Abstract Background and objective Developing harmonious doctor-patient relationships is a powerful way to promote the construction of a new pattern of medical reform in developing countries. We aim to analyze the effects of China’s hierarchical medical system on doctor-patient relationships, thus contributing to China’s medical and health system reform. Methods With panel data on prefectural-level cities in China from 2012 to 2019, we used a time-varying difference-in-differences model to evaluate the effect of hierarchical medical treatment policy. Results Hierarchical medical treatment policies can significantly improve doctor-patient relationships, and this conclusion is supported by various robustness tests. And improving doctor-patient relationships can be indirectly realized by the optimization of resource allocation and saving of medical costs. In addition, the marginal effect of the pilot policy on doctor-patient relationships decreased with age within the city population. In focal cities and cities with high levels of fiscal spending on health care, the effect of the pilot policy on doctor-patient relationships was stronger. Conclusion While reinforcing the literature on the doctor-patient relationship, this study also provides a reference for further exploration of the pilot policy of hierarchical medical treatment and the development of new medical and health system reform in developing countries.

Bone nonunion poses an urgent clinical challenge that needs to be addressed. Recent studies have revealed that the metabolic microenvironment plays a vital role in fracture healing. Macrophages and bone marrow-derived mesenchymal stromal cells (BMSCs) are important targets for therapeutic interventions in bone fractures. Itaconate is a TCA cycle metabolite that has emerged as a potent macrophage immunomodulator that limits the inflammatory response. During osteogenic differentiation, BMSCs tend to undergo aerobic glycolysis and metabolize glucose to lactate. Copper ion (Cu2+) is an essential trace element that participates in glucose metabolism and may stimulate glycolysis in BMSCs and promote osteogenesis. In this study, we develop a 4-octyl itaconate (4-OI)@Cu@Gel nanocomposite hydrogel that can effectively deliver and release 4-OI and Cu2+ to modulate the metabolic microenvironment and improve the functions of cells involved in the fracture healing process. The findings reveal that burst release of 4-OI reduces the inflammatory response, promotes M2 macrophage polarization, and alleviates oxidative stress, while sustained release of Cu2+ stimulates BMSC glycolysis and osteogenic differentiation and enhances endothelial cell angiogenesis. Consequently, the 4-OI@Cu@Gel system achieves rapid fracture healing in mice. Thus, this study proposes a promising regenerative strategy to expedite bone fracture healing through metabolic reprogramming of macrophages and BMSCs.

Abstract Background Intraventricular hemorrhage (IVH) refers to bleeding within the brain’s ventricular system, and hydrocephalus is a life-threatening complication of IVH characterized by increased cerebrospinal fluid accumulation in the ventricles resulting in elevated intracranial pressure. IVH poses significant challenges for healthcare providers due to the complexity of the underlying pathophysiology and lack of standardized treatment guidelines. Herein, we performed a systematic review of the treatment strategies for hydrocephalus secondary to IVH. Methods This systematic review was prospectively registered with PROSPERO (CRD42023450786). The search was conducted in PubMed, Cochrane Library, and Web of Science on July 15, 2023. We included original studies containing valid information on therapy management and outcome of hydrocephalus secondary to primary, spontaneous, and subarachnoid or intracranial hemorrhage following IVH in adults that were published between 2000 and 2023. Glasgow Outcome Scale (GOS) or modified Ranking Scale (mRS) scores during follow-up were extracted as primary outcomes. The risk of bias was assessed using the Newcastle–Ottawa Scale for Cohort Studies or Cochrane Risk of Bias 2.0 Tool. Results Two hundred and seven patients from nine published papers, including two randomized controlled trials, were included in the analysis. The GOS was used in five studies, while the mRS was used in four. Seven interventions were applied, including craniotomy for removal of hematoma, endoscopic removal of hematoma with/without endoscopic third ventriculostomy (ETV), traditional external ventricular drainage (EVD), and various combinations of EVD, lumbar drainage (LD), and intraventricular fibrinolysis (IVF). Endoscopic removal of hematoma was performed in five of nine studies. Traditional EVD had no obvious benefit compared with new management strategies. Three different combinations of EVD, LD, and IVF demonstrated satisfactory outcomes, although more studies are required to confirm their reliability. Removal of hematoma through craniotomy generated reliable result. Generally, endoscopic removal of hematoma with ETV, removal of hematoma through craniotomy, EVD with IVF, and EVD with early continuous LD were useful. Conclusion EVD is still crucial for the management of IVH and hydrocephalus. Despite a more reliable result from the removal of hematoma through craniotomy, a trend toward endoscopic approach was observed due to a less invasive profile.

A disintegrin and metalloproteinase domain 10 (ADAM10), a member of the ADAM family, is a cellular surface protein with potential adhesion and protease/convertase functions. The expression regulations in cancers by natural products [adenosine (AD) and its analogs, cordycepin (CD), and N6, N6-dimethyladenosine (m62A)], and immune regulation are unclear. As results, AD, CD, and m62A inhibited ADAM10 expression in various cancer cell lines, indicating their roles in anti-cancer agents. Summary: Memory B cells (MBCs) are essential for humoral immunological memory and can emerge during both the pre-germinal center (GC) and GC phases. However, the transcription regulators governing MBC development remain poorly understood. Here, we report that the transcription regulator Notch2 is highly expressed in MBCs and their precursors at the pre-GC stage and required for MBC development without influencing the fate of GC and plasma cells. Mechanistically, Notch2 signaling promotes the expression of complement receptor CD21 and augments B cell receptor (BCR) signaling. Reciprocally, BCR activation up-regulates Notch2 surface expression in activated B cells via a translation-dependent mechanism. Intriguingly, Notch2 is dispensable for GC-derived MBC formation. In summary, our findings establish Notch2 as a pivotal transcription regulator orchestrating MBC development through the reciprocal enforcement of BCR signaling during the pre-GC phase and suggest that the generation of GC-independent and -dependent MBCs is governed by distinct transcriptional mechanisms.

Acute coronary syndrome (ACS) is a predominant cause of mortality, and the prompt and precise identification of this condition is crucial to minimize its impact. Recent research indicates that gut microbiota is associated with the onset, progression, and treatment of ACS. To investigate its role, we sequenced the gut microbiota of 38 ACS patients before and after percutaneous coronary intervention and statin therapy at three time points, examining differential species and metabolic pathways. We observed a decrease in the abundance of Parabacteroides, Escherichia, and Blautia in patients after treatment and an increase in the abundance of Gemalla, Klebsiella variicola, Klebsiella pneumoniae, and others. Two pathways related to sugar degradation were more abundant in patients before treatment, possibly correlated with disorders of sugar metabolism and risk factors, such as hyperglycemia, insulin resistance, and insufficient insulin secretion. Additionally, seven pathways related to the biosynthesis of vitamin K2 and its homolog were reduced after treatment, suggesting that ACS patients may gradually recover after therapy. The gut microbiota of patients treated with different statins exhibited notable differences after treatment. Rosuvastatin appeared to promote the growth of anti-inflammatory bacteria while reducing pro-inflammatory bacteria, whereas atorvastatin may have mixed effects on pro-inflammatory and anti-inflammatory bacteria while increasing the abundance of Bacteroides. Our research will provide valuable insights and enhance comprehension of ACS, leading to better patient diagnosis and therapy.

Background The incidence of papillary thyroid cancer (PTC) continues to rise all over the world, 10–15% of the patients have a poor prognosis. Although immunotherapy has been applied in clinical practice, its therapeutic efficacy remains far from satisfactory, necessitating further investigation of the mechanism of PTC immune remodeling and exploration of novel treatment targets.Methods This study conducted a single-cell RNA sequencing (scRNA-seq) analysis using 18 surgical tissue specimens procured from 14 patients diagnosed with adjacent tissues, non-progressive PTC or progressive PTC. Key findings were authenticated through spatial transcriptomics RNA sequencing, immunohistochemistry, multiplex immunohistochemistry, and an independent bulk RNA-seq data set containing 502 samples.Results A total of 151,238 individual cells derived from 18 adjacent tissues, non-progressive PTC and progressive PTC specimens underwent scRNA-seq analysis. We found that progressive PTC exhibits the following characteristics: a significant decrease in overall immune cells, enhanced immune evasion of tumor cells, and disrupted antigen presentation function. Moreover, we identified a subpopulation of lysosomal associated membrane protein 3 (LAMP3+) dendritic cells (DCs) exhibiting heightened infiltration in progressive PTC and associated with advanced T stage and poor prognosis of PTC. LAMP3+ DCs promote CD8+ T cells exhaustion (mediated by NECTIN2-TIGIT) and increase infiltration abundance of regulatory T cells (mediated by chemokine (C-C motif) ligand 17 (CCL17)-chemokine (C-C motif) receptor 4 (CCR4)) establishing an immune-suppressive microenvironment. Ultimately, we unveiled that progressive PTC tumor cells facilitate the retention of LAMP3+ DCs within the tumor microenvironment through NECTIN3-NECTIN2 interactions, thereby rendering tumor cells more susceptible to immune evasion.Conclusion Our findings expound valuable insights into the role of the interaction between LAMP3+ DCs and T-cell subpopulations and offer new and effective ideas and strategies for immunotherapy in patients with progressive PTC.

The diabetic wounds remain to be unsettled clinically, with chronic wounds characterized by drug-resistant bacterial infections, compromised angiogenesis and oxidative damage to the microenvironment. To ameliorate oxidative stress and applying antioxidant treatment in the wound site, we explore the function of folliculin-interacting protein 1 (FNIP1), a mitochondrial gatekeeper protein works to alter mitochondrial morphology, reduce oxidative phosphorylation and protect cells from unwarranted ROS accumulation. And our in vitro experiments showed the effects of FNIP1 in ameliorating oxidative stress and rescued impaired angiogenesis of HUVECs in high glucose environment. To realize the drug delivery and local regulation of FNIP1 in diabetic wound sites, a novel designed glucose-responsive HA-PBA-FA/EN106 hydrogel is introduced for improving diabetic wound healing. Due to the dynamic phenylboronate ester structure with a phenylboronic acid group between hyaluronic acid (HA) and phenylboronic acid (PBA), the hydrogel is able to realize a glucose-responsive release of drugs. Fulvic acid (FA) is added in the hydrogel, which not only severs as crosslinking agent but also provides antibacterial and anti-inflammatory abilities. Moreover, the release of FEM1b-FNIP1 axis inhibitor EN106 ameliorated oxidative stress and stimulated angiogenesis through FEM1b-FNIP1 axis regulation. These in vivo and in vitro results demonstrated that accelerated diabetic wounds repair with the use of the HA-PBA-FA/EN106 hydrogel, which may provide a promising strategy for chronic diabetic wound repair.

Abstract Background This paper analyzed the cases of dural arteriovenous fistula (DAVF) with spinal dural arteriovenous fistula (SDAVF) in the diagnosis and treatment process. Case presentation One case involving dural arteriovenous fistula (DAVF) with spinal dural arteriovenous fistula (SDAVF) from the 306th Hospital of PLA was retrospectively analyzed. The patient consulted the doctor due to lower limb sensory and motor disorders while exhibiting symptoms of urinary dysfunction. A computed tomographic angiography (CTA) and cerebral angiography confirmed the diagnosis of dural arteriovenous fistula (DAVF), necessitating surgical treatment. The patient was referred to our hospital for an magnetic resonance imaging (MRI) and a spinal angiography to obtain a confirmed diagnosis for spinal arteriovenous fistula, after which they underwent surgical fistula resection. The invasive intracranial dural arteriovenous fistula (DAVF) resection proceeded smoothly but did not ease the patient's symptoms. However, postoperative symptoms were partially relieved by the lumbar open spinal dural arteriovenous fistula adminstration. Conclusions Since not enough is understood about these two diseases, the rate of misdiagnosis is significantly increased. Early diagnosis and treatment of spinal dural arteriovenous fistula (SDAVF) can play a positive role during the recovery from neural function damage.

Abstract Highly efficient electrocatalysts for the hydrogen evolution reaction (HER) are essential for sustainable hydrogen energy. The controllable production of hydrogen energy by water decomposition depends heavily on the catalyst, and it is extremely important to seek sustainable and highly efficient water-splitting electrocatalysts for energy applications. Herein, bimetallic RuYO2 − x nanoparticles (Ru: 8.84 at.% and Y: 13 at.%) with high densities and low loadings were synthesized and anchored on graphene through a simple solvothermal strategy by synthesizing hydrogen yttrium ketone (HxYO2 − x) serving as an inserted medium. Electron microscopy demonstrated that the RuYO2 − x/C was composed of densely arranged particles and graphene flakes. Electrochemical results showed that the RuYO2 − x/C had a remarkably low overpotential of η10 = 56 mV at a current density of 10 mA cm−2 in alkaline media, a Tafel slope of 63.18 mV dec−1, and 24 h of stability. The oxygen vacancies of RuYO2 − x/C provided a large proton storage capacity and a strong tendency to bind hydrogen atoms. DFT calculations showed that RuYO2 − x/C catalysts with more Ru-O-Y bonds and VO dramatically decreased the energy barrier for breaking H-OH bonds. Moreover, the robust metal-support interactions provided optimized energies for hydrogen adsorption and desorption, which explained the high activity and favorable kinetics for RuYO2 − x/C catalytic hydrogen precipitation in alkaline electrolyte reactions. This work presents a hydrogen insertion method for the preparation of low-loading, high-density, high-performance and stable water decomposition catalysts for hydrogen production.

An adaptive tracking control strategy with a prescribe tracking error and the convergence time is proposed for hypersonic vehicles with state constraints and actuator failures. The peculiarity is that constructing a new time scale coordinate translation mapping method, which maps the prescribe time on the finite field to the time variable on the infinite field, and the convergence problem of the prescribe time is transformed into the conventional system convergence problem. The improved Lyapunov function, the improved tuning function, and the adaptive fault-tolerant mechanism are further constructed. Combined with the neural network, the prescribe time tracking control of the speed subsystem and the height subsystem are realized respectively. Combined with the Barbalat lemma and Lyapunov stability theory, the boundedness of the closed-loop system is proved. The simulation results have proven that, compared with other control strategies, it can ensure that the tracking error converges to the prescribe interval in the prescribe time and meets the constraints of the whole state of the system.

The world’s coral reefs are threatened by the cumulative impacts of global climate change and local stressors. Driven largely by a desire to understand the interactions between corals and their symbiotic microorganisms, and to use this knowledge to eventually improve coral health, interest in coral microbiology and the coral microbiome has increased in recent years. In this review, we summarize the role of the coral microbiome in maintaining a healthy metaorganism by providing nutrients, support for growth and development, protection against pathogens, and mitigation of environmental stressors. We explore the concept of coral microbiome engineering, that is, precise and controlled manipulation of the coral microbiome to aid and enhance coral resilience and tolerance in the changing oceans. Although coral microbiome engineering is clearly in its infancy, several recent breakthroughs indicate that such engineering is an effective tool for restoration and preservation of these valuable ecosystems. To assist with identifying future research targets, we have reviewed the common principles of microbiome engineering and its applications in improving human health and agricultural productivity, drawing parallels to where coral microbiome engineering can advance in the not-too-distant future. Finally, we end by discussing the challenges faced by researchers and practitioners in the application of microbiome engineering in coral reefs and provide recommendations for future work.

Abstract Breakage-fusion-bridge (BFB) is a complex rearrangement that leads to tumor malignancy. Existing models for detecting BFBs rely on the ideal BFB hypothesis, ruling out the possibility of BFBs entangled with other structural variations, that is, complex BFBs. We propose an algorithm Ambigram to identify complex BFB and reconstruct the rearranged structure of the local genome during the cancer subclone evolution process. Ambigram handles data from short, linked, long, and single-cell sequences, and optical mapping technologies. Ambigram successfully deciphers the gold- or silver-standard complex BFBs against the state-of-the-art in multiple cancers. Ambigram dissects the intratumor heterogeneity of complex BFB events with single-cell reads from melanoma and gastric cancer. Furthermore, applying Ambigram to liver and cervical cancer data suggests that the BFB mechanism may mediate oncovirus integrations. BFB also exists in noncancer genomics. Investigating the complete human genome reference with Ambigram suggests that the BFB mechanism may be involved in two genome reorganizations of Homo Sapiens during evolution. Moreover, Ambigram discovers the signals of recurrent foldback inversions and complex BFBs in whole genome data from the 1000 genome project, and congenital heart diseases, respectively.

Correlation between machined surface alterations and stress corrosion cracking (SCC) susceptibility of austenitic stainless steels is investigated. The machining-induced residual stress, roughness, micro-hardness and dislocation density were characterized and the surface/subsurface cracks were examined after SCC tests in boiling magnesium chloride solution. The results showed SCC crack initiation and early propagation were highly related to the machining-induced alterations. By establishing a quantitative relationship between the surface characteristics and surface crack density, the comprehensive impact of the residual stress, surface roughness as well as the dislocation density on SCC susceptibility is revealed. The predicted crack density is in good agreement with the measured data. As a result, the SCC susceptibility of the machined surface is described by the surface integrity properties.

Abstract Background Thalassemia is an extremely prevalent monogenic inherited blood disorder in southern China. It is important to comprehensively understand the molecular spectrum of thalassemia in an area with such a high prevalence of thalassemia before taking appropriate actions for the prevention and treatment of this disorder. Herein, we explored the clinical feasibility of using next-generation sequencing (NGS) for large-scale population screening to illustrate the prevalence and spectrum of thalassemia in Southern Jiangxi. Methods Blood samples collected from 136,312 residents of reproductive age in Southern Jiangxi were characterized for thalassemia by NGS. A retrospective analysis was then conducted on blood samples determined to be positive for thalassemia. Results In total, 19,827 (14.545%) subjects were diagnosed as thalassemia carriers, and the thalassemia prevalence rate significantly varied by geographical region (p T accounting for 35.257% of these β-thalassemia genotypes. Furthermore, 74 genotypes were identified among 608 individuals with combined α- and β-thalassemia. Notably, most individuals with rare thalassemia mutations had mildly abnormal hematologic parameters including microcytic hypochromia. Conclusions Our findings demonstrate the great heterogeneity and diverse spectrum of thalassemia in Southern Jiangxi, emphasizing the importance and necessity of persistent prevention and control of thalassemia in this region. Additionally, our findings further suggest that NGS can effectively identify rare mutations and reduce the misdiagnosis rate of thalassemia.

Abstract Various joint pathologies such as osteochondritis dissecans, osteonecrosis, rheumatic disease, and trauma, may result in severe damage of articular cartilage and other joint structures, ranging from focal defects to osteoarthritis (OA). The osteochondral unit is one of the critical actors in this pathophysiological process. New approaches and applications in tissue engineering and regenerative medicine continue to drive the development of OA treatment. Hydrogel scaffolds, a component of tissue engineering, play an indispensable role in osteochondral regeneration. In this review, tissue engineering strategies regarding osteochondral regeneration were highlighted and summarized. The application of hydrogels for osteochondral regeneration within the last five years was evaluated with an emphasis on functionalized physical and chemical properties of hydrogel scaffolds, functionalized delivery hydrogel scaffolds as well as functionalized intelligent response hydrogel scaffolds. Lastly, to serve as guidance for future efforts in the creation of bioinspired hydrogel scaffolds, a succinct summary and new views for specific mechanisms, applications, and existing limitations of the newly designed functionalized hydrogel scaffolds were offered. Using data from three automatic weather stations (LGB69, Eagle and Dome A) from distinctly different climatological zones along the CHINARE (Chinese National Antarctic Research Expedition) traverse route from Zhongshan Station to Dome A, we investigated the characteristics of meteorological conditions and subsurface heat conduction. Spatial analysis indicated decreasing trends in air temperature, relative humidity and wind speed from the coastal katabatic wind zone to the inland plateau region, and air temperatures clearly showed a strong daily variability in winter, suggesting the effect from the fluctuation in the Antarctic atmospheric system. We also analyzed the optimal response time of the 1 and 3 m depth snow temperatures to the 0.1 m depth snow temperature for each site under clear/overcast and day/night situations. This showed an important enhancement to the heat transfer from shortwave radiation penetration. Using an iterative optimization method, we estimated the subsurface heat conduction variations along the transect. This was ~3–5 W m–2. Multiple maxima in daily mean subsurface fluxes were found in winter, with a typical value above 2 W m–2, while a single minimum value under –2 W m–2 was found in summer. On an annual scale, a larger mean loss of subsurface heat conduction was observed in the inland plateau compared to in the coastal katabatic area. Finally, we discussed the possible influences of turbulent and radiant transport on the vertical heat response and confirmed the wind enhancement on the growth of thermal conductivity. This preliminary study provides a brief perspective and an important reference for studying subsurface heat conduction in inland areas of Antarctica.

Lagerstroemia is a genus with a wide geographical range extending from Southeast Asia to Australia, and it plays a significant role in many aspects of urban life, including supporting ecosystems, beautifying cities, and affecting human health. To gain insights into the current state of Lagerstroemia research, a bibliometric analysis was conducted using 138 publications from the Web of Science core database between 2002 and 2022. The analysis involved co-occurrence analyses of authors’ affiliated institutions, a keyword co-occurrence analysis, and a timeline view of keywords. The results of the analysis revealed three stages of Lagerstroemia research: the initial stage (2002–04); the rapid growth stage (2005–13); and the in-depth stage (2014–present). Furthermore, 335 keywords from 10 clusters were identified, which indicated that genetic diversity, breeding, and landscape utilization were the three main research areas of Lagerstroemia. Collaboration among authors mainly occurred within major natural and cultivated regions, such as China and the United States. Based on the bibliometric evaluation, future research of Lagerstroemia should focus on areas such as whole-genome sequencing, male sterility problems, resistance, ornamental characteristics, and landscape utilization guidelines. The findings of this study provide an important reference for further studies and can guide the development, innovation, and application of Lagerstroemia.

Abstract With large‐scale wind farms connected to AC/DC network, the transient stability assessment (TSA) of the power system becomes more and more difficult. Among them, estimating the region of attraction (ROA) of the equilibrium point is a traditional but still challenging problem. Based on the Lyapunov stability theory, this paper proposes a new approach to obtain the enlarged estimation of the ROA. The optimization and updating strategy of shape function in the sum of squares (SOS) optimization problem are studied to reduce the conservatism of estimation result. In the proposed method, the gravitational search algorithm (GSA) is employed to optimize the coefficients of initial shape function to improve estimation performance. Based on the time‐domain simulation (TDS) of expected faults, the fitness value for shape function optimization is calculated using the values of state variables at the fault clearing time and the system stability information. Furthermore, the optimal Lyapunov function is computed by introducing the update condition of shape function and adjusting the iteration strategy of the ROA estimation algorithm. Finally, the proposed method is applied to a two‐machine‐infinite‐bus system and a more complex nine‐bus AC/DC system with wind power. And the effectiveness of the proposed method is verified by comparing with the existing ROA estimation methods.

Abstract Atmospheric nitrous acid (HONO) chemistry is of critical importance to air quality during polluted haze events, especially in China. However, current air quality models (such as WRF-CHEM, WRF-CMAQ, Box-MCM) generally underestimate the concentration of HONO, leading to a lack of fundamental understanding of haze pollution. Here, by combining field observations during haze events in Beijing and modeling results, we developed the new parameterization scheme for heterogeneous nitrogen dioxide (NO2) reaction on aerosol surfaces with the synergistic effects of relative humidity and ammonia, which has not been considered in existing air quality models. Including NO2 heterogeneous reactions into modeling significantly improves the estimation accuracy of HONO and OH levels, with the contribution reaching up to 91% and 78% during pollution episodes. The OH derived by HONO can partly explain high concentrations of particulate matter. Together, our work provides a new approach to illustrate the formation of HONO, OH, and haze with the consideration of heterogeneous NO2 → HONO chemistry.

Abstract Most variations in the human genome refer to single-nucleotide variation (SNV), small fragment insertions and deletions, and genomic copy number variation (CNV). Many human diseases including genetic disorders are associated with variations in the genome. These disorders are often difficult to be diagnosed because of their complex clinical conditions, therefore, an effective detection method is needed to facilitate clinical diagnosis and prevent birth defects. With the development of high-throughput sequencing technology, the method of targeted sequence capture chip has been extensively used owing to its high throughput, high accuracy, fast speed, and low cost. In this study, we designed a chip that potentially captured the coding region of 3043 genes associated with 4013 monogenic diseases, with an addition of 148 chromosomal abnormalities that can be identified by targeting specific regions. To assess the efficiency, a strategy of combining the BGISEQ500 sequencing platform with the designed chip was utilized to screen variants in 63 patients. Eventually, 67 disease-associated variants were found, 31 of which were novel. The results of the evaluation test also show that this combined strategy complies with the requirements of clinical testing and has proper clinical application value.

Abstract The transfusion of red blood cells (RBCs) is crucial in current medicinal research. The shelf‐life of donated RBCs preserved by the normal method is very short, limiting their clinical application. Cryopreservation is a reliable and effective technology for the long‐term storage of RBCs. During the process, ice formation will cause irreversible injuries to RBCs. Glycerol is used as a cryoprotectant (CPA) for RBCs to mitigate cryoinjuries. But it severely induces RBC hemolysis and deformation. This review introduces some biocompatible and effective CPAs used for RBC cryopreservation, outlining recent advances. Currently, there is no uniform standard to determine whether a CPA is suitable for RBCs. According to previous studies, we summarize for the first time comprehensive methods to evaluate the performance of CPAs by ensuring their safety and effectiveness. The safety of CPAs is defined as the degree of damage to RBCs, while the effectiveness is demonstrated by the properties of thawed RBCs, including membrane properties, protein activities, rheological properties, and metabolites levels. A novel CPA that is confirmed suitable for RBCs by these methods may be applied to other cells. We believe comprehensive methods can thoroughly evaluate the performance of CPAs and promote the development of transfusion therapy in clinic.

Magnolia figo (Lour.) DC. var. crassipes (Law) Figlar & Noot. is a captivating, purple-flowered plant of the Magnoliaceae family (Magnolia). It is a small tree or shrub up to 2 to 5 m in height. Indigenous to China, M. figo var. crassipes holds a significant position as a parental contributor in controlled crosses within the Magnoliaceae lineage. This acclaim arises from its strikingly deep-purple hue and the entrancing fragrance exuded by its blossoms. Magnolia figo is a native to the southern provinces of China. This evergreen shrub maintains a stature of 2 to 3 m and has garnered popularity for its aromatic blooms. Notably, both M. figo var. crassipes and M. figo exhibit considerable potential in terms of development and use as ornamental trees. Their intrinsic value finds expression in horticulture, garden design, and urban landscapes, as elaborated by Chai et al. (2018).

α-globin gene triplication carriers were not anemic in general, while some studies found that α-globin gene triplication coinherited with heterozygous β-thalassemia may cause adverse clinical symptoms, which yet lacks sufficient evidence in large populations. In this study, we investigated the prevalence and distribution of α-globin gene triplication as well as the phenotypic characteristics of α-globin gene triplication coinherited with heterozygous β-thalassemia in Ganzhou city, southern China. During 2021-2022, a total of 73,967 random individuals who received routine health examinations before marriage were genotyped for globin gene mutations by high-throughput sequencing. Among them, 1,443 were α-globin gene triplication carriers, with a carrier rate of 1.95%. The most prevalent mutation was αααanti3.7/αα (43.10%), followed by αααanti4.2/αα (38.12%). 42 individuals had coinherited α-globin gene triplication and heterozygous β-thalassemia. However, they did not differ from the individuals with heterozygous β-thalassemia and normal α-globin (αα/αα) in terms of mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) levels. In addition, heterogenous clinical phenotypes were found in two individuals with the same genotype. Our study established a database of Ganzhou α-globin gene triplication and provided practical advice for the clinical diagnosis of α-globin gene triplication.

Summary: Even though the discovery of lytic polysaccharide monooxygenases (LPMOs) has fundamentally shifted our understanding of biomass degradation, most of the current studies focused on their roles in carbohydrate oxidation. However, no study demonstrated if LPMO could directly participate to the process of lignin degradation in lignin-degrading microbes. This study showed that LPMO could synergize with lignin-degrading enzymes for efficient lignin degradation in white-rot fungi. The transcriptomics analysis of fungi Irpex lacteus and Dichomitus squalens during their lignocellulosic biomass degradation processes surprisingly highlighted that LPMOs co-regulated with lignin-degrading enzymes, indicating their more versatile roles in the redox network. Biochemical analysis further confirmed that the purified LPMO from I. lacteus CD2 could use diverse electron donors to produce H2O2, drive Fenton reaction, and synergize with manganese peroxidase for lignin oxidation. The results thus indicated that LPMO might uniquely leverage the redox network toward dynamic and efficient degradation of different cell wall components.

The constitutive model plays a decisive role in the accuracy and reliability of the simulation results. Because the material flow characteristics obtained by the traditional experimental method are difficult to describe thermoplastic deformation behavior in the cutting process accurately, and the inverse identification is usually carried out based on a two-dimensional orthogonal cutting model, ignoring the actual cutting state, which leads to significant deviations in the results of parameter identification. Therefore, a three-dimensional (3-D) turning finite element simplified model which conforms to the actual cutting scenario was established. Based on this model, a coupling simulation was used to inverse identify the Johnson-Cook (J-C) parameters for turning 304 stainless steel. Firstly, the automatic finite element modeling and calculation of 3-D turning is implemented by Python. Then, the inverse identification framework was built based on the ISIGHT. The initial yield strength of J-C parameters was obtained from quasi-static compression experiments. With the objective function of minimizing the error between the experimental cutting force and the simulated cutting force, the inverse identification of J-C parameters (strain reinforcement factor, strain rate sensitivity, thermal softening index, and hardening index) was carried out by a multi-island genetic algorithm. Finally, compared with cutting forces, chip morphology, and residual stresses, the feasibility of the inverse identification method and the accuracy of the constitutive model is demonstrated, which also provides a reference for the optimal identification method of constitutive parameters of difficult-to-machine materials.

In this work, Ag/Bi5O7I nanocomposite was prepared and firstly applied in piezo/photocatalytic reduction of N2 to NH3 and methyl orange (MO) degradation. Bi5O7I was synthesized via a hydrothermal-calcination method and shows nanorods morphology. Ag nanoparticles (NPs) were photo deposited on the Bi5O7I nanorods as electron trappers to improve the spatial separation of charge carriers, which was confirmed via XPS, TEM, and electronic chemical analyses. The catalytic test indicates that Bi5O7I presents the piezoelectric-like behavior, while the loading of Ag NPs can strengthen the character. Under ultrasonic vibration, the optimal Ag/Bi5O7I presents high efficiency in MO degradation. The degradation rate is determined to be 0.033 min−1, which is 4.7 folds faster than that of Bi5O7I. The Ag/Bi5O7I also presents a high performance in piezocatalytic N2 fixation. The piezocatalytic NH3 generation rate reaches 65.4 μmol L−1 g−1 h−1 with water as a hole scavenger. The addition of methanol can hasten the piezoelectric catalytic reaction. Interestingly, when ultrasonic vibration and light irradiation simultaneously act on the Ag/Bi5O7I catalyst, higher performance in NH3 generation and MO degradation is observed. However, due to the weak adhesion of Ag NPs, some Ag NPs would fall off from the Bi5O7I surface under long-term ultrasonic vibration, which would greatly reduce the piezoelectric catalytic performance. This result indicates that a strong binding force is required when preparing the piezoelectric composite catalyst. The current work provides new insights for the development of highly efficient catalysts that can use multiple energies.

Efforts to develop high-performance electrocatalysts for the hydrogen evolution reaction (HER) are of utmost importance in ensuring sustainable hydrogen production. The controllable fabrication of inexpensive, durable, and high-efficient HER catalysts still remains a great challenge. Herein, we introduce a universal strategy aiming to achieve rapid synthesis of highly active hydrogen evolution catalysts using a controllable hydrogen insertion method and solvothermal process. Hydrogen vanadium bronze HxV2O5 was obtained through controlling the ethanol reaction rate in the oxidization process of hydrogen peroxide. Subsequently, the intermetallic PtCoVO supported on two-dimensional graphitic carbon nitride (g-C3N4) nanosheets was prepared by a solvothermal method at the oil/water interface. In terms of HER performance, PtCoVO/g-C3N4 demonstrates superior characteristics compared to PtCo/g-C3N4 and PtCoV/g-C3N4. This superiority can be attributed to the notable influence of oxygen vacancies in HxV2O5 on the electrical properties of the catalyst. By adjusting the relative proportions of metal atoms in the PtCoVO/g-C3N4 nanomaterials, the PtCoVO/g-C3N4 nanocomposites show significant HER overpotential of η10 = 92 mV, a Tafel slope of 65.21 mV dec−1, and outstanding stability (a continuous test lasting 48 h). The nanoarchitecture of a g-C3N4-supported PtCoVO nanoalloy catalyst exhibits exceptional resistance to nanoparticle migration and corrosion, owing to the strong interaction between the metal nanoparticles and the g-C3N4 support. Pt, Co, and V simultaneous doping has been shown by Density Functional Theory (DFT) calculations to enhance the density of states (DOS) at the Fermi level. This augmentation leads to a higher charge density and a reduction in the adsorption energy of intermediates.

In contrast to noble metals, graphene exhibits significantly lower loss, especially useful for optical sensing applications that require ultrahigh Q factors, and offer wide range tunability via an adjustable Fermi level. However, precise graphene patterning is difficult, especially for large areas, severely limiting its applications. Here, a tunable terahertz metamaterial absorber (TMMA) with ultrahigh Q factors consisting of a continuous, pattern-free graphene is demonstrated. A graphene sheet is overlaid on an Al metal array, forming a structure that supports strong localized surface plasmon polaritons (LSPPs) with fields tightly confined in the graphene, minimizing loss. Theoretical results show that this TMMA exhibits an ultrahigh Q factor of 1730, a frequency sensitivity of 2.84 THz/RIU, and an excellent figure of merit (FoM) of 365.85 RIU−1, independent of polarization. A tunability from ~2.25 to ~3.25 THz is also achieved by tuning Ef of graphene from 0.3 to 0.7 eV. The proposed graphene-based TMMA holds many potential applications, particularly in the field of sensing.

Abstract Phellodendron has always been of great significance in promoting human health and ecological restoration. However, human activities and climate change have severely affected habitat, population dynamics and sustainable use of Phellodendron. Little is known about the geographical distribution pattern and their responses to climate change of Phellodendron. In order to reveal the impact of climate change on Phellodendron, we conducted a study based on natural distribution data of two species (297 occurrence points), 20 environmental factors, and an optimized MaxEnt model. Our results identified the main environmental factors influencing Phellodendron, predicted their potential geographical distribution, and assessed migration trends under climate change in China. Our analysis showed that Ph. amurense and Ph. chinense have potential suitable habitats of 62.89 × 104 and 70.71 × 104 km2, respectively. Temperature and precipitation were found to play an essential role in shaping the present geographical distribution of Phellodendron populations. Based on two future climate scenarios, we forecasted that the potential suitable habitat of Ph. amurense would decrease by 12.52% (SSP245) and increase by 25.28% (SSP585), while Ph. chinense would decline by 19.61% (SSP245) and 15.78% (SSP585) in the late‐21st century. The potential suitable habitats of Ph. amurense and Ph. chinense would shift to northward and westward, respectively. Hydrothermal change was found to be the primary driver of the suitable habitat of Phellodendron populations in the future. We recommend establishing nature reserves for existing Phellodendron populations, especially Ph. chinense. Our study provided a practical framework for the impact of climate change on the suitable habitat of Phellodendron species and guided regional cultivation, long‐term conservation, and sustainable use.

Nocardiosis is an infectious disease caused by Nocardia that primarily affects immunocompromised hosts. Mycobacterium abscessus is a common opportunistic pathogen that causes disease in humans, including pulmonary and extrapulmonary infection. Nocardia spp. infection is uncommon, and infection with Nocardia wallacei and Mycobacterium abscessus is even rarer. A 59-year-old immunocompetent woman with risk factors for environmental exposure developed nocardiosis and presented to the hospital with a cough, shortness of breath, hemoptysis, and a back abscess. An enhanced computed tomography (CT) of the chest revealed partial destruction of the right lung, as well as consolidation of the right upper lobe. Rare pathogens N. wallacei and Mycobacterium abscessus were detected by metagenomic next-generation sequencing (mNGS) from abscess on the back and lung puncture tissue, respectively. She was treated with a combination of antibiotics and was finally discharged with a good prognosis. In this case, we present a patient who was successfully diagnosed with N. wallacei and Mycobacterium abscessus infection using mNGS. This importance of using mNGS in pathogen detection and the effective use of antibiotics in treating patients with long-term rare infections is highlighted in this report.

ObjectivesThe aim of this study was to improve the diagnostic performance of nuclear medicine physicians using a deep convolutional neural network (DCNN) model and validate the results with two multicenter datasets for thyroid disease by analyzing clinical single-photon emission computed tomography (SPECT) image data.MethodsIn this multicenter retrospective study, 3194 SPECT thyroid images were collected for model training (n=2067), internal validation (n=514) and external validation (n=613). First, four pretrained DCNN models (AlexNet, ShuffleNetV2, MobileNetV3 and ResNet-34) for were tested multiple medical image classification of thyroid disease types (i.e., Graves’ disease, subacute thyroiditis, thyroid tumor and normal thyroid). The best performing model was then subjected to fivefold cross-validation to further assess its performance, and the diagnostic performance of this model was compared with that of junior and senior nuclear medicine physicians. Finally, class-specific attentional regions were visualized with attention heatmaps using gradient-weighted class activation mapping.ResultsEach of the four pretrained neural networks attained an overall accuracy of more than 0.85 for the classification of SPECT thyroid images. The improved ResNet-34 model performed best, with an accuracy of 0.944. For the internal validation set, the ResNet-34 model showed higher accuracy (p < 0.001) when compared to that of the senior nuclear medicine physician, with an improvement of nearly 10%. Our model achieved an overall accuracy of 0.931 for the external dataset, a significantly higher accuracy than that of the senior physician (0.931 vs. 0.868, p < 0.001).ConclusionThe DCNN-based model performed well in terms of diagnosing thyroid scintillation images. The DCNN model showed higher sensitivity and greater specificity in identifying Graves’ disease, subacute thyroiditis, and thyroid tumors compared to those of nuclear medicine physicians, illustrating the feasibility of deep learning models to improve the diagnostic efficiency for assisting clinicians.

Abstract Background Yak (Bos grunniens) mainly inhabiting Tibet Plateau, displayed a high incidence of diarrhea due to harsh living environment and nutritional deficit. Gut microbial community has been reported to be closely related to many diseases including diabetes, obesity and inflammatory bowel disease, but information regarding diarrheic influence on gut microbiota in yaks remains scarce. Here, this study was performed to investigate the gut bacterial and fungal alternations of diarrheic yaks. Results Results revealed that the gut bacterial and fungal communities of diarrheic yaks showed a distinct decline in alpha diversity, accompanied by significant shifts in taxonomic compositions. Specifically, diarrhea caused a distinct increase in the relative abundance of 1 phylum and 8 genera as well as a distinct decrease in 3 phyla and 30 genera. Fungal taxonomic analysis indicated that the relative richness of 1 phylum and 2 genera dramatically increased, whereas the relative richness of 2 phylum and 43 genera significantly decreased during diarrhea. Surprisingly, 2 bacterial genera and 5 fungal genera even cannot be detected in the gut microbiota of diarrheic yaks. Conclusions In summary, this study indicated that the gut bacterial and fungal compositions and diversities of yaks altered significantly during diarrhea. Moreover, these findings also contribute to understanding the gut microbial composition and diversity of yaks and developing strategies to alleviate and prevent diarrhea from gut microbial perspective.

Sarcopenia is a senile disease with high morbidity, serious complications and limited clinical treatments. Menopause increases the risk of sarcopenia in females, while the exact pathogenesis remains unclear. To systematically investigate the development of hormone-related sarcopenia, we established a model of sarcopenia by ovariectomy and recorded successive characteristic changes. Furthermore, we performed the transcriptome RNA sequencing and bioinformatics analysis on this model to explore the underlying mechanism. In our study, we identified an integrated model combining obesity, osteoporosis and sarcopenia. Functional enrichment analyses showed that most of the significantly enriched pathways were down-regulated and closely correlated with endocrine and metabolism, muscle dysfunction, cognitive impairment and multiple important signaling pathways. We finally selected eight candidate genes to verify their expression levels. These findings confirmed the importance of estrogen in the maintenance of skeletal muscle function and homeostasis, and provided potential targets for further study on hormone-related sarcopenia.

In this study, we investigated the eff ects of diff erent litter sizes on the rate of uterine involution in Hu sheep. Using B-mode ultrasonography, we recorded changes in the uterine horn diameter and the maximum uterine caruncle diameter in 60 primiparous Hu sheep from days 0 to 45 postpartum. Th e uterine horn diameter decreased gradually postpartum from 79±1.42 mm at day 0 to 10.87±0.5 mm at day 45 with singleton parturition, from 91±6.58 mm at day 0 to 10.63±0.32 mm at day 45 with twin parturition, and from 107±3.67 mm at day 0 to 11±0.87 mm at day 45 with triplet parturition. Th e time to complete uterine involution postpartum was 30, 35 and 40 days postpartum in singleton, twin and triplet parturitions, respectively. (P0.05). Th us, diff erent litter sizes had a greater eff ect on postpartum uterine horn recovery than on the uterine caruncle. Furthermore, the rate of uterine involution in ewes with singleton parturition was significantly higher than that with triplet parturition. Our findings provide a reference for improving the reproductive performance of Hu sheep.

Abstract Background DNA polymerase β is one of the key enzymes involved in DNA damage repair and its proper expression is strictly controlled within different cells. We previously reported that three genetic mutations in the promoter region of the polb gene are prevalent in the Chinese Han population and two types of mutation are associated with thymic hyperplasia. The purpose of this study was to explore whether other mutated sites exist within the promoter region of the polb gene. Methods Genomic DNAs of 421 healthy Chinese Han individuals were extracted from whole blood samples and used for gene amplification of the promoter region of the polb gene. After gel purification, PCR amplicons were sequenced by the Sanger sequencing method and used for sequence alignment with the Lasergene program. PCR products with novel mutations were then subcloned into luciferase reporter plasmid pGL4.10 and transfected into 293T cells for dual‐luciferase activity analysis. Results In total, 11 mutated sites were detected in the Chinese Han population and eight of these were reported for the first time. Using a dual luciferase reporter system, it was found that one novel mutation −142 C > G could decrease the transcription activity of the polb gene, whereas two novel mutations, −152_−151insC and −218 C > G, could significantly increase the transcription activity of the polb gene. Conclusions High polymorphic sites could be found in the promoter region of polb gene and approximately half of them could influence its transcription activity.

BackgroundThe COVID-19 pandemic has disrupted the diagnosis, treatment, and care for tuberculosis (TB). Delays in seeking TB care may result in increased community transmission and unfavorable treatment outcomes. We sought to understand the influence of the COVID-19 pandemic on the proportion of patients with TB who delayed seeking the diagnosis and care for TB and explore the reasons for their postponement.MethodsWe surveyed a representative sample of outpatients treated for pulmonary TB from June to November 2020 using an anonymous standardized questionnaire. Multivariable logistic regression was used to calculate adjusted odds ratios (aOR) and 95% confidence intervals (CIs) of factors associated with the postponement of TB care. We used routinely collected surveillance data to assess trends of TB reports before and after the emergence of COVID-19 (2017–2019 vs. 2020–2022) in Tianjin, China.ResultsAmong 358 participants who were diagnosed with pulmonary TB during the COVID-19 response, 61 (17%) postponed seeking TB diagnosis due to COVID-19, with 39 (64%) citing fear as the primary reason. Female sex (aOR:2.0; 95% CI: 1.1–3.7), previous antituberculosis treatment (aOR:3.2; 95%CI: 1.4–7.6), and TB diagnosis during the first-level response (aOR = 3.2, 1.7–6.2) were associated with the postponement. Among all 518 participants receiving antituberculosis treatment, 57 (11%) had postponed their regular healthcare visits due to COVID-19, 175 (34%) received no treatment supervision, and 32 (6%) experienced treatment interruption. Compared to 2017–2019, reported pulmonary TB declined by 36.8% during the first-level response to COVID-19, 23.5% during the second-level response, 14% during the third-level response in 2020, and 4.3% in 2021.ConclusionThe COVID-19 response reduced the number of people who sought and received diagnosis, treatment, and care for TB in Tianjin, China. Integrative programs to ensure access and continuity of TB services should be considered and dual testing for SARS-CoV-2 and M. tuberculosis may facilitate finding cases.

Solar radiation drives many geophysical and biological processes in Antarctica, such as sea ice melting, ice sheet mass balance, and photosynthetic processes of phytoplankton in the polar marine environment. Although reanalysis and satellite products can provide important insight into the global scale of solar radiation in a seamless way, the ground-based radiation in the polar region remains poorly understood due to the harsh Antarctic environment. The present study attempted to evaluate the estimation performance of empirical models and machine learning models, and use the optimal model to establish a 35-year daily global solar radiation (DGSR) dataset at the Great Wall Station, Antarctica using meteorological observation data during 1986–2020. In addition, it then compared against the DGSR derived from ERA5, CRA40 reanalysis, and ICDR (AVHRR) satellite products. For the DGSR historical estimation performance, the machine learning method outperforms the empirical formula method overall. Among them, the Mutli2 model (hindcast test R2, RMSE, and MAE are 0.911, 1.917 MJ/m2, and 1.237 MJ/m2, respectively) for the empirical formula model and XGBoost model (hindcast test R2, RMSE, and MAE are 0.938, 1.617 MJ/m2, and 1.030 MJ/m2, respectively) for the machine learning model were found with the highest accuracy. For the austral summer half-year, the estimated DGSR agrees very well with the observed DGSR, with a mean bias of only −0.47 MJ/m2. However, other monthly DGSR products differ significantly from observations, with mean bias of 1.05 MJ/m2, 3.27 MJ/m2, and 6.90 MJ/m2 for ICDR (AVHRR) satellite, ERA5, and CRA40 reanalysis products, respectively. In addition, the DGSR of the Great Wall Station, Antarctica followed a statistically significant increasing trend at a rate of 0.14 MJ/m2/decade over the past 35 years. To our best knowledge, this study presents the first reconstruction of the Antarctica Great Wall Station DGSR spanning 1986–2020, which will contribute to the research of surface radiation balance in Antarctic Peninsula.

Summary: During the early phase of primary humoral responses, activated B cells can differentiate into different types of effector cells, dependent on B cell receptor affinity for antigen. However, the pivotal transcription factors governing these processes remain to be elucidated. Here, we show that transcription factor Bach2 protein in activated B cells is transiently induced by affinity-related signals and mechanistic target of rapamycin complex 1 (mTORC1)-dependent translation to restrain their expansion and differentiation into plasma cells while promoting memory and germinal center (GC) B cell fates. Affinity-related signals also downregulate Bach2 mRNA expression in activated B cells and their descendant memory B cells. Sustained and higher concentrations of Bach2 antagonize the GC fate. Repression of Bach2 in memory B cells predisposes their cell-fate choices upon memory recall. Our study reveals that differential dynamics of Bach2 protein and transcripts in activated B cells control their cell-fate outcomes and imprint the fates of their descendant effector cells.

α-Synuclein aggregation under pathological conditions is one of the causes of related neurodegenerative diseases. PROTACs (proteolysis targeting chimeras) are bifunctional small molecules that induce a post-translational erasure of proteins via the ubiquitination of target proteins by E3 ubiquitin ligase and subsequent proteasomal degradation. However, few research studies have been conducted for targeted protein degradation of α-synuclein aggregates. In this article, we have designed and synthesized a series of small-molecule degraders 1–9 based on a known α-synuclein aggregation inhibitor sery384. In silico docking studies of sery384 with α-synuclein aggregates were accomplished to ensure that the compounds bound to α-synuclein aggregates specifically. The protein level of α-synuclein aggregates was determined to evaluate the degradation efficiency of PROTAC molecules on α-synuclein aggregates in vitro. The results show that compound 5 had the most significant degradation effect, with DC50 of 5.049 μM, and could induce the degradation of α-synuclein aggregates in a time- and dose-dependent manner in vitro. Furthermore, compound 5 could inhibit the elevation of the ROS level caused by overexpression and aggregation of α-synuclein and protect H293T cells from α-synuclein toxicity. Conclusively, our results provide a new class of small-molecule degraders and an experimental basis for the treatment of α-synuclein related neurodegenerative diseases.

The cryopreservation of red blood cells (RBCs) holds great potential for ensuring timely blood transfusions and maintaining an adequate RBC inventory. The conventional cryoprotectants (CPAs) have a lot of limitations, and there is an obvious need for novel, efficient, and biocompatible CPAs. Here, it is shown for the first time that the addition of dimethylglycine (DMG) improved the thawed RBC recovery from 11.55 ± 1.40% to 72.15 ± 1.22%. We found that DMG could reduce the mechanical damage by inhibiting ice formation and recrystallization during cryopreservation. DMG can also scavenge reactive oxygen species (ROS) and maintain endogenous antioxidant enzyme activities to decrease oxidative damage during cryopreservation. Furthermore, the properties of thawed RBCs were found to be similar to the fresh RBCs in the control. Finally, the technique for order performance by similarity to ideal solution (TOPSIS) was used to compare the performance of glycerol (Gly), hydroxyethyl starch (HES), and DMG in cryopreservation, and DMG exhibited the best efficiency. This work confirms the use of DMG as a novel CPA for cryopreservation of RBCs and may promote clinical transfusion therapy.

Cell senescence is a crucial process in cell fate determination and is involved in an extensive array of aging-associated diseases. General perceptions and experimental evidence point out that the decline of physical function as well as aging-associated diseases are often initiated by cell senescence and organ ageing. Therefore, regulation of cell senescence process can be a promising way to handle aging-associated diseases such as osteoporosis. The circadian clock regulates a wide range of cellular and physiological activities, and many age-linked degenerative disorders are associated with the dysregulation of clock genes. BMAL1 is a core circadian transcription factor and governs downstream genes by binding to the E-box elements in their promoters. Compelling evidence has proposed the role of BMAL1 in cellular senescence and aging-associated diseases. In this review, we summarize the linkage between BMAL1 and factors of cell senescence including oxidative stress, metabolism, and the genotoxic stress response. Dysregulated and dampened BMAL1 may serve as a potential therapeutic target against aging- associated diseases.

The gut microbial community is closely related to mastitis, but studies regarding the influences of mastitis on gut microbiota in buffalo remain scarce. Herein, we characterized the differences in gut bacterial and fungal communities between mastitis-affected and healthy buffalos. Interestingly, although mastitis had no effect on gut bacterial and fungal diversities in the buffalos, some bacterial and fungal taxa were significantly altered. Bacterial and fungal taxonomic analysis showed that the preponderant bacterial phyla (Firmicutes and Bacteroidetes) and fungal phyla (Ascomycota and Basidiomycota) in buffalo were the same regardless of health status. At the level of genus, the changes in some gut bacterial and fungal abundances between both groups were gradually observed. Compared with healthy buffalos, the proportions of 3 bacterial genera (uncultured_bacterium_f_Muribaculaceae, Eubacterium_nodatum_group, and Lachnoclostridium_10) and 1 fungal genus (Pichia) in the mastitis-affected buffalo were significantly increased, whereas 4 bacterial genera (Ruminococcus_2, Candidatus_Stoquefichus, Turicibacter, and Cellulosilyticum) and 4 fungal genera (Cladosporium, Thermothelomyces, Ganoderma and Aspergillus) were significantly decreased. Taken together, this research revealed that there was significant difference in the compositions of the gut microbial community between the healthy and mastitis-affected buffalos. To our knowledge, this is the first insight into the characteristics of the gut microbiota in buffalos with mastitis, which is beneficial to understand the gut microbial information of buffalo in different health states and elucidate the pathogenesis of mastitis from the gut microbial perspective.

LEAFY COTYLEDON1 (LEC1) is the central regulator of seed development in Arabidopsis, while its function in monocots is largely elusive. We generated Oslec1 mutants using CRISPR/Cas9 technology. Oslec1 mutant seeds lost desiccation tolerance and triggered embryo greening at the early development stage. Transcriptome analysis demonstrated that Oslec1 mutation altered diverse hormonal pathways and stress response in seed maturation, and promoted a series of photosynthesis-related genes. Further, genome-wide identification of OsLEC1-binding sites demonstrated that OsLEC1 bound to genes involved in photosynthesis, photomorphogenesis, as well as abscisic acid (ABA) and gibberellin (GA) pathways, involved in seed maturation. We illustrated an OsLEC1-regulating gene network during seed development, including the interconnection between photosynthesis and ABA/GA biosynthesis/signaling. Our findings suggested that OsLEC1 acts as not only a central regulator of seed maturation but also an inhibitor of embryo greening during rice seed development. This study would provide new understanding for the OsLEC1 regulatory mechanisms on photosynthesis in the monocot seed development.

Abstract Objective PCOS is a heterogeneous endocrine disorder with both reproductive and metabolic abnormalities. At present, PCOS has been confirmed to have a certain genetic background. Compared with healthy women, the vast majority of PCOS patients have hyperandrogenemia, and this excessive androgen exposure during pregnancy may affect the development of female fetuses. The aim of the current study was to investigate the effect of adiponectin intervention during early pregnancy of obese mice with PCOS on the metabolic phenotype of adult female offspring. Methods After the PCOS model was established, C57BL/6J mice were divided into maternal-control, maternal-PCOS, and maternal-PCOS + APN groups. DHEA-induced PCOS mice were supplemented with adiponectin (10 mg/kg/day) in the early pregnancy in order to eliminate adverse hormone exposure and then traced for endocrine indicators in their adult female offspring, which were observed for metabolism syndrome or endocrine disturbance and exhibited the main effects of APN. To further explore the underlying mechanism, the relative expressions of phosphorylated AMPK, PI3K, and Akt were detected in the ovaries of offspring mice. Biochar’s underlying biochemical and physiological mechanisms in reducing irrigation and salinity stress are elusive. This paper investigates the effects of two types of biochar (wood biochar and poultry biochar) on the growth and physiology of tomato seedlings exposed to the combined effects of drought and salinity stress. Two types of biochar, wood biochar (WB) and poultry biochar (PB), were added to the soil separately, with three salinity gradients of 0, 100, and 200 mmol/L and two water supply conditions of full irrigation (FI) and deficit irrigation (DI). Results showed that biochar addition effectively improved the root water potential and osmotic potential of tomato plant under drought and salinity stress. Biochar application also mitigated leaf relative water content by 9.86% and 24.37% under drought and salinity stress, respectively. Furthermore, biochar application decreased abscisic acid concentrations in xylem sap under drought and salinity stress. Biochar altered the soil structure and increased field water holding capacity, indirectly increasing the soil water supply. While water use efficiency did not increase significantly after biochar application, a synergistic increase in seedling growth and water consumption occurred. In conclusion, biochar addition shows promise for promoting seedling growth to help mitigate the adverse impacts of drought and salinity stress on plant growth and physiology.

Lumpy skin disease virus (LSDV) is capable of causing transboundary diseases characterized by fever, nodules on the skin, mucous membranes, and inner organs. The disease may cause emaciation with the enlargement of lymph nodes and sometimes death. It has had endemic importance in various parts of Asia in recent years, causing substantial economic losses to the cattle industry. The current study reported a suspected LSDV infection (based on signs and symptoms) from a mixed farm of yak and cattle in Sichuan Province, China. The clinical samples were found positive for LSDV using qPCR and ELISA, while LSDV DNA was detected in Culex tritaeniorhynchus Giles. The complete genome sequence of China/LSDV/SiC/2021 was determined by Next-generation sequencing. It was found that China/LSDV/SiC/2021 is highly homologous to the novel vaccine-related recombinant LSDV currently emerging in China and countries surrounding China. Phylogenetic tree analysis revealed that the novel vaccine-associated recombinant LSDV formed a unique dendrograms topology between field and vaccine-associated strains. China/LSDV/SiC/2021 was found to be a novel recombinant strain, with at least 18 recombination events via field viruses identified in the genome sequence. These results suggest that recombinant LSDV can cause high mortality in yaks, and its transmission might be due to the Culex tritaeniorhynchus Giles, which acts as a mechanical vector.
