Abstract

Absorption covers the physical processes which convert intense photon flux into energetic particles when a high-power laser illuminates optically-thick matter. It underpins important petawatt-scale applications today, e.g., medical-quality proton beam production. However, development of ultra-high-field applications has been hindered since no study so far has described absorption throughout the entire transition from the classical to the quantum electrodynamical (QED) regime of plasma physics. Here we present a model of absorption that holds over an unprecedented six orders-of-magnitude in optical intensity and lays the groundwork for QED applications of laser-driven particle beams. We demonstrate 58% efficient \gamma-ray production at $1.8\times 10^{25}~\mathrm{W~ cm^{-2}}$ and the creation of an anti-matter source achieving $4\times 10^{24}\ \mathrm{positrons}\ \mathrm{cm^{-3}}$, $10^{6}~\times$ denser than of any known photonic scheme. Abstract

Proton radiography is a useful diagnostic of high energy density (HED) plasmas under active theoretical and experimental development. In this paper we describe a new simulation tool that interacts realistic laser-driven point-like proton sources with three dimensional electromagnetic fields of arbitrary strength and structure and synthesizes the associated high resolution proton radiograph. The present tool's numerical approach captures all relevant physics effects, including effects related to the formation of caustics. Electromagnetic fields can be imported from PIC or hydrodynamic codes in a streamlined fashion, and a library of electromagnetic field `primitives' is also provided. This latter capability allows users to add a primitive, modify the field strength, rotate a primitive, and so on, while quickly generating a high resolution radiograph at each step. In this way, our tool enables the user to deconstruct features in a radiograph and interpret them in connection to specific underlying electromagnetic field elements. We show an example application of the tool in connection to experimental observations of the Weibel instability in counterstreaming plasmas, using $\sim 10^8$ particles generated from a realistic laser-driven point-like proton source, imaging fields which cover volumes of $\sim10 $ mm$^3$. Abstract

Background: FGFR alterations are reported across various malignancies and might act as oncogenic drivers in multiple histologies. Erdafitinib is an oral, selective pan-FGFR tyrosine kinase inhibitor with activity in FGFR-altered advanced urothelial carcinoma. We aimed to evaluate the safety and activity of erdafitinib in previously treated patients with FGFR-altered advanced solid tumours. Methods: The single-arm, phase 2 RAGNAR study was conducted at 156 investigative centres (hospitals or oncology practices that are qualified oncology study centres) across 15 countries. The study consisted of four cohorts based on tumour histology and patient age; the results reported in this Article are for the primary cohort of the study, defined as the Broad Panel Cohort, which was histology-agnostic. We recruited patients aged 12 years or older with advanced or metastatic tumours of any histology (except urothelial cancer) with predefined FGFR1–4 alterations (mutations or fusions according to local or central testing). Eligible patients had disease progression on at least one previous line of systemic therapy and no alternative standard therapy available to them, and an Eastern Cooperative Oncology Group performance status of 0–1 (or equivalent for adolescents aged 12–17 years). Patients received once-daily oral erdafitinib (8 mg/day with provision for pharmacodynamically guided up-titration to 9 mg/day) on a continuous 21-day cycle until disease progression or intolerable toxicity. The primary endpoint was objective response rate by independent review committee according to Response Evaluation Criteria In Solid Tumors (RECIST), version 1.1, or Response Assessment In Neuro-Oncology (RANO). Abstract

We derive upper and lower bounds on the absorption of ultraintense laser light by solids as a function of fundamental laser and plasma parameters. These limits emerge naturally from constrained optimization techniques applied to a generalization of the laser-solid interaction as a strongly-driven, relativistic, two degree of freedom Maxwell-Vlasov system. We demonstrate that the extrema and the phase-space-averaged absorption must always increase with intensity, and increase most rapidly when $10^{18} < I_L \ \lambda_L^2 < 10^{20}$ W $\mu$m$^2/$cm$^{2}$. Our results indicate that the fundamental empirical trend towards increasing fractional absorption with irradiance therefore reflects the underlying phase space constraints.

Abstract

Two dimensional particle-in-cell simulations characterizing the interaction of ultraintense short pulse lasers in the range 10^{18} \leq I \leq 10^{20} W/cm^{2} with converging target geometries are presented. Seeking to examine intensity amplification in high-power laser systems, where focal spots are typically non-diffraction limited, we describe key dynamical features as the injected laser intensity and convergence angle of the target are systematically varied. We find that laser pulses are focused down to a wavelength with the peak intensity amplified by an order of magnitude beyond its vacuum value, and develop a simple model for how the peak location moves back towards the injection plane over time. This performance is sustained over hundreds of femtoseconds and scales to laser intensities beyond 10^{20} W/cm^{2} at 1 \mu m wavelength., Comment: 5 pages, 6 figures, accepted for publication in Physics of Plasmas

The proposed experiments should be able to detect a measurable number of gamma rays resulting from the annihilation of pairs in the focal spot of two opposing high intensity laser beams. Patients were randomized 1:1 to APF530 500 mg subcutaneously (granisetron 10 mg) or ondansetron 0.15 mg/kg intravenously (IV) (≤16 mg); stratification was by planned cisplatin ≥50 mg/m2 (yes/no). Patients were to receive fosaprepitant 150 mg IV and dexamethasone 12 mg IV on day 1, then dexamethasone 8 mg orally once daily on day 2 and twice daily on days 3 and 4. Patients were mostly younger females (APF530 arm, mean age 54.1 years, female, 99.3%; ondansetron arm, 53.8 years, female 98.3%). The primary end point was delayed-phase (>24–120 hours) complete response (CR). Results: APF530 versus ondansetron regimens achieved numerically better CINV control in delayed and overall (0–120 hours) phases for CR, complete control, total response, rescue medication use, and proportion with no nausea. APF530 trends are consistent with the overall population, although not statistically superior given the underpowered AC subgroup analysis. The APF530 regimen in this population was generally well tolerated, with safety comparable to that of the overall population.Conclusion: APF530 plus fosaprepitant and dexamethasone effectively prevented CINV among patients receiving AC-based HEC, a large subgroup in whom CINV control has traditionally been challenging. Primary data from a Phase II trial for the first-line combination of ­eribulin plus trastuzumab in human epidermal growth factor receptor 2 positive patients showed a 71% objective response rate and tolerability consistent with the known profile of these agents. Here, we present prespecified analyses of efficacy of this combination based on prior trastuzumab use. Patients received eribulin mesylate 1.4 mg/m2 (equivalent to 1.23 mg/m2 eribulin [expressed as free base]) intravenously on days 1 and 8 plus trastuzumab (8 mg/kg intravenously/cycle 1, then 6 mg/kg) on day 1 of each 21-day cycle. Objective response rates, progression-free survival, and tolerability were assessed in patients who had and had not received prior adjuvant or neoadjuvant (neo/adjuvant) trastuzumab treatment. Fifty-two patients (median age: 59.5 years) received eribulin/trastuzumab for a median treatment duration of ~31 weeks; 40.4% (n=21) had been previously treated with neo/adjuvant trastuzumab prior to treatment with eribulin plus trastuzumab for metastatic disease (median time between neo/adjuvant and study treatment: 23 months). In trastuzumab-naïve patients (n=31) compared with those who had received prior trastuzumab, objective response rate was 77.4% versus 61.9%, respectively; duration of response was 11.8 versus 9.5 months, respectively; clinical benefit rate was 87.1% versus 81.0%, respectively; and median progression-free survival was 12.2 versus 11.5 months, respectively. The most common grade 3/4 treatment-emergent adverse events (occuring in ≥5% of patients) in patients who received prior trastuzumab versus trastuzumab naïve patients, respectively, were neutropenia (47.6% vs 32.3%), peripheral neuropathy (14.3% vs 25.8%), febrile neutropenia (14.3% vs 3.2%), fatigue (9.5% vs 6.5%), nausea (9.5% vs 0%), vomiting (9.5% vs 3.2%), and leukopenia (9.5% vs 3.2%). In patients with human epidermal growth factor receptor 2 positive metastatic breast cancer, first-line eribulin/trastuzumab treatment demonstrated substantial antitumor activity and was well tolerated, regardless of prior neo/adjuvant trastuzumab treatment. The impact of memory induced by prior infection is rarely considered and is difficult to ascertain, because infections are often subclinical. This study investigated influenza vaccination among adults from the Ha Nam cohort (Vietnam), who were purposefully selected to include 72 with and 28 without documented influenza A(H3N2) infection during the preceding 9 years (Australian New Zealand Clinical Trials Registry 12621000110886). The primary outcome was the effect of prior influenza A(H3N2) infection on hemagglutinin-inhibiting antibody responses induced by a locally available influenza vaccine administered in November 2016. Baseline and postvaccination sera were titrated against 40 influenza A(H3N2) strains spanning 1968-2018. At each time point (baseline, day 14 and day 280), geometric mean antibody titers against 2008-2018 strains were higher among participants with recent infection (34 (29-40), 187 (154-227) and 86 (72-103)) than among participants without recent infection (19 (17-22), 91 (64-130) and 38 (30-49)). On days 14 and 280, mean titer rises against 2014-2018 strains were 6.1-fold (5.0- to 7.4-fold) and 2.6-fold (2.2- to 3.1-fold) for participants with recent infection versus 4.8-fold (3.5- to 6.7-fold) and 1.9-fold (1.5- to 2.3-fold) for those without. One of 72 vaccinees with recent infection versus 4 of 28 without developed symptomatic A(H3N2) infection in the season after vaccination (P = 0.021). The range of A(H3N2) viruses recognized by vaccine-induced antibodies was associated with the prior infection strain. Abstract

In this presentation we provide an overview of the commissioning and first user experiment of ReLaX in combination with the XFEL beam. We will introduce the setup and laser parameters. We will show the established standard setup combining small-angle x-ray scattering (SAXS), phase-contrast imaging (PCI) and spectroscopy techniques. Finally we will show examples of the data obtained in the interaction of the ReLaX laser with targets of interested for the short-pulse laser community.

Abstract

The High Energy Density (HED) instrument at the European XFEL provides a platform to study hot and warm dense matter. The Helmholtz International Beamline for Extreme Fields (HiBEF) is a User Consortium supplying HED with two laser systems (the high-intensity ReLaX laser, by Amplitude Technologies, and the high-energy Dipole-100X laser, by STFC), Diamond Anvil Cells setup and high-pulsed magnetic fields. These tools in combination with the XFEL beam enable the investigation of relativistic laser plasmas, strong-field QED phenomena, high-pressure astro- and planetary physics as well as magnetic phenomena in condensed matter. The successful commissioning of the ultra-short pulse high-intensity ReLaX laser, provides new unique opportunities in the plasma and high-field physics fields [1]. ReLaX is a double CPA Ti:Sa laser capable of delivering up to 300 TW pulses on target. In the first commissioning phase, 100 TW pulses were used, reaching intensities up to 1020 W/cm2. Small-Angle X-Ray Scattering (SAXS) without the need of a beamstop was first commissioned at HED in September 2019. Two highly annealed pyrolytic graphite (HAPG) crystals were used to reflect the SAXS photons onto a detector while allowing the main XFEL beam to go through [2]. In April and May 2021, Small Angle X-Ray Scattering and Phase Contrast Imaging (PCI) were simultaneously demonstrated in pump-probe experiments at HED in a community experiment involving 15 institutions from all over the world. In this talk we will present the preliminary results of this community experiment probing ultrafast phenomena in a wide array of target configurations: hole boring in wires, shockwave generation in CH blocks, buried heating of a wire inside a CH medium, foam ionization and collective effects in heated foils. Abstract

Computer models of intense, laser-driven ion acceleration require expensive particle-in-cell simulations that may struggle to capture all the multi-scale, multi-dimensional physics involved at reasonable costs. Explored is an approach to ameliorate this deficiency using a multi-fidelity framework that can incorporate physical trends and phenomena at different levels. As the basis for this study, an ensemble of approximately 8000 1D simulations was generated to buttress separate ensembles of hundreds of higher fidelity 1D and 2D simulations. Using transfer learning with deep neural networks, one can reproduce the results of more complex physics at a much lower cost. The networks trained in this fashion can, in turn, act as surrogate models for the simulations themselves, allowing for quick and efficient exploration of the parameter space of interest. Standard figures-of-merit were used as benchmarks such as the hot electron temperature, peak ion energy, conversion efficiency, and so on. We can rapidly identify and explore under what conditions differing fidelities become an important effect and search for outliers in feature space. [ABSTRACT FROM AUTHOR]

Compared student performance on computer-based assessment to machine-graded multiple choice tests. Found that performance improved dramatically on the computer-based assessment when students were not required to scroll through the question paper. Concluded that students may be disadvantaged by the introduction of online assessment unless care is taken with the student-assessment interface. (EV)
