Abstract

Infeksi virus dengue (DENV) menjadi ancaman kesehatan global yang terus mengalami peningkatan kasus pada setiap tahun terutama pada musim penghujan, di daerah beriklim tropis maupun subtropis. Vaksin dengue yang tersedia saat ini berbahan dasar virus yang dilemahkan dinilai memiliki potensi antibody-dependent enhancement (ADE) yang memicu terbentuknya badai sitokin, berujung pada kondisi imunopatogenik. Melalui pendekatan imunoinformatika, penelitian ini bertujuan untuk mengidentifikasi peptida sebagai epitop potensial dari protein selubung DENV serotipe 1-4 yang dapat berinteraksi dengan salah satu HLA dominan dan paling banyak dipelajari yaitu HLA-A*02:01, serta HLA-A*24:02 dan HLA-A*24:07, keduanya merupakan HLA kelas I yang paling dominan di populasi Indonesia. Sebanyak 43 protein selubung DENV disejajarkan dan diamati nilai entropi untuk memilih satu urutan residu dari setiap serotipe DENV yang lestari. Penyaringan imunoinformatika dilanjutkan sampai mendapatkan peptida yang memenuhi kriteria seleksi epitop sebagai kandidat epitop vaksin dengue yaitu bersifat antigenik, tidak homolog dengan protein manusia, bukan alergen, dan tidak toksik. Didapatkan 20 peptida memenuhi kriteria diantaranya 10/20 untuk HLA-A*02:01 serta 4/17 dan 6/17 yang mengikat HLA-A*24:02 dan HLA-A*24:07 secara berurutan. Penelitian ini merupakan awal dalam perancangan vaksin peptida dengue yang masih harus dilanjutkan dan dibuktikan secara in silico dengan melihat interaksi molekuler tiga dimensi antara kandidat epitop dengan masing – masing alel.

Abstract

Lipid nanoparticles (LNPs) are the delivery system behind the success story of mRNA vaccines and the development of DNA and peptide vaccines. LNPs should have particle sizes ranging from 60 to 150 nm for such applications. Achieving such particle size criteria can be challenging in the LNP preparation using a micromixer system with different parameters: i.e., the N/P ratio, aqueous flow rate (AFR) and total flow rate (TFR). Such an issue can be addressed by applying response surface methodology (RSM). Therefore, in this study, we implemented RSM to optimize LNP preparation parameters, including the N/P ratio and AFR. TFR remained constant at 1 mL.min-1 since, from a previous study, it is not a critical parameter for achieving the desired LNP size. Here, we used the pDNA of ACE-2 as a dummy nucleotide. Particle sizes of the resulting LNPs were determined using a dynamic light scattering (DLS) method. Our results indicate that AFR is a significant factor in determining LNP size, whereas the N/P ratio does not have a significant impact on LNP size. We achieved an LNP size of 104.00 nm by applying the N/P ratio of 32.0000 and AFR of 0.9621 mL.min-1. Our results showed that RSM could ease the optimization process in the preparation of LNP, which may support the acceleration of vaccine development. [ABSTRACT FROM AUTHOR]

Abstract

Environmental load issues in Jakarta, economic contribution to GDP, Java's dominance in land conversion, economic justice, and a water supply dilemma are all factors that contribute to Jakarta's excessive density. Therefore, the Indonesian government intends to relocate the Indonesia’s capital to the Penajam Paser Utara district, in the East Kalimantan Province, in 2024. One of the challenges for the new capital, known as Indonesia's new capital (IKN), is securing a sustainable raw water supply for drinking. On the other hand, the potential of existing water resources is very limited in terms of resources and supporting infrastructure. This paper investigates the forecasting and sustainability of raw water supply for Indonesia’s new capital. The hydrological feasibility method, which includes calculating the estimated supply and evaluating the sustainability of IKN raw water sources, was used in this study. The result showed the estimated raw water requirements for the IKN area during the period 2023- 2073. The critical point of the resulting state is that the water resources of IKN by 2023 will be secure if the infrastructure plan is properly implemented. Meanwhile, IKN will be in deficit after 2045. Abstract

Tomatoes (Solanum lycopersicum Mill.), a common vegetable in Indonesia, contain high levels of lycopene, which is good for the body. This research further investigates the activity of polar and nonpolar fractions of tomatoes as elastase and tyrosinase inhibitory, and cytotoxic agents. The extraction procedure used is maceration, fractionation through liquid-liquid fractionation, purification of phytochemical substances is achieved through the application of thin layer chromatography. Elastase and tyrosinase inhibitory activity was analyzed using spectrophotometry and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide cytotoxic assay. The result showed that the extract yield was 0.004%. The percentage of polar fraction from the extract was 2.58%, while the nonpolar fraction was 0.69%. The elastase inhibitory activity of polar and nonpolar fractions of tomato extract is 87.21% ± 7.57% and 73.12% ± 7.44%, respectively, The elastase inhibitory activity of polar and nonpolar fractions of tomato extract is 87.21% ± 7.57% and 73.12% ± 7.44%, respectively. The fractions had higher the anti-elastase activity than the positive control quercetin (65.97% ± 3.00%). The tyrosinase inhibitory activity of polar and nonpolar fractions of tomato extract is 23.71% ± 7.91% and 41.16% ± 5.41% (kojic acid as standard is 65.07% ± 0.86%), respectively. The IC50 of the cytotoxic assay to NIH 3T3 mouse embryonic fibroblast cells of the polar and nonpolar fraction of tomato extract is 1820.90 μg/mL and 1643.86 μg/mL, respectively. Oleh karena itu peneliti terdorong untuk mencari solusi atas permasalahan tersebut dengan upaya merekonstruksi teori motivasi Abraham Maslow agar teori tersebut dapat diterima semua masyarakat. Untuk menunjang keberhasilan penelitian peneliti menggunakan pendekatan kualitatif yang diformulasikan dengan intisari pemikiran Jurgen Habermas sehingga penelitian ini mengutamakan penggunaan komunikasi sebagai jalur pencerahan dari pemecahan permasalahan tersebut. Dalam penelitian ini juga menggunakan pendekatan studi kritis dan dilakukan di kantor akuntansi publik (KAP) yang ada di Malang sehingga informan dalam penelitian ini adalah auditor junior maupun auditor senior. Hasil penelitian menunjukkan teori motivasi Abraham Maslow masih belum sempurna dan memiliki sedikit noda didalamnya. Abstract

Nowadays many department (community) are thinking how to get more knowledges and metadata by linking more systems from other community. There are great challenges to make all systems organizing knowledge and sharing metadata – to make it easy searched, indexed and used in different context. In this paper we will focus on metadata in specific domain - ‘Poverty’2. Regardless of the various definitions of poverty, in this paper we will focus on managing metadata in “Poverty” with many different terms therein. Ontology Mapping is the process of relating similar concepts or relations from different sources through some equivalence relation. Mapping allows finding correspondences between the concepts of two ontologies. If two concepts correspond, then they mean the same thing or closely related things. Currently, the mapping process is regarded as a promise to solve the problem between ontologies since it attempts to find correspondences between semantically related entities that belong to different ontologies. It takes as input two ontologies, each consisting of a set of components (classes, instances, properties, rules and axioms). Based on the presented reasons, we believe that ontologies with common terms and common concepts are very important in a metadata sharing process.

Abstract

The purpose of this paper is to provide research based understanding of leveraging knowledge and managing knowledge within and across several communities using the poverty domain as a case study. We hypothesize that leveraging knowledge with a good taxonomy and a good integration process are good approaches to organize and share knowledge. Problems appear when a group of people in different communities share data and collaborate using different perceptions, different concepts, different terms (terminologies), and different semantics to represent the same reality. In this paper we present an approach to solve this problem. We will generate a common set of terms based on the terms of several different storage devices, used by different communities, in order to make data retrieval independent of the different perceptions and terminologies used by those communities. We use ontologies to represent the particular knowledge of each community and discuss the use of mapping and integration techniques to find correspondences between the concepts used in those ontologies.

Abstract

Air merupakan merupakan kebutuhan yang sangat vital bagi kehidupan. Jika kebutuhanakan air belum terpenuhi baik secara kuantitas maupun kualitas, maka akan menimbulkan dampak yang besar terhadapkerawanan kesehatan maupun sosial. Kualitas air meliputi persyaratan fisik, kimia dan bakteriologis yang merupakan suatu kesatuan, sehingga apabila ada satu parameter yang tidak memenuhi syarat, maka air tersebut tidak layak untuk digunakan. Salah satu parameter kimia dalam persyaratan kualitas air adalah jumlah kandungan unsur Ca2+ dan Mg2+ dalam air yang keberadaannya biasa disebut kesadahan air. Kesadahan dalam air sangat tidak dikehendaki baik untuk penggunaan rumah tangga maupun untuk penggunaan industri. Untuk mengatasi permasalahan tersebut, salah satu alternatif adalah mengolah air tanah atau air sumur setempat sehingga didapatkan air siap minum yang dengan kualitas yang memenuhi persyaratan.

Air tanah di daerah bergambut atau daerah rawa umumnya dangkal berwarnacoklat, berkadar asam humus, zat organik dan besi yang tinggi, sedangkandidaerah daratan agak dalam dengan air berwarna jernih tetapi kadar besi danmangan masih tinggi. Untuk mengatasi masalah air bersih di daerah bergambutperlu adanya alat pengolahan air gambut yang dapat dipakai untuk memenuhikebutuhan air bersih masyarakat setempat.Alat Pengolahan air gambut secara kontinyu ini merupakan rangkaian prosesyabg lengkap namun dikemas dalam bentuk yang sederhana, dirancang sesuaidengan kondisi dan tingkat pendidikan masyarakat pedesaan. Dengan demikian alat pengolah air gambut secara kontinyu ini harus murah, mudah pengerjaan dan pengoperasiannya serta hasil olahan yang memenuhi baku mutu air minum.

49. PEMASYARAKATAN UNIT PENGOLAHAN AIR SIAP MINUM SKALA INDUSTRI KECIL

Abstract

Berdasarkan data ststistik 1995, prosentasi banyaknya rumah tangga dan sumber air minum yang digunakan di berbagai daerah di Indonesia sangat bervariasi tergantung dari kondisi geografisnya. Secara nasional yakni sebagai berikut : Yang menggunakan air leding 16,08 %, air tanah dengan memakai pompa 11,61 %, air sumur (perigi) 49,92 %, mata air (air sumber) 13,92 %, air sungai 4,91 %, air hujan 2,62 % dan lainnya 0,80 %. Permasalahan yang timbul yakni sering dijumpai bahwa kulaitas air tanah maupun air sungai yang digunakan masyarakat kurang memenuhi syarat sebagai air minum yang sehat bahkan di beberapa tempat bahkan tidak layak untuk diminum. Air yang layak diminum, mempunyai standar persyaratan tertentu yakni persyaratan fisis, kimiawi dan bakteriologis, dan syarat tersebut merupakan satu kesatuan. Jadi jika ada satu saja parameter yang tidak memenuhi syarat maka air tersebut tidak layak untuk diminum. Untuk daerah kawasan pemukiman yang telah dibangun di daerah yang kualitas air tanahnya buruk, serta belum mendapatkan pelayanan air bersih dari PAM setempat maka masyarakat terpaksa memenuhi kebutuhan air minum mereka dengan cara membeli air minum kemasan dengan harga yang mahal. Untuk menanggulangi masalah tersebut, salah satu alternatif yakni dengan cara mengolah air tanah atau air sumur sehingga didapatkan air siap minum dengan kualitas yang memenuhi syarat kesehatan. Untuk mengatasi permasalahan tersebut di atas, yakni dengan caramengembangkan dan memasyarakatkan paket teknologi untuk mengolah air sumur atau air PAM menjadi air yang dapat langsung diminum tanpa dimasak terlebih dahulu. Unit alat tersebut terdiri dari antara lain : pompa air baku, filter bertekanan, filter mangan zeolit, filter karbon aktif, cartridge filter, sterilisator ultra violet dan ozon generator. Tujuan dari kegiatan ini adalah memasyarakatkan teknologi pengolahan air siap minum yakni air yang dapat langsung diminum tanpa dimasak untuk skala industri kecil (kapasitas 10.000 - 20.000 liter per hari air siap minum) yang hasilnya dapat dijual kepada masyarakat dengan harga yang jauh lebih murah dibandingkan dengan apabila membeli air minum kemasan. Sasaran kegiatan ini adalah membagun satu percontohan unit pengolahan air siap minum di dalam suatu kawasan pemukimnan yang rawan air bersih yang dapat dikelola oleh koperasi atau unit usaha setempat dan hasilnya dapat dijual kepada masyarakat setempat dengan harga yang lebih murah. Diharapkan hasil penjualan dari air tersebut dapat memberikan peluang usaha dan dapat digulirkan ke tempat lain.

Abstract

Tanjung Aru is a village located in cape of Tanjung Aru, East Kalimantan. The community living in Tanjung Aru use surface water from wells as the main clean water resource. The surface water is so influenced by the tide of sea water. The surface taste is very salty (DHL>1500 mMhos/cm). The water is not only salty but also the turbidity is much more than water quality standart. The use of rain water as the second alternative is very limited, it is only in rainy season.To deal with the chronic problem, such as the lack of clean water supply, it needs an appropriate water treatment technology. The suitable water treatment system is a combination of conventional and advanced technology. Desalination, such as Reverse Osmosis must be involved to reduce salinity of the raw water. A complete process includes the pretreatment and advance treatment. The pretreatment are oxidation and some common filtrations. The advance treatment is a molecular filtration using a membran which the principal is reverse osmosis pressure. If the pilot water treatment plant is avalaible in Tanjung Aru, the clean water supply will not be a serious problem. Generally, it can also play an important role to increase the social level of community in East Kalimantan.
