Abstract

During the last decade there has been increasing interest in the use of lasers in place of electrical systems to ignite various pyrotechnic and explosive materials. The principal driving force for this work was the requirement for safer energetic components which would be insensitive to electrostatic and electromagnetic radiation. In the last few years this research has accelerated since the basic concepts have proven viable. At the present time it is appropriate to shift the research emphasis in laser initiation from the scientific arena--whether it can be done--to the engineering realm--how it can be put into actual practice in the field. Laser initiation research and development at EG&G Mound was in three principal areas: (1) laser/energetic material interactions; (2) development of novel processing techniques for fabricating hermetic (helium leak rate of less than 1 x 10(exp -8) cu cm/s) laser components; and (3) evaluation and testing of laser-ignited components. Research in these three areas has resulted in the development of high quality, hermetic, laser initiated components. Examples are presented which demonstrate the practicality of fabricating hermetic, laser initiated explosive or pyrotechnic components that can be used in the next generation of ignitors, actuators, and detonators.

Abstract

Apolipoprotein E is a monomeric protein secreted by the liver and responsible for the transport of plasma cholesterol and triglycerides. The APOE gene encodes 3 isoforms epsilon 4, epsilon 3 and epsilon 2 with APOE epsilon 4 associated with higher plasma cholesterol levels and increased pathogenesis in several infectious diseases (HIV, HSV). Given that cholesterol is an important nutrient for malaria parasites, we examined whether APOE epsilon 4 was a risk factor for Plasmodium infection, in terms of prevalence or parasite density. A cross sectional survey was performed in 508 children aged 1 to 12 years in Gabon during the wet season. Children were screened for Plasmodium spp. infection, APOE and hemoglobin S (HbS) polymorphisms. Median parasite densities were significantly higher in APOE epsilon 4 children for Plasmodium spp. densities compared to non-APOE epsilon 4 children. When stratified for HbS polymorphisms, median Plasmodium spp. densities were significantly higher in HbAA children if they had an APOE epsilon 4 allele compared to those without an APOE epsilon 4 allele. When considering non-APOE epsilon 4 children, there was no quantitative reduction of Plasmodium spp. parasite densities for HbAS compared to HbAA phenotypes. No influence of APOE epsilon 4 on successful Plasmodium liver cell invasion was detected by multiplicity of infection. These results show that the APOE epsilon 4 allele is associated with higher median malaria parasite densities in children likely due to the importance of cholesterol availability to parasite growth and replication. Results suggest an epistatic interaction between APOE and HbS genes such that sickle cell trait only had an effect on parasite density in APOE epsilon 4 children. This suggests a linked pathway of regulation of parasite density involving expression of these genes. These findings have significance for understanding host determinants of regulation of malaria parasite density, the design of clinical trials as well as studies of co-infection with Plasmodium and other pathogens.

Abstract

Objective:To conduct a cost analysis of injection laryngoplasty performed in the operating theatre under local anaesthesia and general anaesthesia.Methods:The retrospective study included patients who had undergone injection laryngoplasty as day cases between July 2013 and March 2016. Cost data were obtained, along with patient demographics, anaesthetic details, type of injectant, American Society of Anesthesiologists score, length of stay, total operating theatre time and surgeon procedure time.Results:A total of 20 cases (general anaesthesia = 6, local anaesthesia = 14) were included in the cost analysis. The mean total cost under general anaesthesia (AU$2865.96 ± 756.29) was significantly higher than that under local anaesthesia (AU$1731.61 ± 290.29) (p < 0.001). The mean operating theatre time, surgeon procedure time and length of stay were all significantly lower under local anaesthesia compared to general anaesthesia. Time variables such as operating theatre time and length of stay were the most significant predictors of the total costs.Conclusion:Procedures performed under local anaesthesia in the operating theatre are associated with shorter operating theatre time and length of stay in the hospital, and provide significant cost savings. Abstract

Objective: To identify laryngeal mRNA gene changes in patients with laryngopharyngeal reflux (LPR). Method: Laryngeal biopsies from non‐smoking LPR patients (n=10; Reflux Symptom Index (RSI) >12 and a Reflux Finding Score (RFS) >6) and controls (n=9; RSI <12 and RFS <6) were collected from four subsites (true vocal cord, false vocal cord, medial arytenoid and posterior commissure) of the larynx. qRT‐PCR analyses were conducted on 20 reflux‐ and inflammation‐related genes, including interleukins 6 and 8, cytokeratins 8 and 14, mucin genes MUC1, MUC2, MUC3B, MUC4, MUC5B, MUC6 and MUC7 and carbonic anhydrase III. Statistical analysis (Mann‐Whitney U test) compared gene expression levels between LPR and control groups at each subsite. Results: Site‐specific differences in squamous metaplasia and gene expression were noted in LPR patients, with the majority present in the medial arytenoid region. Significant.differences were noted in genes related to mucosal defence and inflammation, including CRNN, CD1d, TGFβ‐1, MUC2, MUC5B and CDH1. Conclusion: Whilst the posterior commissure is commonly identified as the area demonstrating the most significant macroscopic change in LPR, the histological changes and genes assessed here showed more pronounced LPR associated differences in the medial arytenoid. We identified differences in expression of mucin genes, cytokeratin‐14 and molecular markers of inflammation. Whilst some of these changes may be metaplasia‐related, further evaluation of the mRNA expression of these genes may provide a useful biomarker panel for diagnosis and therapeutic monitoring of LPR. 31. The solution of the Elrod algorithm for a dynamically loaded journal bearing using multigrid techniques

Abstract

A numerical solution to a theoretical model of vapor cavitation in a dynamically loaded journal bearing is developed utilizing a multigrid iteration technique. The method is compared with a noniterative approach in terms of computational time and accuracy. The computational model is based on the Elrod algorithm, a control volume approach to the Reynolds equation which mimics the Jakobsson-Floberg and Olsson cavitation theory. Besides accounting for a moving cavitation boundary and conservation of mass at the boundary, it also conserves mass within the cavitated region via a smeared mass or striated flow extending to both surfaces in the film gap. The mixed nature of the equations (parabolic in the full film zone and hyperbolic in the cavitated zone) coupled with the dynamic aspects of the problem create interesting difficulties for the present solution approach. Emphasis is placed on the methods found to eliminate solution instabilities. Excellent results are obtained for both accuracy and reduction of computational time.

Abstract

Interspecific interactions between fungi that colonize stumps ofPicea sitchensisin Scotland were tested in dual cultures on Norkrans agar, spruce sawdust and in autoclaved blocks cut from roots. Isolates were ranked according to competitive ability on the different media, based on their ability to overgrow competitors or to form deadlock interactions. On the defined medium,Phaeolus schweinitziiwas the species most able to overgrow competitors, followed byStereum sanguinolentumandHeterobasidion annosum;Resinicium bicolorwas the least able to overgrow competitors on this medium. By contrast,R. bicolorwas the most competitive on spruce sawdust medium. Deadlocking interactions were formed most often in dual cultures on Norkrans agar. Observation of hyphal interactions on Norkrans agar under the microscope identified several different response types including growth of thin hyphae compared to control cultures, hyphal coiling, vacuolation of hyphae, hyphal lysis of one competitor and deposition of crystals in the agar. Hymenomycetes caused varying amounts of decay in autoclaved root blocks.Resinicium bicolorwas able to replace other species in most co-inoculations.Stereum sanguinolentumappeared to be the least competitive species in root block inoculations, being replaced byMelanotus proteusandR. bicolor, although interactions withH. annosumvaried widely. These results indicate that substrate has a marked effect on interspecific fungal interactions, with wood-based, particularly intact woody tissues closely matching competitive behaviour displayed in the field. 49. Perfluorooctyl Bromide Has Limited Membrane Solubility and Is Located at the Bilayer Center. Locating Small Molecules in Lipid Bilayers through Paramagnetic Enhancements of NMR Relaxation

Abstract

There is considerable interest in the use of perfluorocarbons as oxygen carriers in clinical settings; however, little is known regarding the molecular interactions made by these apolar compounds with biological membranes or their effect on membrane structure. NMR spectroscopy was used to investigate the interaction of perfluorooctyl bromide (PFOB) with 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) bilayers. 19F NMR spectra demonstrate that PFOB partitions into POPC bilayers but that it saturates at a remarkably low membrane concentration of approximately 2 mol %. 19F chemical shifts indicate that this membrane-bound PFOB experiences a local environment similar in polarity to that of hexane, suggesting that the compound resides within the hydrocarbon core of the lipid bilayer. This hydrocarbon location was refined by measuring paramagnetic enhancements of 19F nuclear relaxation for membrane-bound PFOB produced by Gd3+ and O2. The data clearly localize PFOB to the center of the membrane hydrocarbon and show how paramagnetic enhancements of nuclear relaxation produced by O2 may be used to localize small molecules within bilayers. 2H and 31P NMR experiments demonstrate that PFOB produces no significant changes in either acyl chain or headgroup structure even at saturating membrane concentrations.
