Abstract Background Melanin pigmentation in oocytes is a critical feature for both the esthetic and developmental aspects of oocytes, influencing their polarity and overall development. Despite substantial knowledge of melanogenesis in melanocytes and retinal pigment epithelium cells, the molecular mechanisms underlying oocyte melanogenesis remain largely unknown. Results Here, we compare the oocytes of wild-type, tyr −/− and mitf −/− Xenopus tropicalis and found that mitf −/− oocytes exhibit normal melanin deposition at the animal pole, whereas tyr −/− oocytes show no melanin deposition at this site. Transmission electron microscopy confirmed that melanogenesis in mitf −/− oocytes proceeds normally, similar to wild-type oocytes. Transcriptomic analysis revealed that mitf −/− oocytes still express melanogenesis-related genes, enabling them to complete melanogenesis. Additionally, in Xenopus tropicalis oocytes, the expression of the MiT subfamily factor tfe3 is relatively high, while tfeb, mitf, and tfec levels are extremely low. The expression pattern of tfe3 is similar to that of tyr and other melanogenesis-related genes. Thus, melanogenesis in Xenopus tropicalis oocytes is independent of Mitf and may be regulated by other MiT subfamily factors such as Tfe3, which control the expression of genes like tyr, dct, and tyrp1. Furthermore, transcriptomic data revealed that changes in the expression of genes related to mitochondrial cloud formation represent the most significant molecular changes during oocyte development. Conclusions Overall, these findings suggest that further elucidation of Tyr-dependent and Mitf-independent mechanisms of melanin deposition at the animal pole will enhance our understanding of melanogenesis and Oogenesis.

Crisp grass carp is a type of grass carp (Ctenopharyngodon idella) that has been fed faba beans. Flesh quality and expression of muscle quality-related genes in muscle of grass carp were evaluated. Compared with ordinary grass carp, 1901 differentially expressed genes (DEGs) were up-regulated and 1628 DEGs were down-regulated in crisp grass carp. The GO analysis results showed that the DEGs were enriched in ribosomes and proteins. Kyoto Encyclopaedia of genes and genomes (KEGG) pathway analysis results showed that amino acid metabolism was closely related to meat quality. COX3, ACADS, ACAT1, ECHS1, EHHADH, HADHAA, HADH, HMGCL, MCCC1 and MCCC2 were the top 10 hub genes most closely associated with other nodes and showed a strong positive correlation. Hardness was positively correlated with the COX3, ECHS1, ACADS, ACAT1, HADHA, HADH, HMGCL and MCCC1. These results have been demonstrated to play important roles in clarifying the molecular mechanism of muscle firmness increase in crisp grass carp.

BackgroundPelargonic acid vanillyl amide (PAVA), a stable synthetic analog of capsaicin, exhibits potential for therapeutic applications; however, it may present cytotoxic and pro-inflammatory risks. This study aims to investigate the injury effects of PAVA on a cocultured skin cell model in vitro.MethodsHuman keratinocytes and dermal fibroblasts were co-cultured and exposed to PAVA at concentrations ranging from 12.5 to 200 µM for durations of 5, 24, and 48 h. Cell proliferation was quantified using MTS assays. Morphological changes were observed through microscopy, reactive oxygen species (ROS) production was evaluated via fluorescence analysis, apoptosis was assessed using flow cytometry and Western blotting techniques, while inflammatory cytokines (IL-6, IL-8) were quantified by ELISA.ResultsThe proliferation of cells was significantly inhibited by PAVA in a dose- and time-dependent manner, with concentrations of 100 µM and above inducing substantiazl cytotoxicity. Morphological analysis revealed an increase in cell dispersion, irregular morphology, and apoptosis, particularly after prolonged exposure. Treatment with PAVA led to elevated levels of ROS, indicating the presence of oxidative stress. Apoptosis was initiated through both extrinsic pathways (NF-κB, Caspase-8) at an early stage and intrinsic pathways (Caspase-3/9, Bax) at a later period. Furthermore, PAVA markedly increased the secretion of IL-6 and IL-8, suggesting a robust pro-inflammatory response.Conclusion100 μM PAVA elicits pronounced cytotoxic, oxidative, and pro-inflammatory effects on cocultured skin cell model, particularly at higher concentrations and prolonged exposure durations. These findings underscore the necessity of exercising caution when employing PAVA for therapeutic purposes and highlight the imperative for further research to mitigate its adverse consequences as a riot control agent.

Abstract Background Global per capita meat consumption continues to rise, especially pork. Meat quality is influenced by the content of intramuscular fat (IMF) as a key factor. The longissimus dorsi muscle of Dahe pigs (DHM, IMF: 7.98% ± 1.96%) and Dahe black pigs (DHBM, IMF: 3.30% ± 0.64%) was studied to explore cellular heterogeneity and differentially expressed genes (DEGs) associated with IMF deposition using single-nucleus RNA sequencing (snRNA-seq). The lipid composition was then analyzed using non-targeted lipidomics. Results A total of seven cell subpopulations were identified, including myocytes, fibroblast/fibro/adipogenic progenitors (FAPs), satellite cells, endothelial cells, macrophages, pericytes, and adipocytes. Among them, FAPs and adipocytes were more focused because they could be associated with lipid deposition. 1623 DEGs in the FAPs subpopulation of DHBM were up-regulated compared with DHM, while 1535 were down-regulated. These DEGs enriched in the glycolysis/gluconeogenesis pathway. 109 DEGs were up-regulated and 806 were down-regulated in the adipocyte subpopulation of DHBM compared with DHM, which were mainly enriched in the PPAR signaling pathway and fatty acid (FA) biosynthesis. The expression level of PPARG, ABP4, LEP, and ACSL1 genes in DHM was higher than that in DHBM. Lipidomics reveals porcine lipid composition characteristics of muscle tissue. A total of 41 lipid classes and 2699 lipid species were identified in DHM and DHBM groups. The top ten relative peak areas of lipid classes in DHM and DHBM were triglyceride (TG), phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylserine (PS), diglyceride (DG), cardiolipin (CL), ceramides (Cer), Simple Glc series (Hex1Cer), sphingomyelin (phSM), and phosphatidylinositol (PI). The relative peak areas of 35 lipid species in DHM were lower than DHBM, and 28 lipid species that were higher. Abstract Cutaneous squamous carcinoma is the second most common epithelial malignancy, associated with significant morbidity, mortality, and economic burden. However, the mechanisms underlying cSCC remain poorly understood. In this study, we identified TGM3 as a novel cSCC tumor suppressor that acts via the PI3K-AKT axis. RT-qPCR, IHC and western blotting were employed to assess TGM3 levels. TGM3-overexpression/knockdown cSCC cell lines were utilized to detect TGM3’s impact on epithelial differentiation as well as tumor cell proliferation, migration, and invasion in vitro. Additionally, subcutaneous xenograft tumor models were employed to examine the effect of TGM3 knockdown on tumor growth in vivo. Finally, molecular and biochemical approaches were employed to gain insight into the tumor-suppressing mechanisms of TGM3. TGM3 expression was increased in well-differentiated cSCC tumors, whereas it was decreased in poor-differentiated cSCC tumors. Loss of TGM3 is associated with poor differentiation and a high recurrence rate in patients with cSCC. TGM3 exhibited tumor-suppressing activity by regulating cell proliferation, migration, and invasion both in vitro and in vivo. As a novel cSCC tumor differentiation marker, TGM3 expression was positively correlated with cell differentiation. In addition, our results demonstrated an interaction between TGM3 and KRT14 that aids in the degradation of KRT14. TGM3 deficiency disrupts keratinocytes differentiation, and ultimately leads to tumorigenesis. Furthermore, RNA-sequence analysis revealed that loss of TGM3 enhanced EMT via the PI3K-AKT signaling pathway. Deguelin, a PI3K-AKT inhibitor, blocked cSCC tumor growth induced by TGM3 knockdown in vivo. Taken together, TGM3 inhibits cSCC tumor growth via PI3K-AKT signaling, which could also serve as a tumor differentiation marker and a potential therapeutic target for cSCC. Proposed model depicted the mechanism by which TGM3 suppress cSCC development. TGM3 reduces the phosphorylation level of AKT and degrades KRT14. In the epithelial cell layer, TGM3 exhibits a characteristic pattern of increasing expression from bottom to top, while KRT14 and pAKT are the opposite. Loss of TGM3 leads to reduced degradation of KRT14 and activation of pAKT, disrupting keratinocyte differentiation, and eventually resulting in the occurrence of low-differentiated cSCC.

Ecosystem service connectivity (ESC) is the spatial and functional links among and within ecosystems that support unimpeded service flows, and that could play an important role in ecosystem stability enhancement and regional land planning. Understanding the relationships between landscape patterns and ESC is crucial to achieving certain sustainable development goals, but it has not yet received an adequate amount of attention. Here, we evaluated the changes and connectivity of five key types of ecosystem services from 2000 to 2020 and analyzed the correlations and spatial aggregations between the ESCs and landscape metrics in the wetlands of the Yellow River Delta, China. Various research methods, such as the InVEST model, spatial autocorrelation analysis, Spearman’s correlation, and self-organizing map, were applied. The results showed that water yield, water purification, and habitat quality showed high connectivity, but the overall ESC declined along with the restoration of the wetland area. Meanwhile, the High-High ESC cluster of water yield, water purification, and habitat quality had similar spatial distribution patterns, and both were dominated by tidal flats. Moreover, the ESC and landscape metrics showed significant correlations and spatial heterogeneity, and a potential connectivity between water yield and habitat quality was also found. These findings can assist decision-makers in developing effective ecosystem management strategies and provide a reference for future research on ecosystem service connectivity.

Nano-multilayered films offer a multilayer structure arrangement at several to tens of nanometers. They have hardness, high-temperature oxidation resistance, and low wear rates, making them popular in surface protection fields, such as engineering machinery, cutting tools and molds, and electronic instruments. This paper reviews recent research progress on nano-multilayered films, including their general types, preparation methods, microstructures, mechanical properties, and corrosion resistance. Furthermore, the mechanisms of the high hardness and toughness in nano-multilayered films are analyzed in detail. The future research directions for designing nano-multilayered films with improved strengthening and toughening properties are discussed.

Feeding faba beans to grass carp could crisp its muscle texture to avoid softening, the relationship between texture formation throughout the crisping process and the critical lipids regulating the fish quality has not yet been clarified. Herein, an 60-day nutritional trial and untargeted lipidomic analysis was used to study the changes of lipids in crisp grass carp dorsal muscle. A total of 1036 lipids were remarkably different between ordinary and crisp grass carp. The concentrations of the LPC, LPE, PG, Cer, Hex2Cer, SM, MG and MGMG were positively correlated with hardness and springiness, and the CL, TG, PMe, WE, dMePE and AcCa were negative correlation. High content of lipids involved in storage in ordinary grass carp, such as glycerophospholipids, polyunsaturated and saturated fatty acid content. In contrast, high content of membrane components in crisp grass carp, such as monounsaturated fatty acid, sphingolipid and glycerolipids content, and the distribution of PUFA in lipid molecules was related to lipid biosynthesis. This study might provide some insights into improved knowledge of the association between meat texture and lipid molecules in fish fed with faba bean.

The dynamic coefficients (dynamic stiffness coefficient and dynamic damping coefficient) of gas foil bearings (GFBs) are important for transient dynamics calculation and stability analysis of the GFBs-rotor system. Although the perturbation method can be used to calculate the dynamic coefficients, it can only consider a single whirl ratio. Moreover, the whirl ratio is assumed and not practical. Therefore, this paper proposes an identification method of dynamic coefficients of GFBs by simulated excitation. Firstly, based on single journal-single GFB model considering gas-structure interaction by coupled fields, harmonic excitations were applied to the rotor to obtain the shaft orbit. Next, frequency response function method and equivalence coefficient method were used to identify dynamic coefficients. The former method is suitable for the case where response and excitation are same frequency, namely linear problem. The latter method is fit for the situation in which the response has multiple frequency components, namely nonlinear problem. Finally, examples were carried out, and the results from different methods were compared and analyzed. This research work lays a theoretical foundation for the design of the GFBs-rotor system.

ABSTRACT In this study, a mixture of estradiol benzoate, progesterone, and testosterone propionate was injected into Diannan small-ear sows to induce estrus. The 16S rRNA technology was used to comparatively analyze the differences in fecal microbial composition and diversity between induced and spontaneous estrus in Diannan small-ear sows. The most abundant phylum in the sows in estrus were Firmicutes, Bacteroidetes, Actinobacteria, and Proteobacteria. There was a significant negative correlation between Firmicutes and Bacteroide and a significant positive correlation between Bacteroides and Bifidobacterium. The relative abundance of Stenotrophomonas, Neisseria, Anaerofustis, and Terrisporobacter in the sows during induced estrus was significantly higher than that during spontaneous estrus. Taken together, induced estrus affects the relative abundance of specific microbes in the feces of Diannan small-ear sows, but it does not affect the overall composition and diversity. These results provide fundamental knowledge about the gut microbiota of sows with induced estrus.

IntroductionPig growth is an important economic trait that involves the co-regulation of multiple genes and related signaling pathways. High-throughput sequencing has become a powerful technology for establishing the transcriptome profiles and can be used to screen genome-wide differentially expressed genes (DEGs). In order to elucidate the molecular mechanism underlying muscle growth, this study adopted RNA sequencing (RNA-seq) to identify and compare DEGs at the genetic level in the longissimus dorsi muscle (LDM) between two indigenous Chinese pig breeds (Diannan small ears [DSE] pig and Wujin pig [WJ]) and one introduced pig breed (Landrace pig [LP]).MethodsAnimals under study were from two Chinese indigenous pig breeds (DSE pig, n = 3; WJ pig, n = 3) and one introduced pig breed (LP, n = 3) were used for RNA sequencing (RNA-seq) to identify and compare the expression levels of DEGs in the LDM. Then, functional annotation, Gene Ontology (GO) enrichment analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis, and Protein–Protein Interaction (PPI) network analysis were performed on these DEGs. Then, functional annotation, Gene Ontology (GO) enrichment analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis, and Protein-Protein Interaction (PPI) network analysis were performed on these DEGs.ResultsThe results revealed that for the DSE, WJ, and LP libraries, more than 66, 65, and 71 million clean reads were generated by transcriptome sequencing, respectively. A total of 11,213 genes were identified in the LDM tissue of these pig breeds, of which 7,127 were co-expressed in the muscle tissue of the three samples. In total, 441 and 339 DEGs were identified between DSE vs. WJ and LP vs. DSE in the study, with 254, 193 up-regulated genes and 187, 193 down-regulated genes in DSE compared to WJ and LP. GO analysis and KEGG signaling pathway analysis showed that DEGs are significantly related to contractile fiber, sarcolemma, and dystrophin-associated glycoprotein complex, myofibril, sarcolemma, and myosin II complex, Glycolysis/Gluconeogenesis, Propanoate metabolism, and Pyruvate metabolism, etc. In combination with functional annotation of DEGs, key genes such as ENO3 and JUN were identified by PPI network analysis.DiscussionIn conclusion, the present study revealed key genes including DES, FLNC, PSMD1, PSMD6, PSME4, PSMB4, RPL11, RPL13A, ROS23, RPS29, MYH1, MYL9, MYL12B, TPM1, TPM4, ENO3, PGK1, PKM2, GPI, and the unannotated new gene ENSSSCG00000020769 and related signaling pathways that influence the difference in muscle growth and could provide a theoretical basis for improving pig muscle growth traits in the future.

The beta T-cell receptor (TRB) expressed by beta T cells is essential for foreign antigen recognition. The TRB locus contains a TRBV family that encodes three complementarity determining regions (CDRs). CDR1 is associated with antigen recognition and interactions with MHC molecules. In contrast to domestic pigs, African suids lack a 284-bp segment spanning exons 1 and 2 of the TRBV27 gene that contains a sequence encoding CDR1. In this study, we used the African swine fever virus (ASFV) as an example to investigate the effect of deleting the TRBV27-encoded CDR1 on the resistance of domestic pigs to exotic pathogens. We first successfully generated TRBV27-edited fibroblasts with disruption of the CDR1 sequence using CRISPR/Cas9 technology and used them as donor cells to generate gene-edited pigs via somatic cell nuclear transfer. The TRBV-edited and wild-type pigs were selected for synchronous ASFV infection. White blood cells were significantly reduced in the genetically modified pigs before ASFV infection. The genetically modified and wild-type pigs were susceptible to ASFV and exhibited typical fevers (>40 °C). However, the TRBV27-edited pigs had a higher viral load than the wild-type pigs. Consistent with this, the gene-edited pigs showed more clinical signs than the wild-type pigs. In addition, both groups of pigs died within 10 days and showed similar severe lesions in organs and tissues. Future studies using lower virulence ASFV isolates are needed to determine the relationship between the TRBV27 gene and ASFV infection in pigs over a relatively long period.

Objective To investigate whether dexmedetomidine (Dex) can reduce the severity of tourniquet-induced lung injury. Methods 36 patients undergoing total knee arthroplasty with a tourniquet were randomly assigned to the control (ischemia/reperfusion [I/R]) group and Dex group. Patients in the Dex group received a loading dose of Dex (0.8 μg/kg over 10 min intravenously) followed by continuous infusion of Dex (0.5 μg/kg/h intravenously) until the end of the surgery. The I/R group received an equal amount of 0.9% saline instead of Dex. The serum concentrations of tumor necrosis factor-α (TNF-α), Clara cell protein (CC-16), soluble receptor for advanced glycation end products (sRAGE), and brain-derived neurotrophic factor (BDNF) were measured and arterial blood gas analysis was performed before anesthesia and 30 min, 6 h, and 24 h after tourniquet release. Results In the I/R group, compared with baseline, the TNF-α, CC-16, and sRAGE concentrations were higher ( p < 0.05) and the BDNF concentration was lower ( p < 0.05) at most time points. In the Dex group, the TNF-α, CC-16, and sRAGE concentrations were lower than those in the I/R group ( p < 0.05), whereas the concentration of BDNF was higher ( p < 0.05). In the arterial blood gas analysis, the Dex group showed a significantly higher partial pressure of oxygen and arterial/alveolar oxygen tension ratio ( p < 0.05) and a significantly lower alveolar/arterial oxygen tension difference than the I/R group ( p < 0.05). Conclusion Dex administration partly inhibits the release of proinflammatory cytokines, affording protection against tourniquet-induced lung injury.

IntroductionRibonucleotide reductase (RR) is essential for the replication of the double-stranded DNA virus CyHV-2 due to its ability to catalyze the conversion of ribonucleotides to deoxyribonucleotides, and is a potential target for the development of antiviral drugs to control CyHV-2 infection.MethodsBioinformatic analysis was conducted to identify potential homologues of RR in CyHV-2. The transcription and translation levels of ORF23 and ORF141, which showed high homology to RR, were measured during CyHV-2 replication in GICF. Co-localization experiments and immunoprecipitation were performed to investigate the interaction between ORF23 and ORF141. siRNA interference experiments were conducted to evaluate the effect of silencing ORF23 and ORF141 on CyHV-2 replication. The inhibitory effect of hydroxyurea, a nucleotide reductase inhibitor, on CyHV-2 replication in GICF cells and RR enzymatic activity in vitro was also evaluated.ResultsORF23 and ORF141 were identified as potential viral ribonucleotide reductase homologues in CyHV-2, and their transcription and translation levels increased with CyHV-2 replication. Co-localization experiments and immunoprecipitation suggested an interaction between the two proteins. Simultaneous silencing of ORF23 and ORF141 effectively inhibited the replication of CyHV-2. Additionally, hydroxyurea inhibited the replication of CyHV-2 in GICF cells and the in vitro enzymatic activity of RR.ConclusionThese results suggest that the CyHV-2 proteins ORF23 and ORF141 function as viral ribonucleotide reductase and their function makes an effect to CyHV-2 replication. Targeting ribonucleotide reductase could be a crucial strategy for developing new antiviral drugs against CyHV-2 and other herpesviruses.

Summary: Traditional electrode materials still face vital challenges of few active sites, low porosity, complex synthesis process, and low specific capacitance. Herein, N-doped and 3D hierarchical porous graphene nanofoam (N-GNF) is created on carbon fibers (CFs) by employing a facile, fast, and environmentally friendly strategy of N2 plasma activation. After an appropriated N2 plasma activation, the graphene nanosheets (GNSs) synthesized by Ar/CH4 plasma deposition transform into N-GNF successfully. N doping donates rich active sites and increases the hydrophilia, while hierarchical nanoarchitecture exposes an enlarged effective contact area at the interface between electrode and electrolyte and affords sufficient space for accommodating more electrolytes. The as-assembled flexible N-GNF@CFs//Zn NSs@CFs Zn ion capacitor delivered a high energy density of 105.2 Wh kg−1 at 378.6 W kg−1 and initial capacity retention of 87.9% at the current of 2 A g−1 after a long cycle of 10,000.

Cyprinid herpesvirus 2 (CyHV-2) is a pathogen that causes significant losses to the global aquaculture industry due to mass mortality in crucian carp and goldfish. This study demonstrates that the ORF55/ORF57 deletion mutants CyHV-2-Δ55-CP and CyHV-2-Δ57-CP obtained through homologous recombination replicate effectively within the caudal fin of Carassius auratus gibelio (GiCF) cells and exhibit morphologies similar to the CyHV-2 wild-type strain. Both mutants demonstrated a decrease in virulence, with CyHV-2-Δ57-CP exhibiting a more significant reduction. This serves as a reference for the subsequent development of recombinant attenuated vaccines against CyHV-2. Additionally, both mutants expressed the inserted RGNNV-CP (capsid protein of Redspotted grouper nervous necrosis virus) fusion protein gene, and inoculation with CyHV-2-Δ57-CP-infected GiCF cell lysates elicited an antibody response in the grouper. These results indicate that, while ORF55 and ORF57 genes of CyHV-2 are not required for viral replication in vitro, they do play a role in virulence in vivo. Additionally, expression of foreign protein in CyHV-2 suggests that the fully attenuated mutant of CyHV-2 could potentially function as a viral vector for developing subunit vaccines or multivalent recombinant attenuated vaccines.

Abstract Photodetectors generally operate exclusively in either the broadband or narrowband. Developing bifunctional photodetectors that can detect photons in both broadband and narrowband would bring significant versatility to the optoelectronic platform. Nevertheless, the design of bifunctional integrated devices remains challenging due to the differentiated strategies with respect to device structure and material combination. Herein, we propose introducing polyvinylpyrrolidone to increase the viscosity of the perovskite precursor solution, which introduces abundant defects and cavities into the perovskite film while maintaining a relatively low film thickness. Then, we use methylamine gas to postprocess the middle area of the film to promote directional recrystallization and densification, thereby realizing narrowband and broadband dual‐function photodetection in a single device at zero bias. Both ends of the film exhibit a near‐infrared peak response at 780 nm with a narrow full‐width at half maximum of approximately 30 nm without an external bias. The middle broadband photodetector exhibits a high responsivity of 329 mA W−1 and EQE up to 52.46% at 780 nm. We make full use of narrow‐band wavelength selective detection and broadband full‐spectrum detection to achieve double encryption during signal transmission. This work represents an important step toward the realization of perovskite‐based multifunctional integrated devices.

High-speed permanent magnet synchronous motors (PMSMs) are usually started with the hybrid low-speed open-loop and high-speed closed-loop control mode. However, low-speed open-loop control produces large starting current, or even current overload, resulting in demagnetization and motor damage. Therefore, there is an urgent need to develop a high-speed permanent magnet synchronous motor with self-starting ability at low speed, that is, a permanent magnet hysteresis motor (PMHM). This paper takes the permanent magnet ring hysteresis column rotor structure as the research object. Firstly, the performance of this type of rotor is improved using the micro level of effective layer material AlNiCo, and the mechanical properties of AlNiCo are calculated. Secondly, based on the thick-walled cylinder theory, the analytical model for calculating the strength of the composite rotor considering the temperature effect is deduced, and the tangential and radial stress distribution for each part of the rotor under no-load and load conditions are obtained. Then, the electromagnetic losses and temperature field distribution of the rotor are obtained using the magneto-thermal bidirectional coupling finite element method. Finally, strength of the thermal rotor is analyzed by substituting the temperature rise curve function and AlNiCo parameters at the operating temperature. The comparison of the stress calculation results of the semi-analytical solution and the finite element method showed that the error between both of them is less than 5%, which verified that the semi-analytical solution can accurately analyze the thermal stress distribution of the rotor during high-speed rotation.

Kinetic approaches are generally accurate in dealing with microscale plasma physics problems but are computationally expensive for large-scale or multiscale systems. One of the long-standing problems in plasma physics is the integration of kinetic physics into fluid models, which is often achieved through sophisticated analytical closure terms. In this paper, we successfully construct a multimoment fluid model with an implicit fluid closure included in the neural network using machine learning. The multimoment fluid model is trained with a small fraction of sparsely sampled data from kinetic simulations of Landau damping, using the physics-informed neural network (PINN) and the gradient-enhanced physics-informed neural network (gPINN). The multimoment fluid model constructed using either PINN or gPINN reproduces the time evolution of the electric field energy, including its damping rate, and the plasma dynamics from the kinetic simulations. In addition, we introduce a variant of the gPINN architecture, namely, gPINNp, to capture the Landau damping process. Instead of including the gradients of all the equation residuals, gPINNp only adds the gradient of the pressure equation residual as one additional constraint. Among the three approaches, the gPINNp-constructed multimoment fluid model offers the most accurate results. This work sheds light on the accurate and efficient modeling of large-scale systems, which can be extended to complex multiscale laboratory, space, and astrophysical plasma physics problems.

High stiffness of hydrogen-bond network generally endows cellulose hydrogels with weak mechanical flexibility and shape uncontrollability, imposing restrictions on their application scopes. Herein, a flexible polyvinyl alcohol (PVA) network is penetrated through the rigid network of bamboo cellulose, inducing the formation of a dual-crosslinked hydrogel with superior flexibility and shape controllability. Besides, the borax crosslinker offers the bamboo nanocellulose-borax-PVA hydrogel with remarkable self-healing ability and high ionic conductivity. The hydrogel achieves a minimum sensing weight of 5 g (gauge factor: 1.47) and has a stable sensing sensitivity in both hot and cold environments (i.e., 0 and 60 °C). When fixed to the wrist of human, the hydrogel can regularly sense the motion signals, verifying the potential of the hydrogel for the development of next-generation wearable products. More importantly, the hydrogel can serve as a green platform to integrate with amoxicillin to create an effective composite to inhibit the growth of Escherichia coli and Staphylococcus aureus, which provides an effective route to develop eco-friendly and powerful functional composites. The drug-loaded hydrogel displays a negligible cytotoxicity and favorable biocompatibility towards human skin.

Based on the paucity of studies on the analysis of the coupled vibration response of the train-track-overhead System, in this article, finite element software ABAQUS was integrated with multi-body dynamics software, Universal Mechanism (UM), to construct a joint simulation model of the train-track overhead system under a railway line, with the focus on the investigation of the influence of different track irregularity levels, speeds and damping coefficients on the coupled vibration response of the vehicle-track-overhead system. The findings demonstrate that the response of the train body is sensitive to track irregularity, which primarily impacts the safety index of train operation. The results also suggest that the level of track irregularity should be rigorously regulated above AAR5 during construction. The train-track-overhead system functions well and satisfies the overhead system’s design requirements when the train travels through the reinforced line at a speed of no more than 60 km/h. When the train speed is 100 km/h, the vertical acceleration exceeds the limit for the “I” overhead system. There is a possibility of excessive lateral acceleration of the train body and excessive lateral force of the wheel and rail when the train speed is greater than 60 km/h, which endangers the safety of the driver. While it has little effect on the mid-span and vertical displacements, the damping factor of the bridge has a substantial impact on the vertical acceleration and mid-span acceleration of the vertical and horizontal beams. The study’s findings provide useful guidance.

Because of their flexibility, controllability and convenience, Automated Guided Vehicles (AGV) have gradually gained popularity in intelligent manufacturing because to their adaptability, controllability, and simplicity. We examine the relationship between AGV scheduling tasks, charging thresholds, and power consumption, in order to address the issue of how AGV charging affects the scheduling of flexible manufacturing units with multiple AGVs. Aiming to promote AGVs load balance and reduce AGV charging times while meeting customer demands, we establish a scheduling model with the objective of minimizing the maximum completion time based on process sequence limitations, processing time restrictions, and workpiece transportation constraints. In accordance with the model’s characteristics, we code the machine, workpiece, and AGV independently, solve the model using a genetic algorithm, adjust the crossover mutation operator, and incorporate an elite retention strategy to the population initialization process to improve genetic diversity. Calculation examples are used to examine the marginal utility of the number of AGVs and electricity and validate the efficiency and viability of the scheduling model. The results show that the AVGs are effectively scheduled to complete transportation tasks and reduce the charging wait time. The multi-AGV flexible manufacturing cell scheduling can also help decision makers to seek AGVs load balance by simulation, reduce the charging times, and decrease the final completion time of manufacturing unit. In addition, AGV utilization can be maximized when the fleet size of AGV is 20%-40% of the number of workpieces.

A new type of high-entropy alloy, a nitride-based (AlCrTiZrMo)N/ZrO2 nano-multilayered film, was designed to investigate the effect of ZrO2 layer thickness on the microstructure, mechanical properties, and thermal stability. The results show that when the thickness of the ZrO2 layer is less than 0.6 nm, it can be transformed into cubic-phase growth under the template effect of the (AlCrTiZrMo)N layer, resulting in an increased hardness. The (AlCrTiZrMo)N/ZrO2 film with a ZrO2 layer thickness of 0.6 nm has the highest hardness and elastic modulus of 35.1 GPa and 376.4 GPa, respectively. As the thickness of the ZrO2 layer further increases, ZrO2 cannot maintain the cubic structure, and the epitaxial growth interface is destroyed, resulting in a decrease in hardness. High-temperature annealing treatments indicate that the mechanical properties of the film decrease slightly after annealing at less than 900 °C for 30 min, while the mechanical properties decrease significantly after annealing for 30 min at 1000–1100 °C. The hardness and elastic modulus after annealing at 900 °C are still 24.5 GPa and 262.3 GPa, showing excellent thermal stability. This conclusion verifies the “template” effect of the nano-multilayered film, which improves the hardness and thermal stability of the high-entropy alloy.

By employing ordinary Portland cement as a matrix and PZT-5H piezoelectric ceramic as the functional body, 1-3 and 2-2 cement-based piezoelectric composites were prepared. Quasi-static compression tests were performed along with dynamic impact loading tests to study the electro-mechanical response characteristics of 1-3 and 2-2 cement-based piezoelectric composites. The research results show that both composites exhibit strain rate effects under quasi-static compression and dynamic impact loading since they are strain-rate sensitive materials. The sensitivity of the two composites has a non-linear mutation point: in the quasi-static state, the sensitivity of 1-3 and 2-2 composites is 157 and 169 pC/N, respectively; in the dynamic state, the respective sensitivity is 323 and 296 pC/N. Although the sensitivity difference is not significant, the linear range of the 2-2 composite is 24.8% and 61.3% larger than that of the 1-3 composite under quasi-static compression and dynamic impact loading, respectively. Accordingly, the 2-2 composite exhibits certain advantages as a sensor material, irrespective of whether it is subjected to quasi-static or dynamic loading.

Coronavirus disease 2019 (COVID-19) was first identified in Wuhan, China, in December 2019. Although previous studies have described the clinical aspects of COVID-19, few studies have focused on the early detection of severe COVID-19. Therefore, this study aimed to identify the predictors of severe COVID-19 and to compare clinical features between patients with severe COVID-19 and those with less severe COVID-19. Patients admitted to designated hospital in the Henan Province of China who were either discharged or died prior to February 15, 2020 were enrolled retrospectively. Additionally, patients who underwent at least one of the following treatments were assigned to the severe group: continuous renal replacement therapy, high-flow oxygen absorption, noninvasive and invasive mechanical ventilation, or extracorporeal membrane oxygenation. The remaining patients were assigned to the non-severe group. Demographic information, initial symptoms, and first visit examination results were collected from the electronic medical records and compared between the groups. Multivariate logistic regression analysis was performed to determine the predictors of severe COVID-19. A receiver operating characteristic curve was used to identify a threshold for each predictor. Altogether,104 patients were enrolled in our study with 30 and 74 patients in the severe and non-severe groups, respectively. Multivariate logistic analysis indicated that patients aged ≥63 years (odds ratio = 41.0; 95% CI: 2.8, 592.4), with an absolute lymphocyte value of ≤1.02×109/L (odds ratio = 6.1; 95% CI = 1.5, 25.2) and a C-reactive protein level of ≥65.08mg/L (odds ratio = 8.9; 95% CI = 1.0, 74.2) were at a higher risk of severe illness. Thus, our results could be helpful in the early detection of patients at risk for severe illness, enabling the implementation of effective interventions and likely lowering the morbidity of COVID-19 patients.

With a 10-kW, 120,000-r/min, ultra-high-speed permanent magnet synchronous motor taken as a prototype, experimental research is conducted on the rotor dynamic behaviours of a three-pad bidirectional gas foil bearing high-speed motor rotor system. Load-carrying properties of the three-pad bidirectional gas foil bearing are analysed, and natural frequencies of conical and parallel whirling modes of the elastically supported rotor are calculated based on an appropriate simplification to the stiffness and damping coefficients of the gas foil bearings. The prototype passes through a 90,000-r/min coast-down experiment. Experiments show that there are violent subsynchronous whirling motions that are evoked by the gas foil bearing–rotor system itself. The cause of shaft orbit drift is analysed, and the corresponding solution is put forward. The theoretical analysis and experimental results can offer a useful reference to the bearing–rotor system design of ultra-high-speed permanent magnet motors and its subsequent dynamic analysis.

Ultra-high-speed permanent magnet synchronous motors usually adopt the rotor in which a sleeve is interference fitted onto a ring permanent magnet or segmental permanent magnets. The rotors are always subject to the extreme working condition (e.g. high speed and high temperature). In order to predict the strength of the rotor, this article proposes analytical models considering three types of stress sources such as interference fit, centrifugal force, and temperature gradient. Examples are calculated and discussed. Results show that the effect of the temperature gradient on sleeve stresses is slight; however, the influence of the temperature gradient on stresses of the permanent magnet and shaft is dramatic; for the rotor with a outer surface forced air cooling, the shaft will generate a large radial expansion which causes a addition pressure effect on the permanent magnet and sleeve to prevent loosening and stripping of the interfaces. Moreover, segmental permanent magnets are helpful to reduce stress. The study will give a useful hint for the optimal design and manufacture of the ultra-high-speed permanent magnet rotor.

Bipolar rotors with cylindrical permanent magnets (PMs), which are magnetized in parallel, are generally used in ultra-high-speed permanent magnet synchronous motors (PMSMs). Aiming for this type of PMSM, two methods, namely, the Poynting theorem and the Maxwell stress tensor, were used to deduce the analytic expression of the electromagnetic driving torque. A locked-rotor simulation by the finite element (FE) method verifies the analytical solution. In addition, the cogging torque, back-electromotive force (emf), armature reaction inductance, etc., were calculated, compared, and discussed. The no-load back-emf was checked by a 60,000 rpm free coast experiment. The proposed analytical solution is very convenient to guide the design of the ultra-high-speed PMSM and check its performance.

Aiming at the ultra high speed permanent magnet synchronous motor (PMSM) supported by gas foil bearings (GFBs), this paper calculates the transient line starting of the motor. Firstly, the start effect of the rotor composed of cylindrical PM and stainless steel sleeve is studied. Then, in order to enhance the start torque, copper ring, nickel ring and copper squirrel-cage are introduced in the rotor and their start effect are analysed, respectively. It can be found that the rotor including nickel ring can be accelerated to set speed, but all the other rotors are failed due to the higher PM and braking torques. It can be concluded that some material owning slight large relative permeability can be applied in the rotor to reduce the PM field and contribute to start by using the line-start method.

The prognostic value of serum cystatin-C (Cys-C) in renal cell carcinoma (RCC) remains unknown. The purpose of this study is to explore the prognostic value of Cys-C for RCC patients.The levels of preoperative Cys-C, creatinine (CRE) and estimated glomerular filtration rate (e-GFR) were retrospectively collected in 325 RCC patients undergoing surgery. The cutoff values of Cys-C, CRE and e-GFR were determined by the standardized Cutoff Finder algorithm. The receiver operating characteristic (ROC) curve and pairwise comparison were performed to compare the three variables. Univariate and multivariate Cox regression analyses were performed to investigate the prognostic value of serum Cys-C in RCC.Based on the analysis of Cutoff Finder algorithm, ROC curve and pairwise comparison, the preoperative Cys-C was superior to CRE and e-GFR as a predictive factor in RCC. This paper presents an experimental research work that evaluates the pre-stress loss caused by friction in crushed limestone sand (CLS) concrete members with post-tensioning. A total of 26 full-scale pre-stressed concrete beams were constructed and tested for the friction loss experiment. The considered variables mainly included the duct-forming materials, wires of tendons and arrangement of ducts. The tensile forces at both active and passive ends of specimen were recorded by steps, and then the pre-stress friction loss for each case was calculated. The result shows that the proportion of pre-stress friction loss in specimen with multi-wire tendons is in the range of 10–40%, with the trend first increasing before decreasing. The pre-stress friction loss in specimen with curve duct accounts for 10–30%. The pressure on the curved part definitely increases the friction when compared with the straight duct. The pre-stress friction loss in specimen with rubber hose reaches nearly 40%, which is larger than the metal bellow and plastic bellow. The suggested values for each case are proposed for a deviation coefficient κ of 0.0017–0.007 and a friction coefficient μ of 0.108–0.858. This can provide reliable theoretical support for the design and construction.

This study aims at investigating high flexural strength iron-OER soft magnetic composites (SMCs) for active magnetic bearings (AMBs). Iron powders were firstly coated with oxide layer by the passivation process at room temperature. The effect of annealing methods on the static performance, magnetic properties, and flexural strength of SMCs was studied. The results showed that the significant improvements of permeability, magnetic loss, and flexural strength were presented by increasing the bonding number between iron surface and resin and using the two-step annealing method. In addition, the effect of pore shape and angle on the behavior of material fracture has been studied by FEM and the analysis was validated by experimental results.

Aberrant expression of miR-196a has been frequently reported in different cancers including pancreatic cancer. However, its function in pancreatic cancer has not been fully elucidated. Here, we investigated the expression pattern and the biological role of miR-196a in pancreatic cancer cell lines, as well as its interaction with a metastasis-related gene, nuclear factor-kappa-B-inhibitor alpha (NFKBIA). We demonstrated that miR-196a was up-regulated in human pancreatic cancer cell lines compared with immortalized pancreatic ductal epithelial cells by means of microRNAs microarray and qRT-PCR. Furthermore, down-regulation of miR-196a in PANC-1 suppressed its proliferation and migration with an increase in G0/G1 transition and decreased expression of Cyclin D1 and CDK4/6. Meanwhile, an increased expression in E-cadherin and decreased expression in N-cadherin and Vimentin were also observed. We identified a novel miR-196a target, NFKBIA, and down-regulation of miR-196a enhanced the expression of NFKBIA protein. Luciferase assay confirmed that NFKBIA was a direct and specific target of miR-196a. Silencing NFKBIA in PANC-1 cells enhanced its proliferation and migration. Taken together, our findings indicate that miR-196a is highly expressed in pancreatic cancer cell lines, and may play a crucial role in pancreatic cancer proliferation and migration, possibly through its downstream target, NFKBIA. Thus, miR-196a may serve as a potential therapeutic target for pancreatic cancer.

The primary objective of this scholarly article is to assess the effectiveness of implementing the tactical optimization model and decision-making strategy in the context of football coaching environments. The study involved the participation of 120 boys from a school in China, enrolled in grades 7–9, and actively engaged in a football program, with an average age of 13.8 years (SD = 1.26). Two subscales from the Tactical-technical and Social Competencies of Football Players Scale (TTSCS) and four subscales from the Melbourne Decision-Making Questionnaire (MDMQ) were employed in the research. The study revealed that the tactical optimization model and decision-making strategy in football empirically substantiated their effectiveness. The intervention group demonstrated improvements in attacking tactical-technical competencies (ATTC) by 10.03 and defensive (DTTC) by 11.35 compared to the control group, where statistically significant changes amounted to only 2.16 in the DTTC subscale. In the decision-making domain within the intervention group, a statistically significant increase in vigilance by 2.92 was noted, while other subscales decreased: hypervigilance by 3.75, transfer of responsibility, and procrastination by 3.45, indicating enhancements.
