This study investigates the characteristics of submerged vortices in an intake sump through a combination of numerical simulations, experimental validations, and advanced modeling techniques. The aim of this study is to gain insights into the complex flow patterns and vortex structures within the sump, focusing on their behavior under varying flow rates. The Shear Stress Transfer (SST) k-ω model is utilized to capture turbulence, and the Volume of Fluid (VOF) method is employed to visualize the water-air interface. Model tests are conducted to validate the simulations. The findings suggest that under low flow conditions, the flow beneath the bell mouth becomes highly turbulent, leading to the formation of a complex vortex system with three distinct high-pressure zones. With increasing flow rates, the shape and strength of these high-pressure zones fluctuate, and a quadrupole vortex structure emerges at the sump bottom. This quadrupole vortex plays a pivotal role in the transformation of a floor-attached vortex upward, culminating in a dual vortex column structure. This structure, in turn, generates additional low-amplitude pressure pulsations. Wall-attached vortices are also observed on both sides of the inlet pipe, a result of flow stratification due to velocity disparities. The insights gained from this study contribute to a deeper understanding of intake sump dynamics and offer valuable guidance for designing and optimizing fluid systems to mitigate potential turbulence-related issues.

Understanding Holocene temperature changes is vital for resolving discrepancies between proxy reconstructions and climate models. The intricate temperature variations across the Tibetan Plateau (TP) add complexity to studying continental climate change during this period. Discrepancies between model-based and proxy-based reconstructions might stem from seasonal biases and environmental uncertainties in the proxies. Employing multiple proxies from a single sediment core for quantitative temperature reconstructions offers an effective method for cross-validation in terrestrial environments. Here, we present an ice-free-season temperature record for the past 15 kyr from a shallow, freshwater lake on the eastern TP, based on brGDGTs (branched glycerol dialkyl glycerol tetraethers). This record shows that the Holocene Thermal Maximum lags the pollen-based July temperature recorded in the same sediment core. We conclude that the mismatch between the brGDGT-based and pollen-based temperatures is primarily the result of seasonal variations in solar irradiance. The overall pattern of temperature changes is supported by other summer temperature records, and the Younger Dryas cold event and the Bølling–Allerød warm period are also detected. A generally warm period occurred during 8–3.5 ka, followed by a cooling trend in the late Holocene. Our findings have implications for understanding the seasonal signal of brGDGTs in shallow lakes and provide critical data for confirming the occurrence of seasonal biases in different proxies from high-elevation lakes. To further investigate the significance of the brGDGTs and temperature patterns on the TP, we examined existing brGDGT-based Holocene temperature records, which interpret these compounds as indicators of mean annual or growing season temperatures. The existing and available temperature records show complicated patterns of variation, some with general warming trends throughout the Holocene, some with cooling trends, and some with a warm middle Holocene. We analyzed the possible reasons for the diverse brGDGTs records on the TP and emphasize the importance of considering lake conditions and modern investigations of brGDGTs in lacustrine systems when using brGDGTs to reconstruct paleoenvironmental conditions.

Cook Inlet beluga (CIB), Delphinapterus leucas, have not recovered from subsistence overharvest despite conservation efforts initiated in 2000. Reasons for this lack of recovery are still unclear with anthropogenic noise identified as a high threat in this population’s recovery plan. Baseline information on CIB habitat use and soundscape characterization is crucial in evaluating impacts from anthropogenic activities. Using passive acoustic monitoring, we documented the seasonality and foraging occurrence of CIB, together with killer whale (Orcinus orca), harbor porpoise (Phocoena phocoena) and humpback whale (Megaptera novaeangliae), for one year in Chinitna and Tuxedni bays and rivers, two historically important but recently overlooked areas of their critical habitat in western lower Cook Inlet, Alaska. This area’s ecological interest converges with planned mining, marine renewable energy, offshore oil and gas production, and commercial shipping. Results revealed higher presence of harbor porpoise, killer whale, and humpback whale in Chinitna compared to Tuxedni but much lower CIB presence. CIB were not detected in Chinitna Bay but were in the river on 21 days September-February and one day in June. However, CIB were never detected feeding in Chinitna River. CIB acoustic crypsis and preferential use of very shallow habitat in Chinitna revealed a perceived predation risk from killer whales. In contrast, CIB were recorded foraging in both Tuxedni Bay and River and were detected September-April on 127 days making it an important winter foraging ground. Low levels of anthropogenic noise disturbance were quantified in both bays making them some of the most undisturbed sections of their critical habitat, but at the same time highly sensitive to further noise disturbance. Commercial shipping was the main noise source likely causing acoustic disturbance to CIB due to communication masking despite current low levels of temporal overlap with beluga presence. We recommend that applicable regulators consider restricting high sound producing anthropogenic activities in and adjacent to Tuxedni Bay from September 1st to May 15th when CIB are actively using this area. Managing future anthropogenic activities to minimize habitat degradation and acoustic disturbance in this winter foraging refuge will be a key component to support the recovery of this endangered population.

Under future net-zero emission policies, reductions in emissions of ozone (O3) precursors are expected to alter the temporal and spatial distributions of tropospheric O3. In this study, we quantify changes in the tropospheric O3 budget and in the spatiotemporal distribution of surface O3 in east Asia and the contributions of regional emissions, intercontinental transport and climate change between the present day and 2060 under a net-zero scenario using the NCAR Community Earth System Model (CESM) with online tagging of O3 and its precursors. The results reveal that the global tropospheric O3 burden is likely to decrease by more than 20 %, from 316 Tg in the present day to 247 Tg in 2060, under a net-zero scenario. The burden of stratospheric O3 in the troposphere is expected to increase from 69 to 77 Tg. The mean lifetime of tropospheric O3 is expected to increase by 2 d (∼10 %). Changes in climate under a net-zero pathway are relatively small and only lead to small increases in tropospheric O3. Over eastern China, surface O3 increases in winter due to the weakened titration of O3 by NO associated with reduced anthropogenic NO emissions and due to enhanced stratospheric input. In summer, surface O3 decreases by more than 30 ppbv, and peak concentrations shift from July to May. Local contributions from anthropogenic emissions to surface O3 over east Asia are highest in summer but drop substantially, from 30 % to 14 %, under a net-zero scenario. The contribution of biogenic NO sources is enhanced and forms the dominant contributor to future surface O3, especially in summer (∼40 %). This enhanced contribution is mainly due to the increased O3 production efficiency under lower anthropogenic precursor emissions. Over eastern China, local anthropogenic contributions decrease from 50 % to 30 %. The decreases in surface O3 are strongly beneficial and are more than sufficient to counteract the increases in surface O3 observed in China over recent years. This study thus highlights the important co-benefits of net-zero policies that target climate change in addressing surface O3 pollution over east Asia.

Abstract The anti‐tumor immune response relies on interactions among tumor cells and immune cells. However, the molecular mechanisms by which tumor cells regulate DCs as well as DCs regulate T cells remain enigmatic. Here, the authors identify a super signaling complex in DCs that mediates the Arf1‐ablation‐induced anti‐tumor immunity. They find that the Arf1‐ablated tumor cells release OxLDL, HMGB1, and genomic DNA, which together bound to a coreceptor complex of CD36/TLR2/TLR6 on DC surface. The complex then is internalized into the Rab7‐marked endosome in DCs, and further joined by components of the NF‐κB, NLRP3 inflammasome and cGAS‐STING triple pathways to form a super signal complex for producing different cytokines, which together promote CD8+ T cell tumor infiltration, cross‐priming and stemness. Blockage of the HMGB1‐gDNA complex or reducing expression in each member of the coreceptors or the cGAS/STING pathway prevents production of the cytokines. Moreover, depletion of the type I IFNs and IL‐1β cytokines abrogate tumor regression in mice bearing the Arf1‐ablated tumor cells. These findings reveal a new molecular mechanism by which dying tumor cells releasing several factors to activate the triple pathways in DC for producing multiple cytokines to simultaneously promote DC activation, T cell infiltration, cross‐priming and stemness.

Situated within the triangle of the East Asian monsoon, the Indian monsoon, and the westerlies, the Holocene patterns of climate and vegetation changes on the northeast Tibetan Plateau are still unclear or even contradictory. By investigating the distribution of modern pollen taxa on the east Tibetan Plateau, we infer the past vegetation and climate since 14.2 ka BP (1000 years before present) from a fossil pollen record extracted from Gahai Lake (102.3133∘ E, 34.2398∘ N; 3444 m a.s.l.) together with multiple proxies (grain size, contents of total organic carbon and total nitrogen) on the northeast Tibetan Plateau. Results indicate that the Gahai Basin was covered by arid alpine steppe or even desert between 14.2 and 7.4 ka BP with dry climatic conditions, and high percentages of arboreal pollen are thought to be long-distance wind- transported grains. Montane forest (dominated by Abies, Picea, and Pinus) migrated into the Gahai Basin between 7.4 and 3.8 ka BP driven by wet and warm climatic conditions (the climate optimum within the Holocene) but reverted to alpine steppe between 3.8 and 2.3 ka BP, indicating a drying climate trend. After 2.3 ka BP, vegetation shifted to alpine meadow represented by increasing abundances of Cyperaceae, which may reflect a cooling climate. The strange pollen spectra with high abundances of Cyperaceae and high total pollen concentrations after ca. 0.24 ka BP (1710 CE) could be an indication of disturbance by human activities to some extent, but needs more direct evidence to be confirmed. Our study confirms the occurrence of a climate optimum in the mid-Holocene on the northeast Tibetan Plateau, which is consistent with climate records from the fringe areas of the East Asian summer monsoon, and provides new insights into the fluctuations in the intensity and extent of the Asian monsoon system.

We analyzed the globular cluster M5 (NGC 5904) using 15 yr of gamma-ray data from the Fermi Large Area Telescope (LAT). Using rotation ephemerides generated from Arecibo and FAST radio telescope observations, we searched for gamma-ray pulsations from the seven millisecond pulsars (MSPs) identified in M5. We detected no significant pulsations from any of the individual pulsars. In addition, we searched for possible variations of the gamma-ray emission as a function of orbital phase for all six MSPs in binary systems, but we did not detect any significant modulations. The gamma-ray emission from the direction of M5 is well described by an exponentially cutoff power-law spectral model, although other models cannot be excluded. The phase-averaged emission is consistent with being steady on a timescale of a few months. We estimate the number of MSPs in M5 to be between 1 and 10, using the gamma-ray conversion efficiencies for well-characterized gamma-ray MSPs in the Third Fermi-LAT Catalog of Gamma-ray Pulsars, suggesting that the sample of known MSPs in M5 is (nearly) complete, even if it is not currently possible to rule out a diffuse component of the observed gamma rays from the cluster.

We present the first catalog of very-high-energy and ultra-high-energy gamma-ray sources detected by the Large High Altitude Air Shower Observatory. The catalog was compiled using 508 days of data collected by the Water Cherenkov Detector Array from 2021 March to 2022 September and 933 days of data recorded by the Kilometer Squared Array from 2020 January to 2022 September. This catalog represents the main result from the most sensitive large coverage gamma-ray survey of the sky above 1 TeV, covering decl. from −20° to 80°. In total, the catalog contains 90 sources with an extended size smaller than 2° and a significance of detection at >5 σ . Based on our source association criteria, 32 new TeV sources are proposed in this study. Among the 90 sources, 43 sources are detected with ultra-high energy ( E > 100 TeV) emission at >4 σ significance level. We provide the position, extension, and spectral characteristics of all the sources in this catalog.

Neurodegenerative diseases, including Alzheimer’s disease (AD), are challenging to diagnose. Currently the field must rely on imperfect diagnostic modalities. A recent study identified differences in several key bio-mechano-physiological parameters of the skin between AD patients and healthy controls. Here, we visually align these differences with the relevant histological, aging, and embryological paradigms to raise awareness for these potential biomarkers. In a study conducted by Wu et al., a series of n = 41 patients (n = 29 with AD and n = 12 healthy controls) were evaluated, demonstrating that AD patients exhibit a less acidic skin pH, increased skin hydration, and reduced skin elasticity compared to healthy controls. We constructed a visual overview and explored the relevant paradigms. We present a visual comparison of these factors, highlighting four paradigms: (1) the findings emphasize a shared ectodermal origin of the brain and the skin; (2) functional systems such as micro-vascularization, innervation, eccrine excretory functions, and the extracellular matrix undergo distinct changes in patients with AD; (3) the human skin mirrors the alterations in brain stiffness observed in aging studies; (4) assessment of physiological features of the skin is cost-effective, accessible, and easily amenable for monitoring and integration with cognitive assessment studies. Understanding the relationship between aging skin and aging brain is an exciting frontier, holding great promise for improved diagnostics. Further prospective and larger-scale investigations are needed to solidify the brain-skin link and determine the extent to which this relationship can be leveraged for diagnostic applications.

ASASSN-V J205543.90+240033.5 (ASJ2055) is a possible post-common-envelope binary system. Its optical photometric data show an orbital variation of about 0.52 days and a fast period modulation of P _0 ∼ 9.77 minutes, whose origin is unknown. In this Letter, we report evidence of the stellar oscillation of the companion star as the origin of the fast period modulation. We analyze the photometric data taken by the Transiting Exoplanet Survey Satellite, the Liverpool Telescope, and the Lulin One-meter Telescope. It is found that the period of the 9.77 minutes signal measured in 2022 August is significantly shorter than that in 2021 July/August, and the magnitude of the change is of the order of ∣△ P _0 ∣/ P _0 ∼ 0.0008(4). Such a large variation will be incompatible with the scenario of the white dwarf (WD) spin as the origin of the 9.77 minutes periodic modulation. We suggest that the fast periodic signal is related to the emission from the irradiated companion star rather than that of the WD. Using existing photometric data covering a wide wavelength range, we estimate that the hot WD in ASJ2055 has a temperature of T _eff ∼ 80,000 K and is heating the oscillating M-type main-sequence star with T _eff ∼ 3500 K on its unirradiated surface. The stellar oscillation of the M-type main-sequence star has been predicted in theoretical studies, but no observational confirmation has been done. ASJ2055, therefore, has the potential to be a unique laboratory for investigating the stellar oscillation of an M-type main-sequence star and the heating effect on stellar oscillation.

We report the discovery of GRB 221009A, the highest flux gamma-ray burst (GRB) ever observed by the Fermi Gamma-ray Burst Monitor (Fermi-GBM). This GRB has continuous prompt emission lasting more than 600 s, which smoothly transitions to afterglow emission visible in the Fermi-GBM energy range (8 keV–40 MeV), and total energetics higher than any other burst in the Fermi-GBM sample. By using a variety of new and existing analysis techniques we probe the spectral and temporal evolution of GRB 221009A. We find no emission prior to the Fermi-GBM trigger time ( t _0 ; 2022 October 9 at 13:16:59.99 UTC), indicating that this is the time of prompt emission onset. The triggering pulse exhibits distinct spectral and temporal properties suggestive of the thermal, photospheric emission of shock breakout, with significant emission up to ∼15 MeV. We characterize the onset of external shock at t _0 + 600 s and find evidence of a plateau region in the early-afterglow phase, which transitions to a slope consistent with Swift-XRT afterglow measurements. We place the total energetics of GRB 221009A in context with the rest of the Fermi-GBM sample and find that this GRB has the highest total isotropic-equivalent energy ( E _γ _,iso = 1.0 × 10 ^55 erg) and second highest isotropic-equivalent luminosity ( L _γ _,iso = 9.9 × 10 ^53 erg s ^–1 ) based on its redshift of z = 0.151. These extreme energetics are what allowed us to observe the continuously emitting central engine of Fermi-GBM from the beginning of the prompt emission phase through the onset of early afterglow.

Solar photovoltaics (PV) plays an essential role in decarbonizing the European energy system. However, climate change affects surface solar radiation and will therefore directly influence future PV power generation. We use scenarios from Phase 6 of the Coupled Model Intercomparison Project (CMIP6) for a mitigation (SSP1-2.6) and a fossil-fuel-dependent (SSP5-8.5) pathway in order to quantify climate risk for solar PV in Europe as simulated by the Global Solar Energy Estimator (GSEE). We find that PV potential increases by around 5 % in the mitigation scenario, suggesting a positive feedback loop between climate change mitigation and PV potential. While increased clear-sky radiation and reduced cloud cover go hand in hand in SSP1-2.6, the effect of a decrease in clear-sky radiation is outweighed by a decrease in cloud cover in SSP5-8.5, resulting in an increase in all-sky radiation. Moreover, we find that the seasonal cycle of PV generation changes in most places, as generation grows more strongly in winter than in summer (SSP1-2.6) or increases in summer and declines in winter (SSP5-8.5). We further analyze climate change impacts on the spatial variability of PV power generation. Similar to the effects anticipated for wind energy, we report an increase in the spatial correlations of daily PV production with large inter-model agreement yet relatively small amplitude, implying that PV power balancing between different regions in continental Europe will become more difficult in the future. Thus, based on the most recent climate simulations, this research supports the notion that climate change will only marginally impact renewable energy potential, while changes in the spatiotemporal generation structure are to be expected and should be included in power system design.

Abstract The focal plane camera is the core component of the Wide Field-of-view Cherenkov/fluorescence Telescope Array (WFCTA) of the Large High-Altitude Air Shower Observatory (LHAASO). Because of the capability of working under moonlight without aging, silicon photomultipliers (SiPM) have been proven to be not only an alternative but also an improvement to conventional photomultiplier tubes (PMT) in this application. Eighteen SiPM-based cameras with square light funnels have been built for WFCTA. The telescopes have collected more than 100 million cosmic ray events and preliminary results indicate that these cameras are capable of working under moonlight. The characteristics of the light funnels and SiPMs pose challenges (e.g. dynamic range, dark count rate, assembly techniques). In this paper, we present the design features, manufacturing techniques and performances of these cameras. Finally, the test facilities, the test methods and results of SiPMs in the cameras are reported here.

Imaging polarimetry can obtain two-dimensional intensity distribution information and its corresponding states of polarization from the target simultaneously. Sagnac polarization imaging interferometer is one of the typical channeled polarimetry technologies. By splitting the incident light through the triangular optical structure, the polarization information of the target is modulated into the interference fringes and can be obtained by the demodulation algorithm. The non-ideal optical elements in the system will cause reconstruction errors. This article analyses several factors that affect the measurement accuracy, including the splitting ratio of polarization beam splitter and the diffraction efficiency of the blazed grating. We derive the modified intensity formula and obtain interference patterns through numerical simulations. By calculating the modulation degree of interference fringes and reconstruction error under different non-ideal parameters, we have come to the conclusion that to meet the requirement of modulation degree > 0.5 and reconstruction error s/ηp > 0.83, and the first-order diffraction efficiency should be η1 > 79%.

The localizations of sodium exclusion are roots and stem base, however, Na+ distribution in these localizations is unclear. Here, we used a salt-tolerant buckwheat cultivar Chuanqiao No.1 and a salt-sensitive cultivar TQ-0808 to demonstrate Na+ distribution. We found that Na+ content was highest in vacuole, the following was in cell wall or free space, and the least was in cytoplasm. Comparative analysis shows that Na+ accumulation in vacuole, cell wall, or free space of roots and stem base in 'Chuanqiao No.1' was obviously higher than in 'TQ-0808'; in contrast, Na+ accumulation in cytoplasm of 'Chuanqiao No.1' was less than in 'TQ-0808'. These results indicate that the capabilities of Na+ extrusion and Na+ compartmentalization of salt-tolerant buckwheat were obviously higher than of the salt-sensitive one, and the capabilities could effectively restrict Na+ transport to shoot. Compartmentalization of Na+ in the vacuole was the main way for Na+ exclusion of salt-tolerant buckwheat. In addition, the transcriptions of Na+/H+ antiporter 1 and salt overly sensitive 1 were remarkably higher in 'Chuanqiao No.1' than in 'TQ-0808', which is consistent with the above results.

The dominant emission sources of anthropogenic radionuclides come from either atmospheric nuclear weapons tests or the nuclear industry (i.e., reprocessing plants or reactor accidents). Here, the authors identify a new environmental isotope tracer ($$^{233}$$ 233 U/$$^{236}$$ 236 U) which can help distinguish emissions from nuclear weapons tests, and can also provide constraints on past weapon designs and fuel sources, for which many details remain classified or lost.

Aerosol iodine isotopes are pivotal links in atmospheric circulation of iodine in both atmospheric and nuclear sciences, while their sources, temporal change and transport mechanism are still not well understood. This work presents the day-resolution temporal variation in iodine-129 (129I) and iodine-127 (127I) concentrations in aerosols from Xi'an, north-west China, during 2017/18. Both iodine isotopes have significant fluctuations with time, showing the highest levels in winter, approximately 2–3 times higher than in other seasons, but the correlation between 129I and 127I concentrations reflects that they have different sources. Aerosol 127I concentrations are found to be noticeably positively correlated with air quality index and five air pollutants. Enhanced fossil fuel combustion and inverse weather conditions can explain the increased concentrations and peaks of 127I in winter. The change in 129I concentrations confirms that the source and level of 129I in the monsoonal region were alternatively dominated by the 129I-enriched East Asian winter monsoon and the 129I-poor East Asian summer monsoon. The mean 129I∕127I number ratio of (92.7±124)×10-10 provides an atmospheric background level for the purpose of nuclear environmental safety monitoring. This study suggests that locally discharged stable127I and externally input 129I are likely involved in fine particles formation in urban air, which provides insights into the long-range transport of air pollutants and iodine's role in particulate formation in urban atmosphere.

Muscle fiber characteristics comprise a set of complex traits that influence the mea quality and lean meat production of livestock. However, the genetic and biological mechanisms regulating muscle fiber characteristics are largely unknown in pigs. Based on a genome-wide association study (GWAS) performed on 421 Large White × Min pig F2 individuals presenting well-characterized phenotypes, this work aimed to detect genome variations and candidate genes for five muscle fiber characteristics: percentage of type I fibers (FIB1P), percentage of type IIA fibers (FIB2AP), percentage of type IIB fibers (FIB2BP), diameter of muscle fibers (DIAMF) and number of muscle fibers per unit area (NUMMF). The GWAS used the Illumina Porcine SNP60K genotypic data, which were analyzed by a mixed model. Seven and 10 single nucleotide polymorphisms (SNPs) were significantly associated with DIAMF and NUMMF, respectively (P < 1.10E-06); no SNP was significantly associated with FIB1P, FIB2AP or FIB2B. For DIAMF, the significant SNPs on chromosome 4 were located in the previously reported quantitative trait loci (QTL) interval. Because the significant SNPs on chromosome 6 were not mapped in the previously reported QTL interval, a putative novel QTL was suggested for this locus. None of the previously reported QTL intervals on chromosomes 6 and 14 harbored significant SNPs for NUMMF; thus, new potential QTLs on these two chromosomes are suggested in the present work. The most significant SNPs associated with DIAMF (ALGA0025682) and NUMMF (MARC0046984) explained 12.02% and 11.59% of the phenotypic variation of these traits, respectively. In addition, both SNPs were validated as associated with DIAMF and NUMMF in Beijing Black pigs (P < 0.01). Some candidate genes or non-coding RNAs, such as solute carrier family 44 member 5 and miR-124a-1 for DIAMF, and coiled-coil serine rich protein 2 for NUMMF, were identified based on their close location to the significant SNPs. This study revealed some genome-wide association variants for muscle fiber characteristics, and it provides valuable information to discover the genetic mechanisms controlling these traits in pigs.

Cancer stem cells (CSC) have been shown as the origin for therapeutic resistance and patient relapse. Here, the authors show that targeting Arf1-mediated lipid metabolism in CSC induces cell death but also an immunogenic anti-cancer response.

We present 294 pulsars found in GeV data from the Large Area Telescope (LAT) on the Fermi Gamma-ray Space Telescope. Another 33 millisecond pulsars (MSPs) discovered in deep radio searches of LAT sources will likely reveal pulsations once phase-connected rotation ephemerides are achieved. A further dozen optical and/or X-ray binary systems colocated with LAT sources also likely harbor gamma-ray MSPs. This catalog thus reports roughly 340 gamma-ray pulsars and candidates, 10% of all known pulsars, compared to ≤11 known before Fermi. Half of the gamma-ray pulsars are young. Of these, the half that are undetected in radio have a broader Galactic latitude distribution than the young radio-loud pulsars. The others are MSPs, with six undetected in radio. Overall, ≥236 are bright enough above 50 MeV to fit the pulse profile, the energy spectrum, or both. For the common two-peaked profiles, the gamma-ray peak closest to the magnetic pole crossing generally has a softer spectrum. The spectral energy distributions tend to narrow as the spindown power $\dot{E}$ decreases to its observed minimum near 10 ^33 erg s ^−1 , approaching the shape for synchrotron radiation from monoenergetic electrons. We calculate gamma-ray luminosities when distances are available. Our all-sky gamma-ray sensitivity map is useful for population syntheses. The electronic catalog version provides gamma-ray pulsar ephemerides, properties, and fit results to guide and be compared with modeling results.

Binary systems are a well-established subclass of gamma-ray sources. The high mass X-ray binary pulsar 1A 0535+262 has been considered to be a possible gamma-ray emitter for a long time, although former gamma-ray searches using the Fermi Large Area Telescope (LAT) and VERITAS data resulted in upper limits only. We aim at a deep search for gamma-ray emission and pulsations from 1A 0535+262 using more than 13 yr of Fermi-LAT data. The analysis was performed for both the whole Fermi-LAT data set, as well as for the X-ray outbursts that 1A 0535+262 has experienced since the launch of Fermi. Various X‐ray observations have been used to generate the ephemeris for the pulsation search. We also investigate the long-term gamma-ray flux variability and perform orbital phase-resolved analysis for the outbursts. We did not detect any steady or pulsed gamma-ray emission from 1A 0535+262 during the whole Fermi-LAT mission span or its X-ray outbursts. We thus derived the deepest gamma-ray luminosity upper limits to date at the 95% confidence level to be around (2.3–4.7) × 10 ^32 erg s ^−1 depending on different spectral indices assumed, which results in a ratio of L _γ to L _X (2–150 keV) being (1.9–3.9) × 10 ^−6 .

The Fermi Large Area Telescope (LAT) lightcurve repository (LCR) is a publicly available, continually updated library of gamma-ray lightcurves of variable Fermi-LAT sources generated over multiple timescales. The Fermi-LAT LCR aims to provide publication-quality lightcurves binned on timescales of 3, 7, and 30 days for 1525 sources deemed variable in the source catalog of the first 10 yr of Fermi-LAT observations. The repository consists of lightcurves generated through full likelihood analyses that model the sources and the surrounding region, providing fluxes and photon indices for each time bin. The LCR is intended as a resource for the time-domain and multimessenger communities by allowing users to search LAT data quickly to identify correlated variability and flaring emission episodes from gamma-ray sources. We describe the sample selection and analysis employed by the LCR and provide an overview of the associated data access portal.

Abstract Elevated Wnt/β‐catenin signaling has been commonly associated with tumorigenesis especially colorectal cancer (CRC). Here, an MST4‐pβ‐cateninThr40 signaling axis essential for intestinal stem cell (ISC) homeostasis and CRC development is uncovered. In response to Wnt3a stimulation, the kinase MST4 directly phosphorylates β‐catenin at Thr40 to block its Ser33 phosphorylation by GSK3β. Thus, MST4 mediates an active process that prevents β‐catenin from binding to and being degraded by β‐TrCP, leading to accumulation and full activation of β‐catenin. Depletion of MST4 causes loss of ISCs and inhibits CRC growth. Mice bearing either MST4T178E mutation with constitutive kinase activity or β‐cateninT40D mutation mimicking MST4‐mediated phosphorylation show overly increased ISCs/CSCs and exacerbates CRC. Furthermore, the MST4‐pβ‐cateninThr40 axis is upregulated and correlated with poor prognosis of human CRC. Collectively, this work establishes a previously undefined machinery for β‐catenin activation, and further reveals its function in stem cell and tumor biology, opening new opportunities for targeted therapy of CRC.

It has been hypothesized that xylanase inhibitors play important roles in plant defense against microbial pathogens. Currently, there is little information available about xylanase inhibitor OsXIP in rice and its gene expression. We cloned a xylanase inhibitor gene OsXIP from rice (Oryza sativa L. cv. Nipponbare) genomic DNA. To determine the function of OsXIP, we generated OsXIP-overexpressing transgenic rice plants. The transgenic plants had significantly higher OsXIP expression and showed enhanced defense response to Magnaporthe oryzae compared to the wild-type plants. The results also showed that the increased OsXIP expression was accompanied by the up-regulation of pathogenesisrelated genes. To clarify the OsXIP expression pattern, a ProOsXIP::GUS vector was constructed and transgenic plants were obtained. GUS staining results revealed that OsXIP showed organ-specific expressions in rice plants. OsXIP was primarily expressed in the roots and in the veins, but it was weakly expressed in the leaves. Analyses of the OsXIP expression in response to biotic and abiotic stresses indicated that it was drastically induced by biotic stresses and methyl jasmonate treatment. OsXIP, a member of a new class of antifungal proteins, may function as a barrier that prevents the cell wall degradation by xylanases excreted by fungal pathogens. The OsXIP was found to be a stressresponsive gene and it could take part in plant defense via a JA-mediated signaling pathway.

The combination of complex, multiple minerogenic stages and mineral superposition during geological processes has resulted in dynamic spatial distributions and nonstationarity of geological variables. For example, geochemical elements exhibit clear spatial variability and trends with coverage type changes. Thus, bias is likely to occur under these conditions when general regression models are applied to mineral prospectivity mapping (MPM). In this study, we used a spatially weighted technique to improve general logistic regression and developed an improved model, i.e., the improved logistic regression model, based on a spatially weighted technique (ILRBSWT, version 1.0). The capabilities and advantages of ILRBSWT are as follows: (1) it is a geographically weighted regression (GWR) model, and thus it has all advantages of GWR when managing spatial trends and nonstationarity; (2) while the current software employed for GWR mainly applies linear regression, ILRBSWT is based on logistic regression, which is more suitable for MPM because mineralization is a binary event; (3) a missing data processing method borrowed from weights of evidence is included in ILRBSWT to extend its adaptability when managing multisource data; and (4) in addition to geographical distance, the differences in data quality or exploration level can be weighted in the new model.

An enhancement of the tropospheric ozone column (TOC) over Arabian Sea (AS) during the pre-monsoon season is reported in this study. The potential sources of the AS spring ozone pool are investigated by use of multiple data sets (e.g., SCIAMACHY Limb-Nadir-Matching TOC, OMI/MLS TOC, TES TOC, MACC reanalysis data, MOZART-4 model and HYSPLIT model). Three-quarters of the enhanced ozone concentrations are attributed to the 0–8 km height range. The main source of the ozone enhancement is considered to be caused by long-range transport of ozone pollutants from India (∼ 50 % contributions to the lowest 4 km, ∼ 20 % contributions to the 4–8 km height range), the Middle East, Africa and Europe (∼ 30 % in total). In addition, the vertical pollution accumulation in the lower troposphere, especially at 4–8 km, was found to be important for the AS spring ozone pool formation. Local photochemistry, on the other hand, plays a negligible role in producing ozone at the 4–8 km height range. In the 0–4 km height range, ozone is quickly removed by wet deposition. The AS spring TOC maxima are influenced by the dynamical variations caused by the sea surface temperature (SST) anomaly during the El Niño period in 2005 and 2010 with a ∼ 5 DU decrease.

Both intrinsic and external signals regulate stem cell fate. Here, the authors perform a genome-wide RNAi screen to identify regulators of testis germline stem cells (GSC) in Drosophila, finding more than half of the genes were external regulators and heterochromatin/cytokinesis proteins mediated GSC fate.

Speciation analysis of iodine in aerosols is a very useful approach for understanding geochemical cycling of iodine in the atmosphere. In this study, overall iodine species, including water-soluble iodine species (iodide, iodate and water-soluble organic iodine), NaOH-soluble iodine, and insoluble iodine have been determined for 129I and 127I in the aerosols collected at Risø, Denmark, during March and May 2011 (shortly after the Fukushima nuclear accident) and in December 2014. The measured concentrations of total iodine are in the range of 1.04–2.48 ng m−3 for 127I and (11.3–97.0) × 105 atoms m−3 for 129I, corresponding to 129I ∕ 127I atomic ratios of (17.8–86.8) × 10−8. The contribution of Fukushima-derived 129I (peak value of 6.3 × 104 atoms m−3) is estimated to be negligible (less than 6 %) compared to the total 129I concentration in northern Europe. The concentrations and species of 129I and 127I in the aerosols are found to be strongly related to their sources and atmospheric pathways. Aerosols that were transported over the contaminated seas contained higher concentrations of 129I than aerosols transported over the European continent. The high 129I concentrations of the marine aerosols are attributed to secondary emission of marine discharged 129I in the contaminated seawater in the North Sea, North Atlantic Ocean, English Channel, Kattegat, etc., rather than direct gaseous release from the European nuclear reprocessing plants (NRPs). Water-soluble iodine was found to be a minor fraction to the total iodine for both 127I (7.8–13.7 %) and 129I (6.5–14.1 %) in ocean-derived aerosols, but accounted for 20.2–30.3 % for 127I and 25.6–29.5 % for 129I in land-derived aerosols. Iodide was the predominant form of water-soluble iodine, accounting for more than 97 % of the water-soluble iodine. NaOH-soluble iodine seems to be independent of the sources of aerosols. The significant proportion of 129I and 127I found in NaOH-soluble fractions is likely bound with organic substances. In contrast to water-soluble iodine, the sources of air masses exerted distinct influences on insoluble iodine for both 129I and 127I, with higher values for marine air masses and lower values for terrestrial air masses.
