The magnitude of the COOVID-19 pandemic has stimulated research on the treatment and diagnosis of COVID-19 patients.Objective: In this report, a battery of specific parameters was used to develop a model that allows prediction of the outcome of the COVID-19 treatment. These parameters are C-reactive protein, procalcitonin, fibrinogen, D-dimers, immature granulocytes, and interleukin-6.Methods: The study was carried out on a sample of N = 49 survivors (22 men, 27 women) and 83 deceased patients (62 men, 21 women). The distribution of means and differences in means of the parameters studied between survivors and deceased patients were evaluated using the bootstrap method.Results: A mathematical model that allows for the prediction of hospitalization outcome was obtained using the Naive Bayes model. The results demonstrated a statistically significant difference between survivors and deceased patients in all parameters studied. A mathematical model employing a battery of parameters provided a 97% precision in predicting the outcome of hospitalization.Conclusion: This study showed that the cross-correlation of survivability with absolute levels of C-reactive protein, procalcitonin, fibrinogen, D-dimers, immature granulocytes, and interleukin-6 could be used successfully in the hospital setting as a diagnostic tool.Keywords: Covid-19, biological markers, C-reactive protein, procalcitonin, fibrinogen, D-dimers, immature granulocytes, interleukin-6

Objective The purpose of this cross-sectional study was to assess the association of the menopausal transition with changes in vitamins. Methods The study group comprised women aged 17-85 years from the Third National Health and Nutrition Examination Survey (NHANES), which was conducted between 1988 and 1994, and from the NHANES surveys conducted between 1999 and 2006. Menopausal status was defined using the time since the last period, < 2, 2-12, and > 12 months, for the pre-, peri-, and postmenopause, respectively. The data-cleaning technique employing serum follicle stimulating hormone activity resulted in pre-, peri- and postmenopausal samples encompassing the following age brackets: 17-50, 42-51, and 46-85 years. Statistical inferences were analyzed using non-parametric techniques. Results Significant increases in vitamin A and vitamin E concentrations across all phases of the menopausal transition were observed. There was a gradual decrease in the vitamin C concentration across all stages of the menopause but a fairly stable concentration of vitamin B12. There was a statistically significant increase in vitamin D between the pre- and postmenopause. Body mass index correlated negatively with serum vitamin concentration in the pre- and postmenopause. Conclusions Vitamin A should be supplemented in postmenopausal women to decrease the risk of bone fracture. The daily diet should be supplemented with vitamin B12, to avoid possible neurological symptoms due to vitamin B12 deficiency, and with vitamin D to decrease the risk of developing secondary hyperparathyroidism. Due to an adverse influence on serum vitamin concentration, body mass index should be monitored in pre- and postmenopausal women. [ABSTRACT FROM AUTHOR]

Allergic diseases triggered by airborne allergens such as allergic rhinitis and conjunctivitis are increasingly prevalent, posing significant challenges for both patients and healthcare systems. Assessing the efficacy of allergen immunotherapy and other anti-allergic treatments requires precise and reproducible methods. Allergen exposure chambers (AECs) have emerged as advanced tools for evaluating clinical outcomes, offering controlled conditions that address many limitations of traditional field-based studies. This review explores the advantages of AECs in allergy management, emphasizing their role in providing standardized allergen exposure for both clinical research and routine assessments. AECs deliver consistent and reproducible data comparable to the nasal allergen challenge and natural allergen exposure, making them a valuable addition to the diagnosis and treatment effectiveness of allergic diseases. Although they are well suited to early-stage clinical trials, further standardization and validation are needed to gain broader acceptance in pivotal phase III studies. Future research should focus on refining AEC protocols and integrating them into regulatory frameworks, ensuring their role in the advancement of therapeutic approaches for allergic diseases.

Plastics are used all over the world. Unfortunately, due to limited biodegradation, plastics cause a significant level of environmental pollution. The smallest recognized to date are termed nanoplastics (1 nm [nm] up to 1 μm [μm]) and microplastics (1 μm-5 mm). These nano- and microplastics can enter the human body through the respiratory system via inhalation, the digestive tract via consumption of contaminated food and water, or penetration through the skin via cosmetics and clothes contact. Bioaccumulation of plastics in the human body can potentially lead to a range of health issues, including respiratory disorders like lung cancer, asthma and hypersensitivity pneumonitis, neurological symptoms such as fatigue and dizziness, inflammatory bowel disease and even disturbances in gut microbiota. Most studies to date have confirmed that nano- and microplastics can induce apoptosis in cells and have genotoxic and cytotoxic effects. Understanding the cellular and molecular mechanisms of plastics' actions may help extrapolate the risks to humans. The article provides a comprehensive review of articles in databases regarding the impact of nano- and microplastics on human health. The review included retrospective studies and case reports of people exposed to nanoplastics and microplastics. The blueprint of the mammalian body plan is laid out during gastrulation, when a trilaminar embryo is formed. This process entails a burst of proliferation, the ingression of embryonic epiblast cells at the primitive streak, and their priming toward primitive streak fates. How these different events are coordinated remains unknown. Here, we developed and characterized a 3D culture of self-renewing mouse embryonic cells that captures the main transcriptional and architectural features of the early gastrulating mouse epiblast. Using this system in combination with microfabrication and in vivo experiments, we found that proliferation-induced crowding triggers delamination of cells that express high levels of the apical polarity protein aPKC. Upon delamination, cells become more sensitive to Wnt signaling and upregulate the expression of primitive streak markers such as Brachyury. Micro- and nanoplastic particles, including common forms like polyethylene and polystyrene, have been identified as relevant pollutants, potentially causing health problems in living organisms. The mechanisms at the cellular level largely remain to be elucidated. This study aims to visualize nanoplastics in bronchial smooth muscle (BSMC) and small airway epithelial cells (SAEC), and to assess the impact on mitochondrial metabolism. Healthy and asthmatic human BSMC and SAEC in vitro cultures were stimulated with polystyrene nanoplastics (PS-NPs) of 25 or 50 nm size, for 1 or 24 h. Live cell, label-free imaging by holotomography microscopy and mitochondrial respiration and glycolysis assessment were performed. Furthermore, 25 and 50 nm NPs were shown to penetrate SAEC, along with healthy and diseased BSMC, and they impaired bioenergetics and induce mitochondrial dysfunction compared to cells not treated with NPs, including changes in oxygen consumption rate and extracellular acidification rate. NPs pose a serious threat to human health by penetrating airway tissues and cells, and affecting both oxidative and glycolytic metabolism.

The importance of allergen immunotherapy (AIT) is multifaceted, encompassing both clinical and quality-of-life improvements and cost-effectiveness in the long term. Key mechanisms of allergen tolerance induced by AIT include changes in memory type allergen-specific T- and B-cell responses towards a regulatory phenotype with decreased Type 2 responses, suppression of allergen-specific IgE and increased IgG 1 and IgG 4 , decreased mast cell and eosinophil numbers in allergic tissues and increased activation thresholds. The potential of novel patient enrolment strategies for AIT is taking into account recent advances in biomarkers discoveries, molecular allergy diagnostics and mobile health applications contributing to a personalized approach enhancement that can increase AIT efficacy and compliance. Artificial intelligence can help manage and interpret complex and heterogeneous data, including big data from omics and non-omics research, potentially predict disease subtypes, identify biomarkers and monitor patient responses to AIT. Novel AIT preparations, such as synthetic compounds, innovative carrier systems and adjuvants, are also of great promise. Advances in clinical trial models, including adaptive, complex and hybrid designs as well as real-world evidence, allow more flexibility and cost reduction. The analyses of AIT cost-effectiveness show a clear long-term advantage compared to pharmacotherapy. Important research questions, such as defining clinical endpoints, biomarkers of patient selection and efficacy, mechanisms and the modulation of the placebo effect and alternatives to conventional field trials, including allergen exposure chamber studies are still to be elucidated. During AIT, ILC2 and ILC3 decrease while ILC1 increases, resulting in a ratio similar to healthy subjects. This study aimed to analyze the influence of supervised 12-week walking (W) and Nordic-walking (NW) programs on changes in specific physiological parameters in middle-aged women 55.40±7.87 and 52.58±6.91 years of age, respectively. Body mass, fat tissue percentage, muscle tissue percentage, systolic blood pressure, diastolic blood pressure, resting pulse, and maximum oxygen uptake were measured at the beginning and end of the exercise program. Intra- and intergroup differences in physiological parameters as a function of the preparation period were analyzed using bootstrap analysis. Clinically reliable changes for a specific subject in the W and NW programs were analyzed using the Reliable Change Index (RCI). The results of this study indicate that more distinct differences were observed in the NW group. However, both programs improved the parameters studied. Nevertheless, an analysis of the RCI indicates that the W provides results similar to, if not better than, NW for increasing neuromuscular, cardiovascular, and cardiorespiratory fitness. The exponential growth of precision diagnostic tools, including omic technologies, molecular diagnostics, sophisticated genetic and epigenetic editing, imaging and nano-technologies and patient access to extensive health care, has resulted in vast amounts of unbiased data enabling in-depth disease characterization. New disease endotypes have been identified for various allergic diseases and triggered the gradual transition from a disease description focused on symptoms to identifying biomarkers and intricate pathogenetic and metabolic pathways. Consequently, the current disease taxonomy has to be revised for better categorization. This European Academy of Allergy and Clinical Immunology Position Paper responds to this challenge and provides a modern nomenclature for allergic diseases, which respects the earlier classifications back to the early 20th century. Hypersensitivity reactions originally described by Gell and Coombs have been extended into nine different types comprising antibody- (I-III), cell-mediated (IVa-c), tissue-driven mechanisms (V-VI) and direct response to chemicals (VII). Types I-III are linked to classical and newly described clinical conditions. Type IVa-c are specified and detailed according to the current understanding of T1, T2 and T3 responses. Types V-VI involve epithelial barrier defects and metabolic-induced immune dysregulation, while direct cellular and inflammatory responses to chemicals are covered in type VII. It is notable that several combinations of mixed types may appear in the clinical setting. The One Health approach is a collaborative and interdisciplinary strategy with focal point on human, animal, and environmental health interconnections. One Health can support the advanced management of allergic diseases and asthma, as complex, multifactorial diseases driven by interactions between the resilience response to the exposome. According to the One Health concept allergic diseases and asthma arising from exposures to a wide range of allergens, infectious agents and irritants (such as pollutants) occurring indoors and outdoors can be heavily influenced by environmental health (air, water, and soil quality) intermingled with animal health. These are currently heavily impacted by climate change, land use, urbanization, migration, overpopulation, and many more. Thus, a coordinated response to address the underlying factors that contribute to the development of allergic diseases and asthma needs to focus on the environment, human, and animal health altogether. Collaborative efforts across multiple sectors, including public health, veterinary medicine, environmental science, and community engagement are thus needed. A wide range of activities, including monitoring and surveillance of environmental and health data, targeted interventions to reduce exposures to allergens and irritants, and research on the underlying mechanisms that drive the development of allergic diseases and asthma are needed to move the field forward. In this consensus document elaborated by the European Academy of Allergy and Clinical Immunology (EAACI) and American Academy of Allergy, Asthma, and Immunology (AAAAI) under the practical allergy (PRACTALL) series, we provide insights into the One Heath approach aiming to provide a framework for addressing the complex and multifactorial nature of allergic diseases and asthma., (© 2023 European Academy of Allergy and Clinical Immunology and John Wiley & Sons Ltd.)

Allergic diseases and asthma are intrinsically linked to the environment we live in and to patterns of exposure. The integrated approach to understanding the effects of exposures on the immune system includes the ongoing collection of large-scale and complex data. This requires sophisticated methods to take full advantage of what this data can offer. Here we discuss the progress and further promise of applying artificial intelligence and machine-learning approaches to help unlock the power of complex environmental data sets toward providing causality models of exposure and intervention. We discuss a range of relevant machine-learning paradigms and models including the way such models are trained and validated together with examples of machine learning applied to allergic disease in the context of specific environmental exposures as well as attempts to tie these environmental data streams to the full representative exposome. Background: The correlation between primary immunodeficiencies (PIDs) and autoimmunity shows ethnic and geographical diversity. The aim of our study was to accumulate more data in paediatric PID population., Methods: 58 children aged 1-17 and with PID (study group) and 14 age-matched immunocompetent individuals (control group) were included in the study. Serum levels of 17 different specific IgG antibodies against autoantigens were measured by means of a quantitative enzyme immunoassay. Immunoglobulin levels were analysed in relation to a detailed medical examination., Results: Autoantibodies against one or more antigens were detected in the sera of 24.14% (n = 14) subjects in the study group. The most frequent were anti-thyroid peroxidase (anti-TPO) antibodies (n = 8; 13.8%). Anti-TPO antibody levels were elevated more often in PID patients with a positive family history of autoimmune diseases (p = 0.04). The screening for anti-deamidated gliadin peptide (DGP) and anti-tissue transglutaminase (tTG) antibodies in our series allowed identifying two previously undiagnosed cases of coeliac disease in PID patients. There was no statistically significant difference between the study and the control group in terms of the autoantibodies prevalence., Conclusions: This study provides data on the prevalence of autoantibodies in paediatric population diagnosed with PID. Allergen exposure chambers (AECs) are clinical facilities that allow the exposure of participants to allergenic and non-allergenic airborne particles. They provide stable particle concentrations under controlled environmental conditions. This is of great importance both for diagnostic purposes and for the monitoring of treatment effects. Here, a protocol and the technical prerequisites for performing a safe and effective allergen challenge in subjects sensitized to airborne allergens (i.e., house dust mite [HDM]) in the ALL-MED AEC are presented. With this method, triggering allergic symptoms corresponds to natural exposure. This can be used for an allergy diagnosis or as a plausible endpoint in clinical trials, particularly for allergen immunotherapy (AIT). A controlled environment (temperature, humidity, and carbon dioxide [CO2]) in the chamber must be maintained. Allergen particles must be dispersed evenly within the AEC at stable levels throughout the challenge. For this presentation, allergic rhinitis (AR) patients sensitive to HDM allergens were enrolled. AR symptoms were assessed by the following parameters: total nasal symptom score (TNSS), acoustic rhinometry (ARM), peak nasal inspiratory flow (PNIF), and nasal secretion weight. The safety of the procedure was assessed by the peak expiratory flow rate (PEFR) and the forced expiratory volume in the first second (FEV1). The allergic subjects developed symptoms within 120 min of the trial. On average, the most intense symptoms appeared after 60-90 min and, after reaching a plateau, remained stable until the end of the trial.

The purpose of this study was to evaluate changes in health-related parameters caused by the administration of anabolic-androgenic steroids and "fat-burning drugs" during a 6-month competition preparation period. The physiological, biochemical, and anthropometric parameters studied included serum cholesterol, triglycerides, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, aspartate aminotransferase, alanine transaminase, bilirubin, body mass, and percentage of total body fat. Changes in the parameters studied were analyzed at monthly intervals during six months of preparation for competition. The study revealed a continuous increase in body mass, accompanied by a decrease in body fat percentage to the physiologically essential level. Total cholesterol levels remined in the desirable concentration range. The mean levels of triglycerides fluctuated between borderline high and high . Mean high-density lipoprotein cholesterol levels remained within the low range, while low-density lipoprotein cholesterol fluctuated between near-optimal / above-optimal, borderline high , and high levels. Serum levels of aspartate aminotransferase and alanine transaminase remained within the high concentration. The bilirubin concentration remained in the desirable range. The blood nitrogen urea concentration fluctuated between normal and elevated levels. Background: Allergen exposure chamber (AEC) is a clinical facility that allows exposure to allergenic airborne particles in controlled environment. Although AECs offer stable levels of airborne allergens, the validation of symptoms and other endpoints induced by allergen challenge is key for their recommendation as a plausible tool for the assessment of patients, especially in clinical research. This study aimed to demonstrate the reproducibility of defined clinical endpoints after AEC house dust mite (HDM) challenge under optimal conditions in patients with allergic rhinitis (AR)., Method: HDM was distributed at different concentrations. The assessment was subjective by the patients: total nasal symptom score (TNSS), visual analog scale (VAS), and objective by the investigator: acoustic rhinometry, peak nasal inspiratory flow (PNIF), and nasal secretion weight. Safety was assessed clinically and by peak expiratory flow rate (PEFR) and forced expiratory volume in the first second (FEV 1 )., Results: Constant environment: temperature, humidity, and carbon dioxide (CO 2 ) concentration were maintained during all challenges. The concentration of HDM on average remained stable within the targeted values: 1000, 3000, 5000, 7000 particles (p)/m 3 . Most symptoms were observed at concentrations 3000 p/m 3 or higher. The symptoms severity and other endpoints results were reproducible. 5000 p/m 3 , and challenge duration of 120 min were found optimal. The procedure was safe with no lung function abnormalities due to challenge., Conclusion: HDM challenge in ALL-MED AEC offers a safe and reliable method for inducing symptoms in AR patients for the use in controlled clinical studies including allergen immunotherapy., (© 2022 The Authors. Metal allergy is mainly an environmental disorder which can cause allergic contact dermatitis. Environmental metal exposures include jewelry, everyday metal items, mobile phones, leather, metal-rich food and implants, including stents or anchors. While consumer exposure is liable for the majority of metal hypersensitivity cases, the significance of occupational exposure to metals remains relevant. Although the most common metal allergens are nickel, chromium, and cobalt; however, lately, gold, palladium, titanium, and some others have also attracted attention. This review highlights advances in metal allergy mechanisms, biomarkers for potential patients' stratification as well as biological treatments. The most recent evidence of human exposure to metal for risk assessment is discussed, as well as the relationship between the occurrence of metal hypersensitivity and implanted devices, including non-characteristic symptoms. The latest data on the diagnosis of metal hypersensitivity are also reported.

Aging is an independent risk factor for the development of various diseases associated, among others, with detrimental blood levels of fat- and water-soluble vitamins. Thus, the objective of this study is to investigate age-related changes in blood levels of vitamin A, B12, C, D, and E. Subject serum vitamin levels were obtained from the combined National Health and Nutrition Examination Surveys (NHANES). NHANESIII and NHANES 1999-2000, 2001-2002, 2003-2004, and 2005-2006. The raw data set was stratified into five age groups G1- G5: 20 ≤ G1 < 30, 30 ≤ G2 < 40, 40 ≤ G3 < 50, 50 ≤ G4 < 60, and 60 ≤ G5 < 70 years of age. Age stratified data was cleaned using the modified Horn algorithm. The reference range for the vitamin level of a specific age group was defined as data between the first and third quartile of the subject defined by normal blood pressure and normal bone density. Age-dependent changes in serum/plasma vitamin levels were assessed using the bootstrap technique with 10,000 repeats and Bonferroni adjustment. There was a continuous increase in vitamin A, B12, D, and E levels in the blood. However, the vitamin C concentration remained virtually constant in all age groups. There was a lack of cross-correlations between lipid and water-soluble vitamin levels and blood pressure and bone health. Contact hypersensitivity (CHS) is an experimental model of allergic contact dermatitis (ACD) that can be studied in mice. This study aims to present an objective laboratory method that may help to study the CHS reaction in mice, which can be measured and quantified by various tests. To induce CHS, on day "0", mice were sensitized on a previously shaved spot by abdominal skin painting with the hapten 2,4,6-trinitrochlorobenzene (TNCB) in an acetone-ethanol mixture, whereas negative control mice were sham sensitized with vehicle alone-acetone-ethanol mixture. On day "4", the baseline ear thickness was measured with a micrometer prior to the elicitation of CHS (challenge) by painting both ears with diluted TNCB both in the test and control groups. After 24 h, the ear swelling was measured with a micrometer. CHS is an example of a T cell-mediated immune response that causes swelling in inflamed tissue, peaking 24 h after the skin challenge with the same hapten. An increase in ear edema correlated with augmented ear weight, myeloperoxidase (MPO) activity, pro-inflammatory cytokine concentration in the ear extracts, increased thickening of the edematous dermis in the histological examination, and ear vascular permeability. There was also an increase in the concentration of TNP-specific IgG1 antibodies in the sera of the test group when compared with the control mice. Additionally, CHS can be successfully transferred with the CHS-effector cells obtained from donors previously sensitized with TNCB. The CHS-effector cells were administered intravenously into naïve recipient mice, which were subsequently challenged with the same diluted hapten. Ear swelling was measured with a micrometer 24 h later.

Background: Allergy is a clinical condition that reflects a deviated function of the immune system. The purpose of this study was to evaluate serum allergen-specific IgE (sIgE) along with clinical manifestations of allergy in patients with diagnosed primary immunodeficiency (PID). Methods: 72 patients, aged 1−17 years, diagnosed with PID and hospitalized between July 2020 and February 2021 were included in the study. Blood samples were obtained by venipuncture. sIgE (30 allergens), blood eosinophil count, as well as total IgE and IgG were measured and assessed in relation to a detailed medical examination. Results: Serum sIgE was detected in the blood of 50% of the patients in the study group, which significantly correlated (p < 0.0001) with clinical symptoms of allergy. During the period of the study, 61.1% of the patients showed symptoms of allergy, with 77.27% of them having tested positive for sIgE. The total IgE level was elevated in 18.06% of the patients and correlated with clinical symptoms of allergy (p = 0.004). An elevated total IgE level was not observed in children receiving immunoglobulin replacement therapy. Conclusion: The study showed that serum sIgE and total IgE together might be a plausible diagnostic tool for PID patients. However, for patients receiving immunoglobulin replacement therapy, the assessment of total IgE is not useful.

Last year brought a significant advance in asthma management, unyielding to the pressure of the pandemics. Novel key findings in asthma pathogenesis focus on the resident cell compartment, epigenetics and the innate immune system. The precision immunology unbiased approach was supplemented with novel tools and greatly facilitated by the use of artificial intelligence. Several randomised clinical trials and good quality real-world evidence shed new light on asthma treatment and supported the revision of several asthma guidelines (GINA, Expert Panel Report 3, ERS/ATS guidelines on severe asthma) and the conception of new ones (EAACI Guidelines for the use of biologicals in severe asthma). Integrating asthma management within the broader context of Planetary Health has been put forward. Background: Little is known about the prognostic role of fasting glucose after mechanical thrombectomy (MT)., Aims: We investigated whether fasting glucose on the next day after MT was associated with long-term outcome in acute ischemic stroke patients according to diabetes., Methods: We retrospectively analyzed 181 consecutive patients with acute anterior circulation ischemic stroke who underwent MT in 2 comprehensive stroke centers in Poland. Glucose levels were evaluated on admission and on the next day after MT. Fasting hyperglycemia (FHG) was defined as the glucose level above 5.5 mmol/L. Unfavorable outcome was defined as modified Rankin scale (mRS) of 3-6 at day 90 from stroke onset., Results: Patients with FHG had higher mRS at 3-month follow-up compared with those without FHG (3.71 ± 2.56 versus 1.87 ± 2.22, P < .001). In the subgroup analyses, FHG was associated with poor neurological outcome in the group without diabetes (3.74 ± 2.52 versus 1.81 ± 3.74, P < .001) but not with diabetes (3.64 ± 2.67 versus 2.30 ± 3.74, P= .11). Patients without diabetes who had FHG were older, had higher glucose on admission, higher prevalence of atrial fibrillation, cardioembolic stroke etiology and bleeding brain complications compared with the group with normal fasting glucose. After adjustment for potential confounders, fasting glucose (odds ratio [OR] 1.46; 95% CI 1.19-1.79, P < .001), age (OR 1.06; 95% CI 1.02-1.10, P = .001), successful reperfusion (OR 0.09; 95% CI 0.04-0.22, P < .001) and baseline NIHSS score (OR 1.18; 95% CI 1.08-1.29, P < .001) were predictors of mRS 3-6 at 3-month follow-up in the whole group. In the subgroup without diabetes, fasting glucose (OR 1.57; 95% CI 1.17-2.11, P = .002), age (OR 1.05; 95% CI 1.01-1.08, P = .008), successful reperfusion (OR 0.11; 95% CI 0.04-0.30, P < .001) and baseline NIHSS score (OR 1.14; 95% CI 1.04-1.26, P = .011) were independent predictors of unfavorable 3-month outcome., Conclusions: Fasting glucose on the next day after MT in patients with acute ischemic stroke is an independent risk factor for worse 3-month outcome., (Copyright © 2020. Based on the present literature, in March 2016, new recommendations of the American Academy of Ophthalmology for ophthalmic screening tests in patients treated with chloroquine and hydroxychloroquine were published. These recommendations emphasized the fact that toxicity is related to the dose calculated by real weight. The recommended hydroxychloroquine and chloroquine doses have been limited. It is no longer recommended to calculate the cumulative dose of chloroquine to establish the risk of toxicity. Kidney failure and the use of tamoxifen are proven risk factors of ocular complications in these patients. The screening agenda was established and available diagnostic methods were evaluated. Screening in patients treated with chloroquine derivatives may prevent an irreversible complication-toxic retinopathy. The present recommendations warn against making premature decision on medicine withdrawal, especially in the light of the most recent studies on their beneficial systemic influence. This paper systematizes the information on ophthalmological screening in chloroquine derivatives users.

Background: Previous animal study has shown that supplementation with silk amino acid hydrolysate (SAA) increases stamina in mice. The presented study was the first formal evaluation of the influence of SAA supplementation on parameters defining physiological fitness level in humans., Methods: It was a randomized controlled trial with a parallel-group design on elite male fin-swimmers. The experimental group was supplemented with 500 mg of SAA per kg of body mass, dissolved in 250 ml of a Carborade Drink®; the control group with Carborade Drink® alone; 3 times a day, 30 minutes prior to the training session., Results: Changes discerned in the experimental group were more pronounced than those observed in the control group. For example, the change in the serum lactic acid concentration observed in the experimental group was sevenfold less than in the control group [21.8 vs. -3.7 L% for the control and experimental groups, respectively]. An analysis of a lactate profile as a function of a maximal swimming velocity exposed a statistically significant positive shift in the swimming velocity of 0.05 m/s, at the lactate concentration of 4 mmol/L in the experimental group. There was also a positive, although statistically insignificant, increase of 2.6 L% in serum testosterone levels in the experimental group., Conclusions: This study showed that a 12-day SAA supplementation combined with an extensive and rigorous training schedule was sufficient to increase an aerobic stamina. However, this phenomenon was associated with an augmented level of muscular damage (an increased level of creatine phosphokinase in the experimental group).

Loss of physical strength and hypertension are among the most pronounced detrimental factors accompanying aging. The aim of this study was to evaluate the influence of a supervised 8-week Nordic-walking training program on systolic blood pressure in systolic-hypertensive postmenopausal women. This study was a randomized control trial on a sample of 24 subjects who did not take any hypertension medications. There was a statistically significant decrease in systolic blood pressure and an increase in lower and upper-body strength in the group following Nordic-walking training. There was a decrease in serum levels of total cholesterol, triglycerides, and low-density cholesterol. The obtained results indicate that an 8-week Nordic-walking program may be efficiently employed for counteracting systolic hypertension through a direct abatement of systolic blood pressure and an increase of maximal aerobic capacity.

The purpose of this study was to examine an influence of a mixed aerobic and strength training program (MAST) on anthropometry, serum lipid levels, physical performance, and functional fitness in obese postmenopausal women. The MAST sessions were held three times per week, and the exercise program lasted for 10 weeks. The exercise group demonstrated a statistically significant improvement in maximal oxygen uptake, a waist/hip ratio, and strength of the upper and lower body. An increase in LDL-C levels was observed in the control group. A 10-week MAST program encompassing Nordic-walking as an aerobic component, and strength exercises, induces positive changes in functional fitness, HDL-C, LDL-C and a waist/hip ratio in obese postmenopausal women. The observed changes implicate an increase in a health-related quality of life among the women administered to the physical exercise program.

New symmetrical arylene bisimide derivatives formed by using electron-donating-electron-accepting systems were synthesized. They consist of a phthalic diimide or naphthalenediimide core and imine linkages and are end-capped with thiophene, bithiophene, and (ethylenedioxy)thiophene units. Moreover, polymers were obtained from a new diamine, N , N '-bis(5-aminonaphthalenyl)naphthalene-1,4,5,8-dicarboximide and 2,5-thiophenedicarboxaldehyde or 2,2'-bithiophene-5,5'-dicarboxaldehyde. The prepared azomethine diimides exhibited glass-forming properties. The obtained compounds emitted blue light with the emission maximum at 470 nm. The value of the absorption coefficient was determined as a function of the photon energy using spectroscopic ellipsometry. All compounds are electrochemically active and undergo reversible electrochemical reduction and irreversible oxidation processes as was found in cyclic voltammetry and differential pulse voltammetry (DPV) studies. They exhibited a low electrochemically (DPV) calculated energy band gap ( E g ) from 1.14 to 1.70 eV. The highest occupied molecular orbital and lowest unoccupied molecular orbital levels and E g were additionally calculated theoretically by density functional theory at the B3LYP/6-31G(d,p) level. The photovoltaic properties of two model compounds as the active layer in organic solar cells in the configuration indium tin oxide/poly(3,4-(ethylenedioxy)thiophene):poly(styrenesulfonate)/active layer/Al under an illumination of 1.3 mW/cm 2 were studied. The device comprising poly(3-hexylthiophene) with the compound end-capped with bithiophene rings showed the highest value of V oc (above 1 V). The conversion efficiency of the fabricated solar cell was in the range of 0.69-0.90%.

Copepods are the most abundant multicellular animal group and are often most important component of zooplankton and indicators of local and global climate change. Among this broad group of animals, Calanus sinicus holds an important role in the ecosystems of The East China Sea, The Korea Strait, and The East Sea. By comparing the response of their proteomes to ecologically viable environment changes, this study tried to identify molecular responses accountable for compensation of a change in metabolic rates. Using two-dimensional gel electrophoresis and mass spectrometry on C. sinicus sampled at its native environment, 45 distinct proteins were identified that changed abundance as a function of environment changes i.e., temperature elevation and/or oxygen decrease, and 14 that are only present in proteome adapted to higher temperature/lower oxygen. Although the study failed to find heat shock proteins, the abundance of disulfide-isomerase A3 precursor was higher in species thriving at higher temperatures/lower oxygen. The abundance of proteins responsible for redox homeostasis, DNA maintenance, and chromatin rearrangement was also higher at elevated temperature. Also, the molecular machinery responsible for xenobiotic metabolism is mobilized at higher temperatures/ lower oxygen levels. These data indicate fine adjustment of molecular apparatus in response to changes in living environment.

The purpose of this study was to elucidate using time series analysis age and menopause induced differences in selected health quality related physiological factors. The study was conducted, using the Third National Health and Nutrition Examination Survey (NHANES III), and the NHANES 1999-2002 data, on women aged 35-60. Subjects who had not had surgical menopause, did not use contraceptives, did not smoke, and did not breastfeed during the examination, and did not use contraception and for whom follicle stimulating hormone and luteinizing hormone (LH) activity was assessed, were included in the study. Menopausal status was defined by months since the last period (<2, 2-12, and >12 months for pre-, peri-, and postmenopause, respectively). The results indicate that postmenopausal women, aged less than 45, are characterized by a decrease in systolic blood pressure (SBP), an increase in high density lipoprotein (HDL) levels, and a decrease in triglyceride (TG) levels. It was also determined that aging is the main factor leading to physiological variability in systolic blood pressure and high density lipoprotein levels, in pre- and perimenopausal women, and in follicle-stimulating hormone (FSH) activity in peri- and postmenopausal women., (Copyright © 2011 Elsevier Ireland Ltd. The aim of this study was to analyze the influence of individualized training (IT) as a function of motor type and effort status on changes of in specific physiological parameters among young soccer competitors. Blood pH and lactate concentrations, and lactate dehydrogenase (LDH), and creatine kinase (CK) activities were measured at the beginning of a preparation period, a match season, and a recuperation period of a 6-month macrocycle. The differences among specific physiological parameters as a function of the preparation phase for a defined motor type were analyzed by means of a 1-way generalized linear model (GLM) for repeated measurements. The differences in physiological parameters among defined motor types for a defined preparation phase were analyzed by means of the GLM for independent data. The differences in specific parameters before and after short time effort were analyzed by means of a t-test for matched pairs. Applied experimental and analytical approaches have revealed that IT administered to specific motor types differentiates players with respect to the pH, lactate concentration, and LDH activity. Obtained results indicate also that the dynamics of these parameters reflects the player's fitness level. Analysis of CK activity as a function of a preparation phase may serve as a prognostic tool for both overtraining and physical exhaustion.

The aim of this study was to analyze the influence of individualized training (IT) on psychomotor performance of young soccer players. Psychomotor performance was measured by means of differential response time and number of correct responses to audio and visual stimuli. Kinematic analysis allowing assessment of both the distances and the average speeds of young players during consecutive matches was also performed. For the data analysis, one-way analysis of variance, Friedman test, and t-test for dependent samples were used. The obtained results clearly indicate that IT during the preparation period increases specific psychomotor performance among young soccer players.

The objective of these studies was to determine the gender-dependent changes of coordination and fitness parameters in function of age by means of observational, cross-sectional study. We studied 180 male and 180 female participants by means of flamingo, kinesthetic differentiation, spatial orientation, reaction to signals, arm bend and chair stand tests. Generalized linear model (GLM) and the Welch test and the appropriate t-tests were used to compare the differences between the studied age groups. A nonlinear age-dependent progressive deterioration of basic coordination parameters and the strength level has been observed. The results of the battery of the tests indicate men as stronger with higher coordination level than women. In the present study, we analyzed the changes of bone mineral density (BMD) among Polish women age 40-79, as a function of biological aging and body mass index (BMI) class, and compared them with the US sample (National Health and Nutrition Examination Survey III). The null hypothesis of this study was that the rate of BMD change is femoral region, age, ethnicity, and BMI dependent. The studied group was divided into 4 age decades: 40-49, 50-59, 60-69, and 70-79 and 2 BMI classes (normal: 18.524.99). Analysis of covariance technique, using a generalized linear model with age and BMI as covariates, was used for data analysis. The influence of weight, height, and BMI on BMD loss was analyzed using multivariate regression analysis. The changes in BMD of femoral neck and trochanter are congruent. BMD decrease is not only age dependent but also, for specific ethnic groups, weight, height, and BMI dependent. The obtained results indicate that the analysis of age-dependent BMD changes should be performed by means of regression analysis using a broad age range rather than an age-decade approach.

The objective of the studies was to analyze the level of deterioration of basic coordinative parameters and to derive the present correlation. 180 female participants with no neurological disorders and limb surgery were studied by means of flamingo, kinesthetic differentiation, spatial orientation and reaction to signals tests. t-test for un-paired data and Pearson correlation analyses were performed. Nonlinear gradual deterioration of studied parameters in function of age was observed. The correlation between postural balance, kinesthetic differentiation, and reaction to signals was observed. The obtained results indicate that significant correlation between the coordinative parameters may influence daily life functioning. It has also been shown that decline of the basic coordinative parameters can be driven be neurodegenerative processes and environmental factors such as high calorie intake, physical and mental inactivity, toxins and/or infectious agents.
