Abstract

The study examines the effects of the antioxidant flavonoid Pycnogenol on a range of cognitive and biochemical measures in healthy elderly individuals. The study used a double-blind, placebo-controlled, matched-pair design, with 101 elderly participants (60-85 years) consuming a daily dose of 150 mg of Pycnogenol for a three-month treatment period. Participants were assessed at baseline, then at 1, 2, and 3 months of the treatment. The control (placebo) and Pycnogenol groups were matched by age, sex, body mass index, micronutrient intake, and intelligence. The cognitive tasks comprised measures of attention, working memory, episodic memory, and psychomotor performance. The biological measures comprised levels of clinical hepatic enzymes, serum lipid profile, human growth hormone, and lipid peroxidation products. Statistically significant interactions were found for memory-based cognitive variables and lipid peroxidation products, with the Pycnogenol group displaying improved working memory and decreased concentrations of F2-isoprostanes relative to the control group. 11. Expectancy and the aroma of Roman chamomile influence mood and cognition in healthy volunteers

Abstract

To assess the effect of the aroma of the essential oil of Roman chamomile (Chamaemelum nobile) and the impact of induced expectancy on mood and cognition in healthy adults, 80 volunteers participated in an independent groups design study. There were four conditions: (1) aroma and arousal expectancy, (2) aroma and sedation expectancy, (3) aroma and no expectancy, and (4) no aroma controls. Expectancy effects were induced by means of a pre-test session where participants were provided with false information from a supposed official source. Pretest mood scales were completed followed by the Cognitive Drug Research (CDR) battery and a final post-test mood scale. Analysis revealed a significant effect of condition on overall quality of memory (p = 0.017), long-term memory (p = 0.008) and accuracy of attention (p = 0.024). Analysis of the pre to post-test changes in mood revealed significant changes in alertness (p=0.002), and calmness (p = 0.00004). The sedative nature of the aroma appeared to combine with induced sedative expectancy to impair cognitive performance to a greater degree than was found for aroma alone. Subjective alertness was influenced by the sedative effect of the aroma which was ameliorated to some degree by the induced arousal expectancy. Similarly, subjective calmness appeared to be influenced by both the sedative effect of the aroma and induced arousal expectancy. KEYWORDS Aroma; Chamomile; Expectancy; Cognition; Mood; Memory

Abstract

Introduction: With an aging population there is an important need for the development of effective treatments for the amelioration of cognitive decline. Multiple mechanisms underlie age-related cognitive decline including cerebrovascular disease, oxidative stress, reduced antioxidant capacity and mitochondrial dysfunction. CoQ10 is a novel treatment which has the potential to improve brain function in healthy elderly populations due to established beneficial effects on mitochondrial function, vascular function and oxidative stress. Methods and Analysis: We describe the protocol for a 90-day randomized controlled trial which examines the efficacy of Ubiquinol (200 mg/day) vs. placebo for the amelioration of cognitive decline in a healthy (non-demented) elderly sample, aged 60 years and over. The primary outcome is the effect of Ubiquinol at 90 days compared to baseline on CogTrack composite measures of cognition. Additional cognitive measures, as well as measures of cardiovascular function, oxidative stress, liver function and mood will also be monitored across 30-, 60- and 90- day time points. Data analyses will involve repeated measures analysis of variance (ANOVA). Discussion: This study will be the first of its kind to provide important clinical and mechanistic data regarding the efficacy of Ubiquinol as a treatment for age-related cognitive decline in the healthy elderly with important implications for productivity and quality of life within this age group. Clinical Trial Registration: The trial has been registered with the Australian and New Zealand Clinical Trials Registry (ANZCTRN12618001841268).

The precise cause of the neuronal degeneration underlying these disorders, and indeed normal brain ageing remains however elusive. Considering the limits of existing preventive methods, there is a desire to develop effective and safe strategies. Growing preclinical and clinical research in healthy individuals or at the early stage of cognitive decline has demonstrated the beneficial impact of nutrition on cognitive functions. The present review is the most recent in a series produced by the Nutrition and Mental Performance Task Force under the auspice of the International Life Sciences Institute Europe (ILSI Europe). The latest scientific advances specific to how dietary nutrients and non-nutrient may affect cognitive ageing are presented. Furthermore, several key points related to mechanisms contributing to brain ageing, pathological conditions affecting brain function, and brain biomarkers are also discussed. Overall, findings are inconsistent and fragmented and more research is warranted to determine the underlying mechanisms and to establish dose-response relationships for optimal brain maintenance in different population subgroups. Such approaches are likely to provide the necessary evidence to develop research portfolios that will inform about new dietary recommendations on how to prevent cognitive decline.

46. Improved working memory performance following administration of a single dose of American ginseng (Panax quinquefolius L.) to healthy middle-age adults

Abstract

Objective: A ginsenoside-rich extract of American ginseng (Panax quinquefolius L.), CereboostTM, was previously shown to improve\ud working memory and mood in healthy young individuals. The present study represented a partial replication investigating whether these\ud effects extended to healthy middle-aged individuals.\ud Methods: Fifty-two healthy volunteers (40–60 years old, mean age 51.63) received 200 mg of P. quinquefolius or a matching placebo\ud according to a double-blind, placebo-controlled, balanced, crossover design. The Cognitive Drug Research battery and the Computerised\ud Mental Performance Assessment System were used to evaluate cognitive performance at baseline then 1, 3 and 6 h following treatment.\ud Blood glucose and mood were co-monitored.\ud Results: Compared with placebo, P. quinquefolius improved cognitive performance on ‘Working Memory’ factor at 3 h. Similar effects were\ud observed in one of the two tasks making up this factor, spatial working memory. There were no significant effects on mood or blood glucose levels.\ud Conclusions: These data confirm that P. quinquefolius can acutely benefit working memory and extend the age range of this effect to\ud middle-aged individuals. These changes are unlikely to be underpinned by modulation of blood glucose in this population.

Abstract

There is a growing body of evidence from randomized controlled trials which indicates that consumption of berries has a positive effect upon the cognitive function of healthy adults. It has been recommended that studies combining cognitive and physiological measures be undertaken in order to strengthen the evidence base for the putative effects of flavonoid consumption on cognitive outcomes. This pilot study utilized a randomized, double-blind and placebo controlled crossover design to assess the influence of the acute administration of anthocyanin-rich blackcurrant juice, standardized at 500 mg of polyphenols, on mood and attention. Additionally, this trial used electroencephalography (EEG) to assess if any changes in cognitive performance are associated with changes in localized prefrontal cortex neuronal activity in nine healthy young adults. Outcomes from the pilot EEG data highlight an anxiolytic effect of the consumption of a single serve blackcurrant juice, as indexed by a suppression of α spectral power, and an increase in the slow wave δ and θ spectral powers. There was also an indication of greater alertness and lower fatigue, as indexed by an increase in β power and suppression of α spectral power. Outcomes from the CogTrack™ system indicated a small acute increase in reaction times during the digit vigilance task. [ABSTRACT FROM AUTHOR]
