Through millions of years of evolution, bones have developed a complex and elegant hierarchical structure, utilizing tropocollagen and hydroxyapatite to attain an intricate balance between modulus, strength, and toughness. In this study, continuous fiber silk composites (CFSCs) of large size are prepared to mimic the hierarchical structure of natural bones, through the inheritance of the hierarchical structure of fiber silk and the integration with a polyester matrix. Due to the robust interface between the matrix and fiber silk, CFSCs show maintained stable long-term mechanical performance under wet conditions. During in vivo degradation, this material primarily undergoes host cell-mediated surface degradation, rather than bulk hydrolysis. We demonstrate significant capabilities of CFSCs in promoting vascularization and macrophage differentiation toward repair. A bone defect model further indicates the potential of CFSC for bone graft applications. Our belief is that the material family of CFSCs may promise a novel biomaterial strategy for yet to be achieved excellent regenerative implants.

This paper explores the application and development trends of artificial intelligence (AI) technology, particularly machine learning and natural language processing in the field of failure analysis. Failure analysis is a crucial method for ensuring the reliability and safety of equipment, and is widely used in aerospace, automotive manufacturing, electronic devices, and other fields. Traditional failure analysis methods often rely on expert experience, which is time-consuming and laborious. By integrating AI’s powerful data processing capabilities with traditional methods, the accuracy and efficiency of analysis have been significantly enhanced. In terms of failure mode diagnosis, AI can rapidly and accurately identify various fault modes and provide precise diagnostic results. In failure cause diagnosis, AI integrates data from multiple sources to uncover complex failure factors and potential causal relationships, improving diagnostic reliability. In failure prediction, machine learning can accurately forecast material lifespan and strength, reducing experimental time and costs. In failure prevention, AI offers new approaches to effectively reduce the risk of failure and lower product maintenance costs. The paper also looks forward the future development prospects of AI in failure analysis and highlights challenges and recommendations in the areas, such as data quality improvement, model optimization, interdisciplinary collaboration, and ethical and safety issues.

High-performance, fast-growing natural materials with sustainable and functional features currently arouse significant attention. Here, facile processing, involving delignification, in situ hydrothermal synthesis of TiO2 and pressure densification, is employed to transform natural bamboo into a high-performance structural material. The resulting TiO2-decorated densified bamboo exhibits high flexural strength and elastic stiffness, with both properties more than double that of natural bamboo. Real-time acoustic emission reveals the key role of the TiO2 nanoparticles in enhancing the flexural properties. The introduction of nanoscale TiO2 is found to markedly increase the degree of oxidation and the formation of hydrogen bonds in bamboo materials, leading to extensive interfacial failure between the microfibers, a micro-fibrillation process that results in substantial energy consumption and high fracture resistance. This work furthers the strategy of the synthetic reinforcement of fast-growing natural materials, which could lead to the expanded applications of sustainable materials for high-performance structural applications.

High stiffness and strength carbon fibres are commonly used to reinforce epoxy-resin composites. While wild Antheraea pernyi silk fibres exhibit high toughness originating from their α-helix/random coil conformation structures and their micro-fibre morphology, their insufficient strength and stiffness hinders them from being used in similar structural composites. In this work, we use interply hybridization of silk and carbon fibres to reinforce epoxy-matrix composites. With increased carbon fibre content, the quasi-static tensile/flexural stiffness and strength increases following the rule of mixtures while more silk fibre acts to increase ductility and impact strength. This results in a composite comprising equal volumes of carbon and silk fibres achieving an impact strength of 98 kJ m-2, which is twice that of purely carbon-fibre reinforced composites (44 kJ m-2). This work shows tough natural silk fibres and strong synthetic fibres can be successfully integrated into epoxy-resin composites for tailored mechanical properties.

Strong and tough epoxy composites are developed using a less-studied fibre reinforcement, that of natural silk. Two common but structurally distinct silks from the domestic B. mori/Bm and the wild A. pernyi/Ap silkworms are selected in fabric forms. We show that the toughening effects on silk-epoxy composites or SFRPs are dependent on the silk species and the volume fraction of silk. Both silks enhance the room-temperature tensile and flexural mechanical properties of the composite, whereas the more resilient Ap silk shows a more pronounced toughening effect and a lower critical reinforcement volume for the brittle-ductile transition. Specifically, our 60 vol.% Ap-SFRP displays a three-fold elevation in tensile and flexural strength, as compared to pure epoxy resin, with an order of magnitude higher breaking energy via a distinct, ductile failure mode. Importantly, the 60 vol.% Ap-SFRP remains ductile with 7% flexural elongation at lower temperatures (-50 °C). Under impact, these SFRPs show significantly improved energy absorption, and the 60 vol.% Ap-SFRP has an impact strength some eight times that of pure epoxy resin. The findings demonstrate both marked toughening and strengthening effects for epoxy composites from natural silk reinforcements, which presents opportunities for mechanically superior and "green" structural composites.

Hemophagocytic lymphohistiocytosis (HLH) is a life-threatening hyperinflammatory disorder that occurs as a consequence of immune dysregulation. HLH can be primary (familial or non-familial) or secondary to infection, autoimmune disease or malignancy. Malignancy-associated HLH is often accompanied by hematologic and lymphoid neoplasms. This report describes the case of a 3-year-old girl with an initial diagnosis of acute lymphoblastic leukemia who subsequently developed HLH during primary chemotherapy. She was admitted with a pulmonary infection, and initial blood tests showed thrombocytopenia and anemia. Whole-exome sequencing of gene and whole transcriptome RNA sequencing data indicated mutations of UNC13D. The hospital course was complicated by multiple infections, altered mental status and acute respiratory distress syndrome. HLH secondary to multiple infections that achieved remission following targeted therapy with ruxolitinib, in conjunction with corticosteroids and other complementary treatments. The technology for obtaining the high-efficiency expression of target proteins through site-specific recombination has made progress. However, using the CRISPR/Cas9 system for site-specific integration of long fragments and the expression of active proteins remains a challenge. This study optimized the linear DNA circularization system, eliminated the prokaryotic plasmid backbone on the traditional foreign gene vector, and generated a homologous arm-free circular donor template with a single guide RNA target site (sgRNA TS). This strategy significantly increased the co-transfection efficiency of the 1.6 kb template and Cas9 plasmid by 1.15-fold, and the average knock-in (KI) efficiency of the 4.7 kb long-fragment template for the two target gene sites increased by 1.3-fold. Subsequently, we used rhBCHE as a reporter gene to efficiently integrate the 5.4 kb fragment containing the gene of interest (GOI) into specific sites in the HEK293T cell line to detect the expression of the circular template at different target sites. Overall, this study further verifies that the length of the circular donor is more conducive to non-homologous integration, and more importantly, we provide a simple and optimized strategy for the construction of long-fragment site integration cell lines. The fatigue crack growth behaviors of Ti-6.5Al-2Zr-1Mo-1V titanium alloy at different positive stress ratios (R = 0.1, 0.3 and 0.5) were investigated at room temperature. The effects of stress ratio R on the fatigue crack growth rates and fractures morphology were discussed. The results show that with the increase of stress ratio R , the fatigue crack growth rates are increased at the given applied cyclic stress intensity factor, ΔK. Equivalently, the observed threshold stress-intensity range ΔKth and the range of stress intensity factor ΔK for fatigue crack growth are decreased as the positive stress ratio is increased. Some fatigue steps and secondary cracks are observed on the fracture surfaces of the specimens tested at the three stress ratios. In addition, with the increase of the stress ratio, the toughness of the surface is increased and the secondary cracks are decreased. However, the fatigue fracture mechanisms of Ti-6.5Al-2Zr-1Mo-1V titanium alloy tested at the three stress ratios are all the same, which the fractures are all presented in transgranular mode.
