Abstract Introduction With the application of high-resolution 3D 7 Tesla Magnetic Resonance Spectroscopy Imaging (MRSI) in high-grade gliomas, we previously identified intratumoral metabolic heterogeneities. In this study, we evaluated the potential of 3D 7 T-MRSI for the preoperative noninvasive classification of glioma grade and isocitrate dehydrogenase (IDH) status. We demonstrated that IDH mutation and glioma grade are detectable by ultra-high field (UHF) MRI. This technique might potentially optimize the perioperative management of glioma patients. Methods We prospectively included 36 patients with WHO 2021 grade 2–4 gliomas (20 IDH mutated, 16 IDH wildtype). Our 7 T 3D MRSI sequence provided high-resolution metabolic maps (e.g., choline, creatine, glutamine, and glycine) of these patients’ brains. We employed multivariate random forest and support vector machine models to voxels within a tumor segmentation, for classification of glioma grade and IDH mutation status. Results Random forest analysis yielded an area under the curve (AUC) of 0.86 for multivariate IDH classification based on metabolic ratios. We distinguished high- and low-grade tumors by total choline (tCho) / total N-acetyl-aspartate (tNAA) ratio difference, yielding an AUC of 0.99. Tumor categorization based on other measured metabolic ratios provided comparable accuracy. Conclusions We successfully classified IDH mutation status and high- versus low-grade gliomas preoperatively based on 7 T MRSI and clinical tumor segmentation. With this approach, we demonstrated imaging based tumor marker predictions at least as accurate as comparable studies, highlighting the potential application of MRSI for pre-operative tumor classifications.

IntroductionThe mental health of young people has been severely affected by the COVID-19 pandemic and the measures associated with it. Mental health symptoms increased across various domains during the pandemic and subjective wellbeing decreased.MethodsThis study examined the burdens and resources of Austrian school students (M = 16.63 years old) in the third year of the pandemic and compared them descriptively with the burdens and resources identified in a comparative study in 2021. A cross-sectional online survey with open-ended questions from April to May 2022 provided data that was analyzed using qualitative content analysis. A sub-sample of N = 214 was selected from the larger sample. This sub-sample is representative for the Austrian population aged 15–19 in terms of gender and migrant background.ResultsAnalysis of the open-ended questions showed that concerns about the pandemic and the burdens of the measures were no longer on young people's minds at the time of the survey in 2022. On the other hand, stress at school was increasing and the opening of schools and the resumption of face-to-face teaching were challenging for respondents. While resuming social contacts and leisure activities was mentioned as a resource by many respondents, some also expressed a desire for more time off and a retreat into coping strategies such as sleeping more or avoiding problems.DiscussionOur findings suggest that there is a need for low-threshold support from teachers and parents to help students catch up on missed lessons and to empathize with the mas they make the transition back to “old normal”.

Der vorliegende Beitrag untersucht die Sportberichterstattung der Tagesschau, betrachtet, wie vielfältig diese ist und welche Nachrichtenfaktoren die Sportberichterstattung bestimmen. Mithilfe einer quantitativen Inhaltsanalyse wird eine deutliche Dominanz des Fußballs und des Männersports aufgezeigt, was auf eine eingeschränkte inhaltliche und gesellschaftliche Vielfalt der Sportberichterstattung hindeutet. Da die Mehrheit der Beiträge zwar Ereignisse in Deutschland thematisiert, Ethnozentrismus jedoch nicht zu signifikant längeren Beiträgen führt, gibt es nur wenige Hinweise auf eine eingeschränkte räumliche Vielfalt. Im zweiten Teil der Analyse demonstriert die Arbeit, dass es signifikante Unterschiede in der Ausprägung von Nachrichtenfaktoren und Nachrichtenfaktorenbündeln zwischen den Beiträgen über Fußball und den Beiträgen über andere Sportarten gibt. Beiträge über Fußball weisen einen stärkeren nationalen Bezug auf als die übrigen Sportarten, während diese wiederum häufiger einen emotionalen Bezug beinhalten. Schließlich zeigt der Beitrag, dass Emotionalität und Spannung die wichtigsten Prädiktoren für die Länge eines Beitrages sind.

Abstract Breast cancer is a leading cause of death in female patients worldwide. Further research is needed to get a deeper insight into the mechanisms involved in the development of this devastating disease and to find new therapy strategies. The zebrafish is an established animal model, especially in the field of oncology, which has shown to be a promising candidate for pre-clinical research and precision-based medicine. To investigate cancer growth in vivo in zebrafish, one approach is to explore xenograft tumor models. In this article, we present the investigation of a juvenile xenograft zebrafish model using a Jones matrix optical coherence tomography (JM-OCT) prototype. Immunosuppressed wild-type fish at 1-month post-fertilization were injected with human breast cancer cells and control animals with phosphate buffered saline in the tail musculature. In a longitudinal study, the scatter, polarization, and vasculature changes over time were investigated and quantified in control versus tumor injected animals. A significant decrease in birefringence and an increase in scattering signal was detected in tumor injected zebrafish in comparison to the control once. This work shows the potential of JM-OCT as a non-invasive, label-free, three-dimensional, high-resolution, and tissue-specific imaging tool in pre-clinical cancer research based on juvenile zebrafish models.

Transitions between schools, vocational education and training (VET), and work pose important challenges for young people that influence their well‐being and social positioning now and in the future. The young people themselves experience the transition phase as the formation stage of their aspirations and goals. In this process, young people are confronted with the expectations and assessments of relevant others—such as parents, teachers, employers, and career counsellors—and by the requirements that are defined in sociopolitical and institutional contexts. In these contexts, criteria of successful transitions and risky transitions worthy of special support are made relevant. German and Austrian employment-centred transition regimes are characterised by relatively high standardisation and segregation as well as a strong VET system linked to the labour market. This thematic issue brings together contributions that examine challenges in these transitions from different perspectives and related facets of social inequality. The articles address different transitions (mostly school‐to‐VET, but also school‐to‐school or unemployment to work) and their different phases: aspiration formation, changing aspirations, challenges in transitions, and concrete problems in transition processes like disconnectedness or unemployment. The articles on social inequalities are related to class, ethnicity, gender, and (dis)ability. We also place importance on balancing different methods to bring together findings from quantitative surveys, qualitative interviews, and participatory research.

This article presents how young people in educational measures experience and discuss tensions between structural and individualised challenges they face in the transition from school to work. The findings are based on an Austrian citizen social science research project that involved conducting participatory research in Vienna with 33 young people between the ages of 15 and 23 years who are in measures for early school leavers that are preparing for further education and training. These co‐researchers struggle with constant comparisons to “norm” biographies and their accompanying social pressure as they try to meet the high expectations of school, work, and family. Additionally, mental health was a prominent issue, as the young co‐researchers experience stigma and a lack of professional support, which can impede their access to the labour market. The results of our research indicate that young people in employment measures require a more inclusive school and work environment that supports them regardless of their origin, family background, appearance, or mental health status. They negotiate the tension between individual employability and structural disadvantage and demand a recognised place in society, a request that connects to current debates concerning the individualisation of transitions from school to work.

Introduction: Acute hyperammonemic encephalopathy is associated with distinct brain MRI findings, namely, hyperintensity in T2-weighted sequences as well as restricted diffusion in diffusion-weighted imaging with accentuation in the insular cortex and cingulate gyrus. The pathophysiology and the histopathological correlates of these characteristic MRI findings are largely unknown. Case report: We present a 57-year-old male with a history of chronic alcohol abuse, liver cirrhosis and portal hypertension, and a clinical syndrome (variceal bleeding, depression of consciousness, seizures), elevated plasma ammonia levels, and characteristic brain MRI abnormalities suggestive of acute hyperammonemic encephalopathy. A postmortem histopathological examination revealed extensive hypoxic ischemic encephalopathy without evidence for metabolic encephalopathy. No episodes of prolonged cerebral hypoxemia were documented throughout the course of the disease. We conducted a review of the literature, which exhibited no reports of hyperammonemic encephalopathy in association with characteristic brain MRI findings and a consecutive histopathological examination. Conclusion: This is the first report of a patient with acute hyperammonemic encephalopathy together with characteristic brain MRI findings and a histopathological correlation. Although characteristic MRI findings of acute hyperammonemic encephalopathy were present, a histopathological examination revealed only hypoxic pathology without signs of metabolic encephalopathy. Modern data acquisition with active or passive photogrammetric imaging techniques generally results in 3D point clouds. Depending on the acquisition or processing method, the spacing of the individual points is either uniform or irregular. In the latter case, the neighbourhood definition like for digital images (4- or 8-neighbourhood, etc.) cannot be applied. Instead, analysis requires a local point neighbourhood. The local point neighbourhood with conventional k-nearest neighbour or fixed distance searches often produce sub-optimal results suffering from the inhomogeneous point distribution. In this article, we generalize the neighbourhood definition and present a generic spatial search framework which explicitly deals with arbitrary point patterns and aims at optimizing local point selection for specific processing tasks like interpolation, surface normal estimation and point feature extraction, spatial segmentation, and such like. The framework provides atomic 2D and 3D search strategies, (i) k-nearest neighbour, (ii) region query, (iii) cell based selection, and (iv) quadrant/octant based selection. It allows to freely combine the individual strategies to form complex, conditional search queries as well as specifically tailored point sub-selection. The benefits of such a comprehensive neighbourhood search approach are showcased for feature extraction and surface interpolation of irregularly distributed points.

Abstract Audencel is a dendritic cell (DC)-based cellular cancer immunotherapy against glioblastoma multiforme (GBM). It is characterized by loading of DCs with autologous whole tumor lysate and in vitro maturation via “danger signals”. The recent phase II “GBM-Vax” trial showed no clinical efficacy for Audencel as assessed with progression-free and overall survival in all patients. Here we present immunological research accompanying the trial with a focus on immune system factors related to outcome and Audencel’s effect on the immune system. Methodologically, peripheral blood samples (from apheresis before Audencel or venipuncture during Audencel) were subjected to functional characterization via enzyme-linked immunospot (ELISPOT) assays connected with cytokine bead assays (CBAs) as well as phenotypical characterization via flow cytometry and mRNA quantification. GBM tissue samples (from surgery) were subjected to T cell receptor sequencing and immunohistochemistry. As results we found: Patients with favorable pre-existing anti-tumor characteristics lived longer under Audencel than Audencel patients without them. Pre-vaccination blood CD8+ T cell count and ELISPOT Granzyme B production capacity in vitro upon tumor antigen exposure were significantly correlated with overall survival. Despite Audencel’s general failure to induce a significant clinical response, it nevertheless seemed to have an effect on the immune system. For instance, Audencel led to a significant up-regulation of the Th1-related immunovariables ELISPOT IFNγ, the transcription factor T-bet in the blood and ELISPOT IL-2 in a dose-dependent manner upon vaccination. Post-vaccination levels of ELISPOT IFNγ and CD8+ cells in the blood were indicative of a significantly better survival. In summary, Audencel failed to reach an improvement of survival in the recent phase II clinical trial. No clinical efficacy was registered. Our concomitant immunological work presented here indicates that outcome under Audencel was influenced by the state of the immune system. On the other hand, Audencel also seemed to have stimulated the immune system. Overall, these immunological considerations suggest that DC immunotherapy against glioblastoma should be studied further – with the goal of translating an apparent immunological response into a clinical response. Future research should concentrate on investigating augmentation of immune reactions through combination therapies or on developing meaningful biomarkers.
