Travelers’ diarrhea has a high incidence rate among deployed US military personnel and can hinder operational readiness. The Global Travelers’ Diarrhea study is a US Department of Defense­–funded multisite surveillance effort to investigate the etiology and epidemiology of travelers’ diarrhea. During 2018–2023, we enrolled 512 participants at partner institutions in 6 countries: Djibouti, Georgia, Egypt, Honduras, Nepal, and Peru. Harmonized laboratory methods conducted at each partner institution identified >1 pathogens, including Escherichia coli (67%–82%), norovirus (4%–29%), and Campylobacter jejuni (2%–20%), in 403 (79%) cases. Among cases, 79.7% were single infections, 19.6% were double infections, and 0.7% were triple infections. The most common enterotoxigenic E. coli colonization factors identified were CS3 (25%) and CS21 (25%), followed by CS2 (18%) and CS6 (15%). These data can inform best treatment practices for travelers’ diarrhea and support US military health readiness.

Abstract Key message A Soil Monitoring Law to improve soil health across all land uses has been proposed by the European Commission. As forests soils have different chemical and physical properties as well as biogeochemical dynamics compared to agricultural land, they also face different challenges in maintaining and restoring soil health. Examples are soil acidification, eutrophication by atmospheric deposition, responses to climate change, and loss of biodiversity. Therefore, we propose forest soil specific health descriptors and thresholds based on experience and knowledge from existing long-term monitoring programs.

Abstract Soils, just like all other ecosystem compartments, change over time and, consequently, conditions for soil‐inhabiting organisms are also changing, affecting their composition and diversity. Soil biodiversity is a critical component of ecosystems that supports many essential ecosystem functions and services, such as nutrient cycling, carbon sequestration, water regulation and biomass production for food, fodder, fibre and energy. However, and despite the importance of soil biodiversity for ecosystem health and human well‐being, neither current state, drivers, potential consequences for ecosystem services nor options for sustainable governance of soil biodiversity are well understood. Here, we provide a framework for and argue that conducting a national assessment of soil biodiversity, albeit being a complex endeavour, is fundamental to building a baseline to understand the current state and trends of soil biodiversity, but also to identify the main drivers of change, the impacts of soil biodiversity loss and the potential pathways for conservation and sustainable governance of soil biodiversity.

Immune exhaustion is a hallmark of ovarian cancer. Using multiparametric flow cytometry, the study aimed to analyze protein expression of novel immunological targets on CD3+ T cells isolated from the peripheral blood (n = 20), malignant ascites (n = 16), and tumor tissue (n = 6) of patients with ovarian cancer (OVCA). The study revealed an increased proportion of effector memory CD8+ T cells in OVCA tissue and malignant ascites. An OVCA-characteristic PD-1high CD8+ T cell population was detected, which differed from PD-1lowCD8+ T cells by increased co-expression of TIGIT, CD39, and HLA-DR. In addition, these OVCA-characteristic CD8+ T cells showed reduced expression of the transcription factor TCF-1, which may also indicate reduced effector function and memory formation. On the contrary, the transcription factor TOX, which significantly regulates terminal T cell-exhaustion, was found more frequently in these cells. Further protein and gene analysis showed that CD39 and CD73 were also expressed on OVCA tumor cells isolated from solid tumors (n = 14) and malignant ascites (n = 9). In the latter compartment, CD39 and CD73 were also associated with the expression of the “don’t eat me” molecule CD24 on tumor cells. Additionally, ascites-derived CD24+EpCAM+ tumor cells showed a higher frequency of CD39+ or CD73+ cells. Furthermore, CD39 expression was associated with unfavorable clinical parameters. Expression of CD39 on T cells was upregulated through CD3/CD28 stimulation and its blockade by a newly developed nanobody construct resulted in increased proliferation (eFluor), activation (CD25 and CD134), and production of cytotoxic cytokines (IFN-γ, TNF-α, and granzyme-B) of CD8+ T cells.

Retail is currently in a state of upheaval due to digitalization and the associated booming online trade. Sales figures in stationary retail are stagnating. Online trading offers advantages through convenience and quick availability that customers no longer do not want to miss. Due to climate change, retailers and customers have changed their mind set towards more sustainability. Consumers are increasingly demanding sustainable products, so retailers must act appropriately to be both digital and climate-friendly in the future. The aim of this exploratory study is to use the example of floristry retailers to analyze whether they are growing with digitalization and whether they are taking sustainable initiatives to act as climate-friendly as possible. In addition, the florists' whole value chain is examined for climate friendliness. The results of the exploratory online survey of 68 floristry businesses in Germany show that both digitalization as well as climate and sustainability awareness play a significant role in floristry retail.

This study explores the potential of cross-publisher bundled offers as a strategy for increasing subscription sales in digital journalism. While innovative forms of bundling are an integral part of media distribution in music (e.g., Spotify) and film (e.g., Netflix), their adoption in digital journalism has been limited, despite research showing that bundled access to products can increase consumers’ willingness to pay, especially in younger target groups. Against this background, we conduct a choice-based conjoint analysis using data from a representative survey of the German online population (n = 1,542). Results show that bundling digital journalism has the potential to raise publisher revenues and subscription sales in digital markets. In particular, they highlight that a comprehensive, cross-publisher bundled offer, available at a fixed monthly rate, has the potential to stimulate digital journalism sales among different consumer groups in a relatively balanced way, including those who are typically more reluctant towards journalism. These findings align with the principles of information goods economics, which posit that maximising the size of digital content bundles often tends to be the most profitable distribution strategy. However, it is crucial to examine these findings in the context of the potential negative effects associated with this emerging business model in digital journalism, such as the cannibalisation of print subscriptions, diminished brand identification, and a possible imbalanced distribution of revenues.

The standard of care for glioblastoma (GBM) involves surgery followed by adjuvant radio- and chemotherapy, but often within months, patients relapse, and this has been linked to glioma stem cells (GSCs), self-renewing cells with increased therapy resistance. The identification of the epidermal growth factor receptor (EGFR) and platelet-derived growth factor receptor (PDGFR) as key players in gliomagenesis inspired the development of inhibitors targeting these tyrosine kinases (TKIs). However, results from clinical trials testing TKIs have been disappointing, and while the role of GSCs in conventional therapy resistance has been extensively studied, less is known about resistance of GSCs to TKIs. In this study, we have used compartmentalised proteomics to analyse the adaptive response of GSCs to ponatinib, a TKI with activity against PDGFR. The analysis of differentially expressed proteins revealed that GSCs respond to ponatinib by broadly rewiring lipid metabolism, involving fatty acid beta-oxidation, cholesterol synthesis, and sphingolipid degradation. Inhibiting each of these metabolic pathways overcame ponatinib adaptation of GSCs, but interrogation of patient data revealed sphingolipid degradation as the most relevant pathway in GBM. Our data highlight that targeting lipid metabolism, and particularly sphingolipid degradation in combinatorial therapies, could improve the outcome of TKI therapies using ponatinib in GBM.

IntroductionTumor-associated macrophages (TAMs) represent an important cell population within the tumor microenvironment, but little is known about the phenotype and function of these cells. The present study aims to characterize macrophages in high-grade serous ovarian cancer (HGSOC).MethodsPhenotype and expression of co-regulatory markers were assessed on TAMs derived from malignant ascites (MA) or peripheral blood (PB) by multiparametric flow cytometry. Samples were obtained from HGSOC patients (n=29) and healthy donors (HDs, n=16). Additional expression analysis was performed by RNAseq (n=192). Correlation with clinically relevant parameters was conducted and validated by a second patient cohort (n=517). Finally, the role of TIGIT in repolarization and phagocytosis was investigated in vitro.ResultsExpression of the M2-associated receptors CD163, CD204, and CD206, as well as of the co-regulatory receptors TIGIT, CD226, TIM-3, and LAG-3 was significantly more frequent on macrophages in HGSOC than in HDs. CD39 and CD73 were broadly expressed on (mainly M2) macrophages, but without a clear clustering in HGSOC. CD163 mRNA levels were higher in TAMs from patients with residual tumor mass after surgery and associated with a shorter overall survival. In addition, TIGIT expression was associated with a higher tumor grading, indicating a prognostic relevance of M2 infiltration in HGSOC. TIGIT blockade significantly reduced the frequency of M2 macrophages. Moreover, combined blockade of TIGIT and CD47 significantly increased phagocytosis of ovarian cancer cells by TAMs in comparison to a single blockade of CD47.ConclusionCombined blockade of TIGIT and CD47 represents a promising approach to enhance anti-CD47-facilitated phagocytosis.

Digitalization and digital technologies have risen sharply in commerce in form of online offers and advice, even in formerly less technology-based sectors. By using the example of flower shops, the aim of this study is to find out whether customers perceive digital offers to be generally useful or necessary, or if local service is still sufficient. To answer this question, a quantitative survey was conducted in selected flower shops in Germany. 82 customers took part. Although most customers have not yet resorted to an online offering when buying flowers, over 66% of respondents are generally in favor of digital offers in the floristry industry.

Subscription-based news platforms (such as “Apple News+” or “Readly”) that bundle content from different publishers into one comprehensive package and offer it to media users at a fixed monthly rate are a new way of accessing and consuming digital journalism. These services have received little attention in journalism studies, although they differ greatly from traditional media products and distribution channels. This article empirically investigates the perception of journalism platforms based on eight qualitative focus group discussions with 55 German news consumers. Results show that the central characteristics these platforms should fulfill in order to attract users are strikingly similar to the characteristics of media platforms from the music and video industries, in particular regarding price points, contract features, and modes of usage. Against this background, the potential and perspectives of a subscription-based news platform for journalism’s societal role are discussed.

Abstract Background To understand life-history strategies in migratory bird species, we should focus on migration behaviour and possible carry-over effects on both population and individual level. Tracking devices are useful tools to directly investigate migration behaviour. With increased use of tracking devices, questions arise towards animal welfare and possible negative effects of logger on birds. Several studies were conducted to address this question in birds that were tagged and tracked for one complete non-breeding season including migration but with mixed results. To detect individual-based decisions regarding migration strategy, we need to track the same individuals several times. So far, there are no studies investigating effects of repeatedly tagging on reproduction and life-history traits in individual migratory birds, especially in small birds. Methods We used long-term data of 85 tagged common swifts (Apus apus), a long-distance migratory bird, of a breeding colony in Germany to test whether carrying a geolocator or GPS logger once or repeatedly during non-breeding season affected return rate, apparent survival, and parameters determining reproductive success. Additionally, we checked for individual differences in arrival date and breeding parameters when the same individuals were tagged and when they were not tagged in different years. Further, we calculated the individual repeatability in arrival at the breeding colony and date of egg laying in repeatedly tagged swifts. Results Once and repeatedly tagged birds returned to the colony at a similar rate as non-logger birds and arrived earlier than non-logger birds. We found no effect of logger-type on return rate in logger birds. We detected no differences in apparent survival, time lag to clutch initiation, date of clutch initiation, clutch size, number of chicks and fledglings between logger and non-logger birds. We found neither an effect of loggers nor of logger-types on the arrival date and breeding parameter on individual-level. Arrival date was highly repeatable and date of clutch initiation was moderately repeatable within repeatedly tagged individuals.
