Carnivory in plants is an unusual trait that has arisen multiple times, independently, throughout evolutionary history. Plants in the genus Genlisea are carnivorous, and feed on microorganisms that live in soil using modified subterranean leaf structures (rhizophylls). A surprisingly broad array of microfauna has been observed in the plants' digestive chambers, including ciliates, amoebae and soil mites. Here we show, through experiments and simulations, that Genlisea exploit active matter physics to 'rectify' bacterial swimming and establish a local flux of bacteria through the structured environment of the rhizophyll towards the plant's digestion vesicle. In contrast, macromolecular digestion products are free to diffuse away from the digestion vesicle and establish a concentration gradient of carbon sources to draw larger microorganisms further inside the plant. Flagella allow eukaryotic cells to move and pump fluid. We present the first three-dimensional, time-resolved imaging of the flagellar waveform of Chlamydomonas reinhardtii, a model alga found in fresh water. During the power stroke, we find that the flagella show rotational symmetry about the cell's centreline, but during the recovery stroke they display mirror symmetry about the same axis. We use our three-dimensional imaging data to test the applicability of resistive force theory when a force-free configuration is approximated using a cell on a micropipette. The inferred values of cells' swimming speeds and rotation rates show good agreement with experimental results on freely swimming cells., Comment: 7 pages, 4 figures

It is widely believed that the swimming speed, $v$, of many flagellated bacteria is a non-monotonic function of the concentration, $c$, of high-molecular-weight linear polymers in aqueous solution, showing peaked $v(c)$ curves. Pores in the polymer solution were suggested as the explanation. Quantifying this picture led to a theory that predicted peaked $v(c)$ curves. Using new, high-throughput methods for characterising motility, we have measured $v$, and the angular frequency of cell-body rotation, $\Omega$, of motile Escherichia coli as a function of polymer concentration in polyvinylpyrrolidone (PVP) and Ficoll solutions of different molecular weights. We find that non-monotonic $v(c)$ curves are typically due to low-molecular weight impurities. After purification by dialysis, the measured $v(c)$ and $\Omega(c)$ relations for all but the highest molecular weight PVP can be described in detail by Newtonian hydrodynamics. There is clear evidence for non-Newtonian effects in the highest molecular weight PVP solution. Calculations suggest that this is due to the fast-rotating flagella `seeing' a lower viscosity than the cell body, so that flagella can be seen as nano-rheometers for probing the non-Newtonian behavior of high polymer solutions on a molecular scale., Comment: 17 pages

We present a fast, high-throughput method for characterizing the motility of microorganisms in 3D based on standard imaging microscopy. Instead of tracking individual cells, we analyse the spatio-temporal fluctuations of the intensity in the sample from time-lapse images and obtain the intermediate scattering function (ISF) of the system. We demonstrate our method on two different types of microorganisms: bacteria, both smooth swimming (run only) and wild type (run and tumble) Escherichia coli, and the bi-flagellate alga Chlamydomonas reinhardtii. We validate the methodology using computer simulations and particle tracking. From the ISF, we are able to extract (i) for E. coli: the swimming speed distribution, the fraction of motile cells and the diffusivity, and (ii) for C. reinhardtii: the swimming speed distribution, the amplitude and frequency of the oscillatory dynamics. In both cases, the motility parameters are averaged over \approx 10^4 cells and obtained in a few minutes.

We demonstrate 'differential dynamic microscopy' (DDM) for the fast, high throughput characterization of the dynamics of active particles. Specifically, we characterize the swimming speed distribution and the fraction of motile cells in suspensions of Escherichia coli bacteria. By averaging over ~10^4 cells, our results are highly accurate compared to conventional tracking. The diffusivity of non-motile cells is enhanced by an amount proportional to the concentration of motile cells., Comment: 4 pages, 4 figures. In this updated version we have added simulations to support our interpretation, and changed the model for the swimming speed probability distribution from log-normal to a Schulz distribution. Neither modification significantly changes our conclusions
