Two new europium coordination compounds, [Eu2(papip)2(PhCOO)6] and [Eu2(phpip)2(PhCOO)6] (papip is 1-phenyl-2-(4-aminophenyl)imidazo[4,5-f][1,10]phenanthroline and phpip 1-phenyl-2-(4-hydroxyphenyl)imidazo[4,5-f][1,10]phenanthroline), have been synthesized hydrothermally and structurally characterized, involving the in situ hydrolysis of phenylacetonitrile to phenylacetate. The –NH2 group in the former complex was unexpectedly generated from the –NO2 group of free 1-phenyl-2-(4-nitrophenyl)imidazo[4,5-f][1,10]phenanthroline during the reaction process, representing the first such transformation in a hydrothermal reaction. Both complexes exhibit bright red photoemission typical of Eu(III) f–f transition under UV excitation, with long excited state lifetimes and quite high photoluminescence quantum yields. Neuregulin-1 (NRG-1) is a member of the epidermal growth factor family. Our previous study showed that NRG-1 protected neurons from apoptosis following focal cerebral ischemia by the inhibition of caspase-3 and TNF-a expression. However, the molecular signaling mechanisms for this action of NRG-1 following cerebral ischemia are not fully understood. Presently, activation of the PI3K/Akt pathway has been implicated as a major contributor to neuronal survival after an ischemic insult. In the present study, we investigated whether NRG-1 modulates the activation of Akt and its downstream targets Bad and Bcl-2 expression after transient focal cerebral ischemia by intraluminal blockade of the middle cerebral artery. Western blot was employed to analyze the change of phosphorylated Akt (p-Akt) expression; reverse transcription and polymerization chain reaction (RT-PCR) were used to measure changes of Bcl-2 mRNA. The level of phosphorylation of Bad (p-Bad) was determined using an enzyme-linked immunosorbent assay (ELISA). Our results showed that recombinant human NRG-1(3.0 ng-kg-1) significantly increased the expression of p-Akt protein, Bcl-2 mRNA, and the level of p-Bad, respectively, whereas administration of LY294002, a specific inhibitor of PI3K, significantly decreased the expression of p-Akt, p-Bad, and Bcl-2 induced by NRG-1 after a 60 min ischemic insult, followed by 24 h of reperfusion. These results indicate that NRG-1 may be involved in regulating the expression of Bcl-2 and p-Bad through the PI3K/Akt pathway after transient focal cerebral ischemia. Une etude precedente nous a montre que la NRG-1 protegeait les neurones de l'apoptose a la suite d'une ischemie cerebrale focale en inhibant l'expression de la caspase-3 et du TNF-a. Cependant, les mecanismes de signalisation moleeculaire de cet effet de la NRG-1 apres l'ischemie cerebrale ne sont pas parfaitement compris. Presentement, l'activation de la voie PI3K/Akt a ete impliquee comme contributrice majeure de la survie neuronale apres l'ischemie. Dans l'etude presente, nous avons determine si la NRG-1 module l'activation de l'Akt et l'expression de Bad et Bcl-2, ses cibles en aval, a la suite d'une ischemie cerebrale focale induite par blocage de Fartere cerebrale mediane. Le buvardage par Western a ete utilise pour analyser le changement d'expression d'AKT phosphorylee (p-Akt), et la reaction par polymerase en chaine couplee a une transcriptase inverse (RT-PCR) a ete utilisee pour mesurer les changements d'expression de l'ARNm de Bcl-2. Le niveau de phosphorylation de Bad (p-Bad) a ete determine par ELISA <>. Nos resultats ont montre que la NRG-1 recombinante humaine (3,0 ng-kg-1) augmentait significativement l'expression de la p-Akt, de l'ARNm de Bcl-2 et les niveaux de pBAD, alors que l'administration de LY294002, un inhibiteur specifique de PI3K diminuait significativement l'expression de p-Akt, de p-Bad et de Bcl-2 induite par la NRG-1, apres une ischemie de 60 min suivie d'une periode de reperfusion de 24 h. Ces resultats indiquent que la NRG-1 peut etre impliquee en regulant l'expression de Bcl-2 et de p-Bad via la voie PI3K/Akt apres une ischemie cerebrale focale transitoire.
