Norovirus infection can cause gastrointestinal disease in humans. Development of therapies and vaccines against norovirus have been limited by the lack of a suitable and reliable animal model. Here we established rhesus macaques as an animal model for human norovirus infection. We show that rhesus macaques are susceptible to oral infection with human noroviruses from two different genogroups. Variation in duration of virus shedding (days to weeks) between animals, evolution of the virus over the time of infection, induction of virus-specific adaptive immune responses, susceptibility to reinfection and preferential replication of norovirus in the jejunum of rhesus macaques was similar to infection reported in humans. We found minor pathological signs and changes in epithelial cell surface glycosylation patterns in the small intestine during infection. Detection of viral protein and RNA in intestinal biopsies confirmed the presence of the virus in chromogranin A-expressing epithelial cells, as it does in humans. Thus, rhesus macaques are a promising non-human primate model to evaluate vaccines and therapeutics against norovirus disease., (© 2024. Priority effects shape the assembly of free-living communities and host-associated communities. However, the current literature does not fully incorporate two features of host-symbiont interactions, correlated host responses to multiple symbionts and ontogenetic changes in host responses to symbionts, leading to an incomplete picture of the role of priority effects in host-associated communities. We factorially manipulated the inoculation timing of two plant symbionts (mutualistic rhizobia bacteria and parasitic root-knot nematodes) and tested how host age at arrival, arrival order, and arrival synchrony affected symbiont colonization success in the model legume Medicago truncatula. We found that host age, arrival order, and arrival synchrony significantly affected colonization of one or both symbionts. Host age at arrival only affected nematodes but not rhizobia: younger plants were more heavily infected than older plants. By contrast, arrival order only affected rhizobia but not nematodes: plants formed more rhizobia nodules when rhizobia arrived before nematodes. Finally, synchronous arrival decreased colonization both symbionts, an effect that depended on host age. Our results demonstrate that priority effects compromise the host's ability to control colonization by two major symbionts and suggest that the role of correlated host responses and host ontogeny in the assembly of host-associated communities deserve further attention., (© 2022 The Ecological Society of America.)

Premise: We developed a novel low-cost method to visually phenotype belowground structures in the plant rhizosphere. We devised the method introduced here to address the difficulties encountered growing plants in seed germination pouches for long-term experiments and the high cost of other mini-rhizotron alternatives., Methods and Results: The method described here took inspiration from homemade ant farms commonly used as an educational tool in elementary schools. Using compact disc (CD) cases, we developed mini-rhizotrons for use in the field and laboratory using the burclover Medicago lupulina ., Conclusions: Our method combines the benefits of pots and germination pouches. In CD mini-rhizotrons, plants grew significantly larger than in germination pouches, and unlike pots, it is possible to measure roots without destructive sampling. Our protocol is a cheaper, widely available alternative to more destructive methods, which could facilitate the study of belowground phenotypes and processes by scientists with fewer resources., (© 2020 Cassidy et al. Motivation: The BioTIME database contains raw data on species identities and abundances in ecological assemblages through time. These data enable users to calculate temporal trends in biodiversity within and amongst assemblages using a broad range of metrics. BioTIME is being developed as a community-led open-source database of biodiversity time series. Our goal is to accelerate and facilitate quantitative analysis of temporal patterns of biodiversity in the Anthropocene., Main Types of Variables Included: The database contains 8,777,413 species abundance records, from assemblages consistently sampled for a minimum of 2 years, which need not necessarily be consecutive. In addition, the database contains metadata relating to sampling methodology and contextual information about each record., Spatial Location and Grain: BioTIME is a global database of 547,161 unique sampling locations spanning the marine, freshwater and terrestrial realms. Grain size varies across datasets from 0.0000000158 km 2 (158 cm 2 ) to 100 km 2 (1,000,000,000,000 cm 2 )., Time Period and Grain: BioTIME records span from 1874 to 2016. The regrowing forests of eastern North America have been an important global C sink over the past 100+ years, but many are now transitioning into late succession. The consequences of this transition are unclear due to uncertainty around the C dynamics of old- growth forests. Canopy structural complexity (CSC) has been shown to be an important source of variability in C dynamics in younger forests (e.g., in productivity and resilience to disturbance), but its role in late-successional forests has not been widely addressed. We investigated patterns of CSC in two old-growth forest landscapes in the Upper Peninsula of Michigan, USA, to assess factors associated with CSC and its influence on productivity and disturbance resilience (to moderate-severity windstorm). CSC was quantified using a portable below-canopy LiDAR (PCL) system in 65 plots that also had long-term (50-70+ years). inventory data, which were used to quantify aboveground net primary productivity (ANPP), disturbance history, and stand characteristics. We found high and variable CSC relative to younger forests across a suite of PCL-derived metrics. Variation in CSC was driven by species composition and size structure, rather than disturbance history or site characteristics. Recent moderate severity wind disturbance decreased plot-scale CSC, but increased stand-scale variation in CSC. The strong positive correlation between CSC and productivity illustrated in younger forests was not present in undisturbed portions of these late-successional ecosystems. Moderate severity disturbance appeared to reestablish the positive link between CSC and productivity, but this relationship was scale and severity dependent. A positive CSC-productivity relationship was evident at the plot scale with low-severity, dispersed disturbance, but only at a patch scale in more severely disturbed areas. CSC does not appear to strongly correlate With variation in productivity in undisturbed old-growth forests, but may play a very important (and scale/severity-dependent) role in their response to disturbance. Understanding potential, drivers and consequences of CSC in late-successional forests will inform management focused on promoting complexity and old-growth conditions, and illustrate potential inipacts of such treatments on regional C dynamics.

Trends in living aboveground biomass and inputs to the pool of coarse woody debris (CWD) in an undisturbed, old-growth hemlock-northern hardwood forest in northern MI were estimated from multi-decade observations of permanent plots. Growth and demographic data from seven plot censuses over 47 years (1962-2009), combined with one-time measurement of CWD pools, help assess biomass/carbon status of this landscape. Are trends consistent with traditional notions of late-successional forests as equilibrial ecosystems? Specifically, do biomass pools and CWD inputs show consistent long-term trends and relationships, and can living and dead biomass pools and trends be related to forest composition and history? Aboveground living biomass densities, estimated using standard allometric relationships, range from 360-450 Mg/ha among sampled stands and types; these values are among the highest recorded for northeastern North American forests. Biomass densities showed significant decade-scale variation, but no consistent trends over the full study period (one stand, originating following an 1830 fire, showed an aggrading trend during the first 25 years of the study). Even though total above-ground biomass pools are neither increasing nor decreasing, they have been increasingly dominated, over the full study period, by very large (>70 cm dbh) stems and by the most shade-tolerant species (Acer saccharum and Tsuga canadensis). CWD pools measured in 2007 averaged 151 m(3)/ha, with highest values in Acer-dominated stands. Snag densities averaged 27/ha, but varied nearly ten-fold with canopy composition (highest in Tsuga-dominated stands, lowest in Acer-dominated); snags constituted 10-50% of CWD biomass. Annualized CWD inputs from tree mortality over the full study period averaged 1.9-3.2 Mg/ha/yr, depending on stand and species composition. CWD input rates tended to increase over the course of the study. Input rates may be expected to increase over longer-term observations because, (a) living biomass is increasingly dominated by very large trees whose dead trunks have longer residence time in the CWD pool, and (b) infrequent major disturbances, thought to be important in the dynamics of these forests, have not occurred during the study period but would be expected to produce major, episodic pulses in CWD input. Few fragments of old-growth cool-temperate forests remain, but such forests can constitute a very large carbon pool on a per-area basis. The carbon sink/source status of these forests remains unclear. While aboveground living biomass at this study site shows no strong aggrading or declining trend over the last half-century, this remains a modest span in the innate time-scale of late-successional forest. The effects of rare disturbances, long-term shifts in composition and size structure, and changes in soil carbon and CWD pools may all influence long-term carbon status.

Recent global warming is acting across marine, freshwater, and terrestrial ecosystems to favor species adapted to warmer conditions and/or reduce the abundance of cold-adapted organisms (i.e., "thermophilization" of communities). Lack of community responses to increased temperature, however, has also been reported for several taxa and regions, suggesting that "climatic lags" may be frequent. Here we show that microclimatic effects brought about by forest canopy closure can buffer biotic responses to macroclimate warming, thus explaining an apparent climatic lag. Using data from 1,409 vegetation plots in European and North American temperate forests, each surveyed at least twice over an interval of 12-67 y, we document significant thermophilization of ground-layer plant communities. These changes reflect concurrent declines in species adapted to cooler conditions and increases in species adapted to warmer conditions. However, thermophilization, particularly the increase of warm-adapted species, is attenuated in forests whose canopies have become denser, probably reflecting cooler growing-season ground temperatures via increased shading. As standing stocks of trees have increased in many temperate forests in recent decades, local microclimatic effects may commonly be moderating the impacts of macroclimate warming on forest understories. Conversely, increases in harvesting woody biomass--e.g., for bioenergy--may open forest canopies and accelerate thermophilization of temperate forest biodiversity.

Objective: A favorable incidence and severity of graft-vs-host disease is observed in patients transplanted with banked, unrelated, HLA-mismatched umbilical cord blood (UCB) grafts, while the incidence of malignant relapse remains low. CTLA-4 mediates negative T-cell signaling and may contribute to the development of allogeneic tolerance. In this study, we compared protein and mRNA expression of CTLA-4 in stimulated UCB and adult peripheral blood T cells., Materials and Methods: T cells were isolated from UCB and adult peripheral blood and stimulated with anti-CD3 and anti-CD28 monoclonal antibodies. Cells were immunostained and analyzed by flow cytometry for both surface and intracellular expression of CTLA-4 in the presence and absence of cyclosporin A, and kinetics of CTLA-4 expression compared. CTLA-4 mRNA expression was measured using quantitative real-time polymerase chain reaction. NFAT1 protein levels were measured by Western blot analysis., Results: These studies demonstrate reduced surface and intracellular expression of CTLA-4 in stimulated UCB T cells compared to adult controls. Furthermore, reduced CTLA-4 protein expression in UCB T cells was noted to be in part transcriptionally regulated, as CTLA-4 mRNA levels also were significantly lower. Reduced CLTA-4 expression by UCB T cells followed the kinetics of delayed and reduced expression of the transcription factor NFAT1 by UCB T lymphocytes during primary stimulation. Moreover, cyclosporin A, which is known to modulate NFAT activation, reduced CTLA-4 protein expression in adult and UCB T cells., Conclusion: Reduced expression of the key regulatory proteins CTLA-4 and NFAT-1 may contribute to favorable UCB T lymphocyte allogeneic responses.

Umbilical cord blood (UCB) stem cells from related and unrelated allogeneic donors have emerged as novel treatment for patients with hematologic malignancies. The incidence and severity of acute graft-versus-host disease (GVHD) after UCB transplantation compares favorably with that observed in recipients of matched unrelated donor allogeneic grafts, but remains a major cause of morbidity and mortality. It has been shown that stimulated lymphocytes from UCB have reduced production of cytokines including interferon-gamma (IFN-gamma) and tumor necrosis factor-alpha (TNF-alpha), which play a role in GVHD pathophysiology. We investigated the molecular mechanisms underlying this reduced cytokine production by analyzing expression of nuclear factor of activated T cells-1 (NFAT1) in UCB T cells. We detected no constitutive expression of NFAT1 protein in unstimulated UCB T cells compared with adult T cells. Moreover, although NFAT1 expression in UCB T cells was upregulated after prolonged (40 hours) T-cell stimulation, it was only partially upregulated when compared with adult controls. Our observation of minimal NFAT1 expression after stimulation correlated with reduced cytoplasmic IFN-gamma and TNF-alpha production in UCB T cells studied simultaneously. Reduced NFAT1 expression may blunt amplification of donor UCB T-cell alloresponsiveness against recipient antigens, thereby potentially limiting GVHD incidence and severity after allogeneic UCB transplantation.
