In the case of Aspergillus fumigatus, which is an environmental fungus that also causes life-threatening infections in humans, antimicrobial resistance is suggested to arise from fungicide use in agriculture, as the chemicals used for plant protection are almost identical to the antifungals used clinically. However, removing azole fungicides from crop fields threatens the global food supply via a reduction in yield. In this study, we survey crop fields before and after fungicide application. In this study, we clarify the link between azole fungicide use on crop fields and resistance in a prototypical human pathogen through systematic soil sampling on farms in Germany and surveying fields before and after fungicide application. We observed a reduction in the abundance of A. fumigatus on fields following fungicide treatment in 2017, a finding that was not observed on an organic control field with only natural plant protection agents applied. However, this finding was less pronounced during our 2018 sampling, indicating that the impact of fungicides on A. fumigatus population size is variable and influenced by additional factors. The overall resistance frequency among agricultural isolates is low, with only 1 to 3% of isolates from 2016 to 2018 displaying resistance to medical azoles. Isolates collected after the growing season and azole exposure show a subtle but consistent decrease in susceptibility to medical and agricultural azoles. Whole-genome sequencing indicates that, despite the alterations in antifungal susceptibility, fungicide application does not significantly affect the population structure and genetic diversity of A. fumigatus in fields. Given the low observed resistance rate among agricultural isolates as well the lack of genomic impact following azole application, we do not find evidence that azole use on crops is significantly driving resistance in A. fumigatus in this context.

Cytokeratin 20 (CK20) and insulin-like growth factor 2 (IGF2) were previously proposed to be elevated in clinical samples from patients with bladder cancer (BCa). A two cohort design validation study was used to assess the relevance for BCa detection by transcript quantitation of both markers in urine samples. Their diagnostic value was assessed in comparison with voided urine cytology (VUC). RNA isolation was carried out using cellular sediments of urine samples from 196/103 histologically positive BCa patients, as well as 97/50 control subjects for the test (TC) and validation cohort (VC), respectively. In this study a set of 29 X-chromosomal short tandem repeats (STRs) located within the Xq26 region was evaluated. These STRs were found within the 133.14–133.45 Mb region around the HPRTB locus. Evaluation of the microsatellites was performed with regard to polymorphism, reliable amplification, and low stutter artefacts. DXS10101, DXS10102, and DXS10103 were identified as those X-STRs with highest diversity; i.e. PIC values of 0.7174–0.8933. The locus DXS10101 was the optimal candidate for the integration in the commercial available test system Mentype Argus X-8 PCR amplification kit.

To elucidate the minimal lipopolysaccharide (LPS) structure needed for the viability of Escherichia coli, suppressor-free strains lacking either the 3-deoxy-d-manno-oct-2-ulosonic acid transferase waaA gene or derivatives of the heptosyltransferase I waaC deletion with lack of one or all late acyltransferases (lpxL/M/P) and/or various outer membrane biogenesis factors were constructed. Δ(waaC lpxL lpxM lpxP) and waaA mutants exhibited highly attenuated growth, whereas simultaneous deletion of waaC and surA was lethal. Analyses of LPS of suppressor-free waaA mutants grown at 21 °C, besides showing accumulation of free lipid IVA precursor, also revealed the presence of its pentaacylated and hexaacylated derivatives, indicating in vivo late acylation can occur without Kdo. In contrast, LPS of Δ(waaC lpxL lpxM lpxP) strains showed primarily Kdo2-lipid IVA, indicating that these minimal LPS structures are sufficient to support growth of E. coli under slow-growth conditions at 21/23 °C. These lipid IVA derivatives could be modified biosynthetically by phosphoethanolamine, but not by 4-amino-4-deoxy-l-arabinose, indicating export defects of such minimal LPS. ΔwaaA and Δ(waaC lpxL lpxM lpxP) exhibited cell-division defects with a decrease in the levels of FtsZ and OMP-folding factor PpiD. These mutations led to strong constitutive additive induction of envelope responsive CpxR/A and σE signal transduction pathways. Δ(lpxL lpxM lpxP) mutant, with intact waaC, synthesized tetraacylated lipid A and constitutively incorporated a third Kdo in growth medium inducing synthesis of P-EtN and l-Ara4N. Overexpression of msbA restored growth of Δ(lpxL lpxM lpxP) under fast-growing conditions, but only partially that of the Δ(waaC lpxL lpxM lpxP) mutant. This suppression could be alleviated by overexpression of certain mutant msbA alleles or the single-copy chromosomal MsbA-498V variant in the vicinity of Walker-box II.

Die Auswertung der NAT erfolgt nach Agarosegelelektrophorese und Geldokumentation anhand mitgelieferter Langenstandards. Im Gegensatz zur klassischen Pilzdiagnostik weist der molekulargenetische Test auch totes Erregermaterial nach. Daher sind Proben von bereits anbehandelten Patienten ebenfalls sofort zur Diagnostik geeignet. Der Beitrag zeigt auf, wie dieses zukunftweisende Untersuchungsverfahren in der dermatologischen Praxis implementiert werden kann.

The bacterial DnaJ-like protein DjlA is composed of two functional domains, an N-terminal transmembrane domain of type III topology and a J domain at the C terminus. The authors investigated the Legionella pneumophila DjlA protein. The plasmid constructs were used to transform L. pneumophila and Escherichia coli strains. Overexpression of the L. pneumophila djlA gene in E. coli K-12 resulted in a mucoid phenotype due to colanic acid capsule production. The djlA mutants of all four strains did not exhibit alterations in growth rate in buffered yeast extract (BYE) broth when compared with the appropriate wild type, suggesting that djlA is not an essential gene in L. pneumophila. Incubation with a pool of 40% normal human serum revealed that the djlA mutants were as serum-resistant as the parent wild-type strains. The obtained CFU counts for strains R458 and Corby and the corresponding mutants are depicted. Thus, deletion of the djlA gene resulted in reduced virulence in all investigated L. pneumophila strains. To our knowledge, this is the first report of an influence on virulence by the djlA gene product in pathogenic bacteria. However, the authors do not know by which mechanism virulence attenuation is achieved in the L. pneumophila djlA mutants.

Recombinant plasmids encoding 3-deoxy-d-manno-oct-2-ulosonate-8-phosphate (Kdo-8-P) synthase (KdsA; EC 4.1.2.16) were identified from a cDNA library of Pisum sativum L. (pea) by complementing a temperature-sensitive kdsA ts mutant of the Gram-negative bacterium Salmonella enterica. Sequence analysis of several inserts revealed a central open reading frame encoding a protein of 290 amino acids with a high degree of amino acid sequence similarity to bacterial KdsA. The cDNA was confirmed by amplifying a 1,812-bp DNA fragment from the chromosome of pea that encoded four exons around the 5′-end of kdsA. The recombinant enzyme was subcloned, overexpressed and characterized to synthesize Kdo-8-P from d-arabinose-5-phosphate and phosphoenolpyruvate. The pH optimum was 6.1 and the activity of the enzyme was neither stimulated by the addition of divalent cations nor inhibited by EDTA. The cDNA of kdsA could not complement Escherichia coli K-12 strain AB3257, which is defective in all three isoenzymes (AroFGH) of 3-deoxy-d-arabino-hept-2-ulosonate-7-phosphate (Dha-7-P) synthase (EC 4.1.2.15), and neither d-erythrose-4-phosphate nor d-ribose-5-phosphate could substitute for d-arabinose-5-phosphate in vitro. Thus, plant cells possess a specific enzyme for the biosynthesis of Kdo-8-P with remarkable structural and functional similarities to bacterial KdsA proteins.

Heptosyltransferase II, encoded by the waaF gene of Escherichia coli, is a glycosyltransferase involved in the synthesis of the inner core region of lipopolysaccharide. The gene was subcloned from plasmid pWSB33 (Brabetz, W., Muller-Loennies, S., Holst, O. & Brade, H. (1997) Eur. J. Biochem. 247, 716-724) into a shuttle vector for the expression in the gram-positive host Corynebacterium glutamicum. The in vitro activity of the enzyme was investigated in comparison to that of heptosyltransferase I (WaaC) using as a source for the sugar nucleotide donor, ADP-l-glycero-d-manno-heptose, a low molecular mass filtrate from a DwaaCF E. coli strain. Synthetic lipid A analogues varying in the acylation or phosphorylation pattern or both were tested as acceptors for the subsequent transfer of 3-deoxy-d-manno-oct-2-ulosonic acid (Kdo) and heptose by successive action of Kdo transferase (WaaA), heptosyltransferase I (WaaC) and heptosyltransferase II (WaaF). The reaction products were characterized after separation by TLC and blotting with monoclonal antibodies specific for the acceptor, the intermediates and the final products.

The waaA gene encoding the essential, lipopolysaccharide (LPS)-specific 3-deoxy-Dmanno-oct-2-ulosonic acid (Kdo) transferase was inactivated in the chromosome of a heptosyltransferase I and II deficient Escherichia coli K-12 strain by insertion of gene expression cassettes encoding the waaA genes of Chlamydia trachomatis, Chlamydophila pneumoniae or Chlamydophila psittaci. The three chlamydial Kdo transferases were able to complement the knockout mutation without changing the growth or multiplication behaviour. The LPS of the mutants were serologically and structurally characterized in comparison to the LPS of the parent strain using compositional analyses, high performance anion exchange chromatography, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry and specific monoclonal antibodies. The data show that chlamydial Kdo transferases can replace in E. coli K-12 the host's Kdo transferase and retain the product specificities described in their natural background. In addition, we unequivocally proved that WaaA from C. psittaci transfers predominantly four Kdo residues to lipid A, forming a branched tetrasaccharide with the structure alpha-Kdo-(2-->8)-[alpha-Kdo-(2-->4)]-alpha-Kdo-(2-->4)-alpha-Kdo.

The 3-deoxy- d - manno -oct-2-ulosonic acid (Kdo) transferase gene of Legionella pneumophila was cloned and sequenced. Despite remarkable structural differences in lipid A, the gene complemented a corresponding Escherichia coli mutant and was shown to encode a bifunctional enzyme which transferred 2 Kdo residues to a lipid A acceptor of E. coli.

The obligate intracellular gram-negative bacterium Chlamydia contains as a major surface antigen a lipopolysaccharide (LPS) which harbours in its saccharide moiety a genus-specific epitope composed of a linear trisaccharide of 3-deoxy-D-manno- octulopyranosonic acid (Kdo) of the sequence aKdo-(2+8)-~~Kdo-(2+4)-aKdo. The structure was established on LPS of recombinant E. coli bacteria transformed with a plasmid carrying the gene for the chlamydia1 Kdo transferase which is a multifunctional glycosyl transferase. The structure was determined on deacylated and dephosphorylated LPS as well as on deacylated LPS by qualitative, quantitative and methylation analyses, fast-atom-bombardment mass spectrometry, and 'H-, I3C-, and 31P-NMR spectroscopy, and confirmed by chemcial synthesis. Artificial glycoconjugate antigens were synthesized and used for the preparation of murine monoclonal antibodies which had higher affinities than those prepared against the natural counterpart. These artificial antigens were also used as solid-phase antigens in an enzyme-immuno assay which was proven to be useful in the diagnosis of human chlamydia1 infections.

Murine monoclonal antibodies (MAbs) against the rough mutant lipopolysaccharides (LPS) of Salmonella minnesota strain R4 and R7 (chemotypes Rd2P - and Rd1P-, respectively) were obtained after immunization of BALB/c and NZB mice, respectively, with heat-killed bacteria. A total of 5 MAbs was obtained which were serologically characterized by hemagglutination, EIA and Western blot using LPS, de-O-acylated LPS and dephosphorylated LPS. In addition, the completely deacylated carbohydrate backbone, the deacylated, dephosphorylated and reduced carbohydrate backbone, and synthetic partial structures were covalently linked to bovine serum albumin resulting in artificial glycoconjugate antigens. Both groups of MAbs (anti-R4 and anti-R7) were highly specific for the Rd2P- and Rd1P- chemotypes, respectively, since they did not react with LPS of the Ra, Rb, Rc or Re chemotype. The R4 antibodies required for binding the core region [composed of 3-deoxy-α-D- manno-octulosonic acid (Kdo) and L-glycero-α-D- manno-heptopyranose (Hep)] and the phosphorylated glucosamine backbone of the lipid A moiety. The R7 antibodies bound to some extent to the core oligosaccharide Hep(1→3)Hep(1→5)Kdo(2→6), however, better binding was observed when the glucosamine backbone of lipid A was also present. In this case however, phosphoryl groups were not required. These antibodies can detect enterobacteria with an LPS of the Rd-chemotype, and may be useful to characterize also other naturally occurring rough mutants or genetically manipulated strains used to clone genes of the rfa locus.

Die DNA-Chiptechnologie bildet einen innovativen und effizienten Ansatz in der genetischen Diagnostik. Fur diese Technologie stellen wir die Entwicklung von Polymerbeschichtungen auf Glasoberflachen vor, welche die Analyse geringer DNA-Mengen durch die Methode der ALR (Arrayed Ligation Reaction) ermoglicht. Dieses Verfahren wurde fur den spezifischen Nachweis von Schimmelund Hausfaulepilzen bei der Biotype Diagnostic GmbH etabliert (Tafel 1). Neben der Hybridisierung komplementarer DNA-Strange findet dabei eine spezifische Enzymreaktion statt, bei der ein fluoreszenzmarkiertes DNA-Oligonukleotid kovalent an immobilisierte DNA-Sonden des Mikroarrays und damit an den DNA-Chip gebunden wird (Bild 1). Diese Ligationsreaktion findet jedoch nur statt, wenn die DNA-Zielsequenz zur Sonde und zum Ligationsoligonukleotid komplementar ist. [1-3]

The detection of DNA hybridization in medical diagnostics ought to be rapid, sensitive and specific. A platform technology based on surface plasmon resonance (SPR) is presented. We use TOPAS R chips with integrated optics and combined with microfluidics. Applying a nanoliter dispenser, thiol-modified single-stranded probe DNA (anti-tag) is deposited on the gold surface of the chips to create a DNA microarray. We fabricate chips with sufficiently high probe density, which is a key factor for DNA chips and can be controlled by adding MgCl2 to the immobilization solution. This technology offers the possibility of detecting PCR products comprising a single-stranded tag sequence being complementary to an anti-tag sequence of immobilized probes on the microarray. Consequently, this universal platform can be applied for detection of DNA hybridization based on the tag/anti-tag system. We demonstrate detection of specific hybridization of different 300 base pairs-long PCR products by SPR within less than five minutes. We checked for expected cross hybridizations with seven base pairs-long sequences at different positions in the tag/anti-tag sequence. Depending on the distance to the sensor surface we could observe crosshybridization if the according complementary sequence part is more distant from the surface. The initial binding rates (response units/min) at different PCR product concentrations were determined. Within five minutes a PCR product concentration of 2.6 nM is sufficient for distinct detection without crosshybridizations.

We report on the development of a new platform technology for the detection of genetic variations by means of surface plasmon resonance (SPR) spectroscopy. TOPAS chips with integrated optics were exploited in combination with microfluidics. Within minutes, the detection of hybridization kinetics was achieved simultaneously at all spots of the DNA microarray. A nanoliter dispenser is used to deposit thiol-modified single-stranded probe DNA on the gold surface of the chips. We investigated the influence of different parameters on hybridization using model polymerase chain reaction (PCR) products. These PCR products comprised a single-stranded tag sequence being complementary to an anti-tag sequence of probes immobilized on the gold surface. The signals increased with increasing length of PCR products (60, 100 or 300 base pairs) as well as with their concentration. We investigated hybridizations on DNA microarrays comprising 90 spots of probe DNA with three different sequences. Furthermore, we demonstrate that sequences with possible hairpin structures significantly lower the binding rate, and thus, the SPR signals during hybridization.

In Germany now, the recognition of Salmonella infections in pig herds is based on three different commercial tests detecting antibodies against Salmonella-derived lipopolysaccharide (LPS). However, a serious disadvantage of these tests, used so far, is the restricted detection of antibodies belonging predominantly to the immunoglobulin class g (IgG). Therefore, a new test was developed to detect three Ig classes (IgM, IgG and IgA). Different constellations between the three Ig classes allow the evaluation of the current infection status of each pig. Under field conditions, this was proved in three different vaccination trials using a commercial Salmonella Typhimurium live vaccine.

The haplotype discrimination capacity of the 9 Y-chromosomal short tandem repeat (Y-STR) loci comprising the so called minimal haplotype together with additional 26 recently described single-copy Y-STRs was evaluated within 391 males from Germany, The Netherlands, and Turkey. The aim of this study was to identify the minimum number of Y-STRs needed in addition to the recommended 9 minimal haplotype loci or the 11 SWGDAM loci for individualizing male lineages. Highest gene diversities were shown for DYS385 loci, DYS449, DYS481, DYS570, DYS447, DYS576, DYS389-II, and DYS390 (D = 0.7518-0.8746). The five Y-STRs DYS447, DYS449, DYS481, DYS570, and DYS576 comprised the smallest set of loci in addition to the previously recommended standard Y-STRs leading to the individualization of all males from each single population group. Complete resolution of the pooled population was achieved by the additional genotyping of two further loci, DYS446 or DYS505 and DYF406S1 or DYS522.

The evaluation of four pairs of X-chromosomal short tandem repeats (STRs), i.e. DXS10135-DXS8378, DXS7132-DXS10074, HPRTB-DXS10101 and DXS7423-DXS10134 was carried out using the Argus X-8 Multiplex amplification kit. These eight STRs are distributed as four closely linked pairs over the entire X-chromosome (ChrX), and for practical reasons they are assigned to four linkage groups 1-4. The molecular origin of DNA mutations and the mutation rates were analyzed at 14 short tandem repeat (STR) loci with samples from trio cases derived from 10 different German population samples. STR loci comprised of D2S1360, D3S1744, D4S2366, D5S2500, D6S474, D7S1517, D8S1132, D10S2325, D12S391, D18S51, D19S246, D20S480, D21S226, and D22S689. In a total of 488 meioses, 16 isolated genetic inconsistencies in 8 different STRs were observed, whereas no mutations were found at the other loci. The data of five mutations suggested the presence of silent or null alleles due to sequence variation in primer binding site. This could be confirmed for four suspected cases by the use of alternative primer sets and by DNA sequence analyses. Furthermore, this study revealed nine new allelic variants at five different loci.

The X chromosomal STR markers DXS10135 and DXS8378 in linkage group 1, DXS7132 and DXS10074 in linkage group 2, HPRTB and DXS10101 in linkage group 3, and DXS10134 and DXS7423 in linkage group 4 were studied in the Hungarian population. After genotyping unrelated men (219) and women (165), forensic efficiency parameters were calculated. Deviations from Hardy-Weinberg equilibrium could not be detected. There were several microvariant and rare alleles were sequenced: four in locus DXS10135 (alleles 17.1, 18.1, 20.1 and 26.1), one in locus DXS10074 (alleles 11), three in locus DXS10101 (alleles 26, 34.2 and 35) and five in locus DXS10134 (alleles 35.3, 37.2, 38.2, 39.2, 41).

The inner core region of the lipopolysaccharide (LPS) of Haemophilus influenzae is characterized by the presence of a phosphorylated 3-deoxy-α- d - manno -octulosonic acid (Kdo). In this study, we show that the heptosyltransferase I adding the first l - glycero - d - manno -heptose residue to this acceptor is encoded by the gene opsX , which differs in substrate specificity from the other heptosyltransferase I, known as WaaC.

Allele frequencies for the autosomal tetranucleotide short tandem repeat loci D4S2366, D6S474, D14S608, D19S246, D20S480, D21S226 and D22S689 were investigated in a sample of 189 unrelated German individuals using a multiplex polymerase chain reaction approach. The loci showed no significant deviations from Hardy-Weinberg equilibrium except for D14S608. All genotyped alleles were cloned and sequenced, and an allelic nomenclature consistent with the ISFH recommendations was defined.

The genes encoding the 3-deoxy-D- manno-oct-2-ulosonic acid (Kdo) transferase ( waaA) and heptosyltransferases I ( waaC) and II ( waaF) in Klebsiella pneumoniae were cloned from a DNA library by functional complementation of corresponding Escherichia coli and Salmonella enterica mutants. Sequence analyses revealed extensive homologies of the deduced proteins to their counterparts in other Enterobacteriaceae. However, differences were evident with regard to the chromosomal organization of the genes. To perform in vitro studies, the waaA, waaC and waaF genes were subcloned and expressed in the Gram-positive host Corynebacterium glutamicum. WaaA was characterized as a bifunctional enzyme capable of transferring two Kdo residues to a synthetic bisphosphorylated tetraacyl-lipid A precursor of E. coli (compound 406). In contrast, waaC and waaF were shown to encode specific glycosyltransferases catalyzing the consecutive transfer of two L- glycero-D- manno-heptose residues to Kdo2-406.

The lipopolysaccharide (LPS) of the deep rough mutant Haemophilus influenzae I69 consists of lipid A and a single 3-deoxy-d-manno-oct-2-ulosonic acid (Kdo) residue substituted with one phosphate at position 4 or 5 (Helander, I. M., Lindner, B., Brade, H., Altmann, K., Lindberg, A. A., Rietschel, E. T., and Zahringer, U. (1988) Eur. J. Biochem. 177, 483-492). The waaA gene encoding the essential LPS-specific Kdo transferase was cloned from this strain, and its nucleotide sequence was identical to H. influenzae DSM11121. The gene was expressed in the Gram-positive host Corynebacterium glutamicum and characterized in vitro to encode a monofunctional Kdo transferase. waaA of H. influenzae could not complement a knockout mutation in the corresponding gene of an Re-type Escherichia coli strain. However, complementation was possible by coexpressing the recombinant waaA together with the LPS-specific Kdo kinase gene (kdkA) of H. influenzae DSM11121 or I69, respectively. The sequences of both kdkA genes were determined and differed in 25 nucleotides, giving rise to six amino acid exchanges between the deduced proteins. Both E. coli strains which expressed waaA and kdkA from H. influenzae synthesized an LPS containing a single Kdo residue that was exclusively phosphorylated at position 4. The structure was determined by nuclear magnetic resonance spectroscopy of deacylated LPS. Therefore, the reaction products of both cloned Kdo kinases represent only one of the two chemical structures synthesized by H. influenzae I69.

The chromosomal genes rfaC and rfaF of Escherichia coli W3110 were inactivated by allelic-replacement mutagenesis to generate a defined strain lacking both heptosyltransferases which catalyze in lipopolysaccharide (LPS) biosynthesis the transfer of the first two L-glycero-D-manno-heptose (Hep) residues to 3-deoxy-D-manno-2-octulosonic acid (Kdo). The LPS of the mutant was isolated and its chemical structure was investigated by compositional analysis and nuclear magnetic resonance spectroscopy of isolated, deacylated oligosaccharide phosphates. The basic structure was a tetrasaccharide alpha-Kdo-(2-->4)-alpha-Kdo-(2-->6)-beta-D-GlcN4P-(1-->6)-alpha-D- GlcN1P which in LPS was substituted at position 07 of Kdo II by 2-aminoethanol phosphate in non-stoichiometric amounts. 2-Aminoethanol was cleaved during deacylation of the LPS by successive hydrazinolysis and KOH treatment and, in addition, phosphate migration from 07 to 08 of Kdo II occurred. Thus, the oligosaccharides alpha-Kdo7P-(2-->4)-alpha-Kdo-(2-->6)-beta-D-GlcN4P-(1-->6)- alpha-D-GlcN1P and alpha-Kdo8P-(2-->4)-alpha-Kdo-(2-->6)-beta-D-GlcN4P-(1-->6)- alpha-D-GlcN1P could be isolated. KOH treatment of the two trisphosphates and authentic methyl 3-deoxy-D-manno-octulopyranoside 7-(2-acetamidoethyl phosphate) proved that phosphate migration only took place when the phosphate group was substituted with 2-aminoethanol. Complementation studies with plasmid-encoded rfaC and rfaF genes revealed that the mutant strain can be used in combination with LPS-specific antibodies for the cloning and characterization of heptosytransferases which glycosylate Kdo residues of the inner core region of LPS.

The kdsA gene encoding 3-deoxy-D-manno-2-octulosonate-8-phosphate (Kdo-8-P) synthase of Chlamydia psittaci 6BC was cloned by complementing the temperature-sensitive kdsA mutant Salmonella enterica serovar Typhimurium AG701i50. The sequence analysis of a recombinant DNA fragment revealed an open reading frame of 807 nucleotides which codes for a polypeptide of 269 amino acids with a high degree of similarity to known KdsA proteins. In addition, alignments of Kdo-8-P synthases with bacterial and fungal 3-deoxy-D-arabino-2-heptulosonate-7-phosphate (Dha-7-P) synthases suggested that both classes of enzymes are structurally related and may belong to a family of 2-keto-3-deoxy-aldonic acid synthases. The chlamydial protein was overexpressed and functionally characterized in vitro to synthesize Kdo-8-P from D-arabinose 5-phosphate and phosphoenolpyruvate. A chlamydial DNA region upstream of the gene exhibiting similarities to the consensus sequence of sigma 70 promoters of Escherichia coli was responsible for the heterologous expression of kdsA.

The gene kdtA of Chlamydia pneumoniae strain TW-183, encoding the enzyme 3-deoxy-alpha-D-manno-octulosonic acid (Kdo) transferase of lipopolysaccharide biosynthesis, was cloned and sequenced. A single open reading frame of 1314 bp was identified, the deduced amino acid sequence of which revealed 69% similarity and 43% identity with KdtA of Chlamydia trachomatis and Chlamydia psittaci. The gene was expressed in the Gram-positive host Corynebacterium glutamicum and the primary gene product was characterized as a multifunctional glycosyltransferase. Cell-free extracts generated in vitro the genus-specific epitope of Chlamydia composed of the trisaccharide alphaKdo(2-8)alphaKdo(2-4)alphaKdo. The results show that a single polypeptide affords three different glycosidic bonds, which is in contradiction to the dogma of glycobiology: 'one enzyme - one glycosidic bond'.
