Abstract The development of nasal inverted papilloma (NIP) is closely related to human papillomavirus (HPV) infection. Previous studies indicated that HPV11 shows the highest expression in NIP tissues. However, the mechanisms following its integration into host DNA require further clarification. In this study, high-throughput sequencing was employed to identify the HPV integration site KDM4A in HPV-positive specimens. The HPV11E6/E7 overexpression model was established in human nasal mucosal epithelial cells (HNE-pC), and the KDM4A gene was knocked out using CRISPR/Cas9 technology. Cell proliferation was assessed via CCK-8, colony formation, and EdU assays, while cell migration was evaluated through Transwell and wound healing assays. qRT-PCR and Western blot were used not only to analyze mRNA and protein expression in cells after HPV11E6/E7 overexpression and knockout of KDM4A but also to study the effect of the polarization of macrophages. A subcutaneous tumor model in nude mice validated the effects on proliferation and KDM4A knockout in vivo, with macrophage polarization types assessed via immunofluorescence staining. Results showed that HPV11E6/E7 overexpression significantly enhanced nasal epithelial cell proliferation and migration, along with promoting M1 macrophage polarization. Knockout of KDM4A inhibited these effects and delayed the progression of macrophages toward M1 polarization. Our findings suggest that low-risk HPV11 can drive the proliferation of nasal mucosa and regulate M1 macrophage polarization via KDM4A, potentially contributing to NIP pathogenesis. Targeting inhibition of KDM4A expression may represent a viable therapeutic strategy for HPV-positive NIP.

Efficient hydrogen storage and transportation are crucial for the sustainable development of human society. Ammonia, with a hydrogen storage density of up to 17.6 wt%, is considered an ideal energy carrier for large-scale hydrogen storage and has great potential for development and application in the “hydrogen economy”. However, achieving ammonia decomposition to hydrogen under mild conditions is challenging, and therefore, the development of suitable catalysts is essential. Metal oxide-based catalysts are commonly used in the industry. This paper presents a comprehensive review of single and composite metal oxide catalysts for ammonia decomposition catalysis. The focus is on analyzing the conformational relationships and interactions between metal oxide carriers and active metal sites. The aim is to develop new and efficient metal oxide-based catalysts for large-scale green ammonia decomposition.

Abstract Background Approximately 90% of renal malignancies are RCCs (renal cell carcinomas), and the primary subtype in histology is ccRCC (clear cell RCC). In recent years, pyroptosis has been considered a kind of inflammation-related programmed cell death that participates in the invasion, metastasis, and proliferation of tumour cells, thereby influencing tumour prognosis. Nonetheless, the expression level of pyroptosis-associated genes in RCCs and their relationship with prognosis remain obscure. Results In our research, 44 regulators of pyroptosis that were differentially expressed between normal kidney and ccRCC tissues were identified. ccRCC cases were categorized into 2 subgroups according to prognostic-related DEGs (differentially expressed genes), and there was a significant difference in OS (overall survival) between them. The prognostic value of pyroptosis-associated genes was assessed as a signature based on a cohort from TCGA (The Cancer Genome Atlas). Following Cox regression with DEGs and LASSO (least absolute shrinkage and selection operator), a 6-gene signature was established, and all ccRCC cases in the cohort from TCGA were categorized into an LR (low-risk) or HR (high-risk) group (P

Micropillar array electrodes represent a promising avenue for enhancing detection sensitivity and response current. However, existing methods for depositing electrode materials on micropillar arrays often result in uneven distribution, with the thin sidewall layer being less conductive and prone to corrosion. In addressing this issue, this study introduces electroplating to enhance the copper layer on the sidewall of micropillar array electrodes. These electrodes, fabricated through standard microelectronics processes and electroplating, are proposed for non-enzymatic glucose detection, with the copper layer deposited via electroplating significantly enhancing sensitivity. Initially, the impact of cetyltrimethylammonium bromide (CTAB) concentration as an inhibitor on the surface morphology and sensitivity of the plated layer was investigated. It was discovered that CTAB could decrease surface roughness, hinder the development of large and coarse grains, generate small particles, and boost sensitivity. Compared to the uncoated electrode and plating without CTAB, sensitivity was elevated by a factor of 1.66 and 1.62, respectively. Subsequently, the alterations in plating morphology and detection performance within a range of 0.3 ASD to 3 ASD were examined. Sensitivity demonstrated a tendency to increase initially and then decrease. The electrode plated at 0.75 ASD achieved a maximum sensitivity of 3314 μA·mM−1·cm−2 and a detection limit of 15.9 μM. Furthermore, a potential mechanism explaining the impact of different morphology on detection performance due to CTAB and current density was discussed. It was believed that the presented effective strategy to enhance the sensitivity of micropillar array electrodes for glucose detection would promote the related biomedical detection applications.

Background: F-box proteins play important roles in cell cycle and tumorigenesis. However, its prognostic value and molecular function in clear cell renal cell carcinoma (ccRCC) remain unclear. In this study, we established a survival model to evaluate the prognosis of patients with ccRCC using the F-box gene signature and investigated the function of FBXL6 in ccRCC. Methods: Comprehensive bioinformatics analyses were used to identify differentially expressed F-box and hub genes associated with ccRCC carcinogenesis. Based on the F-box gene signature, we constructed a risk model and nomogram to predict the overall survival (OS) of patients with ccRCC and assist clinicians in decision-making. Finally, we verified the function and underlying molecular mechanisms of FBXL6 in ccRCC using CCK-8 and EdU assays, flow cytometry, and subcutaneous xenografts. Results: A risk model based on FBXO39, FBXL6, FBXO1, and FBXL16 was developed. In addition, we drew a nomogram based on the risk score and clinical features to assess the prognosis of patients with ccRCC. Subsequently, we identified FBXL6 as an independent prognostic marker that was highly expressed in ccRCC cell lines. In vivo and in vitro assays revealed that the depletion of FBXL6 inhibited cell proliferation and induced apoptosis. We also demonstrated that SP1 regulated the expression of FBXL6. Conclusions: FBXL6 was first identified as a diagnostic and prognostic marker in patients with ccRCC. Loss of FBXL6 attenuates proliferation and induces apoptosis in ccRCC cells. SP1 was also found to regulate the expression of FBXL6.

Due to the advantages of simple construction, easy application and good environmental suitability, handheld structured-light measurement systems have broad application prospects in 3D measurements. Here, a monocular-vision-guided line-structured-light measurement system is developed, and the posture of the handheld device can be obtained via a specifically designed target attached to it. No more marker points need to be adhered onto the object under inspection. The key for the system calibration is to obtain the coordinate transformation matrix from the sensor to the featured target coordinate system. The mathematical model of the system is first established. Then, an improved multi-view calibration method is proposed, where a selection process for the image pairs is conducted for accuracy improvement. With this method, the maximum relative error of the measured stair heights can be reduced from 0.48% to 0.16%. The measurement results for the specific parts further verified the effectiveness of the proposed system and the calibration method.

Research on mental health states involves paying increasing attention to changes in daily life. Researchers have attempted to understand such daily changes by relying on self-reporting through frequent assessment using devices (smartphones); however, they are mostly focused on a single aspect of mental health. Assessing the mental health of a person from various perspectives may help in the primary prevention of mental illness and the comprehensive measurement of mental health. In this study, we used users' smartphone logs to build a model to estimate whether the scores on three types of questionnaires related to quality of life and well-being would increase compared to the previous week (fluctuation model) and whether they would be higher compared to the average for that user (interval model). Sixteen participants completed three questionnaires once per week, and their smartphone logs were recorded over the same period. Based on the results, estimation models were built, and the F-score ranged from 0.739 to 0.818. We also analyzed the features that the estimation model emphasized. Information related to “physical activity,” such as acceleration and tilt of the smartphone, and “environment,” such as atmospheric pressure and illumination, were given more weight in the estimation than information related to “cyber activity,” such as usage of smartphone applications. In particular, in the Positive and Negative Affect Schedule (PANAS), 9 out of 10 top features in the fluctuation model and 7 out of 10 top features in the interval model were related to activities in the physical world, suggesting that short-term mood may be particularly heavily influenced by subjective activities in the human physical world.

During space missions, astronauts live in a confined technological environment, completely isolated and deprived of the variety and variation found in the environment on Earth. This circumstance has a strong impact on the psycho-physiological states of the crew. Particularly in light of the plans for long-duration missions, new research needs to be carried out. The goal of this study, conducted at Xiangtan Central Hospital in China, was to test whether multicolour lighting can improve people’s psychological state in an isolated and confined environment over a period of seven days. Twenty participants (10 male and 10 female) were randomly divided into two groups: one group that was exposed to multicolour lighting and a control group, which was exposed to a static, monotonous white interior. The participants’ psychological state was recorded on the first day, the fourth day, and the seventh day. The results of the control group showed that the participants’ negative emotions and anxiety continued to increase over time, whereas the group randomly exposed to multicolour lighting that changed every three hours did not show any significant increase in negative emotions and anxiety. Moreover, the random change of light colour in the isolated environment appeared to help the participants increase their sense of surprise, thereby counteracting monotony. Finally, during this experiment, it was observed that when people who are accustomed to being connected to social networks were deprived of this, they experienced insomnia and unaccustomed reactions, in particular on the first days of deprivation. This article contributes to future space exploration and to social and psychological support of life in isolated and confined environments.

In the meteorology of Global Navigation Satellite System, the weighted mean temperature (Tm) is a key parameter in the process of converting the zenith wetness delay into precipitable water vapor, and it plays an important role in water vapor monitoring. In this research, two deep learning algorithms, namely, recurrent neural network (RNN) and long short-term memory neural network (LSTM), were used to build a high-precision weighted mean temperature model for China using their excellent time series memory capability. The model needs site location information and measured surface temperature to predict the weighted mean temperature. We used data from 118 stations in and around China provided by the Integrated Global Radiosonde Archive from 2010 to 2015 to train the model and data from 2016 for model testing. The root mean square error (RMSE) of the RNN_Tm and LSTM_Tm models were 3.01 K and 2.89 K, respectively. Compared with the values calculated by the empirical GPT3 model, the accuracy was improved by 31.1% (RNN_Tm) and 33.9% (LSTM_Tm). In addition, we selected another 10 evenly distributed stations in China and used the constructed model to test the prediction capability of the weighted mean temperature from 2010 to 2016. The RMSE values were 2.95 K and 2.86 K, which proved that the model also exhibits high generalization in non-modeling sites in China. In general, the RNN_Tm and LSTM_Tm models have a good performance in weighted mean temperature prediction.

The poor prognosis of locally advanced and metastatic head and neck squamous cell carcinoma (HNSCC) is primarily mediated by the functional properties of cancer stem cells (CSCs) and resistance to chemoradiotherapy. We investigated whether the aldehyde dehydrogenase (ALDH) inhibitor disulfiram (DSF) can enhance the sensitivity of therapy. Cell viability was assessed by the 1-(4,5-dimethylthiazol-2-yl)-3,5-diphenylformazan (MTT) and apoptosis assays, and the cell cycle and reactive oxygen species (ROS) levels were evaluated by fluorescence-activated cell sorting (FACS). The radio-sensitizing effect was measured by a colony formation assay. The synergistic effects were calculated by combination index (CI) analyses. The DSF and DSF/Cu2+ inhibited the cell proliferation (inhibitory concentration 50 (IC50) of DSF and DSF/Cu2+ were 13.96 μM and 0.24 μM). DSF and cisplatin displayed a synergistic effect (CI values were 2+ abolished the cisplatin-induced G2/M arrest (from 52.9% to 40.7% and 41.1%), and combining irradiation (IR) with DSF or DSF/Cu2+ reduced the colony formation and attenuated the G2/M arrest (from 53.6% to 40.2% and 41.9%). The combination of cisplatin, DSF or DSF/Cu2+, and IR enhanced the radio-chemo sensitivity by inducing apoptosis (42.04% and 32.21%) and ROS activity (46.3% and 37.4%). DSF and DSF/Cu2+ enhanced the sensitivity of HNSCC to cisplatin and IR. Confirming the initial data from patient-derived tumor xenograft (PDX) supported a strong rationale to repurpose DSF as a radio-chemosensitizer and to assess its therapeutic potential in a clinical setting.

With the increase of mining depth and strength, the evolution of bolt axial force is increasingly becoming important for ensuring the reliability and safety of support. To improve the problem of the existing coal mine roadway pressure-monitoring method, whereby it is difficult to continuously monitor the axial force of the bolt over a long period of time, a full rod fiber bragg grating (FBG) force-measuring bolt and system were designed based on the principle of fiber grating sensing. It was found that a trapezoidal groove is a relatively better groove. The linearity between the center wavelength offset of the fiber grating and the axial force was more than 0.99, and the conversion formula between the axial force of the bolt rod and the wavelength change of the fiber grating were obtained. The real-time monitoring revealed that the axial force of the bolt obviously changed before and after compression. The axial force distribution curve can be divided into the stable zone, growth zone, and peak zone. The influence of the roadway abutment pressure was approximately 130 m ahead of the working face, and the peak area was within the 25–35 m range of the advance working face. The axial force of the bolt rod at the end of the anchorage linearly increased with the tail end of the bolt, the axial force of the free segment was the largest, and the overall stress was essentially the same. The application results demonstrate the feasibility and effectiveness of the FBG full-length force bolt.

We investigate the concurrent three-dimensional deformations of fiber-reinforced composite sheets subjected to out-of-plane bending moments via a continuum model, where we invoke the neo-Hookean strain energy model for the matrix material of fiber-reinforced composite, and assimilate the strain energy of fiber reinforcements into the matrix material model by accounting for stretching, bending, and twisting kinematics of the fibers through the computations of the first-order and second-order gradient of deformation. Emphasis is placed on deriving the Euler equation and boundary conditions of bending moment within the framework of the variational principle and configuring composite surfaces using differential geometry. Significant attention has been given to illustrating the concurrent three-dimensional deformation of fiber composite, meshwork deformation, and fiber kinematics. The simulation results reveal that for a square fiber composite subjected to the out-of-plane bending moment, the maximum in-plane deformation of matrix material occurs along the diagonal direction of the domain while the center of the domain experiences weak in-plane deformation. Notably, the matrix material performs isotropic/anisotropic properties depending on the domain size/shape. In addition, the simulated unit fiber deformations reasonably validate the overall deformation of the network, underscoring that the deformations of the embedded fiber units govern the overall mechanical performance of the fiber meshwork. More importantly, the continuum model qualitatively provides reasonable predictions on the damage patterns of construction materials by demonstrating the kinematics of matrix material and meshwork deformation. In the service life of traffic markings, the visibility of the markings can be significantly impacted due to continuous rolling by vehicles. This study aims to investigate the change of retroreflective coefficient of traffic markings at different positions on the road after being rolled by vehicles. A two-month follow-up study was conducted on a specific section of the road, focusing on the sideline and middle line of the lane. The retroreflective coefficient instrument was utilized to collect data on the retroreflective coefficient of the markings. Results revealed that the middle line, due to more frequent vehicle rolling, exhibited a larger range in its retroreflective coefficient as compared to the sideline of the lane. The change in reflection ability of hot melt markings, when rolled by the vehicle, depends on the mosaic condition of the glass beads within the coating. It was observed that the rolling of the vehicle had a direct impact on the retroreflective coefficient of the hot melt marking line. Furthermore, it was found that with increased rolling of the marking line, the greater the impact on its reflection capacity and the thickness also demonstrated a significant decrease.
