4. Suitability of microsatellites BM1329 and OarAElOl as markers for the introgression of the FecB locus into different sheep breeds

Abstract

Litter size in sheep can be improved by introgression of the FecB gene. Genetic markers closely linked to the FecB locus can be used to detect gene carriers. The efficiency of introgression can be increased by using marker assisted selection. Individuals of the prolific breeds Tyrolean Mountain sheep (n = 41), Romanov (n = 43), East Friesian Milksheep (n = 35) and Merinoland sheep (n = 265) were genotyped with the microsatellite markers OarAElOl and BMI 329 linked to the FecB gene to estimate allele frequencies of both microsatellites in these breeds. The frequencies of the microsatellite alleles linked to the FecB gene in Merinoland sheep were 0.07 and 0.1 respectively. The alleles occured also in the other breeds (Tyrolean Mountain sheep, Romanov and East Friesian Milksheep) but with mostly higher frequencies. Furthermore the segregation of the microsatellite alleles was studied in families produced by mating Merinoland sheep ewes (Fec+/Fec+) with two heterozygous Booroola Merino rams (Fec+/FecB) in order to use these markers within an indirect gene test. The recombination rate between BM 1329 and OarAElOl was 13 cM (LOD score 14.6).

7. Optical coherence tomography system mass-producible on a silicon photonic chip

Abstract

Miniaturized integrated optical coherence tomography (OCT) systems have the potential to unlock a wide range of both medical and industrial applications. This applies in particular to multi-channel OCT schemes, where scalability and low cost per channel are important, to endoscopic implementations with stringent size demands, and to mechanically robust units for industrial applications. We demonstrate that fully integrated OCT systems can be realized using the state-of-the-art silicon photonic device portfolio. We present two different implementations integrated on a silicon-on-insulator (SOI) photonic chip, one with an integrated reference path (OCTint) for imaging objects in distances of 5 mm to 10 mm from the chip edge, and another one with an external reference path (OCText) for use with conventional scan heads. Both OCT systems use integrated photodiodes and an external swept-frequency source. In our proof-of-concept experiments, we achieve a sensitivity of −64 dB (−53 dB for OCTint) and a dynamic range of 60 dB (53 dB for OCTint). The viability of the concept is demonstrated by imaging of biological and technical objects.

Abstract

We demonstrate for the first time a waveguide-based frequency shifter on the silicon photonic platform using single-sideband modulation. The device is based on silicon-organic hybrid (SOH) electro-optic modulators, which combine conventional silicon-on-insulator waveguides with highly efficient electro-optic cladding materials. Using small-signal modulation, we demonstrate frequency shifts of up to 10 GHz. We further show large-signal modulation with optimized waveforms, enabling a conversion efficiency of -5.8 dB while suppressing spurious side-modes by more than 23dB. In contrast to conventional acousto-optic frequency shifters, our devices lend themselves to large-scale integration on silicon substrates, while enabling frequency shifts that are several orders of magnitude larger than those demonstrated with all-silicon serrodyne devices.

Abstract

Two methods have been developed for the assessment of conservation priorities on the basis of molecular markers. According to the Weitzman approach, contributions to genetic diversity are derived from genetic distances between populations. Alternatively, diversity within and across populations is optimized by minimizing marker-estimated kinships. We have applied, for the first time, both methods to a comprehensive data set of 69 European cattle breeds, including all cosmopolitan breeds and several local breeds, for which genotypes of 30 microsatellite markers in 25-50 animals per breed have been obtained. Both methods were used to calculate the gain in diversity if a breed was added to a set of nine non-endangered breeds. Weitzman-derived diversities were confounded by genetic drift in isolated populations, which dominates the genetic distances but does not necessarily increase the conservation value of a breed. Marker-estimated kinships across populations were less disturbed by genetic drift than the Weitzman diversities and assigned high conservation values to Mediterranean breeds, which indeed have genetic histories that differ from the non-endangered breeds. Prospects and limitations of marker-assisted decisions on conservation priorities are discussed.

Abstract

Name of Gene. Inhibin-[Alpha] (INHA). Genus and Species. Capra hircus. Source and Description of Primers. The primers were designed from the flanking sequences of a [(TG).sub.n], microsatellite identified in a 2.2-kb PstI fragment of an ovine cosmid clone, containing the INHA gene (Jager and Hiendleder, 1994). Primer Sequences. Forward Primer: 5'-AGCGTGTGAAGCTGGAGAT-3'; reverse Primer 5'-ACGTGATCACTACCACAGTACGGA-3'. Method of Detection. The PCR (15-[micro]L final volume) was performed using 50 to 100 ng of caprine genomic DNA, 10 pM of each primer, 200 [micro]M of each dNTP, .4 unit of Taq polymerase (AGS, Heidelberg), 1.5 mM [MgCl.sub.2], and PCR buffer (20 mM Tris-HCl, pH 8.55; 1.6 mM [NH.sub.4] [SO.sub.4]). Thermal cycling began with an initial cycle of 94 [degrees] C (1.5 min), followed by 30 cycles of 94 [degrees] C (1 min), 60 [degrees] C (1 min), and 72 [degrees] C (1 min), and concluded with a final extension at 72 [degrees] C (5 min) using a Perkin Elmer Gene Amp PCR system 9600 cycler. For obtaining the allele frequency data, the forward primer was Cy5 fluorescence end-labeled (Amersham-Pharmacia, Freiburg) and the analyses were done using an automated laser detection system (A.L.F. express, Amersham Pharmacia, Freiburg). Amplification of caprine genomic DNA resulted in 210- to 230-bp products that were run on a 5.5% Long Ranger gel (.5 mm, 6 M urea). The genotyping for linkage mapping was done by incorporation of radioactive gamma [P.sup.33] according to Schibler et al. (1998). Description of Polymorphism. The microsatellite containing amplification products showed fragment lengths indicating repeat numbers ranging from [(TG).sub.12] to [(TG).sub.22]. Inheritance Pattern. Codominant inheritance was observed in 12 families. Frequency. The frequencies of alleles in a total of 25 unrelated animals from different breeds (Wei [Beta] e Deutsche Edelziege, Bunte Deutsche Edelziege, Burenziege, Toggenburger Ziege) were .091, .432, .114, .114, .068, .136, and .045 for fragments of 210, 214, 216, 218, 224, 226, and 230 bp. The calculated heterozygosity and PIC (Botstein et al., 1980) in these animals were .75 and .73, respectively. Chromosomal Location. Previous physical mapping by FISH has placed INHA on caprine chromosome 2q41-42 (Goldammer et al., 1995). Linkage Mapping. Significant linkages between the polymorphic INHA-microsatellite and gene markers on chromosome 2 were found using CRI-MAP, version 2.4 (Greene et al., 1989). In the male linkage analysis, the significant two-point LOD scores reported ranged from 3.52 to 7.65 (Table 1). The identified microsatellite was placed in a chromosomal region containing the microsatellite loci ARO 28, BMS 829A, and BMS 829B by multipoint analysis between 110 and 130 cM on chromosome 2, where the microsatellites BMS 829A and BMS 829B were amplified using the same primer pair. The PCR product was cloned into the vector pCR 2.1 TOPO (Invitrogen, Leek) and sequenced to confirm homology to the original ovine sequence (GenBank accession no. AF115342) obtained from the ovine cosmid clone. The sequence similarity in microsatellite flanking regions between ovine and caprine loci was 88%. Inhibin is a glycoprotein hormone that suppresses pituitary follicle stimulating hormone secretion (Ling et al., 1985). Two different inhibins have been identified sharing a common [Alpha]-subunit but different [Beta]-subunits ([[Beta].sub.A] and [[Beta].sub.B]). The two forms of inhibin are named with inhibin A ([[Alpha]-[Beta].sub.A]]) and inhibin B ([[Alpha]-[Beta].sub.B]) (Mason et al., 1985). Abstract

The Neolithic introduction of domestic cattle into Europe was followed by differential adaptation, selection, migration and genetic isolation, leading ultimately to the emergence of specialized breeds. We have studied the differentiation of European cattle by amplified fragment length polymorphism (AFLP) fingerprinting. Combining AFLP data sets from two laboratories yielded 81 biallelic polymorphic markers scored in 19-22 individual animals from 51 breeds. Model-based clustering differentiated Podolian cattle as well as French and Alpine breeds from other European cattle. AFLP genetic distances correlated well with microsatellite-based genetic distances calculated for the same breeds. However, the AFLP data emphasized the divergence of taurine and indicine cattle relative to the variation among European breeds and indicated an Eastern influence on Italian and Hungarian Podolian breeds. This probably reflects import from the East after the original introduction of domestic cattle into Europe. Our data suggest that Italian cattle breeds are relatively diverse at the DNA sequence level.

Abstract

Amphibians are a suitable indicator group for monitoring possible negative direct or indirect effects of GMO cultivation at the individual and population level. Direct effects could occur in aquatic ecosystems via uptake of GM pollen or GM detritus by anuran larvae. However, indirect negative effects caused by changes in cultivation practices (changes in pesticide use, for instance) are more likely. The VDI Guideline 4333 aims to ensure comprehensive monitoring of the different life-stages of anuran species that are common in agricultural landscapes of Austria, Germany and Switzerland. The guideline includes a novel approach to tadpole monitoring. To assess immediate effects, tadpole, metamorph and adult deformation rates are compared with naturally occurring deformation rates. Adult population size, adult body condition and juvenile emergence are monitored over multiple years to assess long-term effects of GM crop cultivation on population viability. At each study site, monitoring has to be carried out at multiple amphibian breeding sites which differ in their exposure to GM crop cultivation. All monitoring data have to be stored in a central database for future meta-analyses. This will ultimately allow for generalized statements about the impact of GM crop cultivation on amphibians. Although specifically designed for GM crops, VDI Guideline 4333 may also serve as a model for studying the effects of a wider range of stressors on amphibian populations in agriculture and forestry.

Abstract

Research was conducted to examine the adrenocorticotropic hormone receptor gene locus. Hybridization experiments were performed on human genomic clones of 1 kb and 2.9 kb to detect polymorphism. A 2-kb rat cDNA was used as a control for the neural melanocortin receptor. Results indicate that adrenocorticotropin induces steroid hormone production in the adrenal gland through specific cell surface receptors.

Abstract

Genetic diversity was investigated among four Sudanese domesticated guinea fowl populations collected in different regions of Sudan: the states of Blue Nile (BL), Gezira and Khartoum (G), Kassala and Gedaref (KG), and West and North Kordofan (N). In addition, one wild population from Dinder National Park (D) was included. From 25 microsatellites chosen, 10 were informative and used for the current study. A total of 107 alleles were found with observed heterozygosity between 0.364 and 0.494. The populations kept on farms showed high genetic identity with values between 0.9269 and 0.9601. Neighbor-joining tree analysis and STRUCTURE modeling showed that the wild population clearly differs from the populations kept on farms. [ABSTRACT FROM AUTHOR]
