Abstract

Sea urchins are important inhabitants of many marine ecosystems. They are also an economic resource for many international fisheries and are an important animal model in developmental biology. Sea urchins ingest a variety of plant and animal matter, form a digesta pellet, and produce an egesta (fecal) pellet that contributes to benthic food webs. The size and morphology of fecal pellets produced by various size classes of the sea urchin Lytechinus variegatus fed a variety of natural, vegetable-based, or formulated diets were characterized in terms of two-dimensional area, length (long diameter), width (short diameter), and circularity, as well as by ultrastructure analysis using transmission electron microscopy (TEM). Sea urchins fed natural and vegetable-based ingredients produced a wide variety of irregularly shaped egesta that were loosely surrounded by mucus, whereas urchins fed a formulated diet produced highly circular egesta surrounded by a uniform multilayered mucus coat. Larger urchins (62 [+ or -] 3.1 g wet weight) consuming a formulated diet produced egesta that were larger in size than smaller adult (26.5 [+ or -] 2.7 g) or juvenile urchins (3.6 [+ or -] 0.4 g), with egesta length of 0.93 [+ or -] 0.06, 0.75 [+ or -] 0.05, and 0.61 [+ or -] 0.01 mm, respectively (mean [+ or -] 95% confidence interval [CI]; P < 0.001). Ultrastructural analysis of the egesta using TEM image analysis software from urchins consuming formulated diets indicated that pellets contained extensive bacterial communities, ranging from 31% to 69% of the total cross-sectional area of the pellet, with dense communities located near the surface of the pellet (9.51 x [10.sup.8] cells/[cm.sup.2]). Qualitative examination of the egesta TEM suggested the presence of a diverse bacterial community. Following egestion, these microbial communities are suggested to have an important role in natural food webs, with potential value for integrated multitrophic aquaculture systems. 21. A Case Study of University-Community Collaboration to Reduce the Negative Effects of Binge Drinking

Abstract

Binge drinking is a prevalent, persistent problem within U.S. university cities. Consequences of students' binge drinking can result in injury, assault, disruption in neighborhoods, and even death. Proponents of one potential solution to the problem, the environmental approach, propose changing the context of drinking by altering factors such as access, price, policies, and enforcement that shape drinking patterns of individuals. Those factors are influenced by multiple entities and jurisdictions requiring collaboration to bring about change. The purpose of the study was to uncover factors of university-community collaboration formed to reduce students' destructive binge drinking. Guided by collaboration theory, this qualitative, single case study was designed to discover the factors and insights that led to a successful university-community collaboration to reduce binge drinking within a safe communities coalition. The primary data were gathered from interviews of 13 participants in the coalition. The results indicated that positive relationships among the members of the coalition were the foundation of success. These relationships were nurtured by administrative support, consistent meetings, open communications, shared leadership, mutual benefits, and a lack of tension. The participants defined success, not only as a reduction in the negative consequences of binge drinking, but also as the maintenance of positive relationships that sustain the collaborative efforts. The implications for social change include the increase of coalitions and improvement of collaboration among college administrators, community leaders, and other agencies attempting to reduce the harms caused by students' binge drinking. 26. Zebrafish Danio rerioexhibit positive growth profiles when fed dietary yeast and bacterial‐based single‐cell protein as a replacement for fish protein hydrolysate

Abstract

The value of Zebrafish Danio rerioas a preclinical biomedical model has increased as nutritionally complete diets are being developed. Consequently, nutrients and nutrient sources can be used in evaluating the health status of Zebrafish, providing information that can be translated to human health. We have investigated the impact of commercially produced bacterial (MRD‐Pro) and yeast (DY‐Pro) single‐cell protein sources in formulated Zebrafish diets. Thirty‐five days after fertilization Zebrafish were randomized to one of five dietary treatments including FPH100 (a control diet with primarily fish protein hydrolysate [FPH] as the protein source), MRD50 and MRD100 (diets that replaced half or all of the FPH with MRD, a bacterial protein), or DY50 and DY100 (diets that replaced half or all of the FPH with DY, a yeast protein) for 8 weeks (n= 8 tanks/diet treatment, 14 fish/tank). Female terminal wet weight was the same among all diets; however, male terminal wet weight was increased in fish consuming MRD diets. In addition, carcass lipid content was significantly lower in females consuming the DY and MRD diets than those fed FPH. Male lipid content was the same among all diets. Results demonstrate that single‐cell protein sources are suitable replacements for FPH and, in female populations, may impart health benefits related to lean matter production and reduced adiposity. Impact statementIn this study we demonstrated that replacement of dietary fish protein hydrolysate with commercial single‐cell bacterial and yeast protein sources in the diet of Zebrafish maintained female weight gain and, in fact, increased weight gain in males consuming the bacterial single‐cell protein. Females also had lower carcass fat content when consuming the yeast and bacterial single‐cell proteins. These single‐cell proteins can be an effective alternative ingredient in formulated diets for the Zebrafish, providing additional insight into the health of this important model for human health and valuable ornamental fish. In this study we demonstrated that replacement of dietary fish protein hydrolysate with commercial single‐cell bacterial and yeast protein sources in the diet of Zebrafish maintained female weight gain and, in fact, increased weight gain in males consuming the bacterial single‐cell protein. Females also had lower carcass fat content when consuming the yeast and bacterial single‐cell proteins. 34. Macronutrient Ratio Modification in a Semi‐PurifiedDiet Composition: Effects on Growth and Body Composition of Juvenile Zebrafish Danio rerio

Abstract

The interaction of dietary macronutrients in the control of growth, body composition, health, and longevity has received renewed attention. The protein leverage hypothesis proposes an inverse relationship between dietary protein levels and obesity development (low protein promoting high obesity), although the majority of laboratory studies have utilized a design modulating the protein amount within a single dietary protein source composition. We investigated whether varying the levels of dietary protein amount and sources would have impacts on growth and body composition using the Zebrafish Danio reriomodel. At 28 d postfertilization, Zebrafish were fed 1 of 12 dietary treatments for 12 weeks. Diets contained different protein sources (fish protein hydrolysate [FPH], soy protein isolate [SOY], casein [CAS], or a mixture of all three sources including wheat gluten [MIX]). Sources were formulated at three protein concentrations (18, 33, or 48% as fed, substituted with wheat starch for caloric balance). Body length, height, and weight were measured over time and at termination. Contributions of macronutrients to growth and body fat outcomes were estimated by LASSO (least absolute shrinkage and selection operator) regression. Male and female length, height, and weight increased significantly in response to increasing dietary protein. Male and female fish fed SOY or FPH had the highest amount of body lipid at all protein concentrations relative to those fed CAS or MIX. Relative body lipid was highest in fish that were fed the lowest protein concentration. These data suggest an important role of macronutrient balances, including dietary protein amount and source, in relation to the protein: energy (carbohydrate and lipid) ratio on growth and body composition outcomes. These outcomes may ultimately reflect metabolic alterations that can lead to confounding interpretations of health and disease status. Furthermore, these data reinforce the need to consider and report dietary composition in establishing rigorous and reproducible nutritional guidelines for Zebrafish.

Postexercise ingestion of the CHO-PRO beverage, in comparison with the SB, resulted in a 55% greater time to exhaustion during a subsequent exercise bout at 85% maximum oxygen consumption (V[O.sub.2]max). The greater recovery after the intake of the CHO-PRO beverage could be because of a greater rate of muscle glycogen storage. Therefore, a second study was designed to investigate the effects of after exercise CHO-PRO and SB supplements on muscle glycogen restoration. Eight endurance-trained cyclists (V[O.sub.2]max = 62.1 [+ or -] 2.2 ml*[kg.sup.-1] body wt * [min.sup.-1]) performed 2 trials consisting of a 2-hour glycogen-depletion ride at 65-75% V[O.sub.2]max. Carbohydrate-protein (355 ml; ~0.8 g carbohydrate (CHO) * [kg.sup.-1] body wt and ~0.2 g protein*[kg.sup.-1] body wt) or SB (355 ml; ~0.3 g CHO*[kg.sup.-1] body wt) was provided immediately and 2 hours after exercise. Trials were randomized and separated by 7-15 days. Ingestion of the CHO-PRO beverage resulted in a 17% greater plasma glucose response, a 92% greater insulin response, and a 128% greater storage of muscle glycogen (159 [+ or -] 18 and 69 [+ or -] 32 [micro]mol*[g.sup.-1] dry weight for CHO-PRO and SB, respectively) compared with the SB (p < 0.05). These findings indicate that the rate of recovery is coupled with the rate of muscle glycogen replenishment and suggest that recovery supplements should be consumed to optimize muscle glycogen synthesis as well as fluid replacement. Abstract

Planktonic embryos of Lytechinus variegatus utilize photo-protective mechanisms to minimize the deleterious effects of solar ultraviolet radiation (UVR) exposure. It was hypothesized that embryos from parents who received supplemental β-carotene in their diets would be more resistant to the effects of UVR than embryos from parents who received no supplement. Adult Lytechinus variegatus broodstock with gonads in the growing phase were fed diets either with or without supplemental β- carotene for 5 months and subsequently induced to spawn. Fertilized eggs were collected from each feed treatment and exposed to differing intensities of UVA (0-4 J/m²) or UVB (0-100 mJ/cm²) radiation. Larval mortality counts and developmental status were recorded at 34 and 55 hours postfertilization and compared between feed treatments. Embryos derived from sea urchins consuming supplemental β-carotene developed at a slower rate than those whose parents did not consume supplemental β-carotene. Increasing intensities of UVA and UVB radiation were positively correlated with larval mortality in both dietary treatments. UVB induced higher mortality than UVA. Larval mortality in the β-carotene supplemented treatment was significantly higher than the nonsupplemented treatment. These data suggest that dietary supplements of carotenoid do not provide photo-protection and may enhance, through unknown mechanisms, the deleterious effects of UV exposure. [ABSTRACT FROM AUTHOR]

The Marine Corps manpower system has the responsibility of ensuring that the appropriate numbers of trained and experienced Marines are assigned to commanders to perform their missions. The main concern with the current manpower system is that there are many skills that are critically short while others exceed requirements. The questions that are addressed in this thesis are, "can and should the Marine Corps promotion system be restructured, redesigned, or replaced to fulfill the objective of properly structuring the force," and "what would be the policy implications, unintended consequences, and pros and cons of promoting officers by MOS?" The authors explored the issue by first taking a detailed look at the history, statutory basis, and operation of the Marine Corps officer promotion system. The authors then examined the history, statutory basis, and operations of the officer promotion systems of the Navy, Army and Air Force to understand if their promotion systems were engineered to deal with force structuring concerns.
