16. Investigating the relationship between apoptosis and the matrix metalloproteinases

Abstract

Apoptosis and matrix metalloproteinases remove redundant tissue. They are independently described in similar situations but no causal relationship has been identified. This thesis hypothesizes that the process of apoptosis induces MMP expression. A new cell separation technique was developed allowing rapid separation of apoptotic and non-apoptotic cells with 95% purity, something not previously described. Biotinylated Annexin- V was used to label apoptotic cells and streptavadin-coated dynabeads and a magnetic particle separator used to extract them. Apoptosis was induced in cell lines derived from colon and gastric cancers and the cells separated and analysed using quantitative RT-PCR and real-time PCR. In fibrosarcoma cells apoptosis effects large increases in MMP7 mRNA and decreases for the inhibitor TIMP 4 whereas in malignant epithelial cells apoptosis increases production of MTI-MMP, MMP2 and TIMP2 mRNA. In co-culture, apoptotic epithelial cells induce mRNA production in fibrosarcoma cells for MTI-MMP and TIMP2~ MTI-MMP' combined with TIMP2 is pivotal in the activation of MMP2. These results support the hypothesis that the process of apoptosis leads to increased MMP production. As chemotherapy largely functions by inducing apoptosis, the demonstration that apoptosis may lead to increased matrix destruction and potential tumour shedding could explain why, for many malignancies adjuvant or nee-adjuvant therapy is ineffective and why, for others, efficacy is not as good in-vivo as predicted in-vitro.

24. Prospects for European Research and Development in Training & Education

Abstract

This paper outlines the prospects for research and development in the education and training fields offered by the European Communities. The position of the EC is explained in relation to the existing and forthcoming Framework Programmes of research and development. The 1987-91 Framework Programme has committed most of its funding to existing programmes. Only one, DELTA directly concerns R&D in the education and training technologies. This commenced in 1989 with only 20 million ECU and ends in Spring 1991. In the new Framework Programme, DELTA is expected to continue with funding of between 40 and 60 million ECU though this remains undecided at present. The earliest date for a new DELTA call for proposals in Spring 1991, meaning that work on the new four year programme will not commence until late autumn 1991 at the earliest. There will therefore be a considerable gap between the two. An outline of the likely structure and contents of a new DELTA programme is given, characterised by better liaison with other programmes and a more rational approach to the development of the advanced learning technologies. Other, non-Framework Programmes concerned with aspects of education and training such as COMETT, LINGUA, EUROTECNET and TEMPUS are placed in context with this. The paper concludes with general advice and explanation of EC procedures for application, evaluation and approval of proposals for Framework R&D Proposals. 30. Regions based on social structure

Abstract

Boas argued that anthropologists should make historical comparisons within well-defined regional contexts. A century later, we have many improvements in the statistical methodologies for comparative research, yet most of our regional constructs remain without a valid empirical basis. We present a new method for developing and testing regions. The method takes into account older anthropological concerns with relationships between culture history and the environment, embodied in the culture-area concept, as well as contemporary concerns with historical linkages of societies into world systems. We develop nine new regions based on social structural data and test them using data on 35 I societies. We compare the new regions with Murdock's regional constructs and find that our regional classification is a strong improvement over Murdock's. In so doiig we obtain evidence for the cross-cultural importance of gender and descent systems, for the importance of constraint relationships upon sociocultural systems, for the historical importance of two precapitalist world systems, and for strikingly different geographical alignments of cultural systems in the Old World and the Americas.

Abstract

Whilst the debilitating fatigue experienced in patients suffering from Chronic Fatigue Syndrome (CFS) results in a subjective marked impairment in functioning, little research has investigated the impact of this disorder on quality of life. Forty-seven subjects with a confirmed diagnosis of CFS and 30 healthy controls were compared using the Sickness Impact Profile (SIP). A subgroup of subjects were interviewed regarding the impact CFS has had on their social and family relationships, work and recreational activities. Results from both the SIP and the interview revealed that CFS subjects had significantly impaired quality of life, especially in areas of social functioning. These findings highlight the importance of addressing the social isolation and loss of role functioning experienced by CFS sufferers.
