1. Multiomic interpretation of fungus-infected ant metabolomes during manipulated summit disease

Abstract

Camponotus floridanus ants show altered behaviors followed by a fatal summiting phenotype when infected with manipulating Ophiocordyceps camponoti-floridani fungi. Host summiting as a strategy to increase transmission is also observed with parasite taxa beyond fungi, including aquatic and terrestrial helminths and baculoviruses. The drastic phenotypic changes can sometimes reflect significant molecular changes in gene expression and metabolite concentrations measured in manipulated hosts. Nevertheless, the underlying mechanisms still need to be fully characterized. To investigate the small molecules producing summiting behavior, we infected C. floridanus ants with O. camponoti-floridani and sampled their heads for LC-MS/MS when we observed the characteristic summiting phenotype. We link this metabolomic data with our previous genomic and transcriptomic data to propose mechanisms that underlie manipulated summiting behavior in "zombie ants." This "multiomic" evidence points toward the dysregulation of neurotransmitter levels and neuronal signaling. We propose that these processes are altered during infection and manipulation based on (1) differential expression of neurotransmitter synthesis and receptor genes, (2) altered abundance of metabolites and neurotransmitters (or their precursors) with known behavioral effects in ants and other insects, and (3) possible suppression of a connected immunity pathway. We additionally report signals for metabolic activity during manipulation related to primary metabolism, detoxification, and anti-stress protectants. Taken together, these findings suggest that host manipulation is likely a multi-faceted phenomenon, with key processes changing at multiple levels of molecular organization.

Abstract

Magnetic skyrmions are quasiparticles with non-trivial topology, envisioned to play a key role in next-generation data technology while simultaneously attracting fundamental research interest due to their emerging topological charge. In chiral magnetic multilayers, current-generated spin-orbit torques or ultrafast laser excitation can be used to nucleate isolated skyrmions on a picosecond timescale. Both methods, however, produce randomly arranged skyrmions, which inherently limits the precision on the location at which the skyrmions are nucleated. Here, we show that nanopatterning of the anisotropy landscape with a He$^+$-ion beam creates well-defined skyrmion nucleation sites, thereby transforming the skyrmion localization into a deterministic process. This approach allows to realize control of individual skyrmion nucleation as well as guided skyrmion motion with nanometer-scale precision, which is pivotal for both future fundamental studies of skyrmion dynamics and applications.

Abstract

Multi-electron dynamics in atoms and molecules very often occur on sub- to few-femtosecond timescales. The available intensities of extreme-ultraviolet (XUV) attosecond pulses have previously only allowed the time-resolved investigation of two-photon, two-electron interactions. Here we demonstrate attosecond control over double and triple ionization of argon atoms involving the absorption of up to five XUV photons. In an XUV-pump XUV-probe measurement using a pair of attosecond pulse trains (APTs), the Ar$^{2+}$ ion yield exhibits a weak delay dependence, showing that its generation predominantly results from the sequential emission of two electrons by photoabsorption from the two APTs. In contrast, the Ar$^{3+}$ ion yield exhibits strong modulations as a function of the delay, which is a clear signature of the simultaneous absorption of at least two XUV photons. The experimental results are well reproduced by numerical calculations that provide detailed insights into the ionization dynamics. Our results open up new opportunities for the investigation and control of multi-electron dynamics and complex electron correlation mechanisms on extremely short timescales., Comment: 8 pages, 4 figures

Abstract

The ongoing development of intense high-harmonic generation (HHG) sources has recently enabled highly nonlinear ionization of atoms by the absorption of at least 10 extreme-ultraviolet (XUV) photons within a single atom [Senfftleben \textit{et al.}, arXiv1911.01375]. Here we investigate the role that reshaping of the fundamental, few-cycle, near-infrared (NIR) driving laser within the 30-cm-long HHG Xe medium plays in the generation of the intense HHG pulses. Using an incident NIR intensity that is higher than what is required for phase-matched HHG, signatures of reshaping are found by measuring the NIR blueshift and the fluorescence from the HHG medium along the propagation axis. These results are well reproduced by numerical calculations that show temporal compression of the NIR pulses in the HHG medium. The simulations predict that after refocusing an XUV beam waist radius of 320 nm and a clean attosecond pulse train can be obtained in the focal plane, with an estimated XUV peak intensity of 9x10^15 W/cm^2. Our results show that XUV intensities that were previously only available at large-scale facilities can now be obtained using moderately powerful table-top light sources., Comment: 17 pages, 8 figures

Abstract

Hard photons well above 100 GeV have to be generated in a future photon-collider which essentially will be based on the infrastructure of the planned International Linear Collider (ILC). The energy of near-infrared laser photons will be boosted by Compton backscattering against a high energy relativistic electron beam. For high effectiveness, a very powerful lasersystem is required that exceeds todays state-of-the-art capabilities. In this paper a design of an auxiliary passive cavity is discussed that resonantly enhances the peak-power of the laser. The properties and prospects of such a cavity are addressed on the basis of the specifications for the European TeV Energy Superconducting Linear Accelerator (TESLA) proposal. 11. Retail Display Properties and Consumer Perception of Extended Aged Beef Topically Treated with Ascorbic Acid and Rosemary Extract

Abstract

Extended aging increases browning and decreases redness in fresh beef. The goal of this study was to test an already proven combination of antioxidants (0.05% ascorbic acid + 0.1% rosemary extract) using a method that could be applied at the retail level to simply and effectively extend the shelf-life of extended aged beef. The specific objective was to analyze the effect of topical application of ascorbic acid and rosemary extract on color, lipid oxidation, microbial growth, and sensory perception of beef longissimus lumborum (LL; n = 12) and semimembranosus (SM; n = 12) muscles wet aged at 0°C for 14, 28, and 42 (extended aging period) days. After aging, steaks were cut, sprayed with 2 mL of a 0.05% ascorbic acid + 0.1% rosemary extract solution (treated) or untreated (control), and subjected to retail display. Antioxidant treated LL steaks had greater (P < 0.05) L* (lightness) values, but lower (P < 0.05) a* (redness) and b* (yellowness) values than control steaks. Furthermore, antioxidant treatment decreased (P < 0.05) browning on d 4 of retail display compared to control steaks. Consumers scored antioxidant treated SM steaks as less tender on d 28, more juicy on d 14 but less juicy on d 18 and 42. Antioxidant treatment did not affect lipid oxidation, microbial growth, or sensory flavor scores. As expected, longer aging periods resulted in less color stability of LL and SM steaks. Although the antioxidant treatment resulted in measurable subjective color improvements, these improvements are likely not detectable by the consumer.

Abstract

Multi-electron dynamics in atoms and molecules very often occur on sub- to few-femtosecond timescales. The available intensities of extreme-ultraviolet (XUV) attosecond pulses have previously only allowed the time-resolved investigation of two-photon, two-electron interactions. Here we demonstrate attosecond control over double and triple ionization of argon atoms involving the absorption of up to five XUV photons. In an XUV-pump XUV-probe measurement using a pair of attosecond pulse trains (APTs), the Ar$^{2+}$ ion yield exhibits a weak delay dependence, showing that its generation predominantly results from the sequential emission of two electrons by photoabsorption from the two APTs. In contrast, the Ar$^{3+}$ ion yield exhibits strong modulations as a function of the delay, which is a clear signature of the simultaneous absorption of at least two XUV photons. The experimental results are well reproduced by numerical calculations that provide detailed insights into the ionization dynamics. Our results open up new opportunities for the investigation and control of multi-electron dynamics and complex electron correlation mechanisms on extremely short timescales., Comment: 8 pages, 4 figures

Abstract

At the ELBE high power radiation facility, the second version of a superconducting radio frequency (SRF) photoinjector has been put into operation and is now routinely applied for user operation at the ELBE electron accelerator. SRF guns are suitable for generating continuous wave (CW) electron beam with high average currents and high beam brightness. The SRF gun at ELBE has the goal to generate short electron pulses with bunch charges of 200-300 pC at typical repetition rates of 100 kHz for the production of super radiant, coherent terahertz radiation. The SRF gun includes a 3.5-cell, 1.3 GHz niobium cavity and a superconducting solenoid. A support system with liquid nitrogen (LN2) cooling allows the operation of normal-conducting, high quantum efficiency photo cathodes. In the paper we present the design and performance of the SRF gun as well as beam measurements for the operation with Mg photocathodes and an acceleration gradient of 8 MV/m (4 MeV kinetic energy). Finally, we discuss the SRF gun application for production of coherent terahertz radiation at the ELBE facility.
