Anthracnose is one of the most common diseases in strawberry plants. Colletotrichum gloeosporioides is the major cause of anthracnose in China, including Zhejiang Province. Early, specific, reliable, and time-saving detection is urgently needed to prevent the further spread of C. gloeosporioides, guiding farmers to utilize chemicals to control anthracnose. In this study, we showed that the high resistance to pyraclostrobin, caused by a point mutation at codon 143 (GGT→GCT) in the cytochrome b gene of C. gloeosporioides was prevalent in the strawberry growing regions, and we developed a loop-mediated isothermal amplification (LAMP) assay as a detection method. Primer sets S0 and S4 could be used to specifically detect C. gloeosporioides isolates and the G143A mutations, respectively. A detection limit of 10−2 ng (10 pg), which is at least 10-fold more sensitive than conventional polymerase chain reaction, was achieved by the LAMP assay. Here, we utilized lateral-flow devices (LFDs), nitrocellulose membranes that can absorb nucleic acids, to acquire the total genomic DNA of strawberry plants within 2 min. The LFD membranes were used as DNA templates for the LAMP assays to accurately detect strawberry plants infected with C. gloeosporioides. This diagnostic method for strawberry anthracnose was accomplished within 1 h, including the sample preparation and LAMP assays. Collectively, we developed a sensitive and practical method for monitoring C. gloeosporioides and its quinone outside inhibitor–resistant mutants. The LAMP assay for detection of C. gloeosporioides in strawberry plants has great potential for rapid strawberry anthracnose surveillance and will provide farmers with advice on preventing C gloeosporioides at the early stages of strawberry development.

目的 研究Erythroferrone（ERFE）等铁代谢红系调节因子（iron-regulatory erythroid factor）在不同类型红系造血异常疾病中的表达情况。 方法 采用ELISA方法检测2016年1月至2019年11月共47例真性红细胞增多症（PV）、纯红细胞再生障碍（PRCA）、自身免疫性溶血性贫血（AIHA）和骨髓增生异常综合征（MDS）患者血浆ERFE、生长分化因子15（GDF15）、生长分化因子11（GDF11）和扭转原肠胚形成同系物（TWSG1）的表达，分析铁代谢调节因子与红系造血异常类型及旺盛程度（以骨髓有核红细胞比例反映）的适配性。 结果 血浆GDF15表达水平在PV、PRCA、AIHA、MDS各组依次为266.01（112.40,452.37）、110.63（81.41,220.42）、52.11（32.61,171.66）、276.53（132.16,525.70）ng/L，均显著高于正常对照组的37.45（19.65,57.72）ng/L（P值均0.05）。血浆GDF11表达水平仅在PV组患者中明显高于正常对照组[74.75（10.95,121.32）ng/L对36.90（3.38,98.34）ng/L，P0.05）。PV组血浆ERFE水平为129.63（47.02, 170.03）ng/L，AIHA组血浆ERFE水平最高为121.76（68.12,343.11）ng/L，二者均明显高于正常对照组的43.23（35.18,65.41）ng/L（P值均0.05）。骨髓有核红细胞比例与ERFE（r＝0.458，P＝0.001）呈正相关，而与GDF15（r＝−0.163，P＝0.274）、GDF11（r＝0.120，P＝0.421）、TWSG1（r＝−0.166，P＝0.269）无明显相关性。 结论 铁代谢红系调节因子在不同红系造血异常疾病的表达谱不尽一致，ERFE与红系造血旺盛程度相关度最高。

BackgroundSystemic lupus erythematosus (SLE) is a chronic autoimmune disease characterized by over-activity of lymphocytes, production of autoantibodies and effects on multiple organs 1. Growing evidences suggest long noncoding RNAs (lncRNAs) and mRNAs widely participate in physiological and pathological processes. However, knowledge of related lncRNAs and mRNAs in SLE remains limited.ObjectivesThe aim of our study is to investigate the levels of differential expression of lncRNAs and mRNAs in the peripheral blood mononuclear cells (PBMCs) of SLE patients and their correlation with disease activity, clinical features and cell differentiation.MethodsPeripheral venous blood 4ml were collected from 11 patients with SLE before and after treatment and 11 sex-and age-matched healthy individuals and saved in EDTA tubes. PBMCs were isolated from peripheral blood samples by Ficoll-Histopaque density gradient centrifugation. Total RNA was extracted from PBMCs with TRIzol reagent. RNAs amount and quality were quantified by using a NanoDrop ND-1000. Peripheral blood samples were sent to Novogene Co. Ltd (Beijing, China) for sequencing. The DESeq package in R language was used to analyze the differential expression of lncRNAs and mRNAs in the two groups. GO and KEGG databases analyze the potential biological functions and signal transduction and disease pathways affected by abnormal expression of lncRNAs and mRNAs2.ResultsAccording to the RNAs expression profiles, 338 lncRNAs (173 upregulated and 165 downregulated) and 2020 mRNAs (1292 upregulated and 728 downregulated) were differentially expressed between SLE patients and control groups. In addition, 17 lncRNAs were significantly downregulated and 66 mRNAs (47 upregulated and 19 downregulated) were differentially expressed between active and treated SLE patients. There were 1645 RNAs up-expression in active SLE patients and 36 RNAs under-expression in treated SLE patients, and total 14 RNAs changed direction of expression. GO and KEGG pathway analysis showed most of mRNAs were related to transcription, inflammation and immunity. The relativity between aberrantly expressed RNAs and clinical characteristics of active and treated SLE patients were shown in Table 1.ConclusionDysregulation of lncRNAs and mRNAs involves in molecular regulation of SLE, which may support for diagnosis or determination of the susceptibility of individuals of SLE.References[1]Tsokos GC. BackgroundSystemic lupus erythematosus (SLE) is a multi-system autoimmune disease characterized with the secretion of normal autoantibodies, which leads to the involvement of multiple organs. Several observations have showed that the breakdown of immune tolerance was involved in the pathogenesis of SLE, especially the imbalance of Th17/Treg [1], but the mechanism of the impaired immune tolerance is unknown yet. Numerous researches support that intestinal mucosal barrier is an upstream mechanism leading to impaired immune balance [2]. However, the relationship between intestinal mucosal barrier and SLE is little known.ObjectivesTo measure the expression of serum zonulin to assess and compare the function of intestinal mucosal barrier in patients with SLE and healthy adults, and to explore the role of intestinal mucosal barrier in the pathogenesis and development of SLE.Methods20 patients with SLE who hospitalized at the Second Hospital of Shanxi Medical University and 10 age and gender-matched healthy adults were enrolled in this study. We collected the blood sample of the patients and healthy controls to examine the function of intestinal mucosal barrier and the absolute number of Th17 and Treg cells. The level of serum zonulin was measured by ELISA to assess the function of intestinal mucosal barrier. The absolute number of Th17 cells and Treg cells was detected by flow cytometry. Disease activity indicators of patients with SLE were collected from laboratory including erythrocyte sedimentation rate (ESR, mm/h), complement 3(C3, g/l) and anti-dsDNA (IU/ml). Anthracnose is one of the most common diseases in strawberry plants.

The effect of natural killer (NK) cell alloreactivity on the outcome of unrelated hematopoietic SCT (HSCT) remains a topic of debate. NK cell alloreactivity after allogeneic HSCT is regulated by killer-cell Ig-like receptors (KIRs). To investigate the influence of KIRs on outcome after unrelated HSCT, we retrospectively analyzed the HLA and KIR genotypes of 116 donor-recipient pairs. We found that missing KIR ligands in recipients were significantly associated with a decreased leukemic relapse risk (P=0.019, HR=0.329), mainly in myeloid disease (P=0.003, HR=0.193). This beneficial effect was seen in AML/myelodysplastic syndrome and also in chronic myeloid leukemia. In myeloid disease, missing KIR ligands also improved 5-year OS (P=0.034, HR=0.430) and disease-free survival (DFS) (P=0.024, HR=0.445). Meanwhile, the presence of donor-activating KIR2DS3 gene was associated with increased relapse risk (P=0.003, HR=5.046), decreased OS (P=0.004, HR=3.181) and DFS (P=0.003, HR=2.919) in myeloid disease. No effect was seen in patients with lymphoid disease. Our study indicated that, in unrelated HSCT for myeloid leukemia, missing KIR ligands in recipients offered a lower relapse risk and a long-term survival advantage. The presence of KIR2DS3 in the donor was an important risk factor for myeloid leukemia.

Discussion A great deal of attention has been devoted to the medicine with chiral center, since they usually have special biological activity. Consequently, much interest has been focused on obtaining compounds with such a feature. The title compound is a chiral intermediate used for the synthesis of cetirizine hydrochloride. It consists of three different rings,two phenyl rings and a piperazine ring. The dihedral angle between two phenyl rings is 116.78(6)°. Futhermore, piperazine ring is a chair-like conformation, this can be seen from the distance of N1 and N2 to the plane of C2-C3-C4-C5 (-0.699 A and 0.597 A, respectively). CNE-2Z-H5 was transplanted into the Scid mice of which immune system was reconstituted with the peripheral lymphocytes or thymus of the isogenetic mice and the BALB/c nude mice of which NK cells were inhibited with cyclophosphamide. The results showed that the growth speed and weight of the tumors were in order from high to low among groups of Scid mice, BALB/c nude mice with NK inhibited, BALB/c nude mice, Scid mice with immune reconstitution. The transplanted tumor, in 6/6 cases injected beforehand with peripheral lymphocytes and 1/3 case transplanted accompanied with thymus disappeared within 22 days. The conclusion is that the immune reconstitution of Scid mice has succeeded and play an effective role in antitumor response.
