Phytophthora capsici is reported for the first time in the Lao PDR. It was isolated from soil collected from the root zone of wilting red chilli (Capsicum annuum) plants in a polyhouse farm in Paksong district of Champasak province. The wilting chilli plants had typical symptoms of infection by P. capsici including root rot and necrosis of the outer cortex of the stem base. The isolate was identified using morphological and molecular markers. A pathogenicity trial demonstrated that the culture was pathogenic to a local cultivar of chilli seedlings.

Infections caused by waterborne Escherichia coli O157 continue to pose a public health risk. An increase in faecal coliform loading of watercourses due to expanding populations, intensification of agriculture and climate change are predicted to amplify these risks. Understanding the effect of land use on the ecology of E. coli O157 in environmental waters is therefore important for implementing effective mitigation measures. In order to test the hypothesis that activity of waterborne E. coli O157 is affected by both land use type and the respective autochthonous microbial communities, we inoculated replicate microcosms of water collected from areas of contrasting land uses within a catchment with a chromosomally lux-marked E. coli O157. Pathogen metabolic activity and its ability to reactivate following addition of nutrients were quantified over time in both filter-sterilised and non-sterile microcosms. Metabolic activity differed significantly according to the land use type, the degree of competition from background microbes and the availability of nutrients. These results indicate that land use types associated with particular areas of a watercourse should be considered a central factor in models that aim to predict pathogen risk in environmental waters.

This report describes the twelfth repeat hydrography section across Drake Passage, first established during the World Ocean Circulation Experiment. For this work NOC made use of the regular BAS supply trip to Rothera from the Falklands using the RRS James Clark Ross. Thirty CTD/LADCP stations were carried out across the 753 km section from Burdwood Bank to Elephant Island, plus one test station in the deep waters to the north of Burdwood Bank. Maximum station spacing on the section was 33 km, with stations closer together on the continental shelves. Water samples were drawn for salinity analysis, for subsequent CTD conductivity calibration. Samples were also drawn for analysis of oxygen isotope fraction δ18O, for later analysis back at NOC. The CTD was a SeaBird 911plus with dual temperature and conductivity sensors. There were two LADCP instruments, but no useful data were collected with them. Various underway measurements were also collected, including navigation, vessel-mounted ADCP, sea surface temperature and salinity, water depth and meteorological parameters. Water samples were also collected for subsequent analysis for chlorophyll, particulate carbon and biogenic silica.

Item does not contain fulltext, We describe five boys from different families with an atypically severe form of Pelizaeus-Merzbacher disease (PMD) who have three, and in one case, five copies of the proteolipid protein (PLP1) gene. This is the first report of more than two copies of PLP1 in PMD patients and clearly demonstrates that severe clinical symptoms are associated with increased PLP1 gene dosage. Previously, duplications, deletions and mutations of the PLP1 gene were reported to give rise to this X-linked disorder. Patients with PLP1 duplication are usually classified as having either classical or transitional PMD rather than the more rare severe connatal form. The clinical symptoms of the five patients in this study included lack of stable head control and severe mental retardation, with three having severe paroxysmal disorder and two dying before the first year of life. Gene dosage was determined using interphase FISH (fluorescence in situ hybridization) and the novel approach of multiple ligation probe amplification (MLPA). We found FISH unreliable for dosage detection above the level of a duplication and MLPA to be more accurate in determination of specific copy number. Our finding that three or more copies of the gene give rise to a more severe phenotype is in agreement with observations in transgenic mice where severity of disease increased with Plp1 gene dosage and level of overexpression. The patient with five copies of PLP1 was not more affected than those with a triplication, suggesting that there is possibly a limit to the level of severity or that other genetic factors influence the phenotype. It highlights the significance of PLP1 dosage in CNS myelinogenesis as well as the importance of accurate determination of PLP1 gene copy number in the diagnosis of PMD and carrier detection.

Tapasin is a Mr 48,000 glycoprotein and has a specialized role in MHC class I-restricted antigen presentation. It is encoded by a gene which maps centromeric to the MHC class II region of human Chromosome 6 within 200 kb of HLA-DP. There is variable dependence upon tapasin for MHC class I expression among different MHC class I alleles. HLA-B*4402 and to a lesser extent HLA-A1 and B8 are tapasin dependent, whereas HLA-B27, A2 and to a lesser extent B7 and A3 are tapasin independent. We investigated whether tapasin is polymorphic and whether these Tapasin alleles are in linkage with any MHC class I alleles. We identified three new mutations within intron 4, which are in a particular linkage with the previously described exon 4 (G16003C) dimorphism. The intronic mutations are G16146T, G16232A, and T16317A (numbering according to cosmid clone F0811; GenBank accession number Z97184). The allele frequency of Tapasin*01 (G16003) was 0.47 and Tapasin*02 (C16003) was 0.53 in this UK population. Four of the eight possible intronic haplotypes were identified and their cis linkage with the tapasin dimorphism ascertained. Tapasin*01 was associated with all the identified haplotypes, while Tapasin*02 was only associated with the wild-type intronic sequence (GGT). There was no significant linkage (P>0.01) of the Tapasin dimorphism or new Tapasin alleles to any of the MHC class I A, B, or C alleles studied or to the extended A1 B8 DR3 haplotype.
