Abstract

Ruminant livestock are important sources of human food and global greenhouse gas emissions. Feed degradation and methane formation by ruminants rely on metabolic interactions between rumen microbes and affect ruminant productivity. Rumen and camelid foregut microbial community composition was determined in 742 samples from 32 animal species and 35 countries, to estimate if this was influenced by diet, host species, or geography. Similar bacteria and archaea dominated in nearly all samples, while protozoal communities were more variable. The dominant bacteria are poorly characterised, but the methanogenic archaea are better known and highly conserved across the world. This universality and limited diversity could make it possible to mitigate methane emissions by developing strategies that target the few dominant methanogens. Differences in microbial community compositions were predominantly attributable to diet, with the host being less influential. There were few strong co-occurrence patterns between microbes, suggesting that major metabolic interactions are non-selective rather than specific.

Abstract

A 16S rRNA and methyl coenzyme reductase subunit A (mcrA) gene clone libraries was used to study the composition of the methanogenic archaeal community in the foregut contents of tammar wallabies (Macropus eugenii). The community was observed to show presence of methanogens belonging to the Methanobacteriales and a well-supported cluster of uncultivated archaeon sequences similar to those observed in ovine and bovine rumens.

Abstract

A vaccine based on the key methanogens present in the sheep intended to receive the vaccine was formulated to determine if a targeted vaccine could be used to decrease the methane output of sheep. The results indicated the existence of correlation between 16S rRNA gene sequence relatedness and cross-reactivity for the methanogens and much slower adaptation of the rumen methanogen populations to differing diets called for further studies to properly assess the acclimatization period for the methanogenic archaea.

Abstract

16S rRNA gene libraries were used for examining the molecular diversity of rumen methanogens in feedlot cattle and the composition of the methanogen populations from Ontario and Prince Edward Island. Findings reveal the identification of nine possible new species from the two clone libraries.

Abstract

A study was conducted on the design of PCR and real-time PCR primers for the 18S rRNA gene of rumen protozoa, and their specificities were also tested against a range of rumen microbes and protozoal groups. It was observed that the numbers of Entodinium cells calculated using real-time PCR were higher than microscopic estimations.
