23. Welfare of Decapod Crustaceans with Special Emphasis on Stress Physiology

Abstract

Despite the growing concern on animal welfare in crustacean farming, both from legislative bodies as well as the common public, studies on welfare are limited and transfer to routine farming is missing. While biocertification schemes such as the Aquaculture Stewardship Council (ASC) involve a welfare dimension, these dimensions cannot be communicated to the consumer in a scientifically sound manner. Animal welfare is recognized as integral part of sustainability due to the losses associated with bad animal welfare standards and is considered highly relevant by consumers around the world. On the other hand, increasing animal welfare is also required for the optimisation of aquaculture technology. Behaviour of the animals suggests that decapod crustaceans experience nociception and there are several indications of pain perception as well. Also, distress has rarely been evaluated under routine aquaculture conditions and markers for chronic stress detection need to be identified. Indeed, most work on welfare of crustaceans focuses on cellular, oxidative stress only. Here, a comprehensive assessment of chronic stress should be carried out to optimize rearing technology in nurseries, during ongrowing, harvesting, anesthesia, transportation, and humane slaughter in terms of a good aquaculture practise.

32. Influence of Substrates on the Quality of Hermetia Meal for Fish Meal Substitution in Nile Tilapia Oreochromis niloticus

Abstract

Commercially produced black soldier flies (Hermetia illucens) represent a promising fish meal substitute, particularly in the context of using agricultural by-products and waste. Here, the culture of Hermetia maggots on five selected substrates (potato protein (P) as a by-product of starch production, rapeseed oil cake (R) from rape oil production, maize silage (M), soybean (S) meal and, as a control, concentrated chicken feed (C)) were evaluated, assessing the growth performance of Hermetia maggots related to the overall production and the nutritional composition of the respective meal. Subsequently, their use as ingredients in aquafeed formulations was evaluated in a feeding trial with juvenile Nile tilapia Oreochromis niloticus, assessing the growth performance of the fish. Substrates used for Hermetia culture significantly affected the growth and development of the maggots, revealing substantial differences in the meal quality. Still, if incorporated in isonitrogenous and isocaloric diets (33% crude protein, 21–22 MJ/kg) replacing 75% of the fishmeal protein in the formulated diets, no significant differences in growth performance of the fish were observed compared to the fishmeal control. As a conclusion, substrates clearly affect the production yield and the composition of maggots. Abstract

Creatine is an amino acid derivate commonly found in vertebrate muscle tissue. Creatine facilitates the recycling of adenosine triphosphate and thus contributes to the energy supply of the muscles as well as the brain. Creatine is used as a supplement for several reasons and its effects in humans, particularly in sports medicine, have been studied excessively. Also, creatine supplementation has been studied for its functions and benefits in terrestrial farm animals. Up to date, little is known about the use of creatine as a supplement in fish nutrition. Yet, due to its many physiological functions, creatine may serve as a valuable supplement in aquafeeds of farmed aquaculture species. Indeed, creatine plays a pivotal role in the fish's muscle and may help to enhance performance of fish reared in aquaculture systems. With regard to swimming exercise, creatine may even amplify its metabolic effects. Upon supplementation, creatine stimulates muscle growth increasing body mass and it has the potential to improve feed utilisation particularly of plant‐based diets. Also, creatine plays a part in osmoregulation when fish adapt to changes in salinity. Furthermore, it may improve product quality upon slaughter. Here, we compile what is known about the many functions of creatine as well as its physiological effects in fish in comparison to mammals. We also highlight its potential beneficial effects as a supplement in aquaculture and infer why creatine can help increase the sustainability of fish feeds. [ABSTRACT FROM AUTHOR]

41. Probiotics in Fish Nutrition—Long-Standing Household Remedy or Native Nutraceuticals?

Abstract

Over the last decades, aquaculture production increased rapidly. The future development of the industry highly relies on the sustainable utilization of natural resources. The need for improving disease resistance, growth performance, food conversion, and product safety for human consumption has stimulated the application of probiotics in aquaculture. Probiotics increase growth and feed conversion, improve health status, raise disease resistance, decrease stress susceptibility, and improve general vigor. Currently, most probiotics still originate from terrestrial sources rather than fish. However, host-associated (autochthonous) probiotics are likely more persistent in the gastrointestinal tract of fish and may, therefore, exhibit longer-lasting effects on the host. Probiotic candidates are commonly screened in in vitro assays, but the transfer to in vivo assessment is often problematic. In conclusion, modulation of the host-associated microbiome by the use of complex probiotics is promising, but a solid understanding of the interactions involved is only in its infancy and requires further research. Probiotics could be used to explore novel ingredients such as chitin-rich insect meal, which cannot be digested by the fish host alone. Abstract

International audience; Background: Easy-to-use and fast bioinformatics pipelines for long-read assembly that go beyond the contig level to generate highly continuous chromosome-scale genomes from raw data remain scarce. Results: Chromosome-Scale Assembler (CSA) is a novel computationally highly efficient bioinformatics pipeline that fills this gap. CSA integrates information from scaffolded assemblies (e.g., Hi-C or 10X Genomics) or even from diverged reference genomes into the assembly process. As CSA performs automated assembly of chromosome-sized scaffolds, we benchmark its performance against state-of-the-art reference genomes, i.e., conventionally built in a laborious fashion using multiple separate assembly tools and manual curation. CSA increases the contig lengths using scaffolding, local re-assembly, and gap closing. On certain datasets, initial contig N50 may be increased up to 4.5-fold. For smaller vertebrate genomes, chromosome-scale assemblies can be achieved within 12 h using low-cost, high-end desktop computers. Mammalian genomes can be processed within 16 h on compute-servers. Using diverged reference genomes for fish, birds, and mammals, we demonstrate that CSA calculates chromosome-scale assemblies from long-read data and genome comparisons alone. Even contig-level draft assemblies of diverged genomes are helpful for reconstructing chromosome-scale sequences. CSA is also capable of assembling ultra-long reads. Conclusions: CSA can speed up and simplify chromosome-level assembly and significantly lower costs of large-scale family-level vertebrate genome projects.
