in the therapeutic management of patients with ARDS.

Although lipophilic shellfish toxins (LSTs) pose a significant threat to the health of seafood consumers, their systematic investigation and risk assessment remain scarce. The goals of this study were as follows: (1) analyze LST levels in commercially available shellfish in Zhejiang province, China, and determine factors influencing LST distribution; (2) assess the acute dietary risk of exposure to LSTs for local consumers during the red tide period; (3) explore potential health risks of LSTs in humans; and (4) study the acute risks of simultaneous dietary exposure to LSTs and paralytic shellfish toxins (PSTs). A total of 546 shellfish samples were collected. LSTs were detected in 89 samples (16.3%) at concentrations below the regulatory limits. Mussels were the main shellfish species contaminated with LSTs. Spatial variations were observed in the yessotoxin group. Acute exposure to LSTs based on multiple scenarios was low. The minimum tolerable exposure durations for LSTs calculated using the mean and the 95th percentile of consumption data were 19.7 and 4.9 years, respectively. Our findings showed that Zhejiang province residents are at a low risk of combined exposure to LSTs and PSTs; however, the risk may be higher for children under 6 years of age in the extreme scenario.

As one of China's nine border provinces, Jilin Province is an important window for China's “Belt and Road” to open up to the north, and an important channel for China's foreign trade and foreign exchanges. Therefore, understanding hydrogeochemical characteristics is very important for effective water resources management in Jilin Province. A total of 64 groundwater samples were collected from 32 monitoring Wells in Jilin Province. Use of hydrogeochemical analysis techniques, WQI index and human health risk index methods to assess hydrogeochemical processes and groundwater quality, including the determination of hydrogeochemical parameters, conventional ions, trace elements and stable isotopes (δD and δ18O). The results show that the groundwater is slightly alkaline, the cations are mainly Na+, Ca2+, Mg2+, and K+, and the anions are mainly HCO3-, Cl−, and SO42-. Most of the groundwater samples belong to Ca-HCO3 and Na-HCO3 types, which are formed by the dissolution of calcite, dolomite, gypsum, silicates and carbonates. Stable isotope analysis shows that groundwater recharge in Jilin mainly comes from atmospheric precipitation at an altitude of 1,430~2,348 m. The strong correlation between trace elements indicates a common source. Most of the groundwater quality index (WQI) in the study area is Class I to Class III, and hazard quotient (HQ) and hazard index (HI) are both below 1. This indicates excellent water quality and minimal health risks associated with human consumption. It is worth noting that trace elements are enriched in the Tanlu fault zone. It provides scientific guidance for the utilization and allocation of groundwater resources in Jilin Province.

Nicarbazin (NICA) and triazine anticoccidial drugs (diclazuril (DIZ) and toltrazuril (TOZ)) are the primary strategy for preventing and treating coccidiosis. To prevent the development of drug resistance and mitigate the potential chronic toxicity to humans resulting from prolonged exposure, a liquid chromatography–tandem mass spectrometry method with high reliability and sensitivity was developed to determine NICA, DIZ, TOZ, and its two metabolites in chicken muscle and eggs. Upon establishing the extraction conditions involving 10 mL of acetonitrile and 10 min of sonication, in-syringe dispersive solid-phase extraction with silica was performed in combination with n-hexane clean-up. The selection of isotope peaks of precursor ions and low-mass range scanning allowed the two transitions for the quantification of all compounds. The limits of detection for DIZ and NICA were both 0.1 μg/kg, and for TOZ and metabolites, they were 0.3 μg/kg; the limits of quantitation were 0.3 and 1 μg/kg, respectively. The linear range was 0.25–50 ng/mL with a correlation coefficient r > 0.999. The average recoveries at three spiking levels in muscle and eggs were 90.1–105.2% and 94.0–103.7% with the relative standard deviations of 3.0–8.1% and 3.1–14.4%, respectively. The precision, accuracy, and stability were evaluated by three quality control samples.

Abstract Recently, two‐dimensional transition metal dichalcogenides (TMDs) demonstrated their great potential as cost‐effective catalysts in hydrogen evolution reaction. Herein, we systematically summarize the existing defect engineering strategies, including intrinsic defects (atomic vacancy and active edges) and extrinsic defects (metal doping, nonmetal doping, and hybrid doping), which have been utilized to obtain advanced TMD‐based electrocatalysts. Based on theoretical simulations and experimental results, the electronic structure, intermediate adsorption/desorption energies and possible catalytic mechanisms are thoroughly discussed. Particular emphasis is given to the intrinsic relationship between various types of defects and electrocatalytic properties. Furthermore, current opportunities and challenges for mechanical investigations and applications of defective TMD‐based catalysts are presented. The aim herein is to reveal the respective properties of various defective TMD catalysts and provide valuable insights for fabricating high‐efficiency TMD‐based electrocatalysts.

The latest research results show that there is a unified magma system and heating channel beneath the Arxan volcanic field, indicating a potential risk of eruption. The Arxan volcanic field features multiple gas emission sites (e.g., Jinjianggou hot springs and Yinjianggou hot springs) and exhibits strong hydrothermal activity. In this study, measurements of the hot spring gas composition and soil CO2 flux in the Arxan Holocene volcanic field were conducted, and the results were combined with previous research results to analyze the degassing characteristics of this region. The results show that the volcanic gases in the Arxan volcanic field are composed of 0.07%–1.09% CO2, 0.33–12 ppm CH4, 1.57–53 ppm H2, 800–30,241 ppm He, and 1.14%–1.86% Ar. The He content in this area is notably higher than that in other dormant volcanoes in China. This difference is possibly caused by U–Th decay in the Mesozoic granodiorite and acidic volcanic rocks in the study area, which can produce substantial radiogenic He. The soil gas concentrations near the Jinjianggou and Yinjianggou hot springs are higher than those of two Holocene volcanoes. The peak CO2 concentration in the soil near the Jinjianggou hot spring can reach 35,161 ppm. The single-site soil microseepage CO2 flux in the Arxan volcanic field is 4.66–107.18 g m−2 d−1, and the estimated annual CO2 emission flux from the volcanic field to the atmosphere is 0.63 × 105 t, which also demonstrates that soil CO2 flux of Arxan volcano is comparable to the soil CO2 emission level of the Iwojima volcano.

Karst groundwater resources are rich, and they have important water supply significance. A karst reservoir is a means of exploiting and utilizing groundwater resources, but because of the complex hydrogeological structure and underground river pipeline in a karst area, the seepage problem of the reservoir is extremely serious. Therefore, the Yibasan Karst Reservoir (YKR) was selected as the focus of this study. The hydrogeological structure of the subterranean river system of Yutang (SRSY) was identified and the hydraulic connections between the subterranean river conduits were determined using tracer experiment and groundwater dynamic monitoring. Furthermore, the development location and depth of the karst seepage zone of the YKR were determined using geophysical exploration. The results showed that there were three subterranean river conduits in the SRSY, and there was no hydraulic connection. The northern and southern pressure and torsion faults on the sides constitute the impervious boundary of the SRSY, which provided good catchment conditions for the formation of the YKR. Additionally, the northern and main conduits of the SRSY provide a sufficient groundwater source for the YKR. Moreover, the development width of the karst seepage zone of the YKR ranges between 40 and 60 m, and the elevation ranges between 1275 and 1355 m. The research results not only provide an effective basis for the treatment of the karst seepage problem of the YKR but also provide an important reference for the development and utilization of groundwater resources in similar karst areas.

The intake of paralytic shellfish toxins (PSTs) may adversely affect human health. Therefore, this study aimed to show the prevalence of PSTs from commercially available shellfish in Zhejiang Province, China, during the period of frequent red tides, investigate the factors affecting the distribution of PSTs, and assess the risk of PST intake following the consumption of bivalve shellfish among the Zhejiang population. A total of 546 shellfish samples were collected, 7.0% of which had detectable PSTs at concentrations below the regulatory limit. Temporal, spatial, and interspecific variations in the occurrence of PSTs were observed in some cases. The dietary exposure to PSTs among the general population of consumers only was low. However, young children in the extreme scenario (the 95th percentile of daily shellfish consumption combined with the maximum PST concentration), defined as 89–194% of the recommended acute reference doses, were possibly at risk of exposure. Notably, Arcidae and mussels were the major sources of exposure to toxins. From the public health perspective, PSTs from commercially available shellfish do not pose a serious health risk; however, more attention should be paid to acute health risks, especially for young children, during periods of frequent red tides.

BackgroundWe aimed to compare the detection rates of prostate cancer (PCa) and clinically significant prostate cancer(csPCa) by biparametric (bp-) and multiparameter magnetic resonance imaging (mpMRI).Materials and MethodsA total of 699 patients who underwent transperineal prostate biopsy in the Department of Urology, the Second Affiliated Hospital of Nantong University from January 2018 to December 2021 were retrospectively reviewed. Multivariate analysis was used to explore the influencing factors associated with the detection rates of PCa and csPCa. According to MRI examination before biopsy, the patients were divided into bpMRI group and mpMRI group. The detection rates of PCa and csPCa by bpMRI and mpMRI were compared. Furthermore, stratified analysis was performed for patients in these two groups to compare the detection rates of PCa and csPCa at different tPSA intervals, different prostate volume (PV) intervals and different PI-RADS V2 scores.ResultsA total of 571 patients were finally analyzed in this study after exclusion, and the overall detection rate of PCa was 54.5%. Multivariate analysis showed that patient age, tPSA level, prostate volume and PI-RADS V2 score were independent risk factors affecting the detection rates of PCa and csPCa. The detection rates of PCa and csPCa by bpMRI and mpMRI were comparable (51.3% vs. 57.9%, 44.0% vs. 48.0%, both P > 0.05), with no statistical significance. In the tPSA 10–20 ng/ml interval, the detection rates of PCa (59.72% vs. 40.35%, P = 0.011) and csPCa (51.39% vs. 28.82%, P = 0.005) by mpMRI were significantly higher than those by bpMRI, while in other tPSA interval (tPSA 60 ml) and different PI-RADS V2 scores (3, 4, and 5), the detection rates of PCa and csPCa were comparable between the two groups.ConclusionFor detecting PCa and csPCa, bpMRI and mpMRI had similar diagnostic efficacies, whereas mpMRI detected more PCa and csPCa in the tPSA interval of 10–20 ng/ml.

On January 15th, 2022, a submarine volcano in Tonga erupted violently and explosively, ejecting a large amount of ash, gas and water vapor which entered the stratosphere to form a huge ash cloud. The volcanic eruption caused widespread concern at home and abroad. This paper presents a comprehensive analysis of the geological background, eruption history, eruption process, disaster impact, and eruption mechanism of the Tonga Volcano. This draws attention to the current activity of active volcanoes in China, especially to Changbaishan Tianchi volcano, which is the largest active volcano with most potentially catastrophic eruption risk. It warns us that we should take precautionary measures to further strengthen the monitoring and early warning of domestic volcanic hazards. Drawing on the experience of this volcanic disaster in Tonga, it is particularly important to scientifically study and judge the dynamics of volcanoes and improve the domestic volcanic disaster risk monitoring and early warning system.

Since 2002, the China Geological Survey has successively organized and implemented hydrogeological survey and groundwater exploration in water deficient areas, such as severe water shortage areas in west China, Wumeng Mountain area, Yimeng mountain area and red bed area in southern Jiangxi, obtained a large number of exploration data conducive to promoting discipline development, and summarized the law of water abundance. In recent decades, water prospecting and well drilling in bedrock mountainous areas serving poverty alleviation have not been systematically summarized based on water storage structures and their attributes. This paper takes the water storage structure as the theoretical guidance, considers geomorphic catchment, lithological water accumulation, structural water control and other control factors, and takes structure as the main control factor. The factors such as topography, formation lithology, vegetation development and actual value of development and utilization are systematically summarized and water storage structures are divided into 8 types, further divided into 20 types, and more into 35 types, such as the horizontal rock stratum, monocline formation, fold, fault, contact zone, weathered crust, composite and cave reservoir. The common characteristics of different scales and types of water storage structures are analyzed and summarized with reference to and in combination with the neotectonic water control theory and groundwater system theory. From the aspects of spatial combination characteristics of water storage structure, boundary hydrogeological properties, hydrodynamic conditions, scale effect, symbiotic characteristics of water control and practical value of development and utilization, this paper puts forward the correlation between the lithology and geological structure, the relativity of permeability and water separation, the balance between catchment and water storage, the unity of scale and system, the inheritance of formation and transformation and the matching of scale and purpose. The research results refine and improve the classification of groundwater storage structure types in bedrock mountain areas with water shortage, summarize the attribute characteristics of water storage structure, deepen the understanding of the concept and connotation of water storage structure, not only further promote the research of hydrogeology in bedrock mountain areas, but also provide references for water prospecting and well locating in water shortage areas, and guide the sustainable utilization of water resources in water shortage areas.

Abstract. Background:. Microglia plays an indispensable role in the pathological process of sleep deprivation (SD). Here, the potential role of microglial CX3C-chemokine receptor 1 (CX3CR1) in modulating the cognition decline during SD was evaluated in terms of microglial neuroinflammation and synaptic pruning. In this study, we aimed to investigat whether the interference in the microglial function by the CX3CR1 knockout affects the CNS's response to SD. Methods:. Middle-aged wild-type (WT) C57BL/6 and CX3CR1−/− mice were either subjected to SD or allowed normal sleep (S) for 8 h to mimic the pathophysiological changes of middle-aged people after staying up all night. After which, behavioral and histological tests were used to explore their different changes. Results:. CX3CR1 deficiency prevented SD-induced cognitive impairments, unlike WT groups. Compared with the CX3CR1−/− S group, the CX3CR1−/− SD mice reported a markedly decreased microglia and cellular oncogene fos density in the dentate gyrus (DG), decreased expression of pro-inflammatory cytokines, and decreased microglial phagocytosis-related factors, whereas increased levels of anti-inflammatory cytokines in the hippocampus and a significant increase in the density of spines of the DG were also noted. Conclusions:. These findings suggest that CX3CR1 deficiency leads to different cerebral behaviors and responses to SD. The inflammation-attenuating activity and the related modification of synaptic pruning are possible mechanism candidates, which indicate CX3CR1 as a candidate therapeutic target for the prevention of the sleep loss-induced cognitive impairments.

Cadmium (Cd) pollution of food safety is a prominent food safety concern worldwide. The concentration of Cd in six aquatic food categories collected from 2018 to 2022 was analyzed using inductively coupled plasma mass spectrometry, and the Cd exposure levels were calculated by combining the Cd concentration and food consumption data of 18913 urban and rural residents in Zhejiang Province in 2015–2016. The mean Cd concentration was 0.699 mg/kg and the mean Cd exposure of aquatic foods was 0.00951 mg/kg BW/month for the general population. Marine crustaceans were the largest Cd contributor, corresponding to 82.7%. The regional distribution results showed that the average Cd exposure levels of 11 cities did not exceed the provisional tolerable monthly intake (PTMI). According to the subgroups, the Cd mean exposure level of 2–3-year-old children was significantly higher than that of the other age groups but did not exceed the PTMI. Health risk classification assessment demonstrated that the final risk score was six, and the health risk level of Cd exposure in aquatic products in the Zhejiang population was medium. These results demonstrated that the risk of Cd exposure in certain food types or age groups should be given more concern.

Karst wetlands are important in the global carbon and nitrogen cycles as well as in security of water resources. Huixian wetland (Guilin) is the largest natural karst wetland in China. In recent years, groundwater nitrogen pollution has increasingly affected the wetland ecosystem integrity due to anthropogenic activities. In this study, it was hypothesized that autotrophic microbial diversity is impacted with the advent of pollution, adversely affecting autotrophs in the carbon and nitrogen cycles. Autotrophic microbes have important roles in abating groundwater nitrogen pollution. Thus, it is of great significance to study the characteristics of autotrophic bacterial communities and their responses to environmental parameters in nitrogen-polluted karst groundwaters. The abundances of the Calvin–Benson cycle functional genes cbbL and cbbM as well as the autotrophic CO2-fixing bacterial communities were characterized in the karst groundwater samples with different levels of nitrogen pollution. The cbbM gene was generally more abundant than the cbbL gene in the groundwater samples. The cbbL gene abundance was significantly positively correlated with dissolved inorganic nitrogen (DIN) concentration (P < 0.01). In the autotrophic CO2-fixing bacterial communities, Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria of the phylum Proteobacteria were predominant. At the genus level, Rubrivivax and Methylibium were the dominant cbbL gene containing genera, while Halothiobacillus and Endothiovibrio were the dominant genera for the cbbM gene. The abundance of autotrophic CO2-fixing bacterial communities increased but their diversity decreased with the inflow of nitrogen into the karst groundwater system. The community structure of autotrophic CO2-fixing bacteria in the groundwaters was also significantly affected by environmental factors such as the carbonic anhydrase (CA) activity, dissolved inorganic carbon (DIC) concentration, temperature, and oxidation-reduction potential (ORP). Nitrogen inflow significantly changed the characteristics of autotrophic CO2-fixing bacterial communities in the karst groundwaters. Some key genera such as Nitrosospira and Thiobacillus were clearly abundant in the karst groundwaters with high nitrogen levels. Their respective roles in nitrification and denitrification impact nitrogen removal in this ecosystem. The findings in this study provide an important reference for biological abatement of nitrogen pollution in the karst groundwater system.

A model of consolidation for a single-drain well under vacuum preloading, based on Biot’s axisymmetric theory and considering the “real strain” hypothesis, was established using the finite element method. Degenerating the consolidation equation of real strain yielded the Barron’s classical free strain and equal strain equations. The free strain and equal strain finite element models were derived by imposing boundary and constraint conditions on the real strain FEM model. The validation of the simulation process confirmed that the numerical model achieved consistent outcomes, with the theoretical values postulating its effectiveness. The real strain model revealed the Mandel–Cryer effect of soil near the vertical drain during the initial vacuum preloading consolidation process, causing the effective stress to increase more than the effective stress generated by the vacuum pressure and leading to a greater reduction in the void ratio. This phenomenon is one of the reasons for clogging during the vacuum preloading process. However, the free strain and equal strain models cannot produce this effect because they cannot describe the coupling between soil deformation and fluid flow. The parameter analysis of the real strain model showed that as Poisson’s ratio for soil decreased, the consolidation rate of soil also decreased, while the Mandel–Cryer effect of soil increased.

In this paper, we built a method of verifying the 4 retinols of vitamin A and 8 tocopherol isomers of vitamin E in the food industry based on RP-HPLC-PDA-FLR. The effect of laboratory light conditions on the target components was considered for the first time, and it was found that the loss rate of the target components was the smallest in the case of a white laboratory bench with red or LED light in the dark room. There was no finding of extraction recoveries of the target components with a significant difference under different saponification conditions. Meanwhile, it was found that using ethyl acetate/n-hexane mixed solvent as the extraction solvent could ensure the effective extraction of the target components. Finally, baseline separation of 12 components was achieved within 45 min using the C30 column. With the help of methodological verification, we found that the recovery rate ranged from 76.45% to 93.52%, and RSD was between 0.19% and 12.99%; the Limit of Detection minimum value was 0.01 mg/100 g and the Limit of Quantitation minimum value was 0.03 mg/100 g. The detection method was successfully applied to the distribution detection of 4 kinds of retinols and 8 kinds of vitamin E in egg and dairy products and provided technical support for the accurate nutritional evaluation of vitamin A and vitamin E.

Large-diameter monopiles have been widely used for constructing offshore wind turbines. The bearing capacity of a monopile foundation is a significant research problem. In this study, a new type of foundation, known as the pile–bucket foundation, was investigated to improve the bearing capacity of monopiles. A finite element software was used for establishing several numerical models of monopile and pile–bucket foundations to analyze the reinforcement afforded by the bucket attached to the monopile foundation. Furthermore, considering that offshore wind turbines are prone to resonance under the excitation of wind and wave loads, the natural frequencies of the monopile and pile–bucket foundations were determined and compared using both analytical and numerical methods. The results show that compared to the monopile foundation, the pile–bucket foundation has a significantly higher bearing capacity, mainly for large bucket diameters. The natural frequencies of the pile–bucket foundations are slightly higher than those of the monopile foundations. The addition of the bucket can effectively improve the natural frequency without changing the diameter of the monopile and thus saving the foundation cost.

Alzheimer’s disease (AD) is a central nervous system degenerative disease, with no effective treatment to date. Administration of immune checkpoint inhibitors significantly reduces neuronal damage and tau hyperphosphorylation in AD, but the specific mechanism is unclear. Here, we found that programmed cell death-receptor 1 (PD1) and its ligand PDL1 were induced by an intracerebroventricular injection of amyloid-β; they were significantly upregulated in the brains of APP/PS1, 5×FAD mice and in SH-SY5Y-APP cell line compared with control. The PD1 and PDL1 levels positively correlated with the glycogen synthase kinase 3 beta (GSK3β) activity in various AD mouse models, and the PDL1-GSK3β immune complex was found in the brain. The application of PD1-blocking antibody reduced tau hyperphosphorylation and GSK3β activity and prevented memory impairments. Mechanistically, we identified PD1 as a critical regulator of GSK3β activity. These results suggest that the immune regulation of the PD1/PDL1 axis is closely involved in AD.

Mutations of the leucine-rich repeat kinase 2 (LRRK2) gene are associated with pronounced sleep disorders or cognitive dysfunction in neurodegenerative diseases. However, the effects of LRRK2 deficiency on sleep rhythms and sleep deprivation-related cognitive changes, and the relevant underlying mechanism, remain unrevealed. In this study, Lrrk2-/- and Lrrk2+/+ mice were subjected to normal sleep (S) or sleep deprivation (SD). Sleep recording, behavioral testing, Golgi-cox staining, immunofluorescence, and real-time PCR were employed to evaluate the impacts of LRRK2 deficiency on sleep behaviors and to investigate the underlying mechanisms. The results showed that after SD, LRRK2-deficient mice displayed lengthened NREM and shortened REM, and reported decreased dendritic spines, increased microglial activation, and synaptic endocytosis in the prefrontal cortex. Meanwhile, after SD, LRRK2 deficiency aggravated cognitive impairments, especially in the recall memory cued by fear conditioning test. Our findings evidence that LRRK2 modulates REM/NREM sleep and its deficiency may exacerbate sleep deprivation-related cognitive disorders by perturbing synaptic plasticity and microglial synaptic pruning in mice.

Deoxynivalenol (DON) together with its acetylated derivatives cause detrimental effects on human health, and the purpose of this study was to assess the prevalence of DON and its acetylated derivatives from grains and grain products in Zhejiang province, China, and to assess the risk of DON and its acetylated derivatives due to multiple consumptions of grains and grain products among the Zhejiang population. Food samples numbering 713 were collected, and the LC-MS/MS method was used to determine the toxins. The levels of toxins from grains and grain products were relatively low: DON was the toxin at the highest levels. The food frequency questionnaire was used to collect food consumption data. The result of exposure assessments showed that the population was overall at low levels of toxin exposure. The probable mean group daily intake of toxins was 0.21 μg/kg bw/day, which was far from the group provisional maximum tolerable daily intake of 1 μg/kg bw/day, but 0.71% of participants were at high exposure levels. Rice and dried noodles (wheat-based food) were the main sources of toxin exposure, and reducing the consumption of rice and dried noodles while consuming more of other foods with lower levels of toxins is recommended.

Pathogenic germline mutations occurring in the BRCA1 (MIM:113705 ) and BRCA2 (MIM: 600185) , which always result in truncated protein or nonsense-mediated mRNA decay, have been identified to increase the risk of hereditary breast, ovarian, pancreatic, prostate, and melanoma cancers. Recent studies show that BRCA1/2 germline mutations also contribute to half of all hereditary breast and ovarian cancer (HBOC). In this case series, we reported a novel frameshift mutation of the BRCA1 gene. This novel frameshift mutation occurs in exon10 of BRCA1 and may result in a lack of the serine cluster domain and BRCA1 C-terminus domain, which mediates the function of BRCA1 in DNA repair and are responsible for activation function of BRCA1 . The mutation was present in a Chinese hereditary male/female breast and ovarian cancer family characterized by a high incidence of breast cancer and/or ovarian cancer among the relatives and by a high incidence of triple negative breast cancer (TNBC). Our findings speculate that BRCA1 E1148Rfs*7 mutation may be related to the occurrence of HBOC and even TNBC. Interestingly, three cases of TNBC with this novel BRCA1 mutation in this case series showed a good disease-free survival, one of them has a disease-free survival up to 7 years. Therefore, further study is required to confirm that whether this mutation is associated with good prognosis of HBOC.

Objective: Astrocytes actively participate in energy metabolism in the brain, and astrocytic aerobic glycolysis disorder is associated with the pathology of Alzheimer's disease (AD). GLP-1 has been shown to improve cognition in AD; however, the mechanism remains unclear. The objectives of this study were to assess GLP-1's glycolytic regulation effects in AD and reveal its neuroprotective mechanisms. Methods: The Morris water maze test was used to evaluate the effects of liraglutide (an analog of GLP-1) on the cognition of 4-month-old 5×FAD mice, and a proteomic analysis and Western blotting were used to assess the proteomic profile changes. We constructed an astrocytic model of AD by treating primary astrocytes with Aβ1-42. The levels of NAD+ and lactate were examined, and the oxidative levels were assessed by a Seahorse examination. Astrocyte-neuron co-culture was performed to evaluate the effects of GLP-1 on astrocytes’ neuronal support. Results: GLP-1 improved cognition in 4-month-old 5×FAD mice by enhancing aerobic glycolysis and reducing oxidative phosphorylation (OXPHOS) levels and oxidative stress in the brain. GLP-1 also alleviated Aβ-induced glycolysis declines in astrocytes, which resulted in reduced OXPHOS levels and reactive oxygen species (ROS) production. The mechanism involved the activation of the PI3K/Akt pathway by GLP-1. Elevation in astrocytic glycolysis improved astrocyte cells’ support of neurons and promoted neuronal survival and axon growth. Conclusions: Taken together, we revealed GLP-1's capacity to regulate astrocytic glycolysis, providing mechanistic insight into one of its neuroprotective roles in AD and support for the feasibility of energy regulation treatments for AD.

Radiation effect test cannot be performed in large-scale space simulations of a strong field electromagnetic environment, and existing BCI (bulk current injection) technology employed for nonlinear system tests is subjected to its limitation. Given this, the present study analyzes the equivalent radiation test method of BCI in the parallel double-line coupling channel. To be specific, the response analysis models of the EUT (equipment under test) under the radiation condition and the injection condition are developed with the nonlinear response system as the EUT. In addition, the corresponding equivalent relationship is derived between the voltage of the injection excitation source and the radiation field intensity. Subsequently, the condition and the test method to substitute BCI for radiation are proposed and experimentally verified. As revealed from the research results, specific to nonlinear response systems, the BCI method proposed here can be accurately equated with the radiation effect test of the EUT. It is noteworthy that the maximum test error only reaches 3.62 dB, capable of satisfying the actual requirements of the project.

This article analyzes the relationship between the water level and the water-tube tilting in Shuangyang lake, based on the differential deformation features reflected by the NS and EW components of the water-tube tiltmeter. The results show a good spatiotemporal consistency between the variation of water level and the NS tilt component, which is considered to be affected by the magnitude and duration of the water level variation in Shuangyang Lake. The article uses Landsat remote sensing image data to extract the water boundary of Shuangyang Lake, and takes advantage of the finite element numerical simulation method to build three-dimensional models for different geological structural conditions of the Shuangyang seismostation. The simulation results show that when the underground medium is granite, the effect of water level variation on the vertical displacement of the surface is non-directional. With a 50-m soil layer in Model 2, the simulated NS tilt variation is equivalent to the actual observed water-tube tiltmeter NS component when the water level variation is 0.44 m and 0.8m. When the variation of water level reaches 2.0m, the simulation result of the NS component is 79.6 ​ms, which is slightly larger than the observed result of 60.32 ​ms. However, the simulation results show that the variation of the EW component is significantly smaller than that of the NS one. Due to the fact that the Shuangyang lake is long in the NS direction and short in the EW direction, the existence of the soil layer tends to generate ground deformation along the NS direction in the vicinity of the lake after the increase of water level, thereby resulting in the difference of the ground deformation in the two directions.

The large-diameter single pile has been widely used in marine wind turbine foundations. In order to improve its lateral bearing capacity, the suction bucket foundation (BF) can be used as a reinforcement method, which surrounds the periphery of the monopile foundation (PF), forming a new wind turbine foundation, i.e., pile–bucket foundation (P–BF). By using the general finite element software ABAQUS, in this paper, several numerical models are established to investigate the influence of pile diameter and bucket diameter on the lateral bearing capacity of large-diameter monopiles. The numerical results show that the internal force distribution along the pile shaft for the case of P–BF is similar to the case of PF. Compared to bucket height, bucket diameter has more of an effect on lateral capacity and the p-y curve in the P–BF. In the combined P–BF, the buckets can provide lateral bearing capacity, and the piles can provide anti-overturning moments, resulting in higher lateral bearing capacity. In this paper, the p-y curve in the API specification is modified based on the results from the finite element simulation. The modified p-y curve fits well with the results of the finite element calculation and can be used as a reference for the design of the P–BF in actual engineering.

The metabolic regulator protein family, mTOR, regulate natural killer (NK) cell development and function, but the underlying mechanism is unclear. Here, the authors show that Raptor/mTORC1 and Rictor/mTORC2 form a feedback crosstalk network to variegate cytokine and cellular signaling and modulate NK maturation and effector functions.

Background: Ischemic stroke can induce changes in mitochondrial morphology and function. As a regulatory gene in mitochondria, optic atrophy 1 (OPA1) plays a pivotal role in the regulation of mitochondrial dynamics and other related functions. However, its roles in cerebral ischemia-related conditions are barely understood. Methods: Cultured rat primary cortical neurons were respectively transfected with OPA1-v1ΔS1-encoding and OPA1-v1-encoding lentivirus before exposure to 2-h oxygen-glucose deprivation (OGD) and subsequent reoxygenation (OGD/R). Adult male SD rats received an intracranial injection of AAV-OPA1-v1ΔS1 and were subjected to 90 min of transient middle cerebral artery occlusion (tMCAO) followed by reperfusion. OPA1 expression and function were detected by in vitro and in vivo assays. Results: OPA1 was excessively cleaved after cerebral ischemia/reperfusion injury, both in vitro and in vivo. Under OGD/R condition, compared with that of the LV-OPA1-v1-treated group, the expression of OPA1-v1ΔS1 efficiently restored L-OPA1 level and alleviated neuronal death and mitochondrial morphological damage. Meanwhile, the expression of OPA1-v1ΔS1 markedly improved cerebral ischemia/reperfusion-induced motor function damage, attenuated brain infarct volume, neuronal apoptosis, mitochondrial bioenergetics deficits, oxidative stress, and restored the morphology of mitochondrial cristae and mitochondrial length. It also preserved the mitochondrial integrity and reinforced the mtDNA content and expression of mitochondrial biogenesis factors in ischemic rats. Interpretation: Our results demonstrate that the stabilization of L-OPA1 protects ischemic brains by reducing neuronal apoptosis and preserving mitochondrial function, suggesting its significance as a promising therapeutic target for stroke prevention and treatment.

A truck platoon is a set of virtually linked trucks that travel in tandem with small intervehicle distances. Several studies have proved that traveling in platoons can significantly improve fuel economy due to the reduced aerodynamic drag. However, most literature only provides scattered pieces of information regarding fuel economy in truck platoons. Therefore, a literature survey is needed to understand what has been studied and what problems remain to be further addressed. This paper presents an overview of existing studies to illustrate the state of the art about fuel savings for truck platooning. Specifically, it summarized the methodologies, the contributing factors of fuel consumption, the coordination methods to improve the platooning rate, and the look-ahead control strategies to generate fuel-efficient speed profiles for each vehicle driving in a platoon over different road grades. After that, the autonomous truck platooning was introduced, and we raised and discussed a couple of outstanding questions to be addressed in future work.

Two hypotheses for the blocking effect mechanisms under the electromagnetic radiation are presented, and the corresponding dual-frequency interference prediction models are proposed, which can be used to predict the working state of equipment under the condition that the single-frequency susceptibility is known. By taking a radar and a communication radio as the EUT, the experiments of dual-frequency continuous wave electromagnetic radiation are designed and carried out to verify the prediction models. The test results show that the sensitive bandwidth of blocking interference effect for the tested radar and the communication station is consistent with their working bandwidth, respectively. By substituting the test data into the proposed prediction models, the prediction errors of the models are less than 13%. Moreover, it can be found that the tested radar is more sensitive to the peak value of electric field, and the tested communication station is more sensitive to the effective value of electric field.

Abstract Background C-kit-positive cardiac stem cells (CSCs) have been shown to be a promising candidate treatment for myocardial infarction and heart failure. Insulin-like growth factor (IGF)-1 is an anabolic growth hormone that regulates cellular proliferation, differentiation, senescence, and death in various tissues. Although IGF-1 promotes the migration and proliferation of c-kit-positive mouse CSCs, the underlying mechanism remains unclear. Methods Cells were isolated from adult mouse hearts, and c-kit-positive CSCs were separated using magnetic beads. The cells were cultured with or without IGF-1, and c-kit expression was measured by Western blotting. IGF-1 induced CSC proliferation and migration, as measured through Cell Counting Kit-8 (CCK-8) and Transwell assays, respectively. The miR-193a expression was measured by quantitative real-time PCR (qPCR) assays. Results IGF-1 enhanced c-kit expression in c-kit-positive CSCs. The activities of the phosphoinositol 3-kinase (PI3K)/AKT signaling pathway and DNA methyltransferases (DNMTs) were enhanced, and their respective inhibitors LY294002 and 5-azacytidine (5-AZA) blunted c-kit expression. Based on the results of quantitative real-time PCR (qPCR) assays, the expression of miR-193a, which is embedded in a CpG island, was down-regulated in the IGF-1-stimulated group and negatively correlated with c-kit expression, whereas c-kit-positive CSCs infected with lentivirus carrying micro-RNA193a displayed reduced c-kit expression, migration and proliferation. Conclusions IGF-1 upregulated c-kit expression in c-kit-positive CSCs resulting in enhanced CSC proliferation and migration by activating the PI3K/AKT/DNMT signaling pathway to epigenetically silence miR-193a, which negatively modifies the c-kit expression level.

Abstract Recent data have shown that the expression of lysosome-associated membrane protein type 2 A (LAMP2A), the key protein in the chaperone-mediated autophagy (CMA) pathway, is elevated in breast tumor tissues. However, the exact effects and mechanisms of CMA during breast cancer metastasis remain largely unknown. In this study, we found that the LAMP2A protein level was significantly elevated in human breast cancer tissues, particularly in metastatic carcinoma. The increased LAMP2A level was also positively correlated with the histologic grade of ductal breast cancer. High LAMP2A levels also predicted shorter overall survival of breast cancer patients. Downregulation of CMA activity by LAMP2A knockdown significantly inhibited the growth and metastasis of both MDA-MB-231 and MDA-MB-468 breast cancer cells in vivo and in vitro, while upregulation of CMA activity by LAMP2A overexpression had the opposite effect. Mechanistically, we found that elevated CMA activity mediated increased growth and metastasis of human breast cancer cells by downregulating the activity of autophagy-related gene 5 (ATG5)-dependent macroautophagy. Collectively, these results indicate that the anti-macroautophagic property is a key feature of CMA-mediated tumorigenesis and metastasis and may, in some contexts, serve as an attractive target for breast cancer therapies.

Background/Aims: In liver fibrosis, the activation of hepatic stellate cells (HSCs) is considered as a pivotal event. It is well known that transforming growth factor-β1 (TGF-β1) is the main stimuli factor responsible for HSC activation. microRNAs (miRNAs), regulating various biological processes, have recently been shown to be involved in HSC activation. A recent study reported that deficiency of miR-378a contributes to cardiac fibrosis via TGF-β1-dependent paracrine mechanism. However, the involvement of miR-378a and its roles in TGF-β1-induced HSC activation remains largely unknown. Methods: miR-378a expression was detected in TGF-β1-treated cells and patients with cirrhosis. Then, effects of miR-378a overexpression on cell proliferation and HSC activation were analyzed. We also analyzed the binding of miR-378a to the 3′-untranslated region of TGF-β2. Results: In response to TGF-β1, miR-378a expression was down-regulated in a dose-dependent manner. miR-378a overexpression suppressed both cell proliferation and cell cycle in TGF-β1-treated LX-2 cells. Moreover, miR-378a overexpression inhibited TGF-β1-induced HSC activation including the reduction of α-smooth muscle actin (α-SMA) and type I collagen. Similarly, miR-378a resulted in a reduction in cell proliferation, and the expressions of α-SMA and Col1A1 in TGF-β1-treated primary HSCs. Notably, TGF-β2 was confirmed as a target of miR-378a by luciferase reporter assays. Interestingly, miR-378a promoter methylation may be responsible for miR-378a down-regulation in TGF-β1-treated LX-2 cells and TGF-β1-treated primary HSCs. Further studies confirmed that reduced miR-378a was associated with promoter methylation in patients with cirrhosis compared with healthy controls. Conclusion: Our results demonstrate that miR-378a expression is associated with its methylation status in TGF-β1-treated cells, and epigenetically-regulated miR-378a inhibits TGF-β1-induced HSC activation, at least in part, via TGF-β2.

Using the foundation pit at the Jianye Road Station of Hangzhou Metro Line 6 as a case study, the deformation characteristics of a foundation pit subjected to asymmetric loads is investigated in this paper using PLAXIS 3D numerical simulation software. The influence of active pressure zone reinforcement, passive pressure zone reinforcement, and increased thickness of the diaphragm wall at the loaded side on the maximum lateral displacement of diaphragm wall retaining structure of foundation pit is also systematically analyzed. The results show that the deformation of the diaphragm walls on both sides of the foundation pit is strongly inconsistent when subjected to asymmetric loads and is affected by the asymmetric load value and its distance to the foundation pit. In order to reduce the lateral deformations of foundation pit subjected to asymmetric load, two displacement control methods are adopted in the numerical simulations. It is shown that reinforcing the active pressure zone can reduce the maximum lateral displacement of the diaphragm wall on the loaded side to a certain extent but the reinforcement should have a certain depth, resulting in poor efficiency. On the other hand, reinforcing the passive pressure zone can effectively reduce the difference in lateral deformation between the two sides of the foundation pit by increasing the depth and width of the reinforcement zones. It is also observed that the increase in the thickness of the diaphragm wall can reduce the adverse effect of asymmetric loads on the foundation pit. The research results can provide reference for using measures to reduce the influence of asymmetric loads.

Synthetic aperture radar (SAR) altimeters represent a new method of microwave remote sensing for ocean wave observations. The adoption of SAR technology in the azimuthal direction has the advantage of a high resolution. The Sentinel-3 altimeter is the first radar altimeter to acquire global observations in SAR mode; hence, the data quality needs to be assessed before extensively applying these data. The European Space Agency (ESA) evaluates the Sentinel-3 accuracy on a global scale but has yet to perform a detailed analysis in terms of different offshore distances and different water depths. In this paper, Sentinel-3 and Jason-2 significant wave height (SWH) data are matched in both time and space with buoy data from the United States East and West Coasts and the Central Pacific Ocean. The Sentinel-3 SWH data quality is evaluated according to different offshore distances and water depths in comparison with Jason-2 SWH data. In areas more than 50 km offshore, the Sentinel-3 SWH accuracy is generally high and less affected by the water depth and sea conditions (root-mean-square error of 0.28 m and correlation coefficient of 0.98); in areas less than 50 km offshore, the SWH data accuracy is slightly affected by water depth and sea conditions (especially the former). Compared with Jason-2, the observation ability of the Sentinel-3 altimeter in nearshore areas with water depths of 0 m-500 m is greatly improved, but in some deep water areas with stable sea conditions, the Jason-2 SWH data accuracy is higher than that of Sentinel-3. This work provides a reference for the refined application of Sentinel-3 SWH data in offshore deep water areas and nearshore shallow water areas.

As a secreted axon guidance molecule, Netrin-1 has been documented to be a neuroprotective factor, which can reduce infarct volume, promote angiogenesis and anti-apoptosis after stroke in rodents. However, its role in axonal regeneration and synaptic formation after cerebral ischemic injury, and the related underlying mechanisms remain blurred. In this study, we used Adeno-associated vectors carrying Netrin-1 gene (AAV-NT-1) to up-regulate the expression level of Netrin-1 in rats’ brain after middle cerebral artery occlusion (MCAO). We found that the up-regulated level of Netrin-1 and its receptor DCC promoted axonal regeneration and synaptic formation; the overexpression of Netrin-1 activated the JNK1 signaling pathway; these effects were partially reduced when JNK1 signaling pathway was inhibited by SP600125 (JNK specific inhibitor). Taken together, these findings suggest that Netrin-1 can facilitate the synaptic formation and axonal regeneration via the JNK1 signaling pathway after cerebral ischemia, thus promoting the recovery of neural functions.

Resistance to 5-fluorouracil (5-FU) and its induced immune suppression have prevented its extensive application in the clinical treatment of breast cancer. In this study, the combined effect of 50 Hz-EMFs and 5-FU in the treatment of breast cancer was explored. MCF-7 and MCF10A cells were pre-exposed to 50 Hz-EMFs for 0, 2, 4, 8 and 12 h and then treated with different concentrations of 5-FU for 24 h; cell viability was analyzed by MTT assay and flow cytometry. After pre-exposure to 50 Hz-EMFs for 12 h, apoptosis and cell cycle distribution in MCF-7 and MCF10A cells were detected via flow cytometry and DNA synthesis was measured by EdU incorporation assay. Apoptosis-related and cell cycle-related gene and protein expression levels were monitored by qPCR and western blotting. Pre-exposure to 50 Hz-EMFs for 12 h enhanced the antiproliferative effect of 5-FU in breast cancer cell line MCF-7 in a dose-dependent manner but not in normal human breast epithelial cell line MCF10A. Exposure to 50 Hz-EMFs had no effect on apoptosis and P53 expression of MCF-7 and MCF10A cells, whereas it promoted DNA synthesis, induced entry of MCF-7 cells into the S phase of cell cycle, and upregulated the expression levels of cell cycle-related proteins Cyclin D1 and Cyclin E. Considering the pharmacological mechanisms of 5-FU in specifically disrupting DNA synthesis, this enhanced inhibitory effect might have resulted from the specific sensitivity of MCF7 cells in active S phase to 5-FU. Our findings demonstrate the enhanced cytotoxic activity of 5-FU on MCF7 cells through promoting entry into the S phase of the cell cycle via exposure to 50 Hz-EMFs, which provides a novel method of cancer treatment based on the combinatorial use of 50 Hz-EMFs and chemotherapy.

In the nervous system, Netrin-1 serves as a neural guide, mediating the neuronal development. However, it remains blurred whether Netrin-1 can protect neurons from apoptosis induced by cerebral stroke. In the current study, the cultured rat primary cortical neurons were transfected with Netrin-1-encoding lentivirus before the oxygen-glucose-deprivation (OGD) treatment. Cell death and apoptosis were evaluated by lactate dehydrogenase (LDH) release and flow cytometry. We found that Netrin-1 attenuated OGD-induced cell death and neuronal apoptosis at 24 h after OGD treatment, and that the overexpression of Netrin-1 activated the ERK signaling pathway. These effects were partly abolished by blocking its receptor deleted in colorectal cancer (DCC) or U0126, an inhibitor of the ERK signaling pathway. Netrin-1 overexpression in neurons elevated the expression of DCC, on mRNA level and protein level. Netrin-1 also reduced DNA damage. Taken together, our findings suggest that Netrin-1 attenuates cell death and neuronal apoptosis via the DCC/ERK signaling pathway in the cultured primary cortical neurons after OGD injury, which may involve the mediation of DNA damage in the neurons.

Utilizing the EMR-8B eye-tracker system, the pupil changes of eight drivers were monitored when they drove through 26 typical highway tunnels. Based on the test results, the driver’s pupil areas and pupil illuminance were found to be in a power function relationship at tunnel entrances. Furthermore, a quantitative relationship between the pupil area and its critical velocity was established, and the ratio of pupil area’s velocity in relation to its critical velocity was used to evaluate the lighting transitions and to establish the ideal curve of pupil illuminance at tunnel entrances. The results demonstrated that the relationship between the pupil illuminance of the tunnel entrance and the driver’s pupil areas conforms to the Stevens law found in experimental psychology; severe pupil illuminance transition within the range of 10 metres of the existing highway tunnel entrances, which results in great visual load, is in urgent need of improvement.

Prenatal exposure to an inflammatory stimulus has been shown to cause renal damage in offspring. Our present study explored the role of intra-renal NF-κB activation in the development of progressive renal fibrosis in offspring that underwent prenatal exposure to an inflammatory stimulus. Time-dated pregnant rats were treated with saline (control group) or 0.79 mg/kg lipopolysaccharide (LPS) through intra-peritoneal injection on gestational day 8, 10 and 12. At the age of 7 weeks, offspring from control or LPS group were treated with either tap water (Con+Ve or LPS+Ve group) or pyrollidine dithiocarbamate (PDTC, 120 mg/L), a NF-κB inhibitor, via drinking water starting (Con+PDTC or LPS+PDTC group), respectively, till the age of 20 or 68 weeks. The gross structure of kidney was assessed by hematoxylin-eosin, periodic acid-Schiff staining and Sirius red staining. The expression levels of TNF-α, IL-6, α-smooth muscle actin (α-SMA) and renin-angiotensin system (RAS) genes were determined by real time polymerase chain reaction and/or immunohistochemical staining. Our data showed that post-natal persistent PDTC administration efficiently repressed intra-renal NF-κB activation, TNF-α and IL-6 expression. Post-natal PDTC also prevented intra-renal glycogen deposition and collagenous fiber generation as evident by the reduced expression of collagen III and interstitial α-SMA in offspring of prenatal LPS exposure. Furthermore, post-natal PDTC administration reversed the intra-renal renin-angiotensin system (RAS) over-activity in offspring of prenatal LPS exposure. In conclusion, prenatal inflammatory exposure results in offspring's intra-renal NF-κB activation along with inflammation which cross-talked with excessive RAS activation that caused exacerbation of renal fibrosis and dysfunction in the offspring. Thus, early life prevention of NF-κB activation may be a potential preventive strategy for chronic renal inflammation and progressive renal damage.

This paper deals with propositional fuzzy modal logic with evaluated syntax based on MV-algebras. We focus on its semantical theory from the viewpoint of Pavelka's graded semantics of propositional fuzzy logic, investigate the L-tautologies based on different Kripke frames. We also define the notion of L-semantical consequence operation, its some basic properties are obtained. Finally, this paper considers the fuzzy decision implications in propositional fuzzy modal logic with evaluated syntax based on MV-algebras, and presents a kind of semantical characteristics of fuzzy decision implications. Moreover, we introduce the notions of possible and necessary fuzzy decision implication, and their semantical characteristics are presented as well.

In this paper, an α-resolution method for a set of lattice-valued Horn generalized clauses is established in lattice-valued propositional logic system ℒ P(X) based on lattice implication algebra. Firstly, the notions of lattice-valued Horn generalized clause, normal lattice-valued Horn generalized clause and unit lattice-valued Horn generalized clause are given in ℒ P(X). Then, the α-resolution of two lattice-valued Horn generalized clauses is represented in ℒ P(X). It indicates the reasoning rules in a resolution process, which aims at deleting α-resolution literals and obtaining a resolvent. Finally, we build an α-resolution algorithm for a set of lattice-valued Horn generalized clauses in ℒ P(X). It provides a foundation for automated reasoning in lattice-valued first-order logic system and an application for designing an inference system in the field of intelligent decision support.
