Abstract

In the family of histone-deacetylases, histone deacetylase 6 (HDAC6) stands out. The cytoplasmic class IIb histone deacetylase (HDAC) family is essential for many cellular functions. It plays a crucial and debatable regulatory role in innate antiviral immunity. This review summarises the current state of our understanding of HDAC6's structure and function in light of the three mechanisms by which it controls DNA and RNA virus infection: cytoskeleton regulation, host innate immune response, and autophagy degradation of host or viral proteins. In addition, we summed up how HDAC6 inhibitors are used to treat a wide range of diseases, and how its upstream signaling plays a role in the antiviral mechanism. Together, the findings of this review highlight HDAC6's importance as a new therapeutic target in antiviral immunity, innate immune response, and some diseases, all of which offer promising new avenues for the development of drugs targeting the immune response. 14. Prevalence and antibiogram of Escherichia coli O157 isolated from bovine in Jimma, Ethiopia: abattoirbased survey

Abstract

E. coli O157 is an important serotype that caused many food borne outbreaks worldwide in the past decades. This study was carried out to estimate the prevalence and determine the antimicrobial susceptibility of E. coli O157 isolated from bovine carcasses and cecal contents at one abattoir in Jimma. A total of 300 samples from bovine carcass swabs(n=150) and cecal contents(n=150) were examined to identify E. coli O157 by ISO 17604:2005 method and by using Dry spot E. coli O157 latex test kit. Susceptibility to panels of 9 antimicrobial agents for all 25 E. coli O157 isolates was examined The overall prevalence of E. coli O157 from bovine carcass swabs and cecal contents were 9.3% and 7.3%, respectively. All E. coli O157 isolates were susceptible to chloramphenicol, ceftriaxone, sulfamethoxazole-trimethoprim, tetracycline and 96% of the isolates were susceptible to amoxacillin-clavulanic acid. Twenty-eight, 24% and 20% of the isolates were resistant to amikacin, streptomycin and cephalothin respectively. In conclusion, considerable proportions of bovine carcasses and cecal contents in the current study harbored drug resistant E. coli O157 and pose a significant public health risk. Abstract

An alternative vaccine design approach and diagnostic kits are highly required against the anticipated pandemicity caused by the South African Territories type 2 (SAT2) Foot and Mouth Disease Virus (FMDV). However, the distinct antigenicity and immunogenicity of VP1, VP0, and VP3 of FMDV serotype SAT2 are poorly understood. Similarly, the particular roles of the three structural proteins in novel vaccine design and development remain unexplained. We therefore constructed VP1, VP0, and VP3 encoding gene (SAT2:JX014256 strain) separately fused with His-SUMO (histidine-small ubiquitin-related modifier) inserted into pET-32a cassette to express the three recombinant proteins and separately evaluated their antigenicity and immunogenicity in mice. The fusion protein was successfully expressed and purified by the Ni-NTA resin chromatography. The level of serum antibody, spleen lymphocyte proliferation, and cytokines against the three distinct recombinant proteins were analyzed. Results showed that the anti-FMDV humoral response was triggered by these proteins, and the fusion proteins did enhance the splenocyte immune response in the separately immunized mice. We observed low variations among the three fusion proteins in terms of the antibody and cytokine production in mice. Hence, in this study, results demonstrated that the structural proteins of SAT2 FMDV could be used for the development of immunodiagnostic kits and subunit vaccine designs. [ABSTRACT FROM AUTHOR]
