6. Development of cyclosporin A mediated immunity in L1210 leukaemia.

Cyclosporin A (CsA) is an effective modulator of multidrug resistance (MDR) in vitro and in murine tumour systems in vivo. We now report the production of immunity to L1210 leukaemia by the addition of CsA to VP-16 therapy of leukaemic BDF/1 mice. VP-16/cyclosporin A tumour immunity induction arises as a consequence of active therapy independently of immunisation with modified tumour cells. The addition of CsA to VP-16 prolongs survival of BDF/1 host mice bearing L1210 leukaemia beyond that produced by equivalent dose VP-16 alone. A subpopulation of 60-day surviving mice after combined VP-16/CsA are immune to rechallenge with the same leukaemia inoculum to which they were originally exposed. Spleen cells from immune mice adoptively transfer anti-L1210 leukaemia immunity to untreated BDF/1 mice in a dose dependent, statistically significant manner. Adoptive transfer experiments additionally suggest active recruitment of immunologic response in recipient animals: (1) We have been able to perpetuate leukaemia immunity in four sequential cohorts of naive recipient mice. This propogation of adoptive immunity is accomplished by use of spleen cells harvested from each preceeding passively-protected animal cohort; (2) Cyclophosphamide pretreatment of adoptive transfer recipient mice abrogates the ability of their splenocytes to perpetuate passive protection in sequential adoptive transfer experiments.

8. The Mating Pattern of Captive Naked Mole-Rats Is Best Described by a Monogamy Model

- Abstract
Naked mole-rats form colonies with a single reproductively active female surrounded by subordinate workers. Workers perform offspring care, construction and defense of the burrow system, and food supply. Such division of labor, called “cooperative breeding,” is strongly associated with the evolution of monogamous mating behavior, as seen in several mammalian lineages. This association is explained by the evolutionary theory of kin selection, according to which a subordinate adult may help to raise other’s offspring if they are in full sibling relationship. In conflict with this theory, the naked mole-rat is widely considered to be polyandrous, based on reports on multiple males contributing to a colony’s progeny. In order to resolve this contrast, we undertook an in-depth microsatellite-based kinship analysis on captive colonies. Four independent colonies comprising a total of 265 animals were genotyped using a panel of 73 newly established microsatellite markers. Our results show that each mole-rat colony contains a single monogamous breeder pair, which translates to a reproductive skew of 100% for both sexes. This finding, also in conjunction with previously published parental data, favors monogamy as the best-fitting model to describe naked mole-rat reproduction patterns. Polyandry or other polygamous reproduction models are disfavored and should be considered as exceptional. Overall, the empirical genetic data are in agreement with the kin selection theory.

Geochemical processes such as mineral dissolution and precipitation alter the microstructure of rocks, and thereby affect their hydraulic and mechanical behaviour. Quantifying these property changes and considering them in reservoir simulations is essential for a sustainable utilisation of the geological subsurface. Due to the lack of alternatives, analytical methods and empirical relations are currently applied to estimate evolving hydraulic and mechanical rock properties associated with chemical reactions. However, the predictive capabilities of analytical approaches remain limited, since they assume idealised microstructures, and thus are not able to reflect property evolution for dynamic processes. Hence, aim of the present thesis is to improve the prediction of permeability and stiffness changes resulting from pore space alterations of reservoir sandstones. A detailed representation of rock microstructure, including the morphology and connectivity of pores, is essential to accurately determine physical rock properties. For that purpose, three-dimensional pore-scale models of typical reservoir sandstones, obtained from highly resolved micro-computed tomography (micro-CT), are used to numerically calculate permeability and stiffness. In order to adequately depict characteristic distributions of secondary minerals, the virtual samples are systematically altered and resulting trends among the geometric, hydraulic, and mechanical rock properties are quantified. It is demonstrated that the geochemical reaction regime controls the location of mineral precipitation within the pore space, and thereby crucially affects the permeability evolution. This emphasises the requirement of determining distinctive porosity-permeability relationships by means of digital pore-scale models. By contrast, a substantial impact of spatial alterations patterns on the stiffness evolution of reservoir sandstones are only observed in case of certain microstructures, such as highly porous granular rocks or sandstones comprising framework-supporting cementations. In order to construct synthetic granular samples a process-based approach is proposed including grain deposition and diagenetic cementation. It is demonstrated that the generated samples reliably represent the microstructural complexity of natural sandstones. Thereby, general limitations of imaging techniques can be overcome and various realisations of granular rocks can be flexibly produced. These can be further altered by virtual experiments, offering a fast and cost-effective way to examine the impact of precipitation, dissolution or fracturing on various petrophysical correlations. The presented research work provides methodological principles to quantify trends in permeability and stiffness resulting from geochemical processes. The calculated physical property relations are directly linked to pore-scale alterations, and thus have a higher accuracy than commonly applied analytical approaches. This will considerably improve the predictive capabilities of reservoir models, and is further relevant to assess and reduce potential risks, such as productivity or injectivity losses as well as reservoir compaction or fault reactivation. Hence, the proposed method is of paramount importance for a wide range of natural and engineered subsurface applications, including geothermal energy systems, hydrocarbon reservoirs, CO2 and energy storage as well as hydrothermal deposit exploration. Geochemische Lösungs- und Fällungsprozesse verändern die Struktur des Porenraums und können dadurch die hydraulischen und mechanischen Gesteinseigenschaften erheblich beeinflussen. Die Quantifizierung dieser Parameteränderung und ihre Berücksichtigung in Reservoirmodellen ist entscheidend für eine nachhaltige Nutzung des geologischen Untergrunds. Aufgrund fehlender Alternativen werden dafür bisher analytische Methoden genutzt. Da diese Ansätze eine idealisierte Mikrostruktur annehmen, können insbesondere Änderungen der Gesteinseigenschaften infolge von dynamischen Prozessen nicht zuverlässig abgebildet werden. Ziel der vorliegenden Doktorarbeit ist es deshalb, die Entwicklung von Gesteinspermeabilitäten und -steifigkeiten aufgrund von Porenraumveränderungen genauer vorherzusagen. Für die möglichst exakte Bestimmung physikalischer Gesteinsparameter ist eine detaillierte Darstellung der Mikrostruktur notwendig. Basierend auf mikro-computertomographischen Scans werden daher hochaufgelöste, dreidimensionale Modelle typischer Reservoirsandsteine erstellt und Gesteinspermeabilität und -steifigkeit numerisch berechnet. Um charakteristische Verteilungen von Sekundärmineralen abzubilden, wird der Porenraum dieser virtuellen Sandsteinproben systematisch verändert und die resultierenden Auswirkungen auf die granulometrischen, hydraulischen und elastischen Gesteinseigenschaften bestimmt. Die Ergebnisse zeigen deutlich, dass charakteristische Fällungsmuster unterschiedlicher geochemischer Reaktionsregime die Permeabilität erheblich beeinflussen. Folglich ist die Nutzung von porenskaligen Modellen zur Bestimmung der Porosität-Permeabilitätsbeziehungen unbedingt notwendig. Im Gegensatz dazu ist die Verteilung von Sekundärmineralen für die Gesteinssteifigkeit nur bei bestimmten Mikrostrukturen von Bedeutung, hierzu zählen hochporöse Sandsteine oder solche mit Korngerüst-stützenden Zementierungen. In der Arbeit wird außerdem ein Ansatz zur Konstruktion granularer Gesteine vorgestellt, welcher sowohl die Kornsedimentation als auch die diagenetische Verfestigung umfasst. Es wird gezeigt, dass die synthetischen Proben die mikrostrukturelle Komplexität natürlicher Reservoirsandsteine gut abbilden. Dadurch können generelle Limitationen von bildgebenden Verfahren überwunden und unterschiedlichste virtuelle Repräsentationen von granularen Gesteinen generiert werden. Die synthetischen Proben können zukünftig in virtuellen Experimenten verwendet werden, um die Auswirkungen von Lösungs- und Fällungsreaktionen auf verschiedene petrophysikalische Korrelationen zu untersuchen. Die vorgestellte Arbeit liefert methodische Grundlagen zur Quantifizierung von Permeabilitäts- und Steifigkeitsänderungen infolge geochemischer Prozesse. Die berechneten petrophysikalischen Beziehungen basieren direkt auf mikrostrukturellen Veränderungen des Porenraums. Daher bieten sie eine genauere Vorhersage der Gesteinseigenschaften als herkömmliche analytische Methoden, wodurch sich die Aussagekraft von Reservoirmodellen erheblich verbessert. Somit können Risiken, wie Produktivitäts- oder Injektivitätsverluste sowie Reservoirkompaktion oder Störungsreaktivierung, verringert werden. Die präsentierten Ergebnisse sind daher relevant für verschiedenste Bereiche der geologischen Untergrundnutzung wie CO2- oder Energiespeicherung, Geothermie, Kohlenwasserstoffgewinnung sowie die Erkundung hydrothermaler Lagerstätten.

Numerical simulation has become an inevitable tool for improving the understanding on coupled processes in the geological subsurface and its utilisation. However, most of the available open source and commercial modelling codes do not come with flexible chemical modules or simply do not offer a straight-forward way to couple third-party chemical libraries. For that reason, the simple and efficient TRANsport Simulation Environment (TRANSE) has been developed based on the Finite Difference Method in order to solve the density-driven formulation of the Darcy flow equation, coupled with the equations for transport of heat and chemical species. Simple explicit, weighted semi-implicit or fully-implicit numerical schemes are available for the solution of the system of partial differential equations, whereby the entire numerical code is composed of less than 1000 lines of Python code, only. A diffusive flux-corrected advection scheme can be employed in addition to pure upwinding to minimise numerical diffusion in advection-dominated transport problems. The objective of the present study is to verify the numerical code implementation by means of benchmarks for density-driven fluid flow and advection-dominated transport. In summary, TRANSE exhibits a very good agreement with established numerical simulation codes for the benchmarks investigated here. Consequently, its applicability to numerical density-driven flow and transport problems is proven. The main advantage of the presented numerical code is that the implementation of complex problem-specific couplings between flow, transport and chemical reactions becomes feasible without substantial investments in code development using a low-level programming language, but the easy-to-read and -learn Python programming language.

The second Dynamics and Chemistry of Marine Stratocumulus (DYCOMS-II) field study is described. The field program consisted of nine flights in marine stratocumulus west-southwest of San Diego, California. The objective of the program was to better understand the physics and dynamics of marine stratocumulus. Toward this end special flight strategies, including predominantly nocturnal flights, were employed to optimize estimates of entrainment velocities at cloud-top, large-scale divergence within the boundary layer, drizzle processes in the cloud, cloud microstructure, and aerosol-cloud interactions. Cloud conditions during DYCOMS-II were excellent with almost every flight having uniformly overcast clouds topping a well-mixed boundary layer. Contains fulltext : 215772.pdf (Publisher’s version ) (Open Access) BACKGROUND: The time interval between CRT and surgery in rectal cancer patients is still the subject of debate. The aim of this study was to first evaluate the nationwide use of restaging magnetic resonance imaging (MRI) and its impact on timing of surgery, and, second, to evaluate the impact of timing of surgery after chemoradiotherapy (CRT) on short- and long-term outcomes. METHODS: Patients were selected from a collaborative rectal cancer research project including 71 Dutch centres, and were subdivided into two groups according to time interval from the start of preoperative CRT to surgery (< 14 and >/= 14 weeks). RESULTS: From 2095 registered patients, 475 patients received preoperative CRT. MRI restaging was performed in 79.4% of patients, with a median CRT-MRI interval of 10 weeks (interquartile range [IQR] 8-11) and a median MRI-surgery interval of 4 weeks (IQR 2-5). The CRT-surgery interval groups consisted of 224 (< 14 weeks) and 251 patients (>/= 14 weeks), and the long-interval group included a higher proportion of cT4 stage and multivisceral resection patients. Pathological complete response rate (n = 34 [15.2%] vs. n = 47 [18.7%], p = 0.305) and CRM involvement (9.7% vs. 15.9%, p = 0.145) did not significantly differ. Thirty-day surgical complications were similar (20.1% vs. 23.1%, p = 0.943), however no significant differences were found for local and distant recurrence rates, disease-free survival, and overall survival. CONCLUSIONS: These real-life data, reflecting routine daily practice in The Netherlands, showed substantial variability in the use and timing of restaging MRI after preoperative CRT for rectal cancer, as well as time interval to surgery. Surgery before or after 14 weeks from the start of CRT resulted in similar short- and long-term outcomes.

Aim A Snapshot study design eliminates changes in treatment and outcome over time. This population based Snapshot study aimed to determine current practice and outcome of rectal cancer treatment with published landmark randomized controlled trials as a benchmark.Method In this collaborative research project, the dataset of the Dutch Surgical Colorectal Audit was extended with additional treatment and long-term outcome data. All registered patients who underwent resection for rectal cancer in 2011 were eligible. Baseline characteristics and outcome were evaluated against the results of the Dutch TME trial and the COLOR II trial from which the original datasets were obtained.Results A total of 71 hospitals participated, and data were completed for 2102 out of the potential 2633 patients (79.8%). Median follow-up was 41 (interquartile range 25-47) months. Overall circumferential resection margin (CRM) involvement was 9.3% in the Snapshot cohort and 18.5% in the Dutch TME trial. CRM positivity after laparoscopic resection was 7.8% in the Snapshot and 9.5% in the COLOR II trial. Three-year overall local recurrence rate in the Snapshot was 5.9%, with a disease-free survival of 67.1% and overall survival of 79.5%. Benchmarking with the randomized controlled trials revealed an overall favourable long-term outcome of the Snapshot cohort.Conclusion This study showed that current rectal cancer care in a large unselected Dutch population is of high quality, with less positive CRM since the TME trial and oncologically safe implementation of minimally invasive surgery after the COLOR II trial.

- Abstract
Reactive transport and related processes as dissolution and precipitation of minerals may substantially change rock composition, and consequently its geomechanical properties. However, this chemo-mechanical interaction is rarely considered in assessments of subsurface processes. An approach to determine the macroscopic stress-strain- relationship of a composite material, based on the mechanical characteristics of its constituents is provided by analytical homogenization models. This method is widely used within the field of materials sciences and highly promising for geoscientific applications. However, none of the analytical models can accurately determinate elastic parameters of the composite for the entire range of the inclusions mechanical properties, their specific volume fractions and aspect ratios. Numerical 3D models allow for a more precise quantification and help to overcome the limitations of analytical solutions, especially regarding the spatial distribution of inclusions and their interaction. For a representative rock volume element, effective elastic properties are determined in our approach by taking into account the characteristic geomechanical parameters of all relevant mineral inclusions. We successfully validated our model against available analytical solutions and published experimental data. Sensitivity analyses indicate that the calculated elastic rock properties vary up to 28 % due to the spatial distribution of the inclusions within the rock matrix. Additionally, the developed numerical homogenization method enables an improved quantification of rock parameters: particularly for high inclusion-to-matrix-volume fractions and high mechanical property contrasts, the analytical solution deviates up to 58 % from the numerical results. In conclusion, the introduced homogenization approach provides an essential basis for coupling chemical and mechanical processes in numerical simulations related to the geological subsurface.

49. Numerical simulations of saltwater displacement via fault systems due to exploitation of the subsurface

- Abstract
Injection of fluids into deep saline aquifers induces an increase in pore pressure in the storage formation, and thus displacement of resident brines. Upward brine migration into shallower aquifers via hydraulically conductive faults may therefore lead to unwanted salinization of potable groundwater resources. In the present study, we investigated different scenarios for a prospective storage site close to the city of Beeskow in the Northeast German Basin by using a representative 3D regional-scale model (100 km x 100 km x 1.34 km) that includes four regional fault zones. The focus was on assessing the impact of fault length and permeability as well as model boundary conditions on the potential salinization of shallow groundwater resources. Moreover, the effects of an overlying secondary brine-bearing reservoir as well as varying initial salt-freshwater boundaries were investigated. We employed numerical simulations of brine injection as a representative fluid based on an example case study discussed by Tillner et al. (2013). Our simulation results demonstrate that pressure build-up within the reservoir determines the fluid rates and duration through the faults, and hence salinization of shallower aquifers. Application of different boundary conditions proved that these have a crucial impact on reservoir fluid displacement. If reservoir boundaries are closed, the fluid displaced via the faults into the shallow aquifer corresponds to the overall injected fluid mass. In that case, fault length and permeability as well as the presence of an overlying secondary reservoir have only temporal effects on brine migration. A fault zone with a hydraulically conductive segment of only two kilometres length causes brine flow into the shallow aquifer of 330 years, which is thus five times longer compared to the case with four faults open over their entire length of 193 km. The presence of an overlying secondary reservoir leads to an additional retardation of brine inflow into the uppermost aquifer up to a factor of three. If the reservoir boundaries are open, salinization is considerably reduced. In the presence of a secondary reservoir, 33 % of equivalent brine mass migrates into the shallow aquifer, if all four faults are completely hydraulically open, whereas the displaced equivalent brine mass is only 13 % if accounting for a single fault of two kilometres length. Without the secondary reservoir, 66 % of the brine mass is displaced in the four fault and about 30 % in the 2 km single fault cases. Taking into account the considered geological boundary conditions, the brine mainly originates from the upper 16 m to 300 m of the investigated faults, and hence the initial salt-freshwater boundary present in the fault is of high relevance for the resulting shallow aquifer salinization. The present study successfully demonstrates that a quantification of brine displacement using numerical simulations is feasible at regional scale.
