Abstract Spherical harmonic (SH) expansion is a useful tool to study any variable that has valid values at all latitudes and longitudes. The variable can be quantified as a sum of different spherical harmonic components, which are the spherical harmonic functions multiplied by their expansion coefficients. We find that the SH components of cloud radiative effect (CRE) have correlations with El Niño‐Southern Oscillation (ENSO) and the Hadley Circulation (HC). In particular, the expansion degree 2 (l=2 $l=2$) SH power spectrum component anomaly of CRE is strongly correlated with ENSO. The two dipole patterns appearing in the l=2 $l=2$ SH component anomaly map can be reasonably explained by a known mechanism of ENSO's impact on cloud properties. The l=3 $l=3$ and l=5 $l=5$ SH power spectrum components are correlated with HC intensity, whereas the l=6 $l=6$ and l=8 $l=8$ components are correlated with HC latitudinal widths. In ENSO warm and cold phases, the HC‐correlated SH components have opposite anomalies, which suggests the impact of ENSO on HC. This study illustrates that the SH expansion technique provides a different perspective to study the impacts of large‐scale atmospheric circulation on global cloud properties and radiative effects.

Metal lattices are an important class of cellular materials that offer great advantages by providing high-strength and lightweight structures as compared to bulk materials. Progress in additive manufacturing techniques has led to increased complexity in design and shape of produced objects and is greatly beneficial for the development of metallic lattice structures. However additive manufacturing of lattices suffers from unpredictable defect creation that can compromise its mechanical integrity. Although post-build inspection techniques can provide quality assurance of the process, accurate assessment can be technically challenging, time consuming and costly. In this work, we investigate the use of high-speed measurements of thermal emission from the melt pool to identify defective individual struts formed with a missing bottom half in an otherwise fully built lattice structure produced with laser powder bed fusion. Surprisingly, results indicate lower photodiode signal, suggesting colder melt pool surface temperature, when printing struts with missing bottom half as compared to nominal struts. Additional thermographic imaging and multi-physics simulations reveal that the low photodiode signal is accompanied by presence of hot spatters carrying heat away from detection and continuous avalanche of powder on the melt pool. Based on these observations, a method was developed to identify defective individual struts with missing bottom half in full built lattices. This prediction approach provides valuable insights about part quality which are important for process qualification and illustrates the utility of melt pool thermal emission monitoring for identifying specific defects introduced by laser powder bed fusion.

The decades-long Clouds and Earth’s Radiant Energy System (CERES) Project includes both cloud and radiation measurements from instruments on the Aqua, Terra, and Suomi National Polar-orbiting Partnership (SNPP) satellites. To build a reliable long-term climate data record, it is important to determine the accuracies of the parameters retrieved from the sensors on each satellite. Cloud amount, phase, and top height derived from radiances taken by the Visible Infrared Imaging Radiometer Suite (VIIRS) on the SNPP are evaluated relative to the same quantities determined from measurements by the Cloud-Aerosol Lidar with Orthogonal Polarization (CALIOP) on the Cloud Aerosol Lidar and Infrared Pathfinder Satellite Observation (CALIPSO) spacecraft. The accuracies of the VIIRS cloud fractions are found to be as good as or better than those for the CERES amounts determined from Aqua MODerate-resolution Imaging Spectroradiometer (MODIS) data and for cloud fractions estimated by two other operational algorithms. Sensitivities of cloud fraction bias to CALIOP resolution, matching time window, and viewing zenith angle are examined. VIIRS cloud phase biases are slightly greater than their CERES MODIS counterparts. A majority of cloud phase errors are due to multilayer clouds during the daytime and supercooled liquid water clouds at night. CERES VIIRS cloud-top height biases are similar to those from CERES MODIS, except for ice clouds, which are smaller than those from CERES MODIS. CERES VIIRS cloud phase and top height uncertainties overall are very similar to or better than several operational algorithms, but fail to match the accuracies of experimental machine learning techniques. The greatest errors occur for multilayered clouds and clouds with phase misclassification. Cloud top heights can be improved by relaxing tropopause constraints, improving lapse-rate to model temperature profiles, and accounting for multilayer clouds. Other suggestions for improving the retrievals are also discussed.

Cloud properties are essential for the Clouds and the Earth’s Radiant Energy System (CERES) Project, enabling accurate interpretation of measured broadband radiances, providing a means to understand global cloud-radiation interactions, and constituting an important climate record. Producing consistent cloud retrievals across multiple platforms is critical for generating a multidecadal cloud and radiation record. Techniques used by CERES for retrievals from measurements by the MODerate-Resolution Imaging Spectroradiometer (MODIS) on Terra and Aqua platforms are adapted for the application to radiances from the Visible Infrared Imaging Radiometer Suite (VIIRS) on the Suomi National Polar-orbiting Partnership to continue the CERES record beyond the MODIS era. The algorithm adjustments account for spectral and channel differences, use revised reflectance models, and set new thresholds for detecting thin cirrus clouds at night. Cloud amounts from VIIRS are less than their MODIS counterparts by 0.016 during the day and 0.026 at night, but trend consistently over the 2012–2020 period. The VIIRS mean liquid water cloud fraction differs by ~0.01 from the MODIS amount. The average cloud heights from VIIRS differ from the MODIS heights by less than 0.2 km, except the VIIRS daytime ice cloud heights, which are 0.4 km higher. The mean VIIRS nonpolar optical depths are 17% (1%) larger (smaller) than those from MODIS for liquid (ice) clouds. The VIIRS cloud hydrometeor sizes are generally smaller than their MODIS counterparts. Discrepancies between the MODIS and VIIRS properties stem from spectral and spatial resolution differences, new tests at night, calibration inconsistencies, and new reflectance models. Many of those differences will be addressed in future editions.

The energy balance equation of an atmospheric column indicates that two approaches are possible to compute regional net surface energy flux. The first approach is to use the sum of surface energy flux components Fnet,c and the second approach is to use net top-of-atmosphere (TOA) irradiance and horizontal energy transport by the atmosphere Fnet,t. When regional net energy flux is averaged over the global ocean, Fnet,c and Fnet,t are, respectively, 16 and 2 Wm–2, both larger than the ocean heating rate derived from ocean temperature measurements. The difference is larger than the estimated uncertainty of Fnet,t of 11 Wm–2. Larger regional differences between Fnet,c and Fnet,t exist over tropical ocean. The seasonal variability of energy flux components averaged between 45°N and 45°S ocean reveals that the surface provides net energy to the atmosphere from May to July. These two examples demonstrates that the energy balance can be used to assess the quality of energy flux data products.

Two prostaglandin (PG) H synthases encoded by Ptgs genes, colloquially known as cyclooxygenase (COX)-1 and COX-2, catalyze the formation of PG endoperoxide H2, the precursor of the major prostanoids. To address the functional interchangeability of these two isoforms and their distinct roles, we have generated COX-2>COX-1 mice whereby Ptgs2 is knocked in to the Ptgs1 locus. We then “flipped” Ptgs genes to successfully create the Reversa mouse strain, where knock-in COX-2 is expressed constitutively and knock-in COX-1 is lipopolysaccharide (LPS) inducible. In macrophages, flipping the two Ptgs genes has no obvious impact on COX protein subcellular localization. COX-1 was shown to compensate for PG synthesis at high concentrations of substrate, whereas elevated LPS-induced PG production was only observed for cells expressing endogenous COX-2. Differential tissue-specific patterns of expression of the knock-in proteins were evident. Thus, platelets from COX-2>COX-1 and Reversa mice failed to express knock-in COX-2 and, therefore, thromboxane (Tx) production in vitro and urinary Tx metabolite formation in COX-2>COX-1 and Reversa mice in vivo were substantially decreased relative to WT and COX-1>COX-2 mice. Manipulation of COXs revealed isoform-specific compensatory functions and variable degrees of interchangeability for PG biosynthesis in cells/tissues.

Data is provided to show the detailed fatty acid and lipidomic composition of normal and tumor rat colon tissues. Rats were fed either a Western fat diet or a fish oil diet, and half the rats from each diet group were treated with chemical carcinogens that induce colon cancer (azoxymethane and dextran sodium sulfate). The data show total fatty acid profiles of sera and of all the colon tissues, namely normal tissue from control rats and both normal and tumor tissues from carcinogen-treated rats, as obtained by gas chromatography with mass spectral detection. Data from lipidomic analyses of a representative subset of the colon tissue samples is also shown in heat maps generated from hierarchical cluster analysis. These data display the utility lipidomic analyses to enhance the interpretation of dietary feeding studies aimed at cancer prevention and support the findings published in the companion paper (Effects of fish oil supplementation on prostaglandins in normal and tumor colon tissue: modulation by the lipogenic phenotype of colon tumors, Djuric et al., 2017 [1]). Prostaglandin (PG) endoperoxide H synthase (PGHS)-2, also known as cyclooxygenase (COX)-2, can convert arachidonic acid (AA) to PGH2 in the committed step of PG synthesis. PGHS-2 functions as a conformational heterodimer composed of an allosteric (Eallo) and a catalytic (Ecat) monomer. Here we investigated the interplay between human (hu)PGHS-2 and an alternative COX substrate, the endocannabinoid, 2-arachidonoylglycerol (2-AG), as well as a stable analog, 2-O-arachidonylglycerol ether (2-AG ether). We also compared the inhibition of huPGHS-2-mediated oxygenation of AA, 2-AG, and 2-AG ether by the well-known COX inhibitor, ibuprofen. When tested with huPGHS-2, 2-AG and 2-AG ether exhibit very similar kinetic parameters, responses to stimulation by FAs that are not COX substrates, and modes of inhibition by ibuprofen. The 2-AG ether binds Ecat more tightly than Eallo and, thus, can be used as a stable Ecat-specific substrate to examine certain Eallo-dependent responses. Ibuprofen binding to Eallo of huPGHS-2 completely blocks 2-AG or 2-AG ether oxygenation; however, inhibition by ibuprofen of huPGHS-2-mediated oxygenation of AA engages a combination of both allosteric and competitive mechanisms.

Preparing for climate change depends on the observation and prediction of decadal trends of the environmental variables, which have a direct impact on the sustainability of resources affecting the quality of life on our planet. The NASA Climate Absolute Radiance and Refractivity Observatory (CLARREO) mission is proposed to provide climate quality benchmark spectral radiance observations for the purpose of determining the decadal trends of climate variables, and validating and improving the long-range climate model forecasts needed to prepare for the changing climate of the Earth. The CLARREO will serve as an in-orbit, absolute, radiometric standard for the cross-calibration of hyperspectral radiance spectra observed by the international system of polar operational satellite sounding sensors. Here, we demonstrate that the resulting accurately cross-calibrated polar satellite global infrared spectral radiance trends (e.g., from the Metop IASI instrument considered here) can be interpreted in terms of temperature and water vapor profile trends. This demonstration is performed using atmospheric state data generated for a 100-year period from 2000−2099, produced by a numerical climate model prediction that was forced by the doubling of the global average atmospheric CO2 over the 100-year period. The vertical profiles and spatial distribution of temperature decadal trends were successfully diagnosed by applying a linear regression profile retrieval algorithm to the simulated hyperspectral radiance spectra for the 100-year period. These results indicate that it is possible to detect decadal trends in atmospheric climate variables from high accuracy all-sky satellite infrared radiance spectra using the linear regression retrieval technique.

Western diets are enriched in omega-6 vs. omega-3 fatty acids, and a shift in this balance toward omega-3 fatty acids may have health benefits. There is limited information about the catabolism of 3-series prostaglandins (PG) formed from eicosapentaenoic acid (EPA), a fish oil omega-3 fatty acid that becomes elevated in tissues following fish oil consumption. Quantification of appropriate urinary 3-series PG metabolites could be used for noninvasive measurement of omega-3 fatty acid tone. Here we describe the preparation of tritium- and deuterium-labeled 6-keto-PGF2α and their use in identifying urinary metabolites in mice using LC-MS/MS. The major 6-keto-PGF2α urinary metabolites included dinor-6-keto-PGF2α (∼10%) and dinor-13,14-dihydro-6,15-diketo-PGF1α (∼10%). These metabolites can arise only from the enzymatic conversion of EPA to the 3-series PGH endoperoxide by cyclooxygenases, then PGI3 by prostacyclin synthase and, finally, nonenzymatic hydrolysis to 6-keto-PGF2α. The 6-keto-PGF derivatives are not formed by free radical mechanisms that generate isoprostanes, and thus, these metabolites provide an unbiased marker for utilization of EPA by cyclooxygenases.

Retrievals of ice cloud properties using infrared measurements at 3.7, 6.7, 7.3, 8.5, 10.8, and 12.0 mm can provide consistent results regardless of solar illumination, but are limited to cloud optical thicknesses t < ~6. This paper investigates the variations in radiances at these wavelengths over a deep convective cloud system for their potential to extend retrievals of t and ice particle size De to optically thick clouds. Measurements from an imager, an interferometer, the Cloud Physics Lidar (CPL), and the Cloud Radar System (CRS) aboard the NASA ER-2 aircraft during the NASA TC4 (Tropical Composition, Cloud and Climate Coupling) experiment flight during 5 August 2007, are used to examine the retrieval potential of infrared radiances over optically thick ice clouds. Simulations based on coincident in situ measurements and combined cloud t from CRS and CPL measurements are comparable to the observations. They reveal that brightness temperatures at these bands and their differences (BTD) are sensitive to t up to ~20 and that for ice clouds having t > 20, the 3.7–10.8 µm and 3.7–6.7 µm BTDs are the most sensitive to De. Satellite imagery appears to be consistent with these results suggesting that t and De could be retrieved for greater optical thicknesses than previously assumed. But, because of sensitivity of the BTDs to uncertainties in the atmospheric profiles of temperature, humidity, and ice water content, and sensor noise, exploiting the small BTD signals in retrieval algorithms will be very challenging.

Recombinant human prostaglandin endoperoxide H synthase-1 (huPGHS-1) was characterized. huPGHS-1 has a single high-affinity heme binding site per dimer and exhibits maximal cyclooxygenase (COX) activity with one heme per dimer. Thus, huPGHS-1 functions as a conformational heterodimer having a catalytic monomer (Ecat) with a bound heme and an allosteric monomer (Eallo) lacking heme. The enzyme is modestly inhibited by common FAs including palmitic, stearic, and oleic acids that are not COX substrates. Studies of arachidonic acid (AA) substrate turnover at high enzyme-to-substrate ratios indicate that nonsubstrate FAs bind the COX site of Eallo to modulate the properties of Ecat. Nonsubstrate FAs slightly inhibit huPGHS-1 but stimulate huPGHS-2, thereby augmenting AA oxygenation by PGHS-2 relative to PGHS-1. Nonsubstrate FAs potentiate the inhibition of huPGHS-1 activity by time-dependent COX inhibitors, including aspirin, all of which bind Ecat. Surprisingly, preincubating huPGHS-1 with nonsubstrate FAs in combination with ibuprofen, which by itself is a time-independent inhibitor, causes a short-lived, time-dependent inhibition of huPGHS-1. Thus, in general, having a FA bound to Eallo stabilizes time-dependently inhibited conformations of Ecat. We speculate that having an FA bound to Eallo also stabilizes Ecat conformers during catalysis, enabling half of sites of COX activity.

A convenient, mild, reliable method has been developed for preparing oxazolines of fatty acids and for using these derivatives to determine double bond locations in long-chain polyunsaturated and polyconjugated fatty acids. Fatty acyl mixed anhydrides are prepared using isobutylchloroformate and then converted to their ethanolamides by treatment with ethanolamine. Ethanolamides are subsequently cyclized to the corresponding oxazolines in ≥85% yields by treatment with trifluoroacetic anhydride under mild conditions (>50° for 30–60 min). This general protocol can also be used to synthesize 4,4-dimethyloxazoline and benzoxazole derivatives of fatty acids. Gas chromatography-mass spectrometry of oxazoline derivatives of fatty acids yields prominent ions diagnostic of the structures of the parent fatty acids and, in the case of unsaturated fatty acids, indicating the positions of the double bonds.The utility of the method is illustrated with several fatty acids, including the conjugated 4E,6E,8E,10E,13Z,16Z,19Z-docosaheptaenoic acid.

Due to its direct approach and its detailed analysis of race, the "A More Perfect Union" (AMPU) speech makes for a likely primary source to be included in a lesson addressing Obama's racial significance. As social studies teacher-educators who draw from critical perspectives on race and racism, the authors hope to see Obama's speech used as a catalyst for nuanced, historicized conversations about race in the United States. The authors argue throughout this paper that the race speech has the pedagogical potential to create such conversations. Instead, we hope to underscore the potential for using Obama's AMPU speech as a vehicle for promoting a rich, nuanced understanding of race in America. We contend that this speech has the potential to foster both productive and unproductive conversations about race, reflecting theories of both racial literacy and racial liberalism. Examining these curricula does, however, provide a snapshot into how the field of education has begun to conceptualize Obama's historical narrative. Racial literacy calls for an intersectional analysis of race alongside other social factors such as class, gender, or geography. These lesson ideas can hopefully serve as a starting point for moving away from racial liberalist interpretations of the "A More Perfect Union" speech and toward developing greater racial literacy through Obama's iconic address.
