Abstract Background The incidence of comorbid depression and post-traumatic stress disorder (PTSD) symptoms is higher in snakebite victims. However, the present state and contributing factors of depression and PTSD among Chinese snakebite victims remain unclear. Methods A representative sample of 6837 snakebite victims were assessed with the Post-traumatic Stress Disorder Checklist (Civilian Version) and The Center for Epidemiologic Studies Depression Scale. Multivariate analyses, including network analysis, evaluated the contributing factors of PTSD and depression symptoms caused by snake bites, as well as the bridge symptoms of comorbidity networks. Results Among 6,837 snakebite victims, 79.5% reported PTSD symptoms and 81.4% reported depression symptoms. Comorbidity of PTSD and depression symptoms was found in 75.1%. Key factors included the presence sequelae after snakebite (ORPTSD = 2.31, ORDepression = 1.89), time to medical facilities (6–8 h: ORPTSD = 3.17, ORDepression = 2.46), and marital status (divorced/widowed: ORPTSD = 1.78, ORDepression = 1.76). Symptoms I1 ("Repeated disturbing memories") and D1 ("Bothered by things that don't usually bother me") bridged PTSD and depression networks. Conclusion The primary psychological challenges for snakebite victims in China are PTSD and depression symptoms, which is concerning. Standardized diagnosis and treatments, timely medical care, and stable marital relationships can reduce risks. Additional psychological support and management of negative memories, especially for those with severe bridge symptoms, can be beneficial. Further research should concentrate on understanding victims' psychological states and developing effective interventions.

A significant proportion of the world's population is currently experiencing heat stress. This results from the effects of global warming, urban heat islands (UHIs), and the growth of urban populations. Researchers have investigated multiple strategies to address this problem. It has been found that the outdoor thermal environment (OTE) can be modified by means of vegetation, water surfaces, and urban geometry. Several studies have indicated that certain geometric factors have demonstrated a higher degree of effectiveness in cooling the surrounding environment. OTE studies often employ two commonly utilised factors, known as urban geometry indicating parameters (UGPs)—the sky view factor (SVF) and height-to-width ratio (H/W). This article compares OTE studies focusing on these two factors. The comparison encompasses characteristics such as study methods, places and times of conducting, and thermal indices. The findings indicate that geometric studies have primarily relied on simulation and field measurement methods. These studies were predominantly conducted in tropical or subtropical cities during warm seasons, with a preference for daytime investigations. Complex thermal indices, including the physiologically equivalent temperature and mean radiant temperature, were commonly employed. In addition to UGPs, other physical factors, such as orientation, were also taken into consideration. Studies on H/W and SVF differed slightly although they are both geometrically indicative. This review can provide guidance for future studies in the field of urban geometry, in that researchers may choose to prioritise the examination of practical indicators, such as the energy load, to conduct a more comprehensive analysis while also taking into account other physical aspects, such as the presence of trees.

The thermal effect of coal pyrolysis is a key parameter in the process of coal pyrolysis mechanism research and reactor design. At present, there are a lot of differences in the accuracy of evaluation methods for coal pyrolysis thermal effect. The most widely used and feasible method is based on simultaneous thermal analyzer (TG-DSC synchronous combination). However, this method still relies heavily on the measurement accuracy of the instrument, and there are also some differences in the accuracy of thermal effect calculation methods after the heat flow curve is obtained, resulting in a poor repeatability of the data and difficulty in adapting to the needs of large-scale production and process design. Tar-rich coal is high-quality raw material for coal to tar, it contains more hydrogen-rich structures such as aliphatic side chains and bridge bonds, it could be pyrolyzed to produce more tar during pyrolysis. The regulation of pyrolysis process is the key factor affecting the quality of tar produced by the pyrolysis of tar-rich coal, and the thermal effect of pyrolysis process is an important parameter for selecting pyrolysis process. Therefore, it is urgent to develop an accurate and efficient evaluation method of coal pyrolysis thermal effect for both tar-rich coal pyrolysis and generalized coal conversion. Based on the experimental results for the slow pyrolysis of tar-rich coal, combined with pyrolysis reaction mechanism and empirical formula, the product-oriented pyrolysis reaction system of tar-rich coal was constructed. The chemical reaction heat for the pyrolysis process of tar-rich coal was obtained by the thermodynamic calculation of the reaction system using the classical thermodynamic analysis method. Combined with the physical heat absorption in the pyrolysis process, the thermal effect in the low-temperature slow pyrolysis process of tar-rich coal could be obtained. Then, based on the reported coal pyrolysis experimental results and measured values of pyrolysis thermal effect, the random forest model of machine learning method was used to nonlinearly model and predict the thermal effect of coal pyrolysis. The results showed that based on experimental results, the thermal effect value for slow pyrolysis of tar-rich coal calculated by the product-oriented calculation was generally lower than that measured by TG-DSC synchronous combination, and the error was within 10%. The prediction accuracy for the thermal effect value of tar-rich coal pyrolysis based on machine learning algorithm was 0.935 2. Overall, the two prediction models of thermal effect of tar-rich coal pyrolysis constructed were practical and applicable.

Abstract Vascular dementia (VaD) is a prevalent form of dementia resulting from chronic cerebral hypoperfusion (CCH). However, the pathogenic mechanisms of VaD and corresponding therapeutic strategies are not well understood. Sirtuin 6 (SIRT6) has been implicated in various biological processes, including cellular metabolism, DNA repair, redox homeostasis, and aging. Nevertheless, its functional relevance in VaD remains unexplored. In this study, we utilized a bilateral common carotid artery stenosis (BCAS) mouse model of VaD to investigate the role of SIRT6. We detected a significant decrease in neuronal SIRT6 protein expression following CCH. Intriguingly, neuron-specific ablation of Sirt6 in mice exacerbated neuronal damage and cognitive deficits after CCH. Conversely, treatment with MDL-800, an agonist of SIRT6, effectively mitigated neuronal loss and facilitated neurological recovery. Mechanistically, SIRT6 inhibited excessive mitochondrial fission by suppressing the CCH-induced STAT5-PGAM5-Drp1 signaling cascade. Additionally, the gene expression of monocyte SIRT6 in patients with asymptomatic carotid stenosis showed a correlation with cognitive outcomes, suggesting translational implications in human subjects. Our findings provide the first evidence that SIRT6 prevents cognitive impairment induced by CCH, and mechanistically, this protection is achieved through the remodeling of mitochondrial dynamics in a STAT5-PGAM5-Drp1-dependent manner.

In compact accelerator mass spectrometry (AMS) systems，the detection of low-energy ions is often interfered by interfering ions with the same magnetic rigidity and electric rigidity as the target nuclide. To address this issue，a gas detector with range detection capability was developed for low-energy small-scale AMS. By exploiting the different ranges of the target nuclide and interfering ions，all interfering ions are absorbed in an absorber chamber while the target nuclide can pass through and enter an amplification chamber with proportional amplification functionality，thus facilitating the measurement of the target nuclide. To validate the performance of the gas detector，a series of calibration experiments were conducted using 5.8 MeV alpha particles. Experimental and simulation results demonstrate that the absorber chamber effectively absorbs interfering ions of different energies，and the energy resolution of amplification chamber is around 30% for the one-way signal. At the same time，when the reduced field strength in the amplification chamber is at 5 V·mm−1·hPa−1，the signal can be amplified by about 7 times. The energy resolution of the combined signals from the absorber and the amplification chamber is improved to around 20%，laying the foundation for the measurement of nuclides such as 26Al in low-energy small-scale AMS.

Abstract The glymphatic-lymphatic system is increasingly recognized as fundamental for the homeostasis of the brain milieu since it defines cerebral spinal fluid flow in the brain parenchyma and eliminates metabolic waste. Animal and human studies have uncovered several important physiological factors regulating the glymphatic system including sleep, aquaporin-4, and hemodynamic factors. Yet, our understanding of the modulation of the glymphatic system is limited, which has hindered the development of glymphatic-based treatment for aging and neurodegenerative disorders. Here, we present the evidence from fluorescence tracing, two-photon recording, and dynamic contrast-enhanced magnetic resonance imaging analyses that 40 Hz light flickering enhanced glymphatic influx and efflux independently of anesthesia and sleep, an effect attributed to increased astrocytic aquaporin-4 polarization and enhanced vasomotion. Adenosine-A2A receptor (A2AR) signaling emerged as the neurochemical underpinning of 40 Hz flickering-induced enhancement of glymphatic flow, based on increased cerebrofluid adenosine levels, the abolishment of enhanced glymphatic flow by pharmacological or genetic inactivation of equilibrative nucleotide transporters-2 or of A2AR, and by the physical and functional A2AR–aquaporin-4 interaction in astrocytes. These findings establish 40 Hz light flickering as a novel non-invasive strategy of enhanced glymphatic flow, with translational potential to relieve brain disorders.

Abstract Aim To investigate the impact of preoperative cardiovascular disease on the perioperative period of rectal cancer patients over 75 years old. Methods The clinicopathological data of 625 elderly patients aged ≥ 75 years who underwent radical rectal cancer surgery in the Cancer Hospital of the Chinese Academy of Medical Sciences and affiliated Heji Hospital of Changzhi Medical College from January 2011 to December 2022 were retrospectively collected and analyzed. According to preoperative comorbidities, all patients were divided into cardiovascular disease group (n = 361) and non-cardiovascular disease group (n = 264). One hundred and ninety-two pairs were selected from each group through Propensity score-matched to further analysis. Perioperative indexes and postoperative complications were compared between the two groups. Results There were no significant differences in clinicopathological data between the two groups (P > 0.05). The proportion of elderly patients with cardiovascular disease who went to ICU after radical surgery was significantly higher than those without cardiovascular disease (19.3% vs. 10.4%, P = 0.015). There was no significant difference between the two groups in the time to first flatus (3.0 vs. 3.5 days, P = 0.332) and postoperative hospital stay (11.3 vs. 10.5 days, P = 0.297). One patient in the cardiovascular disease group died due to pulmonary embolism. A total of 100 patients (26.0%) developed postoperative complications, and the incidence of overall complications (30.7% vs. 21.4%, P = 0.036) and grade 3–5 complications (12.5% vs. 6.3%, P = 0.036) in the cardiovascular disease group was significantly higher than that in the non-cardiovascular disease group. In terms of gastrointestinal disorders, the incidence of anastomotic leakage (6.8% vs. 2.1%, P = 0.026) in elderly patients with cardiovascular diseases was significantly higher than that in patients without cardiovascular disease. In addition, the incidence of cardiac disorders (8.3% vs. 2.6%, P = 0.014) in elderly patients with cardiovascular disease was significantly higher. Conclusion Elderly rectal cancer patients over 75 years old with cardiovascular disease are more likely to develop severe complications after radical surgery, especially anastomotic leakage and cardiac disorders.

Abstract Background Extensive research has been conducted on embryonic developmental disorders linked to Polycystic Ovary Syndrome (PCOS), a pathological condition that affects 5−10% of women and is characterized by irregularities in the menstrual cycle and infertility. By employing RNA sequencing (RNA-seq), we performed an in-depth investigation of PCOS-related changes in gene expression patterns at the mouse blastocyst stage. Methods The zygotes of female B6D2 mice were obtained and then differentiated into blastocysts in K + Simplex Optimised Medium (KSOM) cultures containing exo-NC (negative control for exosomes) or exo-LIPE-AS1 (a novel exosomal marker of PCOS). Subsequently, blastocysts were collected for RNA-seq. The bioinformatics was performed to analyze and compare the differences of gene expression profile between blastocysts of control and PCOS group. Results There were 1150 differentially expressed genes (DEGs) between the two groups of mouse blastocysts; 243 genes were upregulated and 907 downregulated in the blastocysts of the exo-LIPE-AS1 group compared to those of the exo-NC group. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed that the genes involved in amino acid synthesis and glutathione metabolic pathways were down-regulated in exo-LIPE-AS1 group. Conclusion This study has revealed that blastocyst developmental retardation may be associated with the downregulation of amino acid synthesis and glutathione metabolism, which may affect energy metabolism, biosynthesis, cellular osmotic pressure, antioxidant synthesis, ROS clearance or mitochondrial function, and ultimately cause blastocyst cell development abnormalities. Our research offers encouraging data on the mechanisms underlying aberrant embryonic development in patients with PCOS as well as potential treatment strategies.

Abstract Background China braces for coronavirus disease 2019 (COVID-19) surge after adjusting the “zero COVID” strategy. We aimed to evaluate and compare the prevalence of clinical symptoms of the Omicron variant infection among people living with HIV (PLWH) and HIV-free people. Methods A cross-sectional study was conducted in Wuchang District, Wuhan, Hubei Province, in December 2022 by a self-administered online survey during the Omicron wave. Participants aged ≥ 18 years with confirmed severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) diagnosis were recruited. PLWH managed by the local healthcare system were recruited, while HIV-free people were recruited by sending out online surveys through WeChat. We compared the prevalence of clinical symptoms of COVID-19 between PLWH and HIV-free people, and factors associated with symptom occurrence among PLWH were accessed. Results Total, 687 PLWH and 1222 HIV-free people were enrolled. After adjusting sex, age, body mass index, comorbidities and COVID-19 vaccination status, the prevalences of all symptoms, including higher degree and long duration of fever (aOR 0.51, 95%CI 0·42 − 0·61; aOR 0.52, 95%CI 0·43 − 0·63), were significantly lower among PLWH than among HIV-free people. Among PLWH, CD4+ T lymphocyte count (CD4 count) between 350 ~ 499 cells/µL and detectable HIV viral load (HIV-VL) were associated with significantly decreased risks of fever (aOR 0·63, 95%CI 0·40 − 0·97; aOR 0·56, 95%CI 0·33 − 0·94), headache (aOR 0·61, 95%CI 0·41 − 0·91; aOR 0·55, 95%CI 0·34 − 0·92) and muscle soreness (aOR 0·57, 95%CI 0·39 − 0·84; aOR 0·57, 95%CI 0·39 − 0·84). No apparent association between the symptoms prevalence and three/four doses of inactivated COVID-19 vaccination among PLWH was observed; both males and older age were associated with significantly decreased risks of nasal congestion/runny nose (aOR 0·52, 95%CI 0·32 − 0·82; aOR 0·97, 95%CI 0·96 − 0·99) and headache (aOR 0·58, 95%CI 0·36 − 0·92; aOR 0·96, 95%CI 0·95 − 0·98); older age was associated with significantly decreased risks of higher degree of fever (aOR 0·97, 95%CI 0·95 − 0·98). Conclusions PLWH have significantly milder symptoms of the Omicron variant infection than HIV-free people. PLWH who are male, older, have low CD4 count, and detectable HIV-VL have reduced occurrence of COVID-19 symptoms. However, continuous monitoring should be conducted among PLWH during the COVID-19 pandemic.

Abstract Cloud products from geostationary satellites are the main alternative to surface synoptic cloud observations (SYNOP), and have become the baseline products for the development and construction of the Quality Management System (QMS) of integrated meteorological observation in China. This study addresses the needs of the China Meteorological Administration (CMA) for such an operational reform, and it is carried out using real-time observations obtained from the Fengyun-2E (FY-2E) geostationary satellite in 2012 to derive cloud total amount and classification using two different methods. Compared to surface SYNOP observations, the cloud total amount estimated by FY-2E is generally significantly lower (about 30% lower on average). The cloud classification resulting from the two methods used in this study is also significantly different from the classification obtained from surface observations (difference between 22 and 32%). The difference is smaller for the classification method, which uses additional auxiliary temperature profiles.

Abstract Background The GeneXpert MTB/RIF (Xpert) assay is a widely used technology for detecting Mycobacterium tuberculosis (MTB) in clinical samples. However, the study on the failure of the Xpert assay during routine implementation and its potential solutions is limited. Methods We retrospectively analyzed the records of unsuccessful tests in the Xpert and the GeneXpert MTB/RIF Ultra (Ultra) assays between April 2017 and April 2021 at the Shanghai Public Health Clinical Center. To further investigate the effect of prolonged preprocessing on clinical sputum, an additional 120 sputum samples were collected for Xpert testing after 15 min, 3 h, and 6 h preprocessing. The analysis was performed by SPSS version 19.0 software. Results A total of 11,314 test records were analyzed, of which 268 (2.37%) had unsuccessful test results. Among these, 221 (1.95%) were reported as “Error”, 43 (0.38%) as “Invalid”, and 4 (0.04%) as “No result”. The most common clinical specimen for Xpert tests was sputum, accounting for 114 (2.17%) unsuccessful tests. The failure rate of urine specimens was lower than that of sputum (OR = 0.12, 95% CI: 0.02–0.88, χ2 = 6.22, p = 0.021). In contrast, the failure rate of stool specimens was approximately twice as high as that of sputum (OR = 1.93, 95% CI: 1.09–3.40, χ2 = 5.35, p = 0.014). In the prolonged preprocessing experiment, 102 cases (85%) yielded consistent results in Xpert tests. Furthermore, 7 cases (5.83%) detected an increase in MTB load, 8 cases (6.67%) detected a decrease in MTB load, and 3 cases (2.5%) yielded incongruent results in MTB and rifampicin resistance detection. Conclusions The primary cause of unsuccessful tests in the Xpert assay was reported as “Error”. Despite varying failure rates depending on the samples, the Xpert assay can be applied to extrapulmonary samples. For paucibacillary specimens, retesting the remaining preprocessed mixture should be carefully considered.

Calculating the parameters of the ground shock induced by an underground explosion is a complex energy coupling problem. It is difficult to establish a unified ground shock coupling law from limited test data. This paper summarizes the research results obtained at home and abroad and systematically analyzes the coupling mechanism of craters formed by an underground explosion and the ground shock. The differences between the concepts of “closed-explosion critical depth” and “equivalent closed-explosion critical depth” are clearly explained. The spreading of the ground shock energy is attributed to the explosive expansion of the air cavity, revealing a linear relationship between the volume of the cavity region (or the volume of the crack region) and the ground shock energy associated with the underground explosion. The proportionality factor is related to the mechanical properties of the medium and is independent of the magnitude of the explosion equivalent. Based on this, a theoretical calculation formula and conversion method for the ground shock coupling coefficient were established. Explosion tests were conducted in clay and Plexiglass under varying burial depths. The test results were consistent with the theoretically calculated results. Our study provides a theoretical basis for the design of explosion-resistant structures in underground engineering.

OBJECTIVE: The aim of the study was to analyze the clinical data of patients with chronic obstructive pulmonary disease and obstructive sleep apnea overlap syndrome (OS) during hospitalization and to evaluate the risk factors of patients treated with Non-Invasive Ventilation (NIV). METHODS: Demographic and clinical data of patients with confirmed OS during hospitalization were retrospectively collected. The patients were divided into two groups according to whether noninvasive ventilator was used during hospitalization, including OS treated with NIV (244 cases) and OS without NIV (239 cases). The t-test, χ2 test, and Kaplan–Meier curve were used to compare the two groups, and multiple logistic regression was used to analyze the risk factors of NIV in patients with OS. RESULTS: Compared with the OS group without NIV, the pulmonary hypertension, lymphocyte count, and left ventricular ejection fraction% of OS patients with NIV were lower, whereas PCO2, uric acid, C-reactive protein, procalcitonin, and N-terminal pro-B-type natriuretic peptide were higher, with statistical differences (P < 0.05). During hospitalization and follow-up, OS patients with NIV had a longer hospital stay (P < 0.001), and there was no significant difference in the rate of readmission within 28 days. The logistic regression analysis showed that the history of diuretic use, previous history of noninvasive ventilator use, and ischemic heart disease were independent risk factors for NIV treatment in OS patients during hospitalization. CONCLUSION: Patients with OS undergoing NIV during hospitalization exhibited more severe overall illness and had prolonged hospital stays compared to OS patients not receiving NIV. History of diuretic use, history of NIV use, and ischemic heart disease are independent risk factors for NIV treatment in OS patients during hospitalization.

Abstract Understanding listeners’ perceptions of consecutive interpreting (CI) quality is crucial for the interpreting profession and service management. However, research in this area is still in its early stages, lacking in-depth exploration of how listeners’ subjective factors impact their perceived CI quality and the extent of these interactions. This study introduced a research model based on existing literature, incorporating six key listener variables: listeners’ quality expectations, perceived characteristics of the interpreter, experiences with CI, domain knowledge, perceived dependence on CI, and perceived communicative effect. A case-based survey was conducted during a simulated interpreter-mediated technical conference. We surveyed 107 conference attendees before and after the technical event, testing 11 proposed hypotheses using their self-reported data. Data were analyzed using descriptive statistics and partial least squares structural equation modeling (PLS-SEM). The findings indicate that (1) domain knowledge has a negative impact on listeners’ perceived CI quality, while the remaining five listener variables positively influence their CI quality perceptions. Quality expectations emerge as the most influential predictor, followed by perceived characteristics of the interpreter and perceived communicative effect. (2) Listeners’ perceived characteristics of the interpreter and perceived communicative effect are positively predicted by their quality expectations, which highlights the fundamental role of quality expectations. (3) Domain knowledge and experiences with CI are confirmed as significant factors influencing quality expectations, while perceived characteristics of the interpreter significantly drive perceived communicative effect. Overall, the findings reveal the complex interplay of these subjective factors and illustrate how they collectively shape listeners’ perceptions of CI quality. Theoretical and practical implications for professional interpreting practice and interpreter training are also discussed.

Abstract The performance of superconducting qubits is degraded by a poorly characterized set of energy sources breaking the Cooper pairs responsible for superconductivity, creating a condition often called “quasiparticle poisoning”. Both superconducting qubits and low threshold dark matter calorimeters have observed excess bursts of quasiparticles or phonons that decrease in rate with time. Here, we show that a silicon crystal glued to its holder exhibits a rate of low-energy phonon events that is more than two orders of magnitude larger than in a functionally identical crystal suspended from its holder in a low-stress state. The excess phonon event rate in the glued crystal decreases with time since cooldown, consistent with a source of phonon bursts which contributes to quasiparticle poisoning in quantum circuits and the low-energy events observed in cryogenic calorimeters. We argue that relaxation of thermally induced stress between the glue and crystal is the source of these events.

Abstract Objective With the development of surgical technology, the level of digital medicine is constantly improving. The birth of new technologies has a certain impact on traditional methods. At present, robot-assisted technology has been applied to patients with calcaneal fractures, which poses a challenge to traditional surgery. We aimed to assess whether robot-assisted internal fixation confers certain surgical advantages through a literature review. Design The databases PubMed, EMBASE, the Cochrane Library, the China National Knowledge Infrastructure (CNKI), and the Wanfang Data Knowledge Service Platform were systematically searched for both randomized and nonrandomized studies involving patients with calcaneal fractures. Main results Five studies were identified that compared clinical indexes. For the clinical indexes, robot-assisted surgery is generally feasible because of intraoperative fluoroscopy, complications, the Gissane angle, the calcaneal width, and the American Orthopedic Foot and Ankle Society (AOFAS) Ankle-Hindfoot score 3 and 6 months after the operation (P 0.05). Conclusions Based on the current evidence, the advantages of robot-assisted fixation over traditional fixation are clear. The long-term clinical effects of the two methods are also good, and the short-term effect of robot assistance is better. However, the quality of some studies is low, and more high-quality randomized controlled trials (RCTs) are needed for further verification.

Titanium alloy is an important structural material with many advantages of high strength, excellent corrosion resistance, good biocompatibility, and good processing performance, which has been widely used in aerospace, medical devices, automobile manufacturing and various other fields. However, the presence of hydrogen in titanium alloys can lead to hydrogen embrittlement, which degrades the mechanical properties. Due to its high ionization energy, high reactivity, and high diffusivity, quantitative analysis of hydrogen in metal or alloy materials faces enormous challenge. Accurately and quickly detecting the hydrogen content in titanium alloys is of great significance for optimizing material design and controlling product quality. Traditional methods for determining the hydrogen in titanium alloys require complicated sample pretreatment, which greatly prolong the analysis time. The self-built high irradiance laser ablation and ionization time-of-flight mass spectrometry (LAI-TOF MS) has many advantages such as high sensitivity, high resolution, multi-element simultaneous detection, and simple sample pretreatment. It introduces helium into the ion source as a buffer gas to cool the high kinetic energy ions and reduce the multi-charged ion interferences, and has been successfully applied to the determination of metallic and nonmetallic elements in solid samples. In this study, a direct quantitative analysis method of LAI-TOF MS was established for detecting hydrogen in titanium alloys. The calibration curve of intensities and contents for hydrogen has a correlation coefficient of 0.991 3, the limit of detection (LOD) is calculated to be 0.000 4%, and the atomization and ionization efficiency of hydrogen can reach 13.4%. This method was applied to the analysis of hydrogen in other titanium alloy samples. The relative standard deviations (RSDs) of hydrogen signals are less than 14%. The relative errors of the quantitative results are less than 5% compared with values obtained by the pulsed heating-infrared method. The analysis time for a single titanium alloy sample is only a few minutes, significantly shorter than the 2-4 h required for the pulsed heating-infrared method. The spectrum obtained by LAI-TOF MS enables qualitative and semi-quantitative determination of trace elements of Be, C, Al, Si, P, Ca, Cr, Fe, Zr, Mo, Pd, In and Sn in the titanium alloy samples. Considering the advantages of less sample preparation, simplicity, rapidness, and high sensitivity, the method based on LAI-TOF MS can be used to the directly quantitative analysis of hydrogen in titanium alloys and can be further extended to the quantitative analysis of hydrogen in other materials, such as hydrogen storage materials, metal materials, ceramic materials and other related materials.

Abstract Background Wheat landraces are considered a valuable source of genetic diversity for breeding programs. It is useful to evaluate the genetic diversity in breeding studies such as marker-assisted selection (MAS), genome-wide association studies (GWAS), and genomic selection. In addition, constructing a core germplasm set that represents the genetic diversity of the entire variety set is of great significance for the efficient conservation and utilization of wheat landrace germplasms. Results To understand the genetic diversity in wheat landrace, 2,023 accessions in the Jiangsu Provincial Crop Germplasm Resource Bank were used to explore the molecular diversity and population structure using the Illumina 15 K single nucleotide polymorphism (SNP) chip. These accessions were divided into five subpopulations based on population structure, principal coordinate and kinship analysis. A significant variation was found within and among the subpopulations based on the molecular variance analysis (AMOVA). Subpopulation 3 showed more genetic variability based on the different allelic patterns (Na, Ne and I). The M strategy as implemented in MStratv 4.1 software was used to construct the representative core collection. A core collection with a total of 311 accessions (15.37%) was selected from the entire landrace germplasm based on genotype and 12 different phenotypic traits. Compared to the initial landrace collections, the core collection displayed higher gene diversity (0.31) and polymorphism information content (PIC) (0.25), and represented almost all phenotypic variation. Conclusions A core collection comprising 311 accessions containing 100% of the genetic variation in the initial population was developed. This collection provides a germplasm base for effective management, conservation, and utilization of the variation in the original set.

Abstract Background This study explored the relationship between inflammatory markers and glucocorticoid dosage upon admission. Methods We conducted a retrospective analysis of 206 patients with refractory Mycoplasma pneumoniae pneumonia (RMPP) admitted to a Children’s Hospital from November 2017 to January 2022. Patients were categorized into three groups based on their methylprednisolone dosage: low-dose (≤ 2 mg/kg/d), medium-dose (2–10 mg/kg/d), and high-dose (≥ 10 mg/kg/d). AbstractContext The mechanisms of Traditional Chinese Medicine (TCM) Guizhi-Gancao Decoction (GGD) remain unknown.Objective This study explores the mechanisms of GGD against cardiac hypertrophy.Materials and methods Network pharmacology analysis was carried out to identify the potential targets of GGD. In vivo experiments, C57BL/6J mice were divided into Con, phenylephrine (PE, 10 mg/kg/d), 2-chloroadenosine (CADO, the stable analogue of adenosine, 2 mg/kg/d), GGD (5.4 g/kg/d) and GGD (5.4 g/kg/d) + CGS15943 (a nonselective adenosine receptor antagonist, 4 mg/kg/d). In vitro experiments, primary neonatal rat cardiomyocytes (NRCM) were divided into Con, PE (100 µM), CADO (5 µM), GGD (10−5 g/mL) and GGD (10−5 g/mL) + CGS15943 (5 µM). Ultrasound, H&E and Masson staining, hypertrophic genes expression and cell surface area were conducted to verify the GGD efficacy. Adenosine receptors (ADORs) expression were tested via real-time polymerase chain reaction (PCR), western blotting and immunofluorescence analysis.Results Network pharmacology identified ADORs among those of the core targets of GGD. In vitro experiments demonstrated that GGD attenuated PE-induced increased surface area (with an EC50 of 5.484 × 10−6 g/mL). In vivo data shown that GGD attenuated PE-induced ventricular wall thickening. In vitro and in vivo data indicated that GGD alleviated PE-induced hypertrophic gene expression (e.g., ANP, BNP and MYH7/MYH6), A1AR over-expression and A2aAR down-expression. Moreover, CADO exerts effects similar to GGD, whereas CGS15943 eliminated most effects of GGD.Discussion and conclusions Our findings suggest the mechanism by which GGD inhibits cardiac hypertrophy, highlighting regulation of ADORs as a potential therapeutic strategy for HF.

Abstract Since the oncogenic rearranged during transfection (RET) gene fusion was discovered in non-small cell lung cancer (NSCLC) in 2012, multiple-targeted kinase inhibitors (MKIs) cabozantinib and vandetanib have been explored in the clinic for RET positive NSCLC patients. As the nonselective nature of these inhibitors, patients have off-target adverse effects. The discovery of highly potent selective RET inhibitors such as pralsetinib and selpercatinib improve the clinic efficiency and more favorable toxicity profile. However, acquired resistance mediated by secondary mutations in the solvent-front region of the kinase (e.g. G810C/S/R) become a new challenge for selective RET inhibitor therapies. In this review, we highlight typical RET inhibitors developed during these years and provide a reference for more potential RET inhibitors exploration in the future.

Abstract Background Mycoplasma pneumoniae pneumonia is a common respiratory infection among children. However, the occurrence of thromboembolism with plastic bronchitis in association with Mycoplasma pneumoniae pneumonia is extremely rare. This case series presents five cases of children with Mycoplasma pneumoniae pneumonia who developed thromboembolism and plastic bronchitis. The clinical presentation, diagnostic approach, and management strategies are discussed. Methods A retrospective analysis was conducted on medical records from a pediatric hospital. Patient demographics, clinical features, laboratory findings, imaging results, treatment modalities, and outcomes were collected. Results The patients in our case series presented with varying degrees of respiratory distress, cough, and fever. Imaging studies revealed evidence of thromboembolism based on pulmonary artery occlusion. Bronchial casts were observed by bronchoscopy. Laboratory tests demonstrated elevated D-dimer levels and fibrinogen degradation products. All patients received a combination of low molecular weight heparin anticoagulation and supportive care. Conclusion Thromboembolism with plastic bronchitis associated with Mycoplasma pneumoniae pneumonia is a rare but potentially serious complication in children. Prompt recognition and management are crucial for improving patient outcomes. This case series highlights the diverse clinical presentations, diagnostic challenges, and treatment strategies for this unique clinical entity. Further research is needed to better understand the pathogenesis and optimal management of this condition.

Abstract Background Anemia is a common complication of tuberculosis (TB), and there is evidence that its prevalence is higher in patients with TB. Although TB is very important in epidemiology, careful investigation of TB-related anemia in children has not been carried out systematically. This study aimed to describe the details of anemia in children with TB and its association with clinical characteristics and the severity of inflammation. Methods In this retrospective study, we explored Hb levels in 103 children with pulmonary TB (PTB) and they were divided into anemic or non-anemic groups. Logistics regression analysis was used to study the associations between anemia and demographic characteristics. Spearman correlations analysis was performed to analyse the associations between the biochemical parameters and hemoglobin levels in blood. Results The prevalence of anemia in children with TB was 37.9% (48.7% showed microcytic hypochromic anemia, and 5.1% showed normal cell anemia). Abstract Although it is well documented that mountains tend to exhibit high biodiversity, how geological processes affect the assemblage of montane floras is a matter of ongoing research. Here, we explore landform-specific differences among montane floras based on a dataset comprising 17,576 angiosperm species representing 140 Chinese mountain floras, which we define as the collection of all angiosperm species growing on a specific mountain. Our results show that igneous bedrock (granitic and karst-granitic landforms) is correlated with higher species richness and phylogenetic overdispersion, while the opposite is true for sedimentary bedrock (karst, Danxia, and desert landforms), which is correlated with phylogenetic clustering. Furthermore, we show that landform type was the primary determinant of the assembly of evolutionarily older species within floras, while climate was a greater determinant for younger species. Our study indicates that landform type not only affects montane species richness, but also contributes to the composition of montane floras. To explain the assembly and differentiation of mountain floras, we propose the ‘floristic geo-lithology hypothesis’, which highlights the role of bedrock and landform processes in montane floristic assembly and provides insights for future research on speciation, migration, and biodiversity in montane regions.

Abstract Background Alterations in epigenetic factors are recognized as key contributors to the emergence of human cancer. The active and reversible alteration of N6‐methyladenosine (m6A) RNA is crucial for controlling gene activity and determining cellular destiny. Even with these insights, the triggering of KIAA1429 (also called VIRMA) and its role in lung adenocarcinoma (LUAD) is mostly unclear. As a result, the objective of this study was to elucidate how KIAA1429 contributes to cancer development in LUAD. Methods This study utilized multiple methods for investigation, encompassing the in vitro functional examination of KIAA1429 in lung adenocarcinoma cells, transcriptome sequencing, methylation RNA immunoprecipitation sequencing (MeRIP‐seq), as well as RNA stability tests to ascertain the half‐life and stability of the target genes. Results The results indicated that modifying the expression of KIAA1429 regulated the proliferation and metastasis of LUAD. By employing transcriptome sequencing alongside MeRIP‐seq analysis, the research pinpointed genes affected by m6A alterations triggered by KIAA1429. In a more detailed manner, it was discovered that KIAA1429 plays a regulatory role in the expression of ARHGAP30. During checking continuous waveform record, it was found that some stations of Tianjin such as EWZ were significantly affected by one unknown noise, and EWZ station was the most seriously affected. Long term observations had also found that this noise cannot be observed for uncertainly several days each year. This noise was all year round and did not change with day and night time, and it had been existed for many years. This noise was different from typical environmental background noise and had some fixed characteristics. This article studied the noise from several aspects such as its own characteristic, its coverage of influence, spectral characteristics, and impact on earthquake records. The research has shown that the noise was not affected by natural factors such as weather and season, and had a characteristic of amplitude decreasing as the distance increasing from EWZ station. The main frequency of this noise ranged 1～2 Hz. The impact of noise was relatively wide and it can be recorded by 12 surrounding stations, with YGZ station being the farthest and approximately 58 km away from EWZ station. Through polarization analysis, it was found that the propagation of noise had a specific directionality, so it is preliminary judged that this noise had the characteristic of fixed source noise. This noise will have an impact on seismic records, reducing the seismic phase pickup rate, and even completely submerging it in the noise. Frequency filtering can improve it.

Abstract Fetal adenocarcinoma of the lung (FLAC) is a rare form of lung adenocarcinoma and was divided into high-grade (H-FLAC) and low-grade (L-FLAC) subtypes. Despite the existence of some small case series studies, a comprehensive multi-omics study of FLAC has yet to be undertaken. In this study, we depicted the multi-omics landscapes of this rare lung cancer type by performing multi-regional sampling on 20 FLAC cases. A comparison of multi-omics profiles revealed significant differences between H-FLAC and L-FLAC in a multi-omic landscape. Two subtypes also showed distinct relationships between multi-layer intratumor heterogeneity (ITH). We discovered that a lower genetic ITH was significantly associated with worse recurrence-free survival and overall survival in FLAC patients, whereas higher methylation ITH in H-FLAC patients suggested a short survival. Our findings highlight the complex interplay between genetic and transcriptional heterogeneity in FLAC and suggest that different types of ITH may have distinct implications for patient prognosis.

Abstract Radiotherapy combined with immune checkpoint blockade holds great promise for synergistic antitumor efficacy. Targeted radionuclide therapy delivers radiation directly to tumor sites. LNC1004 is a fibroblast activation protein (FAP)-targeting radiopharmaceutical, conjugated with the albumin binder Evans Blue, which has demonstrated enhanced tumor uptake and retention in previous preclinical and clinical studies. Herein, we demonstrate that 68Ga/177Lu-labeled LNC1004 exhibits increased uptake and prolonged retention in MC38/NIH3T3-FAP and CT26/NIH3T3-FAP tumor xenografts. Radionuclide therapy with 177Lu-LNC1004 induced a transient upregulation of PD-L1 expression in tumor cells. The combination of 177Lu-LNC1004 and anti-PD-L1 immunotherapy led to complete eradication of all tumors in MC38/NIH3T3-FAP tumor-bearing mice, with mice showing 100% tumor rejection upon rechallenge. Immunohistochemistry, single-cell RNA sequencing (scRNA-seq), and TCR sequencing revealed that combination therapy reprogrammed the tumor microenvironment in mice to foster antitumor immunity by suppressing malignant progression and increasing cell-to-cell communication, CD8+ T-cell activation and expansion, M1 macrophage counts, antitumor activity of neutrophils, and T-cell receptor diversity. A preliminary clinical study demonstrated that 177Lu-LNC1004 was well-tolerated and effective in patients with refractory cancers. Further, scRNA-seq of peripheral blood mononuclear cells underscored the importance of addressing immune evasion through immune checkpoint blockade treatment. This was emphasized by the observed increase in antigen processing and presentation juxtaposed with T cell inactivation. In conclusion, our data supported the efficacy of immunotherapy combined with 177Lu-LNC1004 for cancer patients with FAP-positive tumors.

Abstract High-speed wide-field fluorescence microscopy has the potential to capture biological processes with exceptional spatiotemporal resolution. However, conventional cameras suffer from low signal-to-noise ratio at high frame rates, limiting their ability to detect faint fluorescent events. Here, we introduce an image sensor where each pixel has individually programmable sampling speed and phase, so that pixels can be arranged to simultaneously sample at high speed with a high signal-to-noise ratio. In high-speed voltage imaging experiments, our image sensor significantly increases the output signal-to-noise ratio compared to a low-noise scientific CMOS camera (~2–3 folds). This signal-to-noise ratio gain enables the detection of weak neuronal action potentials and subthreshold activities missed by the standard scientific CMOS cameras. Our camera with flexible pixel exposure configurations offers versatile sampling strategies to improve signal quality in various experimental conditions.

Abstract In prior investigations, a correlation was established between patient outcomes in locally advanced non-small cell lung cancer (LA-NSCLC) following thoracic irradiation and parameters, such as pre/post-treatment neutrophil-to-lymphocyte ratio (NLR) and NLR change (ΔNLR). However, these parameters could potentially be influenced by radiation-related variables, such as gross tumor volume (GTV). The primary aim of this study was to elucidate the factors impacting post-treatment NLR and ΔNLR and to further assess their prognostic relevance. In this retrospective study, a cohort of 188 LA-NSCLC patients who underwent thoracic radiation between 2012 and 2017 was assessed. The calculation of pre/post-treatment NLR involved the use of absolute neutrophil and lymphocyte counts. ΔNLR was defined as the difference between post- and pre-treatment NLR values. To assess the relationships between various variables and overall survival (OS), local progression-free survival (LPFS), and distant metastasis-free survival (DMFS), the Kaplan–Meier technique and Cox proportional hazards regression were employed. Additionally, Spearman’s rank correlation analysis was carried out to investigate correlations between the variables. The analysis revealed that both post-treatment NLR (r = 0.315, P 67.41 cm3 group (P = 0.028 and P = 0.042, respectively), highlighting ΔNLR as the sole independent predictive factor for survival and metastasis, irrespective of GTV.

Abstract Objective Brachytherapy has been indicated as an alternative option for treating cystic craniopharyngiomas (CPs). The potential benefits of brachytherapy for CPs have not yet been clarified. The purpose of this work was to conduct a meta-analysis to analyze the long-term efficacy and adverse reactions profile of brachytherapy for CPs. Materials and methods The relevant databases were searched to collect the clinical trials on brachytherapy in patients with CPs. Included studies were limited to publications in full manuscript form with at least 5-year median follow-up, and adequate reporting of treatment outcomes and adverse reactions data. Stata 12.0 was used for data analysis. Results According to the inclusion and exclusion criteria, a total of 6 clinical trials involving 266 patients with CPs were included in this meta-analysis. The minimum average follow-up was 5 years. The results of the meta-analysis showed that 1-year, 2–3 years and 5 years progression free survival rates (PFS) are 75% (95%CI: 66-84%), 62% (95%CI: 52-72%) and 57% (95%CI: 22-92%), respectively. At the last follow-up, less than 16% of patients with visual outcomes worser than baseline in all included studies. While, for endocrine outcomes, less than 32% of patients worser than baseline level. Conclusion In general, based on the above results, brachytherapy should be considered as a good choice for the treatment of CP.

Abstract Parental experiences can affect the phenotypic plasticity of offspring. In locusts, the population density that adults experience regulates the number and hatching synchrony of their eggs, contributing to locust outbreaks. However, the pathway of signal transmission from parents to offspring remains unclear. Here, we find that transcription factor Forkhead box protein N1 (FOXN1) responds to high population density and activates the polypyrimidine tract-binding protein 1 (Ptbp1) in locusts. FOXN1–PTBP1 serves as an upstream regulator of miR-276, a miRNA to control egg-hatching synchrony. PTBP1 boosts the nucleo-cytoplasmic transport of pre-miR-276 in a “CU motif”-dependent manner, by collaborating with the primary exportin protein exportin 5 (XPO5). Enhanced nuclear export of pre-miR-276 elevates miR-276 expression in terminal oocytes, where FOXN1 activates Ptbp1 and leads to egg-hatching synchrony in response to high population density. Additionally, PTBP1-prompted nuclear export of pre-miR-276 is conserved in insects, implying a ubiquitous mechanism to mediate transgenerational effects.

Abstract Background The World Health Organization (WHO) published the Integrated Care for Older People (ICOPE) framework to help healthcare providers cope with the population aging crisis. However, the relevant evidence on the demands of older people and the compensatory capacity of the environment is limited. This study reports for the first time the level of the ICOPE demand in Western China that includes the impact of geographic accessibility of medical resources (GAMR) on ICOPE demand and the potential mechanism of health status. Methods A cross-sectional questionnaire survey was conducted among 1200 adults aged 60 years and older selected through multi-stage stratified cluster sampling to obtain relevant data, including ICOPE demand, health status, and GAMR. Propensity score matching (PSM) was used to analyze the impact of GAMR on ICOPE demand among older people and those with different health statuses. Results Among the prospective research participants, 1043 were eligible for the study. The mean score of ICOPE demand among all participants was 3.68 (standard deviation [SD] = 0.78). After adjusting for covariates between high and low GAMR groups (1:1 match), ICOPE demand was significantly higher in the low GAMR group than in the high GAMR group (average treatment effect on the treated [ATT] = 0.270, p

This study evaluates the factors linking diabetes to dementia onset, providing guidance for preventing dementia in diabetic patients. Methods This analysis utilized databases such as PubMed, Embase, Web of Science, and the Cochrane Library to review literature from January 31, 2012, to March 5, 2023. Articles were rigorously assessed using specific inclusion and exclusion criteria. The Newcastle-Ottawa Scale (NOS) was used to evaluate the quality of the studies. Abstract Background Vasculopathy is the most common complication of diabetes. Endothelial cells located in the innermost layer of blood vessels are constantly affected by blood flow or vascular components; thus, their mechanosensitivity plays an important role in mediating vascular regulation. Endothelial damage, one of the main causes of hyperglycemic vascular complications, has been extensively studied. However, the role of mechanosensitive signaling in hyperglycemic endothelial damage remains unclear. Methods Vascular endothelial-specific Piezo1 knockout mice were generated to investigate the effects of Piezo1 on Streptozotocin-induced hyperglycemia and vascular endothelial injury. In vitro activation or knockdown of Piezo1 was performed to evaluate the effects on the proliferation, migration, and tubular function of human umbilical vein endothelial cells in high glucose. Reactive oxygen species production, mitochondrial membrane potential alternations, and oxidative stress-related products were used to assess the extent of oxidative stress damage caused by Piezo1 activation. Results Our study found that in VECreERT2;Piezo1flox/flox mice with Piezo1 conditional knockout in vascular endothelial cells, Piezo1 deficiency alleviated streptozotocin-induced hyperglycemia with reduced apoptosis and abscission of thoracic aortic endothelial cells, and decreased the inflammatory response of aortic tissue caused by high glucose. Moreover, the knockout of Piezo1 showed a thinner thoracic aortic wall, reduced tunica media damage, and increased endothelial nitric oxide synthase expression in transgenic mice, indicating the relief of endothelial damage caused by hyperglycemia. We also showed that Piezo1 activation aggravated oxidative stress injury and resulted in severe dysfunction through the Ca2+-induced CaMKII-Nrf2 axis in human umbilical vein endothelial cells. In Piezo1 conditional knockout mice, Piezo1 deficiency partially restored superoxide dismutase activity and reduced malondialdehyde content in the thoracic aorta. Mechanistically, Piezo1 deficiency decreased CaMKII phosphorylation and restored the expression of Nrf2 and its downstream molecules HO-1 and NQO1. Conclusion In summary, our study revealed that Piezo1 is involved in high glucose-induced oxidative stress injury and aggravated endothelial dysfunction, which have great significance for alleviating endothelial damage caused by hyperglycemia.

Abstract Background REBOA is a method used to manage bleeding during surgery involving sacropelvic tumors. Nevertheless, studies on the use of REBOA among elderly people are lacking. The aim of this research was to investigate the efficacy and safety of Zone III REBOA in patients aged more than 70 years. Methods A comparative study was conducted using case-control methods. A group of patients, referred to as Group A, who were younger than 70 years was identified and paired with a comparable group of patients, known as Group B, who were older than 70 years. Continuous monitoring of physiological parameters was conducted, and blood samples were collected at consistent intervals. Results Totally, 188 participants were enrolled and received REBOA. Among the 188 patients, seventeen were aged more than 70 years. By implementing REBOA, the average amount of blood loss was only 1427 ml. Experiments were also conducted to compare Group A and Group B. No notable differences were observed in terms of demographic variables, systolic blood pressure (SBP), arterial pH, lactate levels, blood creatinine levels, potassium levels, or calcium levels at baseline. Additionally, after the deflation of the REBOA, laboratory test results, which included arterial pH, lactate, potassium concentration, calcium concentration, and blood creatinine concentration, were not significantly different (P > 0.05). Conclusion This study indicated that in selected patients aged more than 70 years can achieve satisfactory hemodynamic and metabolic stability with Zone III REBOA. FGH98 powder metallurgy superalloy were successfully bonded via diffusion bonding technique with pure Ni interlayer. It was indicated that adding the Ni interlayer can significantly reduce joint defects, promote elements diffusion, and improve the bonding rate comparing with direct diffusion bonded joint. The typical joint is clearly divided into the IZ with larger volume dendritic γ′ phase and the DZ with narrow primary γ′ phase, petal secondary γ′ phase and granular tertiary γ′ phase. It was observed that the thinner the interlayer, the easier it is for the joint to achieve alloying, the more uneven the transition of the joint structure. Properly increasing the temperature or holding time can help with elements diffusion, but too high bonding parameters can cause grain seriously coarsening and reduce joint performance. The optimized process parameters of 1130 °C −60 min-10 μm Ni were obtained with reaching the highest shear strength of 656 MPa, which is the 201% of the direct diffusion bonded joint.

FGH98 nickel-based powder metallurgy superalloy was successfully bonded via transient liquid phase (TLP) bonding technique with a BNi-2 interlayer. The typical joint consisted of three characteristic zones including: ASZ composed of multiple silicon borides, ISZ with uniform γ solid solution and dispersed small γ′ phases and DAZ containing multiple kinds of borides which exist within and at grain boundaries. With the increase of bonding temperature and holding time, ASZ was gradually replaced by the ISZ, and eutectic structure disappeared. But excessive temperature or time will cause grain coarsening, deteriorating the joint performance. Under the bonding parameters of 1130 °C −60 min, the joint with optimal shear strength was obtained, which is 882 MPa (94% of the base metal strength). The fracture occurs in the DAZ, which is a mixed fracture.

Backfilling mining is important for controlling strata movement and reducing surface subsidence and environmental damage. Insufficient sources of raw materials and high backfilling costs have become major bottlenecks, limiting the application of backfill mining in underground coal mines. Exploring new methods and supporting technologies of targeted backfill with low backfilling rate in key areas is one of the research hotspots in backfilling mining. To solve these problems, combined with the academic thought of constructional backfill in underground coal mines, an in-depth study of the fracture characteristics of roof strata is conducted, the hinged shape and size of broken blocks in the roof strata are clarified, the main subsidence area and position of broken hinged blocks are identified, and the key position of the constructional backfill is determined accordingly. Through the key position determination method, an “I-shaped” constructional backfill new scheme that focuses on controlling the maximum deflection position of the main roof, i.e., the potential tensile fracture position (plastic hinge development position) of the plate structure, is proposed. Combined with the geological parameters of the rock strata of Xinyang Mine, FLAC3D numerical simulation software was used to analyze the stress, vertical subsidence, plastic zone distribution of the roof in the direction of mining width and advancing direction of the working face, and the stability evolution characteristics of backfilling body under different schemes are revealed. The results show that the “I-shaped” backfill can reduce the immediate roof subsidence in the initial mining period by 33.47% compared with the caving mining on the basis of saving the backfilling amount, and there is only a difference of 2.3% with the full backfill. At the same time, the backfilling body at the key position can effectively response the stress concentration caused by the rotation and subsidence of the roof, maintain its stability, and realize the long-term support for the roof. After comprehensive analysis, the best backfilling scheme for the “I-shaped” constructional backfill in the first coal face of Xinyang Mine is 72 m in length and 7 m in width in the middle strip, and 18 m in length and 6 m in width in the two side strips. Compared with full backfill, the cost of backfilling materials can be reduced by 0.338 6 million yuan. The scheme creates a confined space of 1980 m3, or 44% of the total mining space, which can be used for CO2 or carbonized material sequestration, helping to achieve zero-carbon green mining and promote dual-carbon goals.

Highlights The soft 2D material reduces the coupling strength between carriers and longitudinal optical phonons, releasing the mechanical stress of lattice vibration. The power conversion efficiency of rigid devices and flexible devices reaches 25.5% and 23.4%, respectively.

Abstract Irradiation (IR) induces immunogenic cell death (ICD) in tumors, but it rarely leads to the abscopal effect (AE); even combining IR with immune checkpoint inhibitors has shown only anecdotal success in inducing AEs. In this study, we aimed to enhance the IR-induced immune response and generate reproducible AEs using the anti-alcoholism drug, disulfiram (DSF), complexed with copper (DSF/Cu) to induce tumor ICD. We measured ICD in vitro and in vivo. In mouse tumor models, DSF/Cu was injected intratumorally followed by localized tumor IR, creating an in situ cancer vaccine. We determined the anticancer response by primary tumor rejection and assessed systemic immune responses by tumor rechallenge and the occurrence of AEs relative to spontaneous lung metastasis. In addition, we analyzed immune cell subsets and quantified proinflammatory and immunosuppressive chemokines/cytokines in the tumor microenvironment (TME) and blood of the vaccinated mice. Immune cell depletion was investigated for its effects on the vaccine-induced anticancer response. The results showed that DSF/Cu and IR induced more potent ICD under hypoxia than normoxia in vitro. Low-dose intratumoral (i.t.) injection of DSF/Cu and IR(12Gy) demonstrated strong anti-primary and -rechallenged tumor effects and robust AEs in mouse models. These vaccinations also increased CD8+ and CD4+ cell numbers while decreasing Tregs and myeloid-derived suppressor cells in the 4T1 model, and increased CD8+, dendritic cells (DC), and decreased Treg cell numbers in the MCa-M3C model. Depleting both CD8+ and CD4+ cells abolished the vaccine’s anticancer response. Moreover, vaccinated tumor-bearing mice exhibited increased TNFα levels and reduced levels of immunosuppressive chemokines/cytokines. In conclusion, our novel approach generated an anticancer immune response that results in a lack of or low tumor incidence post-rechallenge and robust AEs, i.e., absence of or decreased spontaneous lung metastasis in tumor-bearing mice. This approach is readily translatable to clinical settings and may increase IR-induced AEs in cancer patients.

Abstract Asian soybean rust (ASR), caused by Phakopsora pachyrhizi, is a devastating disease that is present in all major soybean-producing regions. The limited availability of resistant germplasm has resulted in a scarcity of commercial soybean cultivars that are resistant to the disease. To date, only the Chinese soybean landrace SX6907 has demonstrated an immune response to ASR. In this study, we present the isolation and characterization of Rpp6907-7 and Rpp6907-4, a gene pair that confer broad-spectrum resistance to ASR. Rpp6907-7 and Rpp6907-4 encode atypic nucleotide-binding leucine-rich repeat (NLR) proteins that are found to be required for NLR-mediated immunity. Genetic analysis shows that only Rpp6907-7 confers resistance, while Rpp6907-4 regulates Rpp6907-7 signaling activity by acting as a repressor in the absence of recognized effectors. Our work highlights the potential value of using Rpp6907 in developing resistant soybean cultivars.

During the installation of a cable oil terminal, it is easy to leave scratches on the main insulation owing to uneven forces when removing the semi-conductive layer. Scratch defects cause field intensity distortion, which leads to partial discharge and insulation failure. This study attempts to establish a simulation model of a 220 kV cable terminal to determine the effect of the length, depth, and position of the scratch on the maximum field strength at the defect. The simulation and experiment demonstrate that the maximum field strength at the defect increases with greater length and decreases as the depth increases. Therefore, a prediction method for the terminal defect field strength based on a multivariate nonlinear regression model was proposed in this study. When the defect is located at 20 mm from the root of the stress cone, the maximum field strength is 14.5 MV/m when the length and depth are 2 mm and 1 mm, respectively. The maximum field strength at the defect was predicted based on the length, depth, and position of the scratch defect to evaluate the severity of the defect.

The model of the diffusion bonding pressure was established using the analysis calculation method and further verified by the finite element analysis (FEA) and the experiments. The normalized temperature was defined as the ratio of temperature and the melting point of the base materials. And the normalized bonding pressure was defined as the ratio of the applied bonding pressure and the yield strength at the bonding temperature. The contribution of the plastic deformation to the void closure expressed a linear relationship with the normalized bonding pressure, of which the slope was 3. The relationship was appropriate in TiAl6V4, pure copper C11000, aluminium alloy AA6061, stainless steel SUS 304, and Ni-based alloy Inconel 617 using analysis calculation. The diffusion bonding pressure design range can be summarized as σn = 0.05–0.577. Subsequently, the analytic computational model was verified by FEA. The results showed that the maximum stress was concentrated in the position of the void neck. And the linear relationship between the normalized bonding pressure and the bonded ratio was also tenable. There was a stable static error existed between FEA and the analytic computational analysis. Furthermore, the experimental verification showed the verification and accuracy of the analytic computational analysis results.

Flexible electronic devices with mechanical properties like the soft tissues of human organs have great potential for the next generation of wearable and implantable electronic devices. Self-healing hydrogel composites typically have high tensile strength, high electrical conductivity and damage repair properties and have wide applications in flexible electronics, such as human-computer interaction, health detection and soft robots. Various self-healing hydrogel composites have been developed to produce new stretchable conductive materials with satisfactory mechanical and self-healing properties. This paper presents the fabrication of self-healing hydrogel composites and their application in flexible electronic devices. Firstly, the repair mechanism of physically cross-linked and chemically cross-linked self-healing hydrogel composites is presented. Secondly, self-healing double network hydrogels, self-healing nanocomposite hydrogels and double crosslinked self-healing hydrogel composites and their applications in flexible sensors, energy harvesting devices, energy storage devices and optical devices are presented and discussed. Finally, the challenges and prospects of self-healing hydrogel composites in flexible electronic devices in the future are presented.

ABSTRACT: Increased consumption of folic acid is prevalent due to its beneficial effects, but growing evidence emphasizes the side effects pointing to excessive dietary folate intake. The effects of excessive paternal folic acid consumption on offspring and its transgenerational inheritance mechanism have not been elucidated. We hypothesize that excessive folic acid consumption will alter sperm DNA N6-methyladenine (6mA) and 5-methylcytosine (5mC) methylation and heritably influence offspring metabolic homeostasis. Here, we fed roosters either folic acid-control or folic acid-excess diet throughout life. Paternal chronic folic acid excessive supplementation increased hepatic lipogenesis and lipid accumulation but reduced lipolysis both in the roosters and their offspring, which was further confirmed to be induced by one-carbon metabolism inhibition and gene expression alteration associated with the Peroxisome proliferator-activated receptor pathway. Based on the spermatozoal genome-wide DNA methylome identified by Nanopore sequencing, multi-omics association analysis of spermatozoal and hepatic DNA methylome, transcriptome, and metabolome suggested that differential spermatozoal DNA 6mA and 5mC methylation could be involved in regulating lipid metabolism-related gene expression in offspring chickens. This model suggests that sperm DNA N6-methyladenine and 5-methylcytosine methylation were involved in epigenetic transmission and that paternal dietary excess folic acid leads to hepatic lipid accumulation in offspring.

Abstract Background This study investigated clinical and laboratory characteristics of human bocavirus type 1 (HBoV1)‐plastic bronchiolitis (PB), Mycoplasma pneumoniae (MP)‐associated plastic bronchitis (PB) and MP‐NPB in children, highlighting inflammation, coagulation, and bronchoscopic needs. Methods Data on preschool children with PB during HBoV1 or MP infection were collected, comparing MP‐PB to severe Mycoplasma pneumoniae pneumonia.
