13. Precambrian geological history of the Tarim Craton (NW China) involving the assembly and fragmentation of the Columbia and Rodinia supercontinents: review and synthesis.

Abstract

The Tarim Craton (NW China) is a significant archive of the tectonic events that occurred during the assembly and break-up of the Precambrian supercontinents Columbia and Rodinia. It provides a comprehensive record of crustal development during the Proterozoic. We review and synthesize the magmatic, metamorphic and stratigraphic records of the Precambrian Tarim Craton and delineate the geochronology of significant geological events that led to the formation of this major cratonic block. The extant geophysical and geological data show that the Tarim Craton consists mainly of the South and North Tarim blocks. The record of late Paleoproterozoic tectonothermal events is displayed well in and across the craton, involving the amalgamation of these two blocks to form its unified crystalline basement during the build-up of Columbia. However, the record of mid-Neoproterozoic events during the assembly of Rodinia is only exposed around its periphery. The Proterozoic record of the craton includes Mesoproterozoic–early Neoproterozoic low-grade metamorphic rock units and a late Neoproterozoic sedimentary cover. The early Mesoproterozoic stratigraphy in the SW, NE and SE of the Tarim Basin suggests a period of tectonic stability following the amalgamation of the North and South Tarim blocks in the late Paleoproterozoic, indicating the completion of its cratonic build-up. Septic shock and acute lung injury (ALI) are the main etiology of death caused by sepsis. Glucocorticoids, which are commonly used in clinic to antagonize the inflammatory response of sepsis, may cause serious side effects. Isoforskolin (ISOF) from the plant Coleus forskohlii stimulates adenylyl cyclase, increases the cAMP level and inhibits inflammatory response. The aim of this study was to investigate the synergistic effect of ISOF with dexamethasone (DEX) to prevent and ameliorate septic inflammation.Methods: Lipopolysaccharide (LPS) of 30 and 5 mg/kg (iv.) was used to induce sepsis and ALI mice model respectively in vivo. BEAS-2B cells stimulated by LPS were applied as cell model in vitro. The cumulative survival of mice with LPS-induced sepsis and the histopathological changes of lungs in mice with acute lung injury were observed, and the secretion of pro-inflammatory cytokines was analyzed by ELISA. El desarrollo de una herramienta automática de extracción de archivos PDF y Excel puede tener un impacto significativo en muchas empresas. La implementación de dicho sistema conlleva un aumento en la eficiencia operativa, una mayor precisión de datos, una productividad mejorada y la capacidad de tomar decisiones basadas en datos más precisos. En comparación con la extracción manual de datos, se ahorra tiempo y recursos, al tiempo que se mejora la calidad y precisión de los datos extraídos. Además, garantiza que las decisiones se basen en datos más confiables, lo que hace que el proceso sea más eficiente y efectivo. Para garantizar un funcionamiento fluido del extractor, es necesario tener en cuenta varios factores, como una implementación adecuada, un mantenimiento continuo y actualizaciones constantes. Es esencial anticipar los posibles formatos que se pueden proporcionar para los archivos PDF y Excel, ya que el extractor se basa en expresiones regulares para su funcionamiento. Abstract

Context Current medicine for Alzheimer's disease (AD) cannot effectively reverse or block nerve injury. Traditional Chinese Medicine practice and research imply Aconiti lateralis Radix Praeparata (Fuzi) may meet this goal. Objective Analysing the anti-AD effect of Fuzi and its potential molecular mechanism. Materials and methods AD model cells were treated with Fuzi in 0-300 mg/mL for 24 h in 37 °C. The cell viability (CV) and length of cell projections (LCP) for each group were observed, analysed, and standardised using control as a baseline (CVs and LCPs). The Fuzi and AD relevant genes were identified basing on databases, and the molecular mechanism of Fuzi anti-AD was predicted by network analysis. Results Experiment results showed that Fuzi in 0.4 mg/mL boosted LCP (LCPs = 1.2533, p ≤ 0.05), and in 1.6-100 mg/mL increased CV (CVs from 1.1673 to 1.3321, p ≤ 0.05). Bioinformatics analysis found 17 Fuzi target genes (relevant scores ≥ 20), showing strong AD relevant signals (RMS_p ≤ 0.05, related scores ≥ 5), enriched in the pathways regulating axon growth, synaptic plasticity, cell survival, proliferation, apoptosis, and death (p ≤ 0.05). Especially, GRIN1 and MAPK1 interacted with APP protein and located in the key point of the "Alzheimer's disease" pathway. Discussion and conclusions These results suggest that Fuzi may have therapeutic and prevention potential in AD, and GRIN1 and MAPK1 may be the core of the pathways of the Fuzi anti-AD process. Fuzi should be studied more extensively, especially for the prevention of AD.

39. Gut microbiota and differential genes-maintained homeostasis is key to maintaining health of individuals with Yang-deficiency constitution

Abstract

OBJECTIVE: Yang-deficiency constitution (YADC) is a common unbalanced constitution that predisposes individuals to certain diseases. However, not all people with YADC manifest develop diseases. This calls for delineation of the underlying molecular mechanisms. Previous studies suggested that the gut microbiota and gene differential expression should be considered. METHODS: In the present study, we compared profiles of gut microbiota between four healthy YADC individuals and those of five healthy balanced constitution (BC) counterparts, based on 16S rRNA gene sequence analysis. Furthermore, YADC relevant genes identified by comparing 62 healthy YADC and 58 healthy BC individuals in total to perform intersection analysis, functional clustering and pathway enrichment analyses. RESULTS: The levels of harmful gut microbiota (Prevotellaceae, LDA score > 4.0, P = 0.0141) and beneficial gut microbiota (Ruminococcaceae, LDA score > 4.0, P = 0.0025, Faecalibacterium, LDA score > 4.0, P = 0.0484) were both elevated in healthy YADC individuals. Also, we found that the specific metabolic pathway with 2, 6-Dichloro-p-hydroquinone 1, 2-Dioxygenase (PcpA) as the core in gut microbiota and the glutathione transferase activity has been enriched by YADC relevant genes in healthy YADC individuals were both responsible for the detoxification of halogenated aromatic hydrocarbon substances. CONCLUSIONS: Both beneficial and harmful factors had been detected in healthy YADC individuals, functionally, they may have triggered homeostasis to maintain the health of individuals with YADC. The homeostasis may be maintained by beneficial and harmful factors from gut flora and genes. Future studies are expected to focus on halogenated aromatic hydrocarbons and their detoxification processes.

Abstract

Chemical synthesis of proteins bearing base‐labile post‐translational modifications (PTMs) is a challenging task. For instance, O‐acetylation and S‐palmitoylation PTMs cannot survive Fmoc removal conditions during Fmoc‐solid phase peptide synthesis (SPPS). In this work, we developed a new Boc‐SPPS‐based strategy for the synthesis of peptide C‐terminal salicylaldehyde (SAL) esters, which are the key reaction partner in Ser/Thr ligation and Cys/Pen ligation. The strategy utilized the semicarbazone‐modified aminomethyl (AM) resin, which could support the Boc‐SPPS and release the peptide SAL ester upon treatment with TFA/H2O and pyruvic acid. The non‐oxidative aldehyde regeneration was fully compatible with all the canonical amino acids. Armed with this strategy, we finished the syntheses of the O‐acetylated protein histone H3(S10ac, T22ac) and the hydrophobic S‐palmitoylated peptide derived from caveolin‐1. [ABSTRACT FROM AUTHOR]
