Abstract Background Lymphomatoid granulomatosis (LyG) is a rare extralymphatic lymphoproliferative disease characterized by lymphocytic invasion into vascular walls and damage to blood vessels. The lungs are affected in 90% of LyG cases, followed by the skin, central nervous system (CNS), kidneys and liver. Case presentation Here we report a case of a young woman with LyG, with CNS involvement as the initial clinical manifestation. Computer tomography (CT) scans showed multiple nodular, patchy and flocculent high-density shadows in both lungs without mediastinal lymph node enlargement. Magnetic resonance imaging (MRI) scans showed multiple abnormal signal intensities in the right cerebellar hemisphere, frontal, parietal and temporal lobes, and dorsal brainstem, which became patchy and annular after enhancement. The post-operative pathological analysis of lesion samples confirmed the diagnosis of grade II LyG. Conclusions LyG should be concerned in young adults showing multiple radiological brain and lung lesions. Resection and postoperative medication of steroid hormones and IFN-α may be effective in the treatment of LyG.

Heterosis utilization is very important in hybrid seed production. An AL-type cytoplasmic male sterile (CMS) line has been used in wheat-hybrid seed production, but its sterility mechanism has not been explored. In the present study, we sequenced and verified the candidate CMS gene in the AL-type sterile line (AL18A) and its maintainer line (AL18B). In the late uni-nucleate stage, the tapetum cells of AL18A showed delayed programmed cell death (PCD) and termination of microspore at the bi-nucleate stage. As compared to AL18B, the AL18A line produced 100% aborted pollens. The mitochondrial genomes of AL18A and AL18B were sequenced using the next generation sequencing such as Hiseq and PacBio. It was found that the mitochondrial genome of AL18A had 99% similarity with that of Triticum timopheevii, AL18B was identical to that of Triticum aestivum cv. Chinese Yumai. Based on transmembrane structure prediction, 12 orfs were selected as candidate CMS genes, including a previously suggested orf256. Only the lines harboring orf279 showed sterility in the transgenic Arabidopsis system, indicating that orf279 is the CMS gene in the AL-type wheat CMS lines. These results provide a theoretical basis and data support to further analyze the mechanism of AL-type cytoplasmic male sterility in wheat.

Abstract Background The exploration of gene-disease associations is crucial for understanding the mechanisms underlying disease onset and progression, with significant implications for prevention and treatment strategies. Advances in high-throughput biotechnology have generated a wealth of data linking diseases to specific genes. While graph representation learning has recently introduced groundbreaking approaches for predicting novel associations, existing studies always overlooked the cumulative impact of functional modules such as protein complexes and the incompletion of some important data such as protein interactions, which limits the detection performance. Results Addressing these limitations, here we introduce a deep learning framework called ModulePred for predicting disease-gene associations. ModulePred performs graph augmentation on the protein interaction network using L3 link prediction algorithms. It builds a heterogeneous module network by integrating disease-gene associations, protein complexes and augmented protein interactions, and develops a novel graph embedding for the heterogeneous module network. Subsequently, a graph neural network is constructed to learn node representations by collectively aggregating information from topological structure, and gene prioritization is carried out by the disease and gene embeddings obtained from the graph neural network. Experimental results underscore the superiority of ModulePred, showcasing the effectiveness of incorporating functional modules and graph augmentation in predicting disease-gene associations. This research introduces innovative ideas and directions, enhancing the understanding and prediction of gene-disease relationships.

In order to solve the problems of high impurity rate and large loss rate in the whole fresh peanut harvesting and production process in hilly areas of China, the method of computational fluid dynamics (CFD) and discrete element (DEM) coupling is used to examine the gas–solid two-phase simulation of the cleaning device of the crawler fresh-peanut-picking combine harvester. In addition, the Box–Behnken test method is used to analyze the influence of different parameters on the impurity content and loss rate of materials in the cleaning process by taking the fan speed, feed amount, and air inlet angle as the test factors, and the optimal combination of working parameters is sought. The simulation results show that when the fan speed is 2905.07 r/min, the feed rate is 0.80 kg/s, the air inlet angle is 43.14°, the impurity content is 7.32%, and the loss rate is 4.78%. Compared with the simulation test results, the impurity content is increased by 0.68%, and the loss rate is increased by 1.24%, which verifies the reliability of the simulation model, and the research results provide some technical support for the improvement of the cleaning device in the later stages.

Abstract Chronic intrauterine hypoxia (ICH) may lead to permanent alterations in the offspring's body structure, function, and metabolism through the "developmental programming" pathway, resulting in lasting changes in physiology and metabolism, as well as the onset of adult-onset diseases. The aim was to investigate intrauterine growth restriction caused by ICH and its effect on ovarian reserve function in female offspring at different developmental stages after birth. Healthy female Sprague–Dawley rats (n = 20) were pregnant by normal mating, and the rats in the ICH group were treated with chronic intrauterine hypoxia twice a day for 04 h00 each time from day 4 to 21 of gestation. After the first hypoxic treatment, four pregnant rats were randomly selected from the ICH and natural control groups for arterial blood gas analysis. When wide-width sowing wheat after rice stubble (WRS) in a rice-wheat rotation area, there is a problem of poor uniform of seed distribution. To solve the problem, this study designed the seed distribution plate (SDP) structure and optimized its critical structure parameters. Firstly, combined with the operating principles of the wide-width seeder and the agricultural standards for WRS, the main structural parameters affecting seed movement were determined by a theoretical analysis of seed grain dynamics and SDP structure. Secondly, the operational performance of six different structures of SDP under different structural parameters was compared using discrete element simulation technology. The structure of SDP most suitable for WRS wide-width seeding and the value ranges of key structural parameters that have a significant impact on the coefficient of the variation of seed lateral uniformity (CVLU) were determined. Finally, the pattern and mechanism of the influence of key structural parameters of SDP on the CVLU were analyzed. The optimum parameter combination was obtained and a field validation test was conducted on this. The results showed that the anti-arc ridge and arc bottom structure (S6) is more suitable for the agronomy standards of WRS wide-width seeding. The chord length of ridge, installation inclination, angle between the chord and tangent of the end of ridge line (ACT), span, and bottom curve radius are determined as the key structural parameters affecting the CVLU, and there is a lower CVLU (42.8%) when the ACT is 13°. The primary and secondary order of the influence of each factor on CVLU is the chord length of the ridge, span, installation inclination, and bottom curve radius. The corresponding parameter values after optimization are 140 mm, 40°, 75 mm and 50 mm, respectively. A field test was conducted on the SDP after optimizing parameters, and the CVLU was 30.27%, which was significantly lower than the CVLU before optimization.

The soil during the sowing period of wheat after rice stubble cannot be accurately described by existing models and parameters with DEM because of its high moisture content and strong viscosity. The purpose of this study is to conduct an overall simulation of high-viscosity paddy soil and to analyze the accuracy of the model. Based on the results of an unconfined compression test and shear test, the range of bond parameters is preliminarily determined by a simulation test. Through the P-BD test and RSM test, the parameters with significant influence are determined to be normal stiffness per unit area (SN), shear stiffness per unit area (SS), and critical shear stress (CS), and an optimized combination of these parameters is obtained. Based on the optimized model, the error range and error generation mechanism of the model are analyzed under different operating parameters. The results show that the optimal parameter combination is SN of 1.07 × 107 N/m3, SS of 0.70 × 107 N/m3, and CS of 0.35 × 105 Pa, corresponding to a compression force of 120.1 N and a shear force of 7.70 N. With an increase in forward speed or seeding quantity or a decrease in rotary plowing speed, the model accuracy tends to increase, and the range of relative errors was found to be from 8.8% to 28.4%. The results can provide a research basis for the study of the motion state of seeds under soil. It can also further enrich parameter data of soil discrete element simulation models and provide a reference for related research studies.

This study proposes a negative-pressure fruit-soil separating device for peanuts cultivated in hilly and mountainous areas after combined harvesting, and the mechanism of the movement of the material in the process of material screening, fruit-soil separating, and pneumatic conveying of the device was analyzed. In addition, a four-factor, three-level Box-Behnken regression design test was used to explore the optimum operating parameters of the peanut fruit-soil separation device. The results showed that the best fruit-soil separation effect was achieved when the wind speed of the blower was 13.58 m/s, the height of the suction nozzle from the screen surface was 27 mm, the length of the suction port was 64 mm, and the feeding rate was 600 kg/h. Validation tests demonstrated that the impurity rate of 0.16% and the peanut pod loss rate of 0.2% exceeded the industry standard, indicating superior performance.

This meta-analysis evaluated the diagnostic value of ultrasonic elastography in detecting placental stiffness in the diagnosis of preeclampsia (PE). A systematic search was conducted in the EMBASE, Web of Science, Cochrane Library, Scopus database, and PubMed databases to identify studies published before June 2023 using ultrasonic elastography to diagnose PE. The sensitivity, specificity, and diagnostic odds ratio of ultrasonic elastography for diagnosing PE were calculated, and a summary receiver operating characteristic curve model was constructed. The degree of heterogeneity was estimated using the I2 statistic, and a meta-regression analysis was performed to explore its sources. A protocol was determined previously (PROSPERO: CRD42023443646). We included 1188 participants from 11 studies, including 190 patients with PE and 998 patients without PE as controls. Overall sensitivity and specificity of ultrasonic elastography in detecting placental stiffness for the diagnosis of PE were 89% (95% CI: 85–93) and 74% (95% CI: 51–89), respectively. The I2 values for sensitivity and specificity were 59% (95% CI: 29–89) and 96% (95% CI: 95–98), respectively. The area under the receiver operating characteristic curve was 0.90 (95% CI: 0.87–0.92). The meta-regression analysis showed no significant heterogeneity. Ultrasonic elastography exhibits good diagnostic accuracy for detecting placental stiffness and can serve as a non-invasive tool for differentially diagnosing PE.

In order to provide basic data and a theoretical basis for waste mulch film crushing and separation equipment, the mechanical and suspension properties of waste mulch film and cotton stalk were determined in this study. In the mechanical properties test, the tensile properties of waste mulch film and the shear properties of different parts of cotton stalk were investigated. In the suspension properties test, the suspension properties of different qualities of waste mulch film and different parts of cotton stalk were investigated. The results showed that the average maximum tensile forces and fracture displacements of the waste mulch film were 1.70 N (horizontal), 0.80 N (vertical) and 254.75 mm (horizontal), 145.22 mm (vertical). The shear properties of the cotton stem after wrapping the mulch film were greatly improved; the maximum shearing force was 186.89% higher than that of the unwrapped specimens. When the total suspension velocity was from 3.09 to 4.07 m/s, separation of mulch film and cotton stalk could be achieved by wind.

The suspension velocity is the core of the cleaning and sorting mechanisms that utilize a combination of a fan and vibrating sieve. To investigate this, various experimental subjects, such as peanuts with different kernels and clay-heavy clods in different states, were used. The experiment involved simulating the suspension velocity of materials through numerical calculations using fluid dynamics and particle discrete element coupling. The Eularian model was employed to study the coupled gas-solid two-phase flow. The experiment measured the suspension velocities of single and double kernel peanuts, which were found to be 8.34~9.40 m/s and 8.13~9.51 m/s, respectively. Under 20.4% water content and lumpy conditions, the suspension velocities of smaller clods, side by side clods, and larger clods were 12.61~14.30 m/s, 14.16~15.76 m/s and 16.44~18.72 m/s, respectively; under 20.4% water content and smaller clods, the suspension velocities of lumpy and strip of clods were 12.61~14.30 m/s, 11.90~14.13 m/s, respectively; under lumpy and smaller clods, the suspension velocity at 17.6%, 20.4%, and 23.9% water content ranged from 12.38 to 14.20 m/s, 12.61 to 14.30 m/s, and 12.62 to 14.49 m/s, respectively. The simulations showed that the suspension velocity for different types of peanuts, clod sizes, shapes, and water contents was less different from the actual experiments. Specifically, the relative errors in suspension velocity for single-kernel peanuts, double-kernel peanuts, smaller clods, side-by-side clods, larger clods, lumpy clods, strips of clods, and clods with 17.3%, 20.4%, and 23.9% water content were 1.2%, 4.1%, 0.4%, 2.0%, 4.4%, 0.4%, 5.1%, 5.4%, 0.4%, and 1.9%, respectively, compared to actual experiment measurements. The results indicate a significant difference in the suspension velocity between peanuts and clay-heavy clods, which can be distinguished from each other based on this difference. Furthermore, the simulation results have been found to be consistent with the experimental results, thus verifying the feasibility of measuring the material suspension velocity using CFD-DEM gas-solid coupling.

In this study, we quantified and analyzed the root-cutting process of garlic with a test bench with pressure sensors on the basis of the comparative analysis of various information perception methods. On the basis of the output value of the pressure sensor, the force curve of garlic roots was plotted, and the double round blade cutting module is optimized on the basis of the force curve diagram. The innovative proposal of slotted round blades for garlic root cutting is presented here. The round blade diameter is 110 mm, the center distance is 100 mm, the blade thickness is 1 mm, and the blade speed is 1200 r/min. According to the analysis of the force curve, it was found that the slotted round blade with the slanted blade could generate a strong thrust to cut the roots. The cutting effect was better and the cutting surface of the roots was straight. The slotted blade meets the need for cutting garlic roots.

Objective: To evaluate the efficacy of ultrasound-guided intra-articular injection of triptolide-loaded solid lipid nanoparticle (TP-SLN) for treatment of antigen-induced arthritis (AIA) in rabbits. Material and Methods: Knee joints of 33 New Zealand rabbits with AIA were injected intra-articularly with triptolide (TP: n = 7), TP-SLN (n = 7), betamethasone (BS: n = 7) and dimethyl sulfoxide (DMSO: n = 6). The remaining six rabbits were untreated as the control group. The injection therapy in intervention groups was initiated 1 week after the last immunization in order to avoid irreversible joint damage in the later induction. The ultrasonic scores of the joints were assessed based on synovitis, synovial blood flow and bone erosion. Meanwhile, the correlations of ultrasonic scores and pathological scores were determined. The efficacy and side effects of each group were determined by combining ultrasonic scores, pathological scores, behavior, appetite, weight, joint diameter, skin temperature and biochemical examination.Results: 1) Compared with the control group, the diameters of knee joints of the TP, TP-SLN and BS groups began to reduce 1 week after intra-articular injection (p < 0.01). 2) With the exception of the DMSO group, the interventions were effective in treating synovitis compared with the control group, with TP-SLN and BS being the best. The ultrasonic and pathological scores in synovitis of the TP group were lower than that of model group (Z = -2.726 and -2.530, p < 0.05). The ultrasonic scores differed significantly between BS group and TP-SLN group (Z = −2.17 and -2.360, respectively, p < 0.05) and pathological scores (Z = −2.687 and −2.082, respectively, p < 0.05). 3) Compared with the control group, the TP, BS and TP-SLN were all effective in treating synovial blood flow and bone erosion and there were no significant differences of ultrasonic and pathological scores among them (p > 0.05). The ultrasonic scores of synovial blood flow (Z = −3.033, −2.842, −3.277, p < 0.01) were lower than in the controls. The ultrasonic scores (Z = -2.948, -3.141, -3.210, p < 0.01) and pathological scores (Z = −2.216, −2.505, −2.505, p < 0.05) of bone erosion were also lower than in the model group.4) There were significant correlations between the ultrasonic and pathological scores of synovial inflammation and bone erosion (r = 0.832 and 0.859 respectively, p < 0.001).Conclusions: The therapeutic effect of TP-SLN on arthritis is better than that of TP, but there is no difference between BS and TP-SLN. Therefore, TP-SLN may be used as an alternative to BS in the treatment of rheumatoid arthritis in the future. The ultrasonic and pathological scores showed significant correlation in synovitis and bone erosion. Ultrasound can provide a useful assessment of synovitis in early arthritis.

Potato stem removal is one of the critical technical problems of potato mechanized harvesting; it directly affects the quality of potato harvesting and potato storage. There have been several studies on potato stem removal mechanisms. In practice, however, it was found that the potato stem removal rate was greatly influenced by the posture of the stem before it entered the removal mechanism. In this study, we designed a potato stem posture adjustment mechanism consisting of elastic curtains. A test rig was built to investigate the effect of curtain height, curtain width, and curtain suspension height on potato passage rate and potato stem removal rate. The Box–Behnken design (BBD), combined with the response surface method, was used to conduct the test. The optimal construction and installation parameters for each elastic curtain were determined as 278.93 mm for the curtain height, 20 mm for the curtain width, and 260 mm for the curtain suspension height. The predicted values of potato passage rate and potato stem removal rate under the optimal parameters were 92.36% and 82.83%, which were consistent with the validation test results. Based on the optimization results, a rigid-flexible coupled simulation model for a potato stem transported-posture adjustment process based on Abaqus and Adams was constructed. The maximum impact of the elastic curtain of the stem posture adjustment mechanism on the potato stem was 15.91 N and caused the stem to spring back. The projection angle β′ of the stem posture angle in the xoz plane before posture adjustment was 19.07°, and the β′ of the stem after posture adjustment was 87.18°. At this time, the stem was basically parallel to the rod of the separating sieve and had a high probability of falling from the gap of the bar to complete the removal of the stem. Overall, the stem transport position adjustment mechanism effectively adjusted the stem transported posture and improved the stem removal rate in potato mechanical harvesting.

When sowing with a wide boundary under full rice straw retention in the rice–wheat rotation area of China, conventional fertilization methods have some problems, such as a low fertilizer utilization rate, heap soil around a buried fertilizer device, or blocked fertilizing orifice. Firstly, combined with theoretical analysis, discrete element numerical simulation technology, and central composite test method, the wide-boundary fertilization device for wheat wide-boundary sowing was designed. Then, with the coefficient of variation for particle uniformity (CVPU) as the response value, the central composite experiment was carried out on the key structural parameters (focal length coefficient, lateral span, tilt angle, and ground clearance) of the wide-boundary fertilization device by EDEM software. Finally, the influential rules of core factors of the device on the CVPU were analyzed by Design-Expert software; then, the optimal parameter combination was determined and verified by a field test. The results showed that all factors had significant effects on the CVPU. The primary and secondary factors affecting the CVPU were the tilt angle, lateral span, focal length coefficient, ground clearance, tilt angle × ground clearance, and lateral span × ground clearance, in which there were certain interactions between the tilt angle and ground clearance and lateral span and ground clearance. When the focal length coefficient, lateral span, tilt angle, and ground clearance were 1.5, 60 mm, 30°, and 192 mm, respectively, the lateral was minimum. In this case, the theoretical value and field test value were 14.11% and 17.63%, respectively. The field test value is consistent with the theoretical calculation value. This study could provide references for the design of a fertilizer-spreading device with a wide boundary.

The uneven sowing of wheat on ground covered with rice straw in the rice–wheat rotation area in the middle and lower reaches of the Yangtze River has become a serious problem. Therefore, a test bed for throwing soil after sowing with a wide wheat seed belt was designed, which could complete the functions of straw crushing, straw lateral concentration and uniform sowing at one time. The discrete element simulation model of throwing soil after sowing with a wide wheat seed belt was established with rotary blade shaft speed, soil guide plate angle and soil retaining plate angle as variables. Taking the variation coefficient of wheat sowing depth and variation coefficient of sowing lateral uniformity as evaluation indexes, the effects of three variables on sowing uniformity were analyzed by single factor test and Box–Behnken test. The results of single factor test observed that when the rotating speed of rotary blade shaft was 260–300 rpm, the angle of soil guide plate was 36°–48°, the angle of soil retaining plate was 58°–74° and the experiment of utilizing a soil throwing and covering device with a wide seed belt after sowing revealed a good consistency of sowing depth and lateral uniformity effect. The Box–Behnken simulation experiment showed that the primary and secondary factors affecting the variation coefficient of wheat sowing lateral uniformity were the angle of soil guide plate, the rotation speed of rotary blade shaft, the angle of soil retaining plate and the angle of soil guide plate. When the rotation speed of rotary blade shaft, the angle of soil guide plate and the angle of soil retaining plate were 282.1 rpm, 42.4° and 65.5°, respectively, the soil throwing and covering device after sowing has the best seed-homogenizing effect. At this time, the variation coefficients of sowing depth and lateral uniformity in simulation test and field verification test were 4.35% and 4.57%, respectively, and 12.46% and 12.73%, respectively. The results of field verification test were basically consistent with those of the simulation test, which proved that the results of applying discrete element methods to optimize the soil-throwing device after sowing with a wide seed belt were credible. This study could provide a theoretical reference for the structure optimization of a soil-throwing device after sowing with a wide seed belt.

To address the fact that existing studies may not be able to accurately describe the discrete element parameters of the soil during the potato harvesting period and to improve the accuracy of the potato harvesting equipment simulation studies, this study was conducted on the soil around the harvested potato tubers in the field. The simulation parameters of the discrete element model of soil were determined by the soil stacking angle test and soil block shear crushing test with the actual test measurements as the target. Based on a series of experimental designs, the factors affecting the stacking angle were the static friction coefficient and the rolling friction coefficient between soil particles; and the rolling friction coefficient between soil and steel. The factors affecting the maximum shear damage force were normal stiffness and shear stiffness per unit area. The quadratic regression equations were established and solved to obtain the optimal discrete element simulation parameters. The results can provide more realistic and reliable parameters for the construction of soil simulation models of potato fields during harvesting and for the discrete element simulation of soil-touching components of potato harvesting equipment. It can also further enrich the parameter data of soil discrete element simulation models and provide a reference for related research
