Q fever, also known as query fever, is a zoonotic illness brought on by the Coxiella burnetii bacteria. This disease was first discovered in 1935 in Queensland, Australia. Worldwide, Q fever is a disease that requires notification, and certain nations classify it as a national health concern. A feature of C. burnetii is known as cell wall phase fluctuation. Serological testing is the main method used to diagnose Q fever illnesses. Inhalation is the primary method of C. burnetii transmission in both people and animals, with smaller amounts occurring through milk and milk product ingestion. The bacterial strain that is causing the infection determines how severe it is. Q fever is a significant zoonosis that can be dangerous for personnel working in veterinary laboratories, livestock breeding operations, and slaughterhouses due to its high human contagiousness. C. burnetii is a biological weapon that can be sprayed on food, water, or even mail. It can also be employed as an aerosol. Antibiotics work well against this disease's acute form, but as the infection develops into a chronic form, treatment becomes more difficult and the illness frequently returns, which can result in a high death rate. Vaccination has been demonstrated to lower the incidence of animal infections, C. burnetii shedding, and abortion. Several hygienic precautions should be put in place during an outbreak to lessen the spread of disease to animals. [Open Vet J 2024; 14(10.000): 2509-2524]

6. Effect of PG-600 dose in cows with persistent corpus luteum treated with PGF2α and hCG

The purpose of this study was to determine the effect of administering a dose of gonadotropin (PG-600) in cows with persistent corpus luteum (PCL) which were treated with prostaglandin F2 α (PGF2α) and human chorionic gonadotropin (hCG). This study used 20, three-year-old cows diagnosed with PCL, had recently given birth, and were not pregnant. Cows were randomly allocated into groups T0, T1, T2 and T3. Cows in all groups were injected with PGF2α (25 mg/cow). Simultaneously, cows in groups T1, T2 and T3 were also injected with PG-600 at doses of 100, 150 and 200 IU/cow respectively. PGF2α and PG-600 were injected when PCL was identified. After that, cows showing signs of estrus were injected with hCG (100 IU/cow) and artificially inseminated 12-18 hours after the appearance of signs of estrus. AI was conducted using frozen thawed semen. Sixty days following AI, a pregnancy examination was conducted using USG. Estrus was detected in all treated cows, while the pregnancy rates varied. Compared with other groups, the groups given 100 and 150 IU PG-600 produced higher pregnancy rates. It could be concluded that the combination of 25 mg PGF2α, 150 IU PG-600, and 100 IU hCG was the most effective treatment to produce high pregnancy rates in cows with persistent corpus luteum.

This study aims to determine the effect of oral administration of white guava fruit juice (Psidium guajava L.) on the spermatogenic cells of rats (Rattus norvegicus) exposed orally to lead acetate. Twenty-five male rats (Rattus norvegicus) of the Wistar strain were randomly divided into five groups. Placebo group rats (P0) were given 0.5 mL distilled water twice every day orally at four-hour intervals. Groups T0, T1, T2, and T3 were given lead acetate 50 mg/kg bw orally, then respectively given distilled water, 100, 50 and 25% (v/v) white guava (Psidium guajava L.) fruit juice orally four hours later for 14 days. On day-15, the rats were sacrificed and their testicles were taken for histological preparations. Data were analyzed using Analysis of Variance followed by Duncan's Multiple Range Test. The results showed that the number of spermatogonia, spermatocytes and spermatids cells of rats in T0 group was lower (p 0.05) as in the P0 group. This study found that white guava pure juice without dilution was the best for maintaining the number of spermatids, but was unable to restore the number of spermatogonia and spermatocyte cells in rats exposed to lead acetate.

This study aims to determine the effect of the administration of white guava (Psidium guajava L.) fruit juice on spermatozoa plasma membrane integrity (PMI), morphological abnormality, viability, and motility of lead acetate induced rats (Rattus norvegicus). Twenty-five male rats were divided into five groups: NC (negative control) group, rats were administered with distilled water twice daily at four-hour intervals; T0 (positive control) group, rats were administered daily with lead acetate 50 mg/kg bw and distilled water four hours later; T1, T2, and T3 groups, rats were administered daily with lead acetate 50 mg/kg bw and 0.5 mL of 25, 50, and 100% white guava fruit juice four hours later. The treatment of the rats was conducted for 14 days, and on day 15, all rats were sacrificed to assess the spermatozoa quality. Data was analyzed using ANOVA followed by Duncan's multiple range test at a confidence level of 95%. This study aimed to determine the effect of the administration of ethanolic extract of Moringa oleifera leaves on the number of spermatogenic and Leydig cells of gentamicin-induced rats (Rattus norvegicus). This study used 25 white male rats divided randomly into five groups. The rats were injected with Gentamicin 5 mg/kg bw/day subcutaneously for 14 days, except for the negative control group (NCG). During the next 14 days, the rats were given oral moringa leaf extract of 200, 316, and 500 mg/kg bw/day respectively for E200, E316, and E500 groups. The data were analyzed with one-way ANOVA followed by Duncan's test. The results showed that the number of spermatogonia, spermatocytes, and spermatids in NCG was higher (p 0.05) to the E500 group. The number of PCG Leydig cells was the smallest (p 0.05) from that of the NCG. It could be concluded that administration of Moringa oleifera leaves extract restored the number of spermatogenic and Leydig cells of gentamicin-induced rats.

This study aimed to evaluate the profile of progesterone in dairy cattle with ovarian hypofunction. A total of 10 cows in this study were evaluated three times in the collection phase, i.e. (F1) when the cow was diagnosed with ovarian hypofunction, (F2) when the cow was in heat and (F3) 21 days after artificial insemination to detect pregnancy. Samples in the form of blood serum were then analyzed using ELISA. As a result, the average time of heat in ovarian hypofunction cows was 7,4 days. Progesterone levels in F1 were 1,027 ng/ml, 2,774 ng/ml, 1,476 ng/ml, 2,256 ng/ml, 1,258 ng/ml, 1,758 ng/ml, 2,393 ng/ml, 0,592 ng/ml, 0,755 ng /ml, 1,876 ng/ml, respectively. Progesterone levels in F2 were 0,671 ng/ml, 0,517 ng/ml, 0,763 ng/ml, 0,598 ng/ml, 0,615 ng/ml, 0,537 ng/ml, 0,726 ng/ml, 0,643 ng/ml, 0,593 ng/ml, 0,975ng/ml, respectively. Progesterone levels in F3 were 15,642 ng/ml, 4,215 ng/ml, 17,327 ng/ml, 20,721 ng/ml, 5,796 ng/ml, 17,214 ng/ml, 15,815 ng/ml, 16,745 ng/ml, 4,632 ng /ml, 18,281 ng/ml, respectively. The pregnancy rate in hypofunctional cows treated with PG-600 in this study was 70%.

This study aimed to determine the effect of the addition of cherry tomato, california papaya, and honey melon juice into skim milk extender in maintaining the quality of Sapudi ram semen at chilled temperature (5oC) storage. Five replication of ejaculates were divided equally into four groups. In control group (T0) semen was diluted in skim milk extender, while in T1, T2, and T3 groups semen were diluted in skim milk extender contained 20% of cherry tomato, california papaya, and honey melon juice, respectively. The extended semen was stored at a chilled temperature, and semen quality (based on sperm motility, viability, and plasma membrane integrity) was observed daily for five days. The result showed that semen quality declined day by day during the five days of storage. Based on the minimum standard of post-thawed semen motility (40%), the spermatozoa of the control group only lasted by the second day. This research aims to determine the effect of Moringa oleifera leaf extract (MLE) on the diameter and epithelium thickness of the seminiferous tubules of rats induced with gentamicin. This study used 25 male rats (Rattus norvegicus) aged 8-12 weeks with 150-200 grams body weight. Negative control group (C-) was injected with aquadest and given 0.5% Na-CMC orally, positive control group (C+) was induced with gentamicin injection of 5 mg/kg BW/day and given 0.5% Na-CMC orally, while groups T1, T2, and T3 were induced with gentamicin at the same dose and treated orally with MLE of 200, 316, 500 mg/kg BW/day. Analysis of variance showed significant differences and further analysis using Duncan Multiple Range Test showed difference in each group. The results showed that treatment with MLE of 500 mg/kg BW/day improved the diameter and epithelium thickness of the seminiferous tubules of rats injected with gentamicin. It could be concluded that MLE restored the diameter and epithelium thickness of the seminiferous tubules of rats induced with gentamicin.

Background: Migraine is a primary headache, usually described as pain attacks lasting 4 - 72 hours, unilateral, throbbing, with moderate to severe pain intensity that can be exacerbated by activity, and can be accompanied by nausea and vomiting, photophobia and phonophobia. Migraine can interfere with patient activities, so that it can reduce productivity and result in an economic burden to the family and reduce the patient's quality of life. To handle it, pharmacological and non-pharmacological therapies can be used. Acupuncture therapy is one of the non-pharmacological therapeutic options that can be used in migraines. Purpose: This article review collects and analyzes journals related to the effect of acupuncture therapy on migraines. The journal search method was carried out electronically from December to April 2021 using several databases such as Pubmed, Sagepub and Google Schoolar with the keywords: acupuncture in cases of migraine, acupuncture for migraine, and acupuncture point for migraine. Review: The journal searches discovered that acupuncture therapy had an effect in reducing pain intensity in migraine cases with the most widely used method that was the manual acupuncture method. However, the most influential method was the combination method of manual acupuncture, herbal acupuncture, and nutritional therapy with the hyperactivity of Yang liver as the frequently occurring syndrome. The frequently used points were the acupuncture points of Fengchi-GB20, Hegu-LI4, Taichong-LR3, Baihui-DU20, Zusanli-ST36, Sanyinjiao-SP6, and Taiyang-EX-HN5.Conclusion: Based on the journal analyses, the combination method of manual acupuncture, herbal acupuncture, and nutritional therapy can reduce pain intensity in migraine cases

Background and Aim: 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is a persistent organic pollutant toxic to the human reproductive system. This study aimed to evaluate the effect of α-Tocopherol administration on the male fertility parameters of a rat model exposed to TCDD. Materials and Methods: Fifty healthy 12-week-old male rats were randomly divided into five groups. Rats in the control group were given corn oil twice daily in 4 h intervals. In the treatment groups, all rats were given TCDD at a dose of 700 ng/kg of body weight (BW)/day for 45 days. Four hours after receiving the TCDD, T0 rats were given corn oil, and T1, T2, and T3 rats were given α-Tocopherol at doses of 77, 140, and 259 mg/kg BW/day, respectively, for 45 days. On day 46, experimental animals were sacrificed to collect blood and testicular samples. Results: TCDD exposure decreased superoxide dismutase activity, plasma membrane integrity, Leydig cell count, sperm cell count, sperm viability and motility, and increased malondialdehyde levels, serum testosterone levels, and sperm morphological abnormalities. The administration of α-Tocopherol mitigated the effects of TCDD exposure, and the 140 and 259 mg/kg BW/day treatments returned those male fertility parameters to normal levels. Conclusion: The administration of 140 mg/kg BW/day α-Tocopherol restored male semen quality in rats exposed to TCDD. We found dynamics serum testosterone levels in rats exposed to TCDD that need to be further studied.

Background and Aim: The production of male calf beef cattle is an agricultural innovation needed to increase the farm's productivity as a provider of meat sources. This study aimed to determine the sex ratio of the offspring of cows inseminated with Y-bearing sperm enriched by Percoll density gradient centrifugation and swim up, combined with delayed fixed-time artificial insemination (FTAI). Materials and Methods: Ejaculates of Simmental bulls were divided into four equal portions and grouped as T0 (control, non-sexed semen), T1 and T2 were sexed semen using Percoll density gradient centrifugation three and five levels, respectively, and T3 was sexed semen using swim-up. After the sex was sorted, the semen was diluted in a tris egg yolk extender, packaged in French mini-straws containing 50 million live sperm cells, and frozen. Pre-sexed, post-sexed, and post-thawed spermatozoa were evaluated based on progressive motility, viability, intact plasma membrane, and abnormality. The post-thawed semen of T0 was artificially inseminated to recipient cows at 12 h after onset of estrus (not delayed FTAI). Meanwhile, the delayed FTAI was conducted 18 20 h after onset of estrus using the T0, the best of T1 and T2, and the T3 post-thawed semen. Results: The Percoll density gradient centrifugation reduced motility, viability, and intact plasma membrane but increased sperm abnormalities. Meanwhile, the swim up process increased motility, viability, and intact plasma membrane of sperm cells but decreased sperm abnormalities. Post-thawed semen decreased motility, viability, and intact plasma membrane of sperm cells but increased sperm abnormalities. The sex ratio of the Simmental crossbred offspring was 96.08% and 100% in T1 and T3, respectively, compared to 48.25% and 67.39% in T0 not delayed and delayed FTAI, respectively. Conclusion: The Percoll density gradient centrifugation and swim-up methods are prospective for obtaining male offspring.

The purpose of this research was to determine the best dosage of Moringa oleifera Aqueous extract in egg yolk skim milk extender for post thawed Limousin Bull sperm quality on aspect viability, and the level of. The treatment was divided into five groups: egg yolk and skim milk diluter (P0), 2,5% M. oleifera aqueous ekstract in 4 ml egg yolk skim milk (P1), 5% M. oleifera aqueous ekstract in 4 ml egg yolk skim milk (P2), 10% M. oleifera aqueous ekstract in 4 ml egg yolk skim milk (P3), 20% M. oleifera aqueous ekstract in 4 ml egg yolk skim milk (P4). The sperm quality was observed post thawing. The data were analyzed using ANOVA and Duncant Test. The best sperm motility showed on P2 with 43b ± 5,70, the best sperm viability showed on P3 with 58,20b ± 8,72 and than the lowest level of malondialdehyde showed on P4 with 5,434a ± 1,034. In conclusion addition of M. oleifera on dose 10% can increase quality of Limousin Sperm Post Thawed.

Meanwhile, bull seminal plasma contains the insulin-like growth factor-1 (IGF-1) complex, which is cryoprotective. Materials and Methods: Buck semen was diluted in the following skim milk-egg yolk extender preparations: Without the addition of Simmental bull seminal plasma IGF-1 complex protein (T0); with the addition of 12-μg Simmental bull seminal plasma IGF-1 complex protein (T1); and with the addition of 24-μg Simmental bull seminal plasma IGF-1 complex protein (T2). The extended semen was packed in 0.25-mL straws and frozen. Post-thawed semen fertility was evaluated based on the following variables: Sperm motility, viability, intact plasma membrane (IPM), malondialdehyde (MDA) levels, capacitation status, and acrosome reaction. The difference in each variable among the groups was evaluated using analysis of variance, followed by Tukey's honestly significant difference test, at a 95% level of significance. Meanwhile, principal component analysis (PCA) was used to identify the principal component of semen fertility among the seven parameters. Results: The T1 group showed the highest sperm motility, viability, IPM, and percentage of incapacitated sperm and the lowest MDA levels, percentage of capacitated sperm, and acrosome reaction. PCA revealed that sperm motility had a moderate to very robust correlation with other variables and is the most crucial parameter, accounting for 80.79% of all variables. Conclusion: The IGF-1 complex in Simmental bull seminal plasma was useful for increasing the fertility of post-thawed Kacang buck semen, and sperm motility was the principal component of semen fertility.

Background: Malnutrition can cause an increase in oxidative stress as it triggers the expression of Heat Shock Protein70 (HSP70), a chaperon molecule that is needed to repair damaged cells within optimal levels. Honey is a source of feed that can stimulate HSP70 expression, which can be given to the malnourished in the animal trial. Aim: The purpose of this study was to prove that Kaliandra honey can improve testosterone levels, diameter and epithelial thickness of the seminiferous tubule of rat testes (Rattus norvegicus) due to malnutrition through stimulation of Heat Shock Protein 70 (HSP70), which is expressed immunohistochemically (IHC). Methods: This study used 40 male rats, which were divided into 4 treatment groups: T0 (negative control): normal rats and not given honey; T1 (positive control): malnourished rats and not given honey; T2 (Treatment 2): malnourished rats and given 30% Kaliandra honey (v/v) for 10 days; T3 (Treatment 3), malnourished rats and given 50% Kaliandra honey (v/v) for 10 days. The condition of malnutrition is carried out by fasting the feed for 5 consecutive days resulting in damage to the male reproductive organs, especially the testes. Results: The results showed that Kaliandra honey at a dose of 50% (v/v) had a significant effect in improving testosterone levels, diameter and epithelial thickness of seminiferous tubules of malnourished male rats through stimulation of HSP70 expression. The HSP70 expression scores by IHC at T0, T1, T2 and T3 were 0.15a±0.5, 3.15c±0.4, 2.95c±0.35 and 1.75b±0.15, sequentially. Elisa indirect testosterone levels at T0, T1, T2 and T3 (in µg/dL) were 36.39c±0.35, 6.12a±0.51, 7.45a±0.15, 25.27b±0.63, sequentially. The diameter and epithelial thickness of the seminiferous tubule of the testes (in µm) in the four treatments T0, T1, T2 and T3 were 362.40c±4.71, 248.46a±3.90, 255.22a±2.34, 318.37b±4.23 and 117.60d±11.30, 3.86a±1.57, 9.72b±3.96, 29.84c±4.02 sequentially. Conclusion: The conclusion of the study showed that Kaliandra honey at a dose of 50% (v/v) had a significant effect in improving testosterone levels, diameter and epithelial thickness of the seminiferous tubule of malnourished rats through stimulation of HSP70, although not significantly the same as negative control (T0).

Aim: This study aimed to develop equations to predict daily milk production (DMP) based on linear body and udder morphometry of Holstein Friesian (HF) dairy cows. Materials and Methods: The experiment was conducted on 174 lactating HF dairy cows reared by farmers at different locations under similar conditions. The age, parity, and body condition score of experimental animals were limited to 0.25 of the standard deviation value above or below the average. The average DMP was based on farmers' records. Morphometry components, i.e., body length (BL); chest circumference (CC); front udder height (FUH), rear udder height (RUH); and udder circumference (UC) were directly measured using a tape; meanwhile, body weight (BW) was estimated using the Indonesia Winter formula. Aim: The aim of this study was to know crude guava leaf tannins effect on motility, viability, and intact plasma membrane of stored spermatozoa of Etawa crossbred goats. Materials and Methods: Macroscopic assessment of normal Etawa crossbred semen was followed by dilution with a glucose solution at a 1:10 ratio to increase volume. The diluted semen was treated by adding crude guava leaf tannins into 1 ml of the semen glucose diluent, and five treatments were obtained, namely, control group (C), with no added tannins; treatment Group 1 (T1), with 3%; treatment Group 2 (T2), with 6% tannins; treatment Group 3 (T3), with 12% tannins; and treatment Group 4 (T4), with 24% tannins. Each treatment used five replications. Then, microscopic analysis of the treated and control semen was carried out after 15 days of storage at 4-5°C temperature. The parameters observed were motility, pH, viability, abnormality, and intact spermatozoa plasma membrane. Results: The spermatozoa motility in Group C was the highest (76.60±1.47). The motility in Group T1 did not differ from that in Group C, but was different and higher than that in Groups T2, T3, and T4. The pH of Group C tended to be acidic after 15 days of storage (4.78±0.01) as compared to the initial pH of fresh semen (6.76±0.12). The pH in Group C did not differ from that in the Groups T1 and T2, but differed from that in the T3 and T4 groups; the pH in the T3 and T4 groups was similar. The viability of spermatozoa in the T1 group was higher than that in all treatments (64.60±2.76); the lowest values were found in Group C (28.94±1.02). Group C had the lowest number of normal spermatozoa, with a mean of 72.58±3.48. The total number of abnormalities in the T2 group did not differ from those in the T3 group, and abnormalities in the T4 group did not differ from those in Group C, which exhibited the highest abnormalities in the head, neck, and tail. The most significant decrease was observed in the intact plasma membrane of spermatozoa on addition of 12% and 24% crude guava leaf tannin in glucose diluents. Conclusion: The addition of 3% crude guava leaf tannin to crossbred Etawa goat semen diluted with glucose diluent and stored for 15 days at 4-5°C resulted in a significant effect on spermatozoa motility, viability, and intact plasma membrane, whereas the administration of 24% crude guava leaf tannin resulted in low live percentage of spermatozoa.

2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) is a persistent organic pollutant that induces overproduction of reactive oxygen species (ROS). Studies on avoiding the adverse effects of dioxin pollution exposure are needed in all aspects, including reproductive health. This study aimed to determine the effect of α-tocopherol on superoxide dismutase (SOD) and malondialdehyde (MDA) levels, live spermatozoa, apoptosis, and necrosis in male rats exposed to dioxin as a model. Thirty healthy 12-week-old male rats were randomly divided into five groups. Rats in the control group were given corn oil twice daily at 4-hour intervals. The remaining rats were given TCDD 700 mg/kg BW daily, followed by administration of corn oil and α-tocopherol at doses of 77, 140, and 259 mg/kg BW/d for T0, T1, T2, and T3 groups, respectively. The treatments were conducted for 45 days; all rats were euthanized to collect blood and testicular samples on day 46. The results showed that exposure of TCDD resulted in a decrease in SOD activity and live spermatozoa and increased MDA level and death, apoptosis, and necrosis of spermatozoa (T0) compared to the control (C) group (p 0.05) to those of the control group. It could be concluded that the administration of α-tocopherol resolves the harmful effects of TCDD on the viability of spermatozoa in rats as a model.

Aim: The purpose of this study was to determine the benefits of L-arginine addition in skim milk extender to maintain the quality of goat spermatozoa in chilled storage. Materials and Methods: A total of 18 ejaculates from three healthy goats with weight and age of 45 kg and 4-5 years, respectively, were divided into three groups. The control group contained goat semen diluted in a skim milk extender without L-arginine; Treatment I and Treatment II contained goat semen diluted in a skim milk extender with added L-arginine 4 and 6 mM, respectively. These three groups were chilled at 5°C and evaluated daily for 5 days. Observed variables were viability, motility, intact plasma membrane (IPM), malondialdehyde (MDA) level, necrosis, and apoptosis of spermatozoa. Data were obtained by stratified random sampling from population to get 18 dairy cows, which divided into three groups. The first, second, and third groups each with milk yield of less than 17, between 17-21, and over 21 liters/day, and with calving interval of less than 365, between 365-450 and more than 450 days. Blood was collected from the coccygeal vein at the time of insemination (D0), 7 day (D+7) and 22 days (D+22) after insemination. BUN and progesterone concentration were measured. Data analysis showed no significant difference (P>0.05) on the observed parameters. This study was the first to combine the addition of antioxidants to a skim milk–egg yolk (SM–EY) extender and different equilibration periods to obtain higher quality post-thawed Kacang buck semen. This study aimed to determine the effects of green tea extract (GTE) on the quality of frozen Kacang goat sperm equilibrated for one and two hours. The pool of Kacang buck ejaculate was equally divided into four portions and was diluted in an SM–EY extender that contained four doses of 0, 0.05, 0.10, and 0.15 mg of GTE/100 mL for T0, T1, T2, and T3 groups, respectively. The aliquots were treated for an equilibration period of 1–2 h before further processing as frozen semen. Post-thawed semen quality was evaluated for sperm quality. The Sanger method was used for DNA sequencing, and the amino acid sequence was read using MEGA v.7.0. The post-thawed semen of the T2 group that was equilibrated for one hour had the highest semen quality. Pre-freezing motility had the highest determination coefficient compared to post-thawed sperm motility. This study is the first to report amino acid mutation due to freeze–thawing. The frequency of amino acid mutations revealed that T2 was the least mutated amino acid. Glycine, valine, leucine, serine, and asparagine strongly correlated to post-thawed sperm motility. It can be concluded that a combination of 0.1 mg GTE/100 mL extender as an antioxidant and one-hour equilibration period resulted in the best post-thawed Kacang buck semen quality.

The study aimed to determine the addition of green tea extract (GTE) in extender on the quality and DNA mutation of post-thawed Kacang buck sperm. The sperm DNA mutation was observed on nicotinamide adenine dinucleotide hydride (NADH) dehydrogenase 1 (ND1) of mitochondrial Deoxyribonucleic Acid (mtDNA). A pool of 12 Kacang buck ejaculates was diluted in skim milk-egg yolk extender contained 0, 0.05, 0.10, and 0.15 mg of GTE/100 mL for T0, T1, T2, and T3 group, respectively. Each of the aliquot groups was packaged in 0.25 mL French mini straw contained 60 million alive sperm and froze according to the protocol. The ND1 mtDNA amplification of samples was carried out Polymerase Chain Reaction machine, followed by DNA sequencing using the Sanger method. Meanwhile, the phylogenetic tree was constructed using the neighbor-joining (NJ) method with MEGA 7.0 software. The results showed that the T2 group maintained the highest quality for Kacang buck post-thawed semen. There was the highest percentages of sperms viability, motility, intact plasma membrane (IPM), the lowest of malondialdehyde (MDA) concentration, sperm DNA fragmentation (SDF), the total and types of ND1 mtDNA mutation frequency. The phylogenetic tree analysis revealed that the clade of the T2 group was most closely related to the sequence reference. However, there was no correlation between the semen quality parameters (sperm viability, motility, IPM, MDA concentration, and SDF) with ND1 mtDNA mutation of post-thawed Kacang buck semen. It could be concluded that GTE was useful as an antioxidant for Kacang buck semen extender for frozen sperm.

Artificial insemination has proven to be an effective method for increasing population size and genetic quality of Kacang goats. However, innovation is required to maintain the quality of Kacang goat semen in storage. This study aimed to examine the effects of supplementing the 150 kDa protein assumed as IGF-I complex derived from bull seminal plasma in skim milk-egg yolk extender on the quality of Kacang goat sperm stored at 5°C. Twelve ejaculates collected from three Kacang goats were divided into three groups. In the control group (T0), the ejaculates were extended with skim milk-egg yolk only. In the treatment groups (T1 and T2), the ejaculates were extended with skim milk-egg yolk supplemented with the IGF-I complex protein at 12 μg and 24 μg/100 mL, respectively. The extended semen was stored at 5°C, and the viability, motility, intactness of the plasma membrane, malondialdehyde concentration, and apoptotic sperm percentage were evaluated daily for five days. The results showed that the T1 was the most effective treatment for maintaining Kacang goat semen at a quality acceptable for artificial insemination over five days of storage at 5°C. However, the T0 and T2 groups retained acceptable qualities for only three days at 5°C. It could be concluded that supplementation of 12 μg of the 150 kDa protein derived from bull seminal plasma per 100 mL extender successfully extended the life span of Kacang goat sperm for five days.

Since the damage to alveolar tissue due to cigarette smoke exposure (CSE) is lipid peroxidation, antioxidant treatment is needed. The red guava (Psidium guajava L.) fruit contains antioxidants derived from quercetin, lycopene, and vitamin C. This study aimed to determine the effect of red guava fruit extract (RGFE) on the alveolar tissue of rats exposed to cigarette smoke. The 25 rats (Rattus norvegicus) were divided into five groups. The control and T0 groups were only administered placebo, while T1, T2, and T3 groups were orally administered RGFE of 18.9, 37.8, and 56.7 mg/kg body weight daily for 44 days. The CSE dose of 20 suctions daily was conducted on T0, T1, T2, and T3 groups on days 15–44. On day 45, all rats were sacrificed for serum collection and histopathological lung slides with eosin-nigrosin staining. To study the antimitotic effect of an extract of the leaf Achyranthes asapera Linn on embryonal cleavage rats (Rattus norwegicus). Rats ova were fertilized in vitro, and thes suitable zygote and embryo were selected to use as samples, which were divided into 5 groups. Each group comprised 80 ova/embryo. Those groups were: group 1. ova for fertilization, group 2. zygote, group 3. 2 cell embryo, group 4. 4 cell embryo and group 5. 8 cell embryo. Each group was divided into 4 treatmens, each of which consisted of 20 ova/ embryos. The treatments were as follows: control treatment receiving TCM 199 media, and treatment 1, 2 and 3 receiving Achyranthes aspera Linn alkaloid of 20, 30, and 40 ppm respectively. Observation to embryonal cleavage and development was out 12 and 24 hours after treatment. Concluded from the results that the administration of Achyranthes aspera Linn as much as 30 ppm in vitro could 1) inhibits fertilization zygote and 2, 4, and 8 cell embryonal cleavage, growth and development 2) induced zygote and embryonal membrane recruitment and blastomere degeneration and 3) inhibits zygote and embryonal mitotic cleavage at metaphase stage.

Q fever, also known as query fever, is a zoonotic illness brought on by the Coxiella burnetii bacteria. This disease was first discovered in 1935 in Queensland, Australia. Worldwide, Q fever is a disease that requires notification, and certain nations classify it as a national health concern. A feature of C. burnetii is known as cell wall phase fluctuation. Serological testing is the main method used to diagnose Q fever illnesses. Inhalation is the primary method of C. burnetii transmission in both people and animals, with smaller amounts occurring through milk and milk product ingestion. The bacterial strain that is causing the infection determines how severe it is. Q fever is a significant zoonosis that can be dangerous for personnel working in veterinary laboratories, livestock breeding operations, and slaughterhouses due to its high human contagiousness. Coxiella burnetii is a biological weapon that can be sprayed on food, water, or even mail. It can also be employed as an aerosol. Antibiotics work well against this disease's acute form, but as the infection develops into a chronic form, treatment becomes more difficult and the illness frequently returns, which can result in a high death rate. Vaccination has been demonstrated to lower the incidence of animal infections, C. burnetii shedding, and abortion. This study aimed to determine the effects of green tea extract (GTE) on the quality of frozen Kacang goat sperm equilibrated for one and two hours. The pool of Kacang buck ejaculate was equally divided into four portions and was diluted in an SM-EY extender that contained four doses of 0, 0.05, 0.10, and 0.15 mg of GTE/100 mL for T0, T1, T2, and T3 groups, respectively. The aliquots were treated for an equilibration period of 1-2 h before further processing as frozen semen. Post-thawed semen quality was evaluated for sperm quality. The Sanger method was used for DNA sequencing, and the amino acid sequence was read using MEGA v.7.0. The post-thawed semen of the T2 group that was equilibrated for one hour had the highest semen quality. Pre-freezing motility had the highest determination coefficient compared to post-thawed sperm motility. This study is the first to report amino acid mutation due to freeze-thawing. The frequency of amino acid mutations revealed that T2 was the least mutated amino acid. Glycine, valine, leucine, serine, and asparagine strongly correlated to post-thawed sperm motility. It can be concluded that a combination of 0.1 mg GTE/100 mL extender as an antioxidant and one-hour equilibration period resulted in the best post-thawed Kacang buck semen quality.

35. α -Tocopherol Prevents Sperm Apoptosis and Necrosis in Rats Exposed to 2,3,7,8-Tetrachlorodibenzo-p-dioxin.

2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) is a persistent organic pollutant that induces overproduction of reactive oxygen species (ROS). Studies on avoiding the adverse effects of dioxin pollution exposure are needed in all aspects, including reproductive health. This study aimed to determine the effect of α -tocopherol on superoxide dismutase (SOD) and malondialdehyde (MDA) levels, live spermatozoa, apoptosis, and necrosis in male rats exposed to dioxin as a model. Thirty healthy 12-week-old male rats were randomly divided into five groups. Rats in the control group were given corn oil twice daily at 4-hour intervals. The remaining rats were given TCDD 700 mg/kg BW daily, followed by administration of corn oil and α -tocopherol at doses of 77, 140, and 259 mg/kg BW/d for T0, T1, T2, and T3 groups, respectively. The treatments were conducted for 45 days; all rats were euthanized to collect blood and testicular samples on day 46. The results showed that exposure of TCDD resulted in a decrease in SOD activity and live spermatozoa and increased MDA level and death, apoptosis, and necrosis of spermatozoa (T0) compared to the control (C) group ( p < 0.05). The administration of α -tocopherol, starting from the doses of 77 (T1), 149 (T2), and 259 mg (T3) per kg BW, was sequentially followed by returning MDA levels, recovering SOD activities, and restoration in the percentage of living, dead, apoptotic, and necrotic spermatozoa, similar ( p > 0.05) to those of the control group. - Abstract
Background and Aim: The production of male calf beef cattle is an agricultural innovation needed to increase the farm's productivity as a provider of meat sources. This study aimed to determine the sex ratio of the offspring of cows inseminated with Y-bearing sperm enriched by Percoll density gradient centrifugation and swim-up, combined with delayed fixed-time artificial insemination (FTAI)., Materials and Methods: Ejaculates of Simmental bulls were divided into four equal portions and grouped as T0 (control, non-sexed semen), T1 and T2 were sexed semen using Percoll density gradient centrifugation three and five levels, respectively, and T3 was sexed semen using swim-up. After the sex was sorted, the semen was diluted in a tris-egg yolk extender, packaged in French mini-straws containing 50 million live sperm cells, and frozen. Pre-sexed, post-sexed, and post-thawed spermatozoa were evaluated based on progressive motility, viability, intact plasma membrane, and abnormality. The post-thawed semen of T0 was artificially inseminated to recipient cows at 12 h after onset of estrus (not delayed FTAI). Meanwhile, the delayed FTAI was conducted 18-20 h after onset of estrus using the T0, the best of T1 and T2, and the T3 post-thawed semen., Results: The Percoll density gradient centrifugation reduced motility, viability, and intact plasma membrane but increased sperm abnormalities. Meanwhile, the swim-up process increased motility, viability, and intact plasma membrane of sperm cells but decreased sperm abnormalities. Post-thawed semen decreased motility, viability, and intact plasma membrane of sperm cells but increased sperm abnormalities. 37. A 150 kDa Protein Derived from Bull Seminal Plasma Extended the Survival Time of Kacang Goat Sperm Stored at 5°C.

Artificial insemination has proven to be an effective method for increasing population size and genetic quality of Kacang goats. However, innovation is required to maintain the quality of Kacang goat semen in storage. This study aimed to examine the effects of supplementing the 150 kDa protein assumed as IGF-I complex derived from bull seminal plasma in skim milk-egg yolk extender on the quality of Kacang goat sperm stored at 5°C. Twelve ejaculates collected from three Kacang goats were divided into three groups. In the control group (T0), the ejaculates were extended with skim milk-egg yolk only. In the treatment groups (T1 and T2), the ejaculates were extended with skim milk-egg yolk supplemented with the IGF-I complex protein at 12 μ g and 24 μ g/100 mL, respectively. The extended semen was stored at 5°C, and the viability, motility, intactness of the plasma membrane, malondialdehyde concentration, and apoptotic sperm percentage were evaluated daily for five days. The results showed that the T1 was the most effective treatment for maintaining Kacang goat semen at a quality acceptable for artificial insemination over five days of storage at 5°C. However, the T0 and T2 groups retained acceptable qualities for only three days at 5°C. 38. Effect of insulin-like growth factor-1 complex of Simmental bull seminal plasma on post-thawed Kacang buck semen fertility.

- Abstract
Background and Aim: Kacang buck sperm is cryosensitive due to the seminal plasma of semen itself. Meanwhile, bull seminal plasma contains the insulin-like growth factor-1 (IGF-1) complex, which is cryoprotective. The addition of the crude protein of Simmental bull seminal plasma increased the quality of post-thawed semen of Kacang buck. The study was conducted to determine the effects of Simmental bull seminal plasma with IGF-1 on the fertility of post-thawed Kacang buck semen., Materials and Methods: Buck semen was diluted in the following skim milk-egg yolk extender preparations: Without the addition of Simmental bull seminal plasma IGF-1 complex protein (T0); with the addition of 12-μg Simmental bull seminal plasma IGF-1 complex protein (T1); and with the addition of 24-μg Simmental bull seminal plasma IGF-1 complex protein (T2). The extended semen was packed in 0.25-mL straws and frozen. Post-thawed semen fertility was evaluated based on the following variables: Sperm motility, viability, intact plasma membrane (IPM), malondialdehyde (MDA) levels, capacitation status, and acrosome reaction. The difference in each variable among the groups was evaluated using analysis of variance, followed by Tukey's honestly significant difference test, at a 95% level of significance. Meanwhile, principal component analysis (PCA) was used to identify the principal component of semen fertility among the seven parameters., Results: The T1 group showed the highest sperm motility, viability, IPM, and percentage of incapacitated sperm and the lowest MDA levels, percentage of capacitated sperm, and acrosome reaction. PCA revealed that sperm motility had a moderate to very robust correlation with other variables and is the most crucial parameter, accounting for 80.79% of all variables., Conclusion: The IGF-1 complex in Simmental bull seminal plasma was useful for increasing the fertility of post-thawed Kacang buck semen, and sperm motility was the principal component of semen fertility., (Copyright: © Susilowati, et al.)

Background: Malnutrition can cause an increase in oxidative stress as it triggers the expression of heat shock protein70 (HSP70), a chaperon molecule that is needed to repair damaged cells within optimal levels. Honey is a source of feed that can stimulate HSP70 expression, which can be given to the malnourished in the animal trial., Aim: The purpose of this study was to prove that Kaliandra honey can improve testosterone levels, diameter, and epithelial thickness of the seminiferous tubule of rat testes ( Rattus norvegicus ) due to malnutrition through stimulation of HSP70, which is expressed immunohistochemically., Methods: This study used 40 male rats, which were divided into four treatment groups: T0 (negative control): normal rats and not given honey; T1 (positive control): malnourished rats and not given honey; T2 (treatment 2): malnourished rats and given 30% Kaliandra honey (v/v) for 10 days; T3 (treatment 3), malnourished rats and given 50% Kaliandra honey (v/v) for 10 days. The condition of malnutrition is carried out by fasting the feed for five consecutive days resulting in damage to the male reproductive organs, especially the testes., Results: The results showed that Kaliandra honey at a dose of 50% (v/v) had a significant effect in improving testosterone levels, diameter, and epithelial thickness of seminiferous tubule of malnourished male rats through stimulation of HSP70 expression. The HSP70 expression scores by IHC at T0, T1, T2, and T3 were 0.15

40. The Restorative Effect of Red Guava ( Psidium guajava L.) Fruit Extract on Pulmonary Tissue of Rats ( Rattus norvegicus ) Exposed to Cigarette Smoke.

- Abstract
Since the damage to alveolar tissue due to cigarette smoke exposure (CSE) is lipid peroxidation, antioxidant treatment is needed. The red guava ( Psidium guajava L.) fruit contains antioxidants derived from quercetin, lycopene, and vitamin C. This study aimed to determine the effect of red guava fruit extract (RGFE) on the alveolar tissue of rats exposed to cigarette smoke. The 25 rats ( Rattus norvegicus ) were divided into five groups. The control and T0 groups were only administered placebo, while T1, T2, and T3 groups were orally administered RGFE of 18.9, 37.8, and 56.7 mg/kg body weight daily for 44 days. The CSE dose of 20 suctions daily was conducted on T0, T1, T2, and T3 groups on days 15-44. On day 45, all rats were sacrificed for serum collection and histopathological lung slides with eosin-nigrosin staining. The result showed that CSE caused an increase ( p < 0.05) in malondialdehyde (MDA) levels, cell death, apoptosis, and necrosis percentages, congestion and thickening of alveolar septum tissue, and reduction in the alveolar diameter and alveolar number. Administration of RGFE suppressed those effects, and the highest dose of RGFE (T3) restored ( p > 0.05) MDA levels, percentage of apoptotic and necrosis, alveolar septal thickening, and alveolar diameter. However, the percentages of cell death, alveolar congestion, and the alveolar number were still worse ( p < 0.05) than in normal rats. 41. Green tea extract increases the quality and reduced DNA mutation of post-thawed Kacang buck sperm.

- Abstract
The study aimed to determine the addition of green tea extract (GTE) in extender on the quality and DNA mutation of post-thawed Kacang buck sperm. The sperm DNA mutation was observed on nicotinamide adenine dinucleotide hydride (NADH) dehydrogenase 1 (ND1) of mitochondrial Deoxyribonucleic Acid (mtDNA). A pool of 12 Kacang buck ejaculates was diluted in skim milk-egg yolk extender contained 0, 0.05, 0.10, and 0.15 mg of GTE/100 mL for T0, T1, T2, and T3 group, respectively. Each of the aliquot groups was packaged in 0.25 mL French mini straw contained 60 million alive sperm and froze according to the protocol. The ND1 mtDNA amplification of samples was carried out Polymerase Chain Reaction machine, followed by DNA sequencing using the Sanger method. Meanwhile, the phylogenetic tree was constructed using the neighbor-joining (NJ) method with MEGA 7.0 software. The results showed that the T2 group maintained the highest quality for Kacang buck post-thawed semen. There was the highest percentages of sperms viability, motility, intact plasma membrane (IPM), the lowest of malondialdehyde (MDA) concentration, sperm DNA fragmentation (SDF), the total and types of ND1 mtDNA mutation frequency. The phylogenetic tree analysis revealed that the clade of the T2 group was most closely related to the sequence reference. However, there was no correlation between the semen quality parameters (sperm viability, motility, IPM, MDA concentration, and SDF) with ND1 mtDNA mutation of post-thawed Kacang buck semen. The genetic resources of Indonesia's indigenous Kacang goat require preservation. Artificial insemination is expected to accelerate population increases and preserve genetic resources simultaneously. The present study was the maiden attempt for cryopreservation of Kacang buck sperm. The objectives of this study were to determine whether the supplementation of superior Simmental bull seminal plasma protein in egg yolk-citrate extender could improve the quality of post-thawed Kacang buck sperm, increase conceptions rates, and improve kidding rates. Buck semen was diluted without supplementation (T0) and with supplementation of 2.5 mg (T1) and 5 mg (T2) of Simmental bull seminal plasma protein per mL egg yolk-citrate extender. Extended semen was packed in 0.25 mL straw as cryopreserved frozen semen. Post-thawed semen samples were evaluated for viability, motility, intact plasma membranes, malondialdehyde level, and DNA fragmentation. Estrus was synchronized for sixty Kacang does, which were divided randomly into three groups and inseminated using post-thawed semen. The progesterone serum concentration of the does was measured 7 and 22 days post-insemination to detect ovulation and conception. Pregnancy was confirmed using abdominal palpation at 43 days post-insemination and by observing birth. The T1 group showed the highest (P < 0.05) post-thawed viability, motility, and intact plasma membrane. Conception, pregnancy and kidding rates were also higher in T1 than other treatment groups. In conclusion, the 2.5 mg Simmental bull seminal plasma protein supplementation per mL egg yolk-citrate extender provided the best seminal quality and fertility of post-thawed Kacang buck semen., (Copyright © 2020 Elsevier Inc. Aim: The purpose of this study was to determine the benefits of L-arginine addition in skim milk extender to maintain the quality of goat spermatozoa in chilled storage., Materials and Methods: A total of 18 ejaculates from three healthy goats with weight and age of 45 kg and 4-5 years, respectively, were divided into three groups. The control group contained goat semen diluted in a skim milk extender without L-arginine; Treatment I and Treatment II contained goat semen diluted in a skim milk extender with added L-arginine 4 and 6 mM, respectively. These three groups were chilled at 5°C and evaluated daily for 5 days. Observed variables were viability, motility, intact plasma membrane (IPM), malondialdehyde (MDA) level, necrosis, and apoptosis of spermatozoa., Results: The addition of L-arginine 4 mM was the best treatment in maintaining viability, motility, and IPM and a decreased MDA level, percentage of necrosis, and apoptosis of goat spermatozoa. An ejaculate in this extender can be divided into 37 doses for intracervical insemination in <1 ml volume with 125 million motile spermatozoa., Conclusion: Goat semen retained its quality when kept for 5 days in chilled storage by adding L-arginine in skim milk extender., (Copyright: © Susilowati, et al.)
