Post-imprinting modification of imprinted molecular cavities with a ‘redox probe’ for electrochemical sensing.

Catalytically active nanomaterials, in particular, nanozymes, are promising candidates for applications in biosensors due to their excellent catalytic activity, stability and cost-effective preparation. Nanozymes with peroxidase-like activities are prospective candidates for applications in biosensors. The purpose of the current work is to develop cholesterol oxidase-based amperometric bionanosensors using novel nanocomposites as peroxidase (HRP) mimetics. To select the most electroactive chemosensor on hydrogen peroxide, a wide range of nanomaterials were synthesized and characterized using cyclic voltammetry (CV) and chronoamperometry. Pt NPs were deposited on the surface of a glassy carbon electrode (GCE) in order to improve the conductivity and sensitivity of the nanocomposites. The most HRP-like active bi-metallic CuFe nanoparticles (nCuFe) were placed on a previously nano-platinized electrode, followed by conjugation of cholesterol oxidase (ChOx) in a cross-linking film formed by cysteamine and glutaraldehyde. The constructed nanostructured bioelectrode ChOx/nCuFe/nPt/GCE was characterized by CV and chronoamperometry in the presence of cholesterol. The bionanosensor (ChOx/nCuFe/nPt/GCE) shows a high sensitivity (3960 A·M−1·m−2) for cholesterol, a wide linear range (2–50 µM) and good storage stability at a low working potential (−0.25 V vs. Ag/AgCl/3 M KCl). The constructed bionanosensor was tested on a real serum sample. A detailed comparative analysis of the bioanalytical characteristics of the developed cholesterol bionanosensor and the known analogs is presented.

Recently, shot noise has been shown to be an inherent part of all charge transfer processes, leading to a practical limit of quantification of 2100 electrons (~0.34 fC) (Curr. Opin. Electrochem. 2020, 22, 170177). Attainable limits of quantification are made much larger by greater background currents and insufficient instrumentation, which restricts progress in sensing and single-entity applications. This limitation can be overcome by converting electrochemical charges into photons, which can be detected with much greater sensitivity, even down to a single-photon level. In this work, we demonstrate the use of fluorescence, induced through a closed-bipolar set-up, to monitor charge transfer processes below the detection limit of electrochemical workstations. During this process, the oxidation of ferrocenemethanol (FcMeOH) in one cell is used to concurrently drive the oxidation of Amplex Red (AR), a fluorogenic redox molecule, in another cell. The spectroelectrochemistry of AR is investigated and new insights on the commonplace practise of using deprotonated glucose to limit AR photooxidation are presented. The closed-bipolar set-up was used to produce fluorescent signals corresponding to the steady-state voltammetry of FcMeOH on a microelectrode. Chronopotentiometry is then used to show a linear relationship between the charge passed through FcMeOH oxidation and the integrated AR fluorescence signal. The sensitivity of the measurements obtained at different timescales varies between 2200 - 500 electrons per detected photon. The electrochemical detection limit is approached using a diluted FcMeOH solution in which no current signal is observed. Nevertheless, a fluorescence signal corresponding to FcMeOH oxidation is still seen, and detection of charges down to 300 fC is demonstrated.

The aim of the current research is to design alcohol oxidase-based amperometric biosensors (ABSs) using hybrid metallic nanoparticles as artificial peroxidases (PO) or PO-like nanozymes (NZs). A lot of metallic PO-like NZs were synthesized and tested with respect to their ability to substitute natural PO in solution and on amperometric electrode. The most effective PO mimetics were coupled with alcohol oxidase (AOX) on graphite electrodes (GEs) and characterized. Two types of modified GEs, namely, the AOX/nAuCePt/GE and the AOX/nFePtAu/GE show the highest sensitivities to ethanol (2600 A⋅M

International audience; Actuators controlled by external stimuli have received a lot of attention in recent years. Herein we report a polymer based dual stimuli actuator, triggered by light and an electric field. This allows a better control of actuation, enlarging the field of potential applications, like e.g. in the frame of soft robotics. The actuator is composed of polypyrrole (PPy) and TiO2 modified with methylene blue (MB). In aqueous solution, the resulting freestanding hybrid film shows reversible actuation due to the synergy of light and an applied electric field. Illumination with light produces electron-hole pairs in the TiO2 layer, which are shuttled to the opposite ends of the actuator by the potential gradient present in solution. This results in electrochemical oxidation and reduction reactions at the two extremities and consequently in site selective swelling of the polymer, which finally leads to a controlled motion of the actuator, following the principles of logic gate operations. Such synergistically induced switching allows developing original actuation schemes for performing complex mechanical tasks triggered by more than one stimulus.

π-Conjugated macrocycles are molecules with unique properties that are increasingly exploited for applications and the question of whether they can sustain global aromatic or antiaromatic ring currents is particularly intriguing. However, there are only a small number of experimental studies that investigate how the properties of π-conjugated macrocycles evolve with systematic structural changes. Here, we present such a systematic experimental study of a set of [2.2.2.2]cyclophanetetraenes, all with formally Hückel antiaromatic ground states, and combine it with an in-depth computational analysis. The study reveals the central role of local and global aromaticity for rationalizing the observed optoelectronic properties, ranging from extremely large Stokes shifts of up to 1.6 eV to reversible fourfold reduction, a highly useful feature for charge storage/accumulation applications. A recently developed method for the visualization of chemical shielding tensors (VIST) is applied to provide unique insight into local and global ring currents occurring in different planes along the macrocycle. Conformational changes as a result of the structural variations can further explain some of the observations. The study contributes to the development of structure–property relationships and molecular design guidelines and will help to understand, rationalize, and predict the properties of other π-conjugated macrocycles. It will also assist in the design of macrocycle-based supramolecular elements with defined properties., Insights into how the optoelectronic properties of π-conjugated macrocycles evolve as a result of systematic structural changes are presented, helping to develop structure–property relationships and molecular design guidelines.

Living systems that can spontaneously exhibit directional motion belong to diverse classes such as bacteria, sperm and plankton. They have fascinated scientists in recent years to design completely artificial or biohybrid mobile objects. Natural ingredients, like parts of plants, have been used to elaborate miniaturized dynamic objects, which can move when they are combined with other, non-natural, building blocks. Herein, we report that the precise structural tailoring of natural plant leaves allows generating a spatially predefined and confined release of oxygen gas, due to the conversion of carbon dioxide. This constitutes the driving force for generating motion, which is solely due to the respiration of leaves by photosynthesis. The rate of gas evolution can be fine-tuned by changing the light intensity and the leaf size, allowing ultimately to control the motility of objects with dimensions ranging from the millimeter to the micrometer scale.

Aromatic organic compounds can be used as electrode materials in rechargeable batteries and are expected to advance the development of both anode and cathode materials for sodium‐ion batteries (SIBs). However, most aromatic organic compounds assessed as anode materials in SIBs to date exhibit significant degradation issues under fast‐charge/discharge conditions and unsatisfying long‐term cycling performance. Now, a molecular design concept is presented for improving the stability of organic compounds for battery electrodes. The molecular design of the investigated compound, [2.2.2.2]paracyclophane‐1,9,17,25‐tetraene (PCT), can stabilize the neutral state by local aromaticity and the doubly reduced state by global aromaticity, resulting in an anode material with extraordinarily stable cycling performance and outstanding performance under fast‐charge/discharge conditions, demonstrating an exciting new path for the development of electrode materials for SIBs and other types of batteries., Aromatic stabilization of both states: A molecular design concept for highly stable organic battery electrode materials is presented using the conjugated macrocycle [2.2.2.2]paracyclophane‐1,9,17,25‐tetraene (PCT). Electrochemistry has a limit of detection imposed by the inherent shot-noise present in electrical currents [1]. As a result, the direct measurement of single-electron charge transfer processes is not feasible using electrochemical methods. However, luminescence can be measured with single photon resolution and the development of a method to efficiently convert an electrochemical signal to an optical one is expected to be a key step in overcoming the detection limit of electrochemistry [2]. An optical conversion technique based on closed bipolar electrochemistry has recently been demonstrated using large glassy carbon electrodes [3,4]. In this technique, which is illustrated in Fig. 1, any electrochemical process of interest occurring in one cell (the detection cell) can be used to induce either fluorescence or chemiluminescence in another cell (the reporting cell). Fluorogenic reporting reactions have the advantage that several photons can be emitted during the lifetime of each fluorophore, leading to signals with a greater intensity. However, the facile photooxidation of some fluorophores introduces the need for chemical reductants, which complicates the electrochemistry [5]. Conversely, the constant electron-to-photon relationship of chemiluminescence makes it an easier process for calculating the number of electrons passed in the detection cell, but also leads to a reduced signal intensity [6]. There is still much work needed to understand which reporting mechanism is more suited to sub-detection limit electrochemistry and how to reach the maximum sensitivity of single-electron charge transfer processes. In this work, we present our recent progress towards the measurement of sub-detection limit electrochemical processes via luminogenic reporting reactions. We compare the conversion mechanisms of reporting reactions which use either fluorescence or chemiluminescence, and we study the factors which can affect the sensitivity of the conversion, such as the presence of chemical reductants. Finally, we test the temporal resolution and sensitivity of optical conversion techniques in the measurement of single entity electrochemical processes. H2 O2 is a versatile chemical and can be generated by the oxygen reduction reaction (ORR) in proton donor solution in molecular solvents or room temperature ionic liquids (IL). We investigated this reaction at interfaces formed by eleven hydrophobic ILs and acidic aqueous solution as a proton source with decamethylferrocene (DMFc) as an electron donor. H2 O2 is generated in colorimetrically detectable amounts in biphasic systems formed by alkyl imidazolium hexafluorophosphate or tetraalkylammonium bis(trifluoromethylsulfonyl)imide ionic liquids. H2 O2 fluxes were estimated close to liquid|liquid interface by scanning electrochemical microscopy (SECM). Contrary to the interfaces formed by hydrophobic electrolyte solution in a molecular solvent, H2 O2 generation is followed by cation expulsion to the aqueous phase. Weak correlation between the H2 O2 flux and the difference between DMFc/DMFc+ redox potential and 2 electron ORR standard potential indicates kinetic control of the reaction.

Evolution of bacteria to selective chemical pressure (e.g. antibiotics) is well studied in contrast to the influence of physical stressors. Here we show that instantaneous physical stress in a homogeneous environment (without concentration gradient) induces fast adaptation of Escherichia coli. We exposed E. coli to a large number of collisions of around 105 per bacterium per second with sharp ZnO nanorods. The pressure exerted on the bacterial cell wall was up to 10 GPa and induced phenotype changes. The bacteria’s shape became more spherical, the density of their periplasm increased by around 15% and the average thickness of the cell wall by 30%. Such E. coli cells appeared almost as Gram-positive bacteria in the standard Gram staining. Additionally, we observed a combination of changes occurring at the genomic level (mutations identified in form of single nucleotide polymorphisms) and down-regulation of expression of 61 genes encoding proteins involved in β-oxidation of fatty acids, glycolysis, the citric acid cycle, as well as uptake of amino acids and enzyme cofactors. Thus, we show that bacteria undergo phenotypic changes upon instantaneous, acute physical stress without any obviously available time for gradual adaptation.

Due to the limited availability of noble metal catalysts, such as platinum, palladium, or gold, their substitution by more abundant elements is highly advisable. Considerably challenging is the controlled and reproducible synthesis of stable non-noble metallic nanostructures with accessible active sites. Here, we report a method of preparation of bare (ligand-free) Cu nanostructures from polycrystalline metal in a controlled manner. This procedure relies on heterogeneous localized electrorefining of polycrystalline Cu on indium tin oxide (ITO) and glassy carbon as model supports using scanning electrochemical microscopy (SECM). The morphology of nanostructures and thus their catalytic properties are tunable by adjusting the electrorefining parameters, i.e., the electrodeposition voltage, the translation rate of the metal source and the composition of the supporting electrolyte. The activity of the obtained materials towards the carbon dioxide reduction reaction (CO2RR), oxygen reduction reaction (ORR) in alkaline media and hydrogen evolution reaction (HER), is studied by feedback mode SECM. Spiky Cu nanostructures obtained at a high concentration of chloride ions exhibit enhanced electrocatalytic activity. Nanostructures deposited under high cathodic overpotentials possess a high surface-to-volume ratio with a large number of catalytic sites active towards the reversible CO2RR and ORR. The CO2RR yields easily electrooxidizable compounds – formic acid and carbon monoxide. The HER seems to occur efficiently at the crystallographic facets of Cu nanostructures electrodeposited under mild polarization.

Major difficulties in nanoscale scanning electrochemical microscopy (SECM), resulting from the short tip-to-sample distance, are tilt misalignment and vertical thermal drift. This renders constant height mode nanoscale electrochemical imaging experimentally problematic. Nanoscale SECM also requires application of reliable nanoprobes. Although SECM nanotips are commercially available, they are rather disposable, fragile and their shelf-life is limited. Many SECM laboratories employ self-prepared nanoprobes, fabricated using advanced apparatus. In this paper, we employ a hopping mode approach to nanoscale electrochemical imaging, which resolves the mentioned common difficulties. The applied platinum nanoelectrodes, insulated with borosilicate glass, were prepared using affordable equipment and materials. They were applied for nanoscale mapping of redox mediator regeneration at a model interdigitated microsensor, composed of Pt microband electrodes, in a solution of ferrocene in viscous hydrophobic room temperature ionic liquid (RTIL) – 1-butyl-3-methylimidazoliumbis(trifluoro-methylsulfonyl)imide (C4mim N(Tf)2). The presented nanosensors were also characterized with cyclic voltammetry in aqueous electrolyte containing hexamineruthenium (III) chloride as redox mediator.

A new possibility for the formation of macroscopic and photoactive structures from zinc oxide nanocrystals is described. Photoactive freely suspended and free-standing films of macroscopic area (up to few square millimeters) and submicrometer thickness (up to several hundreds of nanometers) composed of carboxylate ligand-coated zinc oxide nanocrystallites (RCO2-ZnO NCs) of diameter less than 5 nm are prepared according to a modified Langmuir-Schaefer method. First, the suspension of RCO2-ZnO NCs is applied onto the air/water interface. Upon compression, the films become turbid and elastic. The integrity of such structures is ensured by interdigitation of ligands stabilizing ZnO NCs. Great elasticity allows transfer of the films onto a metal frame as a freely suspended film. Such membranes are afterward extracted from the supporting frame to form free-standing films of macroscopic area. Because the integrity of the films is maintained by ligands, no abolishment of quantum confinement occurs, and films retain spectroscopic properties of initial RCO2-ZnO NCs. The mechanism of formation of thin films of RCO2-ZnO NCs at the air/water interface is discussed in detail.

Single molecule surface-enhanced resonance Raman scattering (SERRS) spectra have been obtained for the parent porphycene (Pc-d0) and its deuterated isotopologue (Pc-d12), located on gold and silver nanoparticles. Equal populations of "hot spots" by the two isotopologues are observed for 1 : 1 mixtures in a higher concentration range of the single molecule regime (5 × 10(-9) M). For decreasing concentrations, hot spots are preferentially populated by undeuterated molecules. This is interpreted as an indication of a lower surface diffusion coefficient of Pc-d12. The photostability of single Pc molecules placed on nanoparticles is strongly increased in comparison with polymer environments. Trans tautomeric species dominate the spectra, but the analysis of time traces reveals transient intermediates, possibly due to rare cis tautomeric forms.

Nanoscale pH evaluation is a prerequisite for understanding the processes and phenomena occurring at solid-liquid, liquid-liquid, and liquid-gas interfaces, e.g., heterogeneous catalysis, extraction, partitioning, and corrosion. Research on the homogeneous processes within small volumes such as intracellular fluids, microdroplets, and microfluidic chips also requires nanometer scale pH assessment. Due to the opacity of numerous systems, optical methods are useless and, if applicable, require addition of a pH-sensitive dye. Potentiometric probes suffer from many drawbacks such as potential drift and lack of selectivity. Here, we present a voltammetric nanosensor for reliable pH assessment between pH 2 and 12 with high spatial resolution. It consists of a pyrolytic carbon nanoelectrode obtained by chemical vapor deposition (CVD) inside a quartz nanopipette. The carbon is modified by adsorption of syringaldazine from its ethanolic solution. It exhibits a stable quasi-reversible cyclic voltammogram with nearly Nernstian dependency of midpeak potentials (-54 mV/pH). This sensor was applied as a probe for scanning electrochemical microscopy (SECM) in order to map pH over a platinum ultramicroelectrode (UME), generating hydroxide ions (OH(-)) by the oxygen reduction reaction (ORR) at a diffusion-controlled rate in aerated phosphate buffered saline (PBS). The results reveal the alkalization of the electrolyte close to the oxygen reducing electrode, showing the insufficient buffer capacity of PBS to maintain a stable pH at the given conditions.

Metallic nanostructures (NS) due to their unique properties are often used as electrocatalytic electrode materials in energy conversion and storage as well as in sensing applications. Their common methods of preparation involve homogeneous synthesis and electrodeposition from solutions containing appropriate precursor and often reducing agent and capping agent (surfactant). The main drawbacks of these methods are restricted yield, waste production and often intrinsic contamination of the surface of obtained NS. Moreover, surface capping ligands affect electrocatalytic activity by hindrance of reactant access to the active sites. We propose a versatile method of preparation of bare (non-capped) mono- and multi-metallic (Au,1 Cu,2 Ag, Fe) NS by localized electrorefining of polycrystalline raw metals at high efficiency in a controlled manner. Catalytic activities of obtained NS towards oxygen reduction reaction (ORR) in alkaline media, hydrogen evolution (HER) and carbon dioxide reduction (CO2RR) to easily electrooxidizable fuels (formic acid and carbon monooxide) were analyzed by feedback mode scanning electrochemical microscopy (SECM). The morphology and specific activity of obtained NS depend on used support material (indium tin oxide (ITO) and glassy carbon) and other electrorefining parameters such as electrodeposition potential, source velocity and the composition of the electrolyte. The last parameter determines stoichiometry of Faradaic processes. Both gold and copper electrooxidation and electroreduction can follow one electron stoichiometry. ORR in alkaline media and CO2RR occur effectively on NS with highly developed surfaces, containing large number of defects, corners and edges, whereas HER occurs efficiently at crystallographic Cu facets. Bare gold NS also exhibit outstanding plasmonic properties. They were applied as support for surface-enhanced Raman spectroscopy (SERS) of single molecules providing enhancement factor >106.1 Another method of preparation of metallic nanostructures is cathodic corrosion. This approach for platinum was studied by Koper group.3 We used this procedure for platinum ultramicroelectrode treatment. Although electron transfer reactions at liquid liquid interfaces have been thoroughly studied in the presence of deliberately added potential determining ions or phase transfer catalysts, little is known about these reactions in the absence of the above species. Here, we report the formation of hydrogen peroxide (H2O2) in a liquid liquid two-phase system with only an electron donor (decamethylferrocene) solution in trifluorotoluene (TFT) and a proton donor solution (HClO4) in water. To detect H2O2, we used fluorescent microscopy and scanning electrochemical microscopy (SECM). We applied a potential sweep program to the SECM probe to overcome the electrode deactivation. To provide insight into the reaction rate and mechanism, we fitted SECM results to finite elements simulations. From the results of UV vis spectroscopy, we determined a H2O2 partition coefficient of 0.03 and the standard Gibbs energy of H2O2 transfer from water to TFT as 9.5 kJ/mol. The most important conclusion from this work is that the studied system provides conditions for spontaneous transfer of protons from the aqueous to the organic phase, even in the absence of deliberately added potential determining ions or phase transfer catalysts.

A straightforward method for preparation of carbon nanoparticulate film electrodes is reported. Durable interfaces were prepared by electrophoretic deposition of phenylsulphonate functionalized carbon nanoparticles. The amount of the deposited material and therefore the magnitude of the faradaic current was controlled by tuning the electrophoresis parameters. The prepared electrodes exhibited electrocatalytic effect towards oxidation of thiocholine and showed a linear signal response in a wide concentration range (10.7 - 2000 μM). The topographies of selected electrodes were investigated by hopping cone-shaped nanoelectrode using a scanning electrochemical microscopy setup in dry conditions. The proposed method of surface examination can be applied to large lateral areas of other nanostructured interfaces.

Hydrogen or hydrogen peroxide can be generated in liquid−liquid biphasic systems, where the organic phase contains sufficiently strong electron donor (whose redox potential is lower than the potential of reversible hydrogen electrode). H2O2 generation with acidified aqueous phase occurs prior to H2 evolution when oxygen is present. No other organic solvent than highly toxic 1,2-dichloroethane (DCE) has been reported in biphasic system for H2 or H2O2 generation. In this work, we have used trifluorotoluene (TFT) instead of carcinogenic DCE, and studied these reactions in TFT−water biphasic system. To evaluate H2 flux, scanning electrochemical microscopy potentiometric approach curves to the TFT−water interface were recorded. H2O2 was detected voltametrically at a microelectrode located in the vicinity of the interface. H2 and H2O2 are formed and both reactions occur also in the absence of a hydrophobic salt in the organic phase. Their thermodynamics was discussed on the basis of Gibbs energies determined electrochemically with droplet-modified electrodes. The results show that DCE can be replaced by a noncarcinogenic solvent and the biphasic system for H2 and H2O2 generation can be simplified by elimination of the uncommon hydrophobic salt from the organic phase.

The capabilities and applications of the focused ion beam (FIB) technology for detection of an electrochemical signal in nanoscale area are shown. The FIB system, enabling continuous micro- and nanofabrication within only one equipment unit, was used to produce a prototype of electrochemical nanometer-sized electrode for sensor application. Voltammetric study of electrochemically active compound (ferrocenemethanol) revealed the diffusion limiting current (12 pA), corresponding to a disc (planar) nanoelectrode with about 70 nm diameter of contact area. This size is in a good accordance with the designed contact-area (50 nm × 100 nm for width × thickness) of the FIB-produced nanoelectrode. It confirms that produced nanoelectrode is working properly in liquid solution and may enable correct measurements in nanometer-sized regions.

Electrocatalytic oxidation of glucose was studied at nanoparticulate gold and gold-carbon film electrodes. These electrodes were prepared by a layer-by-layer method without application of any linker molecules. Gold nanoparticles were stabilized by undecane thiols functionalized by trimethyl ammonium or carboxylate groups, whereas the carbon nanoparticles were covered by phenylsulfonate functionalities. The gold nanoparticulate electrodes were characterized by UV-vis and XPS spectroscopy, atomic force microscopy and voltammetry, before and after heat-treatment. Heat-treatment facilitates the aggregation of the nanoparticles and affects the structure of the film. The comparison of the results obtained with film electrodes prepared from gold nanoparticles with the same charge and with gold-carbon nanoparticulate electrodes, proved that positively charged nanoparticles are responsible for the high electrocatalytic activity, whereas negatively charged ones act rather as a linker of the film.

H2O2 is produced at the interface between a room-temperature ionic liquid with decamethylferrocene as an electron donor and an acidic aqueous solution. The electron donor can be regenerated electrochemically.
