Abstract

An essential prerequisite to safeguard pollinator species is characterisation of the multifaceted diversity of crop pollinators and identification of the drivers of pollinator community changes across biogeographical gradients. The extent to which intensive agriculture is associated with the homogenisation of biological communities at large spatial scales remains poorly understood. In this study, we investigated diversity drivers for 644 bee species/morphospecies in 177 commercial apple orchards across 33 countries and four global biogeographical biomes. Our findings reveal significant taxonomic dissimilarity among biogeographical zones. Interestingly, despite this dissimilarity, species from different zones share similar higher-level phylogenetic groups and similar ecological and behavioural traits (i.e. functional traits), likely due to habitat filtering caused by perennial monoculture systems managed intensively for crop production. Honey bee species dominated orchard communities, while other managed/manageable and wild species were collected in lower numbers. Moreover, the presence of herbaceous, uncultivated open areas and organic management practices were associated with increased wild bee diversity. 3. Apparatus and Method for Low-Temperature Training of Shape Memory Alloys

Abstract

An apparatus and method for the low-temperature thermo-mechanical training of shape memory alloys (SMA) has been developed. The experimental SMA materials are being evaluated as prototypes for applicability in novel thermal management systems for future cryogenic applications. Alloys providing two-way actuation at cryogenic temperatures are the chief target. The mechanical training regimen was focused on the controlled movement of rectangular strips, with S-bend configurations, at temperatures as low as 30 K. The custom holding fixture included temperature sensors and a low heat-leak linear actuator with a magnetic coupling. The fixture was mounted to a Gifford-McMahon cryocooler providing up to 25 W of cooling power at 20 K and housed within a custom vacuum chamber. Operations included both training cycles and verification of shape memory movement. The system design and operation are discussed. Results of the training for select prototype alloys are presented.

Abstract

Background. As more women with metabolic muscle diseases reach reproductive age, knowledge of these diseases and their impact on pregnancy is necessary. Case. 23-year-old G1P0 with juvenile-onset Pompe disease (PD) delivered a viable infant by cesarean section at 32 weeks and 6 days. The pregnancy was complicated by worsening maternal pulmonary status, muscular strength, and mobility. Conclusion. The management of pregnancies complicated by Pompe disease requires a multidisciplinary approach, including expertise in neuromuscular disease, maternal-fetal medicine, biochemical genetics, pulmonology, anesthesia, and dietetics.

46. Expression and functional analysis of the Rhizobium meliloti nifA gene

Abstract

The translational initiation point for Rhizobium meliloti nifA, the specific activator for nitrogen fixation (nif) genes, was determined. When expressed in an Escherichia colilinked transcriptional‐translational system the DNA coding for the R.meliloti nifA gene produced four polypeptide bands of 71 000, 67 000, 62 000 and 59 000 daltons. There are three in‐frame ATG codons at the N‐terminus of the gene; by replacing the poor ribosome binding sites of the native DNA with a synthetic consensus ribosome binding site prior to each of ATG codons the polypeptides were produced at enhanced levels and related to each of the initiation codons. The ability of these specifically expressed polypeptides to activate nifpromoters fused to lacZ was determined. Only the fulllength polypeptide activated the Klebsiella pneumoniae nifH, R.meliloti nifH and fixA and Bradyrhizobium japonicum nifH and nifD promoters. The R.meliloti fixA promoter, contrary to previous evidence, could be activated in E.coli.Deletion of the putative N‐terminal domain of the R.meliloti nifA gene product greatly increased the ability of the protein to activate nifpromoters. However, deletions retaining part of this domain were not functional. This shows that the N‐terminal domain is not essential for activity and that its presence decreases the full potential function of the protein. Our results are consistent with the suggesting that this domain has a regulatory role. In contrast to K.pneumoniae nifA protein, the function of the full length and domain deleted forms of R.meliloti nifA gene product was sensitive to oxygen in E.coli.
