15. Rapid access to phospholipid analogs using thiol-yne chemistry

Abstract

Phospholipids and glycolipids constitute an essential part of biological membranes, and are of tremendous fundamental and practical interest. Unfortunately, the preparation of functional phospholipids, or synthetic analogs, is often synthetically challenging. Here we utilize thiol-yne click chemistry methodology to gain access to phospho- and glycolipid analogs. Alkynyl hydrophilic head groups readily photoreact with numerous thiol modified lipid tails to yield the appropriate dithioether phospho- or glycolipids. The resulting structures closely resemble the structure and function of native diacylglycerolipids. Dithioether phosphatidylcholines (PCs) are suitable for forming giant unilamellar vesicles (GUV), which can be used as vessels for cell-free expression systems. The unnatural thioether linkages render the lipids resistant to phospholipase A2 hydrolysis. We utilize the improved stability of these lipids to control the shrinkage of GUVs composed of a mixture of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) and dioleyl-dithioether PC, concentrating encapsulated nanoparticles. We imagine that these readily accessible lipids could find a number of applications as natural lipid substitutes.

33. In SituFabrication of Robust Polyphenolic Hydrogels for Skin Protection and Repair

Abstract

Robust and multifunctional polyphenolic hydrogels have been well acknowledged as promising biomaterial candidates due to their various fascinating properties in regulating cell biology. However, the construction of these hydrogels commonly needs multistep fabrication and a sophisticated gelation process. The typical phenol-aldehyde condensation is a facial and in situreaction for industrial resin construction; however, its application in hydrogel fabrication is seldom reported. In this study, we reported the feasibility and modularity of this strategy for in situfabricating various kinds of robust and multifunctional natural polyphenolic hydrogels using natural polyphenol (extracts) and formaldehyde. The physicochemical characterization results demonstrated that the chemical structure (e.g., phenolic hydroxyl groups) of polyphenol molecules could be well maintained after the gelation process, allowing as-prepared hydrogels to inherit the merits of polyphenol molecules, such as good UV shielding, radical scavenging, antibacterial properties, and so forth, as demonstrated by in vitrodata. The in vitroand in vivocellular and animal results further confirmed that the as-prepared hydrogels were able to perform a skin protection and repair role, as demonstrated in UV protection and wound healing models. Collectively, this strategy could allow natural polyphenols as both structural and functional synthons toward more types of robust hydrogels for a wide range of biomedical applications.

Abstract

Dendritic polymers have highly branched three‐dimensional architectures, the fourth type apart from linear, cross‐linked, and branched one. They possess not only a large number of terminal functional units and interior cavities, but also a low viscosity with weak or no entanglement. These features endow them with great potential in various biomedicine applications, including drug delivery, gene therapy, tissue engineering, immunoassay and bioimaging. Most review articles related to bio‐related applications of dendritic polymers focus on their drug or gene delivery, while very few of them are devoted to their function as cancer diagnosis agents, which are essential for cancer treatment. In this review, we will provide comprehensive insights into various dendritic polymer‐based cancer diagnosis agents. Their classification and preparation are presented for readers to have a precise understanding of dendritic polymers. On account of physical/chemical properties of dendritic polymers and biological properties of cancer, we will suggest a few design strategies for constructing dendritic polymer‐based diagnosis agents, such as active or passive targeting strategies, imaging reporters‐incorporating strategies, and/or internal stimuli‐responsive degradable/enhanced imaging strategies. Their recent applications in in vitro diagnosis of cancer cells or exosomes and in vivo diagnosis of primary and metastasis tumor sites with the aid of single/multiple imaging modalities will be discussed in great detail. Abstract

Highly fluorogenic tetrazine bioorthogonal probes emitting at near‐infrared wavelengths are in strong demand for biomedical imaging applications. Herein, we have developed a strategy for forming a palette of novel Huaxi‐Fluor probes in situ, whose fluorescence increases hundreds of times upon forming the bioorthogonal reaction product, pyridazine. The resulting probes show large Stokes shifts and high quantum yields. Manipulating the conjugate length and pull–push strength in the fluorophore skeleton allows the emission wavelength to be fine‐tuned from 556 to 728 nm. The highly photo‐stable and biocompatible probes are suitable for visualizing organelles in live cells without a washing step and for imaging of tumors in live small animals to depths of 500 μm by two‐photon excitation.
