9. Large genetic change at small fitness cost in large populations of Drosophila melanogaster for wind tunnel flight: rethinking fitness surfaces

Abstract

The fitness effects of extreme genetic change by selection were studied in large populations subjected to prolonged, intense selection. Two replicate populations of Drosophila melanogaster, with estimated effective sizes 500 [less than or equal to] [N.sub.e] [less than or equal to] 1000, were selected for increased performance in a wind tunnel, selecting on average the fastest 4.5% of flies. The mean apparent flying speed of both lines increased from [approximately]2 to 170 cm/sec and continued to respond at diminishing rates, without reaching a plateau, for 100 generations. Competitive fitness tests in generations 50 and 85 showed minimal or no fitness loss in selected lines compared to controls. Sublines relaxed in generations 65 and 85 showed minimal or no regression in apparent flying speed. Hybrid lines, from a cross of selected x control lines in generation 75, responded to reselection saltationally, showing that the chromosomes of the selected lines had been assembled from alleles at many loci, from many different chromosomes in the base population. Thus, major genetic change was achieved, but without the costs usually associated with strong directional selection. Large population size has been interpreted, in opposing models, as either a brake or an accelerator in its effects on long-term change by selection. These results favor the second model, and challenge the concept of rugged fitness surfaces underlying the first model.

18. Selection for increased flight performance in Drosophila melanogaster

Abstract

A selection experiment on upwind flight ability in Drosophilia melanogaster has inadvertently created an elegant system to study the constraints on the evolution of muscle performance. Selected flies have increased their mean apparent flight speed from ##10 to ##220 cm/s over 300 generations of selection. However, comparisons of free flight in still air by James Marden showed no difference in maximum flight speeds between selected and control lines but did show higher average flight speeds and a higher frequency of more horizontal flights. These differences, in combination with the apparent selection for both an attraction to the light at the end of the tunnel and a decreased hunker-down response to wind could explain the evolved increase in mean apparent flight speed. Using high-speed video, we found that flies make upwind progress in the compartmentalized wind tunnel by a series of short, burst flights and not by a single, long flight. To test the model that maximum flight performance has not responded to selection, we tested for burst-flight performance differences between selected and control lines by measuring the highest wind speed against which a fly could burst at least 5 cm off of a downwind screen. Selected flies could burst against 40% (males) and 37% (females) higher wind speeds, demonstrating that selection increased maximum flight performance. High-speed films of the burst behavior suggest that at least part of the failure to fly against the wind may be a function of the ability to rotate and stabilize the body into its fast-forward flight position. Mean mass is 32% larger in selected flies. We do not know if the selected flies achieve higher flight speeds by simply adding more muscle or if there been selection on musculoskeletal design to achieve higher mass specific muscle power or to increase control of maneuverability and stability.

Abstract

Participatory development activities at local level in a sub-district located in the Central Plain of Thailand were studied employing the theoretical concept of the "linking loops" to analyze the related interactions among target group members and between the local level and superordinated organizations.The participatory development planning approach was first introduced in Thailand through her Fourth National Economic and Social Development Plan (1977-1981) as a strategy to apply the bottom-up concept.The qualitative, in-depth study covers nine projects at different stages of the planned development process alternatively covering the stages of initiation, implementation and completion. Despite the variety of projects involving different groups of the local population within the three case study villages in close vicinity, all these are set in the same socio-cultural context which allows direct comparison.Drawing on selected theoretical sociological explanations of the dimensions and determinants of people's participation, the conceptual and analytical framework is focussed on an actor-oriented approach using the theoretical concept of the "linking loops" to explicate the complex dynamics evolving in Thai rural society when engaged in local-level planning.By their mechanism of initiation, as explained in terms of the "linking loops" concept, three categories of projects are distinguished including village initiated projects (VIP) and government initiated projects (GIP) as well as projects initiated by villagers with a view to securing government support (V/GP). The comparison of hypothesis test results by project category leads to their classification as explanations of narrow range pertaining to one project category only, of medium range covering two out of the three categories, or of wide range rendering explanations valid for all three project categories.Six general hypotheses and their sub-hypotheses addressing as many stages of the project cycle are tested.
