This thesis develops a conceptual design tool capable of generating optimized structural layout suggestions for building design in the early design phase. The structural layout of a building is the arrangement and design of the load-bearing elements that support the weight of the building and resist external forces. The structural layout in this project solely consists of prefabricated reinforced (RC) elements. The use of prefabricated RC elements is embedded in the Danish construction industry and will likely remain so in the foreseeable future. Therefore, there is great potential for more effective use of concrete in terms of sustainability and decreasing cost. The proposed design tool can help fulfill this potential. Action Research (AR) is used to create the conceptual framework of the design tool. The AR analysis consists of semi-structured interviews and a co-creation workshop where architects, engineers, and contractors contribute to the development of the design tool to ensure that the final tool conforms to real-world practice. The final design tool is based on this framework and developed using four core principles: optimization, interactivity, dissemination, and automation. A novel parametric modeling method is developed in the design tool called Adjacent Polygon (APoly) representation. The APoly method creates a dynamic parametric representation of a given building plan to generate diverse yet feasible structural layout suggestions.The evaluation modules of different structural typologies are constructed using surrogate models in the form of Neural Networks. The surrogate models are combined in a hierarchical structure to create an algorithm capable of predicting the optimized geometry and corresponding cost for a structural element based on the external conditions inputted into the algorithm. The entire network of prediction models is then combined with a meta-heuristic optimization algorithm in the form of a Genetic Algorithm (GA) to create a surrogate-assisted optimization framework. A repair algorithm is incorporated into the GA to increase the number of valid solutions generated during each optimization iteration to decrease the convergence time. The performance and reliability of the design tool are validated through two groups of local and global case studies. The first group consists of parameter sensitivity studies on the local approximation modules for each structural typology. The second group of validation studies examines the design tool’s effectiveness across relevant building plans and scenarios. The corresponding results demonstrate that the tool can effectively adapt to these different settings and produce optimized structural layout suggestions. It is also demonstrated that the design tool can conduct multi-objective optimization and produce a front of Pareto optimal solutions.

HER2 is a predictive biomarker for HER2-targeted therapeutics. For antibody–drug conjugates (ADCs; e.g., trastuzumab emtansine (T-DM1)), HER2 is utilized as a transport gate for cytotoxic agents into the cell. ADC biomarkers may therefore be more complex, also reflecting the intracellular drug transport. Here we report on a positive correlation between the early endosome marker RAB5A and T-DM1 sensitivity in five HER2-positive cell lines. Correlation between RAB5A expression and T-DM1 sensitivity is confirmed in breast cancer patients treated with trastuzumab emtansine/pertuzumab in the I-SPY2 trial (NCT01042379), but not in the trastuzumab/paclitaxel control arm. The clinical correlation is further verified in patients from the KAMILLA trial (NCT01702571). Antibody-drug conjugates (ADCs) have demonstrated impressive activity in recent clinical trials in breast cancer. Such targeted therapeutics strongly depends on the presence of target molecules on the tumor cells, and the presence of such target molecules may determine the response of ADCs. However, ADCs are also dependent on cellular uptake, and factors regulating endocytosis as well as intracellular trafficking may strongly influence ADC activity. We have recently demonstrated that the activity of the HER2 targeted ADC trastuzumab emtansine (T-DM1) is dependent on the expression of RAB5A, a protein regulating endocytosis (1). A significant correlation between Rab5A expression and T-DM1 efficacy was found in a panel of HER2 expressing breast- and ovarian cancer cell lines. This result was verified in the I-SPY2 clinical trial where patients with high RAB5A expression were more likely to achieve a pathological complete response following T-DM1 as a neoadjuvant. The result was further validated in patients treated with T-DM1 in the Kamilla study where patients with a high RAB5A had a longer progression free survival. All ADCs should in principle be dependent on endocytosis to exert their activity. This triggered the investigation of proteins regulating endocytosis as predictive biomarkers for ADCs in general. METHODS: HER2-positive breast and ovarian cancer cell lines were evaluated with respect to the sensitivity and efficacy of treatment with T-DM1, trastuzumab deruxtecan, sacituzumab govitecan and the targeted toxin MH3B1/rGel.. The expression levels of proteins involved in endocytosis and endocytic trafficking including RAB4A, RAB5A and RAB11A were investigated in addition to the molecular drug targets (HER2 and TROP2). Cellular drug sensitivity was correlated to the expression levels of the investigated proteins using both RNA and protein as readout. RESULTS: The early endosome marker RAB5A, was found to correlate positively to the activity of trastuzumab deruxtecan, sacituzumab govitecan and MH3B1/rGel in the HER2 positive cell line panel, confirming the importance of RAB5A expression for the activity of these drugs. A significant correlation was found between RAB5A and drug efficacy using both protein and RNA as a readout. CONCLUSION: The present results indicate RAB5A as a generic predictive biomarker for both ADCs and targeted toxins which both depend on cellular uptake for cytotoxic efficacy. PURPOSETargeted therapeutics strongly depends on validated biomarkers in order to select patients most likely to benefit from the treatment. HER2 serves as a predictive biomarker for HER2-targeted tyrosine kinase inhibitors and monoclonal antibodies. HER2 may, however, also be utilized as a transport gate for delivery of cytotoxic agents into the cell, such as for HER2-targeted antibody drug conjugates (ADCs; e.g. trastuzumab emtansine (T-DM1)). The predictive biomarkers for such ADCs may be more complex, also reflecting the intracellular transport.METHODSFive HER2-positive breast and ovarian cancer cell lines were evaluated with respect to T-DM1 sensitivity and correlated to the expression levels of proteins involved in endocytic trafficking including RAB4A, RAB5A and RAB11A, with possible impact on ADC pharmacology. The results were confirmed in a clinical cohort consisting of patients from the adaptive breast cancer clinical trial I-SPY2 where pathological complete response (pCR) was correlated to the RNA expression level of RAB4A, RAB5A and RAB11A. A subset of the clinical KAMILLA trial including 19 patients was used as a verification cohort where semi-quantitative IHC of RAB5A was correlated to progression free survival (PFS).RESULTSThe early endosome marker RAB5A, was found to correlate positively to T-DM1 sensitivity in the cell line panel. Correlation between RAB5A expression and T-DM1 sensitivity (pCR) was confirmed in patients treated with trastuzumab emtansine/pertuzumab in the I-SPY2 trial, but not in the trastuzumab/paclitaxel control arm. The clinical correlation was verified in the patients from the KAMILLA trial where semi-quantitative RAB5A IHC staining correlated significantly positive to PFS.CONCLUSIONThe present results indicate that RAB5A is a predictive biomarker for T-DM1 and outline, for the first time, proteins involved in endocytic trafficking as predictive biomarkers for ADCs.

The present literature review brings together the latest research-based evidence and knowledge in the international research literature in the field on how RTOs' technological ser-vice advice and infrastructure affect firms’ financial performance, how they contribute to firms’ innovation processes and innovation capacity, and their significance to firms’ subsequent col-laboration with knowledge institutions and others. The focus is primarily on GTS-like RTOs1 to increase the applicability of the review in a Danish context, including giving the Danish Agency for Higher Education and Science (UFS) a knowledge base on RTOs’ significance for Danish firms. An overview table in the appendix summarizes the identified literature, giving references, short summaries and categorizations about, e.g. empirical methods and data, its coverage and placement in our evidence hierarchy.

Advances in recombinant DNA technology have opened up new possibilities of exploiting toxic proteins for therapeutic purposes. Bringing forth these protein toxins from the bench to the bedside strongly depends on the availability of production methods that are reproducible, scalable and comply with good manufacturing practice (GMP). The type I ribosome-inhibiting protein, gelonin, has great potential as an anticancer drug, but is sequestrated in endosomes and lysosomes. This can be overcome by combination with photochemical internalization (PCI), a method for endosomal drug release. The combination of gelonin-based drugs and PCI represents a tumor-targeted therapy with high precision and efficiency. The aim of this study was to produce recombinant gelonin (rGel) at high purity and quantity using an automated liquid chromatography system. The expression and purification process was documented as highly efficient (4.4 mg gelonin per litre induced culture) and reproducible with minimal loss of target protein (~50% overall yield compared to after initial immobilized metal affinity chromatography (IMAC)). The endotoxin level of 0.05&ndash, 0.09 EU/mg was compatible with current standards for parenteral drug administration. The automated system provided a consistent output with minimal human intervention and close monitoring of each purification step enabled optimization of both yield and purity of the product. rGel was shown to have equivalent biological activity and cytotoxicity, both with and without PCI-mediated delivery, as rGelref produced without an automated system. This study presents a highly refined and automated manufacturing procedure for recombinant gelonin at a quantity and quality sufficient for preclinical evaluation. The methods established in this report are in compliance with high quality standards and compose a solid platform for preclinical development of gelonin-based drugs.

Lysosomal accumulation of sunitinib has been suggested as an underlying mechanism of resistance. Here, we investigated if photochemical internalization (PCI), a technology for cytosolic release of drugs entrapped in endosomes and lysosomes, would activate lysosomal sequestered sunitinib. By super-resolution fluorescence microscopy, sunitinib was found to accumulate in the membrane of endo/lysosomal compartments together with the photosensitizer disulfonated tetraphenylchlorin (TPCS2a). Furthermore, the treatment effect was potentiated by PCI in the human HT-29 and the mouse CT26.WT colon cancer cell lines. The cytotoxic outcome of sunitinib-PCI was, however, highly dependent on the treatment protocol. Thus, neoadjuvant PCI inhibited lysosomal accumulation of sunitinib. PCI also inhibited lysosomal sequestering of sunitinib in HT29/SR cells with acquired sunitinib resistance, but did not reverse the resistance. The mechanism of acquired sunitinib resistance in HT29/SR cells was therefore not related to lysosomal sequestering. Sunitinib-PCI was further evaluated on HT-29 xenografts in athymic mice, but was found to induce only a minor effect on tumor growth delay. In immunocompetent mice sunitinib-PCI enhanced areas of treatment-induced necrosis compared to the monotherapy groups. However, the tumor growth was not delayed, and decreased infiltration of CD3-positive T cells was indicated as a possible mechanism behind the failed overall response.

Photochemical internalisation (PCI) is a unique intervention which involves the release of endocytosed macromolecules into the cytoplasmic matrix. PCI is based on the use of photosensitizers placed in endocytic vesicles that, following light activation, lead to rupture of the endocytic vesicles and the release of the macromolecules into the cytoplasmic matrix. This technology has been shown to improve the biological activity of a number of macromolecules that do not readily penetrate the plasma membrane, including type I ribosome-inactivating proteins (RIPs), gene-encoding plasmids, adenovirus and oligonucleotides and certain chemotherapeutics, such as bleomycin. This new intervention has also been found appealing for intracellular delivery of drugs incorporated into nanocarriers and for cancer vaccination. PCI is currently being evaluated in clinical trials. Data from the first-in-human phase I clinical trial as well as an update on the development of the PCI technology towards clinical practice is presented here.

I dette kapitlet avklarer vi begrepet kritisk leseforståelse og undersøker hvordan elevenes evne til å lese kritisk måles i PISA 2018 og nasjonale leseprøver. Vi finner at flertallet av elevene verken foretar reelle vurderinger av teksters troverdighet eller begrunner standpunktene sine. Under halvparten av norske elever rapporterer at de har lært å foreta slike vurderinger på skolen. Det er grunn til å spørre om norske elevers kritiske lesekompetanse i for stor grad overlates til hva de lærer hjemme.

PISA-undersøkelsen måler om 15-åringer har lesekompetansen som trengs for å delta i vårt tekstbaserte samfunn. I dette kapitlet gjør vi rede for hvordan lesekompetanse måles i PISA, og hva som er nytt i rammeverket for 2018. Vi diskuterer også hva slags kompetanse som er nødvendig for dagens lesere, og knytter dette til teori om lesing av multiple og digitale tekster. Vi viser flere oppgaveeksempler fra prøven, og avslutningsvis drøfter vi kort om PISA måler lesekompetanse som er viktig i dagens samfunn.
