BACKGROUND:The response of virgin females of the legume pest Acanthoscelides obtectus (Coleoptera: Bruchidae) to headspace extracts of volatiles collected from flowers of a nectar plant, Daucus carota, was investigated using behaviour (four-arm olfactometry) and coupled gas chromatography–electroantennography (GC-EAG). RESULTS: Odours from inflorescences were significantly more attractive to virgin female beetles than clean air. Similarly, a sample of volatile organic compounds (VOCs) collected by air entrainment (dynamic headspace collection) was more attractive to beetles than a solvent control. In coupled GC-EAG experiments with beetle antennae and the VOC extract, six components howed AG activity. Using coupled GC–mass spectrometry (GC–MS) and GC peak enhancement with authentic standards, the components were identified as 𝜶-pinene (S:R 16:1), sabinene, myrcene, limonene (S:R 1:3), terpinolene and (S)-bornyl acetate. Females preferred the synthetic blend of D. carota EAG-active volatiles to the solvent control in bioassays. When compared directly, odours of D. carota inflorescences elicited stronger positive behaviour than the synthetic blend. CONCLUSION: This is the first report of behaviourally active volatiles linked to pollen location for A. obtectus, and development of the six-component blend is being pursued, which could underpin the design of semiochemical-based field management approaches against this major pest of stored products.

BACKGROUNDAgrilus bigutattus (Fabricius) is a forest pest of increasing importance in the United Kingdom. The larvae damage weakened native oaks and are thought to contribute to premature tree death. Suspected links with acute oak decline (AOD) are not yet confirmed, but AOD-predisposed trees appear to become more susceptible to A. biguttatus attack. Thus, management may be necessary for control of this insect. To explore the possibility of monitoring beetle populations by baited traps, the host tree volatiles regulating A. biguttatus-oak interactions were studied. RESULTSBiologically active volatile organic compounds in dynamic headspace extracts of oak foliage and bark were identified initially by coupled gas chromatography-electroantennography (GC-EAG) and GC-mass spectrometry (GC-MS), and the structures were confirmed by GC coinjection with authentic compounds. Of two synthetic blends of these compounds comprising the active leaf volatiles, the simpler one containing three components evoked strongly positive behavioural responses in four-arm olfactometer tests with virgin females and males, although fresh leaf material was more efficient than the blend. The other blend, comprising a five-component mixture made up of bark volatiles, proved to be as behaviourally active for gravid females as bark tissue. CONCLUSIONSThese initial results on A. biguttatus chemical ecology reveal aspects of the role of attractive tree volatiles in the host-finding of beetles and underpin the development of semiochemically based surveillance strategies for this forest insect. A synthetic biology approach to the rational design of analogues of olfactory ligands by providing unnatural substrates for the enzyme synthesising (S)-germacrene D, an olfactory ligand acting as a plant derived insect repellent, to produce novel ligands is described as a viable alternative to largely unsuccessful ligand docking studies. (S)-14,15-Dimethylgermacrene D shows an unexpected reversal in behavioural activity.

We have developed an integrated pest management strategy (IPM) for pollen beetles in winter oilseed rape (OSR) based on risk assessment, monitoring and alternative crop management that can be used as a framework by growers and crop consultants to manage pollen beetles with reduced insecticide inputs - and the confidence to do so. This will prolong insecticide life by reducing selection for resistance, reduce environmental impacts and contribute towards the sustainability and profitability of OSR in the UK. One of the major limitations to the use of action thresholds is that proper monitoring of the populations is time consuming and has to be conducted over a prolonged period. To encourage and facilitate their use, we tested and developed tools to improve risk assessment and monitoring. We conducted a pollen beetle monitoring study over 4 years in 178 OSR crops across the UK. Pollen beetles were sampled using sticky traps and plant sampling along transects in the crop. The data were used to help test a decision support system (DSS) for pollen beetles and to develop a monitoring trap. proPlant Expert is a DSS available in mainland Europe that uses a model of pollen beetle immigration and local meteorological data to forecast the start and end of pollen beetle immigration into the crop and main risk periods and advises when to monitor. We tested the model under UK conditions using data from our study and compared monitoring advice with the current advice system on the CropMonitor website (advises monitoring when the crop is at green-yellow bud stage and temperature >15°C). Both performed reassuringly well in prompting monitoring that would detect breaches of spray thresholds. However there were considerable reductions provided by proPlant in the need for consultation of the system (30%) and advised monitoring days (34-53%) in comparison with current advice. Use of the proPlant DSS could therefore focus monitoring effort to when it is most needed. It could also help to reduce unnecessary sprays in cases where beetle numbers are approaching threshold but consultation of the system returns a poor immigration risk forecast or an immigration complete result. The proPlant tool is now freely available to growers and crop consultants in the UK via the Bayer CropScience website. A monitoring trap for pollen beetles would help to more easily and accurately identify when spray thresholds have been breached than monitoring plants in the crop. We developed a baited monitoring trap for pollen beetles which will be commercially available from Oecos. The trap comprises a yellow sticky card mounted at 45°, baited with phenylacetaldehyde, a floral volatile produced naturally by several plant species. Unfortunately using data from our study we were unable to calibrate the trap catch to a given action threshold expressed as the number of beetles per plant using a simple linear relationship. However, the monitoring trap still has value for risk assessment, especially if used together with DSS. We tested the potential of turnip rape (TR) trap crops, planted as borders to the main OSR crop to reduce pollen beetle numbers in a field scale experiment conducted over three years on two sites. We found evidence that the strategy worked well in some years, but not others. This tactic is probably practically and economically worthwhile only for organic growers.

Plants can produce compounds which act as semiochemicals, that is, signals modifying the development or behaviour of other organisms without having direct physiological activity. Among such semiochemicals are plant stress signals associated with the induction of defence systems, and these may include phytopheromones that naturally influence plant development. It is well known that plant-derived semiochemicals can be exploited by colonizing organisms, particularly pathogens and insect pests. Recently proposed external signals not yet proven as natural phytopheromones are nitric oxide and the volatile methyl esters of jasmonic and salicylic acids. Since it is now possible to use sophisticated electrophysiological techniques to investigate insect interactions with prospective phytopheromones, the detection and characterization of signalling systems has been made much easier and can provide a molecular characterization of signals that are active beyond the insects themselves. In addition to the advances these studies have brought to the understanding of plant/insect and plant/plant interactions, plant signals are potentially valuable in the regulation of gene expression for improved or alternative approaches to crop protection or for other developmental processes in plants.

The present situation and future prospects for the use of semiochemicals for insect control is reviewed, with particular reference to the work being carried out at IACR-Rothamsted. The techniques used to identify pheromones and other semiochemicals, and the types of compound found in various insect classes, are described. The effects of such compounds on pests, their predators and other members of the ecosystem are considered in relation to the development of control strategies such as 'push-pull' or stimulo-deterrent diversionary strategies (SDDS). Male-specific volatile components, released by the dried bean beetle, Acanthoscelides obtectus, were identified as methyl (E,R)-2,4,5-tetradecatrienoate, methyl (2E,4Z,7Z)-2,4,7-decatrienoate, methyl (2E,4Z)-2,4-decadienoate, octadecanal and the sesquiterpenes (3Z,6E)- and (3E,6E)-alpha-farnesene. In olfactometer bioassays, pure methyl (E, R)-2,4,5-tetradecatrienoate was only weakly attractive to unmated females. However, a blend of the six identified compounds released in physiologically relevant ratios and doses proved to be as active as headspace odours collected from live males.

BACKGROUND: The banana weevil, Cosmopolites sordidus, has been frequently cited as the most challenging constraint to banana and plantain production, particularly in small-scale (smallholder) farming. For the development of a new, low-cost weevil management technology based on attractive host plant material, we previously identified (2R,5S)-theaspirane as the active component of attractive senesced banana leaves. In this new study, we used behavioural (olfactometer) bioassays with adult weevils to compare the attractiveness of four different developmental stages of banana leaves, ie. unfolding (pale green), matured green (deep green), matured yellowing and senesced, to determine which leaf developmental stage would be most appropriate for use in weevil management. We also investigated the attractiveness of senesced leaf extracts prepared using different solvents to determine which solvent would be most appropriate for local production of leaf extracts. Coupled gas chromatography-electroantennography (GC-EAG) was then used with adult weevils to confirm the presence of (2R,5S)-theaspirane in attractive leaf extracts.
