Abstract

Organoid models have the potential to recapitulate the biological and pharmacotypic features of parental tumors. Nevertheless, integrative pharmaco-proteogenomics analysis for drug response features and biomarker investigation for precision therapy of patients with liver cancer are still lacking. We established a patient-derived liver cancer organoid biobank (LICOB) that comprehensively represents the histological and molecular characteristics of various liver cancer types as determined by multiomics profiling, including genomic, epigenomic, transcriptomic, and proteomic analysis. Proteogenomic profiling of LICOB identified proliferative and metabolic organoid subtypes linked to patient prognosis. High-throughput drug screening revealed distinct response patterns of each subtype that were associated with specific multiomics signatures. Through integrative analyses of LICOB pharmaco-proteogenomics data, we identified the molecular features associated with drug responses and predicted potential drug combinations for personalized patient treatment. The synergistic inhibition effect of mTOR inhibitor temsirolimus and the multitargeted tyrosine kinase inhibitor lenvatinib was validated in organoids and patient-derived xenografts models. We also provide a user-friendly web portal to help serve the biomedical research community. Our study is a rich resource for investigation of liver cancer biology and pharmacological dependencies and may help enable functional precision medicine.

13. Design and disturbance rejection control of a piezoelectric nanopositioning stage.

Abstract

The article covers structural design and disturbance rejection control of a flexure-based compliant piezoelectric nanopositioning stage. The stage is devised using four-bar amplification mechanisms, auxiliary guiding mechanisms, and a compound parallelogram mechanism. Static and dynamic models of compliant mechanisms are analyzed based on a compliance matrix method. A modified Prandtl-Ishlinskii (PI) model with dead zone operators is used to describe the asymmetric hysteresis. Then, a global electromechanical dynamic model is built and identified. A comprehensive disturbance rejection controller (CDRC) combining a robust H∞ control and a disturbance observer-based (DOB) control is proposed for high trajectory tracking accuracy and anti-disturbance performance. The trajectory tracking accuracy of the stage can be up to 3 nm in experiments. As amplitudes of reference signals increases, relative tracking errors gradually decrease. System instability and oscillation caused by strong external disturbances are also reduced. Experimental results prove the effectiveness of the developed CDRC. [ABSTRACT FROM AUTHOR]

Abstract

This paper reports the design, modeling, and control of a novel three-degrees-of-freedom piezoelectric compliant microstage by introducing a new double-rocker mechanism. The double-rocker mechanism combines a first (leverage) amplifier and a second (rocker) amplifier for double-stage displacement amplification and parasitic motion reduction. An analytical model is established to calculate the deformation behavior of the microstage, and the model is verified using finite-element analysis (FEA). An improved Prandtl-Ishlinskii (PI) model is proposed to describe piezoelectric hysteresis characteristics by optimizing the threshold selection. Then, a composite control strategy is designed to achieve precision trajectory control. The control strategy consists of a hysteresis-based feedforward controller and a proportional-integral feedback controller. A prototype of the microstage is manufactured, and an experimental system is established. Several open-loop and closed-loop experiments are conducted, and the experimental results validate the effectiveness of the proposed microstage and the designed control strategy.

Abstract

Micromanipulation tasks require microgrippers equipped with large displacement/size performance and position/force control capabilities. This paper reports the structural design, implementation and position/force switching control of an S-shaped microgripper which is driven by the micro-fiber-composite (MFC) actuator. Unlike conventional piezoelectric microgrippers, the MFC one can provide excellent characteristics of a larger displacement-volume ratio (i.e. the ratio of the maximal output displacement to the total volume of the microgripper) and simple structure. Combining the Hamilton’s principle and the Bouc-Wen model, the dynamic model considering both the structural flexibility and the hysteresis nonlinearity of the microgripper is derived. Then, FEA simulations are conducted and the optimal structural dimensions are acquired. Moreover, the challenge of guaranteeing the smooth transition between the position and the force is realized through a switching control scheme. In particular, a fuzzy discrete sliding mode (FDSM) controller based on the dynamic model and a fuzzy regulator is used to control the output displacement, and an incremental PI controller is used to regulate the gripping force. To validate the effectiveness of the developed microgripper and the control scheme, experimental investigations and the control of pick-and-release tasks are carried out. The results verify their effectiveness and demonstrate that the MFC microgripper and the control scheme are suitable use for precision micromanipulation and microassembly applications.

19. AGO1 enhances the proliferation and invasion of cholangiocarcinoma via the EMT-associated TGF-β signaling pathway.

Abstract

Purpose: To detect the expression of AGO1 in cholangiocarcinoma and explore its role and significance in the progression of cholangiocarcinoma., Patients and Methods: Using immunohistochemistry, Western blotting and qPCR, we examined the expression of AGO1 in cholangiocarcinoma tissues. Through the analysis of clinical case data, the relationship between AGO1 and clinical prognosis was explored. The effect of AGO1 on cholangiocarcinoma was verified by cell functional experiments. Finally, we examined the effects of AGO1 on EMT-related proteins and signaling pathways., Results: AGO1 is significantly upregulated in cholangiocarcinoma and is associated with the prognosis of cholangiocarcinoma. AGO1 can significantly increase the proliferation, migration and invasion of cholangiocarcinoma cell lines. AGO1 affects the prognosis of cholangiocarcinoma by affecting epithelial-mesenchymal transition (EMT)-related TGF-β-PI3K-AKT signaling pathways., Conclusion: AGO1 is an independent predictor of cholangiocarcinoma prognosis and a potential target for the treatment of cholangiocarcinoma., Competing Interests: None., (AJTR Copyright © 2020.)
