The latency-to-lie (LTL) test is an objective method for assessing walking ability of broiler chickens which has traditionally consisted of placing a bird into a tub of shallow water and measuring how long it takes the standing bird to sit, with duration being negatively associated with gait score. Unfortunately, this method is impractical for use on commercial farms. Thus, the aim of this study was to assess the performance of LTL tests without the addition of water, making them more practical for use on farm. In total, 159 Ross 308 and 186 Rustic Gold broilers were assessed. Following receiving a gait score (Bristol scale), birds were placed individually into a litter lined clear plastic storage box and the LTL was conducted with a maximum test duration of 300 s. Following a 120 s period of rest, the bird was then placed on the litter near a group of flockmates, and the LTL test was repeated. This allowed for the assessment of whether containment of the bird was necessary. Latencies to lie were negatively correlated with gait score (With box: ρ=-0.44 , P < 0.001; Without box: ρ=-0.46 , P < 0.001). The latencies to lie (mean ± SD) when using a box were 129.0 ± 82.0, 114.0 ± 78.8, 71.9 ± 54.9, 45.8 ± 35.8, and 7.9 ± 14.0 s and without a box were 104.0 ± 97.2, 52.2 ± 53.8, 27.9 ± 29.9, 22.8 ± 27.0, and 14.0 ± 19.4 s for gait scores 0, 1, 2, 3, and 4, respectively. There was no effect of hybrid on the performance of either of the tests. Results suggest that the LTL tests without water could serve as an objective and valid measure of walking ability on farm, with the LTL test with a box showing better ability at distinguishing between specific gait scores compared to the LTL test without a box, though possible impacts of fatigue due to study design should be considered., Competing Interests: Declaration of competing interest The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (Copyright © 2024. Poultry are deprived of water when transported to slaughter, beginning shortly prior to catching of the first bird and lasting through catching and loading, the journey on the vehicle, time spent in lairage, and up until time of death. Our aim was to review existing knowledge on variables which may be useful in determining the length of time that poultry may go without water in connection with transport before their welfare begins to deteriorate. During transport, it is likely that birds experience a motivation to drink, which may transition into the negative emotional state of thirst if water is unavailable. Determining when drinking motivation reaches a threshold where welfare is negatively impacted is challenging. In the absence of water, birds may over time experience dehydration which may be detected through physiological indicators as their body attempts to maintain homeostasis. In poultry, plasma osmolality, arginine vasotocin, and chloride have been suggested as being most suitable for assessing dehydration resulting from periods of water deprivation that correspond with typical transport durations, due to their particular sensitivity during this period. While initial dehydration may not be associated with negative emotional states, it is likely that it eventually leads to discomfort, but additional behavioral and motivational studies are necessary to infer when this begins. Impacts of thermal conditions, genetics, and the condition of the individual bird on the development of a dehydrated state were also assessed, though more information is needed to fully understand these interactions. With the available literature, this review concludes that total transport (i.e., from the initial deprivation from water until time of slaughter) durations of longer than 6 h are likely associated with measurable physiological indicators of dehydration and may potentially be associated with negative emotional states, although more research is needed to clarify this. Outdoor range areas provide laying hens with improved opportunities to perform natural behaviors and increase the available space per bird, however, birds are also exposed to potentially stressful factors including weather and predators. Ability to cope with challenging environments varies between different strains and must be considered to ensure good welfare. The aim of this study was to determine how suitable 2 hybrids, the Dekalb White (DW) and the Bovans Brown (BB), are for organic production with special emphasis on ranging behavior. A total of 1,200 hens were housed according to organic regulations across 12 flocks of 100 birds. Range and shelter use, effect of weather, vegetation cover, egg production and quality, and mortality were assessed in addition to a range of clinical welfare indicators. Initially a greater proportion of DW hens accessed the range. However, after approximately 2 mo, a greater proportion of BB were using the range and venturing further from the house. DW hens were more likely to use the shelters than BB hens (P < 0.001). Vegetation was also worn away to a greater extent in the BB ranges. Weather affected the proportion of hens that went outside, the distance ranged from the popholes, and shelter use. BB hens were found to have better plumage condition (P < 0.001), fewer footpad lesions (P < 0.001), fewer comb wounds (P < 0.001), and lower mortality rates (P = 0.013). Both hybrids experienced keel bone fractures, though DW hens had more at the cranial portion (P < 0.001) and BB at the caudal portion (P < 0.001). DW hens had an earlier onset of lay and higher egg production than BB hens (P < 0.001), though BB hens laid heavier eggs (P < 0.001) with thicker shells (P = 0.001). Overall, BB hens seemed to perform superiorly or equivalently to the DW hens for all variables apart from egg production. These results demonstrate the importance of considering the strain of bird selected for organic production systems in order for the birds to reap the potential benefits that are offered by outdoor access., (Copyright © 2022 The Authors. Large-scale phenotyping of animal behaviour traits is time consuming and has led to increased demand for technologies that can automate these procedures. Automated tracking of animals has been successful in controlled laboratory settings, but recording from animals in large groups in highly variable farm settings presents challenges. The aim of this review is to provide a systematic overview of the advances that have occurred in automated, high throughput image detection of farm animal behavioural traits with welfare and production implications. Peer-reviewed publications written in English were reviewed systematically following Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. After identification, screening, and assessment for eligibility, 108 publications met these specifications and were included for qualitative synthesis. Data collected from the papers included camera specifications, housing conditions, group size, algorithm details, procedures, and results. Most studies utilized standard digital colour video cameras for data collection, with increasing use of 3D cameras in papers published after 2013. Papers including pigs (across production stages) were the most common (n = 63). The most common behaviours recorded included activity level, area occupancy, aggression, gait scores, resource use, and posture. Our review revealed many overlaps in methods applied to analysing behaviour, and most studies started from scratch instead of building upon previous work. Training and validation sample sizes were generally small (mean±s.d. groups = 3.8±5.8) and in data collection and testing took place in relatively controlled environments. To advance our ability to automatically phenotype behaviour, future research should build upon existing knowledge and validate technology under commercial settings and publications should explicitly describe recording conditions in detail to allow studies to be reproduced., Competing Interests: The authors have declared that no competing interests exist.

Hydroxytyrosol, an important polyphenolic compound found in olive oil, has shown anti-tumor activity both in vitro and in vivo. However, effects of hydroxytyrosol on prostate cancer are largely unkown. We found that hydroxytyrosol preferentially reduces the viability of human prostate cancer cells (PC-3, DU145) compared to an immortalized non-malignant prostate epithelial cell line (RWPE-1). Exposure of PC-3 cells to 80 µmol/L hydroxytyrosol resulted in significant increases in both superoxide production and activation of apoptosis. These increases were accompanied by mitochondrial dysfunction, defects in autophagy, and activation of MAP kinases. Moreover, N-acetylcysteine (NAC), an efficient reactive oxygen species (ROS) scavenger, was able to reverse the hydroxytyrosol-induced effects of cell viability loss, defects in autophagy, and activation of apoptosis. This evidence suggests that ROS play a vital role in the loss of PC-3 cell viability. However, MAPK inhibitors including U0126 (for Erk1/2), SB203580 (for p38MAPK) and SP600125 (for JNK) did not decrease hydroxytyrosol-induced growth inhibition, suggesting that these kinases may not be required for the growth inhibitory effect of hydroxytyrosol. Moreover, addition of ROS scavengers (i.e. NAC, catalase, pyruvate, SOD) in the growth media can prevent hydroxytyrosol induced cell viability loss, suggesting that extracellular ROS (superoxide and hydrogen peroxide) facilitate the anti-proliferation effect of hydroxytyrosol in prostate cancer cells. The present work firstly shows that hydroxytyrosol induces apoptotic cell death and mitochondrial dysfunction by generating superoxide in PC-3 cells. This research presents preliminary evidence on the in vitro chemopreventive effect of hydroxytyrosol, and will contribute to further investigation of hydroxytyrosol as an anti-cancer agent.

The synthesis, electrochemical, optical, and cation-sensing properties of [2.2]ferrocenophanes, in which the two ferrocene subunits are linked through two aldiminic or iminophosphorane moieties, are reported. The new compounds show remarkably selective cation-sensing properties due to the presence of redox-active units (ferrocene) and aza-unsaturated functionalities that are able to act as putative cation-binding sites. In this structural motif, the aldimine groups act as a highly selective binding site for Zn(2+) cations, whereas the iminophosphorane bridges display an unusually strong binding affinity towards Li(+) cations, which could be explained by an additional LiFe interaction. The X-ray structure of the complex 4Li(+) as well as detailed NMR spectroscopic studies, both in solution and in the solid state, support this assessment. Experimental data and conclusions about the cation-sensing capabilities of this family of compounds are supported by DFT calculations.
