Abstract Background As a novel and time-efficient exercise form, high-intensity interval training (HIIT) has shown great potential in improving health-related physical fitness among diverse populations. However, empirical evidence on its efficacy among the elderly has not been well summarized. This systematic review and meta-analysis aimed to determine the effect of HIIT interventions on the parameters related to physical fitness and health of older adults, including resting heart rate (HR), systolic blood pressure (SBP), diastolic blood pressure (DBP), cardiorespiratory fitness (CRF), body mass index (BMI), body fat percent (BF%), waist circumference (WC), muscular endurance (ME), muscular strength (MS), muscular power (MP), balance and flexibility, compared to non-exercise and other-exercise (e.g., moderate-intensity continuous training, resistance training) conditions. Methods Literature published from January 2000 to May 2023 was collected through extensive searches across eight databases and relevant review papers. Randomized controlled trials (RCTs) featuring a minimum 2-week exercise intervention for older adults (≥ 60 years) were included. The pooled effect size of Hedges’g was estimated using random-effects models in R. Meta-regression was performed for both categorical (health status, duration of training programme, and frequency) and continuous moderators (mean age, male rate, and attrition rate). Results Forty-four eligible RCTs with 1863 participants (52.1% female; 60.5–81.2 years) were included in the quantitative analysis. Compared to non-exercise condition, HIIT significantly improved resting HR (g = -0.36, 95%CI = [-0.67, -0.05], P = 0.032), SBP (g = -0.29, 95%CI = [-0.54, -0.03], P = 0.008), CRF (g = 0.77, 95%CI = [0.51, 1.04], P 0.05). Meta-regression indicated that mean age moderated the HIIT effect on resting HR (b = -0.02, P = 0.014; HIIT vs. other-exercise condition) and SBP (b = 0.03, P = 0.048; HIIT vs. non-exercise), and attrition rate moderated the effect on CRF (b = 0.03, P = 0.007; HIIT vs. non-exercise). Conclusion This study supports the efficacy of HIIT in improving resting HR, SBP, CRF, BF%, MS, ME and balance among older adults. More empirical evidence is needed to determine the efficacy of HIIT for MP and flexibility in this population. Trial Registration PROSPERO CRD42022316246.

Objective The process of a stainless steel train body derailment accident in urban rail transit in 2015 is simulated, reproducing the direct cause and process of the car body damage in the complex accident scene. Method Based on the investigation of the accident scene and the stainless steel car body structure and characteristics, the finite element method is used to reconstruct the accident model and analyze the changes of the speed, acceleration and energy of the train. Result & Conclusion Simulation results show that the intermediate cars climbing, snaking and colliding with each other caused by huge energy and rugged ground at the time of the accident forms the main reason for the fracture failure of stainless steel car body structure in the accident. The car body fractures mainly locate at the connections between the cab frame and the under-frame, the end wall and the roof, the under-frame and the side wall, as well as the under-frame and the end wall.

Abstract Background Liver surgery during the perioperative period often leads to a significant complication known as hepatic ischemia-reperfusion (I/R) injury. Hepatic I/R injury is linked to the innate immune response. The cGAS-STING pathway triggers the activation of innate immune through the detection of DNA within cells. Nevertheless, the precise mechanism and significance of the cGAS-STING pathway in hepatic I/R injury are yet to be investigated. Methods Mouse model of hepatic I/R injury was used in the C57BL/6 WT mice and the STING knockout (STING-KO) mice. In addition, purified primary hepatocytes were used to construct oxygen-glucose deprivation reperfusion (OGD-Rep) treatment models. Results Our research revealed a notable increase in mRNA and protein levels of cGAS and STING in liver during I/R injury. Interestingly, the lack of STING exhibited a safeguarding impact on hepatic I/R injury by suppressing the elevation of liver enzymes, liver cell death, and inflammation. Furthermore, pharmacological cGAS and STING inhibition recapitulated these phenomena. Macrophages play a crucial role in the activation of the cGAS-STING pathway during hepatic I/R injury. The cGAS-STING pathway experiences a significant decrease in activity and hepatic I/R injury is greatly diminished following the elimination of macrophages. Significantly, we demonstrate that the activation of the cGAS-STING pathway is primarily caused by the liberation of mitochondrial DNA (mtDNA) rather than nuclear DNA (nDNA). Moreover, the safeguarding of the liver against I/R injury is also attributed to the hindrance of mtDNA release through the utilization of inhibitors targeting mPTP and VDAC oligomerization. Conclusions The results of our study suggest that the release of mtDNA plays a significant role in causing damage to liver by activating the cGAS-STING pathway during I/R injury. Furthermore, inhibiting the release of mtDNA can provide effective protection against hepatic I/R injury.

Coal mine rescue robots perform search and rescue tasks in unstructured underground tunnel environments. Traditional path planning algorithms may encounter issues such as low efficiency, non-optimal paths, and poor smoothness when applied to search spaces that are large or complex. Additionally, tunnels feature complex environmental characteristics such as intersections, where robots are prone to deviating from preset routes or scraping against tunnel walls. To address these challenges and enhance the navigation accuracy of robots, improvements to the path planning algorithm for wheeled coal mine rescue robots are proposed: ① The heuristic global path planning A* algorithm is enhanced by employing layered neighborhood search and pruning techniques to optimize the search process. The cost function is refined to better balance the influence of actual cost and heuristic cost, thus more accurately assessing the cost of each node, adapting to real situations, reducing computational complexity, and smoothing the path using B-spline methods. ② The Random Sample Consensus (RANSAC) fitting algorithm is utilized to construct a geometric model of coal mine tunnel walls, facilitating the extraction of feature point coordinates of intersections for inclusion in the planning system. The path is optimized using the local support property of B-spline basis functions. When additional path optimization points are added subsequently, only the shape of the curve in the corresponding interval is affected, leaving the rest of the path unaffected. ③ A comprehensive local force field is established based on the constructed environmental geometric model and extracted feature points. Adjustment coefficients are introduced to optimize the distribution of the force field, and motion control is achieved using the Particle Swarm Optimization (PSO) optimized PID (Proportion Integral Differential) algorithm, enhancing the robot's adaptability to complex environments such as tunnel intersections. The feasibility of the algorithm principles and applications is validated through MATLAB and ROS (Robot Operating System) simulations. Experimental results demonstrate that the proposed method can realize functions such as path planning and autonomous driving in complex intersection environments.

As one of the most common primary glomerulopathies, immunoglobulin A nephropathy（IgAN） is a major cause of end-stage renal disease （ESRD）. Its proper treatment has remained rather challenging. Disease progression may be delayed if associated risk factors are intervened. Hyperuricemia is a common clinical feature of IgAN and it is correlated with disease progression. However, the relationship has been elusive between hyperuricemia, pathological injury and disease progression in IgAN. This review focused upon the prevalence of hyperuricemia in IgAN, related pathogenesis and the relationship with the progression of IgAN and therapeutic targets.

Abstract Objective To evaluate the accuracy of Type B2 based on narrow-band imaging-magnifying endoscopy (NBI-ME) in judging invasion depth of superficial esophageal squamous cell carcinoma (SESCC) and analyze potential influencing factors. Method Data from 113 patients where Type B2 was observed by magnifying endoscopy and confirmed by postoperative pathology as SESCC at the First Affiliated Hospital of Fujian Medical University and Fujian Provincial Hospital from January 2015 to April 2024 were retrospectively analyzed. Patients were divided into correct prediction and incorrect prediction groups according to the postoperative pathological results, and the prediction accuracy was calculated. The incorrect prediction group was further divided into overestimation and underestimation groups to identify the influential factors for overprediction and underprediction, respectively. Abstract Background This study quantitatively analyzed the anatomic structure of the alveolar bone in the maxillary molar region at three potential locations for Temporary Anchorage Device (TAD) placement. Additionally, the study compared the variability in this region across different age groups, sagittal skeletal patterns, vertical facial types, and sexes. Methods In this retrospective cone-beam computed tomography study, the buccal alveolar bone was analyzed in the posterior molar area of 200 patients, the measurement items include buccal alveolar bone height, alveolar bone thickness, interradicular distance, and maxillary retromolar space. Results Buccal alveolar height was greatest in the U56 region. The interradicular space was largest in the U56 region and increased from the alveolar crest to the sinus floor. Buccal alveolar bone thickness was highest in the U67 region and generally increased from the alveolar crest to the sinus floor. The maxillary retromolar space gradually increased from the alveolar crest to the root apex. Conclusions TADs are safest when placed in the buccal area between the maxillary second premolar and the first molar, particularly at the 9 mm plane. The U67 region is the optimal safe zone for TAD placement for maxillary dentition distalization. TADs placement in adolescents can be challenging. Maxillary third molar extraction can be considered for maxillary dentition distalization.

【Objective】 Surface membrane irrigation is a new irrigation technique developed over the past decades to reduce water leaching and improve water retention in the root zone. This paper investigates the effect of different subsurface-membrane irrigation amounts on soil moisture and winter wheat growth. 【Method】 The field experiment compared three irrigation amounts: 300 m3/hm2 (W1), 450 m3/hm2 (W2) and 600 m3/hm2 (W3), each irrigation treatment having two irrigation schedules: continuous irrigation (S1), and intermittent irrigation (S2). Traditional border irrigation was taken as the control (CK). During the experiment, we measured water distribution in the soil, as well as growth, water consumption, yield composition and water use efficiency of the wheat. 【Result】 ① Compared to CK, the W3+S1 and W3+S2 improved soil water in the root zone and reduced deep water infiltration, therefore increasing plant height, grain yield and water use efficiency (WUE). ② With the increase in irrigation amount, water content in the 20-60 cm soil layer increased. Compared with S1, S2 reduced the infiltration of irrigation water into the 80-100 cm soil layer. ③ Plant height, leaf area index (LAI) and water consumption of the wheat were positively correlated with irrigation amount, regardless of its growing stages. Irrigation scheduling did not have a significant effect on plant height and LAI, but significantly affected total water consumption of the wheat. ④ With the increase in irrigation amount, S2 increased grain yield by 3.46% to 13.11%, compared with S1, with the increase depending on irrigation amount. With the increase in irrigation amount, the grain yield and WUE increased first and then decreased. 【Conclusion】 Compared with CK, subsurface membrane irrigation saved water and increased grain yield. Considering soil water distribution, crop growth, water consumption, grain yield and WUE, W2+S2 was optimal for wheat production in the studied region.

Abstract The window strategy, known for its flexibility and efficiency, is extensively used in dynamic path planning. To further enhance the performance of the Automated Guided Vehicles (AGVs) sorting system, the two processes of AGV movement and path planning can be executed concurrently based on the window strategy. Nonetheless, difficulties in matching the computing time of the planning server with the moving time of AGVs may cause delays or reduced path optimality. To address the problem, this paper proposes an optimal time reuse strategy. The proposed solution controls computing time by managing path length for each planning instance, ensuring alignment with the moving time of AGVs to maximize path optimality and avoid delays. To achieve this, two aspects need to be considered. Firstly, on a systemic level, we control the entry rate of AGVs by adjusting the replanning period, thus avoiding congestion caused by excessive AGVs and maintaining high system efficiency. Secondly, we reversely control the computing time by adjusting the path length that needs to be planned for each single planning, so that it matches the moving time of AGVs. Simulation results show that our method outperforms existing top-performing methods, achieving task completion rates 1.64, 1.57, and 1.12 times faster across various map sizes. This indicates its effectiveness in synchronizing planning and movement times. The method contributes significantly to dynamic path planning methodologies, offering a novel approach to time management in AGV systems.

Abstract Background In recent years, global climate change in tandem with increased human activity has resulted in habitat degradation or the migration of rare medicinal plants, potentially impacting the quality of medicinal herbs. Astragalus membranaceus var. mongholicus is a valuable bulk medicinal material in Northwest China. As the demand for this medicinal herb continues to increase in both domestic and international markets, ensuring the sustainable development of high-quality Astragali Radix is important. In this study, the maximum entropy (Maxent) model was applied, thereby incorporating 136 distribution records, along with 39 environmental factors of A. membranaceus var. mongholicus, to assess the quality zonation and potential distribution of this species in China under climate change. Results The results showed that the elevation, annual mean temperature, precipitation of wettest month, solar radiation in June, and mean temperature of warmest quarter were the critical environmental factors influencing the accumulation of astragaloside IV and Astragalus polysaccharide in A. membranaceus var. mongholicus. Among the twelve main environmental variables, annual mean temperature, elevation, precipitation of the wettest month, and solar radiation in November were the four most important factors influencing the distribution of A. membranaceus var. mongholicus. In addition, ecological niche modelling revealed that highly suitable habitats were mainly located in central and western Gansu, eastern Qinghai, northern Shaanxi, southern Ningxia, central Inner Mongolia, central Shanxi, and northern Hebei. However, the future projections under climate change suggested a contraction of these suitable areas, shifting towards northeastern high-latitude and high-elevation mountains. Conclusions The findings provide essential insights for developing adaptive strategies for A. membranaceus var. mongholicus cultivation in response to climate change and can inform future research on this species. By considering the identified environmental factors and the potential impacts of the predicted climate changes, we can visualize the regional distribution of high-quality Radix Astragali and develop conservation strategies to protect and restore its suitable habitats.

Objective To explore the efficacy and safety of obinutuzumab (OBZ) for phospholipase A2 receptor（PLA2R）-associated membranous nephropathy (MN) with an intermediate-to-high risk of progression. MethodsFrom January 2022 to November 2022, ten patients with a diagnosis of primary membranous nephropathy (PMN) received OBZ treatment. Baseline profiles, medical history, 24 hours urinary total protein (24 hUTP), blood biochemistry, cellular immunity, humoral immunity and other parameters were recorded and statistically analyzed by Graphpad Prism 9.4.1 software. Descriptions of \begin{document}$ \bar x $\end{document} ± \begin{document}$ s $\end{document} deviation were utilized for conforming to the measurements. Statistical comparisons of inter-group differences were made by non-parametric test of paired samples. ResultsRenal partial response (PR) and complete response (CR) were defined according to the criteria of Kidney Disease: Improving Global Outcomes (KDIGO). Four cases achieved PR at Month 1. There were PR (n=6) and CR (n=1) at Month 6 and PR (n=1) and CR (n=7) at Month 14. PR+CR remission rate of 80%. Safety: 7 cases had no infusion reaction, 3 cases developed infusion reaction after an initial infusion of OBZ and all of them successfully completed treatment. Adverse reactions: 3 cases had complications of infection within 1 month after OBZ dosing. However, infections were cured. ConclusionOBZ offers better therapeutic effect on PLA2R-associated MN with fewer adverse reactions and better safety.

Offshore wind farms are becoming increasingly distant from onshore centralized control centers, and the communication delays between them inevitably introduce time delays in the measurement signal of the primary frequency control. This causes a deterioration in the performance of the primary frequency control and, in some cases, may even result in frequency instability within the power system. Therefore, a frequency response model that incorporates communication delays was established for power systems that integrate offshore wind power. The Padé approximation was used to model the time delays, and a linearized frequency response model of the power system was derived to investigate the frequency stability under different time delays. The influences of the wind power proportion and frequency control parameters on the system frequency stability were explored. In addition, a Smith delay compensation control strategy was devised to mitigate the effects of communication delays on the system frequency dynamics. Finally, a power system incorporating offshore wind power was constructed using the MATLAB/Simulink platform. The simulation results demonstrate the effectiveness and robustness of the proposed delay compensation control strategy.

Abstract The aim of this study is to evaluate the developments in the treatment and prevalence of hypertension by demographic subgroups in least developed area of China in 2012 and 2022. This population‐based cross‐sectional study was conducted in 2012 and 2022, we applied stratified multistage random sampling to investigate residents aged 18 years or older in Gansu, the least developed province in the northwest of China. Questionnaires and anthropometric measurements were given to all respondents. The standardized prevalence of hypertension in adults in Gansu increased from 26.1% in 2012 to 28.8% in 2022. Compared with 2012, the control rate remains decreased despite the significantly improved awareness and treatment rates of hypertension in 2022. Apart from the reversal of the control rate, the trend of higher prevalence in men and higher awareness and treatment rates in women has not changed. There was an obviously increase in the proportion of participants who had received health education and hypertension management services from medical workers. The treatment was still primarily monotherapy, and there was no significant improvement in the prescription of medication. The prevalence of hypertension has increased mildly in the least developed region of China over the past decade, and the challenge of hypertension management has shifted from increasing awareness and treatment rates to increasing control rates. The onset and control of hypertension are affected by education methods, BMI, local economic conditions and other factors, and targeted strategies can be adopted to strengthen the management of hypertension in economically underdeveloped areas of China.

Abstract With an increasing demand for environmentally friendly refrigeration, the solid‐state refrigeration based on the electrocaloric effect (ECE) has been drawn extensive attention. It is a challenge to maintain a large adiabatic temperature change (∆T) over a broad temperature span. Herein, the authors designed and synthesised (0.74‐x) Na0.5Bi0.5TiO3‐0.06BaTiO3‐0.2SrTiO3‐xBi(Mg0.5Zr0.5)O3 (abbreviated as NBT‐xBMZ) (x = 0, 0.02, 0.04, 0.06 and 0.08) lead‐free relaxor ferroelectrics. Their microstructures, dielectric properties, ferroelectric properties, ECEs and the structure‐property relationships were investigated. Via doping with BMZ, an enhanced relaxor feature and a wider temperature range where multi‐phases coexist were achieved. The relaxor ferroelectric characteristics were illustrated using the Vogel‐Fulcher relation. The indirectly calculated ECE results showed that the optimal ΔT of 1.11 K was obtained for the x = 0.02 sample at 90°C and 70 kV/cm over a wide Tspan of 120°C, providing a potential ECE material. The direct ECE results procured using thermocouple indicated that the maximal ∆T of 2.14 K and ∆T/∆E of 0.31 K m/MV were achieved in the same sample at 70°C and 7 MV/m and the variation trend of ECE results was consistent with the indirect results. Moreover, the multi‐phases coexistent strategy can be extended to other materials system to generate a large ΔT over a wide temperature range.

Abstract In prior investigations, a correlation was established between patient outcomes in locally advanced non-small cell lung cancer (LA-NSCLC) following thoracic irradiation and parameters, such as pre/post-treatment neutrophil-to-lymphocyte ratio (NLR) and NLR change (ΔNLR). However, these parameters could potentially be influenced by radiation-related variables, such as gross tumor volume (GTV). The primary aim of this study was to elucidate the factors impacting post-treatment NLR and ΔNLR and to further assess their prognostic relevance. In this retrospective study, a cohort of 188 LA-NSCLC patients who underwent thoracic radiation between 2012 and 2017 was assessed. The calculation of pre/post-treatment NLR involved the use of absolute neutrophil and lymphocyte counts. ΔNLR was defined as the difference between post- and pre-treatment NLR values. To assess the relationships between various variables and overall survival (OS), local progression-free survival (LPFS), and distant metastasis-free survival (DMFS), the Kaplan–Meier technique and Cox proportional hazards regression were employed. Additionally, Spearman’s rank correlation analysis was carried out to investigate correlations between the variables. The analysis revealed that both post-treatment NLR (r = 0.315, P 67.41 cm3 group (P = 0.028 and P = 0.042, respectively), highlighting ΔNLR as the sole independent predictive factor for survival and metastasis, irrespective of GTV.

Abstract According to the urgent low latency and the heavy computation tasks demands required for sixth‐generation (6G) wireless networks, the authors introduce the conventional resource allocation algorithms, including the game theory, artificial‐intelligence (AI) methods, and matching theory enabled framework, in which the multi‐access edge computing (MEC) scheme collaborative with the cloud platform to serve the primary users (PUs) and cognitive users (CUs) for next generation multiple access (NGMA). The proposed framework allows both the PUs and CUs to offload their computation tasks in a 6G‐enabled cognitive radio (CR) networks, so called cloud‐assisted CR‐MEC networks. In particular, the fundamentals of this conceived networks based on NGMA are first introduced. Hence, a number of methods based on the resource allocation algorithms are proposed in order to improve the quality of service for the mobile users, and reduce their transmission latency as well as the energy consumptions. Moreover, the motivations, challenges, and representative models for these conventional algorithms are described for integrated‐intelligent communication and computing aided NGMA networks. Furthermore, the open issues and future research directions for this conceived networks are summarised.

BackgroundStress influences examination performance among Sri Lankan students. Validated tests are required to evaluate stress levels among elementary students in Sri Lanka. Therefore, the Perceived Stress Questionnaire 8–11 (PSQ8-11) was translated into a Sinhala version. The aim of this study was to examine the psychometric properties of the translated and adapted scale among elementary level school children in Sri Lanka and examine invariance across male and female children.MethodsThe participants were 1021 students from seven schools. After removing missing values, responses from 693 students (mean age = 9.65 ± 0.478 years, 51.8% male) were analysed for participant characteristics. Cronbach’s alpha, Spearman’s correlation, and confirmatory factor analysis with measurement invariance models were conducted after adding one item to the original PSQ8-11 version.ResultsThe Cronbach’s alpha value for the 20-item modified PSQ8-11 Sinhala version was.788. The two subscales, psychological stress (Cronbach’s alpha = 0.615) and physiological stress (Cronbach’s alpha = 0.711), indicated a satisfactory level of internal consistency. Furthermore, a statistically significant correlation (p < 0.01; 2-tailed) was reported among each of the subscales. Confirmatory factor analyses demonstrated a satisfactory goodness-of-fit across the two models by confirming the theoretical constructs of the PSQ8-11 translated version with its two subscales. The two-factor model has better model fit indices compared to the unidimensional model (χ2/df = 1.447, CFI = 0.947, TLI = 0.938, WRMR = 0.028, RMSEA = 0.026, SRMSR = 0.0341, and PCLOSE = 1 of the two-factor model). Measurement variance across gender was supported by the establishment of configural and metric invariances.ConclusionAcceptable psychometric properties for the PSQ8-11 Sinhala version were observed in elementary schoolers in Sri Lanka.

Abstract Renal tubular epithelial cells (TECs) undergo an energy‐related metabolic shift from fatty acid oxidation to glycolysis during chronic kidney disease (CKD) progression. However, the mechanisms underlying this burst of glycolysis remain unclear. Herein, a new critical glycolysis regulator, the transcription factor forkhead box protein K1 (FOXK1) that is expressed in TECs during renal fibrosis and exhibits fibrogenic and metabolism‐rewiring capacities is reported. Genetic modification of the Foxk1 locus in TECs alters glycolytic metabolism and fibrotic lesions. A surge in the expression of a set of glycolysis‐related genes following FOXK1 protein activation contributes to the energy‐related metabolic shift. Nuclear‐translocated FOXK1 forms condensate through liquid‐liquid phase separation (LLPS) to drive the transcription of target genes. Core intrinsically disordered regions within FOXK1 protein are mapped and validated. A therapeutic strategy is explored by targeting the Foxk1 locus in a murine model of CKD by the renal subcapsular injection of a recombinant adeno‐associated virus 9 vector encoding Foxk1‐short hairpin RNA. In summary, the mechanism of a FOXK1‐mediated glycolytic burst in TECs, which involves the LLPS to enhance FOXK1 transcriptional activity is elucidated.

Abstract Aortic aneurysm (AA) is an aortic disease with a high mortality rate, and other than surgery no effective preventive or therapeutic treatment have been developed. The renin–angiotensin system (RAS) is an important endocrine system that regulates vascular health. The ACE2/Ang‐(1–7)/MasR axis can antagonize the adverse effects of the activation of the ACE/Ang II/AT1R axis on vascular dysfunction, atherosclerosis, and the development of aneurysms, thus providing an important therapeutic target for the prevention and treatment of AA. However, products targeting the Ang‐(1–7)/MasR pathway still lack clinical validation. This review will outline the epidemiology of AA, including thoracic, abdominal, and thoracoabdominal AA, as well as current diagnostic and treatment strategies. Due to the highest incidence and most extensive research on abdominal AA (AAA), we will focus on AAA to explain the role of the RAS in its development, the protective function of Ang‐(1–7)/MasR, and the mechanisms involved. We will also describe the roles of agonists and antagonists, suggest improvements in engineering and drug delivery, and provide evidence for Ang‐(1–7)/MasR's clinical potential, discussing risks and solutions for clinical use. This study will enhance our understanding of AA and offer new possibilities and promising targets for therapeutic intervention.

ObjectivesConsidering the importance of mental health help-seeking, researchers have closely examined the relationship between mental health literacy (MHL) and help-seeking intention (HSI). Furthermore, the high impact of stigma and the potential value of social support on HSI have been recognised. However, the relationship between these variables has not been fully tested within the context of Chinese elite athletes. This study addressed this gap by examining the relationship between MHL and HSI. IntroductionThe quality of traditional Chinese medicine is based on the content of their secondary metabolites, which vary with habitat adaptation and ecological factors. This study focuses on Lonicera japonica Flos (LJF), a key traditional herbal medicine, and aims to evaluate how ecological factors impact its quality.MethodsWe developed a new evaluation method combining high-performance liquid chromatography (HPLC) fingerprinting technology and MaxEnt models to assess the effects of ecological factors on LJF quality. The MaxEnt model was used to predict suitable habitats for current and future scenarios, while HPLC was employed to analyze the contents of key compounds. We also used ArcGIS for spatial analysis to create a quality zoning map.ResultsThe analysis identified 21 common chromatographic peaks, with significant variations in the contents of Hyperoside, Rutin, Chlorogenic acid, Cynaroside, and Isochlorogenic acid A across different habitats. Key environmental variables influencing LJF distribution were identified, including temperature, precipitation, and elevation. The current suitable habitats primarily include regions south of the Yangtze River. Under future climate scenarios, suitable areas are expected to shift, with notable expansions in southern Gansu, southeastern Tibet, and southern Liaoning. The spatial distribution maps revealed that high-quality LJF is predominantly found in central and southern Hebei, northern Henan, central Shandong, central Sichuan, southern Guangdong, and Taiwan.DiscussionThe study indicates that suitable growth areas can promote the accumulation of certain secondary metabolites in plants, as the accumulation of these metabolites varies. The results underscore the necessity of optimizing quality based on cultivation practices. The integration of HPLC fingerprinting technology and the MaxEnt model provides valuable insights for the conservation and cultivation of herbal resources, offering a new perspective on evaluating the impact of ecological factors on the quality of traditional Chinese medicines.

Abstract Background COVID-19 has been shown to increase the risk of extracorporeal coagulation during hemodialysis in patients, but the underlying mechanism remains unclear. This study aimed to investigate the effect and mechanism of COVID-19 on the risk of extracorporeal coagulation in patients with chronic kidney disease undergoing hemodialysis. Methods A retrospective analysis of the extracorporeal coagulation status of 339 hemodialysis patients at our center before and after COVID-19 infection was performed, including subgroup analyses. Post-infection blood composition was analyzed by protein spectrometry and ELISA. Results Compared to the pre-COVID-19 infection period, COVID-19-induced extracorporeal coagulation predominantly occurred in patients with severe/critical symptoms. Further proteomic analysis demonstrated that in patients with severe/critical symptoms, the coagulation cascade reaction, platelet activation, inflammation, and oxidative stress-related pathways were significantly amplified compared to those in patients with no/mild symptoms. Notably, the vWF/FBLN5 pathway, which is associated with inflammation, vascular injury, and coagulation, was significantly upregulated. Conclusions Patients with severe/critical COVID-19 symptoms are at a higher risk of extracorporeal coagulation during hemodialysis, which is associated with the upregulation of the vWF/FBLN5 signaling pathway. These findings highlight the importance of early anticoagulant therapy initiation in COVID-19 patients with severe/critical symptoms, particularly those undergoing hemodialysis. Additionally, vWF/FBLN5 upregulation may be a novel mechanism for virus-associated thrombosis/coagulation.

Abstract Background Chat Generative Pretrained Transformer (ChatGPT), a generative artificial intelligence chatbot, may have broad applications in healthcare delivery and patient education due to its ability to provide human-like responses to a wide range of patient queries. However, there is limited evidence regarding its ability to provide reliable and useful information on orthopaedic procedures. This study seeks to evaluate the accuracy and relevance of responses provided by ChatGPT to frequently asked questions (FAQs) regarding total knee replacement (TKR). Methods A list of 50 clinically-relevant FAQs regarding TKR was collated. Each question was individually entered as a prompt to ChatGPT (version 3.5), and the first response generated was recorded. Responses were then reviewed by two independent orthopaedic surgeons and graded on a Likert scale for their factual accuracy and relevance. These responses were then classified into accurate versus inaccurate and relevant versus irrelevant responses using preset thresholds on the Likert scale. Results Most responses were accurate, while all responses were relevant. Of the 50 FAQs, 44/50 (88%) of ChatGPT responses were classified as accurate, achieving a mean Likert grade of 4.6/5 for factual accuracy. On the other hand, 50/50 (100%) of responses were classified as relevant, achieving a mean Likert grade of 4.9/5 for relevance. Conclusion ChatGPT performed well in providing accurate and relevant responses to FAQs regarding TKR, demonstrating great potential as a tool for patient education. However, it is not infallible and can occasionally provide inaccurate medical information. Patients and clinicians intending to utilize this technology should be mindful of its limitations and ensure adequate supervision and verification of information provided.

Abstract In the last decade, prophages that possess the ability of lysogenic transformation have become increasingly significant. Their transfer and subsequent activity in the host have a significant impact on the evolution of bacteria. Here, we investigate the role of prophage phi456 with high spontaneous induction in the bacterial genome of Avian pathogenic Escherichia coli (APEC) DE456. The phage particles, phi456, that were released from DE456 were isolated, purified, and sequenced. Additionally, phage particles were no longer observed either during normal growth or induced by nalidixic acid in DE456Δphi456. This indicated that the released phage particles from DE456 were only phi456. We demonstrated that phi456 contributed to biofilm formation through spontaneous induction of the accompanying increase in the eDNA content. The survival ability of DE456Δphi456 was decreased in avian macrophage HD11 under oxidative stress and acidic conditions. This is likely due to a decrease in the transcription levels of three crucial genes—rpoS, katE, and oxyR—which are needed to help the bacteria adapt to and survive in adverse environments. It has been observed through animal experiments that the presence of phi456 in the DE456 genome enhances colonization ability in vivo. Additionally, the number of type I fimbriae in DE456Δphi456 was observed to be reduced under transmission electron microscopy when compared to the wild-type strain. The qRT-PCR results indicated that the expression levels of the subunit of I fimbriae (fimA) and its apical adhesin (fimH) were significantly lower in DE456Δphi456. Therefore, it can be concluded that phi456 plays a crucial role in helping bacterial hosts survive in unfavorable conditions and enhancing the colonization ability in DE456.

Abstract Recently, innate immunity and inflammation were recognized as the key factors for acute kidney injury (AKI) caused by sepsis, which is closely related to high mortality. Stimulator of interferon genes (STING) has emerged as a critical component of innate immune and inflammatory responses. However, the role of STING in the pathogenesis of septic AKI remains unclear. This study demonstrated that the STING was significantly activated in tubular cells induced by lipopolysaccharide (LPS) in vivo and in vitro. Tubule-specific STING knockout attenuated LPS-induced renal dysfunction and pathological changes. Mechanistically, the STING pathway promotes NOD-like receptor protein 3 (NLRP3) activation. STING triggers endoplasmic reticulum (ER) stress to induce mitochondrial reactive oxygen species (mtROS) overproduction, enhancing thioredoxin-interacting protein activation and association with NLRP3. Eventually, the NLRP3 inflammasome leads to tubular cell inflammation and pyroptosis. This study revealed the STING-regulated network and further identified the STING/ER stress/mtROS/NLRP3 inflammasome axis as an emerging pathway contributing to tubular damage in LPS-induced AKI. Hence, targeting STING may be a promising therapeutic strategy for preventing septic AKI.

Abstract This article focuses on the cascading failure problem and node importance evaluation method in complex networks. To address the issue of identifying important nodes in dynamic networks, the method used in static networks is introduced and the necessity of re-evaluating node status during node removal is proposed. Studies have found that the methods for identifying dynamic and static network nodes are two different directions, and most literature only uses dynamic methods to verify static methods. Therefore, it is necessary to find suitable node evaluation methods for dynamic networks. Based on this, this article proposes a method that integrates local and global correlation properties. In terms of global features, we introduce an improved k-shell method with fusion degree to improve the resolution of node ranking. In terms of local features, we introduce Solton factor and structure hole factor improved by INCC (improved network constraint coefficient), which effectively improves the algorithm’s ability to identify the relationship between adjacent nodes. Through comparison with existing methods, it is found that the KPDN–INCC method proposed in this paper complements the KPDN method and can accurately identify important nodes, thereby helping to quickly disintegrate the network. Finally, the effectiveness of the proposed method in identifying important nodes in a small-world network with a random parameter less than 0.4 was verified through artificial network experiments.

Abstract Dazomet is a dry powder formulation that releases toxic gas containing methyl isothiocyanate, which controls soil-borne pests and weeds, improving crop yields when applied to moist soils. To explore the efficacy of dazomet fumigation in the cultivation of the perennial herb Codonopsis pilosula, four typical cultivars (G1, G2, W1 and TCK) in Gansu Province were selected for seedling cultivation after soil fumigation (F) by dazomet, and non-fumigated soil was used as a control (CK). The experiments took 2 years to complete. The functional diversity of the soil enzymes and microorganisms, seedling emergence and physiological characteristics, and the quality and yield of Codonopsis seedlings and Radix were assessed. The results showed that the seed emergence rate, seedling re-green rate and several antioxidant enzymatic activities improved in the treatments involving soil fumigation with dazomet, and membrane lipid peroxidation in the seedlings decreased. On average, compared with those of the respective controls, the root viability and yield of the seedlings of the tested cultivars also increased by 34.87% and 42.4%, respectively, and the incidence of root rot in the seedlings was reduced by 83.9%, compared with their respective controls. After harvest, the yield increased by 23.9%, the incidence of root rot decreased by 61.3%, increase in yield and a 61.3% reduction in incidence, and the medicinal materials were determined to be safe and residue-free. The effects of fumigation were cultivar-specific and were especially prominent in G2. Therefore, soil fumigation with dazomet could improve the quality and productivity of Codonopsis pilosula seedlings. Taken together, these findings suggest that when herbs are bred by seedling transplantation, especially cultivars of good quality but poor resistance or species with rare germplasm resources, soil fumigation provides a way to improve cultivation effectiveness and, more importantly, ensures the probability of successfully breeding the species.

Abstract Although brood parasitism has been well documented among bird species, interspecific alloparenting, which is parenting behavior of adult individuals of one species toward the progeny of another species, is increasingly being reported. However, compared with the many reports of interspecific alloparenting behavior in North America and Europe, this phenomenon is less well known in China, with only two prior cases of interspecific alloparenting behavior in birds having been recorded. On June 23, 2022, we observed an instance of interspecific alloparental care provided by a mountain bulbul (Ixos mcclellandii) towards silver‐eared mesia (Leiothrix argentauris) nestlings in Caihu Village, Jingdong County, Yunnan Province, southwestern China. We recorded 19.5 h of footage during the period in which the mountain bulbul provided care for the nestlings with the aim of documenting detailed observations of interspecific alloparenting to contribute to our overall understanding of this behavior. The alloparenting behavior of the mountain bulbul lasted for at least 5 days. During this period, both silver‐eared mesia parents fed their nestlings 157 times and removed their nestlings' fecal sacs 5 times, while the mountain bulbul fed the nestlings 30 times and removed the nestlings' fecal sacs 4 times. In addition, the male silver‐eared mesia parent chased the mountain bulbul away during nestling feeding. As there was no life history information for the mountain bulbul at that time, we were unable to directly determine why it exhibited interspecific alloparental care. Regardless of the reason for the mountain bulbul's behavior, these findings provide valuable information for future studies on the reproductive ecology of these two bird species.

Abstract In 2017, one pair of Oriental reed warblers (Acrocephalus orientalis) with color aberrations was found in Yongnianwa National Wetland Park, Hebei, China. The female bird exhibited white feathers on the head, neck, and upper back, and the base of the beak was flesh‐red in color. The male had a few feathers on the outer edges of the left and right primary wing coverts that were white, which was determined to be leucism after analysis. The breeding pairs laid their first egg on May 29, with a clutch size of four eggs. After an incubation period of 13 days, two chicks hatched on June 13, 2017. The nest was found empty on June 20 when the chicks were 7 days old and before fledging age; therefore, it was presumed that the chicks had been predated. A white parrot egg was added to the nest during the incubation period to test the egg recognition ability of breeding pairs and was successfully rejected. To the best of our knowledge, this is the first report of color aberrations in the Oriental reed warbler, and we found that this color aberrations did not affect some reproductive and antiparasitic behaviors of the birds, but whether it affects their breeding success needs to be further studied.

Leukemia is a malignant tumor of the hematologic system. Studies have shown that cernuumolide J (TMJ-105), an extract of Carpesium cernuum, has anti-cancer effects, but the underlying mechanism is unclear. In this study, we investigated the effect of TMJ-105 on the proliferation of human leukemia HEL cells and its molecular mechanism. MTT analysis showed TMJ-105 had revealed that it shows significant IC50 in HEL cells at lower doses (1.79 ± 0.29 μmol/L) than in K562 cells (3.89 ± 0.80 μmol/L), and the suppression of HEL cell proliferation was time- and concentration-dependent. Meanwhile, TMJ-105 induced G2/M phase blockage, leading to DNA damage in HEL cells. TMJ-105 promoted HEL cells to release of reactive oxygen species (ROS) and changed mitochondrial membrane potential (MMP). Furthermore, TMJ-105 induced apoptosis by upregulating the cleaved-caspase9 and cleaved-caspase3 protein expression, while caspase pan inhibitor (Z-VAD-FMK) blocked the inhibition effect. Finally, TMJ-105 downregulated the phosphorylation of JAK2, STAT3 and Erk, and activated the phosphorylation of JNK and p38. Collectively, these results demonstrated that TMJ-105 inhibited proliferation of leukemia cells and the underlying mechanism via the JAK2/STAT3 axis and MAPKs signaling pathway. Based on these results, the present study suggested the sesquiterpene lactone TMJ-105 is a new chemotherapeutic agent for the treatment of leukemia.

Summary: ADAR1-mediated RNA editing establishes immune tolerance to endogenous double-stranded RNA (dsRNA) by preventing its sensing, primarily by MDA5. Although deleting Ifih1 (encoding MDA5) rescues embryonic lethality in ADAR1-deficient mice, they still experience early postnatal death, and removing other MDA5 signaling proteins does not yield the same rescue. Here, we show that ablation of MDA5 in a liver-specific Adar knockout (KO) murine model fails to rescue hepatic abnormalities caused by ADAR1 loss. Ifih1;Adar double KO (dKO) hepatocytes accumulate endogenous dsRNAs, leading to aberrant transition to a highly inflammatory state and recruitment of macrophages into dKO livers. Mechanistically, progranulin (PGRN) appears to mediate ADAR1 deficiency-induced liver pathology, promoting interferon signaling and attracting epidermal growth factor receptor (EGFR)+ macrophages into dKO liver, exacerbating hepatic inflammation. Notably, the PGRN-EGFR crosstalk communication and consequent immune responses are significantly repressed in ADAR1high tumors, revealing that pre-neoplastic or neoplastic cells can exploit ADAR1-dependent immune tolerance to facilitate immune evasion.

Building energy flexibility plays a critical role in demand-side management for reducing utility costs for building owners and sustainable, reliable, and smart grids. Realizing building energy flexibility in tropical regions requires solar photovoltaics and energy storage systems. However, quantifying the energy flexibility of buildings utilizing such technologies in tropical regions has yet to be explored, and a robust control sequence is needed for this scenario. Hence, this work presents a case study to evaluate the building energy flexibility controls and operations of a net-zero energy office building in Singapore. The case study utilizes a data-driven energy flexibility quantification workflow and employs a novel data-driven model predictive control (MPC) framework based on the physically consistent neural network (PCNN) model to optimize the building energy flexibility. To the best of our knowledge, this is the first instance that PCNN is applied to a mathematical MPC setting, and the stability of the system is formally proved. Three scenarios are evaluated and compared: the default regulated flat tariff, a real-time pricing mechanism, and an on-site battery energy storage system (BESS). Our findings indicate that incorporating real-time pricing into the MPC framework could be more beneficial to leverage building energy flexibility for control decisions than the flat-rate approach. Moreover, adding BESS to the on-site PV generation improved the building self-sufficiency and the PV self-consumption by 17% and 20%, respectively. This integration also addresses model mismatch issues within the MPC framework, thus ensuring a more reliable local energy supply. Future research can leverage the proposed PCNN-MPC framework for different data-driven energy flexibility quantification types.

BackgroundIt is known that 24-hour movement behaviors, including physical activity (PA), sedentary behavior (SB), and sleep, are crucial components affecting older adults’ health. Canadian 24-hour movement guidelines for older adults were launched in 2020, emphasizing the combined role of these 3 movement behaviors in promoting older adults’ health. However, research on the prevalence and correlates of guideline adherence and its associations with health-related outcomes is limited, especially among Chinese older adults. ObjectiveThis study aimed to investigate the prevalence and correlates of meeting 24-hour movement guidelines among Chinese older adults. Furthermore, this study aimed to examine the associations of guideline adherence with older adults’ physical and mental health outcomes. MethodsUsing a stratified cluster random sampling approach, a total of 4562 older adults (mean age 67.68 years, SD 5.03 years; female proportion: 2544/4562, 55.8%) were recruited from the latest provincial health surveillance of Hubei China from July 25 to November 19, 2020. Measures included demographics, movement behaviors (PA, SB, and sleep), BMI, waist circumference, waist-hip ratio (WHR), percentage body fat (PBF), systolic and diastolic blood pressure, physical fitness, depressive symptoms, and loneliness. Generalized linear mixed models were employed to examine the associations between variables using SPSS 28.0 (IBM Corp). ResultsOnly 1.8% (83/4562) of participants met all 3 movement guidelines, while 32.1% (1466/4562), 3.4% (155/4562), and 66.4% (3031/4562) met the individual behavioral guidelines for PA, SB, and sleep, respectively. Participants who were older, were female, and lived in municipalities with lower economic levels were less likely to meet all 3 movement guidelines. Adhering to individual or combined movement guidelines was associated with greater physical fitness and lower values of BMI, waist circumference, WHR, PBF, depressive symptoms, and loneliness, with the exception of the relationship of SB+sleep guidelines with loneliness. Furthermore, only meeting SB guidelines or meeting both PA and SB guidelines was associated with lower systolic blood pressure. ConclusionsThis is the first study to investigate adherence to 24-hour movement guidelines among Chinese older adults with regard to prevalence, correlates, and associations with physical and mental health outcomes. The findings emphasize the urgent need for promoting healthy movement behaviors among Chinese older adults. Future interventions to improve older adults’ physical and mental health should involve enhancing their overall movement behaviors and should consider demographic differences.

BackgroundIn the current digital era, eHealth literacy plays an indispensable role in health care and self-management among older adults with noncommunicable diseases (NCDs). Measuring eHealth literacy appropriately and accurately ensures the successful implementation and evaluation of pertinent research and interventions. However, existing eHealth literacy measures focus mainly on individuals’ abilities of accessing and comprehending eHealth information (Web1.0), whereas the capabilities for web-based interaction (Web2.0) and using eHealth information (Web3.0) have not been adequately evaluated. ObjectiveThis study aimed to examine the reliability, validity, and measurement invariance of the eHealth Literacy Scale-Web3.0 (eHLS-Web3.0) among older adults with NCDs. MethodsA total of 642 Chinese older adults with NCDs (mean age 65.78, SD 3.91 years; 55.8% female) were recruited in the baseline assessment, of whom 134 (mean age 65.63, SD 3.99 years; 58.2% female) completed the 1-month follow-up assessment. Baseline measures included the Chinese version of the 24-item 3D eHLS-Web3.0, the Chinese version of the 8-item unidimensional eHealth Literacy Scale (eHEALS), and demographic information. Follow-up measures included the 24-item eHLS-Web3.0 and accelerometer-measured physical activity and sedentary behavior. A series of statistical analyses, for example, Cronbach α, composite reliability coefficient (CR), confirmatory factor analysis (CFA), and multigroup CFA, were performed to examine the internal consistency and test-retest reliabilities, as well as the construct, concurrent, convergent, discriminant, and predictive validities, and the measurement invariance of the eHLS-Web3.0 across gender, education level, and residence. ResultsCronbach α and CR were within acceptable ranges of 0.89-0.94 and 0.90-0.97, respectively, indicating adequate internal consistency of the eHLS-Web3.0 and its subscales. The eHLS-Web3.0 also demonstrated cross-time stability, with baseline and follow-up measures showing a significant intraclass correlation of 0.81-0.91. The construct validity of the 3D structure model of the eHLS-Web3.0 was supported by confirmatory factor analyses. The eHLS-Web3.0 exhibited convergent validity with an average variance extracted value of 0.58 and a CR value of 0.97. Discriminant validity was supported by CFA results for a proposed 4-factor model integrating the 3 eHLS-Web3.0 subscales and eHEALS. The predictive validity of the eHLS-Web3.0 for health behaviors was supported by significant associations of the eHLS-Web3.0 with light physical activity (β=.36, P=.004), moderate to vigorous physical activity (β=.49, P

ABSTRACT: Acoustic characteristics reflect male quality and play a role in female mate choice. Thus, the frequency of vocalizations and temporal characteristics are often related to body size within and across species. However, it is less clear whether acoustic features can reveal information about individual quality in the domestic chicken (Gallus gallus domesticus) populations. Here, we investigated the relationship between morphological parameters and acoustic features in male and female free-ranged domestic chickens in Liuzhi, Guizhou, southwest China, and further examined whether acoustic characteristics correlate with internal organs, including the heart, liver, testis, and spleen in male chickens, and whether the cackling call of females indicates body size and mass. We found that both male and female chickens differ significantly in their morphological parameters; however, based on acoustic parameters, they only differ in high frequency. Morphological parameters displayed no relationship with the frequency and duration of calls in both male and female chickens. Furthermore, none of the frequency or temporal parameters of the calls we studied were related to the internal body parameters of males.

Abstract Avian brood parasites depend upon locating host nests to lay their eggs. However, how brood parasites locate host nests and select the nests for parasitism remains poorly studied. Here, we examined how a non‐evicting brood parasite, the Asian koel (Eudynamys scolopaceus) were able to locate host nests in locally novel nest sites. We provided a set of novel nest sites (i.e., nestboxes, n = 100) to the common myna (Acridotheres tristis), a regular host of the Asian koel, at a field site in Central Bangladesh. We found that common myna quickly utilized the novel breeding sites (n = 99 nests across 59 boxes) and similarly, 21.2% of these nests were parasitized by the Asian koel. Unsurprisingly, none of the inactive nest boxes were parasitized, neither were empty nests of common myna (i.e., either predated or fledged clutches, thus lack of parental activity, n = 83). Our results provide an experimental demonstration of the Asian koel ability to rapidly locate nests in locally novel nest sites, potentially facilitated by close monitoring of host activity and behavior around the nest site. However, the underlying mechanisms for this rapid adjustment to novel nest sites remain unresolved.

ObjectivesThis study aimed to examine the effectiveness of a specifically designed mindfulness-acceptance-insight-commitment (MAIC) training program on relevant psychological factors (i.e., mindfulness, acceptance, performance-related satisfaction) as well as sport training performance for elite adolescent athletes from Hong Kong. And it also aimed to explore the athletes’ real experiences (i.e., receptiveness and perceptions) of completing the MAIC program.Methods and designThe mixed-method was used in this study, including a randomized controlled trial (RCT) and a qualitative exploration. The RCT employed a 2 (groups) x 3 (data collection points) design involving 40 elite adolescent athletes from the Hong Kong Sports Institute (HKSI). These athletes were randomly assigned to either the MAIC training group (MT; n = 20, Mage = 15.65) or the control group (CG; n = 20, Mage = 15.85) to further test the effectiveness of the MAIC intervention on mindfulness, acceptance, performance-related satisfaction, and sport training performance. Subsequent to the RCT, the qualitative exploration was used to explore the athletes’ real experiences towards the MAIC program. In the qualitative exploration, all athletes who participated in the MAIC program were invited to participate in voluntary semi-structured interviews. Of these, 14 athletes chose to take part in the interviews. The RCT employed a 2×3 mixed-design ANOVA, while thematic analysis was applied to the qualitative exploration.ResultsThe results revealed that the MAIC training program significantly enhanced athletes’ mindfulness, acceptance, satisfaction with performance, and sport training performance. However, these effects diminished at the follow-up assessment compared to post-training. Notably, the acceptance level of MT athletes did not significantly differ from CG athletes at the follow-up assessment. Additionally, the qualitative analysis identified four key dimensions: (a) Attitude towards MAIC training, (b) Reflection on the MAIC learning process, (c) Outcomes of MAIC training, and (d) Recommendations for future MAIC training. Overall, the qualitative findings complemented and reinforced the quantitative results, offering deeper insights into athletes’ experiences and valuable suggestions for further enhancing the MAIC program.ConclusionThe findings suggested that the specifically designed MAIC training program in this study effectively enhanced sport training performance and various psychological factors among elite adolescent athletes from Hong Kong. Nevertheless, further investigations are still required to comprehensively evaluate and further develop the MAIC training program.

BackgroundThe spread of Covid-19 and resultant infection prevention strategies have disturbed the life routine of Chinese young adults, led to reduced physical activity (PA), prolonged screen time (ST) and inadequate sleep duration (SP), and made immense influence on their mental health (MH) and quality of life (QoL). E-Health literacy (EHL) can enable citizens to use available online information to respond to the highly complex information environment and make appropriate health decisions.ObjectiveThis study aims to examine associations between adhering to 24-h movement (24HM) guidelines and MH and QoL among young adults, as well as to identify any mediating or moderating role of EHL in these associations.Methods1742 young adults (20.03 ± 1.54 years old, 68.6% females) from north and south China completed self-report measures of 24HM (PA, ST and SP), health indicators (MH and QoL), EHL and demographic information through an online survey between 4 Apr and 16 Jun 2022. Generalized linear mixed models were applied for data analysis.ResultsResults showed that adhering to PA, ST and SP guidelines were all positively connected with QoL while MH was associated with adhering ST or SP guidelines. Adhering to more of 24HM guidelines was linked to better MH and QoL. EHL significantly mediated the association of guideline adherence and QoL and moderate that of guideline adherence and MH.ConclusionThis is the first study to investigate the role of EHL on the associations between 24HM and MH as well as QoL during the Covid-19. The findings may contribute to further empirical research or intervention that aims to promote MH or QoL among young adults more effectively or provide valuable references for developing relevant strategies or policy of health promotion or public health events in China.

The pharmacodynamic substances in “Scrophulariae Radix–Fritillaria” and the molecular mechanisms underlying its therapeutic effects against goiter were analyzed through metabolomics and serum pharmaco-chemistry. A rat model of goiter was established using propylthiouracil (PTU), and the animals were treated using “Scrophulariae Radix–Fritillaria.” The efficacy of the drug pair was evaluated in terms of thyroid gland histopathology and blood biochemical indices. Serum and urine samples of the rats were analyzed by UPLC-Q-TOF/MS. Principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) were performed to screen potential biomarkers in urine and the corresponding metabolic pathways. The blood components of “Scrophulariae Radix–Fritillaria” were also identified, and their correlation with urine biomarkers was analyzed in order to screen for potential bioactive compounds. “Scrophulariae Radix–Fritillaria” mitigated injury to thyroid tissues and normalized the levels of the thyroid hormones FT3, FT4, and TSH. We also identified 22 urine biomarkers related to goiter, of which 19 were regulated by “Scrophulariae Radix–Fritillaria.” Moreover, urine biomarkers are involved in tryptophan metabolism, steroid hormone biosynthesis, and beta-alanine metabolism, and these pathways may be targeted by the drug pair. In addition, 47 compounds of “Scrophulariae Radix–Fritillaria” were detected by serum pharmacochemistry, of which nine components, namely, syringic acid, paeonol, cedrol, and cis-ferulic acid, fetisinine, aucubigenin, linolenic acid, ussuriedine, and 5-(methylsulfanyl)pentanenitrile, were identified as potential effective substances against goiter. To summarize, we characterized the chemical components and mechanisms of “Scrophulariae Radix–Fritillaria” involved in the treatment of goiter, and our findings provide an experimental basis for its clinical application.

BackgroundAging individuals are vulnerable to various Noncommunicable Diseases (NCDs). Different behaviors are closely related to a decreased risk of suffering from NCDs: sufficient Physical Activity (PA) (e.g., at least 150 mins Moderate-to-vigorous Physical Activity (MVPA) per week) and a healthy daily diet (e.g., at least five portions of Fruit and Vegetable Intake (FVI), 5–6 taels (189.0–226.8 g) Meat, Fish, Egg and Alternatives (MFEA)). Traditional face-to-face interventions were effective in behavior change. However, it was revealed to be resource-intensive and limited transfer due to poor self-regulation skills outside of face-to-face sessions. Thus, eHealth could be a supplement for older adults outside traditional face-to-face settings. The blended approach combining these two interventions might optimize the intervention effects on lifestyle behavior initiation and maintenance, but little research can be found among Hong Kong older adults. Therefore, the study aims to test a blended intervention to promote PA, diet, and health outcomes among Hong Kong community-dwelling older adults.MethodsThis study will adopt a 10-week three-arm randomized controlled trial. The blended group will receive weekly (1) two 60-min face-to-face sessions with one for PA and one for diet, and (2) two web-based sessions with one for PA and one for diet. The face-to-face group will receive the same intervention content as the face-to-face sessions in the blended group. The control condition will receive a biweekly telephone call. The outcomes will include MVPA (minutes/week), FVI (portions/day), MFEA consumption (taels/day), social-cognitive factors (self-efficacy, planning, social support, action control), physical health outcomes (clinical indicators, senior physical fitness), mental health outcomes (depression, loneliness) and health-related quality of life. Data collection will be implemented at the pre-test, post-test, and 3-month follow-up test.DiscussionThis is the first study evaluating a blended intervention promoting multiple health behaviors among Hong Kong community-dwelling older adults. If the effect of the blended intervention is superior to the traditional face-to-face group and the control group, it will enrich lifestyle intervention approaches and can be applied to older adults, helping them obtain health benefits. In this study, the sodium dodecyl sulfate intercalated modified magnesium-aluminum hydrotalcite/sodium alginate/sodium carboxymethylcellulose (modified LDHs/SA/CMC) composite gel spheres were synthesized and their efficacies in adsorbing the cationic dye rhodamine B (RhB) from aqueous solutions were evaluated. The effects of adsorption time, pH and temperature on the adsorption of RhB by spheres were investigated. Remarkably, the modified LDHs/SA/CMC gel spheres achieved adsorption equilibrium after 600 min at 25 °C, and the removal rate of RhB at 60 mg/L reached 91.49 % with the maximum adsorption capacity of 59.64 mg/g. The gel spheres maintained over 80 % efficacy across four adsorption cycles. Kinetic and isotherm analyses revealed that the adsorption of RhB conformed to the secondary kinetic model and the Langmuir isotherm, indicating a spontaneous and exothermic nature of the adsorption process. The adsorption mechanisms of modified LDHs/SA/CMC gel spheres on RhB dyes include electrostatic adsorption, hydrogen bonding and hydrophobic interactions. In conclusion, modified LDHs/SA/CMC gel sphere is a green, simple, recyclable and efficient adsorbent, which is expected to be widely used for the treatment of cationic dye wastewater.

Fault diagnosis holds important significance in mitigating financial losses and ensuring equipment safety. As a crucial aspect of industrial machinery, bearing fault diagnosis becomes imperative. Nevertheless, in reality, identifying faults becomes challenging due to the presence of diverse variations in abnormal data, such as different vibration rotational speeds. In this paper, a novel framework for bearing fault diagnosis is proposed, called DTM-bearing, built upon the diffusion transformation model (DTM). This approach can transfer signals across different vibration speeds into a standardized signal aligned with a speed template. The primary purpose of DTM-bearing is to eliminate speed variations and extract speed-invariant features. Consequently, bolster the robustness of bearing fault diagnosis across diverse vibration speed scenarios. To the best of our knowledge, the proposed method is the first to combine the concepts of diffusion model and transformation in the domain of bearing fault diagnosis. Various experiments are performed on some datasets with multiple different speeds, which shows proposal can effectively improve the performance of bearing diagnosis. The framework based on the diffusion transformation model is expected to eliminate additional variations and improve the effectiveness of bearing diagnosis in practical applications.

Abstract Objective This cross-sectional study examined the socio-ecological factors influencing fundamental motor skills (FMS) in Chinese school-aged children. Methods A total of 1012 parent-child pairs were randomly sampled between March-1st and April-15th, 2022. Based on the socio-ecological model of Children’s FMS, three levels of factors: individual-level (e.g., demographic, physical, psychological, and behavioral characteristics of children), family-level (e.g., caregiver demographics, parental support, and socioeconomic status), and environmental factors (e.g., availability of physical activity equipment) were assessed using self-reported scales (e.g., the Self-perception Profile for Children, the Physical Activity Enjoyment Scale, and the 12-item Psychological Well-Being Scale for Children) and objective measures (e.g., ActiGraph GT3X, the Chinese National Student Physical Fitness Standard, and the Test of Gross Motor Development-Third Edition). Multi-level regression models were employed using SPSS. Results The results demonstrated that children’s age, sex, physical fitness, parental support, and the quality of home and community physical activity environments consistently influenced all three types of FMS, including locomotor, ball, and composite skills. Additionally, seven individual-level factors (children’s age, sex, body mass index, light physical activity, sleep duration, perceived motor competence, and physical fitness) were associated with different types of FMS. Conclusions The findings underscore the multidimensional and complex nature of FMS development, with individual-level factors playing a particularly significant role. Future research should adopt rigorous longitudinal designs, comprehensive assessment tools covering various FMS skills, and objective measurement of parents’ movement behaviors to better understand the strength and direction of the relationship between socio-ecological factors and children’s FMS.

The effect of microstructure and mechanical properties of submicro grained commercial pure titanium (CP–Ti) on the wear mechanism is still unclear. In the present work, CP-Ti samples were subjected to multi-axial forging (MAF) and low-temperature annealing treatment to prepare ultrafine-grained (UFG) material. The mechanical properties, microstructure, and tribological characterization between coarse-grained (CG) sample and UFG samples were comparatively studied. The results showed that the MAF sample could reach a grain size of 165 nm, while its grains coarsened slightly after low-temperature annealing. The MAF sample had the highest strength and hardness and the lowest coefficient of friction, but only a 7.3 % decrease in wear rate compared to the CG sample. The 400 °C annealed MAF sample possessed a good combination of tensile strength (814 MPa) and elongation (18.6 %), resulting in the lowest wear rate of 16.8 × 10−5mm3/(N∙m). The dominated wear mechanisms of the CG sample, MAF sample and 500 °C annealed MAF sample were abrasive wear, adhesive wear and delamination wear, while the primary mechanism of the 400 °C annealed MAF sample was abrasive wear and adhesive wear. It was found that the wear mechanism transforming, higher hardness and sufficient work hardening could be responsible for lower wear rate of the 400 °C annealed MAF sample. The 400 °C annealed MAF sample showed a thinner and more homogeneous deformation layer near the worn surface, and its thickness of deformation layer was about 8 μm.

To investigate the role of pre-twins in Mg alloy sheets during warm planar deformation, the stretch forming is conducted at 200 °C. Results suggest the formability of the pre-twinned AZ31 Mg alloy sheet is enhanced to 11.30 mm. The mechanisms for the improved formability and the deformation behaviors during the planar stretch forming are systematically investigated based on the planar stress states. The Schmid factor for deformation mechanisms are calculated, the results reveal that planar stress states extremely affect the Schmid factor for {10-12} twinning. The detwinning is activated and the prismatic slip is enhanced in the pre-twinned sheet, especially under the planar extension stress state in the outer region. Consequently, the thickness-direction strain is accommodated better. The dynamic recrystallization (DRX) type is continuous DRX (CDRX) regardless of the planar stress state. However, the CDRX degree is greater under the planar extension stress state. Some twin lattices deviate from the perfect {10-12} twinning relation due to the planar compression stress state and the CDRX. The basal texture is weakened when the planar stress state tends to change the texture components.

Abstract Objective The purpose of this study is to reduce the spread of the vanA gene by curing the vanA-harboring plasmid of vancomycin-resistant using the CRISPR-Cas9 system. Methods Two specific spacer sequence (sgRNAs) specific was designed to target the vanA gene and cloned into plasmid CRISPR-Cas9. The role of the CRISPR-Cas system in the plasmid elimination of drug-resistance genes was verified by chemically transformation and conjugation delivery methods. Moreover, the elimination efficiency in strains was evaluated by plate counting, PCR, and quantitative real-time PCR (qPCR). Susceptibility testing was performed by broth microdilution assay and by Etest strips (bioMérieux, France) to detect changes in bacterial drug resistance phenotype after drug resistance plasmid clearance. Results In the study, we constructed a specific prokaryotic CRISPR-Cas9 system plasmid targeting cleavage of the vanA gene. PCR and qPCR results indicated that recombinant pCas9-sgRNA plasmid can efficiently clear vanA-harboring plasmids. There was no significant correlation between sgRNA lengths and curing efficiency. In addition, the drug susceptibility test results showed that the bacterial resistance to vancomycin was significantly reduced after the vanA-containing drug-resistant plasmid was specifically cleaved by the CRISPR-Cas system. The CRISPR-Cas9 system can block the horizontal transfer of the conjugated plasmid pUC19-vanA. Conclusion In conclusion, our study demonstrated that CRISPR-Cas9 achieved plasmid clearance and reduced antimicrobial resistance. The CRISPR-Cas9 system could block the horizontal transfer of plasmid carrying vanA. This strategy provided a great potential to counteract the ever-worsening spread of the vanA gene among bacterial pathogens and laid the foundation for subsequent research using the CRISPR-Cas9 system as adjuvant antibiotic therapy.

Abstract Background Anthocyanin synthase (ANS) is the enzyme downstream of the anthocyanins synthesis pathway and the rate-limiting enzyme of the synthesis pathway. It catalyzes the conversion of colorless anthocyanins to anthocyanins and plays an important role in plant color presentation and stress resistance. However, ANS gene is rarely studied in grapes. Results In this study, 121 VvANS genes were identified and distributed on 18 chromosomes, VvANS family members were divided into 8 subgroups. Secondary structure prediction showed mainly irregular coils and α-helices, and subcellular localization indicated that VvANS gene family is mainly located in chloroplast, cytoplasm and nucleus. The promoter region of the VvANS gene family contains multiple cis-acting elements that are associated with light, abiotic stress, and hormones. Intraspecific collinearity analysis showed that there were 13 pairs of collinearity between VvANS genes. Interspecific collinearity analysis showed that there was more collinearity between grape, apple and Arabidopsis, but less collinearity between grape and rice. Microarray data analysis showed that VvANS17, VvANS23 and VvANS75 had higher expression levels in flesh and peel, while VvANS25, VvANS64 and VvANS106 had higher expression levels in flower. The results of qRT-PCR analysis showed that VvANS genes were expressed throughout the whole process of fruit coloring, such as VvANS47 and VvANS55 in the green fruit stage, VvANS3, VvANS64 and VvANS90 in the initial fruit color turning stage. The expression levels of VvANS21, VvANS79 and VvANS108 were higher at 50% coloring stage, indicating that these genes play an important role in the fruit coloring process. VvANS4, VvANS66 and VvANS113 had the highest expression levels in the full maturity stage. Conclusions These results indicated that different members of VvANS gene family played a role in different coloring stages, and this study laid a foundation for further research on the function of ANS gene family.

Abstract The emerging Space-Air-Ground, Artificial intelligence, blockchain and Vehicle-to-everything technology in Integrated Internet of Things (IoT) enables vehicles to communicate with other vehicles and roadside units (RSU), which improves communication efficiency and driving safety. Due to the rich data in the Integrated IoT, it is easy for illegal persons to steal data or deceive users. While reasonably allocating resources to complete the transmission task, it is still a challenge to ensure the communication security of vehicle users in Integrated IoT. In this paper, we study the cost of vehicle users transmitting tasks by Vehicle to Infrastructure and Vehicle to Vehicle (V2V) in Integrated IoT. In order to protect the information security, we establish the identity authentication and matching model to obtain a better channel environment of Integrated IoT. Moreover, we consider dynamic pricing based on bandwidth resource occupancy to ensure user experience and server load in Integrated IoT. Under the constraints of task tolerable delay, we fix the bandwidth on the V2V side and decouple the task decomposition and bandwidth allocation in Integrated IoT. Then, we propose algorithms Blockchain-based Vehicle Identity Authentication (BVIA) and Delay Weight Fairness Bandwidth Allocation (DWFBA). In this way, vehicle users and RSUs entering the Integrated IoT system are authenticated and matched with devices with good trust value, and the fairness of users resource allocation is guaranteed. Simulation results show that BVIA algorithm can reduce communication overhead, and DWFBA algorithm can control user costs and effectively reduce the number of task failures in Integrated IoT.
