Objective: This study aims to investigate the therapeutic effect of Lactobacillus rhamnosus GG-loaded lemon pectin microspheres (LGG@LPM) on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in mice. Methods: LGG@LPM was characterized, and the therapeutic potential of gavage administration of LGG@LPM against UC in mice was explored. Results: LGG@LPM microspheres exhibited good stability. The administration of LGG@LPM significantly alleviated the pathological process of DSS-induced UC in mice, restoring normal body mass and colon length and decreasing histological activity index (HAI) and disease activity index (DAI). Additionally, LGG@LPM administration reversed the increase in inflammatory cytokine levels and the decrease in antioxidant enzyme activities in the colon, the increase in serum lipopolysaccharide (LPS) levels and the alterations in short-chain fatty acids (SCFAs) levels induced by DSS in mice. 16S rRNA sequencing revealed that LGG@LPM restored the gut microbiota in UC mice through enhancing the abundance of Lactobacillus and Mucispirillum, and the alteration of the microbiota was significantly correlated with some environmental factors. Conclusion: LGG@LPM could alleviate the pathological progression of UC by improving the gut microbiota, the effect being more pronounced than that of either lemon pectin or LGG alone.

The role of nanoscale chemomechanical behavior in the macroscopic performance of functional materials is well recognized. For lithium-ion battery cathodes, tremendous effort has been devoted to the development of new chemistry and structure, yet fundamental understanding of the correlation between redox processes and mechanical properties of the novel materials lags behind. In the present study, we prepare large discrete single grains of Li-excess cation-disordered rocksalts (DRX) and investigate their chemomechanical behavior at the particle level, using nanoresolution X-ray and electron-based spectro-imaging and chemical mapping techniques. While irregular cracking upon lithium extraction leads to the eventual breakdown of the baseline DRX oxide (Li1.2Ti0.4Mn0.4O2) particles at a high delithiation state, the fluorinated-DRX (Li1.3Ti0.3Mn0.4O1.7F0.3) clearly displays aligned cracking along the direction. The resulting periodicity in the cracking pattern enables the particles to retain their integrity and, consequently, improved electrochemical stability. Density functional theory (DFT) calculations showed that fluorination leads to increased concentration of Li+ on the (001) planes and preferential Li movements along the -family directions, revealing the underlying mechanism for directional cracking. Our study demonstrates the unique role of fluorine in modulating nanoscale chemomechanics, which in turn influences the evolution of charge and strain heterogeneity at the particle level. These insights provide important design guidelines in further improving DRX cathode materials.

Nickel-rich layered oxide cathode materials are attractive near-term candidates for boosting the energy density of next generation lithium-ion batteries. The practical implementation of these materials is, however, hindered by unsatisfactory capacity retention, poor thermal stability, and oxygen release as a consequence of structural decomposition, which may have serious safety consequences. The undesired side reactions are often exothermic, causing complicated electro-chemo-mechanical interplay at elevated temperatures. In this work, we explore the effects of thermal exposure on chemically delithiated LiNi0.8Mn0.1Co0.1O2 (NMC-811) at a practical state-of-charge (50% Li content) and an over-charged state (25% Li content). A systematic study using a suite of advanced synchrotron radiation characterization tools reveals the dynamics of thermal behavior of the charged NMC-811, which involves sophisticated structural and chemical evolution; e.g. lattice phase transformation, transition metal (TM) cation migration and valence change, and lithium redistribution. These intertwined processes exhibit a complex 3D spatial heterogeneity and, collectively, form a valence state gradient throughout the particles. Our study sheds light on the response of NMC-811 to elevated temperature and highlights the importance of the cathode's thermal robustness for battery performance and safety.

The structural integrity of layered Ni-rich oxide cathode materials is one of the most essential factors that critically affect the performance and reliability of lithium-ion batteries. Prolonged battery operation often involves repeated phase transitions, buildup of mechanical stresses, and could provoke thermal spikes. Such sophisticated chemo-thermo-mechanical interplay can cause performance degradation through structural disintegration of the cathode active materials (CAMs). Herein, we systematically investigate the thermal decomposition, fracture, and oxygen evolution of chemically delithiated Li0.3Ni0.8Co0.15Al0.05O2 (NCA) particles upon heating from 25 °C to 450 °C using a number of advanced X-ray and electron probes. We observed a continuous reduction of the Ni oxidation state upon heating, as well as the release of oxygen from the NCA lattice that undergoes the thermally induced phase transformations. The release of oxygen also created numerous mesopores throughout the analyzed particles, which could significantly affect the chemical and mechanical properties of the electrode. In addition, intergranular and intragranular fracturing at elevated temperatures also contribute to the degradation of the NCA cathode under these conditions. Our investigation of the mechanical integrity at elevated temperatures provides a fundamental understanding of the thermally driven chemomechanical breakdown of the NCA cathode active materials.

Featured Application: This work can be applied to estimate acceleration using measured inclination, withoutpartially measuring the target acceleration. Moreover, this is a finiteelement model-free method and can be conveniently applied to beam bridges. Data fusion is an important issue in bridge health monitoring. Through data fusion, specific unknown bridge responses can be estimated with measured responses. However, existing data fusion methods always require a precise finite element model of the bridge or partially measured target responses, which are hard to realize in actual engineering. In this study, we propose a novel data fusion method. Measured inclinations across multiple cross-sections of the target bridge and accelerations at a subset of these sections were used to estimate accelerations at the remaining sections. Theoretical analysis of a typical vehicle-bridge interaction (VBI) system has shown parallels with the blind source separation (BSS) problem. Based on this, Independent Component Analysis (ICA) was applied to derive surrogate inclination mode shapes. This was followed by calculating surrogate displacement mode shapes through numerical integration. Finally, a surrogate inter-section transfer matrix for both measured and unmeasured accelerations was constructed, enabling the estimation of the target accelerations. This paper presents three key principles involving the relationship between the surrogate and actual inter-section transfer matrices, the integration of mode shape functions, and the consistency of transfer matrices for low- and high-frequency responses, which form the basis of the proposed method. A series of numerical simulations and a large-scale laboratory experiment were proposed to validate the proposed method. Compared to existing approaches, our proposed method stands out as a purely data-driven technique, eliminating the need for finite element analysis assessment. By incorporating the ICA algorithm and surrogate mode shapes, this study addresses the challenges associated with obtaining accurate mode shape functions from low-frequency responses. Moreover, our method does not require partial measurements of the target responses, simplifying the data collection process. While Li ion batteries are intended to be operated within a mild temperature window, their structural and chemical complexity could lead to unanticipated local electrochemical events that could cause extreme temperature spikes, which, in turn, could trigger more undesired and sophisticated reactions in the system. Visualizing and understanding the response of battery electrode materials to thermal abuse conditions could potentially offer a knowledge basis for the prevention and mitigation of the safety hazards. Here we show a comprehensive investigation of thermally driven chemomechanical interplay in a Li0.5Ni0.6Mn0.2Co0.2O2 (charged NMC622) cathode material. We report that, at the early stage of the thermal abuse, oxygen release and internal Li migration occur concurrently, and are accompanied by mechanical disintegration at the mesoscale. At the later stage, Li protrusions are observed on the secondary particle surface due to the limited lithium solubility in non-layered lattices. The extraction of both oxygen and lithium from the host material at elevated temperature could influence the chemistry and safety at the cell level via rearrangement of the electron and ion diffusion pathways, reduction of the coulombic efficiency, and/or causing an internal short circuit that could provoke a thermal runaway.

Novel developments in X-ray sources, optics and detectors have significantly advanced the capability of X-ray microscopy at the nanoscale. Depending on the imaging modality and the photon energy, state-of-the-art X-ray microscopes are routinely operated at a spatial resolution of tens of nanometres for hard X-rays or ∼10 nm for soft X-rays. The improvement in spatial resolution, however, has led to challenges in the tomographic reconstruction due to the fact that the imperfections of the mechanical system become clearly detectable in the projection images. Without proper registration of the projection images, a severe point spread function will be introduced into the tomographic reconstructions, causing the reduction of the three-dimensional (3D) spatial resolution as well as the enhancement of image artifacts. Here the development of a method that iteratively performs registration of the experimentally measured projection images to those that are numerically calculated by reprojecting the 3D matrix in the corresponding viewing angles is shown. Multiple algorithms are implemented to conduct the registration, which corrects the translational and/or the rotational errors. A sequence that offers a superior performance is presented and discussed. Going beyond the visual assessment of the reconstruction results, the morphological quantification of a battery electrode particle that has gone through substantial cycling is investigated. The results show that the presented method has led to a better quality tomographic reconstruction, which, subsequently, promotes the fidelity in the quantification of the sample morphology.

Redox phase transformations are relevant to a number of metrics pertaining to the electrochemical performance of batteries. These phase transformations deviate from and are more complicated than the conventional theory of phase nucleation and propagation, owing to simultaneous changes of cationic and anionic valence states as well as the polycrystalline nature of battery materials. Herein, we propose an integrative approach of mapping valence states and constructing chemical topographies to investigate the redox phase transformation in polycrystalline layered oxide cathode materials under thermal abuse conditions. We discover that, in addition to the three-dimensional heterogeneous phase transformation, there is a mesoscale evolution of local valence curvatures in valence state topographies. The relative probability of negative and positive local valence curvatures alternates during the layered-to-spinel/rocksalt phase transformation. The implementation of our method can potentially provide a universal approach to study phase transformation behaviors in battery materials and beyond.

Formaldehyde (FA) is a human leukemogen and is hematotoxic in human and mouse. The biological plausibility of FA-induced leukemia is controversial because few studies have reported FA-induced bone marrow (BM) toxicity, and none have reported BM stem/progenitor cell toxicity. We sought to comprehensively examine FA hematoxicity in vivo in mouse peripheral blood, BM, spleen and myeloid progenitors. We included the leukemogen and BM toxicant, benzene (BZ), as a positive control, separately and together with FA as co-exposure occurs frequently. We exposed BALB/c mice to 3 mg/m3 FA in air for 2 weeks, mimicking occupational exposure, then measured complete blood counts, nucleated BM cell count, and myeloid progenitor colony formation. We also investigated potential mechanisms of FA toxicity, including reactive oxygen species (ROS) generation, apoptosis, and hematopoietic growth factor and receptor levels. FA exposure significantly reduced nucleated BM cells and BM-derived colony-forming unit-granulocyte-macrophage (CFU-GM) and burst-forming unit-erythroid (BFU-E); down-regulated GM-CSFRα and EPOR expression; increased ROS in nucleated BM, spleen and CFU-GM cells; and increased apoptosis in nucleated spleen and CFU-GM cells. FA and BZ each similarly altered BM mature cells and stem/progenitor counts, BM and CFU-GM ROS, and apoptosis in spleen and CFU-GM but had differential effects on other end points. Co-exposure was more potent for several end points. Thus, FA is toxic to the mouse hematopoietic system, including BM stem/progenitor cells, and it enhances BZ-induced toxic effects. Our findings suggest that FA may induce BM toxicity by affecting myeloid progenitor growth and survival through oxidative damage and reduced expression levels of GM-CSFRα and EPOR.

Background: Cancer stem cells (CSCs) are a specific subpopulation of cancer cells with the ability of self‐renewal, infinite proliferation, multidifferentiation and tumorigenicity, and play critical roles in cancer progression and treatment resistance. CSCs are tightly regulated by the tumor microenvironment, such as hypoxia; however, how hypoxia regulates CSCs in non‐small cell lung cancer (NSCLC) remains unclear. Methods: The proportion of ALDHhi cells was examined using the Aldefluor assay. Tankyrase inhibitor XAV939 and siRNA were used to inhibit β‐catenin while pcDNA3‐β‐catenin (S33Y) plasmid enhanced the expression of β‐catenin. Western blot was administered for protein detection. The mRNA expression was measured by quantitative real‐time PCR. Results: We found that hypoxia led to an increase in the proportion of ALDHhi cells in lung squamous carcinoma (LUSC) H520 cells, while causing a decrease in the ALDHhi cell proportion in lung adenocarcinoma (LUAD) A549 cells. Similarly, β‐catenin expression was upregulated in H520 cells but downregulated in A549 cells upon exposure to hypoxia. Mechanically, the proportion of ALDHhi cells in both cell lines was decreased by β‐catenin inhibitor or siRNA knockdown, whereas increased after β‐catenin overexpression. Furthermore, hypoxia treatment suppressed E‐cadherin expression in H520 cells and enhanced N‐cadherin and β‐catenin expression, while this effect was completely opposite in A549 cells. Conclusion: The hypoxia‐EMT‐β‐catenin axis functions as an important regulator for the proportion of CSCs in NSCLC and could potentially be explored as therapeutic targets in the future.
