10. Exogenous Calcium Alleviates the Photosynthetic Inhibition and Oxidative Damage of the Tea Plant under Cold Stress.

Abstract

Calcium (Ca2+), a second messenger, plays a crucial role in plant growth and development as well as in responding to biotic and abiotic stresses. In this work, we explored the role of exogenous calcium in alleviating cold stress and examined the relationship between calcium chloride (CaCl2) and calcium channel blockers, lanthanum chloride (LaCl3), in tea plants under cold stress at the physiological and transcriptional levels. Exogenous Ca2+ partially offsets the negative impacts of cold stress which increased the tolerance of tea plants by significantly raising the photochemical efficiency of PSII, protective enzyme activities, and the ABA content, which reduced the relative electrical conductivity (REC) level and the malondialdehyde (MDA) concentration. At the transcriptome level, exogenous Ca2+ significantly enhanced the expression of key genes involved in cold response pathways. Nevertheless, LaCl3 treatment not only significantly inhibited the activities of antioxidant enzymes including superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT), but also increased cold damage. This study aims to provide essential insight into the role of exogenous Ca2+ in tea plants responding to cold stress, and to better understand the molecular mechanisms that facilitate Ca-mediated cold tolerance. Abstract

Exploiting adsorbents with highly efficient extraction performance is of great promise for extracting small organic molecules from biological samples. In this work, a novel Zn2+‐immobilized chitosan@silica hybrid monolith was prepared through a simple self‐assembly Zn2+‐immobilization process. Exploited as an adsorbent in solid‐phase micro‐extraction for extracting trace β‐agonists, the monolith exhibited high extraction efficiencies for salbutamol, clenbuterol, and ractopamine with the enrichment factors approaching 120, 85, and 52, respectively. These could be attributed to the effective interaction between Zn2+ ions and the target molecule via coordination or other intermolecular interactions. Under optimized extraction operations, a sensitive determination was successfully developed coupling with high‐performance liquid chromatography‐ultraviolet detection. The linear range was 0.17–58.8, 0.12–68.5, and 0.18–65.5 ng/ml for salbutamol, clenbuterol, and ractopamine. The limits of detection of the β‐agonists were from 0.04 to 0.07 ng/ml, and the limits of quantification were from 0.12 to 0.18 ng/ml. The recoveries of spiking in mutton samples were observed in the range of 85.9%–95.7%, with relative standard deviations <8.0% (n = 3). Application tests demonstrated this newly developed determination was practical, accurate, and convenient for detecting trace content β‐agonists in meat. [ABSTRACT FROM AUTHOR]

Abstract

A facile in-tube solid phase microextraction (in-tube SPME) procedure was developed to enrich ractopamine before HPLC-UV analysis. This was achieved by employing amide groups modified polysaccharide-silica hybrid monolith as an efficient sorbent. The monolith was synthesized by a simple reaction with agarose oxide and tetramethoxylisane, followed by the modification of amide groups via subsequent ring opening, “thiol–ene” click and dehydration reactions. Under the optimized extraction conditions, the enrichment factors for ractopamine, dopamine, clenbuterol, para-methylphenol and phenol were determined to be 50.5, 32.2, 4.8, 2.1 and 1.8, respectively. The monolithic column has ideal selectivity for ractopamine. Coupled with HPLC-UV, this method demonstrated a linearity within 2.0–800 ng/g for ractopamine with spiking in pork muscles (R2 = 0.9958). The LOD was 0.64 ng/g (S/N = 3) and recoveries ranged from 85.2 to 108.1% (n = 3). This approach provides a feasible way for analysis of trace ractopamine in biological samples.

42. DFsn collaborates with Highwire to down-regulate the Wallenda/DLK kinase and restrain synaptic terminal growth

Abstract

Abstract Background The growth of new synapses shapes the initial formation and subsequent rearrangement of neural circuitry. Genetic studies have demonstrated that the ubiquitin ligase Highwire restrains synaptic terminal growth by down-regulating the MAP kinase kinase kinase Wallenda/dual leucine zipper kinase (DLK). To investigate the mechanism of Highwire action, we have identified DFsn as a binding partner of Highwire and characterized the roles of DFsn in synapse development, synaptic transmission, and the regulation of Wallenda/DLK kinase abundance. Results We identified DFsn as an F-box protein that binds to the RING-domain ubiquitin ligase Highwire and that can localize to the Drosophila neuromuscular junction. Loss-of-function mutants for DFsn have a phenotype that is very similar to highwire mutants – there is a dramatic overgrowth of synaptic termini, with a large increase in the number of synaptic boutons and branches. In addition, synaptic transmission is impaired in DFsn mutants. Genetic interactions between DFsn and highwire mutants indicate that DFsn and Highwire collaborate to restrain synaptic terminal growth. Finally, DFsn regulates the levels of the Wallenda/DLK kinase, and wallenda is necessary for DFsn-dependent synaptic terminal overgrowth. Conclusion The F-box protein DFsn binds the ubiquitin ligase Highwire and is required to down-regulate the levels of the Wallenda/DLK kinase and restrain synaptic terminal growth. We propose that DFsn and Highwire participate in an evolutionarily conserved ubiquitin ligase complex whose substrates regulate the structure and function of synapses.

Abstract

Synaptic signaling involves the release of neurotransmitter from presynaptic active zones (AZs). Proteins that regulate vesicle exocytosis cluster at AZs, composing the cytomatrix at the active zone (CAZ). At the Drosophila neuromuscular junction (NMJ), the small GTPase Rab3 controls the distribution of CAZ proteins across release sites, thereby regulating the efficacy of individual AZs. Here we identify Rab3-GEF as a second protein that acts in conjunction with Rab3 to control AZ protein composition. At rab3-GEF mutant NMJs, Bruchpilot (Brp) and Ca2+ channels are enriched at a subset of AZs, leaving the remaining sites devoid of key CAZ components in a manner that is indistinguishable from rab3 mutant NMJs. As the Drosophila homologue of mammalian DENN/MADD and Caenorhabditis elegans AEX-3, Rab3-GEF is a guanine nucleotide exchange factor (GEF) for Rab3 that stimulates GDP to GTP exchange. Mechanistic studies reveal that although Rab3 and Rab3-GEF act within the same mechanism to control AZ development, Rab3-GEF is involved in multiple roles. We show that Rab3-GEF is required for transport of Rab3. However, the synaptic phenotype in the rab3-GEF mutant cannot be fully explained by defective transport and loss of GEF activity. A transgenically expressed GTP-locked variant of Rab3 accumulates at the NMJ at wild-type levels and fully rescues the rab3 mutant but is unable to rescue the rab3-GEF mutant. Our results suggest that although Rab3-GEF acts upstream of Rab3 to control Rab3 localization and likely GTP-binding, it also acts downstream to regulate CAZ development, potentially as a Rab3 effector at the synapse.
