For more than 20 years, researchers have isolated and identified postnatal dental pulp stem cells (DPSCs) from different teeth, including natal teeth, exfoliated deciduous teeth, healthy teeth, and diseased teeth. Their mesenchymal stem cell (MSC)-like immunophenotypic characteristics, high proliferation rate, potential for multidirectional differentiation and biological features were demonstrated to be superior to those of bone marrow MSCs. In addition, several main application forms of DPSCs and their derivatives have been investigated, including stem cell injections, modified stem cells, stem cell sheets and stem cell spheroids.

Intrinsic dual-emission (DE) of gold nanoclusters in the near-infrared (NIR) are fascinating for fundamental importance and practical applications, but their synthesis remains a formidable challenge and sophisticated excited-state processes make elucidating DE mechanisms much more arduous. Here, we report an all-alkynyl–protected gold nanocluster, Au20 , showing a prolate Au 12 tri-octahedral kernel surrounded by two Au 2 (CZ-PrA) 3 dimers, four Au(CZ-PrA) 2 monomers, and two CZ-PrA − bridges. Au20 exhibits distinguished photophysical properties including NIR DE at 820 and 940 nm, microsecond radiative relaxation, and 6.26% photoluminescent quantum yield at ambient environment in nondegassed solution. Combining systematic studies on steady/transient spectroscopy and theoretical calculation, we identified two triplet charge transfer (CT) states, ligand-to-kernel and kernel-based CT states as DE origins. Furthermore, this NIR DE exhibits highly independent and sensitive response to surrounding environments, which well coincide with its mechanism. This work not only provides a substantial structure model to understand a distinctive DE mechanism but also motivates the further development of NIR DE materials.

Extracellular vesicles can be released by almost all types of cells and are important mediators of intercellular signal transmssion. Extracellular vesicles regulate the function and activity of recipient cells by delivering biologically active molecules such as proteins and nucleic acids, which is of great significance in tissue repair and regeneration. According to numerous studies, extracellular vesicles derived from endothelial/endothelial progenitor cells can induce cell proliferation and differentiation, inhibit cell apoptosis, and promote angiogenesis, playing an increasingly important role in regenerative medicine. We reported in this review the latest findings on applying extracellular vesicles derived from endothelial/endothelial progenitor cells in tissue regeneration and repair, and discussed the challenges and future development directions of their application in the field of regenerative medicine.

Microspheres have been widely utilised as versatile carriers in biomedical applications. In recent years, as a new type of injectable scaffold, microspheres have attracted increasing attention in the field of regenerative medicine owing to their various advantages including their small size, large specific surface area and mimicry of the 3D native environment. These characteristics enable them to adopt the narrow and irregular anatomy of the tooth and become an ideal scaffold for endodontic regeneration. Microspheres play an important role in carrying biologics (cells, biomolecules and drugs), which effectively regulate the fate of stem cells and control the release of growth factors and drugs. Cell-laden microspheres, which can be divided into microcarriers and microcapsules, have great application prospects in dental pulp regeneration. This paper summarises the properties and characteristics of microsphere scaffolds used in tissue engineering, placing emphasis on their advantages and applications in endodontic regeneration.

To investigate the role of a novel chemically defined medium (CDM) in the regulation of dental papilla cells (DPCs) functional phenotype in vitro and periodontal bone regenerationDPCs were isolated and cultured in conventional medium (CM) or CDM. The surface makers, and the proliferation, migration and osteogenic differentiation abilities of DPCs were evaluated.DPCs in the CM group and CDM group showed similar surface markers. Compared to the CM group, the CDM significantly enhanced the proliferation, colony-forming efficiency and migration of DPCsCDM is a suitable medium to culture DPCs for periodontal bone regeneration. This research provided a substitute for basic research and set the stage for future clinical application of stem cell transplantation.

Numerous studies have demonstrated that plant species diversity enhances ecosystem functioning in terrestrial ecosystems, including diversity effects on insects (herbivores, predators and parasitoids) and plants. However, the effects of increased plant diversity across trophic levels in different ecosystems and biomes have not yet been explored on a global scale. Through a global meta-analysis of 2,914 observations from 351 studies, we found that increased plant species richness reduced herbivore abundance and damage but increased predator and parasitoid abundance, predation, parasitism and overall plant performance. Moreover, increased predator/parasitoid performance was correlated with reduced herbivore abundance and enhanced plant performance. We conclude that increasing plant species diversity promotes beneficial trophic interactions between insects and plants, ultimately contributing to increased ecosystem services.

Deformities of the maxillofacial region following trauma and ablative surgery are devastating and not uncommon. Reconstruction of such defects is a surgically challenging procedure. Conventionally, reconstruction of dental arch defects lacks preoperative customised planning and relies heavily on the surgeon experience to ensure optimum surgical outcomes. The restoration of the dental arch shape and function has taken precedence after an extensive tumour resection surgery, especially in the current age of technological advancement. Thus, personalised and accurate reconstruction of dental arch defects has become a new goal. Computer-assisted surgery, especially navigation-assisted surgery, has gained popularity of late, in reconstructing deformities and restoring facial symmetry, appearance and function in the maxillofacial region. This technology provides a clearer three-dimensional visualisation of the area of interest and its relationship with the adjacent vital structures. Together with preoperative virtual surgical planning, it allows more specific and accurate osteotomies, thus reducing the ischemia and total operating times substantially. The risk of complications is also minimised whilst improving the final surgical outcomes. The use of the intraoperative navigation system and other computer-assisted surgical techniques during surgery can significantly improve the precision of the reconstruction of dental arch deformities, and achieve personalised and functional reconstructive goals while enhancing the quality of life of patients postoperatively. The Society of Oral and Maxillofacial Surgery, Chinese Stomatological Association provides the present professional perspective and treatment protocol for navigation-guided reconstruction of dental arch defects, to allow standardisation of the technique while promoting its application among oral and maxillofacial surgeons.

Skull base surgery is an interdisciplinary subject. The anatomical structures in the skull base related to oral and maxillofacial surgery include the parapharyngeal space, the pterygopalatine fossa and the infratemporal space. This operative area is one of the most challenging surgical areas in oral and maxillofacial surgery due to its deep site, complex anatomy and high risk. Obtaining pathological information of the tumour preoperatively may help surgeons optimise their treatment plan. Needle biopsy is one of the major minimally invasive techniques that allows preoperative pathological results to be obtained. The navigation technology, which is developing rapidly nowadays, provides a reliable assistance for deep tissue biopsy surgery. Experts from the Society of Oral and Maxillofacial Surgery, Chinese Stomatological Association formulated an expert consensus on the procedures and operations of navigation-guided needle biopsy techniques for skull base tumours, so as to standardise and promote the application and operation of navigation-guided needle biopsy for skull base tumours.

Orbital fracture and orbital floor reconstruction surgery is characterised by limited surgical exposure and high risks. Before the advent of digital technology, the design and implementation of the surgical scheme for orbital floor reconstruction surgery mainly depended on the intuitive understanding of imaging and the clinical experience of surgeons, with strong unpredictability and experience dependence. Surgical navigation systems with real-time positioning and imaging functions, when used in orbital reconstruction surgery, can detect the real-time intraoperative position, avoid risks, and assist in locating the reconstruction position to ensure the accuracy of orbital reconstruction, with the help of a preoperative surgical plan. Many studies have confirmed its effect. Unilateral orbital fracture and orbital floor reconstruction surgery is one of the earliest and most widely used surgical techniques in maxillofacial surgery. Experts from the Society of Oral and Maxillofacial Surgery, Chinese Stomatological Association have fully discussed and formulated this expert consensus on navigation-guided unilateral orbital fracture and orbital floor reconstruction techniques to standardise the clinical surgical procedures and promote its application.

The zygoma is located in the medial and lateral parts of the face, supporting the midfacial contour. The forward projection of the zygoma and the zygomatic arch often expose them to injury. Fractures of the zygoma can lead to the displacement of the zygoma and the zygomatic arch, causing facial collapse deformity. For delayed zygomatic fractures, the loss of normal anatomical landmarks caused by the malunion of the fracture lines and remodelling of the bony contour makes it difficult to determine the correct positions of the zygomatic bones. In such cases, ideal and steady outcomes with satisfactory midface symmetry have been difficult to obtain using traditional surgical methods. Nowadays, the application of digital surgical software and surgical navigation helps surgeons to perform accurate preoperative simulations to obtain ideal three-dimensional virtual surgical plans and achieve accurate reduction by intraoperative navigation systems, which increase the accuracy and predictability of fracture reduction outcomes. Experts from the Society of Oral and Maxillofacial Surgery, Chinese Stomatological Association have fully discussed and formulated an expert consensus on navigation-guided unilateral delayed zygomatic fracture reconstruction techniques to standardise the clinical operation procedures and promote the application.

The pigment responses of human skin to broadband UVA radiation (320-400 nm) occur in three distinct phases. The first phase includes immediate pigment darkening (IPD), the pigment that appears immediately after irradiation. The second phase involves an intermediate step, termed persistent pigment darkening (PPD), which leads to the third phase of neomelanogenesis or delayed tanning (DT). Since DT results from synthesis of new melanin, it persists beyond 5-7 days. We conducted studies on human subjects to investigate the dynamic responses of the IPD and PPD reactions to broadband UVA radiation at threshold and superthreshold doses. The threshold doses for IPD, PPD, and DT were found to be approximately 1, 11, and 18 J/cm2 , respectively. The colorimetry ΔL* value corresponding to minimal clinically perceptible pigmentation was found to be 0.8 ± 0.1. IPD appeared immediately and had an associated decay constant of approximately 1.4 minutes. At doses greater than PPD threshold, IPD reaction decayed while PPD developed indicating toward IPD being used as a substrate in the formation of PPD.

The aim of this study was to evaluate the accuracy of novel software-CMF-preCADS-for the prediction of soft tissue changes following repositioning surgery for zygomatic fractures. Twenty patients who had sustained an isolated zygomatic fracture accompanied by facial deformity and who were treated with repositioning surgery participated in this study. Cone beam computed tomography (CBCT) scans and three-dimensional (3D) stereophotographs were acquired preoperatively and postoperatively. The 3D skeletal model from the preoperative CBCT data was matched with the postoperative one, and the fractured zygomatic fragments were segmented and aligned to the postoperative position for prediction. Then, the predicted model was matched with the postoperative 3D stereophotograph for quantification of the simulation error. The mean absolute error in the zygomatic soft tissue region between the predicted model and the real one was 1.42±1.56mm for all cases. The accuracy of the prediction (mean absolute error ≤2mm) was 87%. In the subjective assessment it was found that the majority of evaluators considered the predicted model and the postoperative model to be 'very similar'. CMF-preCADS software can provide a realistic, accurate prediction of the facial soft tissue appearance after repositioning surgery for zygomatic fractures. The reliability of this software for other types of repositioning surgery for maxillofacial fractures should be validated in the future.

Surgical guides have been used widely in maxillofacial surgery. Details of the types of digital surgical guide used in mandibular resection and reconstruction with fibula flaps at the authors' institution are presented in this article. Three patients diagnosed with mandibular ameloblastoma underwent mandibular resection and reconstruction with fibula flaps using surgical guides for assistance. These digital surgical guides included a mandibular osteotomy guide, a fibular osteotomy guide, and a mandibular fixation guide. Surgical guides are helpful in improving the accuracy of operations and are appropriate for many types of mandibular resection and reconstruction.

This study aimed to investigate the clinical phenotype and genetic characteristics of Chinese families with Van der Woude syndrome (VWS).Clinical manifestations between 14 families and within each family were recorded. Possible inheritance modes and pathogenic genes were analyzed. Phenotypic distribution and gene frequencies were calculated.Of the pedigrees investigated, an autosomal dominant inheritance pattern was suggested. All patients had typical symptoms. The pathogenic gene was interferon regulatory factor 6 (IRF6). Phenotypic distribution frequencies were as follows: lip pits (91.9%), cleft lip and/or palate (73.0%), and hyperdontia (8.1%). There were significant differences in clinical phenotypes among individuals of different families and individuals of the same family.VWS in a Chinese population was dominantly inherited with high penetrance and variable expressivity. The pathogenic gene was IRF6. VWS in a Chinese population was genotyped as VWS1.目的 探讨中国人Van der Woude综合征（VWS）的临床表型及遗传学特点。方法 先证者法收集14个VWS家系并进行口腔专科检查、家系调查及基因突变分析，分析不同VWS家系个体或同一家系不同个体的临床表型，绘制家系图谱，明确遗传方式及致病基因，计算表型分布频率和表型基因频率。结果 VWS家系基本符合常染色体显性遗传特征，患者多数表现为典型的VWS，致病基因为干扰素调节因子6（IRF6）。VWS表型分布频率为：唇瘘91.9%，唇腭裂73.0%，牙畸形8.1%。不同家系个体和同一家系的不同个体临床表型存在明显差异。结论 收集的家系均为常染色体显性遗传，表现度变异大。中国人群VWS致病基因为IRF6，为Ⅰ型VWS。.

Fractures of the coronoid process are uncommon and can easily be missed. The purpose of this study was to classify the fracture patterns and explore the treatment options. This retrospective study included 39 patients with fractures of the mandibular coronoid process. Treatment protocols were developed based on the time of fracture, degree of mouth opening, location of the coronoid fracture, types of fracture, and other concomitant fractures. All patients were followed up for 12-60 months. Sixteen patients underwent conservative management and four of these patients developed progressive trismus, which improved significantly after removal of the coronoid process. Twenty-three patients underwent open reduction and internal fixation (ORIF) via the modified retromandibular approach. Follow-up data showed significant improvement in maximum mouth opening and symptoms (diet and pain) compared to their preoperative status. In summary, conservative management is first recommended for fractures of the coronoid process with minimal displacement or restriction of mouth opening. For patients with significant fracture displacement and limited mouth opening, or with concomitant fractures of the zygoma, zygomatic arch, or mandibular ramus, ORIF via the modified retromandibular approach through the anterior border of the parotid gland is an alternative treatment method.

Objective The aim of this study was to analyze the clinicopathologic characteristics and prognostic factors of adenoid cystic carcinoma of the head and neck (ACCHN). Study Design This was a retrospective study of 218 patients with ACCHN. Results The cohort included 110 men and 108 women; the parotid and the palate were the most common site of involvement. Of 203 patients with follow-up information (range 2-132 months), 57 had died of the tumor. Distant metastasis (DM) and local recurrence (LR) were documented in 83 (40.9%) and 34 (16.7%) patients, respectively. Cox regression analysis indicated that a solid pattern was a marker for LR and that positive margins and older age were risk factors for DM. Histologic pattern, T stage, N stage, LR, DM, and patient age contributed to the prediction of disease-specific survival. Conclusions A solid pattern, metastasis, LR, and older age are the most important factors for predicting poor prognosis in Chinese patients with ACCHN.

Different from the other distribution center location methods, there are new characteristics in distribution center location during earthquake disaster responses. In this paper, we investigate the factors impacting the location of distribution center during earthquake responses, and propose a decision-making model based on improved AHP framework for this issue. The improved AHP is extended to automatically calculate large scale judgment matrices by the crosstie model. This decision-making model was successfully applied in the simulation case of Yushu earthquake responses in 2010. The results show that the proposed decision-making model is reasonable.

The cartilage tissue engineering is an inspiring and profitable way for the reconstruction of cartilage defects, but it has been hampered by two large obstacles: how to get qualified seed cells and credible scaffold. This study aimed to evaluate the chondrogenic potential of rat bone marrow stromal cells (BMSCs) by loading them on alginate gel. In this study, the compounds of SD rat BMSCs and alginate gel were injected on the dorsum of rats subcutaneously. The implantations were harvested and examined by histological and immunohistochemical examination, in situ hybridization and transmission electron microscopy at different time points after the operations. The results showed that the compounds of BMSCs and alginate gel are promising for cartilage tissue engineering applications.

Bone tissue engineering is a promising way to repair of bone defects. To choose a proper scaffold is still a disputable problem in bone tissue engineering. This study aimed to compare the effects of repairing critical calvarial defects with the compounds of autogenous bone marrow stromal cells (BMSCs) and coral hydroxyapatite(CHA), hydroxyapatite/ tricalcium phosphate (HA/TCP), poly(lactide-co-glycolide) (PLGA) and alginate (AG). The results showed that CHA and AG were satisfactory bone tissues engineering scaffolds among the four kinds of materials. BMSCs/CHA and BMSCs/AG are promising techniques for reconstruction of bone defects.

To investigate the mechanical property (shear strength, bending strength and compressive strength) and the interface combinative ability of the composite of bovine bone morphogenetic protein (bBMP) and porous hydroxyapatite (HA) with different pore diameter. HA1 (Φ100-300μm) and HA2 (Φ130-500μm) were used as carriers of bBMP(5mg). The 4 types material (HA1, HA2, bBMP /HA1, bBMP /HA2) were implanted into the femur of 10 dogs (2 dogs each group) for the shear test and 15 dogs (3 dogs each group) for the bending test and compressive test. The shear strength of all kinds of materials were increased as time went on and the shear strength of bBMP/HA1 was highest in four materials. The bending strength of bBMP /HA1 was close to the normal femur in the 24th week. The compressive strength of all materials was lower than the cortex of the dog’s femur until 24th week. The compressive strength of bBMP /HA1 was the highest in four types. The compressive strength of HA2 and bBMP /HA2 was decreased after implantation. Implant of bBMP /HA with 100-300μm diameter had strong bonding with the host bone, and had better mechanical properties than that with 130-500μm diameter. So the porous bBMP /HA (Φ100-300 μm ) could be a useful graft material due to its strength.

This study was to determine the ectopic osteogenic ability of BMSCs in combination with a scaffolding material comprising hydroxyapatite and β-tricalcium phosphate matrix (HA/β-TCP). BMSCs were obtained from the SD rats and induced to osteogenesis. Then these induced cells were seeded into HA/β-TCP and the constructs were auto-implanted subcutaneously for up to 12 weeks. Histological analysis, immunostaing, RT-PCR and transmission electron microscopy of the retrieved specimens at various intervals showed obvious trends of ectopic bone formation with obvious alteration of cellular phenotype.

Poly(lactide-co-glycolide) (PLGA) and alginate(AG) are the most promising scaffolds in the bone tissue engineering for their stable mechanical characters and three-dimensional porous structure. This study aimed to assay the in vivo osteogenesis potentials by loading the autogenous bone marrow stromal cells (BMSCs) on PLGA or AG. The results suggested that PLGA and AG are both ideal bone tissue engineering scaffold. BMSCs/AG has stronger osteogenesis potentials in vivo than BMSCs/PLGA.

To optimize the extraction method of secretory factors from adipose tissue explant (SFAE) in vitro.SFAE were obtained through adherent culture (SFAE-A) and suspension culture (SFAE-S) and concentrated by filtration and centrifugation. The yield of SFAE was compared using BCA protein detection kit. P3 adipose-derived stem cells (ADSCs) were induced with equal amount of SFAE for 7-13 d, before the state of adipogenesis between suspension culture and adherent culture was compared by microscope observation and oil red O staining.The average amount of SFAE yielded from adherent culture and suspension culture did not show significant difference. While the yield of SFAE from suspension culture was consistent at 8. 7 mg per gram of adipose tissue, the adherent culture generated an inconsistent result in the four repeat experiments, ranging from 7. 3 mg to 12. 4 mg per gram of adipose tissue. Moreover, ten more flasks and better distribution were needed for adherent culture to acquire an equal amount of SFAE in comparison with suspension culture. SFAE from both adherent and suspension culture promoted the adipogenesis of P3 adipose-derived stem cells. No differences on the adipogenic effect were found between the two extraction methods.Secretory factors from adherent culture and suspension culture have the same adipogenesis effect. Suspension culture can save time and labor. The most important advantage of suspension culture is its stable yield of SFAE.

To develop a new threshold segmentation method for mandible image segmentation.CT data of 12 volunteers were exported into Mimics 10. 01. An improved method usinga narrowed threshold range (the maximum threshold range that can segment mandible without manual efforts) was developed in 3D reconstruction, and compared with the traditional method. We used dilation operations to make up the information loss of image borders, by which we obtained an approxinate segment result. A precise segment resultwas eventually arrived with the help of logical operations and region growing. We compared mean time consumptions of the two methods, as well as their 3D reconstruction results using Geomagic Studio 11. 0.The new method generated a success rate of 91. 67% (11/12), with a mean time consumption of (319. 7±125. 3) s. The traditional method took much longer time [(1,261. 3±427. 3) s, P0. 05] than the new method. Compared with the reconstruction results of traditional method, the new method had an outward deviation of (0. 066±0. 011) mm and an inward deviation of (0. 070±0. 008) mm. Such deviations were less than the minimum distance that a naked eye can discern. The lower limit of the widest threshold range which mandible could be isolated was (507. 72± 100. 31) HU, while the upper limit was (1,133. 33±47. 57) HU.The new method we proposed can improve the efficiency of threshold segmentation of mandible.

Previously, we found that electromagnetic pulses (EMP) induced an increase in blood brain barrier permeability and the leakage of albumin from blood into brain tissue. Albumin is known to activate microglia cells. Thus, we hypothesised that microglia activation could occur in the brain after EMP exposure. To test this hypothesis, the morphology and secretory function of microglia cells, including the expression of OX-42 (a marker of microglia activation), and levels of TNF-α, IL-10, IL-1β, and NO were determined in the rat cerebral cortex after EMP exposure. In addition, to examine the signalling pathway of EMP-induced microglia activation, protein and phosphorylated protein levels of p38, JNK and ERK were determined. To determine the effects of exogenous nitric oxide (NO) donor sodium nitroprusside (SNP) on the proliferation and expression of related-gene of adipose-derived stem cells(ADSCs) and its role in chondrogenic differentiation of ADSCs.Rat ADSCs were harvested and cultured, and then induced to osteogenic and adipogenic differentiations, detected with Alizarin red stained and Oil red O stain, respectively. The change of NO during chondrogenic differentiation of ADSCs was tested by NO detection kit. Cell counting kit-8 (CCK8) was used to detect the proliferation of ADSCs under different concentrations of SNP (0.25 mmol/L, 1.00 mmol/L, 4.00 mmol/L). Gene expression level of transformation growth factor (TGF-beta1) and specific gene of chondrogenic differentiation-signaling protein Smad3 and Collage II alPHA (Col-II alpha1), were detected by Real time- PCR (RT-PCR) method.Positive alizarin red staining and Oil red O staining were found after osteogenic and adipogenic induction of cultured ADSCs. Higher concentrations of NO were found in the supernatant of the experimental group with ADSCs chondrogenic differentiations compared with the controls (P0.05). Low concentrations (0. 25 mmol/L, 1.00 mmol/L) of SNP showed no significant effects on cell proliferations (P0.05), whereas high concentration (4. 00 mmol/L) of SNP inhibited cell proliferation (P0.05). RT-PCR revealed that SNP inhibited the gene expression of TGF-beta1 mRNA and chondrogenic differentiation of specific gene Smad3 mRNA, Col-II alpha1 mRNA.SNP can inhibit chondrogenic differentiations by suppressing the production of TGF-beta1 and inhibiting downstream of TGF-beta1 signaling pathways, thereby inhibiting ADSCs differentiation into chondrocytes.

Dendritic cells (DCs), as primary antigen-presenting cells with the capacity to activate naïve T lymphocytes, are considered to be promising adjuvants for immunity against cancer. In this study, the effect of T lymphocyte-mediated immunity induced by a DC vaccine against Tca8113 cells in vivo and in vitro was evaluated. DCs were from human peripheral blood monocytes cultured in the presence of granulocyte-macrophage colony stimulating factor, interleukin 4 and/or tumour necrosis factor alpha, and stimulated with Tca8113 cell lysate prepared by the freeze-thaw method. The autologous T cells activated by these DCs were used in an in vitro MTT assay to detect their tumouricidal activity and investigated for their anti-tumour effect in vivo by administration to nude mice with implanted tumours (Tca8113). An adequate number of DCs were successfully generated from the monocytes. The T cells activated by the DC-based vaccine killed Tca8113 cells in vitro (P0.01), postponed tumour doubling time of the implanted tumours in nude mice (P0.01) and inhibited the growth of the tumours (P0.05). These results show that DCs from monocytes induce a lymphocyte-mediated immune response against tongue squamous carcinoma, and could be used as a vehicle for tumour antigens.

OBEJECTIVE: To construct miRNA-expressing plasmid vector for interfering the expression of beta-catenin in adipose-derived stem cells from Sprague-Dawley rats.The double strands oligonucleotides of miR-expressing cassette were ligated into plasmid pSWH to generate pSWH-miR. Then the PCR product of enhanced green fluorescence protein (EGFP) open reading frame (ORF) fragment was inserted into BamH I and Xbo I digested pSWH-miR to generate pSWH-EGFP-miR. The adipose-derived stemcells from rats (rADSCs) were transfected with pSWH-EGFP-miR respectively. The expression of beta-catenin was determined by Western blot at 72 h post-transfection.Five miRNA-expressing plasmid vectors were constructed (miR-780, miR-796, miR-1467, miR-1948, miR-1960). miR-780 and miR-796 had the best silencing effect on the expression of beta-catenin (P0.05), less did the miR-1960 (P0.05), but not the miR-1467 and the miR-1948 (P0.05).miR-780 and miR-796 could interference the beta-catenin in ADSCs with high efficiency.

Navigation surgery in the mandible has rarely been reported because of the complexities of navigating a mobile structure. In this article, we present a simple and novel strategy for removal of a foreign body in the mandible using a navigation system. A female diagnosed with a foreign body in the left mandible underwent navigation surgery using a BrainLAB system. We used a special open splint fabricated with acrylic resin to successfully perform the mandibular navigation. This strategy may be appropriate for many types of mandibular navigation surgery.

Hydroxyapatite-tricalcium phosphate (HA-TCP) is a new kind of material which shows good biocompatibility, biological degradability, and porosity. This study aimed to determine the effectiveness of HA-TCP as a bone tissue engineering scaffold. In this study, critical size cranial defects were reconstructed with compounds of autogenous bone marrow stromal cells (BMSCs) and HA-TCP. The resulting grafts were examined by X-ray, histological examination, semi-quantitative analysis of osteogenesis, immunochemical examination (collagen type I and III), scanning electron microscopy and transmission electron microscopy. The results showed that HA-TCP is a good bone tissue engineering scaffold and BMSCs/HA-TCP is a promising technique for reconstruction of bone defects.

To investigate the effects of human treated dentin matrix (hTDM) extracellular matrix molecules on odontogenetic and neural differentiation of human dental pulp cells (hDPCs) with an aim to find an effective method to collect extracellular matrix molecules to contribute to reparation dental-pulp complex with dentin defects.hDPCs were obtained and biological characteristics such as source of cells and multi-differentiation potentials were assessed using immunofluorescence and flow cytometry. Fabrication of hTDM extracts and hDPCs was induced with it for 1 week. The odontogenetic differentiation associated genes were tested by qRT-PCR. Results qRT-PCR results showed that cells were higher expression of odontogenetic differentiation associated genes ALP, OPN, OCN, BSP, DMP-1, DSP, beta-III tubulin.The method of extracting extracellular matrix molecules from dentin matrix was effective. The extract liquid provides a suitable microenvironment for odontogenetic and neural differentiation of hDPCs and contributes to reparation dental-pulp complex.

To observe whether fetal bovine serum (FBS) will affect the adipogenic ability of adipose derived stem cells (ADSCs) induced by adipose tissue extract (ATE), and to explore the effects of different FBS protein concentrations in ATE on adipogenic ability, cell proliferation and migration of ADSCs.Rat ADSCs were cultured, passaged, and then subjected to osteogenic and neural induction. The adipose explants were cultured in culture medium with or without FBS, then ATE was collected to induce passage four (P4)-ADSCs, which were subsequently detected with oil red staining on the 7th day and the adipogenic ratio was calculated. Different concentrations of ATE without FBS (100 microg/mL, 250 microg/mL and 500 microg/mL) were used to induce P4-ADSCs before the adipogenic events, and the adipogenic ratio of each concentration group was observed. The effect of different protein concentrations on ADSCs proliferation and migration was also observed.Alizarin red staining and NF immunofluorescence staining were positive after the osteogenic and neural induction of cultured ADSCs. Either ATE with or without FBS was able to induce adipogenic differentiation of ADSCs on the 3rd day and there was no significant differences of adipogenic differentiation between ATE with FBS and without FBS on the 7th day. The adipogenic ratio of 500 microg/mL group was higher than that of 100 pg/mL group and 250 microg/mL group (P0.05). After two days of induction, all the three different protein concentrations could inhibit cell proliferation, and different protein concentrations in ATE had no effect on the migration of ADSCs.ATE obtained from culture medium without FBS has no effect on adipogenic capacity of ADSCs in a short period of time. The adipogenic ratio of ADSCs is associated with protein concentration in ATE.

To investigate the impact of combined adipose-derived stem cells (ASCs) and platelet-rich plasma (PRP) on the survival of transplanted fat.The ASCs were isolated and cultured from fat tissues by enzyme digestion; and the PRP was prepared by two-step ultracentrifugation. The grafts of fat granules were divided into test groups (ASCs + PRP + fat granules, PRP + fat granules, ASCs + fat granules) and control groups (PBS + fat granules). The grafts were injected into the left and right dorsal subcutaneous areas of nude mice. General observations, volume measurements and microscope examinations were conducted 10 d, 30 d, 60 d and 90 d after transplantation.The grafts of the mice in the ASCs + PRP group showed soft structure, with light yellow color and closer to normal adipose tissue compared with those in other groups. Greater survival volumes (P0.05) and better histology were also observed in the grafts of the mice in the ASCs + PRP group.The fat grafts consisting of PRP and ASCs constitute an ideal transplant strategy, which could provide a valuable and needed tool in plastic and reconstructive surgery.
