Abstract Computational methods for predicting protein function are of great significance in understanding biological mechanisms and treating complex diseases. However, existing computational approaches of protein function prediction lack interpretability, making it difficult to understand the relations between protein structures and functions. In this study, we propose a deep learning-based solution, named DPFunc, for accurate protein function prediction with domain-guided structure information. DPFunc can detect significant regions in protein structures and accurately predict corresponding functions under the guidance of domain information. It outperforms current state-of-the-art methods and achieves a significant improvement over existing structure-based methods. Detailed analyses demonstrate that the guidance of domain information contributes to DPFunc for protein function prediction, enabling our method to detect key residues or regions in protein structures, which are closely related to their functions. In summary, DPFunc serves as an effective tool for large-scale protein function prediction, which pushes the border of protein understanding in biological systems.

Abstract Background We aim to explore the impact of excessive glutathione (GSH) intake on chemotherapy sensitivity in breast cancer. Methods Clinicopathological data were collected from 460 breast cancer patients who underwent adjuvant chemotherapy from January 2016 to December 2019 from Zhengzhou University People’s Hospital. The clinicopathological characteristics following GSH treatment were collected and compared with those in Non-GSH group after 1:2 propensity score matching (PSM). Intracellular GSH levels and the expression of antioxidant enzymes (NRF2, GPX4 and SOD1) were evaluated in tumor tissues in 51 patients receiving neoadjuvant chemotherapy. Results The recurrence rate after adjuvant chemotherapy was significantly higher in the GSH group (n = 28, 31.8%) than that in the Non-GSH group (n = 39, 22.2%; P = 0.010). Abstract Drought-induced osmotic stress severely affects the growth and yield of maize. However, the mechanisms underlying the different responses of young and old maize leaves to osmotic stress remain unclear. To gain a systematic understanding of age-related stress responses, we compared osmotic-stress-induced changes in maize leaves of different ages using multi-omics approaches. After short-term osmotic stress, old leaves suffered more severe water deficits than young leaves. The adjustments of transcriptomes, proteomes, and hormones in response to osmotic stress were more dynamic in old leaves. Metabolic activities, stress signaling pathways, and hormones (especially abscisic acid) responded to osmotic stress in an age-dependent manner. We identified multiple functional clusters of genes and proteins with potential roles in stress adaptation. Old leaves significantly accumulated stress proteins such as dehydrin, aquaporin, and chaperones to cope with osmotic stress, accompanied by senescence-like cellular events, whereas young leaves exhibited an effective water conservation strategy mainly by hydrolyzing transitory starch and increasing proline production. The stress responses of individual leaves are primarily determined by their intracellular water status, resulting in differential transcriptomes, proteomes, and hormones. This study extends our understanding of the mechanisms underlying plant responses to osmotic stress.

Abstract Long non-coding RNAs (lncRNAs) involved in metabolism are recognized as significant factors in breast cancer (BC) progression. We constructed a novel prognostic signature for BC using metabolism-related lncRNAs and investigated their underlying mechanisms. The training and validation cohorts were established from BC patients acquired from two public sources: The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO). The prognostic signature of metabolism-related lncRNAs was constructed using the least absolute shrinkage and selection operator (LASSO) cox regression analysis. We developed and validated a new prognostic risk model for BC using the signature of metabolism-related lncRNAs (SIRLNT, SIAH2-AS1, MIR205HG, USP30-AS1, MIR200CHG, TFAP2A-AS1, AP005131.2, AL031316.1, C6orf99). The risk score obtained from this signature was proven to be an independent prognostic factor for BC patients, resulting in a poor overall survival (OS) for individuals in the high-risk group. The area under the curve (AUC) for OS at three and five years were 0.67 and 0.65 in the TCGA cohort, and 0.697 and 0.68 in the GEO validation cohort, respectively. The prognostic signature demonstrated a robust association with the immunological state of BC patients. Conventional chemotherapeutics, such as docetaxel and paclitaxel, showed greater efficacy in BC patients classified as high-risk. A nomogram with a c-index of 0.764 was developed to forecast the survival time of BC patients, considering their risk score and age. The silencing of C6orf99 markedly decreased the proliferation, migration, and invasion capacities in MCF-7 cells. Our study identified a signature of metabolism-related lncRNAs that predicts outcomes in BC patients and could assist in tailoring personalized prevention and treatment plans.

Abstract As the latest and most anticipated method of tumor immunotherapy, CAR-NK therapy has received increasing attention in recent years, and its safety and high efficiency have irreplaceable advantages over CAR-T. Current research focuses on the application of CAR-NK in hematological tumors, while there are fewer studies on solid tumor. This article reviews the process of constructing CAR-NK, the effects of hypoxia and metabolic factors, NK cell surface receptors, cytokines, and exosomes on the efficacy of CAR-NK in solid tumor, and the role of CAR-NK in various solid tumor. The mechanism of action and the research status of the potential of CAR-NK in the treatment of solid tumor in clinical practice, and put forward the advantages, limitations and future problems of CAR-NK in the treatment of solid tumor.

A pre-metastatic niche (PMN) is a protective microenvironment that facilitates the colonization of disseminating tumor cells in future metastatic organs. Extracellular vesicles (EVs) play a role in intercellular communication by delivering cargoes, such as noncoding RNAs (ncRNAs). The pivotal role of extracellular vesicle-derived noncoding RNAs (EV-ncRNAs) in the PMN has attracted increasing attention. In this review, we summarized the effects of EV-ncRNAs on the PMN in terms of immunosuppression, vascular permeability and angiogenesis, inflammation, metabolic reprogramming, and fibroblast alterations. In particular, we provided a comprehensive overview of the effects of EV-ncRNAs on the PMN in different cancers. Finally, we discussed the promising clinical applications of EV-ncRNAs, including their potential as diagnostic and prognostic markers and therapeutic targets.

Abstract Tumor immunotherapy has transformed neoplastic disease management, yet low response rates and immune complications persist as major challenges. Extracellular vesicles including exosomes have emerged as therapeutic agents actively involved in a diverse range of pathological conditions. Mounting evidence suggests that alterations in the quantity and composition of extracellular vesicles (EVs) contribute to the remodeling of the immune-suppressive tumor microenvironment (TME), thereby influencing the efficacy of immunotherapy. This revelation has sparked clinical interest in utilizing EVs for immune sensitization. In this perspective article, we present a comprehensive overview of the origins, generation, and interplay among various components of EVs within the TME. Furthermore, we discuss the pivotal role of EVs in reshaping the TME during tumorigenesis and their specific cargo, such as PD-1 and non-coding RNA, which influence the phenotypes of critical immune cells within the TME. Additionally, we summarize the applications of EVs in different anti-tumor therapies, the latest advancements in engineering EVs for cancer immunotherapy, and the challenges encountered in clinical translation. In light of these findings, we advocate for a broader understanding of the impact of EVs on the TME, as this will unveil overlooked therapeutic vulnerabilities and potentially enhance the efficacy of existing cancer immunotherapies.

Abstract Cancer immunotherapy has transformed traditional treatments, with immune checkpoint blockade being particularly prominent. However, immunotherapy has minimal benefit for patients in most types of cancer and is largely ineffective in some cancers (such as pancreatic cancer and glioma). A synergistic anti-tumor response may be produced through the combined application with traditional tumor treatment methods. Radiotherapy (RT) not only kills tumor cells but also triggers the pro-inflammatory molecules’ release and immune cell infiltration, which remodel the tumor microenvironment (TME). Therefore, the combination of RT and immunotherapy is expected to achieve improved efficacy. In this review, we summarize the effects of RT on cellular components of the TME, including T cell receptor repertoires, different T cell subsets, metabolism, tumor-associated macrophages and other myeloid cells (dendritic cells, myeloid-derived suppressor cells, neutrophils and eosinophils). Meanwhile, non-cellular components such as lactate and extracellular vesicles are also elaborated. In addition, we discuss the impact of different RT modalities on tumor immunity and issues related to the clinical practice of combination therapy.

Abstract The prestressed and steel-reinforced concrete slab (PSRCS) is an innovative composite structural member offering high load capacity and stiffness and exceptional anti-crack performance, making it a leading trend in composite structures. This paper presents the derived calculation formulas for bearing capacity, section stiffness, mid-span deflection of PSRCS. Additionally, a numerical analysis of PSRCS is conducted using ABAQUS software, with several models created to systematically investigate bearing capacity, section stiffness, anti-crack performance, and failure mode. Concurrently, PSRCS member parameters are analyzed for optimal design, and the results of finite element (FE) calculations are compared with theoretical formula calculations. The results demonstrate that PSRCS exhibits superior load capacity, section stiffness, and anti-crack performance comparing to conventional slabs. The parametric analysis offers optimal design for each parameter and presents the corresponding recommended span-to-depth ratios for various spans in PSRCS applications.

Summary: Immune escape is identified as one of the reasons for the poor prognosis of colorectal cancer (CRC) patients. Circular RNAs are considered to promote tumor progression by mediating tumor immune escape. We discovered that higher expression of circYAP1 was associated with a worse prognosis of CRC patients. Functional experiments in vitro and in vivo showed that circYAP1 upregulation inhibited the cytotoxicity of CD8+ T cells by upregulating programmed death ligand-1 (PD-L1). Mechanistically, we found that circYAP1 directly binds to the YAP1 protein to prevent its phosphorylation, enhancing proportion of YAP1 protein in the nucleus, and that YAP1 interacts with TCF4 to target the PD-L1 promoter and initiate PD-L1 transcription in CRC cells. Taken together, circYAP1 promotes CRC immune escape and tumor progression by activating the YAP1/TCF4-PD-L1 axis and may provide a new strategy for combination immunotherapy of CRC patients.

Grating-assisted contra-directional couplers (CDCs) as 4-port wavelength-selective devices hold great potential as add-drop filters, multiplexers, routers, etc, for various integrated-optics applications. However, these devices have inherent low coupling strengths due to the coupled modes confined in different separate waveguides, preventing from realizing broad operation bands. Here, we propose silicon photonic CDCs assisted by novel asymmetric photonic crystals (APhCs) on multimode waveguides with elliptical nanoholes. Such APhC-assisted CDCs, compared with the conventional grating-assisted ones, can achieve significantly larger coupling strengths, thus offering much wider operation bands. We investigate the impact of changes in various structural parameters on the spectral response of an APhC-assisted CDC, and then discuss the design and optimization strategy of the device. The possibility of using other nanohole shapes of the APhC is also explored. In the experiments, a series of broadband and flat-top filters based on APhC-assisted CDCs on silicon-on-insulator (SOI) were fabricated and measured, and the influence of variations in the filter parameters on the response was validated. Notably, a 3 dB bandwidth of $\sim$48.8 nm was demonstrated on an APhC-assisted CDC with a coupling length of 22.3 um, which is the largest bandwidth ever reported for unchirped, SOI-based CDC filters, to the best of our knowledge.

Motile cilia are miniature, hair‐like organelles whose beating moves fluid in various organisms and the human body. Motile cilia are also able to sense many mechanical or fluidic cues. Inspired by the motile cilia in nature, a type of artificial cilia that can both actuate and sense is investigated. These artificial cilia can operate their sensing and actuation individually to sense the flow speed and collaboratively transport objects. More importantly, they can also sense the viscosity of the fluid and the distance from the wall by beating and feeling their own motion. In the latter function, two different sensing modes (modes 1 and 2) are proposed for low and high Reynolds numbers. They are investigated through both computational fluid dynamics and experiments. It is found that mode 1 can sense a wall distance up to 3 times the body length of the cilium, while mode 2 can sense up to 2 times. Such sensing of distance can be further converted into the ability to compute the morphology of the boundary inside the fluid. This work can be useful for small‐scale soft robots that work inside fluid and other flow control applications.

PurposeIn recent years, a number of clinical trials have shown that programmed death 1 (PD-1) inhibitors offer significant survival benefits in patients with esophageal squamous cell carcinoma (ESCC). We conducted a meta-analysis to explore the antitumour efficacy of PD-1 inhibitor-based therapy in specific subgroups of patient with advanced ESCC.MethodsWe searched for eligible studies from the PubMed, Embase, Web of Science, Cochrane Library databases and conference abstracts. The indicators related to survival outcomes were extracted. The pooled hazard ratios (HRs) for overall survival (OS), progression-free survival (PFS) and duration of response (DOR) and the pooled odds ratio (OR) for objective response rate (ORR) were calculated to evaluate the efficacy of PD-1 inhibitor-based therapy in ESCC. Data regarding treatment lines, treatment regimens, programmed death ligand 1 (PD-L1) status, baseline demographic and disease characteristics were extracted. Subgroup analyses were conducted in specific populations of ESCC patients. The Cochrane risk of bias tool and sensitivity analysis were used to assess the quality of the meta-analysis.ResultsEleven phase 3 randomized controlled trials (RCTs) involving 6267 patients with ESCC were included in this meta-analysis. Compared with standard chemotherapy, PD-1 inhibitor-based therapy provided benefits in terms of OS, PFS, ORR, and DOR in all populations, the first-line treatment group, the second-line treatment group, the immunotherapy group, and the immunochemotherapy group. Although a limited PFS benefit was observed in second-line treatments and immunotherapy alone, PD-1 inhibitor-based therapy still reduced the risk of disease progression or death. Patients with high PD-L1 expression had a better OS benefit than those with low PD-L1 expression. The HR for OS favoured PD-1 inhibitor-based therapy over standard chemotherapy for all prespecified clinical subgroups.ConclusionsCompared with standard chemotherapy, PD-1 inhibitor-based therapy exhibited clinically meaningful benefits in patients with ESCC. Survival benefits were better in patients with high PD-L1 expression than in those with low PD-L1 expression, suggesting that the PD-L1 expression level can be used as a predictor of survival benefit from PD-1 inhibitor therapy. PD-1 inhibitor-based therapy provided a consistent benefit in reducing the risk of death according to prespecified subgroup analyses of clinical characteristics.

Breast cancer (BRCA) remains the most prevalent cancer worldwide and the tumor microenvironment (TME) has been discovered to exert a wide influence on the overall survival and therapeutic response. Numerous lines of evidence reported that the effects of immunotherapy of BRCA were manipulated by TME. Immunogenic cell death (ICD) is a form of regulated cell death (RCD) that is capable of fueling adaptive immune responses and aberrant expression of ICD-related genes (ICDRGs) can govern the TME system by emitting danger signals or damage-associated molecular patterns (DAMPs). In the current study, we obtained 34 key ICDRGs in BRCA. Subsequently, using the transcriptome data of BRCA from the TCGA database, we constructed a risk signature based on 6 vital ICDRGs, which had a good performance in predicting the overall survival of BRCA patients. We also examined the efficacy of our risk signature in the validation dataset (GSE20711) in the GEO database and it performed excellently. According to the risk model, patients with BRCA were divided into high-risk and low-risk groups. Also, the unique immune characteristics and TME between the two subgroups and 10 promising small molecule drugs targeting BRCA patients with different ICDRGs risk have been investigated. The low-risk group had good immunity indicated by T cell infiltration and high immune checkpoint expression. Moreover, the BRCA samples could be divided into three immune subtypes according to immune response severity (ISA, ISB, and ISC). ISA and ISB predominated in the low-risk group and patients in the low-risk group exhibited a more vigorous immune response. In conclusion, we developed an ICDRGs-based risk signature that can predict the prognosis of BRCA patients and offer a novel therapeutic strategy for immunotherapy, which would be of great significance in the BRCA clinical setting.

Summary: The chromatin loop plays a critical role in the study of gene expression and disease. Supervised learning-based algorithms to predict the chromatin loops require large priori information to satisfy the model construction, while the prediction sensitivity of unsupervised learning-based algorithms is still unsatisfactory. Therefore, we propose an unsupervised algorithm, Ecomap-loop. It takes advantage of extrusion complex-associated patterns, including CTCF, RAD21, and SMC enrichments, as well as the orientation distribution of CTCF motif of loops to build feature matrices; then the eigen decomposition model is employed to obtain the cell type-specific loops. We compare the performance of Ecomap-loop with the state-of-the-art unsupervised algorithm using Hi-C, ChIA-PET, expression quantitative trait locus (eQTL), and CRISPR interference (CRISPRi) screen data; the results show that Ecomap-loop achieves the best in four cell types. In addition, the functional analysis reveals the ability of Ecomap-loop to predict active functionality-related and cell type-specific loops.

To address the issue of serious torque pulsation and optimize the output characteristics of multi-acting cam ring motors at low speed, a sensitivity analysis was conducted on the parameters of the triangular groove at the valve plate. Firstly, a mathematical model of the flow area between the rotor hole and the valve plate hole was established. Then, a numerical simulation model was built to study the motor output characteristics. Finally, the coupling effect of the triangular groove parameters on the motor torque pulsation rate was analyzed based on the response surface methodology. The results show that the motor torque pulsation rate can be reduced by 55% when adjusting depth angle θ1, width angle θ2, and length l. The influence order of design parameters on the pulsation rate is θ1>l>θ2; among all parameter combinations, the coupling of the triangular groove between the depth angle and the length has the most significant effect on the pulsation rate.

PurposeThis study determined the efficacy of low-dose gemcitabine combined with programmed death-1 (PD-1) inhibitors for treating multiple malignancies, providing a cost-effective and safe treatment option.Study DesignThis study included 61 patients with advanced solid tumors treated with low-dose gemcitabine combined with PD-1 inhibitors at the Henan Cancer Hospital between January 2018 and February 2022. We retrospectively reviewed medical records to evaluate several clinical factors, including progression-free survival (PFS), overall survival (OS), adverse effects (AEs), and objective response to treatment.ResultsSixty-one patients received treatment with low-dose gemcitabine combined with PD-1 inhibitors. The objective response rate (ORR) was 29.5% and the disease control rate (DCR) was 62.3%. The median PFS was 4.3 months (95% confidence interval, 2.3 to 6.3 months) and the median OS was 15.0 months (95% confidence interval, 8.8 to 21.2 months). Hematological toxicity, mainly leukopenia or thrombocytopenia, was the most common AE, with any-grade and grade 3/4 hematological toxicity reported in 60.7 and 13.1% of patients, respectively.ConclusionsLow-dose gemcitabine combined with PD-1 inhibitors may offer a novel treatment option for patients with advanced malignancies.

The interplay between long non-coding RNAs (lncRNAs) and the Notch pathway involves a variety of malignancies. However, Notch-derived lncRNAs and their latent clinical significance remain elusive in colorectal cancer (CRC). In this study, we introduced a framework that could screen Notch-derived lncRNAs (named “NLncer”) and ultimately identified 24 NLncers. To further explore the clinical significance of these NLncers, we performed LASSO and Cox regression in TCGA-CRC cohort (n = 584) and then retained six lncRNAs tightly associated with prognosis. The final model (termed “NLncS”) was subsequently tested in GSE38832 (n = 122), GSE39582 (n = 573), and an in-house clinical cohort (n = 115). Ultimately, our NLncS model could serve as an independent risk factor and afford a robust performance for assessing the prognosis of CRC patients. Additionally, patients with high NLncS risk scores were characterized by upregulation of immune pathways, strong immunogenicity, abundant CD8 + T-cell infiltration, and potentially higher response rates to CTLA4 blockers, which turned out to be suitable for immunotherapy. Aiming at globally observing the characteristics of high-risk patients, somatic mutation and methylation modification analysis provide us with evidence at the genomic and transcriptomic levels. To facilitate the clinical transformability, we mined deeply into the sensitive compounds targeting high-risk individuals and identified dasatinib as a candidate agent for patients with a high Notch risk score. In conclusion, our NLncS model is a promising biomarker for optimizing the clinical management of CRC patients.

The study aimed to reveal altitudinal distribution patterns of phyllosphere microbial communities and silage fermentation of Kobresia pygmaea along the elevation gradient on the Tibetan Plateau. The K. pygmaea was individually collected from 2,500, 3,000, 4,000, 4,500, and 5,000 m above sea level (a.s.l.) on the Tibetan Plateau and ensiled for 60 days, respectively. The phyllosphere bacterial diversity increased while fungal diversity decreased along the elevation gradient, and bacterial and fungal richness showed a unimodal distribution with peak abundance at 4,000 and 3,000 m a.s.l., respectively. After 60 days of ensiling, the bacterial and fungal community composition changed but did not exhibit clear altitudinal distribution patterns. All K. pygmaea underwent a weak fermentation indicated by pH above 5.0 and low ratio of lactic/acetic acid (LA/AA). The S5000 and S3000 showed the highest and lowest pH, respectively. Although Lactobacillus dominated S4000 after 60 days of ensiling, S4000 still exhibited poor fermentation quality as well as silages from the other four regions. The higher ammonia N concentrations in S3000 and S4000 than the other silages were consistent with the detectable butyric acid in S3000 and S4000. The silage fermentation of K. pygmaea collected from five regions exhibited poor fermentation quality, thereby inoculating lactic acid bacteria to K. pygmaea before ensiling is highly recommended to improve fermentation quality on the Tibetan Plateau.

This study introduces a wearable soft knee exoskeleton that aids active knee motions during walking. It is mainly driven by vacuum-actuated rotary actuators. In this paper, the overall design of the exoskeleton is introduced. Moreover, the design of the vacuum-actuated rotary actuator is introduced, and the correspondence among the interior air pressure, rotation angle and output force of the actuator under a vacuum condition and during the transition from the vacuum to an equal atmospheric pressure condition were studied. Then, the corresponding relations among the pressure, angle and torque of the actuator were obtained to construct a knee torque model. Furthermore, we introduced in detail the control system of the exoskeleton, including a gait estimation model and knee torque model. The control system analysed knee angle information measured by IMUs and the air pressure of the actuators measured by air pressure sensors. Then, by the calculations performed by both the gait estimation model and knee torque model, the corresponding commands for the aerodynamic switch, pressures and air flow rates were determined. According to the commands, a micro air pump and valves provided quantitatively positive or negative pressure for the actuators to generate gait-consistent auxiliary torques, stretching torques and bending torques that were able to meet the needs of the knee during walking. Finally, a cardiopulmonary exercise test was used to quantitatively evaluate the exoskeleton. The results show that under the same load condition, the metabolic cost of walking is reduced by an average of 6.85% when the exoskeleton is worn.

Hydrogen sulfide (H2S), a gaseous signaling molecule, is associated with the development of various malignancies via modulating various cellular signaling cascades. Published research has established the fact that inhibition of endogenous H2S production or exposure of H2S donors is an effective approach against cancer progression. However, the effect of pharmacological inhibition of endogenous H2S-producing enzymes (cystathionine-γ-lyase (CSE), cystathionine-β-synthase (CBS), and 3-mercaptopyruvate sulfurtransferase (3-MPST)) on the growth of breast cancer (BC) remains unknown. In the present study, DL-propargylglycine (PAG, inhibitor of CSE), aminooxyacetic acid (AOAA, inhibitor of CBS), and L-aspartic acid (L-Asp, inhibitor of 3-MPST) were used to determine the role of endogenous H2S in the growth of BC by in vitro and in vivo experiments. An in silico study was also performed to confirm the results. Corresponding to each enzyme in separate groups, we treated BC cells (MCF-7 and MDA-MB-231) with 10 mM of PAG, AOAA, and L-Asp for 24 h. Findings reveal that the combined dose (PAG + AOAA + L-Asp) group showed exclusive inhibitory effects on BC cells’ viability, proliferation, migration, and invasion compared to the control group. Further, treated cells exhibited increased apoptosis and a reduced level of phospho (p)-extracellular signal-regulated protein kinases such as p-AKT, p-PI3K, and p-mTOR. Moreover, the combined group exhibited potent inhibitory effects on the growth of BC xenograft tumors in nude mice, without obvious toxicity. The molecular docking results were consistent with the wet lab experiments and enhanced the reliability of the drugs. In conclusion, our results demonstrate that the inhibition of endogenous H2S production can significantly inhibit the growth of human breast cancer cells via the AKT/PI3K/mTOR pathway and suggest that endogenous H2S may act as a promising therapeutic target in human BC cells. Our study also empowers the rationale to design novel H2S-based anti-tumor drugs to cure BC.

Solute Carrier Family 26 (SLC26) is a conserved anion transporter family with 10 members in human (SLC26A1-A11, A10 being a pseudogene). All SLC26 genes except for SLC26A5 (prestin) are versatile anion exchangers with notable ability to transport a variety of anions. SLC26A6 has the most extensive exchange functions in the SLC26 family and is widely expressed in various organs and tissues of mammals. SLC26A6 has some special properties that make it play a particularly important role in ion homeostasis and acid-base balance. In the past few years, the function of SLC26A6 in the diseases has received increasing attention. SLC26A6 not only participates in the development of intestinal and pancreatic diseases but also serves a significant role in mediating nephrolithiasis, fetal skeletal dysplasia and arrhythmia. This review aims to explore the role of SLC26A6 in physiology and pathophysiology of relative mammalian organs to guide in-depth studies about related diseases of human.

Intensive care unit-acquired weakness (ICU-AW), a common neuromuscular complication associated with patients in the ICU, is a type of skeletal muscle dysfunction that commonly occurs following sepsis, mobility restriction, hyperglycemia, and the use of glucocorticoids or neuromuscular blocking agents. ICU-AW can lead to delayed withdrawal of mechanical ventilation and extended hospitalization. Patients often have poor prognosis, limited mobility, and severely affected quality of life. Currently, its pathogenesis is uncertain, with unavailability of specific drugs or targeted therapies. ICU-AW has gained attention in recent years. This manuscript reviews the current research status of the epidemiology, pathogenesis, diagnosis, and treatment methods for ICU-AW and speculates the novel perspectives for future research.

Real-time recognition of walking-related activities is an important function that lower extremity assistive devices should possess. This article presents a real-time walking pattern recognition method for soft knee power assist wear. The recognition method employs the rotation angles of thighs and shanks as well as the knee joint angles collected by the inertial measurement units as input signals and adopts the rule-based classification algorithm to achieve the real-time recognition of three most common walking patterns, that is, level-ground walking, stair ascent, and stair descent. To evaluate the recognition performance, 18 subjects are recruited in the experiments. During the experiments, subjects wear the knee power assist wear and carry out a series of walking activities in an out-of-lab scenario. The results show that the average recognition accuracy of three walking patterns reaches 98.2%, and the average recognition delay of all transitions is slightly less than one step.

The tube-crushing dissipator is widely used in engineering, but it has the eccentricity problem. Therefore, a symmetric tube-crushing dissipator was proposed in this article, and quasistatic and dynamic tests were performed to compare mechanical properties of the tube-crushing dissipators and the symmetric tube-crushing dissipators. The results of the quasistatic tests show that the working force of the tube-crushing dissipators fluctuates around an average value which is about 20% smaller than the activation threshold after activation, while the working force fluctuates around an average value which is approximately equal to the activation threshold after activation. The results of the dynamic tests show that mean force at the crushing section of the tube-crushing dissipators and the symmetric tube-crushing dissipators increases with the increase of the impact velocity. Furthermore, the dynamic load-displacement curves are more volatile than those of the static tests. Therefore, dynamic tests which are more similar to the real working conditions of the dissipators are preferable over static tests. In addition, the metal tubes of the symmetric tube-crushing dissipators collapse vertically both in the quasistatic and dynamic tests; that is, the eccentricity problem of the tube-crushing dissipators is overcome by the symmetric tube-crushing dissipators.

A simple and reliable ultraviolet sensing method with high sensitivity is proposed. ZnO and ZnO composite graphene are successfully prepared by the hydrothermal method. The optical fiber sensor is fabricated by coating the single-mode-taper multimode-single-mode (STMS) with different shapes of ZnO. The effects of the sensitivity of ultraviolet sensors are further investigated. The results show that the sensor with ZnO nanosheets exhibits a higher sensitivity of 357.85 pm/nW·cm−2 for ultraviolet sensing ranging from 0 to 4 nW/cm2. The ultraviolet characteristic of STMS coated flake ZnO composite graphene has been demonstrated with a sensitivity of 427.76 pm/nW·cm−2. The combination of sensitive materials and optical fiber sensing technology provides a novel and convenient platform for ultraviolet detection technology.

In areas where solar energy is abundant, such as the Tibetan plateau, passive solar buildings are attracting more and more attention and becoming a popular form of rural building. However, it is often difficult to achieve the satisfactory indoor thermal environment in a local rural passive solar single-family house. In order to improve the indoor thermal environment of passive solar buildings through building design, a systematic study of rural single-family buildings in Tibet was conducted. The basic parameters were investigated on the outdoor thermal environment, space-arrangement, envelope structure, and the indoor thermal environment. The building model considering space-arrangement modes was developed based on the survey data in multi-space passive solar buildings. The general physical and mathematical analysis models of multi-space passive solar buildings were established based on the heat transfer theory. Furthermore, the effects of space-arrangement and exterior wall thermal resistance on indoor air temperature were analyzed by numerical simulation. Results show that the indoor air temperature of the passive solar building is influenced by space-arrangement and wall thermal resistance together. When the space-arrangement of the building model was changed from “north-south through type” (mode a) to “through and separation combination” (mode b) and “north-south separation” (mode c), the indoor air temperature of the living room increased from 8.8 °C to 10.6 °C and 11.6 °C, with increases of 20.5% and 31.8%, respectively. In addition, equally increasing the thermal resistance of exterior walls in different orientations has different effects on the indoor air temperature. In the space-arrangement mode c, comparing with the temperature increment of the living room and bedroom caused by increasing thermal resistance of the south wall and north wall, the temperature increment of the living room caused by increasing thermal resistance of the east/west wall increased by 151.7% and 32.7%, and that of the bedroom increased by 609.1% and 239.1% respectively. This study can provide a reference for the optimal design of passive solar buildings in solar energy abundant areas.

HPS (Honeycomb-like Protective Structure) is a newly proposed protective structure filled with sandy soil. In order to investigate the penetration resistance of the structure, numerical simulations based on SPH method had been carried out by using LS-DYNA, which are corresponding to the experiments. The calibrated model leads to reasonable predictions of the dynamic responses and damage modes of the HPS. More situations were carried out taking factors influencing the penetration into consideration, including point of impact, angle of impact, and projectile caliber. Penetration mode was established by analyzing the energy dissipation and investigating the mechanism from the phenomenological viewpoint. Simulation results show that the resisting forces and the torque that act on the long rod projectile would be greater than those acting on the short one when instability occurred. Besides, approximate 45° angle of impact was formed in the case of off-axis, which has a certain influence on the ballistic stability, resulting in more kinetic energy of projectile dissipating in HPS and less depth of penetration. The kinetic energy of projectile dissipated in sandy soil largely and the strip slightly, and the former was greater than the sum of the latter.

Low-speed streaks in wall-bounded turbulence are the dominant structures in the near-wall turbulent self-sustaining cycle. Existing studies have well characterized their spanwise spacing in the buffer layer and below. Recent studies suggested the existence of these small-scale structures in the higher layer where large-scale structures usually receive more attention. The present study is thus devoted to extending the understanding of the streak spacing to the log layer. An analysis is taken on two-dimensional (2D) wall-parallel velocity fields in a smooth-wall turbulent boundary layer with R e τ = 440∼2400, obtained via either 2D Particle Image Velocimetry (PIV) measurement taken here or public Direct Numerical Simulation (DNS). Morphological-based streak identification analysis yields a R e -independent log-normal distribution of the streak spacing till the upper bound of the log layer, based on which an empirical model is proposed to account for its wall-normal growth. The small-scale part of the spanwise spectra of the streamwise fluctuating velocity below y + = 100 is reasonably restored by a synthetic simulation that distributes elementary streak units based on the proposed empirical streak spacing model, which highlights the physical significance of streaks in shaping the small-scale part of the velocity spectra beyond the buffer layer.
