The Hippo pathway senses cellular conditions and regulates YAP/TAZ to control cellular and tissue homeostasis, while TBK1 is central for cytosolic nucleic acid sensing and antiviral defence. The correlation between cellular nutrient/physical status and host antiviral defence is interesting but not well understood. Here we find that YAP/TAZ act as natural inhibitors of TBK1 and are vital for antiviral physiology. Independent of transcriptional regulation and through the transactivation domain, YAP/TAZ associate directly with TBK1 and abolish virus-induced TBK1 activation, by preventing TBK1 Lys63-linked ubiquitylation and the binding of adaptors/substrates. Accordingly, YAP/TAZ deletion/depletion or cellular conditions inactivating YAP/TAZ through Lats1/2 kinases relieve TBK1 suppression and boost antiviral responses, whereas expression of the transcriptionally inactive YAP dampens cytosolic RNA/DNA sensing and weakens the antiviral defence in cells and zebrafish. Thus, we describe a function of YAP/TAZ and the Hippo pathway in innate immunity, by linking cellular nutrient/physical status to antiviral host defence.

Coupling hydrolysis and electrochemical oxidation is an attractive strategy for polyethylene terephthalate (PET) recycling. The development of a highly efficient and stable electro-catalyst for the ethylene glycol oxidation reaction (EGOR) is crucial in this process. In this work, we reported a self-supported Ni/Ni3N1−x heterostructure with abundant nitrogen vacancies as highly efficient electro-catalysts to oxidize ethylene glycol to formate with a high faradaic efficiency (94.5%) and selectivity (88.6%). The combination of advanced spectroscopic techniques and density functional theory calculations reveals that the presence of nitrogen vacancies shifts the Ni d-band center towards the Fermi level, thus enhancing the adsorption of EG and lowering the energy required for state transition in the EGOR. The result of this work can guide the development of highly active EGOR electrocatalysts, promoting the application of recycling waste PET plastics through hydrolysis coupled with electrochemical oxidation processes. Emerging evidence suggests differential antagonism of lactic acid‐producing bacteria (LAB) to Helicobacter pylori, posing challenges to human health and food safety due to unclear mechanisms. This study assessed 21 LAB strains from various sources on H. pylori growth, urease activity, and coaggregation. Composite scoring revealed that Latilactobacillus sakei LZ217, derived from fresh milk, demonstrates strong inhibitory effects on both H. pylori growth and urease activity. L. sakei LZ217 significantly reduced H. pylori adherence of gastric cells in vitro, with inhibition ratios of 47.62%. Furthermore, in vivo results showed that L. sakei LZ217 alleviated H. pylori‐induced gastric mucosa damage and inflammation in mice. Metabolomic exploration revealed metabolic perturbations in H. pylori induced by L. sakei LZ217, including reduced amino acid levels (e.g., isoleucine, leucine, glutamate, aspartate, and phenylalanine) and impaired carbohydrate and nucleotide synthesis, contributing to the suppression of ureA (28.30%), ureE (84.88%), and ureF (59.59%) expressions in H. pylori. This study underscores the efficacy of LAB against H. pylori and highlights metabolic pathways as promising targets for future interventions against H. pylori growth and colonization. At present, Ag nanoparticles have been widely used as Raman substrates, but their easy oxidation and aggregation have limited practical applications. In order to address the above problems, firstly, tetraethyl orthosilicate as a precursor was applied to synthesis silver coated with silica dioxide nanoparticles (Ag@SiO2). As a result, the surface of Ag nanoparticles is uniformly coated with a thin layer of SiO2 in order to solve the easy oxidation problem without adversely affecting their surface‐enhanced Raman scattering (SERS) performance. Furthermore, Ag@SiO2 nanoparticles were electrostatically deposited onto polyamide (PA) films to form a two‐dimensional PA‐Ag@SiO2 film substrate, thus resolving nanoparticle agglomeration issues and further improving the repeatability of the entire system. As can been from the SERS detection results obtained from the probe molecules and pollutants, the Raman signal on the PA‐Ag@SiO2 thin film substrate has a good degree of sensitivity, stability, and repeatability. Cytosolic RNA/DNA sensing elicits primary defense against viral pathogens. Interferon regulatory factor 3 (IRF3), a key signal mediator/transcriptional factor of the antiviral-sensing pathway, is indispensible for interferon production and antiviral defense. However, how the status of IRF3 activation is controlled remains elusive. Through a functional screen of the human kinome, we found that mammalian sterile 20-like kinase 1 (Mst1), but not Mst2, profoundly inhibited cytosolic nucleic acid sensing. Mst1 associated with IRF3 and directly phosphorylated IRF3 at Thr75 and Thr253. This Mst1-mediated phosphorylation abolished activated IRF3 homodimerization, its occupancy on chromatin, and subsequent IRF3-mediated transcriptional responses. In addition, Mst1 also impeded virus-induced activation of TANK-binding kinase 1 (TBK1), further attenuating IRF3 activation. As a result, Mst1 depletion or ablation enabled an enhanced antiviral response and defense in cells and mice. Therefore, the identification of Mst1 as a novel physiological negative regulator of IRF3 activation provides mechanistic insights into innate antiviral defense and potential antiviral prevention strategies.

For the purpose of investigating the chemical enhancement of amorphous semiconductors as well as increasing the sensitivity of the surface-enhanced Raman spectroscopy (SERS) substrate, titanium dioxide (TiO2) precursors were calcined at different temperatures to generate crystallized TiO2 (c-TiO2) and amorphous TiO2 (a-TiO2) nanosheets, respectively. Afterward, a two-dimensional (2D) a-TiO2/Ag nanosheet SERS substrate was successfully fabricated using electrostatic interaction between a-TiO2 and Ag nanoparticles. In order to demonstrate a greater SERS sensitivity on a-TiO2/Ag compared to either c-TiO2 or Ag nanoparticles alone, the SERS probe molecules rhodamine 6G (R6G) and malachite green (MG) were utilized. Based on the results of SERS detections for probe molecules and contaminants, it demonstrates that a-TiO2/Ag nanosheets produce highly sensitive and repeatable Raman signals. The detectable concentration limits for R6G and MG were found to be 10−11 M and 10–10 M, respectively. And it has been determined that the system exhibits an enhancement factor (EF) of up to 1 × 108. The limit of detection for 4-mercaptobenzoic acid and alizarin red can both reach 1 × 10–8. Furthermore, a finite-difference time-domain simulation is performed in order to evaluate the magnetic field strength generated by Ag nanoparticles. As a result of the simulation, it is evident that the actual EF is smaller than the calculated one, leading support to the view that a-TiO2 nanosheets have a beneficial effect on the chemical enhancement of SERS.
