Abstract

In this note, we give a self-contained proof of the following classification (up to conjugation) of finite subgroups of GSpn(F̅ℓ) containing a nontrivial transvection for ℓ ≥ 5, which can be derived from work of Kantor: G is either reducible, symplectically imprimitive or it contains Spn(F̅ℓ). This result is for instance useful for proving ‘big image’ results for symplectic Galois representations.

4. Scaling Up of Steric Exclusion Membrane Chromatography for Lentiviral Vector Purification

Abstract

Lentiviral vectors (LVs) are widely used in clinical trials of gene and cell therapy. Low LV stability incentivizes constant development and the improvement of gentle process steps. Steric exclusion chromatography (SXC) has gained interest in the field of virus purification but scaling up has not yet been addressed. In this study, the scaling up of lentiviral vector purification by SXC with membrane modules was approached. Visualization of the LVs captured on the membrane during SXC showed predominant usage of the upper membrane layer. Furthermore, testing of different housing geometries showed a strong influence on the uniform usage of the membrane. The main use of the first membrane layer places a completely new requirement on the scaling of the process and the membrane modules. When transferring the SXC process to smaller or larger membrane modules, it became apparent that scaling of the flow rate is a critical factor that must be related to the membrane area of the first layer. Performing SXC at different scales demonstrated that a certain critical minimum surface area-dependent flow rate is necessary to achieve reproducible LV recoveries. With the presented scaling approach, we were able to purify 980 mL LVs with a recovery of 68%.

5. Steric Exclusion Chromatography for Purification of Biomolecules—A Review

Abstract

Steric exclusion chromatography (SXC) is a purification method that is based on steric exclusion effects from the surface of the target and a hydrophilic stationary phase after the addition of polyethylene glycol (PEG), which leads to an association of the target with the stationary phase without direct binding, such as covalent, electrostatic, and hydrophilic/hydrophobic interactions. The gentle nature of the method has led to an increased focus on sensitive targets such as enveloped viruses with potential for other sensitive entities, e.g., extracellular vesicles and virus-like particles. SXC is related to PEG-mediated protein precipitation, but investigation of further process parameters was crucial to gain a better understanding of the SXC method. After explaining mechanistic fundamentals and their discovery, this review summarizes the findings on SXC from its first reference 11 years ago until today. Different applications of SXC are presented, demonstrating that the method can be used for a wide variety of targets and achieves high recovery rates and impurity removal. Further, critical process parameters for successful process implementation are discussed, including technical requirements, buffer composition, and scalability.

11. Infectious titer determination of lentiviral vectors using a temporal immunological real-time imaging approach

Abstract

The analysis of the infectious titer of the lentiviral vector samples obtained during upstream and downstream processing is of major importance, however, also the most challenging method to be performed. Currently established methods like flow cytometry or qPCR lack the capability of enabling high throughput sample processing while they require a lot of manual handling. To address this limitation, we developed an immunological real-time imaging method to quantify the infectious titer of anti-CD19 CAR lentiviral vectors with a temporal readout using the Incucyte® S3 live-cell analysis system. The infective titers determined with the Incucyte® approach when compared with the flow cytometry-based assay had a lower standard deviation between replicates and a broader linear range. A major advantage of the method is the ability to obtain titer results in real-time, enabling an optimal readout time. The presented protocol significantly decreased labor and increased throughput. The ability of the assay to process high numbers of lentiviral samples in a high throughput manner was proven by performing a virus stability study, demonstrating the effects of temperature, salt, and shear stress on LV infectivity.

15. Butterfly curves and critical modes for second-order spin-wave instability processes in yttrium iron garnet films

Abstract

The second-order spin-wave instability threshold microwave field amplitudes and the critical modes for ferromagnetic resonance saturation in 4.15 micron-thick yttrium iron garnet films at 8.47 GHz with the static magnetic field in-plane, were determined by combined Brillouin light-scattering and microwave pumping methods. Findings revealed a new king of butterfly curve for second-order processes, strong scattering from modes at low wave numbers in the 10000 rad/cm range and in-plane propagation direction for low wave number modes at 90 degrees to the in-plane static field direction.

Abstract

In-plane magnetized yttrium iron garnet thin films exhibit a fine structure in the parallel pumping spin wave instability absorption at 9.4 gigahertz, and is in accordance with the critical mode theory. The standing wave critical mode explains this fine structure in terms of cooperative volume dipole-dipole and shape demagnetization induced ellipticity. The fine structure spacing is of the order of 5 Oe, and square of the spacing decreases linearly with increase in static field.

Abstract

A 12.8 micrometer thick single crystal yttrium iron garnet film is experimented upon for measuring its butterfly curves of the subsidiary absorption spin wave instability threshold microwave field amplitude versus static field at 10 gigahertz for two field configurations. One of the field configurations has the static field normal to the film plane and the microwave field in plane (PI case), and the other has a static magnetic field in plane and the microwave field out of plane (IP case). The measurements obtained for the IP case show a 'thick' in the butterfly curve having a jump of the critical mode wave number K at the kind field. No kind effect is observed for the PI case. A new theory which considers the discrete standing wave modes in the films explains the different butterfly curves for the two cases.

Abstract

A new model was used to describe multistability and chaos at parametrically excited spherical modes in an yttrium iron garnet sphere. The model used the true eigenmodes of the sphere insteads of plane spin waves. It was shown that the half pumping frequency below the 45 degree spin wave branch featured critical spherical modes with efficiencies of 1.5x10(super 5) cm(super -1). This was attributed to the effects of surface pit scattering.
